Abstract Melatonin (Mt) functions as a growth regulator and multifunctional signaling molecule in plants, thereby playing a crucial role in promoting growth and orchestrating protective responses to various abiotic stresses. However, the mechanism whereby exogenous Mt protects Lonicera japonica Thunb. (L. japonica) against salt stress has not been fully elucidated. Therefore, this study aimed to elucidate how exogenous Mt alleviates sodium chloride (NaCl) stress in L. japonica seedlings. Salt-sensitive L. japonica seedlings were treated with an aqueous solution containing 150 mM of NaCl and aqueous solutions containing various concentrations of Mt. The results revealed that treatment of NaCl-stressed L. japonica seedlings with a 60 µM aqueous solution of Mt significantly enhanced vegetative plant growth by scavenging reactive oxygen species and thus reducing oxidative stress. The latter was evidenced by decreases in electrical conductivity and malondialdehyde (MDA) concentrations. Moreover, Mt treatment led to increases in the NaCl-stressed L. japonica seedlings’ total chlorophyll content, soluble sugar content, and flavonoid content, demonstrating that Mt treatment improved the seedlings’ tolerance of NaCl stress. This was also indicated by the NaCl-stressed L. japonica seedlings exhibiting marked increases in the activities of antioxidant enzymes (superoxide dismutase, peroxidase, catalase, and ascorbate peroxidase) and in photosynthetic functions. Furthermore, Mt treatment of NaCl-stressed L. japonica seedlings increased their expression of phenylalanine ammonia-lyase 1 (PAL1), phenylalanine ammonia-lyase 2 (PAL2), calcium-dependent protein kinase (CPK), cinnamyl alcohol dehydrogenase (CAD), flavanol synthase (FLS), and chalcone synthase (CHS). In conclusion, our results demonstrate that treatment of L. japonica seedlings with a 60 µM aqueous solution of Mt significantly ameliorated the detrimental effects of NaCl stress in the seedlings. Therefore, such treatment has substantial potential for use in safeguarding medicinal plant crops against severe salinity.

Abstract Bone defects pose significant challenges in healthcare, with over 2 million bone repair surgeries performed globally each year. As a burgeoning force in the field of bone tissue engineering, 3D printing offers novel solutions to traditional bone transplantation procedures. However, current 3D-printed bone scaffolds still face three critical challenges in material selection, printing methods, cellular self-organization and co-culture, significantly impeding their clinical application. In this comprehensive review, we delve into the performance criteria that ideal bone scaffolds should possess, with a particular focus on the three core challenges faced by 3D printing technology during clinical translation. We summarize the latest advancements in non-traditional materials and advanced printing techniques, emphasizing the importance of integrating organ-like technologies with bioprinting. This combined approach enables more precise simulation of natural tissue structure and function. Our aim in writing this review is to propose effective strategies to address these challenges and promote the clinical translation of 3D-printed scaffolds for bone defect treatment. Abstract C–H bond activation under mild conditions remains a great challenge in the chemical industry, while catalytic cyclohexane oxidation is inefficient and often requires organic solvents and strong oxidants. This study constructed a C3N4/WO3 Z-type heterojunction catalyst to efficiently convert cyclohexane into cyclohexanone and cyclohexanol (KA oil) through aqueous phase oxidation by O2 under visible light irradiation. With strong redox performance and high photogenerated carrier separation ability, the proposed composite catalyst can produce the key active species for cyclohexane oxidation in the H2O–O2 system. The reaction mechanism was clarified through experiment and DFT theory calculation. Cyclohexane was converted into cyclohexyl radical under the action of ·OH, and ·O2 − converted most products into cyclohexanone. The catalyst can be recycled under optimized process conditions while reaching a KA oil yield of 139.73 μmol g−1 h−1 and a selectivity of 93.1%.

Abstract In generic closed quantum systems, the complexity of operators increases under time evolution governed by the Heisenberg equation, reflecting the scrambling of local quantum information. However, when systems interact with an external environment, the system-environment coupling allows operators to escape from the system, inducing a dynamical transition between the scrambling phase and the dissipative phase. This transition is known as the environment-induced information scrambling transition, originally proposed in Majorana fermion systems. In this work, we advance this discovery by investigating the transition in charge-conserved systems with space-time randomness. We construct solvable Brownian Sachdev-Ye-Kitaev models of complex fermions coupled to an environment, enabling the analytical computation of operator growth. We determine the critical dissipation strength, which is proportional to $$n(1-n)$$ n ( 1 - n ) with n being the density of the complex fermions, arising from the suppression in the quantum Lyapunova exponent due to the Pauli blockade in the scattering process. We further analyze the density dependence of maximally scrambled operators at late time. Our results shed light on the intriguing interplay between information scrambling, dissipation, and conservation laws.

Abstract In recent years, immunotherapy, particularly PD-1 antibodies, have significantly enhanced the outcome of gastric cancer patients. Despite these advances, some patients do not respond well to treatment, highlighting the need to understand resistance mechanisms and develop predictive markers of treatment effectiveness. This study retrospectively analyzed data from 106 patients with stage IV gastric cancer who were treated with first-line immunotherapy in combination with chemotherapy. By comparing plasma cytokine levels between patients resistant and sensitive to PD-1 antibody therapy, the researchers identified elevated IL-4 expression in the resistant patients. Mechanical investigations revealed that IL-4 induces metabolic changes in macrophages that activate the PI3K/AKT/mTOR pathway. This alteration promotes ATP production, enhances glycolysis, increases lactic acid production, and upregulates FcγRIIB expression in macrophages. Ultimately, these changes lead to CD8+ T cell dysfunction and resistance to PD-1 antibody therapy in gastric cancer. These findings highlight the role of IL-4-induced macrophage polarization and metabolic reprogramming in immune resistance and verify IL-4 as potential targets for improving treatment outcomes in gastric cancer patients.

Abstract The rapid development of quantum science and technology is leading us into an era where quantum many-body systems can be comprehended through quantum simulations. Holographic duality, which states gravity and spacetime can emerge from strongly interacting systems, then offers a natural avenue for the experimental study of gravity physics without delving into experimentally infeasible high energies. A prominent example is the simulation of traversable wormholes through the wormhole teleportation protocol, attracting both theoretical and experimental attention. In this work, we develop the theoretical framework for computing the fidelity of wormhole teleportation in N-qubit systems with all-to-all interactions, quantified by mutual information and entanglement negativity. The main technique is the scramblon effective theory, which captures universal out-of-time-order correlations in generic chaotic systems. We clarify that strong couplings between the two systems are essential for simulating the probe limit of semi-classical traversable wormholes using strongly interacting systems with near-maximal chaos. However, the teleportation signal diminishes rapidly when reducing the system size N, requiring a large number of qubits to observe a sharp signature of emergent geometry by simulating the Sachdev-Ye-Kitaev model. This includes both the causal time-order of signals and the asymmetry of the teleportation signal for coupling with different signs. As a comparison, the teleportation signal increases when reducing N in weakly interacting systems. We also analyze the fidelity of the generalized encoding scheme in fermionic string operators.

