As a kind of online learning, micro-lectures are getting more and more extensive research and practical application in domestic and foreign colleges and universities. According to the new requirements of higher education teaching reform, this paper focuses on the professional curriculum setting and teaching characteristics of colleges and universities, from the composition of micro-lectures, the necessity of application in micro-lectures, the precautions in the construction of micro-lectures, and the requirements for teachers' information application ability are analyzed, and finally the countermeasures to promote the construction and application of micro-lectures resources in colleges and universities are obtained, hoping to provide reference for related research.

To explore the effect of acacetin on myocardial mitochondrial dysfunction in spontaneously hypertensive rats (SHR) with insulin resistance (IR), and the possible mechanism. Rapid IR was first induced in fructose-fed SHR, and they were then treated with acacetin (25, 50 mg/kg). After 7 weeks, the rats were tested for hypertension, IR, cardiac function, and mitochondrial damage status. The purpose of this study was to analyze the influencing factors of frailty in elderly patients with type II diabetes. 332 elderly patients with type II diabetes admitted to our hospital from May 2018 to December 2019 were selected as the study subjects. The degree of frailty was evaluated by the Fried fragility phenotype scale, and the general information and the clinical data of patients were collected by inquiry and questionnaire survey. After that, all the data were analyzed by SPSS 20.0. Various time‐frequency (T‐F) masks are being applied to sound source localization tasks. Moreover, deep learning has dramatically advanced T‐F mask estimation. However, existing masks are usually designed for speech separation tasks and are suitable only for single‐channel signals. A novel complex‐valued T‐F mask is proposed that reserves the head‐related transfer function (HRTF), customized for binaural sound source localization. In addition, because the convolutional neural network that is exploited to estimate the proposed mask takes binaural spectral information as the input and output, accurate binaural cues can be preserved. Compared with conventional T‐F masks that emphasize single speech source–dominated T‐F units, HRTF‐reserved masks eliminate the speech component while keeping the direct propagation path. Thus, the estimated HRTF is capable of extracting more reliable localization features for the final direction of arrival estimation. Hence, binaural sound source localization guided by the proposed T‐F mask is robust under noisy and reverberant acoustic environments. The experimental results demonstrate that the new T‐F mask is superior to conventional T‐F masks and lead to the better performance of sound source localization in adverse environments.

Due to the high dimensionality of hyperspectral images (HSIs), more training samples are needed in general for better classification performance. However, surface materials cannot always provide sufficient training samples in practice. HSI classification with small size training samples is still a challenging problem. Multiview learning is a feasible way to improve the classification accuracy in the case of small training samples by combining information from different views. This article proposes a new spatial window-based multiview intact feature learning method (SWMIFL) for HSI classification. In the proposed SWMIFL, multiple features that reflect different information of the original image are extracted and spatial windows are imposed on training samples to select unlabeled samples. Then, multiview intact feature learning is performed to learn the intact feature of the training and unlabeled samples. Considering that neighboring samples are likely to belong to the same class, labels of spatial neighboring samples are determined by two factors including the labels of training samples that locate in the spatial window and the labels learned from the intact feature. Finally, unlabeled samples that have same labels under these two factors are treated as new training samples. Experimental results demonstrate that the proposed SWMIFL-based classification method outperforms several well-known HSI classification methods on three real-world data sets.

We postulated that pseudoephedrine nanoparticles (PSE-NPs) with high bioavailability could overcome this limitation. The defensive function of PSE-NPs nanoparticles against adriamycin-induced reproductive toxicity in mice was studied.Methods: We encapsulated PSE in polylactide-polyglycolide nanoparticles (PLGA-NPs) and verified their protective activity against testicular injury in vivo and in vitro.Results: We report a promising delivery system that loads PSE into PLGA-NPs and finally assembles it into a nanocomposite particle. In vitro, PSE-NPs reduced the adriamycin-induced apoptosis of GC-1 cells significantly, improved mitochondrial energy metabolism and promoted expression of the proteins related to the gonadotropin-releasing hormone (GnRh) receptor signaling pathway. In vivo, evaluation of sperm indices and histology showed that adriamycin could induce testicular toxicity. PSE-NPs significantly increased the sperm motility of mice, reduced the percent apoptosis and oxidative stress of testes, increased serum levels of GnRh, activated the GnRhR signaling pathway in testes and promoted expression of meiosis-related factors.Conclusion: In view of their safety and efficiency, these PSE-NPs have potential applications in alleviating adriamycin-induced reproductive toxicity.Graphical Abstract: Keywords: pseudoephedrine nanoparticles, adriamycin, reproductive toxicity, GnRhR signaling pathway, sperm meiosis

