Reading the prevailing emotion of groups of people (“crowd emotion”) is critical to understanding their overall intention and disposition. It alerts us to potential dangers, such as angry mobs or panicked crowds, giving us time to escape. A critical aspect of processing crowd emotion is that it must occur rapidly, because delays often are costly. Although knowing the timing of neural events is crucial for understanding how the brain guides behaviors using coherent signals from a glimpse of multiple faces, this information is currently lacking in the literature on face ensemble coding. Therefore, we used magnetoencephalography to examine the neurodynamics in the dorsal and ventral visual streams and the periamygdaloid cortex to compare perception of groups of faces versus individual faces. Forty-six participants compared two groups of four faces or two individual faces with varying emotional expressions and chose which group or individual they would avoid. We found that the dorsal stream was activated as early as 68 msec after the onset of stimuli containing groups of faces. In contrast, the ventral stream was activated later and predominantly for individual face stimuli. The latencies of the dorsal stream activation peaks correlated with participants’ response times for facial crowds. We also found enhanced connectivity earlier between the periamygdaloid cortex and the dorsal stream regions for crowd emotion perception. Our findings suggest that ensemble coding of facial crowds proceeds rapidly and in parallel by engaging the dorsal stream to mediate adaptive social behaviors, via a distinct route from single face perception.

Intra-epidermal electrical stimulation (IEES) has been shown to activate selectively Aδ fibers subserving spinothalamic-mediated sensations. Owing to electrically induced highly synchronous afferent volleys, IEES induces Aδ-mediated evoked potentials at nonpainful intensities, contrasting with thermo-nociceptive laser pulses which entail painful pricking sensations. Here, we recorded intracortical responses from sensory and limbic-cognitive regions of human subjects in response to IEE and laser stimuli, in order to test the hypothesis that IEES could dissociate the sensory from nonsensory networks of nociceptive processing. Intracortical evoked potentials were obtained in 11 epileptic patients with stereotactically implanted electrodes in sensory regions receiving spinothalamic afferents (posterior insula), limbic regions receiving spino-parabrachial input (amygdalar nucleus), and high-order affective-cognitive regions (anteromedial frontal cortex, including perigenual anterior cingulate and rostromedial prefrontal areas). Responses in the sensory posterior insula were of similar amplitude and latency to IEE and laser stimuli (after accounting for heat-transduction time of laser), and consistent in both cases with spinothalamic activation. However, responses to IEES in the amygdala and the anteromedial frontal regions were inconsistent and significantly smaller compared to those evoked to the laser stimulation. Thus, IEES can effectively activate the spinothalamic-sensory system with little recruitment of affective-motivational networks, including those triggered by spino-parabrachio-amygdalar projections. The fact that identical sensory responses were associated to either painful or nonpainful percepts underscores that subjective pain perception is not solely dependent on the sensory recruitment, but rather on the combined activation of sensory, limbic and cognitive areas with precise spatiotemporal relations.

The location and distribution of the calcium-binding protein calbindin-D28k (CB) has been considered to be of great value as a neuronal marker for identifying distinct brain regions and discrete neuronal populations. In the amygdaloid complex (AC), the balance of excitatory and inhibitory inputs is controlled by CB immunoreactive interneurons. Alterations of inhibitory mechanisms in the AC may play a role in the emotional symptomatology of neurological diseases like Alzheimer's and psychiatric disorders like posttraumatic stress disorder. The present investigation examined the distribution and morphology of CB-containing neurons, neuropils and fibers in marmoset monkey ACs by using immunohistochemical and morphometrical methods. We recognized four types of CB cells in the AC: type 1 (multipolar), type 2 (spherical or bipolar), type 3 (pyramidal) and type 4 (halo cells), a cell type specific to the marmoset located in the basal and central nuclei. We detected CB cells in all nuclei and areas of the AC, where most of the cells were present in the deep nuclei (lateral, basal, accessory basal and paralaminar). In the superficial nuclei (the nucleus of the lateral olfactory tract, medial nucleus, periamygdaloid cortex and cortical nuclei), the CB cells were abundant in layers 2 and 3. The intercalated nuclei contained small densely packed cells. The CB neuropils were particularly dense in layer 1 of the superficial nuclei, in the deep nuclei and in the amygdalohippocampal area. Large CB immunoreactive neurons in the white matter and fibers with varicosities were found in the myelin tracts that surrounded the AC. These findings are the first step in determining whether some of these cells are specifically disrupted in pathological states.

The objective was to study effects of fear on brain activity, functional connectivity and brain-behavior relationships during symptom provocation in subjects with specific phobia. Positron emission tomography (PET) and (15)O water was used to measure regional cerebral blood flow (rCBF) in 16 women phobic of either snakes or spiders but not both. Subjects watched pictures of snakes and spiders serving either as phobic or fear-relevant, but non-phobic, control stimuli depending on phobia type. Presentation of phobic as compared with non-phobic cues was associated with increased activation of the right amygdala and cerebellum as well as the left visual cortex and circumscribed frontal areas. Activity decreased in the prefrontal, orbitofrontal and ventromedial cortices as well as in the primary somatosensory cortex and auditory cortices. Furthermore, amygdala activation correlated positively with the subjective experience of distress. Connectivity analyses of activity in the phobic state revealed increased functional couplings between voxels in the right amygdala and the periamygdaloid area, fusiform gyrus and motor cortex. During non-phobic stimulation, prefrontal activity correlated negatively with amygdala rCBF, suggesting a phobia-related functional decoupling. These results suggest that visually elicited phobic reactions activate object recognition areas and deactivate prefrontal areas involved in cognitive control over emotion-triggering areas like the amygdala, resulting in motor readiness to support fight or flight.

In rats and primates, the central nucleus of the amygdala (CeN) is most known for its role in responses to fear stimuli. Recent evidence also shows that the CeN is required for directing attention and behaviors when the salience of competing stimuli is in flux. To examine how information flows through this key output region of the primate amygdala, we first placed small injections of retrograde tracers into the subdivisions of the central nucleus in Old world primates, and examined inputs from specific amygdaloid nuclei. The amygdalostriatal area and interstitial nucleus of the posterior limb of the anterior commissure (IPAC) were distinguished from the CeN using histochemical markers, and projections to these regions were also described. As expected, the basal nucleus and accessory basal nucleus are the main afferent connections of the central nucleus and transition zones. The medial subdivision of the central nucleus (CeM) receives a significantly stronger input from all regions compared to the lateral core subdivision (CeLcn). The corticoamygdaloid transition zone (a zone of confluence of the medial parvicellular basal nucleus, paralaminar nucleus, and the sulcal periamygdaloid cortex) provides the main input to the CeLcn. The IPAC and amygdalostriatal area can be divided in medial and lateral subregions, and receive input from the basal and accessory basal nucleus, with differential inputs according to subdivision. The piriform cortex and lateral nucleus, two important sensory interfaces, send projections to the transition zones. In sum, the CeM receives broad inputs from the entire amygdala, whereas the CeLcn receives more restricted inputs from the relatively undifferentiated corticoamygdaloid transition region. Like the CeN, the transition zones receive most of their input from the basal nucleus and accessory basal nucleus, however, inputs from the piriform cortex and lateral nucleus, and a lack of input from the parvicellular accessory basal nucleus, are distinguishing afferent features.

The amygdala complex is a heterogeneous group of temporal lobe brain structures involved in the processing of biologically significant sensory stimuli and in the generation of appropriate responses to them. The amygdala has also been implicated in certain forms of emotional learning and memory. While much progress has been made in understanding neural processing in the basolateral subgroup of the amygdala, physiological studies in the cortical regions of the complex, also known as olfactory amygdala, are missing. Using a rat brain slice preparation, we conducted whole-cell recordings on pyramidal neurons of the periamygdaloid cortex and the anterior cortical nucleus, two structures receiving direct connections from the olfactory bulb. Upon depolarization by current injection through the recording electrode, a fraction of periamygdaloid cortex and most anterior cortical nucleus layer II pyramidal neurons displayed an intermittent discharge pattern, where clusters of action potentials were interspersed by periods of membrane potential subthreshold oscillations. Oscillations frequency increased with membrane potential and correlated linearly with the cluster spiking frequency. Frequency ranged from 3 to 20 Hz, considering different cells and membrane potential values (up to approximately 30 mV above resting potentials of typically approximately -70 mV). Subthreshold oscillations were preserved after pharmacological inhibition of fast excitatory and inhibitory synaptic transmission, but were abolished by application of the sodium channel blocker tetrodotoxin. We conclude that pyramidal neurons of the olfactory cortical amygdala display intrinsically generated voltage-dependent membrane potential rhythmic fluctuations in the theta-low beta range, requiring the activation of a sodium conductance.

