Abstract

The body louse, Pediculus humanus humanus, is an obligate blood-feeding ectoparasite and an important insect vector that mediates the transmission of diseases to humans. The analysis of the body louse genome revealed a drastic reduction of the chemosensory gene repertoires when compared to other insects, suggesting specific olfactory adaptations to host specialization and permanent parasitic lifestyle. Here, we present for the first time functional evidence for the role of odorant receptors (ORs) in this insect, with the objective to gain insight into the chemical ecology of this vector. We identified seven putative full-length ORs, in addition to the odorant receptor co-receptor (Orco), and expressed four of them in the Xenopus laevis oocytes system. When screened with a panel of ecologically-relevant odorants, PhumOR2 responded to a narrow set of compounds. At the behavior level, both head and body lice were repelled by the physiologically-active chemicals. This study presents the first evidence of the OR pathway being functional in lice and identifies PhumOR2 as a sensitive receptor of natural repellents that could be used to develop novel efficient molecules to control these insects.

Abstract

Oviposition attractants are environmental cues that allow Culex gravid female mosquitoes to locate suitable sites for egg-laying and, therefore, may be exploited for environmentally friendly strategies for controlling mosquito populations. Naturally occurring skatole has been identified as an oviposition attractant for the Southern House mosquito, Culex quinquefasciatus. Previously, we identified in Cx. quinquefasciatus female antennae an olfactory receptor neuron (ORN) highly sensitive to skatole and an odorant-binding protein involved in the detection of this semiochemical. Here, we describe the characterization of an odorant receptor (OR), CquiOR10, which is narrowly tuned to skatole when expressed in the Xenopus oocyte system. Odorant-induced response profiles generated by heterologously expressed CquiOR10 suggest that this OR is expressed in the mosquito ORN sensitive to skatole. However, geranylacetone, which stimulates the antennal ORN, was not detected by CquiOR10-expressing oocytes, thus raising interesting questions about reception of oviposition attractants in mosquitoes.

Abstract

Odorant-binding proteins (OBPs) were discovered almost three decades ago, but there is still considerable debate regarding their role(s) in insect olfaction, particularly due to our inability to knockdown OBPs and demonstrate their direct phenotypic effects. By using RNA interference (RNAi), we reduced transcription of a major OBP gene, CquiOBP1, in the antennae of the Southern house mosquito, Culex quinquefasciatus. Previously, we had demonstrated that the mosquito oviposition pheromone (MOP) binds to CquiOBP1, which is expressed in MOP-sensitive sensilla. Antennae of RNAi-treated mosquitoes showed significantly lower electrophysiological responses to known mosquito oviposition attractants than the antennae of water-injected, control mosquitoes. While electroantennogram (EAG) responses to MOP, skatole, and indole were reduced in the knockdowns, there was no significant difference in the EAG responses from RNAi-treated and water-injected mosquito antennae to nonanal at all doses tested. These data suggest that CquiOBP1 is involved in the reception of some oviposition attractants, and that high levels of OBPs expression are essential for the sensitivity of the insect’s olfactory system.

Abstract

BackgroundThe navel orangeworm, Amyelois transitella Walker (Lepidoptera: Pyralidae), is the most serious insect pest of almonds and pistachios in California for which environmentally friendly alternative methods of control--like pheromone-based approaches--are highly desirable. Some constituents of the sex pheromone are unstable and could be replaced with parapheromones, which may be designed on the basis of molecular interaction of pheromones and pheromone-detecting olfactory proteins.MethodologyBy analyzing extracts from olfactory and non-olfactory tissues, we identified putative olfactory proteins, obtained their N-terminal amino acid sequences by Edman degradation, and used degenerate primers to clone the corresponding cDNAs by SMART RACE. Additionally, we used degenerate primers based on conserved sequences of known proteins to fish out other candidate olfactory genes. We expressed the gene encoding a newly identified pheromone-binding protein, which was analyzed by circular dichroism, fluorescence, and nuclear magnetic resonance, and used in a binding assay to assess affinity to pheromone components.ConclusionWe have cloned nine cDNAs encoding olfactory proteins from the navel orangeworm, including two pheromone-binding proteins, two general odorant-binding proteins, one chemosensory protein, one glutathione S-transferase, one antennal binding protein X, one sensory neuron membrane protein, and one odorant receptor. Of these, AtraPBP1 is highly enriched in male antennae. Fluorescence, CD and NMR studies suggest a dramatic pH-dependent conformational change, with high affinity to pheromone constituents at neutral pH and no binding at low pH.