The aim of the work was to investigate the effects of acacetin on endothelial dysfunction and aortic fibrosis in insulin-resistant SHR rats and explore its mechanism. Seven-week-old male spontaneously hypertensive rats (SHR) were selected to establish a rat model of hypertension with insulin resistance induced by 10% fructose. The nuclear factor kappa B p65 (NF-κB p65) and Collagen I were observed by Immunohistochemistry. Immunofluorescence was used to observe estrogen receptor-alpha (ERα), estrogen receptor-beta (ERβ), and G protein-coupled receptor 30 (GPR30). Western blotting was used to detect interleukin (IL-1β), Arginase 2 (ARG2), Nostrin, endothelial nitric oxide synthase (eNOS), TGF-β, Smad3, ERK pathway proteins such as p-c-Raf, p-MEK1/2, p-ERK, ERK, p-P90RSK and p-MSK1. We found that acacetin did have an improvement on endothelial dysfunction and fibrosis. Meanwhile, it was also found to have a significant effect on the level of estrogen in this model by accident. Then, the experiment of uterine weight gain in mice confirmed that acacetin had a certain estrogen-like effect in vivo and played its role through the estrogen receptors pathway. In vitro experience HUVEC cells were stimulated with 30 mM/L glucose and 100 mM/L NaCl for 24 h to establish the endothelial cell injury model. HUVEC cells were treated with 1 μM/L estrogen receptors antagonist (ICI 182780) for 30 min before administration. Cell experiments showed that acacetin could reduce the apoptosis of HUVEC cells, the levels of inflammatory cytokines and the expression of TGF-β, Collagen I and Smad3 in endothelial cell injury model. After treatment with ICI 182780, the improvement of acacetin was significantly reversed. The results showed that acacetin relieved endothelial dysfunction and reduced the aortic fibrosis in insulin-resistant SHR rats by reducing the release of inflammatory factors and improving vasodilatory function through estrogen signaling pathway.

Objectives To investigate the effects of geniposide in an iridoid found in Gardenia jasminoides var. radicans Makino (GJRM) in spontaneous hypertensive rat (SHR) and explore the possible mechanisms. Methods In this study, we detected the content of geniposide in GJRM by high-performance liquid chromatography (HPLC). Then, we used acute diuretic experiments to determine whether geniposide has diuretic effect. Moreover, we carried out experiments on SHR to further study the mechanism of hypertension, while real-time PCR, Western blot and immunohistochemistry were used for the experiments in vivo test. Hypotonic model was used for in vitro test. Key findings Our data showed that the content of geniposide in the extract of GJRM is 27.54%. Meanwhile, 50 mg/kg geniposide showed the strongest effect on promoting urine volume. Further study indicated that the extract of GJRM and geniposide could significantly reduce blood pressure and promote the excretion of urine and Na+ in SHR. In addition, geniposide significantly inhibited the activation of the with-no-lysine kinase (WNK) signalling pathway and significantly increases the protein expressions of estrogen receptor α (ERα), estrogen receptor β (ERβ) and G protein-coupled receptor 30 (GPR30) in SHR. In hypotonic model, geniposide significantly inhibits the phosphorylation of NKCC and NCC and could be antagonistic to estrogen receptor antagonists. Conclusions Collectively, we would suggest that geniposide may potentially be utilized as an adjunct to existing thiazide and thiazide-like diuretics to control hypertension, mainly through inhibiting the activation of the WNK signalling pathway mediated by the estrogen receptor.