Though both the amygdala and the serotonin system appear to play critical roles in regulating fear and anxiety, little is known regarding the organization of serotonergic inputs to the primate amygdala. The present study employed immunohistochemistry to determine the distribution of serotonin fibers in the macaque amygdala. The brains of three adult male Macaca fascicularis monkeys were prepared for histological analysis using a polyclonal antibody to serotonin. The macaque amygdala is densely innervated by serotonergic fibers and demonstrates a distinctive pattern of fiber distribution and density among the 13 nuclei and cortical areas. The highest density of 5-hydroxytryptamine immunoreactive fibers is observed in the central nucleus, the nucleus of the lateral olfactory tract, the paralaminar nucleus, the anterior amygdaloid area and a small region of the amygdalohippocampal area. Moderate fiber densities are found in portions of the basal, lateral, and intercalated nuclei. The lowest fiber densities are observed in the accessory basal, posterior cortical, anterior cortical and medial nuclei, and in subregions of the periamygdaloid cortex. The present study provides evidence that the serotonergic system can have substantial influence on the ongoing activity of the amygdaloid complex.

Projections from the amygdala to the piriform cortex are proposed to provide a pathway via which the emotional system can modulate the processing of olfactory information as well as mediate the spread of seizure activity in epilepsy. To understand the details of the distribution and topography of these projections, we injected the anterograde tracer Phaseolus vulgaris-leucoagglutinin into different nuclear divisions of the amygdaloid complex in 101 rats and analyzed the distribution and density of projections in immunohistochemically processed preparations. The heaviest projections from the amygdala to the piriform cortex originated in the medial division of the lateral nucleus, the periamygdaloid and sulcal subfields of the periamygdaloid cortex, and the posterior cortical nucleus. The heaviest terminal labeling was observed in layers Ib and III of the medial aspect of the posterior piriform cortex. Lighter projections to the posterior piriform cortex originated in the dorsolateral division of the lateral nucleus, the magnocellular and parvicellular divisions of the basal and accessory basal nuclei, and the anterior cortical nucleus. The projections to the anterior piriform cortex were light and originated in the dorsolateral and medial divisions of the lateral nucleus, the magnocellular division of the basal and accessory basal nuclei, the anterior and posterior cortical nuclei, and the periamygdaloid subfield of the periamygdaloid cortex. The results indicate that only selective amygdaloid nuclei or their subdivisions project to the piriform cortex. In addition, substantial projections from several amygdaloid nuclei converge in the medial aspect of the posterior piriform cortex. Via these projections, the amygdaloid complex can modulate the processing of olfactory information in the piriform cortex. In pathologic conditions such as epilepsy, these connections might provide pathways for the spread of seizure activity from the amygdala to extra-amygdaloid regions. J. Comp. An emerging literature on the neuroanatomical correlates of social cognition in pervasive developmental disorders is reviewed. Studies conducted with high-functioning adults with autism or Asperger's syndrome highlight patterns of decreased activation in ventromedial prefrontal cortex, temporo-parietal junction, amygdala, and periamygdaloid cortex, along with aberrantly increased activation in primary sensory cortices. Future studies should extend these important initial results to younger and more severely affected subjects.

We have previously described the origins of neocortical inputs to the lateral nucleus of the macaque monkey amygdala based on retrograde tracing studies. Here we report results from studies that have attempted to confirm the projections from several candidate afferent regions using (3)H-amino acid autoradiography as an anterograde tracer. We have charted, based on the results of 33 separate injections, the topographic distribution of cortical projections throughout the amygdala. Areas TE and TEO of the inferotemporal cortex, portions of the superior temporal gyrus, and the granular region of the insula project primarily to the lateral nucleus, with little or no innervation of other amygdaloid nuclei. In contrast, orbitofrontal, medial prefrontal, and anterior cingulate regions project primarily to the basal and accessory basal nuclei and provide little innervation to the lateral nucleus. The orbitofrontal and medial prefrontal cortices, but not the anterior cingulate cortex, project to medially situated amygdaloid areas such as the cortical and medial nuclei and to the periamygdaloid cortex. The agranular and dysgranular insula, the parainsula, and rostral portions of the superior temporal gyrus project both to the lateral, basal, and accessory basal nuclei and to the medially situated nuclei. Projections to the central nucleus are particularly prominent from these regions. These data are discussed in relation to the hierarchical processing of sensory information that occurs within the amygdaloid complex.

The claustrum and the endopiriform nucleus contribute to the spread of epileptiform activity from the amygdala to other brain areas. Data of the distribution of pathways underlying the information flow between these regions are, however, incomplete and controversial. To investigate the projections from the amygdala to the claustrum and the endopiriform nucleus, we injected the anterograde tracer Phaseolus vulgaris leucoagglutinin into various divisions of the amygdaloid complex, including the lateral, basal, accessory basal, central, anterior cortical and posterior cortical nuclei, the periamygdaloid cortex, and the amygdalohippocampal area in the rat. Analysis of immunohistochemically processed sections reveal that the heaviest projections to the claustrum originate in the magnocellular division of the basal nucleus. The projection is moderate in density and mainly terminates in the dorsal aspect of the anterior part of the claustrum. Light projections from the parvicellular and intermediate divisions of the basal nucleus terminate in the same region, whereas light projections from the accessory basal nucleus and the lateral division of the amygdalohippocampal area innervate the caudal part of the claustrum. The most substantial projections from the amygdala to the endopiriform nucleus originate in the lateral division of the amygdalohippocampal area. These projections terminate in the central and caudal parts of the endopiriform nucleus. Lighter projections originate in the anterior and posterior cortical nuclei, the periamygdaloid cortex, the medial division of the amygdalohippocampal area, and the accessory basal nucleus. These data provide an anatomic basis for recent functional studies demonstrating that the claustrum and the endopiriform nucleus are strategically located to synchronize and spread epileptiform activity from the amygdala to the other brain regions. These topographically organized pathways also provide a route by means of which the claustrum and the endopiriform nucleus have access to inputs from the amygdaloid networks that process emotionally significant information.

The dynamics of limbic epileptogenesis was studied using a model of electrical kindling from different rostrocaudal segments of the cortical nucleus of the amygdala (CoA). Statistically significant relationships were found between the rostrocaudal location of the site of focal stimulation of the CoA and the behavioral and electrographic manifestations of seizure activity. The data obtained suggest that different rostrocaudal segments of the CoA play different roles in the genesis of limbic epilepsy. Temporal lobe epilepsy (or limbic epilepsy) is the most common form of this disease in humans [3, 10, 11]. Patients with this form of epilepsy suffer from complex partial (focal) and secondarily generalized seizures. In most cases, pharmacological resistance develops that makes it difficult to efficiently control seizures or significantly decrease their frequency using anticonvulsant drugs [13]. Because of the high incidence of temporal lobe epilepsy, difficulties in its pharmacotherapy, and the diversity of associated structural and functional brain lesions, of particular interest are experimental studies in which the pathogenetic mechanisms of temporal epilepsy are analyzed using various animal models [9, 10]. Electrical kindling (low-level stimulation that causes a progressively increasing response when applied repeatedly) [8] is the most adequate chronic model of temporal epilepsy that closely follows the natural course of its development with respect to a number of parameters [3‐6, 9, 10]. The role of the CoA in initiating the epileptiform activity of the amygdaloid complex (AC) and of the entire brain remains largely obscure. Our early studies [4, 5] and a recent study by Pitkanen et al. [11] implicated the CoA in spreading the seizure activity, especially in generating mirror foci in the amygdala of the contralateral hemisphere. In contrast, Cain [7] presents evidence that the CoA is of little significance in the epileptogenic process. However, taken together, the fact that the CoA is morphologically closely related to the piriform cortex and to some other structures of the AC in the ipsi- and contralateral hemispheres, and the clinical data indicating that the mediobasal temporal lobe is involved in the genesis of human temporal lobe epilepsy put in doubt Cain’s point of view. In the rat AC, the CoA lies medial to the piriform cortex, being partially exposed on the basal surface of the brain [1, 2]. The CoA is elongated rostrocaudally. This makes it possible to perform separately microstimulation of its anterior and posterior subniclei, which are known for their complex and heterogeneous structure [2]. The CoA can be divided into the anterior CoA (CoAa), the periamygdaloid cortex (PAC), and the posterior CoA (CoAp), based on the differences in their neuron composition, cytoarchitectonics, histochemistry, afferentation, and involvement in the main conducting systems of the AC [1]. The purpose of this study was to determine whether the behavioral and electrographic manifestations of epileptogenesis depend on the rostrocaudal location of the site of focal stimulation.