In order to study the stability and influencing factors of the end face roof of the full-mechanized mining face, a mechanical model of tip-to-face roof stability was developed in this study with a combination of the energy method and the “roof-support-coal” system. Meanwhile, the load monitoring system on the canopy of the support and the digital image measuring technique were utilized in a physical simulation model to analyze the influence factors of tip-to-face roof fall, the roof failure characteristics and the evolution characteristics of pressure distribution on the canopy of the support. The results showed that the stability coefficient of the immediate roof within 1 m behind the coal wall was less than 0 based on the mechanical model of tip-to-face roof stability, indicating a high risk of roof collapse in this area. In addition, the vertical and horizontal displacement of the immediate roof increased with the increase of the distance behind the coal wall. Enhancing the working resistance of the support can also reduce the roof subsidence, and the higher roof cohesion and internal friction angle can improve the tip-to-face roof stability. On the other hand, the tip-to-face roof caving, roof broken and roof crushing were observed in sequence during the process of the physical simulation experiments. The middle section of the canopy showed the largest measured pressure, followed by the front and rear sections of the canopy in the descending order. When the roof was intact, the support showed adequate setting load and working resistance, and the interaction between the support and the strata was in good condition. As the roof was broken and led to an unfavorable support position, the support showed a loss of working resistance or an unbalanced loading condition, which may easily result in the crack development in the roof, tip-to-face roof cavity, an iron-bound support and an awful support-strata coupling. According to the digital image measuring technique, the maximum shear strain was found near the tip-to-face area during the tip-to-face roof caving stage. Lastly, it can be concluded that the coupling between the support and strata is closely correlated to the tip-to-face roof stability. Thus, maintaining a good support-strata interaction is beneficial to the stability of the tip-to-face roof.

Computing-aware networking (CAN) is introduced to unite the computing resources distributed in different platforms. This paper proposes a joint routing and resource allocation model to minimize the total operational cost in CAN. We formulate the problem as an integer linear programming problem. We introduce a polynomial-time algorithm for larger-size problems. The numerical results reveal that the introduced algorithm reduces the computation time 9.18 times, with increasing the objective no more than 4% compared to the optimal solution; the proposed model can reduce 70% of the total cost compared to a baseline adopting a two-stage strategy in our examined cases.

Abstract Keratinocyte proliferation and differentiation in epidermis are well-controlled and essential for reacting to stimuli such as ultraviolet light. Imbalance between proliferation and differentiation is a characteristic feature of major human skin diseases such as psoriasis and squamous cell carcinoma. However, the effect of keratinocyte metabolism on proliferation and differentiation remains largely elusive. We show here that the gluconeogenic enzyme fructose-1,6-bisphosphatase 1 (FBP1) promotes differentiation while inhibits proliferation of keratinocyte and suppresses psoriasis development. FBP1 is identified among the most upregulated genes induced by UVB using transcriptome sequencing and is elevated especially in upper epidermis. Fbp1 heterozygous mice exhibit aberrant epidermis phenotypes with local hyperplasia and dedifferentiation. Loss of FBP1 promotes proliferation and inhibits differentiation of keratinocytes in vitro. Mechanistically, FBP1 loss facilitates glycolysis-mediated acetyl-CoA production, which increases histone H3 acetylation at lysine 9, resulting in enhanced transcription of proliferation genes. We further find that the expression of FBP1 is dramatically reduced in human psoriatic lesions and in skin of mouse imiquimod psoriasis model. Fbp1 deficiency in mice facilitates psoriasis-like skin lesions development through glycolysis and acetyl-CoA production. Collectively, our findings reveal a previously unrecognized role of FBP1 in epidermal homeostasis and provide evidence for FBP1 as a metabolic psoriasis suppressor.

Hydraulic fracturing is an important technical means to relieve the pressure of coal seam roof. Better understanding of fracture propagation mechanism is of great significance to the safe mining of coal seam. In order to further explore the law of hydraulic fracture propagation, aiming at rock-like specimens commonly used in the laboratory, MatDEM, a particle discrete element numerical simulation software, was used to establish a two-dimensional discrete element numerical model of hydraulic fracturing, and various hydraulic fracturing tests with different injection pressure increments were carried out. The effect of injection pressure increment on the propagation of hydraulic fractures was studied, and the mechanism of model initiation was revealed. The law of fracture generation and propagation was analyzed from mesoscale, and the propagation characteristics of hydraulic fractures were discussed. The results show that ① the effect of injection pressure increment on the model initiation pressure and initiation time presents an opposite trend. With the increase of injection pressure, the increase trend of initiation pressure is slow and gradually approaches to 5.6 MPa. The initiation time decreases with the increase of injection pressure, and the decreasing trend slows down gradually. ② The cumulative number of fractures increases exponentially with time. The hydraulic fracturing process can be divided into four stages (Ⅰ−Ⅳ): no fracture stage, slow fracture growth stage, steady fracture growth stage and rapid fracture growth stage, which correspond to the pre-crack initiation, pre-crack formation, primary fracture propagation and secondary fracture propagation processes respectively. As the injection pressure increment increases, the durations of stage Ⅰ, Ⅱ and Ⅲ decrease, while the duration of stage Ⅳ increases in a fluctuating manner. The number of cracks in each stage is the highest in stage Ⅳ, followed by stage Ⅲ and stage Ⅱ. ③ As the injection pressure increment increases, the number of secondary fractures increases from 8 to 16, and the growth rate of fractures gradually slows down before the stage Ⅲ, and increases rapidly after the stage Ⅳ. When the injection pressure increment increases from 0.03 MPa to 0.70 MPa, the final fracture length increases by 1.79 times. ④ The internal energy of the model increases with the increase of the injection pressure increment, and the energy input speed gradually becomes faster. After the model initiation, high-pressure water forms stress concentration at the crack tip, which promotes the crack to continue to extend. At higher injection pressure increment, the fracture propagation speed becomes faster, and the particle displacement decreases gradually from the pressure hole to the outside of the model. The increment of injection pressure makes the secondary fracture forming position close to the pressure hole, which inhibits the formation and expansion of the primary fracture and promotes the formation and expansion of the secondary fracture. All fracture types are tensile fractures.