In binaural sound source localisation, front–back confusion is often the challenging problem when localising sources in the noisy or reverberant environments. Hence, a novel algorithm fusing deep and convolutional neural network (CNN) is proposed to address this issue. First, joint features, which consist of interaural level differences (ILDs) and cross-correlation function (CCF) within a lag range, are extracted from binaural signals. Second, with the extracted CCF–ILD features, CNN is used for the front–back classification task, while deep neural network is used for azimuth classification task. The front–back features extracted by the CNN can be leveraged as additional information for the sound source localisation task. Also, an angle-loss function is designed to avoid the overfitting problem and to improve the generalisation ability of this method in adverse acoustic conditions. Finally, two branches are concatenated and then followed by an output layer, which generates the posterior probability of azimuth angles, and the azimuth corresponding to the maximum posterior probability is chosen as the direction of sound source. Experimental results demonstrate the effectiveness of the authors’ method for front–back decision and azimuth estimation in noisy and reverberant environments.

Sparse additive models have shown competitive performance for high-dimensional variable selection and prediction due to their representation flexibility and interpretability. Despite their theoretical properties have been studied extensively, few works have addressed the robustness for the sparse additive models. In this paper, we employ the robust average top-k (ATk) loss as classification error measure and propose a new sparse algorithm, named ATk group sparse additive machine (ATk-GSAM). Besides the robust concern, the ATk-GSAM has well adaptivity by integrating the data dependent hypothesis space and group sparse regularizer together. Generalization error bound is established by the concentration estimate with empirical covering numbers. In particular, our error analysis shows that ATk-GSAM can achieve the learning rate O ( n − 1 / 2 ) under appropriate conditions. We further analyze the robustness of ATk-GSAM via a sample-weighted procedure interpretation, and the theoretical guarantees on grouped variable selection. Experimental evaluations on both simulated and benchmark datasets validate the effectiveness and robustness of the new algorithm.

Context In the antihypertensive study of phenylacetamide (PA) on spontaneously hypertensive rats (SHR), it was occasionally found that PA prevents myocardial injury. Objective Clarify the protective mechanism of PA on myocardial injury in SHR rats. Materials and methods In vivo, SHR rats were treated with or without PA (15, 30, 45 mg/kg) for 3 weeks (12 per group). In vitro, H9c2 cells were treated with PA (1, 5, 10 μM) for 24 h, and then stimulated with H2O2 (300 μM) for 4 h. Molecular mechanisms were explored through cardiac pathology, cardiac function and biochemical markers. Results In vivo, PA (15, 30, 45 mg/kg) reduced CVF from 14.8 ± 1.62 to 9.94 ± 1.56, 8.6 ± 1.33, 8.14 ± 1.45%; increased the LVEF relative level from 0.8 ± 0.06 to 0.83 ± 0.04, 0.86 ± 0.05, 0.9 ± 0.04. All three doses can improve the cardiac pathological structure and function (LVEDD, LVESD, LVFS, heart index, NT-proBNP, CKMB, SBP); however, 45 mg/kg works best. But different doses show different molecular mechanisms. PA (15 mg/kg) improves RAAS system (REN, ACE), inflammation (ET-1, IL-1β) and MAPK pathway (p-ERK/ERK, p-JNK/JNK) better. PA (45 mg/kg) improves oxidative stress (SOD, NOX1) and TGF-β pathway (Smad3) better. In vitro, PA improved cell viability, oxidative stress (SOD, NOX1) and Smad3 protein expression. Discussion and conclusions: PA regulates different mechanisms at different concentrations to improve myocardial injury, and high dose is the best. This experiment provides a theoretical basis for the development of new clinical drugs for cardiovascular disease.