The amygdalo-prefrontal network is thought to play an important role in fear conditioning and other associative learning processes. We investigated amygdalo-prefrontal connectivity data from anatomical tracing studies using an advanced databasing approach (www.cocomac.org) and multivariate statistics. Both amygdaloid and prefrontal cortical areas were found to form three distinct clusters with (i) widespread, (ii) absent, and (iii) highly specific connections, respectively. Major amygdaloid sources of projections were the accessory basal (AB), basal (B), lateral (L) nuclei and the periamygdaloid cortex (PAC). Major prefrontal targets were orbital areas 13 and 14, as well as medial areas 24 and 25.

We have used the anterograde tracer, Phaseolus vulgaris-leucoagglutinin (PHA-L) to study the intrinsic projections of the lateral nucleus of the Macaca fascicularis monkey amygdaloid complex. A reanalysis of the monkey lateral nucleus indicated that there are at least four distinct cytoarchitectonic divisions: dorsal, dorsal intermediate, ventral intermediate, and ventral. The major projections within the lateral nucleus originate in the dorsal, dorsal intermediate, and ventral intermediate divisions and terminate in the ventral division. The ventral division also projects to itself but does not project significantly to the other divisions of the lateral nucleus. Thus, the ventral division appears to be a site of convergence for information entering all other portions of the lateral nucleus. There are substantial regional and topographic differences in the projections from each of the lateral nucleus divisions to other amygdaloid nuclei. The dorsal division projects to all divisions of the basal and accessory basal nuclei, to the periamygdaloid cortex, the nucleus of the lateral olfactory tract, the dorsal division of the amygdalohippocampal area, and the lateral capsular nuclei. The dorsal intermediate division projects to the intermediate and parvicellular divisions of the basal nucleus, to the parvicellular division of the accessory basal nucleus, and to the periamygdaloid cortex. The ventral intermediate division projects to the magnocellular division of the accessory basal nucleus and to the parvicellular division of the basal nucleus. The major projections from the ventral division are directed to the parvicellular division of the basal nucleus, the parvicellular division of the accessory basal nucleus, the medial nucleus, and the periamygdaloid cortex. Projections from all portions of the lateral nucleus to the central nucleus are generally very light. It appears, therefore, that each division of the lateral nucleus originates topographically organized projections to the other amygdaloid areas that terminate in distinct portions of the target regions. The topographic organization of intrinsic amygdaloid projections raises the possibility that serial and parallel sensory processing may take place within the amygdaloid complex. J. Comp. Echoplanar functional magnetic resonance imaging (fMRI) was used in normal human subjects to investigate the role of the amygdala in conditioned fear acquisition and extinction. A simple discrimination procedure was employed in which activation to a visual cue predicting shock (CS+) was compared with activation to another cue presented alone (CS−). CS+ and CS− trial types were intermixed in a pseudorandom order. Functional images were acquired with an asymmetric spin echo pulse sequence from three coronal slices centered on the amygdala. Activation of the amygdala/periamygdaloid cortex was observed during conditioned fear acquisition and extinction. The extent of activation during acquisition was significantly correlated with autonomic indices of conditioning in individual subjects. Consistent with a recent electrophysiological recording study in the rat (Quirk et al. 1997), the profile of the amygdala response was temporally graded, although this dynamic was only statistically reliable during extinction. These results provide further evidence for the conservation of amygdala function across species and implicate an amygdalar contribution to both acquisition and extinction processes during associative emotional learning tasks.

Negative affect is critical for conferring vulnerability to opiate addiction as reflected by the high comorbidity of opiate abuse with major depressive disorder (MDD). Rodent models implicate amygdala prodynorphin (Pdyn) as a mediator of negative affect; however, evidence of PDYN involvement in human negative affect is limited. Here, we found reduced PDYN mRNA expression in the postmortem human amygdala nucleus of the periamygdaloid cortex (PAC) in both heroin abusers and MDD subjects. Similar to humans, rats that chronically self-administered heroin had reduced Pdyn mRNA expression in the PAC at a time point associated with a negative affective state. Using the in vivo functional imaging technology DREAMM (DREADD-assisted metabolic mapping, where DREADD indicates designer receptors exclusively activated by designer drugs), we found that selective inhibition of Pdyn-expressing neurons in the rat PAC increased metabolic activity in the extended amygdala, which is a key substrate of the extrahypothalamic brain stress system. In parallel, PAC-specific Pdyn inhibition provoked negative affect–related physiological and behavioral changes. Altogether, our translational study supports a functional role for impaired Pdyn in the PAC in opiate abuse through activation of the stress and negative affect neurocircuitry implicated in addiction vulnerability.

Information from most of the sensory modalities enters the amygdala via the lateral nucleus. The olfactory information, however, arrives at the amygdala through the superficial nuclei, including the periamygdaloid cortex. To find out whether the olfactory information can modulate the processing of sensory information in the lateral nucleus we injected Phaseolus vulgaris leucoagglutinin, an anterograde axonal tracer, into the different divisions of the periamygdaloid cortex. We found that the PAC division of the periamygdaloid cortex projects to the ventrolateral and medial divisions, but not the dorsolateral division, of the lateral amygdaloid nucleus. Therefore, the projection from the PAC to the lateral nucleus provides a route, by which the olfactory information may become associated with other sensory modalities. Also, together with our previous finding that the lateral nucleus projects to the periamygdaloid cortex, the present data demonstrate that the lateral nucleus and the PAC are reciprocally connected.

Previous studies have shown that the orbital and medial prefrontal cortex (OMPFC) is extensively connected with medial temporal and cingulate limbic structures. In this study, the organization of these projections was defined in relation to architectonic areas within the OMPFC. All of the limbic structures were substantially connected with the following posterior and medial orbital areas: the posteromedial, medial, intermediate, and lateral agranular insular areas (Iapm, Iam, Iai, and Ial, respectively) and areas 11m, 13a, 13b, 14c and 14r. In contrast, lateral orbital areas 12o, 12m, and 12l and medial wall areas 24a,b and 32 were primarily connected with the amygdala, the temporal pole, and the cingulate cortex. Data were not obtained on the posteroventral medial wall. Three distinct projections were recognized from the basal amygdaloid nucleus: 1) The dorsal part projected to area 12l; 2) the ventromedial part projected to most areas in the posterior and medial orbital cortex except for area Iai, 12o, 13a, and 14c; and 3) the ventrolateral part projected to orbital areas 12o, Iai, 13a, 14c, and to the medial wall areas. The accessory basal and lateral amygdaloid nuclei projected most strongly to areas in the posterior and medial orbital cortex. The medial, anterior cortical, and central amygdaloid nuclei and the periamygdaloid cortex were connected with the posterior orbital areas. The projection from the hippocampus originated from the rostral subiculum and terminated in the medial orbital areas. The same region was reciprocally connected with the anteromedial nucleus of the thalamus, which received input from the rostral subiculum. The parahippocampal cortical areas (including the temporal polar, entorhinal, perirhinal, and posterior parahippocampal cortices) were primarily connected with posterior and medial orbital areas, with some projections to the dorsal part of the medial wall. The rostral cingulate cortex sent fibers to the medial wall, to the medial orbital areas, and to lateral areas 12o, 12r, and Iai. The posterior cingulate gyrus, including the caudomedial lobule, was especially strongly connected with area 11m.

