Anthropogenic activities have escalated CH4 emissions, exacerbating global warming, yet specialized bacteria known as Methanotrophs play a key role in mitigating atmospheric CH4 levels by consuming 30-70% of emitted methane. This study focuses on exploring the culturable methanotrophic population within Muthukuda mangrove sediments, an unexplored reservoir of methanotrophic diversity. The sediment sample yielded a methanotrophic bacterial count of 1.5 x 103 CFU/g, leading to the selection of three unique bacterial morphotypes (NCT270, NCT271, and NCT272) for in-depth investigation. Optimal growth was observed at pH 8, with peak growth at 30°C, while extreme temperatures of 4°C and 40°C inhibited growth across all isolates. Salinity levels between 20 and 30 ppt supported optimal growth, with strains displaying tolerance to various stressors. Methane served as the sole carbon source for all experiments, with positive urease production noted after 7 days of incubation. Microscopic and biochemical analyses suggested the classification of strains NCT270, NCT271, and NCT272 within Group I methanotrophic genera: Methylomicrobium, Methyloscarcina, and Methylomonas, respectively. BLASTn analysis of 16S rRNA gene sequences shared high similarities with known methanotrophic species Methyloscarcina fibrate (ON834586) with 99.28%, Methylomicrobium album (ON834587) with 98.77% and Methylomonas methanica (ON834588) with 99.15%. The resulting insights enhance our understanding of culturable methanotrophic diversity and underscore its potential for environmental applications.

Anaerobic microbial sulfate reduction and oxidative sulfur cycling have been studied in long sediment cores mainly acquired as part of IODP explorations. The most remarkable observation in many of these studies is the existence of an active sulfur cycle in the deep subsurface sediments that have very low organic carbon content and are presumably refractory. Here we investigate the interstitial sulfate concentrations and sulfur isotope ratios in a 290 m long core collected from the eastern Arabian Sea at a water depth of 2663 m. Continuous decrease in pore water-sulfate concentrations with depth (up to 75 mbsf) coupled with enrichment in δ34SSO4 values suggests organoclastic sulfate reduction (OSR) processes attributed to the activity of sulfate-reducing bacteria (SRB) and retention of labile organic substrates amenable to the SRBs. Below a depth of 75 mbsf, the absence of a further reduction in sulfate concentrations indicates insufficient labile substrate to drive SRB. An increase in sulfate concentrations at the deeper subsurface (below 128.5 mbsf) coupled with decreasing δ34SSO4 values may be attributed to a ferric-oxyhydroxide driven oxidation of Fe-sulfide. This study reveals that even under deep aerobic water columns, organic matter may continue to be a source of labile organic substrates at significantly deeper subsurface. Enhanced sulfate concentrations in the deeper depths may be attributed to the oxidation of sulfides via ferric-oxyhydroxides buried deep within the sediment. Kinetics of thiosulfate oxidation, product and intermediate formation, and 34S fractionation, were studied for the members of Alphaproteobacteria Paracoccus sp. SMMA5 and Mesorhizobium thiogangeticum SJTT, the Betaproteobacteria member Pusillimonas ginsengisoli SBO3, and the Acidithiobacillia member Thermithiobacillus sp. SMMA2, during chemolithoautotrophic growth in minimal salts media supplemented with 20 mM thiosulfate. The two Alphaproteobacteria oxidized thiosulfate directly to sulfate, progressively enriching the end-product with 34S; Δ34Sthiosulfate-sulfate values recorded at the end of the two processes (when no thiosulfate was oxidized any further) were −2.9‰ and −3.5‰, respectively. Pusillimonas ginsengisoli SBO3 and Thermithiobacillus sp. SMMA2, on the other hand, oxidized thiosulfate to sulfate via tetrathionate intermediate formation, with progressive 34S enrichment in the end-product sulfate throughout the incubation period; Δ34Sthiosulfate-sulfate, at the end of the two processes (when no further oxidation took place), reached −3.5‰ and −3.8‰, respectively. Based on similar 34S fractionation patterns recorded previously during thiosulfate oxidation by strains of Paracoccus pantotrophus, Advenella kashmirensis and Hydrogenovibrio crunogenus , it was concluded that progressive reverse fractionation, enriching the end-product sulfate with 34S, could be a characteristic signature of bacterial thiosulfate oxidation. To explore the potential role of tetrathionate in the sulfur cycle of marine sediments, population ecology of microorganisms capable of metabolizing this polythionate was revealed at 15-30 cm resolution along two, ~ 3-m-long, cores collected from 530 and 580 meters below the sea level, off India's west coast, within the oxygen minimum zone (OMZ) of the Arabian Sea. Metagenome analysis along the two sediment-cores revealed widespread occurrence of genes involved in microbial formation, oxidation, and reduction of tetrathionate; high diversity and relative-abundance was also detected for bacteria that are known to render these metabolisms in vitro. Results of slurry-incubation of the sediment-samples in thiosulfate- or tetrathionate-containing microbial growth media, data obtained via pure-culture isolation, and finally metatranscriptome analyses, corroborated the in situ functionality of tetrathionate-forming, oxidizing, and reducing microorganisms. Geochemical analyses revealed the presence of up to 11.1 μM thiosulfate along the two cores, except a few sample-sites near the sediment-surface. Thiosulfate oxidation by chemolithotrophic bacteria prevalent in situ is the apparent source of