Lipid deposition in the kidney can cause serious damage to the kidney, and there is an obvious epithelial–mesenchymal transition (EMT) and fibrosis in the late stage. To investigate the interventional effects and mechanisms of phenolic compounds from Mori Cortex on the EMT and fibrosis induced by sodium oleate-induced lipid deposition in renal tubular epithelial cells (NRK-52e cells), and the role played by CD36 in the adjustment process, NRK-52e cells induced by 200 μmol/L sodium oleate were given 10 μmoL/L moracin-P-2″-O-β-d-glucopyranoside (Y-1), moracin-P-3′-O-β-d-glucopyranoside (Y-2), moracin-P-3′-O-α-l-arabinopyranoside (Y-3), and moracin-P-3′-O-[β-glucopyranoside-(1→2)arabinopyranoside] (Y-4), and Oil Red O staining was used to detect lipid deposition. A Western blot was used to detect lipid deposition-related protein CD36, inflammation-related protein (p-NF-κB-P65, NF-κB-P65, IL-1β), oxidative stress-related protein (NOX1, Nrf2, Keap1), EMT-related proteins (CD31, α-SMA), and fibrosis-related proteins (TGF-β, ZEB1, Snail1). A qRT-PCR test detected inflammation, EMT, and fibrosis-related gene mRNA levels. The TNF-α levels were detected by ELISA, and the colorimetric method was used to detects SOD and MDA levels. The ROS was measured by flow cytometry. A high-content imaging analysis system was applied to observe EMT and fibrosis-related proteins. At the same time, the experiment silenced CD36 and compared the difference between before and after drug treatment, then used molecular docking technology to predict the potential binding site of the active compounds with CD36. The research results show that sodium oleate can induce lipid deposition, inflammation, oxidative stress, and fibrosis in NRK-52e cells. Y-1 and Y-2 could significantly ameliorate the damage caused by sodium oleate, and Y-2 had a better ameliorating effect, while there was no significant change in Y-3 or Y-4. The amelioration effect of Y-1 and Y-2 disappeared after silencing CD36. Molecular docking technology showed that the Y-1 and Y-2 had hydrogen bonds to CD36 and that, compared with Y-1, Y-2 requires less binding energy. In summary, moracin-P-2″-O-β-d-glucopyranoside and moracin-P-3′-O-β-d-glucopyranoside from Mori Cortex ameliorated lipid deposition, EMT, and fibrosis induced by sodium oleate in NRK-52e cells through CD36.

The purpose of this study was to evaluate the diagnostic performance of single-parameter, unimodal and bimodal magnetic resonance imaging (MRI) in differentiating tumor recurrence (TR) from radiation necrosis (RN) in patients with glioblastoma (GBM) after treatment using diffusion-weighted imaging (DWI), diffusion tensor imaging (DTI), dynamic susceptibility contrast enhancement-perfusion weighted imaging (DSC-PWI), and proton magnetic resonance spectroscopy (Patients with histologically proven GBM who underwent surgical intervention followed by chemoradiotherapy and developed a new, progressively enhanced lesion on follow-up MRI were included in our study. Subsequently, DWI, DTI, DSC-PWI, andThere were significant differences between the TR and RN groups in terms of ADC (p = 0.001), rADC (p0.001), FA (p = 0.001), DA (p = 0.003), DR (p = 0.003), rCBV (p0.001), rCBF (p0.001), Cho/NAA (p0.001), Lac/Cr (p0.001) and Lip/Cr (p0.001). ROC analysis suggested that rCBV, MRS, and DSC + MRS were the optimal single-parameter, unimodal, and bimodal MRI classifiers for distinguishing TR from RN, with AUC values of 0.909, 0.940, and 0.994, respectively.The combination of parameters based on multiparametric MRI in the region of enhanced lesions is a valuable noninvasive tool for discriminating TR from RN.

Carboxymethyl chitosan (CMCh), an ampholetic chitosan derivative, has found broad applications in the food industry. However, its cryo-protective properties remained less explored compared to other viscous polysaccharides, such as carboxymethyl cellulose, carrageenan etc., which have been widely utilized as frozen food additives. In this study, we investigated the effect of CMCh addition to frozen dough in terms of water state, protein structure, and the textural properties of prepared frozen dumpling wrappers. Results indicated that CMCh restricted the water migration in dough and delayed protein deterioration during frozen storage. Specifically, the content of freezable water in dough was reduced and the water distribution became more uniform as reflected by DCS and LF-NMR analysis. CMCh also stabilized disulfide bond and secondary structures of the protein, leading to inhibition of dough rheology changes. Accordingly, the obtained frozen dumplings wrappers demonstrated decreased cracking rate and water loss, and improved textural properties. Moreover, CMCh with higher degree of carboxymethyl substitution (DS: 1.2, CMCh-B) exhibited better cryo-protective effects compared to CMCh of lower DS (DS: 0.8, CMCh-A). Our study provides novel insights and scientific basis for the development of ampholetic polysaccharides as high-performance food additives.

