- Abstract
BACKGROUND: Multiplexed Assays of Variant Effects (MAVEs) can test all possible single variants in a gene of interest. The resulting saturation-style data may help resolve variant classification disparities between populations, especially for variants of uncertain significance (VUS). METHODS: We analyzed clinical significance classifications in 213,663 individuals of European-like genetic ancestry versus 206,975 individuals of non-European-like genetic ancestry from All of Us and the Genome Aggregation Database. Then, we incorporated clinically calibrated MAVE data into the Clinical Genome Resource's Variant Curation Expert Panel rules to automate VUS reclassification for BRCA1, TP53, and PTEN . RESULTS: Using two orthogonal statistical approaches, we show a higher prevalence ( p ≤5.95e-06) of VUS in individuals of non-European-like genetic ancestry across all medical specialties assessed in all three databases. Further, in the non-European-like genetic ancestry group, higher rates of Benign or Likely Benign and variants with no clinical designation ( p ≤2.5e-05) were found across many medical specialties, whereas Pathogenic or Likely Pathogenic assignments were higher in individuals of European-like genetic ancestry ( p ≤2.5e-05). Using MAVE data, we reclassified VUS in individuals of non-European-like genetic ancestry at a significantly higher rate in comparison to reclassified VUS from European-like genetic ancestry ( p =9.1e-03) effectively compensating for the VUS disparity. Further, essential code analysis showed equitable impact of MAVE evidence codes but inequitable impact of allele frequency ( p =7.47e-06) and computational predictor ( p =6.92e-05) evidence codes for individuals of non-European-like genetic ancestry. CONCLUSIONS: Generation of saturation-style MAVE data should be a priority to reduce VUS disparities and produce equitable training data for future computational predictors.

Aim of this paper is to report a case of spontaneous externalization of Ventriculoperitoneal shunt tube through anus in an eighteen months female child (k/c/o Dandy-Walker malformation) treated with placement of ventriculoperitoneal shunt for hydrocephalus. The exact cause of such externalization is not known. Unusual migration of distal catheter tip of VP shunt is a rare and bowel perforation is reported to occur between 0.1% and 0.7%. In view of the potential risk for meningitis prompt and aggressive management is essential to avoid morbidity and mortality.

PubMed: 31209238, The effectiveness of most cancer targeted therapies is short-lived. Tumors often develop resistance that might be overcome with drug combinations. However, the number of possible combinations is vast, necessitating data-driven approaches to find optimal patient-specific treatments. Here we report AstraZeneca’s large drug combination dataset, consisting of 11,576 experiments from 910 combinations across 85 molecularly characterized cancer cell lines, and results of a DREAM Challenge to evaluate computational strategies for predicting synergistic drug pairs and biomarkers. 160 teams participated to provide a comprehensive methodological development and benchmarking. Winning methods incorporate prior knowledge of drug-target interactions. Synergy is predicted with an accuracy matching biological replicates for >60% of combinations. However, 20% of drug combinations are poorly predicted by all methods. - Abstract
The effectiveness of most cancer targeted therapies is short-lived. Tumors often develop resistance that might be overcome with drug combinations. However, the number of possible combinations is vast, necessitating data-driven approaches to find optimal patient-specific treatments. Here we report AstraZeneca’s large drug combination dataset, consisting of 11,576 experiments from 910 combinations across 85 molecularly characterized cancer cell lines, and results of a DREAM Challenge to evaluate computational strategies for predicting synergistic drug pairs and biomarkers. 160 teams participated to provide a comprehensive methodological development and benchmarking. Winning methods incorporate prior knowledge of drug-target interactions. Synergy is predicted with an accuracy matching biological replicates for >60% of combinations. However, 20% of drug combinations are poorly predicted by all methods. Genomic rationale for synergy predictions are identified, including ADAM17 inhibitor antagonism when combined with PIK3CB/D inhibition contrasting to synergy when combined with other PI3K-pathway inhibitors in PIK3CA mutant cells.

The influence of Calcination temperature on the physical properties of Ni0.6Zn0.4Fe2O4 ferrite nanoparticles were investigated. These ferrite nanoparticles have been synthesized by sol-gel auto combustion method using citric acid as fuel agent at different calcination temperatures (4000C, 5000C and 6000C). The Morphological investigation, average crystallite size and microstructure of the material were examined by using X-ray diffraction (XRD) and confirmed by field emission scanning electron microscope (FESEM) and FTIR spectra. The Effects of calcination temperature on the dielectric and magnetic properties were calculated by using LCR meter and vibrating sample magnetometer (VSM). The XRD result shows a single-phase cubic spinel structure with average crystallite size increases from 27 to 29.5 nm, with an increase of temperature. The highest saturation magnetization was found at a calcination temperature 6000C with value 80.39 emu/g, and the value coercive field (Hc) was inverse with the crystallite size.

Aim of this article is to report a case of spontaneous externalization of Ventriculoperitoneal shunt tube through anus in an eighteen months old female child (k/c/o Dandy-Walker malformation) treated with placement of ventriculoperitoneal shunt for hydrocephalus. The exact cause of such externalization is not known. Unusual migration of distal catheter tip of VP shunt is rare and bowel perforation is reported to occur between 0.1% and 0.7%. In view of the potential risk for meningitis prompt and aggressive management is essential to avoid morbidity and mortality.

Evaluation of preliminary results of the emission of slow (about 1 eV) positrons from solid moderators. The results presented are for copper, gold, and aluminum and were obtained using the backscatter mode of operation. Of the three metals investigated, copper is found to emit positrons with the lowest energy, while aluminum emits those with the highest energy. Several small peaks are noted on the high-energy side of the spectra, especially in the case of gold and aluminum.

- Abstract
Urinary tract infections are the most common infection in renal transplant patients and Escherichia coli(E. coli) is the most common clinical isolate. Although acute allograft injury (AAI) secondary to urinary tract infection (UTI) has been reported, the incidence of AAI associated with UTI, the virulence factors express by uropathic E. coliand whether virulence factors are associated with renal allograft outcome have not been described. We collected E. colifrom our renal transplant patients with UTI, determined O:H serotypes, P and Dr fimbriae expression and the clinical presentation and allograft function during the UTI and post-UTI period. Pyelonephritis occurred in 40% of our patients, 82% of which had AAI (>20% increase in SCr). Sixty-two percent of E. coliisolates that expressed P fimbriae were associated with AAI, whereas only 29% that did not express P fimbriae had AAI (p = 0.03). The pattern of P fimbriae and O serotypes differed from reported isolates, as the P fimbriae PapG class II and the O25 serotype were the most common. Dr adhesin was expressed on 7 isolates, including 2 of 3 with urosepsis. We propose a unique pattern of uropathogenic serotypes and adherence factors contribute to acute allograft injury in renal transplant patients with UTI.