The amygdaloid complex plays an essential role in auditory fear conditioning of the Pavlovian type. The available evidence suggests that the lateral nucleus is the input station of the amygdala for auditory conditioned stimuli, whereas the central medial nucleus is the output for conditioned fear responses. However, the intrinsic pathway transmitting auditory information about the conditioned stimulus from the lateral to the central medial nuclei is unknown as there are no direct projections between these nuclei. The present study was undertaken to determine if the main intra-amygdaloid targets of the lateral nucleus, namely the basomedial and basolateral nuclei, project to the central medial nucleus. To this end, iontophoretic injections of the anterograde tracer Phaseolus vulgaris-leucoagglutinin were performed in these nuclei. To rule out the possibility that the anterograde labeling reflected passing fibers merging with the major fiber bundles that course in and around the central medial nucleus, labeled terminals and varicosities were observed in the electron microscope. It was determined that the basolateral and basomedial nuclei have partially overlapping intraamygdaloid targets. They both project to the central medial nucleus, nucleus of the lateral olfactory tract and peri-amygdaloid cortex, but have limited projections to each other. Small Phaseolus vulgaris-leucoagglutinin injections in both nuclei gave rise to prominent intranuclear projections but only the basomedial nucleus was found to project to the lateral and anterior cortical nuclei. At the electron microscopic level, all labeled axon terminals and varicosities formed asymmetric synapses (n = 245) with dendritic spines (83%) or with dendritic shafts (17%). This is the first unambiguous demonstration that the basolateral and basomedial nuclei project to the central medial nucleus. Since these nuclei constitute the main intra-amygdaloid targets of the lateral nucleus, they represent likely candidates for the transmission of auditory conditioned stimuli to the central medial nucleus in auditory fear conditioning.

We addressed the question whether the projection neurons of the olfactory bulb, i.e. the mitral and tufted cells, are immunoreactive for the calcium-binding protein, calretinin. The following approaches were adopted: (1) light and electron microscopic calretinin-immunocytochemistry; (2) neuroanatomical tracing combined with calretinin-immunocytochemistry according to double-peroxidase and double-fluorescence protocols; (3) unilateral lesion of the olfactory bulb combined with calretinin-immunocytochemistry. The experiments were carried out in rats. Immunostaining of brain sections revealed weakly calretinin-immunopositive mitral cell bodies. Tufted cells were immunonegative. In contrast, fibers in the lateral olfactory tract were strongly immunopositive. Dense immunostaining was also present in a superficial band in layer I of the olfactory tubercle, piriform cortex, periamygdaloid cortex, and in the lateral entorhinal cortex. In electron microscopic preparations of these target areas we observed immunoreaction product in axons and axon terminals. The latter invariably formed asymmetrical synapses, mostly with dendritic spines. Injections of the neuroanatomical tracer biotinylated dextran amine (BDA) into the olfactory bulb produced labeled fibers which remained completely restricted to the superficial, calretinin-immunopositive band in layer I in the above-mentioned cortical forebrain areas. We noted colocalization of transported BDA and calretinin-immunoreactivity in mitral cells, in fibers in the lateral olfactory tract and in fibers in the piriform cortex. Olfactory bulb lesions produced depletion of calretinin-immunoreactivity in the lateral olfactory tract and the superficial band in the olfactory cortex-related areas. Together these data firmly indicate that mitral cells and their axons are calretinin-immunoreactive.(ABSTRACT TRUNCATED AT 250 WORDS)

Calbindin-D28k is a calcium-binding protein located in a variety of neuronal cell types in many regions of the central nervous system. In the present study, we describe the distribution of calbindin-D28k-immunoreactive cells, fibers, and terminals in the monkey amygdaloid complex. Calbindin-D28k-immunoreactive neurons could be divided into four major cell types. Neurons of the first three cell types demonstrated clearly stained dendrites that were either aspiny or had a few spines on their distal portions. Type 1 cells were small, stellate, or multipolar and found throughout the amygdala. Type 2 cells were large, multipolar and were most commonly found in the deep nuclei, particularly in the lateral nucleus, intermediate division of the basal nucleus, accessory basal nucleus and in the periamygdaloid cortex. Type 3 cells were fusiform, of various sizes, and were found throughout the amygdala. Type 4 cells were quite large and lightly stained; the dendrites of these cells were usually unstained. The size, shape, and location of Type 4 labeled cell bodies suggested that they might be the large, modified pyramidal cells that constitute the projection neurons of the amygdala. Type 4 cells were observed primarily in the lateral, basal, and accessory basal nuclei and in the periamygdaloid cortex. Calbindin-D28k-immunoreactive fibers and terminals were difficult to observe in the amygdala partly because of a diffuse, finely granular neuropil labeling that was particularly dense in the anterior cortical and medial nuclei, in the central nucleus, and in the periamygdaloid cortex. The neuropil labeling was substantially lighter in the lateral, basal, and accessory basal nuclei. Conspicuous linear profiles resembling the “calbindin bundles” of the neocortex were evident in large numbers in the accessory basal nucleus, the medial portion of the parvicellular division of the basal nucleus, in the amygdalohippocampal area, and in the periamygdaloid cortex. There were calbindin-D28k-positive fibers in the stria terminalis and in the ventral amygdalofugal pathway. When the distributions of calbindin-D28k and parvalbumin immunoreactivity in the monkey amygdaloid complex were compared, it appeared that the overall distribution of these two calcium-binding proteins was generally complementary rather than overlapping. © 1993 Wiley-Liss, Inc.

Previous anatomical studies of corticotropin-releasing factor (CRF)-like immunoreactivity in rat brain have reported prominent clustering of neuronal elements containing this peptide within the amygdala. The highest concentrations of both CRF-positive cells and fibers were evident in the central nucleus, an observation consistent with the putative role of this peptide in autonomic and endocrine regulation. In addition, lower densities of CRF-positive somata and processes have boon noted in other amygdaloid nuclei. However, the distribution of CRF-like immunoreactivity in the amygdala has not been described for any primate species. Such a description would be of interest since substantial differences in the distribution of CRF in rodent and primate have been reported for other brain regions. The present study uses immunohistochemical methods, with a polyclonal antiserum directed against the human form of CRF, to determine the distribution of this peptide in non-colchicine-treated monkeys (Saimiri sciureus). Within the amygdaloid complex, the most numerous and concentrated collections of CRF-positive neurons were seen in the basal and lateral nuclei. The highest densities of CRF-positive fibers and terminals were seen in the lateral and central amygdaloid nuclei. Moderately dense plexuses of CRF-positive fibers also were seen in layer Ia of the periamygdaloid cortex, nucleus of the lateral olfactory tract, anterior and posterior cortical nuclei, and the medial nucleus. Thus, the distribution of CRF-like immunoreactivity differs substantially in monkey and rat amygdala. Since CRF-positive perikarya in monkey are most prominent in nuclei with pronounced interconnections with neocortex, these differences may be an integral component of the increased cortical development that characterizes the primate brain. © 1992 Wiley-Liss, Inc.

Our earlier electrophysiological work provided evidence of a direct input to the supraoptic nucleus (SON) from the olfactory bulbs; however, these experiments could not determine if the input originated in the main and/or accessory portions of the olfactory bulb. Here, a connection between the accessory olfactory bulb (AOB) and the SON of the rat was examined using a combination of anatomic techniques. We employed neurophysin immunocytochemistry to delineate the morphological boundaries of the SON and the proximal arborizations of supraoptic dendrites. Accessory olfactory bulb efferents to the SON were studied by injection of wheat germ agglutinin conjugated to horseradish peroxidase (WGA-HRP) into the AOB. The distribution of retrogradely labeled cells within the AOB was also determined after injection of either rhodamine-labeled latex microspheres (rhodamine beads) or Fluoro-Gold (FG) into the SON. Neurophysin immunocytochemistry revealed that SON dendrites extended beyond the generally accepted boundaries of the nucleus, coursing ventrolaterally along the surface of the periamygdaloid cortex. Anterograde tract tracing with WGA-HRP labeled AOB efferents including a dense plexus of terminals and fibers around the ipsilateral SON along the path of the ventrally projecting dendrites. Injections of retrograde tracers into the SON resulted in rhodamine bead or FG labeling of mitral cells throughout the ipsilateral AOB. Taken together, these anatomic studies suggest a direct projection from the accessory olfactory bulb to the SON of the rat and thus a vomeronasal organ to SON pathway.