The Lower Cretaceous Shahezi Formation glutenite reservoirs in the Xujiaweizi Fault Depression are important tight gas reservoirs in the deep strata of the Songliao Basin. Their complex porosity and permeability relationships pose challenges to determining reservoir boundaries and evaluating reservoir grades. For the Shahezi Formation glutenite reservoirs, the water film thickness method, charge dynamics method, gas testing productivity method, and buoyancy balance method were used to determine theoretical lower limits, gas accumulation lower limits, effective flow lower limits, and reservoir-forming upper limits. The petrophysical values of the reservoir boundaries were determined based on the porosity and permeability relationships under different diagenetic controlling factors. On this basis, reservoir types were classified according to differences in microscopic pore structures, and a grading evaluation standard for tight reservoirs was established. This standard was then applied using logging data to provide a basis for selecting sweet spots in the tight gas exploration areas. The Shahezi Formation glutenite was divided into conventional reservoirs, Class Ⅰ-Ⅳ tight glutenite reservoirs, and non-reservoirs. The reservoir boundaries and classification evaluation results were well matched. Conventional reservoirs had porosity greater than 9% and permeability greater than 0.05×10-3 μm2. Class Ⅰ tight reservoirs had porosity of 8%-9% and permeability of (0.01-0.05)×10-3 μm2. Class Ⅱ tight reservoirs had porosity of 5%-8% and permeability of (0.001-0.01)×10-3 μm2. Class Ⅲ tight reservoirs had porosity of 3.5%-5% and permeability of (0.2-1)×10-3 μm2. Class Ⅳ tight reservoirs had porosity of 2%-3.5% and permeability of (0.05-0.2)×10-6 μm2. Non-reservoirs had porosity less than 2% and permeability less than 0.05×10-6 μm2. The gas production of tight glutenite was controlled by the reservoir type. Class Ⅰ and Class Ⅱ tight reservoirs were favorable high-yield layers. The thickness of favorable reservoirs in the Anda-Songzhan area of the northern Xujiaweizi Fault Depression was large, making it a sweet spot for tight gas exploration and development.

Abstract Accurate and robust time delay estimation is crucial for synthetic aperture sonar (SAS) imaging. A two‐step time delay estimation method based on displaced phase centre antenna (DPCA) micronavigation has been widely applied in motion estimation and compensation of SASs. However, the existing methods for time delay estimation are not sufficiently robust, which reduces the performance of SAS motion estimation. Deep learning is currently one of the cutting‐edge techniques and has brought about a remarkable progress in the field of underwater acoustic signal processing. In this study, a deep learning‐based time delay estimation method is introduced to SAS motion estimation and compensation. The subband processing is first applied to obtain ambiguous time delays between adjacent pings from phases of SAS echoes. Then, a lightweight neural network is utilised to construct phase unwrapping. The model of the employed neural network is trained with simulation data and applied to real SAS data. The results of time delay estimation and motion compensation demonstrate that the proposed neural network‐based method has much better performance than the two‐step and joint‐subband methods.

Abstract Background The health and size of the testes are crucial for boar fertility. Testicular development is tightly regulated by epigenetics. N6-methyladenosine (m6A) modification is a prevalent internal modification on mRNA and plays an important role in development. The mRNA m6A methylation in boar testicular development still needs to be investigated. Results Using the MeRIP-seq technique, we identify and profile m6A modification in boar testes between piglets and adults. The results showed 7783 distinct m6A peaks in piglets and 6590 distinct m6A peaks in adults, with 2,471 peaks shared between the two groups. Enrichment of GO and KEGG analysis reveal dynamic m6A methylation in various biological processes and signalling pathways. Meanwhile, we conjointly analyzed differentially methylated and expressed genes in boar testes before and after sexual maturity, and reproductive related genes (TLE4, TSSK3, TSSK6, C11ORF94, PATZ1, PHLPP1 and PAQR7) were identified. Functional enrichment analysis showed that differential genes are associated with important biological functions, including regulation of growth and development, regulation of metabolic processes and protein catabolic processes. Conclusion The results demonstrate that m6A methylation, differential expression and the related signalling pathways are crucial for boar testicular development. These results suggest a role for m6A modification in boar testicular development and provided a resource for future studies on m6A function in boar testicular development.

Abstract Background Coronavirus disease 2019 (COVID-19) has been proved as a significant risk factor for deep vein thrombosis (DVT) after several waves of pandemic. This study aims to further investigate impact of COVID-19 on prognosis of DVT following anticoagulation treatment. Methods A total of 197 patients with initially detected DVT and meanwhile accomplishing at least 3 months anticoagulation treatment were identified from our hospital between January 2021 and December 2022. DVT characteristics, clinical data, and exposure to COVID-19 were recorded for multivariable logistic regression analysis to identify DVT aggravation related risk factors. Propensity score matching (PSM) was used to balance baseline covariates. Kaplan–Meier curves and Log-Rank test were performed to exhibit distribution of DVT aggravation among different subgroups. Results In 2022, patients exhibited higher incidence rates of DVT aggravation compared to those in 2021 (HR:2.311, P = 0.0018). The exposure to COVID-19, increased red blood cell count, increased D-dimer level and reduced prothrombin time were found to be associated with DVT aggravation (P

Antimicrobial resistance (AMR) has become a global health crisis in need of novel solutions. To this end, antibiotic combination therapies, which combine multiple antibiotics for treatment, have attracted significant attention as a potential approach for combating AMR. To facilitate advances in antibiotic combination therapies, most notably in investigating antibiotic interactions and identifying synergistic antibiotic combinations however, there remains a need for automated high-throughput platforms that can create and examine antibiotic combinations on-demand, at scale, and with minimal reagent consumption. To address these challenges, we have developed a Robotic-Printed Combinatorial Droplet (RoboDrop) platform by integrating a programmable droplet microfluidic device that generates antibiotic combinations in nanoliter droplets in automation, a robotic arm that arranges the droplets in an array, and a camera that images the array of thousands of droplets in parallel. We further implement a resazurin-based bacterial viability assay to accelerate our antibiotic combination testing. As a demonstration, we use RoboDrop to corroborate two pairs of antibiotics with known interactions and subsequently identify a new synergistic combination of cefsulodin, penicillin, and oxacillin against a model E. coli strain. We therefore envision RoboDrop becoming a useful tool to efficiently identify new synergistic antibiotic combinations toward combating AMR.

Wind velocity is usually assumed to obey a stationary stochastic process in wind engineering, and this may cause significant bias in describing extremely severe strong wind such as typhoons and thunderstorms. To take into account the non-stationary characteristics of extreme wind, a novel evolutionary power spectral density (EPSD) model is proposed, and the spectral representation method (SRM) is introduced to simulate the whole process of strong winds. Firstly, the wavelet transform (WT) method is adopted to capture the three-dimensional time-varying properties of the low-frequency mean winds, and the associated turbulence features, including turbulent intensity, gust factor, probability density function, and power spectrum, are analyzed in depth. Secondly, the measured horizontal EPSD of strong winds are estimated. Thirdly, the performance of the proposed EPSD model is validated. Finally, the whole process of non-stationary strong winds are simulated and discussed. The results show that the proposed EPSD models are in good agreement with the measured EPSD, and the time-frequency features of the power spectrum of the simulated winds are well reproduced, which provides a powerful tool for large eddy simulation and wind engineering studies under non-stationary extreme wind climate.

