Abstract

The Elephant Path, book two in the series Tales of the Protectors, continues the story of the Protectors, Maxwell, Jack, Eden and Gem, who return to Everwell where they further their training expecting to be assigned to another simple protection assignment. The Protectors arrive at their long-awaited training grounds, Everwell, only to discover they now have access to a garden with one of them possessing special powers there. After strengthening their protection skills even further, the children stand together with another team hoping to prevent the loss of innocent life during an assignment no one could foresee. The Protectors find themselves in the middle of a civil war with more than one group of victims to defend, all the while keeping an eye out for Norris who continually has Maxwell in his crosshairs. Past friends and old enemies reappear in this sequel to Almost Heaven – Tales of the Protectors as the teams combine their strength and talents in a desperate attempt to save the world from a loss that cannot be regained.

Abstract

Almost Heaven elevates children to a position of responsibility without losing their childlike wonder. Tales of the Protectors: Almost Heaven is an enchanting contemporary juvenile fiction that strives to captivate the reader into make-believing he/she could be chosen as a Protector, as well as Maxwell and his friends. When young Maxwell transports to Everwell, the training ground of the Protectors, through a turnstile in a local cemetery, he finds himself with a group of young people chosen from earth to assist and defend the human race. His team soon discovers there are special circumstances surrounding Maxwell which no one in authority wishes to reveal. Almost Heaven empowers young readers to imagine a place broader than the universe and to trust their instincts to solve problems which may pop-up unexpectedly. It is unique in the concept of children assuming great responsibility for others and appeals to readers who wish to relate to their character. The pivotal problem faced by the children towards the end of Almost Heaven forces them to choose whether they will go along with those in charge to attempt a rescue of their friend or will Maxwell possibly sacrifice himself to save his friend.

22. Early metabolomics changes in heart and plasma during chronic doxorubicin treatment in B6C3F1 mice.

Abstract

The present study aimed to identify molecular markers of early stages of cardiotoxicity induced by a potent chemotherapeutic agent, doxorubicin (DOX). Male B6C3F1 mice were dosed with 3 mg kg−1 DOX or saline via tail vein weekly for 2, 3, 4, 6 or 8 weeks (cumulative DOX doses of 6, 9, 12, 18 or 24 mg kg−1, respectively) and euthanized a week after the last dose. Mass spectrometry-based and nuclear magnetic resonance spectrometry-based metabolic profiling were employed to identify initial biomarkers of cardiotoxicity before myocardial injury and cardiac pathology, which were not noted until after the 18 and 24 mg kg−1 cumulative doses, respectively. After a cumulative dose of 6 mg kg−1, 18 amino acids and four biogenic amines (acetylornithine, kynurenine, putrescine and serotonin) were significantly increased in cardiac tissue; 16 amino acids and two biogenic amines (acetylornithine and hydroxyproline) were significantly altered in plasma. In addition, 16 acylcarnitines were significantly increased in plasma and five were significantly decreased in cardiac tissue compared to saline-treated controls. Plasma lactate and succinate, involved in the Krebs cycle, were significantly altered after a cumulative dose of 6 mg kg−1. A few metabolites remained altered at higher cumulative DOX doses, which could partly indicate a transition from injury processes at 2 weeks to repair processes with additional injury happening concurrently before myocardial injury at 8 weeks. These altered metabolic profiles in mouse heart and plasma during the initial stages of injury progression due to DOX treatment may suggest these metabolites as candidate early biomarkers of cardiotoxicity. Abstract

ylamide (AA) is known to cause neurotoxicity, genotoxicity, reproductive, and carcinogenic effects in rodents and neurotoxicity in humans. A metabolomics study of urine samples from rats dosed with acrylamide for 14 days was undertaken to understand the mechanisms of and develop biomarkers for acrylamide-induced toxicity. NMR-based and LC/MS-based metabolomics methods were used to analyze metabolites in urine samples. Three mercapturic acid conjugates of acrylamide were detected using exact mass and principal component analysis (PCA) of urine samples. NMR analysis showed an increase in creatine and a decrease in taurine throughout the dosing period. Results showed that citric acid cycle metabolites were down-regulated later in the dosing period. Further, many amino acids were also up-regulated during the study and may be related to the weight loss observed in this study. Taken together, the data suggest that both LC/MS-based and NMR-based metabolomics analysis can detect changes in endogenous metabolites related to glutathione, TCA cycle, and amino acid metabolism induced by AA administration over a 2 week dosing period. Abstract

The role of protein glutathionylation in acetaminophen (APAP)-induced liver injury was investigated in this study. A single oral gavage dose of 150 or 300 mg/kg APAP in B6C3F1 mice produced increased serum alanine aminotransferase and aspartate aminotransferase levels and liver necrosis in a dose-dependent manner. The ratio of GSH to GSSG was decreased in a dose-dependent manner, suggesting that APAP produced a more oxidizing environment within the liver. Despite the increased oxidation state, the level of global protein glutathionylation was decreased at 1 h and continued to decline through 24 h. Immunohistochemical localization of glutathionylated proteins showed a complex dynamic change in the lobule zonation of glutathionylated proteins. At 1 h after APAP exposure, the level of glutathionylation decreased in the single layer of hepatocytes around the central veins but increased mildly in the remaining centrilobular hepatocytes. This increase correlated with the immunohistochemical localization of APAP covalently bound to protein. Thereafter, the level of glutathionylation decreased dramatically over time in the centrilobular regions with major decreases observed at 6 and 24 h. Despite the overall decreased glutathionylation, a layer of cells lying between the undamaged periportal region and the damaged centrilobular hepatocytes exhibited high levels of glutathionylation at 3 and 6 h in all samples and in some 24-h samples that had milder injury. These temporal and zonal pattern changes in protein glutathionylation after APAP exposure indicate that protein glutathionylation may play a role in protein homeostasis during APAP-induced hepatocellular injury.

37. Clonal Associations among Staphylococcus aureusIsolates from Various Sites of Infection

Abstract

ABSTRACTA molecular epidemiological analysis was undertaken to identify lineages of Staphylococcus aureusthat may be disproportionately associated with infection. Pulsed-field gel electrophoresis analysis of 405 S. aureusclinical isolates collected from various infection types and geographic locations was performed. Five distinct S. aureuslineages (SALs 1, 2, 4, 5, and 6) were identified, which accounted for 19.01, 9.14, 22.72, 10.12, and 4.69% of isolates, respectively. In addition, 85 lineages which occurred with frequencies of <2.5% were identified and were termed “sporadic.” The most prevalent lineage was methicillin-resistant S. aureus(SAL 4). The second most prevalent lineage, SAL 1, was also isolated at a high frequency from the anterior nares of healthy volunteers, suggesting that its prevalence among clinical isolates may be a consequence of high carriage rates in humans. Gene-specific PCR was carried out to detect genes for a number of staphylococcal virulence traits. tstand cnawere found to be significantly associated with prevalent lineages compared to sporadic lineages. When specific infection sites were examined, SAL 4 was significantly associated with respiratory tract infection, while SAL 2 was enriched among blood isolates. SAL 1 and SAL 5 were clonally related to SALs shown by others to be widespread in the clinical isolate population. We conclude from this study that at least five phylogenetic lineages of S. aureusare highly prevalent and widely distributed among clinical isolates. The traits that confer on these lineages a propensity to infect may suggest novel approaches to antistaphylococcal therapy.