Evoked potential (EPs) and responses of the medial (MPO) and lateral (LPO) preoptic region (RPO) and adjacent structures of the hypothalamus to stimulation of the prefrontal (area 8) and cingulate (area 24) cortex, piriform lobe (periamygdaloid cortex), and hippocampus (area CA3) were investigated in acute experiments on cats under ketamine anesthesia. The most pronounced EPs were observed in the RPO after stimulating the piriform and cingulate cortex. A close relation was found between neuronal responses and EP components. The majority of neurons responding to stimulation of various cortical structures were localized in the LPO, where primarily excitatory responses dominate. The MPO contained somewhat fewer neurons responding to cortical stimulation, and the dominant response here was primarily inhibitory. The ratio of inhibitory and excitatory responses in the LPO was 0.6:1 and in the MPO 5.8:1. Primarily in-inhibitory responses dominated also in the LPO zone adjacent to the bed nucleus stria terminalis (BST) and primarily excitatory in the region surrounding the supraoptic nucleus (SO) (respective ratios 4.9:1 and 0.7:1). The RPO is a broad convergence zone, where 3/4 of the neurons responded to stimuli of two and more cortical regions.

Responses of neurons of the medial (MPO) and lateral (LPO) preoptic region (RPO) and adjacent hypothalamic structures to serial stimuli (6–300/sec) of the prefrontal (area 8) and cingulate (area 24) cortex, piriform lobe (periamygdaloid cortex — RPA), and hippocampus (area CA3) were investigated in acute experiments on cats under ketamine anesthesia. Four main types of responses were found: excitatory, inhibitory, excitatory on-off effect, and inhibitory on-off effect. With the use of stimuli with increasing frequencies, the direction of the response remained constant, only its intensity changed. Neurons responding to presentation of serial stimuli were localized mainly in the central part of the MPO and basal part of the LPO, where the most pronounced foci of convergence were observed. During serial stimulation of cortical structures, inhibitory responses occurred considerably more often than excitatory (ratio 3.4:1). The presence of a gradient of inhibition was established from new to old (in a phylogenetic respect) brain formations in a number of stimulated structures. In the case of stimulating the neocortex (proreal gyrus), the predominance of inhibitory responses over excitatory was minimum (1.7:1); it increased (1.9:1) in the case of stimulating the intermediate cortex (cingulate gyrus), still more (4.5:1) under conditions of stimulating the paleocortex (periamygdaloid cortex), and in the case of stimulating the archicortex (10.2:1).

Both the amygdala and hippocampus are damaged in drug-resistant temporal lobe epilepsy (TLE), suggesting that amygdalo-hippocampal interconnectivity is compromised in TLE. Therefore, we examined one of the major projections from the amygdala to the hippocampus, the projection from the amygdala to the CA1 subfield of the hippocampus/subiculum border region, and assessed whether it is preserved in rats with spontaneous seizures. Male Wistar rats were injected with kainic acid (9 mg/kg, i.p.) to induce chronic epilepsy. The occurrence of spontaneous seizures was monitored 5 or 15 weeks later by video-recording the rats for up to 5 days. Saline-injected animals served as controls. Thereafter, the retrograde tracer Fluoro-gold was injected into the border region of the temporal CA1/subiculum. Rats were perfused for histology 1-2 weeks later and sections were immunohistochemically processed to detect Fluoro-gold-positive cells. Comparison of the labeling in control and epileptic tissue indicated that a large cluster of retrogradely labeled cells in the parvicellular division of the basal nucleus was well preserved in epilepsy, even when the neuronal damage in the amygdala was substantial. Another large cluster of retrogradely labeled cells in the lateral division of the amygdalo-hippocampal area, the posterior cortical nucleus (part of the vomeronasal amygdala), and the periamygdaloid cortex (part of the olfactory amygdala), however, had disappeared in epileptic brain in parallel to severe neuronal loss in these nuclei. These data demonstrate that a projection from the parvicellular division of the basal nucleus to the temporal CA1/subiculum region is resistant to status epilepticus-induced neuronal damage and provides a candidate pathway by which seizure activity can spread and propagate from the amygdala to the hippocampal formation.

Synaptic plasticity in the amygdala appears to be necessary for the generation of emotional memories. However, the molecular bases of this plasticity are not fully understood. Because the polysialylated form of the neural cell adhesion molecule (PSA-NCAM) has been implicated in memory consolidation in the hippocampus and temporal cortex, we have studied in detail the expression of this molecule in the adult rat amygdala with an antibody against PSA-NCAM. Our results demonstrate for the first time the presence of PSA-NCAM in the adult rat amygdala. Immunoreactive somata and processes are abundant in the amygdalo-hippocampal transition area, central nucleus, intra-amygdaloid bed nucleus of the stria terminalis, anterior and posterior cortical nuclei, periamygdaloid cortex and medial nucleus of the amygdala. In addition PSA-NCAM immunoreactive neuronal somata and processes exist in the lateral, basal and accessory basal nuclei, anterior amygdaloid area and amygdalo-striatal area. The presence of this molecule in areas that receive olfactory or vomeronasal input could reflect the intrinsic plasticity of these chemosensory systems. PSA-NCAM expression in the lateral amygdala could indicate its participation in the plastic events that lead to the generation of emotional memories such as those related to fear conditioning.

The ventral striatum is the part of the striatum associated with reward and goal-directed behaviors, which are mediated in part by inputs from the amygdala. The ventral striatum is divided into 'shell' and 'core' subterritories which have different connectional, histochemical and pharmacological properties. Behavioral studies also indicate that subterritories of the ventral striatum are differentially involved in specific goal-directed behaviors. The amygdala is a heterogeneous structure which has multiple nuclei involved in processing emotional information. While the existence of an amygdalostriatal pathway has long been established, the relationship between amygdaloid afferents and specific subterritories of the ventral striatum is not known. In this study we operationally defined the ventromedial striatum as the region receiving cortical inputs primarily from the orbital and medial prefrontal cortex. We placed retrograde tracer injections into subregions of the ventromedial striatum of macaques monkeys to determine the relative contribution of specific amygdaloid inputs to each region. Calbindin-D28k immunostaining was used to further define the shell subterritory of the ventromedial striatum. Based on these definitions, the amygdala innervates the entire ventromedial striatum, and has few to no inputs to the central striatum. The basal and accessory basal nuclei are the major source of input to the ventromedial striatum, innervating both the shell and ventromedial striatum outside the shell. However, a restricted portion of the dorsomedial shell receives few basal nucleus inputs. Afferent inputs from the basal nucleus subdivisions are arranged such that the parvicellular subdivision projects mainly to the ventral shell and core, and the magnocellular subdivision targets the ventral shell and ventromedial putamen. In contrast, the intermediate subdivision of the basal nucleus projects broadly across the ventromedial striatum avoiding only the dorsomedial shell. The shell has a specific set of connections derived from the medial part of the central nucleus and periamygdaloid cortex. Within the shell, the dorsomedial region is distinguished by additional inputs from the medial nucleus. The ventromedial caudate nucleus forms a unique transition zone with the shell, based on inputs from the periamygdaloid cortex. Together, these results indicate that subterritories of the ventromedial striatum are differentially modulated by amygdaloid nuclei which play roles in processing olfactory, visual/gustatory, multimodal sensory, and 'drive'-related stimuli.

Bcl-2, a cell death suppressor protein, is expressed during brain development but is largely down-regulated in the adult central nervous system. We previously reported strong expression of bcl-2 in small, "oligodendrocyte-like" cells (OLC) found in glioneuronal hamartias. These hamartias are microscopic cell rests found in temporal lobe resections from patients with intractable epilepsy and are considered a form of cerebral microdysgenesis. However, a causative relationship between these rests and seizures is not clear. We now report the identification, lineage characterization, and postnatal ontogeny of hamartia-like cell rests in temporal lobes of nonepileptic humans. Postmortem temporal lobes from 28 patients without history of neurologic disease (mean age = 53 years; range = 20 to 83 years) were studied. Microscopic cellular aggregates containing immature-appearing, bcl-2-immunoreactive cells (BIC) (identical to OLC) were observed in 23 of 28 (82%) temporal lobes from nonepileptic individuals. BIC were strongly immunoreactive for neuronal-specific class III beta tubulin, neuronal nuclear antigen, and MAP-2, but were consistently negative for neurofilament proteins and Ki67. Such cells were localized to subventricular regions of the caudal amygdala and often extended into the adjacent subcortical white matter and periamygdaloid cortex. BIC became less abundant with advancing age. These findings suggest that hamartia-like rests containing immature postmitotic neurons are normally present in the human brain and that glioneuronal hamartias may not always represent a maldevelopmental lesion associated with epilepsy.