Doxorubicin (DOX) is a highly effective chemotherapeutic that is effective for various tumours. However, the clinical application of DOX has been limited by adverse reactions such as cardiotoxicity and heart failure. Since DOX-induced cardiotoxicity is irreversible, drugs to prevent DOX-induced cardiotoxicity are needed.This study aimed to investigate the effect of total flavonoids of Selaginella tamariscina (P.Beauv.) Spring (TFST) on doxorubicin-induced cardiotoxicity.The present study established DOX-induced cardiotoxicity models in C57BL/6 mice treated with DOX (cumulative dose: 20 mg/kg body weight) and H9c2 cells incubated with DOX (1 μM/l) to explore the intervention effect and potential mechanism of TFST. Echocardiography was performed to evaluate left ventricular functions. Heart tissue samples were collected for histological evaluation. Myocardial injury markers and oxidative stress markers were examined. Mitochondrial energy metabolism pathway associated proteins PPARα/PGC-1α/Sirt3 were detected. We also explored the effects of TFST on endoplasmic reticulum (ER) stress and apoptosis. To further investigate the protective mechanism of TFST, we used the specific small interfering RNA MFN2 (siMFN2) to explore the effect of MFN2 on TFST against DOX-induced cardiotoxicity in vitro. Flow cytometry detected reactive oxygen species, mitochondrial membrane potential and apoptosis. Cell mitochondrial stress was measured by Seahorse XF analyser.Both in vivo and in vitro studies verified that TFST observably alleviated DOX-induced mitochondrial dysfunction and ER stress. However, these effects were reversed after transfected siMFN2.Our results indicated that TFST ameliorates DOX-induced cardiotoxicity by alleviating mitochondrial dysfunction and ER stress by activating MFN2/PERK. MFN2/PERK pathway activation may be a novel mechanism to protect against DOX-induced cardiotoxicity.

Cross Version Defect Prediction (CVDP) is a practical scenario by training the classification model on the historical data of the prior version and then predicting the defect labels of modules in the current version. Unfortunately, the differences of data distribution across versions may hinder the effectiveness of the trained CVDP model. Thus, it is not trivial to select a suitable training subset from the prior version to promote the CVDP performance. In this paper, we propose a novel method, called Two-Stage Training Subset Selection (TSTSS), to address this challenging issue. In the first stage, TSTSS utilizes a sparse modeling representative selection method to select an initial module subset from the prior version which can well reconstruct the data of the prior version. In the second stage, TSTSS leverages a dissimilarity-based sparse subset selection method to further refine the selected module subset, which enables the selected modules to well represent the modules of the current version. Finally, we use a novel weighted extreme learning machine classifier to construct the CVDP model. We evaluate the CVDP performance of TSTSS on 50 cross-version pairs using 6 indicators. The experiments show that TSTSS can efficiently improve the CVDP performance compared with 11 baseline methods.

A key issue of load-carrying exoskeleton is to maintain the stability of gait during walking and prevent the wearer from falling over. Some of current strategies were to maintain ZMP (zero-moment point) in the support area, which had been widely used in humanoid robot. However, this method would cause serious interference between body and exoskeleton. because the body often gets energy-saving and rapid walking through unstable conditions during human walking. A control strategy for load-carrying exoskeleton was presented in this paper, which maintained the consistency of body ZMP and exoskeleton ZMP. This control strategy could reduce body-exoskeleton interference while maintaining the stability of walking. In this paper, we firstly introduced the experimental environment and the ZMP detection method, and then designed a computational model of the body-exoskeleton interaction force and evaluation criterion for gait stability. Then, a control strategy based on ZMP was designed. Finally, experiments showed that the control strategy could effectively reduce the body-exoskeleton interference, and maintained the stability of wearer’s walking.