Bone defect disease seriously endangers human health and affects beauty and function. In the past five years, the three dimension (3D) printed radially graded triply periodic minimal surface (TPMS) porous scaffold has become a new solution for repairing bone defects. This review discusses 3D printing technologies and applications for TPMS scaffolds. To this end, the microstructural effects of 3D printed TPMS scaffolds on bone regeneration were reviewed and the structural characteristics of TPMS, which can promote bone regeneration, were introduced. Finally, the challenges and prospects of using TPMS scaffolds to treat bone defects were presented. This review is expected to stimulate the interest of bone tissue engineers in radially graded TPMS scaffolds and provide a reliable solution for the clinical treatment of personalised bone defects.

Abstract To date, several molecules have been found to facilitate iron influx, while the types of iron influx channels remain to be elucidated. Here, Piezo1 channel was identified as a key iron transporter in response to mechanical stress. Piezo1-mediated iron overload disturbed iron metabolism and exaggerated ferroptosis in nucleus pulposus cells (NPCs). Importantly, Piezo1-induced iron influx was independent of the transferrin receptor (TFRC), a well-recognized iron gatekeeper. Furthermore, pharmacological inactivation of Piezo1 profoundly reduced iron accumulation, alleviated mitochondrial ROS, and suppressed ferroptotic alterations in stimulation of mechanical stress. Moreover, conditional knockout of Piezo1 (Col2a1-CreERT Piezo1 flox/flox ) attenuated the mechanical injury-induced intervertebral disc degeneration (IVDD). Notably, the protective effect of Piezo1 deficiency in IVDD was dampened in Piezo1/Gpx4 conditional double knockout (cDKO) mice (Col2a1-CreERT Piezo1 flox/flox /Gpx4 flox/flox ). These findings suggest that Piezo1 is a potential determinant of iron influx, indicating that the Piezo1-iron-ferroptosis axis might shed light on the treatment of mechanical stress-induced diseases.

Abstract Satellite navigation systems enable long‐distance and high‐precision time transfers. The authors proposed an undifferenced precise time transfer method with information constraints in this study. A more accurate receiver clock offset can be obtained by constraining the station coordinate parameters and receiver clock offset parameters to achieve a precise time transfer. In this example, the research object is the time transfer link established by the Global Positioning System observation station using a caesium clock, rubidium clock, and hydrogen clock. The clock difference sequence of the time‐transfer link was calculated by combining the observation data and precise products. Following analysis of the examples, the following conclusions were reached. The undifferenced precise time transfer method based on information constraints can effectively be used to smooth the clock difference sequence of the receiver and the clock difference sequence of the time transfer link. To some extent, this method can also be used to reflect the performance of atomic clocks at Global Positioning System stations and is better suited for the precise time transfer of the equal‐clock long‐baseline time transfer link. Simultaneously, the frequency stability of this method is improved significantly in the short term compared to the traditional scheme without any constraints. The improvement of frequency stability is helpful to improve the accuracy of time transfer.

Nitric oxide (NO) enhanced photodynamic therapy (PDT) is a promising approach to overcome drug tolerance and resistance to biofilm but is limited by its short excitation wavelengths and low yield of reactive oxygen species (ROS). Herein, we develop a compelling degradable polymer-based near-infrared II (NIR-II, 1000–1700 nm) photosensitizer (PNIR-II), which can maintain 50 % PDT efficacy even under a 2.6 cm tissue barrier. Remarkably, PNIR-II is synthesized by alternately connecting the electron donor thiophene to the electron acceptors diketopyrrolopyrrole (DPP) and boron dipyrromethene (BODIPY), where the intramolecular charge transfer properties can be tuned to increase the intersystem crossover rate and decrease the internal conversion rate, thereby stabilizing the NIR-II photodynamic rather than photothermal effect. For exerting a combination therapy to eradicate multidrug-resistant biofilms, PNIR-II is further assembled into nanoparticles (NPs) with a synthetic glutathione-triggered NO donor polymer. Under 1064 nm laser radiation, NPs precisely release ROS and NO that triggered by over-expressed GSH in the biofilm microenvironment, thereby forming more bactericidal reactive nitrogen species (RNS) in vitro and in vivo in the mice model that orderly destroy biofilm of multidrug-resistant Staphylococcus aureus cultures from clinical patients. It thus provides a new outlook for destroy the biofilm of deep tissues.

Abstract Acceleration of tooth movement during orthodontic treatment is challenging, with osteoclast-mediated bone resorption on the compressive side being the rate-limiting step. Recent studies have demonstrated that mechanoreceptors on the surface of monocytes/macrophages, especially adhesion G protein-coupled receptors (aGPCRs), play important roles in force sensing. However, its role in the regulation of osteoclast differentiation remains unclear. Herein, through single-cell analysis, we revealed that CD97, a novel mechanosensitive aGPCR, was expressed in macrophages. Compression upregulated CD97 expression and inhibited osteoclast differentiation; while knockdown of CD97 partially rescued osteoclast differentiation. It suggests that CD97 may be an important mechanosensitive receptor during osteoclast differentiation. RNA sequencing analysis showed that the Rap1a/ERK signalling pathway mediates the effects of CD97 on osteoclast differentiation under compression. Consistently, we clarified that administration of the Rap1a inhibitor GGTI298 increased osteoclast activity, thereby accelerating tooth movement. In conclusion, our results indicate that CD97 suppresses osteoclast differentiation through the Rap1a/ERK signalling pathway under orthodontic compressive force.

Abstract Background Branch retinal vein occlusion (BRVO) is a common retinal vascular disease leading to severe vision loss and blindness. This study aimed to investigate and reveal the pathophysiological mechanisms underlying macular edema (ME) recurrence in patients with BRVO through a proteomic approach. Methods We detected proteins in the aqueous humor of 14 untreated, four refractory, and four post-operative patients with BRVO-ME and 12 age-matched cataract controls using four-dimensional label-free proteomic and bioinformatics analyses. Abstract The present study was conducted by cultivating tomato (Solanum lycopersicum ‘Provence’) using varied inorganic mulching to investigate soil hydrothermal environment and tomato characters under unheated greenhouse cultivation in the cold zone of China. A total of 6 different treatments were adopted: no mulching (control), white film mulching (white film), black film mulching (black film), the white film with hole mulching (white hole), the black film with hole mulching (black hole), and snake skin bag mulching (snake skin). Inorganic mulching treatment significantly improved soil temperature and moisture, water use efficiency, tomato yield, and reduced soil water consumption. There was no significant difference observed in the variation of daily mean soil temperature between different mulching treatments, and the variation was in the range of 1.95–2.20 °C, which was significantly lower compared with the control (3.42 °C). The daily mean soil moisture varied significantly after different mulching treatments, with the highest level achieved by snake skin (23.37%), followed by black hole (22.55%), white hole (22.08%), white film (21.48%), black film (20.12%), and control (18.78%) in descending order. According to the research results, plastic-hole mulching, which include white hole and black hole treatments, performed better in maintaining soil temperature and moisture.