16. Ntpdase1 modulates smooth muscle contraction in mice bladder by regulating nucleotide receptor activation distinctly in male and female

Abstract

Nucleotides released by smooth muscle cells (SMCs) and by innervating nerve terminals activate specific P2 receptors and modulate bladder contraction. We hypothesized that cell surface enzymes regulate SMC contraction in mice bladder by controlling the concentration of nucleotides. We showed by immunohistochemistry, enzymatic histochemistry, and biochemical activities that nucleoside triphosphate diphosphohydrolase-1 (NTPDase1) and ecto-5′-nucleotidase were the major ectonucleotidases expressed by SMCs in the bladder. RT-qPCR revealed that, among the nucleotide receptors, there was higher expression of P2X1, P2Y1, and P2Y6 receptors. Ex vivo, nucleotides induced a more potent contraction of bladder strips isolated from NTPDase1 deficient (Entpd1−/−) mice compared to wild type controls. The strongest responses were obtained with uridine 5′-triphosphate (UTP) and uridine 5′-diphosphate (UDP), suggesting the involvement of P2Y6 receptors, which was confirmed with P2ry6−/− bladder strips. Interestingly, this response was reduced in female bladders. Our results also suggest the participation of P2X1, P2Y2 and/or P2Y4, and P2Y12 in these contractions. A reduced response to the thromboxane analogue U46619 was also observed in wild type, Entpd1−/−, and P2ry6−/− female bladders showing another difference due to sex. Abstract

L’utilisation de l’analyse économique du droit pour étudier la gouvernance de la Ligue nationale permet de réduire l’erreur humaine lors de la prise de décision. La Ligue nationale est une ligue professionnelle. La maximisation de la valeur des équipes et la quête du profit sont des objectifs économiques importants à réaliser. La gouvernance de la Ligue doit avoir comme priorité d’aider les propriétaires d’équipe à concrétiser la maximisation des objectifs économiques. L’analyse économique du droit peut aider le commissaire et les propriétaires d’équipe à améliorer la gouvernance de la Ligue afin de concrétiser l’objectif de la maximisation de la valeur. Cette étude porte sur l’analyse du poste de commissaire à la gouvernance de la Ligue nationale. Dans une première partie, une présentation de la structure de la Ligue suivie d’une analyse des conditions de marché sont effectuées afin de circonscrire la marge de manœuvre offerte aux décisions de gouvernance. Il est important de comprendre l’effet des règles établies par les gouverneurs de la Ligue au niveau du marché des équipes. Par la suite, dans une deuxième partie, l’analyse du poste de commissaire est effectuée avec l’aide de la théorie du réseau de contrats. La création du poste de commissaire est venue adjoindre une autre partie au réseau de contrats. Les propriétaires assument maintenant le rôle de superviseur tandis que le commissaire s’occupe des activités quotidiennes de la gestion de la Ligue. Le commissaire a intérêt à ce que la Ligue nationale participe activement à la génération de revenus pour les équipes. Il peut arriver des situations où le commissaire a une divergence d’intérêt avec certains propriétaires de la Ligue au niveau de la gouvernance. Empruntées au droit des sociétés, des solutions sont proposées dans la troisième partie afin de mieux assumer l’objectif de la maximisation de la valeur des équipes lors de la prise de décision du commissaire., The use of the economic analysis of law when studying the governance of the National Hockey League reduces human error in the decision-making process. The National Hockey League is a professional league. Value maximisation of the League teams and profits are important economic objectives. As a priority, the League governance must help the team owners to realize these economic objectives. The economic analysis of law can help the Commissioner and the team owners improve the governance of the National Hockey League in order to achieve the objective of value maximisation. This study analyzes the Commissioner’s position within the governance of National Hockey League. In the first part, a description of the National Hockey League structure as well as an analysis of the market conditions will be completed in order to outline the flexibility in governance related decisions. It is important to understand the impact of the regulations established by the Governors of the League related to the team markets. Further, in the second part, an analysis of the Commissioner’s position will be completed based on the theory of the firm. The creation of the Commissioner’s position was added to another element of the theory of the firm. As such, the team owners now assume the role of supervisor while the Commissioner takes care of the daily management of the League. The Commissioner has an interest in ensuring that the National Hockey League contributes actively to the generation of income for the teams. There can be situations where the Commissioner as diverging views with the team owners on governance. Using corporate law, in the third part, solutions will be recommended to better attaint the objective of value maximisation of the League teams in the Commissioner’s decision-making process.