To investigate possible distinct contributions of different temporal-lobe structures to odour identification, the University of Pennsylvania Smell Identification Test was administered monorhinally to seizure-free patients who had undergone one of three types of temporal-lobe resection practised in three different institutions for surgical treatment of epilepsy. The resections were neocorticectomy (Dublin), selective amygdalohippocampectomy (Zurich), or anterior temporal-lobe resection with encroachment on amygdala and hippocampus (Montreal). Resections, analysed from MRI scans, showed unexpected encroachment on medial structures in most patients of the neocorticectomy groups, and largest amygdala and hippocampal resections in the amygdalohippocampectomy groups. Impaired odour identification was observed in all patient groups, irrespective of surgical approach, with greatest impairment in the nostril ipsilateral to the resection. The finding of deficits in all three surgical groups suggests that damage in the anterior temporal area, perhaps in piriform cortex, is sufficient to disrupt performance on this task; it may be that function is disrupted in the medial temporal-lobe region by disconnection when the periamygdaloid area is damaged, even when amygdala and hippocampus are left intact. An alternative explanation for our results is that damage in any one of these areas disrupts a complex network involving several distinct temporal-lobe structures.

The distribution of calbindin-D28k-immunoreactive cells and fibres in five human amygdalae was analysed from sections that had been stained immunohistochemically with a monoclonal antibody raised against calbindin-D28k. The highest density of calbindin-D28k-positive neurons were found in the anterior cortical, medial, posterior cortical and accessory basal nuclei, in the parvicellular division of the basal nucleus and in the amygdalohippocampal area. The lowest densities of immunopositive neurons were found in the paralaminar nucleus, in the periamygdaloid cortex (PAC1 and PACo) and in some of the intercalated nuclei. The deep nuclei (lateral, basal and accessory basal nuclei) contained a high density of calbindin-D28k-immunoreactive fibres and terminals. The cortical nuclei and the central nucleus were characterized by intense neuropil labelling. Morphologically, a large majority of the calbindin-D28k-immunoreactive neurons were aspiny or sparsely spiny and resembled inhibitory local circuit neurons. A small population of lightly-stained, pyramidal-shaped neurons was also observed. In most of the amygdaloid nuclei, calbindin-D28k-immunoreactive fibres travelled close to each other and formed bundles, which suggests that some of the immunostained neurons were double-bouquet cells. In the paralaminar nucleus, the calbindin-D28k-immunoreactive axons formed tortuous plexus (100–200 mm in diameter) that surrounded several unstained somata. This study provides baseline information on the morphology and distribution of calcium-binding protein-containing inhibitory cells and fibres immunoreactive for calbindin-D28k in the human amygdaloid complex. This information can be used in future studies on the pathogenesis of diseases known to damage the amygdala, such as Alzheimer's disease and temporal lobe epilepsy.

Retinal projections to the basal forebrain in male Syrian hamsters were examined at the ultrastructural level following bilateral intraocular injections of horseradish peroxidase conjugated to either cholera toxin (CT-HRP) or wheat germ agglutinin (WGA-HRP). Light level microscopic analysis confirmed retinal projections along basal telencephalon, and examination on the electron microscope of individual fibers from the peri-amygdaloid area revealed en passant synaptic profiles. Sections from animals treated with WGA-HRP showed evidence of transsynaptic communication in the form of labeled dendrites in the peri-amygdaloid area. Taken together, these data show that the retina communicates directly with the periamygdaloid area, where photic and chemosensory information may be integrated to modulate reproductive behavior.

Previous studies of the intrinsic connections of the amygdaloid complex in the rat, cat and monkey demonstrated that the lateral nucleus projects prominently to the accessory basal nucleus and periamygdaloid cortex and lightly to several other nuclei of the amygdala. Most previous studies have emphasized the lack of a connection between the lateral and basal nuclei. As part of ongoing studies of the intrinsic circuitry of the monkey amygdala, we have placed discrete, iontophoretic injections of the lectin anterograde tracer Phaseolus vulgaris leucoagglutinin into the lateral nucleus of the Macaca fascicularis monkey. In addition to confirming the well established intrinsic connections of the lateral nucleus, heavy fiber and terminal labeling was also observed in the adjacent basal nucleus. The projection innervated all divisions of the basal nucleus and tended to be somewhat denser rostral to the level of the injection site. This hitherto unreported intrinsic connection provides a means by which the lateral nucleus, which is the principal recipient of afferents from sensory neocortex, can influence cells of the basal nucleus, which originate the major amygdalofugal projection to the neocortex.

Using the autoradiographic tracing technique the retinal projections were studied in the tenrecs, Echinops telfairi and Setifer setosus (insectivora, tenrecidae). Bilateral projections were found to the n. suprachiasmaticus, the anterior hypothalamic area, the dorsal and ventral lateral geniculate bodies, the pretectal olivary nucleus and the superior colliculus. The contralateral projections were usually more intense than the ipsilateral ones except the retinohypothalamic connections. A partial segregation of the projection fields from both eyes was present in the dorsal and ventral lateral geniculate bodies. In the superior colliculus retinal fibers predominantly involved the stratum zonale and the upper portion of the stratum griseum superficiale on both sides. The projections to the deeper portion of the colliculi were rather faint, particularly on the ipsilateral side. Target areas receiving contralateral projections exclusively were the periamygdaloid area (labeled only in Setifer), the terminal accessory nuclei including the n. tractus optici and the inferior colliculus. The data are compared with other species. The most striking finding may concern the projection to the medial terminal nucleus being quite prominent in marsupials and most eutherian mammals (including the erinaceomorphous hedgehogs), but greatly reduced in tenrecs and primates.

The distribution of somatostatin-like immunoreactivity was studied in the macaque monkey (Macaca fascicularis) by using primary antisera that recognize somatostatin-28 (S309) or somatostatin-28(1-12) (S320). Somatostatin-immunoreactive neuronal cell bodies were observed in all amygdaloid nuclei and cortical regions. The density of labeled cells varied substantially, however, both within and across the various amygdaloid subdivisions. The highest densities of labeled neurons were observed in layer III of the periamygdaloid cortex, in layers II and III of the medial nucleus, in the magnocellular division of the accessory basal nucleus, and in the medial portion of the lateral nucleus. Many labeled cells were also consistently observed in the caudoventral portion of the lateral division of the central nucleus. Labeled cells were heterogeneous in size and shape ranging from small and spherical to large and multipolar. The density of somatostatin-immunoreactive fibers also varied greatly from region to region and was often inversely related to the density of immunoreactive cells. Highest densities of immunoreactive fibers were observed in the periamygdaloid cortex, medial nucleus, parvicellular division of the accessory basal nucleus, paralaminar nucleus, ventrolateral portion of the lateral nucleus, parvicellular division of the basal nucleus, and the lateral division of the central nucleus. Fibers and terminals in the central nucleus had a coarsely varicose appearance and this pattern of staining was continuous along the trajectory of the central nucleus projection to the bed nucleus of the stria terminalis. The large, immunoreactive varicosities located in this area often appeared to outline dendritic or vascular profiles within the substantia innominata. The lowest levels of somatostatin-immunoreactive fibers were observed in the magnocellular division of the basal nucleus and in the ventromedial portion of the accessory basal nucleus.

Injections of horseradish peroxidase were made into the basolateral nuclei of the amygdaloid complex in cats. It was shown that the periamygdaloid cortex immediately below the rhinal sulcus and extending medially to the amygdaloid fissure projects to the lateral nucleus. The rest of the periamygdaloid cortex medial to the amygdaloid fissure and including the cortical nucleus of the amygdala projects primarily to the basomedial nucleus. These cortico-amygdaloid projections originate in the deeper one-third of the cortex. No projections from the neocortex could be demonstrated.