Abstract Background Whether Transanal drainage tubes (TDTs) placement reduces the occurrence of anastomotic leakage (AL) after rectal cancer (RC) surgery remains controversial. Most existing meta-analyses rely on retrospective studies, while the prospective studies present an inadequate level of evidence. Methods A systematic review and meta-analysis of prospective studies on TDTs placement in RC patients after surgery was conducted. The main analysis index was the incidence of AL, Grade B AL, and Grade C AL, while secondary analysis index was the incidence of anastomotic bleeding, incision infection, and anastomotic stenosis. A comprehensive literature search was performed utilizing the databases Cochrane Library, Embase, PubMed, and Web of Science. We recorded Risk ratios (RRs) and 95% confidence intervals (CI) for each included study, and a fixed-effect model or random-effect model was used to investigate the correlation between TDTs placement and four outcomes after RC surgery. Results Seven studies (1774 participants, TDT 890 vs non-TDT 884) were considered eligible for quantitative synthesis and meta-analysis. The meta-analysis revealed that the incidence of AL was 9.3% (83/890) in the TDT group and 10.2% (90/884) in the non-TDT group. These disparities were found to lack statistical significance (P = 0.58). A comprehensive meta-analysis, comprising four studies involving a cumulative sample size of 1259 participants, revealed no discernible disparity in the occurrence of Grade B AL or Grade C AL between the TDT group and the non-TDT group (Grade B AL: TDT 34/631 vs non-TDT 26/628, P = 0.30; Grade C AL: TDT 11/631 vs non-TDT 27/628, P = 0.30). Similarly, the incidences of anastomotic bleeding (4 studies, 876 participants), incision infection (3studies, 713 participants), and anastomotic stenosis (2studies, 561 participants) were 5.5% (24/440), 8.1% (29/360), and 2.9% (8/280), respectively, in the TDT group, and 3.0% (13/436), 6.5% (23/353), and 3.9% (11/281), respectively, in the non-TDT group. These differences were also determined to lack statistical significance (P = 0.08, P = 0.43, P = 0.48, respectively). Conclusion The placement of TDTs does not significantly affect the occurrence of AL, Grade B AL, and Grade C AL following surgery for rectal cancer. Additionally, TDTs placement does not be associated with increased complications such as anastomotic bleeding, incision infection, or anastomotic stenosis. The Absorbing Aerosol Index (AAI) determines the presence and approximate amount of absorbing aerosol particles. The Absorbing Aerosol Sensor (AAS) equipped on the Gaofen-5B satellite was successfully launched and deployed in orbit on September 7, 2021. Following a period of initial on-orbit testing, reliable AAI observational results were collected. The nadir of the AAS had a design spatial resolution of 4 km × 4 km and a current spatial resolution of 2 km × 4 km, which presented significant benefits in the spatial resolution and clear distribution characteristics of pollutants. The results of AAS were validated by comparing with the observed data of Global Ozone Monitoring Experiment-2 (GOME-2) in the Sahara Desert and surrounding areas. In 2022 and 2023, we utilized AAS observations to study the typical pollution processes. These included the spring pollution distribution around the Bohai Sea, the observation and tracking of long-term dust pollution in China in March and April 2023, the distribution of spring pollution in the Indochina Peninsula in April 2023, and the distribution of wildfires in Canada on June 22, 2023. The results were validated and analyzed using data from Tropospheric Monitoring Instrument (TROPOMI), GOME-2, AErosol RObotic NETwork (AERONET) and Moderate-resolution Imaging Spectroradiometer (MODIS). The results of the observation and evaluation revealed that the AAS AAI can provide reliable information for precise observation of air pollutants at a high resolution.

This study explores an obstacle avoidance control approach that takes into account both actuator dead zones and unpredictable disturbances in environments with moving obstacles. Initially, we enhance a Barrier Lyapunov Function (BLF) by introducing the concept of an equivalent safe distance. This innovation broadens the safety perimeter around obstacles by considering their velocity and movement duration, thereby simplifying the intricacies associated with moving obstacle models in avoidance strategies. Furthermore, we refine the auxiliary function within the obstacle avoidance control laws by incorporating exponential and saturation functions. This refinement ensures that as the directional error increases, the velocity of the Automated Guided Vehicle (AGV) decreases. Similarly, it guarantees a reduction in speed as the AGV nears obstacles, resulting in a smoother auxiliary function that prevents excessive speeding when approaching obstacles. Subsequently, we propose an obstacle avoidance control method for AGVs with dead zones, leveraging the enhanced BLF alongside the introduction of a fixed-time disturbance observer. This integration facilitates the estimation of disturbances within a predetermined timeframe. Unlike existing fixed-time disturbance observers, our method incorporates command filtering to mitigate the risk of computational overload. Ultimately, the efficacy of our proposed approach is validated through rigorous experimental simulations conducted on the Matlab platform.

Abstract Long-term use of glucocorticoids (GCs) is known to be a predominant cause of osteonecrosis of the femoral head (ONFH). Moreover, GCs can mediate apoptosis of various cell types by exaggerating oxidative stress. We have previously found that Cortistatin (CST) antagonizes oxidative stress and improves cell apoptosis in several conditions. In this study, we detected that the CST expression levels were diminished in patients with ONFH compared with femoral neck fracture (FNF). In addition, a GC-induced rat ONFH model was established, which impaired bone quality in the femoral head. Then, administration of CST attenuated these ONFH phenotypes. Furthermore, osteoblast and endothelial cells were cultured and stimulated with dexamethasone (Dex) in the presence or absence of recombinant CST. As a result, Dex induced impaired anabolic metabolism of osteoblasts and suppressed tube formation in endothelial cells, while additional treatment with CST reversed this damage to the cells. Moreover, blocking GHSR1a, a well-accepted receptor of CST, or blocking the AKT signaling pathway largely abolished the protective function of CST in Dex-induced disorder of the cells. Taken together, we indicate that CST has the capability to prevent GC-induced apoptosis and metabolic disorder of osteoblasts in the pathogenesis of ONFH via the GHSR1a/AKT signaling pathway.