Abstract

La notion de qualité de vie au travail (QVT) donne lieu à des déclinaisons innombrables et il n’y a pas aujourd’hui de définition stabilisée de celle-ci, au sens de norme juridique ou scientifique, en France comme à l’étranger. Les travaux sur la QVT retiennent généralement une approche multidimensionnelle et considèrent que, pour bien l’appréhender, plusieurs critères doivent être mobilisés. Le choix d’une approche plurielle résulte d’un pragmatisme face à un enjeu complexe et multiforme. Cet article propose de considérer la QVT comme une démarche d’apprentissage collective dans un contexte de changement organisationnel.

Abstract

Switched reluctance machines (SRMs) benefit from simple and cheap design and inherent fault tolerance, but still suffer from torque ripple and noise, vibration and harshness (NVH) issues. This paper aims at experimentally investigating the influence of the DC-bus voltage on NVH aspects of a 15 kW 8/6 SRM. Spectrograms of current, vibration and acoustic noise, as well as the evolution of loudness and sharpness are presented for DC-bus-ramp tests. It turns out that a higher DC-bus voltage combined with a hysteresis control leads to the excitation of higher frequencies and therefore to the generation of a sharper noise. This trend is essentially remarkable in hard-chopping mode. Furthermore, the sound becomes also louder as the DC-link voltage increases, probably due to the discrete character of the controller, responsible for overshootsoutside the current-hysteresis band, becoming more important as the switching frequency increases. 27. P2Y6 receptors regulate CXCL10 expression and secretion in mouse intestinal epithelial cells

Abstract

In this study, we investigated the role of extracellular nucleotides in chemokine (KC, MIP-2, MCP-1, and CXCL10) expression and secretion by murine primary intestinal epithelial cells (IECs) with a focus on P2Y6 receptors. qRT-PCR experiments showed that P2Y6 was the dominant nucleotide receptor expressed in mouse IEC. In addition, the P2Y6 ligand UDP induced expression and secretion of CXCL10. For the other studies, we took advantage of mice deficient in P2Y6 (P2ry6-/-). Similar expression levels of P2Y1, P2Y2, P2X2, P2X4, and A2A were detected in P2ry6-/- and WT IEC. Agonists of TLR3 (poly(I:C)), TLR4 (LPS), P2Y1, and P2Y2 increased the expression and secretion of CXCL10 more prominently in P2ry6-/- IEC than in WT IEC. CXCL10 expression and secretion induced by poly(I:C) in both P2ry6-/- and WT IEC were inhibited by general P2 antagonists (suramin and Reactive-Blue-2), by apyrase, and by specific antagonists of P2Y1, P2Y2, P2Y6 (only in WT), and P2X4. Neither adenosine nor an A2Aantagonist had an effect on CXCL10 expression and secretion. Macrophage chemotaxis was induced by the supernatant of poly(I:C)-treated IEC which was consistent with the level of CXCL10 secreted. Finally, the non-nucleotide agonist FGF2 induced MMP9 mRNA expression also at a higher level in P2ry6-/- IEC than in WT IEC. In conclusion, extracellular nucleotides regulate CXCL10 expression and secretion by IEC. In the absence of P2Y6, these effects are modulated by other P2 receptors also present on IEC. Abstract