The corticoamygdaloid and intraamygdaloid projections of the rat were studied by the use of retrograde transport of horseradish peroxidase (HRP). Observations based on anterograde transport of the enzyme were exploited to determine the course of the intrinsic connections. The HRP was injected stereotactically by means of iontophoresis. Most of the amygdaloid nuclei were selectively injected, and all but a few were reached by more than one approach. The vast majority of corticoamygdaloid fibers was found to originate in cortical areas defined as allocortical (Stephan, '75). From the medial frontal cortex the central amygdaloid nucleus (AC) receives a hitherto undescribed projection originating in the tenia tecta; and both the AC and the lateral amygdaloid nucleus (AL) receive fibers from the prelimbic and infralimbic areas. The anterior cingulate area entertains a weak connection with the basolateral amygdaloid nucleus (BL). As to the insular cortex, the posterior agranular insular area projects to all amygdaloid subdivisions; the BL, AC, and the anterior cortical nucleus (COa) receive, in addition, fibers from the ventral agranular area. The prepyriform cortex connects with the entire amygdala except the medial nucleus (Am). The amygdala receives afferents from a transitional area between the amygdala and the entorhinal area. The entorhinal area proper is related to the amygdala via projections from the ventral part of the lateral entorhinal area to the AL and from the dorsal part of the lateral entorhinal area to the BL. The former nucleus also receives fibers from the perirhinal region. Additional amygdalopetal connections from the hippocampal region include a previously undescribed projection from the temporal two-thirds of CA1 to the AL and BL and to the posterior cortical nucleus (COp) with the adjacent periamygdaloid cortex (PAC). The subiculum projections to the AL, and more modestly to other amygdaloid nuclei. There is an extensive network of intraamygdaloid connections, the Am and AC being the only nuclei not giving rise to the intrinsic fibers.

The association and commissural fiber systems arising in the olfactory cortical areas caudal to the olfactory peduncle (the piriform cortex, nucleus of the lateral olfactory tract, anterior cortical nucleus of the amygdala, periamygdaloid cortex and entorhinal cortex) have been studied utilizing horseradish peroxidase as both an anterograde and a retrograde axonal tracer. In the piriform cortex two sublaminae within layer II (IIa and IIb) and layer III have been found to give rise to distinctly different projections. Retrograde cell labeling experiments indicate that the association fiber projection from layer IIb is predominantly caudally directed, while the projection from layer III is predominantly rostrally directed. Cells in layer IIa project heavily to areas both caudal and rostral to the piriform cortex. The commissural fibers from the piriform cortex are largely restricted in their origin to layer IIb of the anterior part of the piriform cortex and in their termination on the contralateral side to the posterior part of the piriform cortex and adjacent olfactory cortical areas. A projection to the olfactory bulb has also been found to arise from cells in layers IIb and III of the ipsilateral piriform cortex, but not in layer IIa. In addition to those from the piriform cortex, association projections have also been found from other olfactory cortical areas. The nucleus of the lateral olfactory tract has a heavy bilateral projection to the medial part of the anterior piriform cortex and the lateral part of the olfactory tubercle (as well as a lighter projection to the olfactory bulb); both the anterior cortical nucleus of the amygdala and the periamygdaloid cortex project ipsilaterally to several olfactory cortical areas. The entorhinal cortex has been found to project to the medial parts of the olfactory tubercle and the olfactory peduncle. The olfactory tubercle is the only olfactory cortical area from which no association fiber systems (instrinsic or extrinsic) have been found to originate. A broad topographic organization exists in the distribution of the fibers from several of the olfactory areas. This is most obvious in the anterior part of the olfactory cortex, in which fibers from the more rostral areas (the anterior olfactory nucleus and the anterior piriform cortex) terminate in regions near the lateral olfactory tract, while those from more caudal areas (the posterior piriform cortex and the entorhinal cortex) terminate in areas further removed, both laterally and medially, from the tract. Projections to olfactory areas from the hypothalamus, thalamus, diagonal band, and biogenic amine cell groups have been briefly described.

The interconnections between the amygdala and the hippocampal formation were investigated in the macaque monkey using anterograde tracers. The hippocampal inputs to the amygdala arose from the subicular and entorhinal cortices and passed through the angular bundle to terminate principally in the medial basal and lateral basal nuclei, with lighter termination in the lateral nucleus, the periamygdaloid cortex, and the cortical-transition area. The majority of these amygdaloid inputs arose from the rostral hippocampal formation although there was equivocal evidence of an additional projection from the caudal hippocampus to the central nucleus. Projections arose from many of the amygdaloid nuclei to terminate in the molecular layer of the amygdalo-hippocampal area and the adjacent stratum moleculare of the uncal portion of the hippocampus. The accessory basal, lateral basal, and medial basal nuclei also projected to the most rostral portions of the stratum moleculare of fields CA1-3, the heaviest termination occurring in field CA3. Additional projections from the basal nuclei terminated in the prosubiculum, presubiculum, and parasubiculum. The heaviest entorhinal inputs arose from the accessory basal and lateral nuclei and terminated in layers I, II, and III of areas 28b, 28i, and the prorhinal cortex. The major amygdaloid input to the perirhinal cortex arose from the lateral basal nucleus.

Projections are described from the basolateral, lateral and anterior cortical nuclei of the amygdaloid complex, and from the prepiriform cortex, to several discrete areas of the cerebral cortex in the rat and cat and to the mediodorsal thalamic nucleus in the rat. These projections are very well-defined in their origin, and in their area of laminar pattern of termination. The basolateral amygdaloid nucleus can be divided into anterior and posterior divisions, based on cytoarchitectonic and connectional distinctions. In both the rat and cat the posterior division projects to the prelimbic area (area 32) and the infralimbic area (area 25) on the medical surface of the hemisphere. The anterior division projects more lightly to these areas, but also sends fibers to the dorsal and posterior agranular insular areas and the perirhinal area on the lateral surface. Furthermore, in the cat the perirhinal area is divided into two areas (areas 35 and 36) and the anterior division projects to both of these and also to a ventral part of the granular insular area; this last area is adjacent to, but separate from the auditory insular area and the second cortical taste area. In most of these areas, the fibers from the basolateral nucleus terminate predominantly in two bands: one in the deep part of layer I and layer II, and a heavier band in layer V (in the rat) or layers V and VI (in the cat). The lateral amygdaloid nucleus projects heavily to the perirhinal area, and also to the posterior agranular insular area. These fibers terminate predominantly in the middle layers of the cortex, although the cellular lamination in these two areas is relatively indistinct. The anterior cortical amygdaloid nucleus and the prepiriform cortex both project to the infralimbic area and the ventral agranular insular area, and the anterior cortical nucleus also projects to the posterior agranular area and the perirhinal area. In all of these areas, the fibers from these olfactory-related structures terminate in the middle of layer I. In the rat, the two divisions of the basolateral nucleus also project to the medial segment of the mediodorsal thalamic nucleus, with the anterior division projecting mainly to the posterior part of this segment and the posterior division to the anterior part. The endopiriform nucleus, deep to the prepiriform cortex, projects to the central segment of the mediodorsal nucleus; this may constitute the major olfactory input into the mediodorsal nucleus, since little or no projection could be demonstrated from the prepiriform cortex itself. Projections to the mediodorsal nucleus have not been found in the cat.

It is widely believed that amygdaloid lesions induce hypersexuality in certain mammals including humans. However, the evidence is so confused as to obscure the real nature of the purported changes. Twelve sexually experienced male cats were given two series of quantitative tests, the conventional sex test and a new selection test in which the animals were presented sequentially with a small stuffed toy panda, moving toy, tranquilized rabbit, tranquilized male and estrous female. Lesions in 8 experimental males were largely confined to the basolateral nuclei and periamygdaloid cortex. In the standard test and in the response to the female in the selection tests, the only significant postoperative change in the major measurements of sexual behavior was a decline in initial mount latency. However, in the selection tests, the major change was an increased tendency to mount less appropriate objects. There was no evidence of hypersexuality, in the sense of a higher level of sexual performance. This change was not interpreted as the disruption of an amygdaloid mechanism directly regulating sexual behavior, but rather as interference with a system which normally modulates approach and withdrawal behavior in response to stimuli that are salient with respect to an existing behavioral setting and set.