Pseudomonas aeruginosa is one of the leading causes of nosocomial infections worldwide and has emerged as a serious public health threat, due in large part to its multiple virulence factors and remarkable resistance capabilities. Stk1, a eukaryotic-type Ser/Thr protein kinase, has been shown in our previous work to be involved in the regulation of several signalling pathways and biological processes. Here, we demonstrate that deletion of stk1 leads to alterations in several virulence- and resistance-related physiological functions, including reduced pyocyanin and pyoverdine production, attenuated twitching motility, and enhanced biofilm production, extracellular polysaccharide secretion, and antibiotic resistance. Moreover, we identified AlgR, an important transcriptional regulator, as a substrate for Stk1, with its phosphorylation at the Ser143 site catalysed by Stk1. Intriguingly, both the deletion of stk1 and the mutation of Ser143 of AlgR to Ala result in similar changes in the above-mentioned physiological functions. Furthermore, assays of algR expression in these strains suggest that changes in the phosphorylation state of AlgR, rather than its expression level, underlie changes in these physiological functions. These findings uncover Stk1-mediated phosphorylation of AlgR as an important mechanism for regulating virulence and resistance in P. aeruginosa.

ABSTRACTThis study proposes a sandwich structure of triply periodic minimal surface (TPMS) based on the theory of micro-perforated plates. This structure was processed using laser selective melting (SLM) with the acoustic-vibration theoretical model established through the acoustic-electrical analogy method. The theoretical model was verified by comparison with the experimental results, showing that the micro-perforated plate TPMS sandwich structure exhibited good sound absorption performance with the maximum sound absorption coefficients being higher than 0.8. The micro-perforated plate TPMS sandwich resonated when excited by sound waves. The change in the volume fraction affected the size of the cavity volume behind the micro-perforated plate, thereby affecting the resonance frequency. The increase in volume fraction decreased the volume of the cavity behind the structural plate, the sound absorption performance to increase, the resonance frequency to move towards high frequencies, and the sound absorption bandwidth to decrease. The resonance frequency was negatively correlated with the panel thickness. The increase in the panel thickness increased the maximum sound absorption coefficient and decreased the sound absorption bandwidth when the structure resonated. Also, high-order resonance frequencies appeared when the number of cell layers increased to a certain level.

The phenomenon of carbon isotopic fractionation, induced by the transport of methane in tight sedimentary rocks through processes primarily involving diffusion and adsorption/desorption, is ubiquitous in nature and plays a significant role in numerous geological and geochemical systems. Consequently, understanding the mechanisms of transport-induced carbon isotopic fractionation both theoretically and experimentally is of considerable scientific importance. However, previous experimental studies have observed carbon isotope fractionation phenomena that are entirely distinct, and even exhibit opposing characteristics. At present, there is a lack of a convincing mechanistic explanation and valid numerical model for this discrepancy. Here, we performed gas transport experiments under different gas pressures (1–5 MPa) and confining pressures (10–20 MPa). The results show that methane carbon isotope fractionation during natural gas transport through shale is controlled by its pore structure and evolves regularly with increasing effective stress. Compared with the carbon isotopic composition of the source gas, the initial effluent methane is predominantly depleted in 13C, but occasionally exhibits 13C enrichment. The carbon isotopic composition of effluent methane converges to that of the source gas as mass transport reaches a steady state. The evolution patterns of the isotope fractionation curve, transitioning from the initial non-steady state to the final steady state, can be categorized into five distinct types. The combined effect of multi-level transport channels offers the most compelling mechanistic explanation for the observed evolution patterns and their interconversion. Numerical simulation studies demonstrate that existing models, including the Rayleigh model, the diffusion model, and the coupled diffusion-adsorption/desorption model, are unable to describe the observed complex isotope fractionation behavior. In contrast, the multi-scale multi-mechanism coupled model developed herein, incorporating diffusion and adsorption/desorption across multi-level transport channels, effectively reproduces all the observed fractionation patterns and supports the mechanistic rationale for the combined effect. Finally, the potential carbon isotopic fractionation resulting from natural gas transport in/through porous media and its geological implications are discussed in several hypothetical scenarios combining numerical simulations. These findings highlight the limitations of carbon isotopic parameters for determining the origin and maturity of natural gas, and underscore their potential in identifying greenhouse gas leaks and tracing sources.

Abstract The practical application of aqueous Zn‐ion batteries (AZIBs) is hindered by the crazy Zn dendrites growth and the H2O‐induced side reactions, which rapidly consume the Zn anode and H2O molecules, especially under the lean electrolyte and Zn anode. Herein, a natural disaccharide, d‐trehalose (DT), is exploited as a novel multifunctional co‐solvent to address the above issues. Molecular dynamics simulations and spectral characterizations demonstrate that DT with abundant polar −OH groups can form strong interactions with Zn2+ ions and H2O molecules, and thus massively reconstruct the coordination structure of Zn2+ ions and the hydrogen bonding network of the electrolyte. Especially, the strong H‐bonds between DT and H2O molecules can not only effectively suppress the H2O activity but also prevent the rearrangement of H2O molecules at low temperature. Consequently, the AZIBs using DT30 electrolyte can show high cycling stability even under lean electrolyte (E/C ratio = 2.95 µL mAh−1), low N/P ratio (3.4), and low temperature (−12 °C). As a proof‐of‐concept, a Zn||LiFePO4 pack with LiFePO4 loading as high as 506.49 mg can be achieved. Therefore, DT as an eco‐friendly multifunctional co‐solvent provides a sustainable and effective strategy for the practical application of AZIBs.

Carbon dots (CDs), as a new kind of fluorescent nanomaterials, show great potential for application in several fields due to their unique nano-size effect, easy surface functionalization, controllable photoluminescence, and excellent biocompatibility. Conventional preparation methods for CDs typically involve top-down and bottom-up approaches. Doping is a major step forward in CDs design methodology. Chemical doping includes both non-metal and metal doping, in which non-metal doping is an effective strategy for modulating the fluorescence properties of CDs and improving photocatalytic performance in several areas. In recent years, Metal-doped CDs have aroused the interest of academics as a promising nano-doping technique. This approach has led to improvements in the physicochemical and optical properties of CDs by altering their electron density distribution and bandgap capacity. Additionally, the issues of metal toxicity and utilization have been addressed to a large extent. In this review, we categorize metals into two major groups: transition group metals and rare-earth group metals, and an overview of recent advances in biomedical applications of these two categories, respectively. Meanwhile, the prospects and the challenges of metal-doped CDs for biomedical applications are reviewed and concluded. The aim of this paper is to break through the existing deficiencies of metal-doped CDs and fully exploit their potential. I believe that this review will broaden the insight into the synthesis and biomedical applications of metal-doped CDs.