Lepidoptera's pheromonal communication, so sensitive and specific, constitutes a model of choice to study the molecular basis of olfactory reception. The strong specificity of this system is thought to be ensured in the antennae by the intervention of a diversity of genes implicated in the olfactory chemoreception. The silkworm, Bombyx mori, is a particularly interesting species for studying olfaction in insects: male's response to the pheromone is well known, this species is amenable to genetic manipulations and genomic sequences are available on the Internet. In my PhD work, I have identified and characterized at the molecular level a diversity of olfactory genes by bioinformatic analysis of the genome and molecular biology tools. The expression patterns of these genes have been established in order to check for a possible chemosensory function. Some of them (BmPBP2, BmPBP3, BmAOX2) could encode proteins specifically involved in pheromonal reception whereas others (BmAOX1, BmSNMP2) could also act in the reception of less specific molecules. By their combinatorial and sequential actions in signal recognition, these proteins from different families would ensure specific recognition across the different steps of peripheral olfactory reception. I have also participated in the development of a new in vivo functional analysis tool in B. mori, based on the expression of OR by transgenesis. This system has been tested with a male-specific pheromone receptor, BmOR1. Females from transgenic lines strongly expressed the receptor in their antennae, but were unable to respond to the pheromonal compound (Bol) during electrophysiological recordings. Future use of a new promotor, more specific to olfactory neurons, pOR2, could allow the development of the first stable in vivo functional analysis tool in a lepidoptera.; La communication phéromonale chez les lépidoptères, si sensible et spécifique, constitue un modèle de choix pour l'étude des bases moléculaires de la réception olfactive. La spécificité de ce système serait assurée dès le niveau périphérique, dans l'antenne, par l'intervention d'une diversité de protéines impliquées dans la chimioréception des odorants. Le ver à soie, Bombyx mori, est une espèce particulièrement appréciée pour l'étude des mécanismes olfactifs : la réponse des mâles à la phéromone est connue, l'espèce est manipulable génétiquement et les séquences génomiques sont accessibles sur Internet. Au cours de mon travail de thèse, j'ai pu identifier et caractériser au niveau moléculaire une diversité de gènes olfactifs appartenant à différentes familles par des techniques bioinformatiques d'analyse du génome et de biologie moléculaire. Leurs patrons d'expression ont notamment été analysés afin de vérifier si leur localisation tissulaire était compatible avec une fonction chimiosensorielle. Certains de ces gènes (BmPBP2, BmPBP3, BmAOX2) pourraient coder pour des protéines spécifiquement impliquées dans la réception phéromonale alors que d'autres (BmAOX1, BmSNMP2) pourraient intervenir aussi dans la réception de molécules moins spécifiques. Par leurs interactions combinées et séquentielles avec les molécules odorantes, ces différentes familles de protéines permettraient d'assurer leur reconnaissance spécifique au cours des différentes étapes périphériques de la réception olfactive. Par ailleurs, j'ai pu participer à la mise au point chez le ver à soie d'un outil novateur d'analyse fonctionnelle in vivo basé sur l'expression par transgenèse de RO dans les antennes du ver à soie. Ce système a été développé avec un récepteur phéromonal mâle-spécifique, BmOR1. Les femelles des lignées transgéniques exprimaient fortement le récepteur dans leurs antennes, mais se sont révélées incapables de répondre au composé phéromonal (Bol) au cours d'enregistrements électrophysiologiques.