Context Software defect prediction strives to detect defect-prone software modules by mining the historical data. Effective prediction enables reasonable testing resource allocation, which eventually leads to a more reliable software. Objective The complex structures and the imbalanced class distribution in software defect data make it challenging to obtain suitable data features and learn an effective defect prediction model. In this paper, we propose a method to address these two challenges. Method We propose a defect prediction framework called KPWE that combines two techniques, i.e., Kernel Principal Component Analysis (KPCA) and Weighted Extreme Learning Machine (WELM). Our framework consists of two major stages. In the first stage, KPWE aims to extract representative data features. It leverages the KPCA technique to project the original data into a latent feature space by nonlinear mapping. In the second stage, KPWE aims to alleviate the class imbalance. It exploits the WELM technique to learn an effective defect prediction model with a weighting-based scheme. Results We have conducted extensive experiments on 34 projects from the PROMISE dataset and 10 projects from the NASA dataset. The experimental results show that KPWE achieves promising performance compared with 41 baseline methods, including seven basic classifiers with KPCA, five variants of KPWE, eight representative feature selection methods with WELM, 21 imbalanced learning methods. Conclusion In this paper, we propose KPWE, a new software defect prediction framework that considers the feature extraction and class imbalance issues. The empirical study on 44 software projects indicate that KPWE is superior to the baseline methods in most cases.

Background: Prototype foamy virus (PFV) is a complex and unique retrovirus with the longest genome among the retroviruses and is used as a vector for gene therapies. The viral Tas protein transactivates the viral long terminal repeat promoter and is required for viral replication. We have utilized RNA sequencing to identify and characterize the long-noncoding RNA NONHSAG000101 (lnc-NONH), which markedly increases in PFV-infected cells. However, little is known about the function of lnc-NONH. Objectives: We aim to explore the role of lnc-NONH during PFV infection. Methods: To assess the lnc-NONH role during PFV infection, the siRNAs were used to silence the lnc-NONH expression. The microRNA (miRNA) mimic and inhibitor were employed to explore the function of lnc-NONH-related miRNA miR-34c-5p. Quantitative real-time polymerase chain reaction assay and Western blotting were applied to measure the mRNA and protein levels of PFV transactivator Tas. Luciferase assay was used to determine the transcriptional activity of the PFV unique internal promoter (IP). Results: lnc-NONH promotes the expression of PFV Tas and miR-34c-5p. The interaction between lnc-NONH and miR-34c-5p enhances the transcriptional activity of the PFV IP. Conclusions: In the current study, we report a novel mechanism for the lnc-NONH-mediated upregulation of Tas expression. Our findings contribute to the understanding of regulatory network of Tas expression and PFV replication.

The aim of present study is to investigate the effects and mechanisms of geniposide on myocardial injuries in spontaneously hypertensive rats (SHR) and H9c2 cells induced by NaCl solution.Materials and Methods: Male Wistar–Kyoto (WKY) and SHR rats were given different doses of geniposide (25 mg/kg/d or 50 mg/kg/d) or distilled water for three consecutive weeks. Meanwhile, an H9c2 cell line-injury model was established using a solution of 150 μmol/L NaCl for 8 h. The cardiac function and related indexes of rats were detected.Results: The results showed that geniposide decreased the levels of COI and COIII, which promoted the phosphorylation of AMPK (p-AMPK) and enhanced the energy metabolism pathway. Geniposide improved myocardial apoptosis by regulating apoptotic proteins (p38, BAX and Bcl-2). Finally, heart function was regulated, and the markers of myocardial injury were decreased. Geniposide increased the viability of H9c2 cells treated with the NaCl solution and decreased the rate of apoptosis by regulating the levels of apoptotic proteins. Geniposide could activate energy metabolism signalling pathway (AMPK/SirT1/FOXO1) and reduce H9c2 cell apoptosis.Conclusion: Our results showed that the mechanisms by which geniposide improves myocardial injury in SHR may be through regulating the energy metabolism signalling pathway (AMPK/SirT1/FOXO1) and improving myocardial apoptosis by regulating apoptotic proteins.Keywords: geniposide, SHR, myocardial injury, apoptosis, energy metabolism