The connections of the amygdala with the insular and temporal cortices were examined by injecting wheat germ agglutinin conjugated to HRP (WGA-HRP) into the rat cortex. Following injections into the posterior agranular insular area (AIp) or perirhinal cortex (PR), bands of labeled neurons extending across nuclear boundaries were observed in the amygdala. These neuronal bands involved cells in the lateral, basolateral, and basomedial nuclei as well as the periamygdaloid cortex. Other nuclei of the corticomedial amygdala and the ventral endopiriform nucleus also exhibited retrogradely labeled cells. Anterograde label was observed in nuclei containing labeled neurons and in the central nucleus. Injections into gustatory, somatosensory, and auditory neocortical areas located dorsal to AIp and PR labeled small numbers of cells in the lateral and basolateral nuclei. Injections into AIp, PR, and, to a lesser extent, dorsally adjacent neocortical areas produced both retrograde and anterograde labeling in the contralateral amygdala. The main nuclei with contralateral insular and temporal projections are the basomedial nucleus, ventral endopiriform nucleus, and nucleus of the lateral olfactory tract. The contralateral central nucleus and to a lesser extent the lateral nucleus exhibited anterograde labeling. The pattern of retrograde labeling seen with injections at different rostrocaudal levels of the AIp-PR continuum indicates that amygdalocortical projections to these areas exhibit an overlapping topographical organization. Comparison of the results of this study with findings on amygdaloprefrontal cortical efferents suggests that amygdaloid projections to the entire fronto-insulo-temporal mesocortical field are topographically organized.

Summary Through stimulation of the lateral amygdaloid nucleus (M of Volsch) we could produce remarkable changes upon respiratory movements, namely, diminution of amplitude and increase in the rate of respiration. The stimulation of the intermediate or medial nucleus of amygdala proper (T or T of Volsch, basal amygdaloid nucleus of Fox) may not bring about changes upon the respiratory movements; if effective, it is typified in the increase in the rate and amplitude of respiration, as it was previously recognized through stimulation of the periamygdaloid cortex. The motion of the diaphragm stands still at the moment of stimulation, regardless as to whether it stands in expiratory or in inspiratory position. The histological observation of the lungs revealed that changes such as hemorrhage, congestion, emphysematous and edematous change, were ext: aordinarily severe in the stimulated side, except in a few instances where much severe damage was shown in the contralateral side.

Summary Through bilateral destruction of the amygdaloid nuclear complex in infant and adult animals and through subsequent observations under careful feeding in sxne period of time, following results were obtained: 1) By bilateral amygdalectomy in infant animals, inhibition in the growth was induced in most cases. 2) Histological observations on the brain lesions of these cases reveal that the inhibitory effect upon the growth may be initiated seemingly by destruction of the medial amygdaloid complex such as nuclei B, D or T, or periamygdaloid cortex such as Pam2 and Pam3, or the anterior amygdaloid region, especially at the level of the nucleus of the lateral olfactory trac:. 3) The inhibitory effect upon the growth was exceedingly obvious in those cases, in which the amygdaloid regions or surrounding structures of these most effective parts of the amygdala were extensively damaged on both sides. 4) Somewhat accelerating effect upon the growth was found in some cases, however, no positive evidence could be gained by any means in this series of observation. 5) Unilateral arnygdalar or hypothalamic destruction cannot result in retardation of the growth, even in those highly convincing cases with tissie deterioration in the vicinity of the amygdala. 6) An example of remarkable obesity was found in an adult cat following bilateral destruction of the medial parts of the amygdala and a part of the entorhinal area. Histological alterations of the internal organs of this case were described and a functional relationship of the so-called feeding centre in the hypothalamus was discussed.

Summary The results of comparative-anatomical and experimental studies on amygdala disclosed the following facts: 1) The division of amygdaloid niiclei into two groups, basolateral and corticomedial seems to be not fully adequate in regard to the functional differences, because results of the stimulation experiments in our laboratory indicate for certain coincidence of effectiveness with autonomous function. From the view angle of fiber connection, this way of division ought also not to be highly evaluated. 2) The intermediate principal nucleus (nucleus T of Volsch large-celled lateral part of the basal nucleus of American authors) is generally divided into two parts, namely, dorsal and ventral portions. The ventral portion of that of the bat is particularly large and together with the large lateral principal nucleus nucleus M of Volsch, lateral nucleus of American authors) constitutes a characteristic of bat amygdala. The reason for its largeness was found, by means of destruction experiment of the amygdaln, in a close connection with the flight activity. Presumably intermediate prinripal nucleus participates in the extrapyramidal motor function; the lateral principal nucleus is generally large in most animals and functionally seems to inhibit motor activity. 3) The medial principal nucleus (nucleus T of Volsch, small-celled medial part of the basal nucleus of American authors) remains maldeveloped in some lower animals. In general, it grows distinctly larger in the caudal level. According to the result of the investigations undertaken in our laboratory, the said nucleus is chiefly related to autonomic nervous system. The above mentioned lateral and intermediate principal nuclei (T and T) also seem to have some relation, partially at least with autonomic nervous system. 4) The so-called medial nucleus in higher animals (D of Volsch) is generally maldeveloped. This nucleus, together with cortical nucleus (B of Volsch), participates partially also in autonomic nerve function. The central nuclei of higher animals (E and E of Volsch) are also underdeveloped 5) The periamygdaloid cortex (Pam1, Pam2 and Pam3 of Rose) receives the fibers from the lateral olfactory tract and evidently participates in olfactory function, but it also causes both extrapyramidal and autonomic reflexes probably by way of the amygdalar connection. Of the said cortical portions what are particularly closely related with amygdala are the portions Pam2 and Pam3. However, the portion Pam3 has not been differentiated yet in the lower animals. 6) When the bilateral amygdala of a young animal is destroyed early in life, growth gets markedly inhibited. In such an animal. atrophy of the parotid gland, thyroid gland, hypophysis, adrenal gland and pancreatic gland was observed. As the cause of this developmental disturbance there are two nervous routes assumable, namely, the route through amygdala hypothalamus anterior lobe of hypophysis general inner secretory glands and the route through amygdala salivary glands inner secretory disturbance of the latter. In the latter connection, the pituitary gland also may participate in the process. We should be particularly cautious about applying bilateral amygdalectomy to human subjects as there is a possibility of the occurrence of disturbances long after the operation. 7) That amygdala participates in salivation was verified by our experiments and the following facts also came to be clear as the result thereof: the portion Pam2 of periamygdaloid cortex has a particularly close connection with the secreting function of parasympathetic origin; of all the amygdaloid nuclei, the medial principal nucleus (T) has an especially close relation with saliva secretion of sympathetic origin, while the intermediate principal nucleus (T), cortical nucleus (B), medial nucleus (D) and hippocampus secret saliva of a parasympathetic origin. Unilateral stimulation produces secretion of bilateral salivary glands. The fibers starting from such portions seem to be led directly toward hypothalamus further to reach the lower part of the brain.

Characteristic bulbar potentials were evoked by retrograde stimulation of the lateral olfactory tract. Its projection was thereby traced into the prepiriform areas, the lateral part of the olfactory tubercle, the periamygdaloid area, cortical amygdaloid nucleus, anterior part of the medial nucleus, and throughout the parahippocampal area. This latter connection has not been described previously. By our definition the parahippocampal area lies immediately behind the periamygdaloid area and extends caudally alongside the posterior prepiriform area, being bounded caudomedially by the true entorhinal area which thus occupies only the most posterior part of the piriform lobe. Negative bulbar potentials were evoked when centrifugal systems were stimulated, crossed potentials being mediated by the anterior commissure. However, the commissure is not the only efferent pathway to the ipsilateral bulb. Centrifugal systems were demonstrated in relation to all regions receiving lateral olfactory tract afferents. Stimulation of these areas, both superficially and deeper in the cortex, evoked bulbar negativities even after interruption of the ipsilateral anterior limb of the anterior commissure, or section of the lateral olfactory tract in the peduncle. The close association of centripetal and centrifugal systems suggests that bulbar activity is under the influence of sensory feedback systems, as well as more central control.

The amygdaloid complex in the cat was studied in a series of Golgi preparations. Both the lateral and the basal nucleus are composed of the same two cell types, one of which (type P) resembles the pyramidal and the other (type S) the stellate neuron of the cortex. The cortical nucleus can be divided into three layers (I, II, and III–IV) which are made up of cells similar to those in the periamygdaloid cortex. In addition, there are sufficient differences in the organization of these layers to justify a subdivision of the cortical nucleus into lateral and medial parts. The dendrites of neurons in the medial part of the central nucleus, the medial nucleus and the anterior amygdaloid area undergo less branching and carry fewer spines than those of the type P cell. The neurons in the nucleus of the lateral olfactory tract are all of the pyramidal or modified pyramidal type. These findings are discussed in relation to those of previous investigators who employed the Nissl and Golgi methods.