tetrathionate in this ecosystem. However, potential abiotic origin of the polythionate can neither be ruled out nor confirmed from the geochemical information currently available for this territory. Tetrathionate, in turn, can be either oxidized to sulfate (via oxidation by the chemolithotrophs present) or reduced back to thiosulfate (via respiration by native bacteria). Up to 2.01 mM sulfide present in the sediment-cores may also reduce tetrathionate abiotically to thiosulfate and elemental sulfur. As tetrathionate was not detected in situ, high microbiological and geochemical reactivity of this polythionate was hypothesized to be instrumental in its cryptic status as a central sulfur cycle intermediate. To explore the potential role of tetrathionate in the sulfur cycle of marine sediments, the population ecology of tetrathionate-forming, oxidizing, and respiring microorganisms was revealed at 15-30 cm resolution along two, ~ 3-m-long, cores collected from 530- and 580-mbsl water-depths of Arabian Sea, off India's west coast, within the oxygen minimum zone (OMZ). Metagenome analysis along the two sediment-cores revealed widespread occurrence of the structural genes that govern these metabolisms; high diversity and relative-abundance was also detected for the bacteria known to render these processes. Slurry-incubation of the sediment-samples, pure-culture isolation, and metatranscriptome analysis, corroborated the in situ functionality of all the three metabolic-types. Geochemical analyses revealed thiosulfate (0-11.1 µM), pyrite (0.05-1.09 wt %), iron (9232-17234 ppm) and manganese (71-172 ppm) along the two sediment-cores. Pyrites (via abiotic reaction with MnO2) and thiosulfate (via oxidation by chemolithotrophic bacteria prevalent in situ) are apparently the main sources of tetrathionate in this ecosystem. Tetrathionate, in turn, can be either converted to sulfate (via oxidation by the chemolithotrophs present) or reduced back to thiosulfate (via respiration by native bacteria); 0-2.01 mM sulfide present in the sediment-cores may also reduce tetrathionate abiotically to thiosulfate and elemental sulfur. Notably tetrathionate was not detected in situ - high microbiological and geochemical reactivity of this polythionate is apparently instrumental in the cryptic nature of its potential role as a central sulfur cycle intermediate. Biogeochemical roles of this polythionate, albeit revealed here in the context of OMZ sediments, may well extend to the sulfur cycles of other geomicrobiologically-distinct marine sediment horizons. Abstract: The distribution of sediment components, organic carbon, aluminium normalized metals, bulk sedimentary organic stable carbon isotope ratios i.e. δ13Corg and elemental TOC/TN ratios were studied within the sediment of Vaitarna estuary. Deposition of coarser sediment was favored under higher energy conditions prevailing in the past. Prevalence of relatively calm environment in recent years favored deposition of finer particles. Changes in depositional environment of Vaitarna estuary were very well reflected in distribution pattern of metals, δ13Corg and TOC/TN ratios. The narrow range (−22.9‰ to −22.0‰) and absence of distinct trend for δ13Corg in core collected towards the mouth (core S4) was attributed to prevalence of higher hydrodynamic energy conditions at this location. While protected environment of core S9 indicated a gradual change in the proportion of organic matter input to the estuary over a period of time. δ13Corg values in this core increased from −28.4‰ at the bottom to −20.7‰ at the surface and showed a corresponding decrease in TOC/TN ratio. All these changes will significantly alter the biogeochemical processes within the Vaitarna estuary and adjoining coastal waters. [Copyright &y& Elsevier]

During chemolithoautotrophic thiosulfate oxidation, the phylogenetically diverged proteobacteria Paracoccus pantotrophus, Tetrathiobacter kashmirensis, and Thiomicrospira crunogena rendered steady enrichment of 34S in the end product sulfate, with overall fractionation ranging between -4.6% and+5.8%. The fractionation kinetics of T. crunogena was essentially similar to that of P. pantotrophus, albeit the former had a slightly higher magnitude and rate of 34S enrichment. In the case of T. kashmirensis, the only significant departure of its fractionation curve from that of P. pantotrophus was observed during the first 36 h of thiosulfate-dependent growth, in the course of which tetrathionate intermediate formation is completed and sulfate production starts. The almost-identical 34S enrichment rates observed during the peak sulfate-producing stage of all three processes indicated the potential involvement of identical S-S bond-breaking enzymes. Concurrent proteomic analyses detected the hydrolase SoxB (which is known to cleave terminal sulfone groups from SoxYZ-bound cysteine S-thiosulfonates, as well as cysteine S-sulfonates, in P. pantotrophus) in the actively sulfate-producing cells of all three species. The inducible expression of soxB during tetrathionate oxidation, as well as the second leg of thiosulfate oxidation, by T. kashmirensis is significant because the current Sox pathway does not accommodate tetrathionate as one of its substrates. Notably, however, no other Sox protein except SoxB could be detected upon matrix-assisted laser desorption ionization mass spectrometry analysis of all such T. kashmirensis proteins as appeared to be thiosulfate inducible in 2-dimensional gel electrophoresis. Instead, several other redox proteins were found to be at least 2-fold overexpressed during thiosulfate- or tetrathionate-dependent growth, thereby indicating that there is more to tetrathionate oxidation than SoxB alone.