The purpose of the study was to explore effect of four different strains on quality characteristics of soy yogurt. The results showed that four strains were all related to the genus Lactobacillus and N1 was Lacticaseibacillus rhamnosus, N2 was Lacticaseibacillus paracasei, N3 was Lacticaseibacillus plantarum, and N4 was Lacticaseibacillus acidophilus. The result analysis of biochemical, sensory, nutritional, functional and safety properties of fermentation process and end products showed that the soy yogurt fermented with L. rhamnosus N1 had the highest isoflavone content and the lowest phytic acid content; the soy yogurt fermented with L. paracasei N2 had the highest content of free amino acids and oligosaccharides, the lowest content of trypsin inhibitors; the soy yogurt fermented with L. plantarum N3 had the lowest oil content; the soy yogurt fermented with L. acidophilus N4 had optimal functional properties. In summary, N4 was suitable as a fermentation strain for soymilk.

A novel AlGaN/GaN HEMT is proposed to improve its single event transient (SET) effect and breakdown characteristics. The device features an AlGaN back barrier layer and a buried P-GaN island in the back barrier layer (BP-HEMT). First, the P-GaN island not only modulates the electric field distribution and reduces impact ionization at both the blocking state and after ion strike but also increases the hole-electron recombination rate. Therefore, it not only effectively increases the breakdown voltage (BV), but also improves the anti-SET performance owing to decreasing the current peak after ion strike. Second, the AlGaN back barrier confines electrons in the channel, and thus, on one hand, it is beneficial to the high saturation drain current (Id,sat) and low on-state resistance (Ron); on the other hand, it effectively prevents the electrons in the buffer layer introduced by ion strike from reaching the drain electrode both at the blocking state and after ion strike. The TCAD simulation results show that the SET peak drain current of the BP-HEMT is significantly dropped to 0.49 A/mm from 4.17 A/mm of the conventional HEMT with a linear energy transfer (LET) of 0.6 pC/μm, and the BV is significantly increased to 1318.3 V from 175.2 V, as well as the Ron decreases by 11.1%.

Rotor imbalances in wind turbines present safety risks and lead to economic losses, and a method to diagnose rotor imbalances is urgently needed. A diagnostic method for rotor imbalances is proposed in this paper. First, a signal reconstruction method is proposed, and a novel index is used to determine the number of components used in signal decomposition in order to effectively address the interference by noise on the sensor. Second, an entropy calculation method is proposed, and the Gaussian kernel function is used to replace the fuzzy functions. The results indicate significant differences for different types of rotor imbalances. Moreover, it exhibits good noise robustness and a low dependence on the data length. Third, a support vector machine with multiscale kernels is proposed, and kernel functions with various characteristics and scales are combined. It has a well-distributed hyperplane and better classification performance, and it is robust to wind conditions. Finally, the method is tested and verified with varying levels of noise and turbulence. The results demonstrate satisfactory performance because the proposed method can effectively identify rotor imbalances under different noise and wind conditions.

Epithelial growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) have significantly enhanced the treatment outcomes in non-small cell lung cancer (NSCLC) patients harboring EGFR mutations. However, the occurrence of acquired resistance to EGFR-TKIs is an unavoidable outcome observed in these patients. Disruption of the PI3K/AKT/mTOR signaling pathway can contribute to the emergence of resistance to EGFR TKIs in lung cancer. The emergence of PIK3CA mutations following treatment with EGFR-TKIs can lead to resistance against EGFR-TKIs. This review provides an overview of the current perspectives regarding the involvement of PI3K/AKT/mTOR signaling in the development of lung cancer. Furthermore, we outline the state-of-the-art therapeutic strategies targeting the PI3K/AKT/mTOR signaling pathway in lung cancer. We highlight the role of PIK3CA mutation as an acquired resistance mechanism against EGFR-TKIs in EGFR-mutant NSCLC. Crucially, we explore therapeutic strategies targeting PIK3CA-mediated resistance to EGFR TKIs in lung cancer, aiming to optimize the effectiveness of treatment.

Controllable contraception in male animals was demonstrated through the utilization of gold nanorods' photothermal effect to accomplish mild testicular hyperthermia. However, the challenges arising from testicular administration and the non-biodegradability of nanoparticles hinder further clinical implementation. Therefore, a straightforward, non-invasive, and enhanced contraception approach is required. This study explores the utilization of human heavy chain ferritin (HFn) nanocarriers loaded with aggregation-induced emission luminogens (AIEgens) for noninvasive, controllable male contraception guided by Near-Infrared-II (NIR-II) fluorescence imaging. The HFn-caged AIEgens (HFn@BBT) are delivered via intravenous injection and activated by near-infrared irradiation. Lower hyperthermia treatment induces partial damage to the testes and seminiferous tubules, reducing fertility indices by approximately 100% on the 7th day, which gradually recovers to 80% on the 60th day. Conversely, implementation of elevated hyperthermia therapy causes total destruction of both testes and seminiferous tubules, leading to a complete loss of fertility on the 60th day. Additionally, the use of AIEgens in NIR-II imaging offers improved fluorescence efficiency and penetration depth. The findings of this study hold significant promise for the advancement of safe and effective male contraceptive methods, addressing the need for noninvasive and controllable approaches to reproductive health and population control.

The gas-water distribution and production heterogeneity of tight gas reservoirs have been summarized from experimental and geological observations, but the charging and accumulation mechanisms have not been examined quantitatively by mathematical model. The tight gas charging and accumulation mechanisms were revealed from a combination of physical simulation of nuclear magnetic resonance coupling displacement, numerical simulation considering material and mechanical equilibria, as well as actual geological observation. The results show that gas migrates into tight rocks to preferentially form a gas saturation stabilization zone near the source-reservoir interface. When the gas source is insufficient, gas saturation reduction zone and uncharged zone are formed in sequence from the source-reservoir interface. The better the source rock conditions with more gas expulsion volume and higher overpressure, the thicker the gas saturation stabilization and reduction zones, and the higher the overall gas saturation. When the source rock conditions are limited, the better the tight reservoir conditions with higher porosity and permeability as well as larger pore throat, the thinner the gas saturation stabilization and reduction zones, but the gas saturation is high. The sweet spot of tight gas is developed in the high-quality reservoir near the source rock, which often corresponds to the gas saturation stabilization zone. The numerical simulation results by mathematical model agree well with the physical simulation results by nuclear magnetic resonance coupling displacement, and reasonably explain the gas-water distribution and production pattern of deep reservoirs in the Xujiaweizi fault depression of the Songliao Basin and tight gas reservoirs in the Linxing–Huangfu area of the Ordos Basin.

The heavy oil is essentially comprised of asphaltene and nitrogen, sulphur, and oxygen-bearing (NSO) compounds, which records the alteration processes of aerobic biodegradation. In this study, six biodegraded oils were geochemically examined using Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) in negative-ion electrospray ionization (ESI(-)) mode to decipher the NSO heteroatom compounds molecular compositions. The results reveal that the NSO compounds, especially those strongly resistant to biodegradation, i.e., alcohol/phenols, carboxylic acids with various alkyl substitution, naphthenic acids with 2 to 5 naphthenic/aromatic rings, nitrogen- and sulphur-bearing carboxylic acids, and hydroxy acids (O3) and dibasic acids (O4) are enriched in biodegraded oils. The O1, N1O1, and O1S1 classes appear in trace amounts in severely to extremely biodegraded oils (PM > 7) but are abundant in moderately biodegraded oils (PM

Abstract Well-defined multicrystalline multiblock polymers are essential model polymers for advancing crystallization physics, phase separation, self-assembly, and improving the mechanical properties of materials. However, due to different chain properties and incompatible synthetic methodologies, multicrystalline multiblock polymers with more than two crystallites are rarely reported. Herein, by combining polyhomologation, ring-opening polymerization, and catalyst switch strategy, we synthesized a pentacrystalline pentablock quintopolymer, polyethylene-b-poly(ethylene oxide)-b-poly(ε-caprolactone)-b-poly(L-lactide)-b-polyglycolide (PE-b-PEO-b-PCL-b-PLLA-b-PGA). The fluoroalcohol-assisted catalyst switch enables the successful incorporation of a high melting point polyglycolide block into the complex multiblock structure. Solid-state nuclear magnetic resonance spectroscopy, X-ray diffraction, and differential scanning calorimetry revealed the existence of five different crystalline phases.

The sandwich structure based on Triply periodic minimal surface (TPMS) is a lightweight and high-strength multifunctional composite material that combines the versatility of heat exchange, impact resistance, and energy absorption, and has been widely used in various fields such as aviation and aerospace. However, its sound absorption performance has not meant fully studied. In this paper, a Micro-perforated plate Diamond sandwich structure (MPP-DSS) was designed based on TPMS method, which was composed of aluminum alloy solid panel, aluminum alloy macroscopically ordered porous Diamond structure and aluminum alloy micro-perforated plate. The acoustic absorption performance in low and medium frequency band was studied by impedance tube method. The results show that MPP-DSS has higher absorption coefficient and bandwidth than traditional perforated plate structure with the same structural parameters. Increasing the thickness of micro-perforated plate can improve the sound absorption capacity of MPP-DSS in the low frequency range, but the width of the sound absorption band will be narrowed accordingly. Different from the resonant sound absorption mechanism of the traditional perforated plate structure, the sound absorption mechanism of MPP-DSS is the combined effect of resonant sound absorption and friction loss sound absorption. This study broadens the versatility of the TPMS structure and can serve as a reference for the design of integrated load-bearing and sound-absorbing structures.

Abstract Synthetic gene networks in mammalian cells are currently limited to either protein-based transcription factors or RNA-based regulators. Here, we demonstrate a regulatory approach based on circular single-stranded DNA (Css DNA), which can be used as an efficient expression vector with switchable activity, enabling gene regulation in mammalian cells. The Css DNA is transformed into its double-stranded form via DNA replication and used as vectors encoding a variety of different proteins in a wide range of cell lines as well as in mice. The rich repository of DNA nanotechnology allows to use sort single-stranded DNA effectors to fold Css DNA into DNA nanostructures of different complexity, leading the gene expression to programmable inhibition and subsequently re-activation via toehold-mediated strand displacement. The regulatory strategy from Css DNA can thus expand the molecular toolbox for the realization of synthetic regulatory networks with potential applications in genetic diagnosis and gene therapy.

Leghemoglobin is a kind of plant-derived hemoglobin, which can be used as an important flavor catalyst and colorant in the processing of plant protein-based meat because of the property of increasing the fidelity of the product greatly. In order to realize the heterologous expression of leghemoglobin in food-grade microorganisms, the plasmid PESC-TRP carrying leghemoglobin LBC2 gene was transformed into S. cerevisiae CEN.PK2-1C. Kozak sequence was added to promote protein translation, and different promoter sequences were selected to improve the expression of leghemoglobin. The recombinant strain was fermented, and the expression level of LegH protein was analyzed by western blot. The results showed that the inducible promoter GAL1,10 had a significant advantage over the three constitutive promoters TEF1, ADH1 and GAP in LegH expression ability, which was 3.93 times higher than the lowest yield ADH1 promoter. Among the constitutive promoters, TEF1 promoter had the best ability, which was 2.91times that of the ADH1 promoter and 1.2 times that of the GAP promoter. Finally, the protein was concentrated and purified by Ni-column affinity chromatography, and the fermentation concentration of LegH was 2.79 mg/L. This study successfully achieved the heterologous expression of leghemoglobin in Saccharomyces cerevisiae, and after subsequent optimization, it would become another way of leghemoglobin heterologous expression.

Abstract Under the Heisenberg evolution in chaotic quantum systems, initially simple operators evolve into complicated ones and ultimately cover the whole operator space. We study the growth of the operator “size” in this process, which is related to the out-of-time-order correlator (OTOC). We derive the full time evolution of the size distribution in large N quantum mechanics of Majorana fermions. As examples, we apply the formalism to the Brownian SYK model (infinite temperature) and the large q SYK model (finite temperature).

Abstract Earth rotation parameters (ERPs) are essential for transforming between the celestial and terrestrial reference frames, and for high-precision space navigation and positioning. Among the ERPs, polar motion (PM) is a critical parameter for analyzing and understanding the dynamic interaction between the solid Earth, atmosphere, ocean, and other geophysical fluids. Traditional methods for predicting the change in ERPs rely heavily on linear models, such as the least squares (LS) and the autoregressive (AR) model (LS + AR). However, variations in ERP partly reflect non-linear effects in the Earth system, such that the predictive accuracy of linear models is not always optimal. In this paper, long short-term memory (LSTM), a non-linear neural network, is employed to improve the prediction of ERPs. Polar motion prediction experiments in this study are conducted using the LSTM model and a hybrid method LS + LSTM model based on the IERS EOP14C04 time series. Compared with Bulletin A, the PMX and PMY prediction accuracy can reach a maximum of 33.7% and 31.9%, respectively, with the LS + LSTM model. The experimental results show that the proposed hybrid model displays a better performance in mid- and long-term (120–365 days) prediction of polar motion. Graphical Abstract

Aerodynamic contour dimensional accuracy is very important for the stable and safe flight of aerospace vehicles. Nevertheless, due to the influence of various factors such as material properties, machining and manufacturing deviations, and assembly and installation deviations, key structural geometric dimensions are frequently exceeded. Therefore, this paper investigates a data-driven combined vector loop method (VLM)–Skin Model Shapes (SMS) method to realize aerospace vehicle structural geometric accuracy analysis; assembly optimization targeting contour deviation is also achieved. Tests are carried out on a typical aerospace vehicle’s underconstrained structural workpieces to validate the effectiveness of the proposed method.
