It has earlier been reported that the human monoclonal antibody (MoAb 33G2) and polyclonal antibodies reactive with Pf332 may interfere in vitro with the erythrocytic cycle of Plasmodium falciparum at two potential target sites for protective antibodies, indicating that the antigen may constitute an important target for immune responses during malaria infections. MoAb 33G2 shows its highest reactivity with repeated sequences in the antigen Pr332 and also cross-reacts with determinants in Pf155/RESA. This study was conducted in order to assess the prevalence of seroreactivity against Pf332 in individuals residing in areas of different malaria endemicity, and in children with different degrees of disease severity. We now report that individuals resident in malaria endemic regions show a high prevalence of seroreactivity to antigen Pf332 repeat sequences. The mean antibody concentrations were significantly higher in donors from Liberia. Madagascar and Gambia compared with Thai and Colombian donors, probably reflecting the higher degree of exposure in the African regions. Although the levels of such antibodies in individual sera correlated well with the levels of antibodies to one Pf155/RESA repeat peptide, only a minor part of the peptidereactive antibodies were cross-reactive between the two antigens. In Gambian children, the mean concentrations of antibodies reactive with PD32 or Pf 155/RESA peptides were significantly higher in children with severe than with mild malaria. Normal IgG preparations of human, rabbit or guinea-pig origin (IgG2) were tested for their capacity to inhibit the cytotoxicity of purified human lymphocytes, as induced by rabbit IgG antibodies to chicken erythrocytes. All IgGs were found to be about equally efficient inhibitors. Human F(ab′)2 used for control, gave no inhibition. Human myeloma proteins of subclasses IgG1, IgG2 and IgG3, were about equally efficient inhibitors. In contrast, the inhibitory action of myeloma proteins belonging to subclass IgG4 was weak and more irregular. In this assay system, a large excess (∼ 106 × ) of normal IgG over antibodies had to be added in order to achieve ≥ 50 per cent inhibition. Heating of the inhibitors to 63° for 30 minutes did not significantly enhance their inhibitory capacity. For comparison, the same human IgG preparations and myeloma proteins were also tested for their capacity to inhibit phagocytosis by human blood monocytes of chicken erythrocytes sensitized with rabbit IgG antibody. Rat liver microsomes were separated into three subfractions by means of ultracentrifugation through sucrose media of different densities and in the presence of different cations. Detergent extracts of these preparations were analysed by double diffusion in agar gel and immunoelectrophoresis with rabbit antisera against each of the subfractions. One subfraction was derived from the ‘rough’ and another from the ‘smooth’ part of the endoplasmic reticulum of the liver parenchymal cells. Their extracts contained at least thirteen soluble antigens in common. However, fraction-specific antigens were also present. Thus, the extracts of the rough (R) membranes contained at least one typical antigen, not found in any of the other fractions. An antiserum against this component also precipitated a non-migrating antigen present only in the extracts of smooth (Sa) membranes. These two antigens may represent different molecular forms of a substance involved in binding of the ribosomes or in the assembly of membrane subunits. At least eleven of the antigens common for the two fractions exhibited enzymatic activities when assayed after precipitation with antibody in the immunoelectro- phoretic plates. Six immunologically and electrophoretically distinct antigens had esterase activity with α-naphthyl propionate as substrate. Two of these esterases also split indoxyl acetate. Three other antigens with acid phosphatase activity split both α-naphthyl acid phosphate and β-glycerophosphate. Three antigens had nucleoside diphosphatase activity when tested with uridine- or inosine-diphosphate. Preliminary experiments also suggested that two additional antigens possessed NADH-diaphorase activity and thus could belong to the microsomal electron trans- port systems. The third subfraction consisted of electron-microscopically smooth membranes (Sb), enzymatically different from those of the endoplasmic reticulum. The antigens typical for the latter were either absent or present only in minor and variable concentrations. Its extracts contained at least two typical antigens. One of these was identified as contaminating ferritin. The nature and origin of the other antigen has not yet been established. The cytotoxic effect of normal human blood lymphocytes on Chang cells in tissue culture was investigated. Cell damage was measured by release of 51Cr from pre-labelled tissue culture ‘target’ cells. This method was sensitive and rendered highly reproducible results. Released 51Cr was not re-utilized. About 25 per cent of the 51Cr was spontaneously released from labelled Chang cells when incubated for 24 hours at 37°. Lymphocytes at a lymphocyte/Chang cell ratio of 25:1 led to a slight increase of this release. When phytohaemagglutinin was also present, about 50—60 per cent of the isotope appeared in the medium. Under these conditions target cells were significantly damaged -within 1 hour. At a lymphocyte/Chang cell ratio of only 1:1, weak cytotoxic effects were also noted after 24 hours of incubation. The results of dose—response experiments suggested that a considerable proportion of the lymphocytes participated in the reaction. Individual variation of the cytotoxic effect of lymphocytes from different donors suggested that it could be related to the degree of histoincompatibility between lymphocytes and Chang cells. Under the present conditions contaminating erythrocytes or granulocytes did not interfere with the cytotoxic action of the lymphocytes.

Ten out of sixty-three sera from patients with ulcerative colitis gave positive staining of goblet cells in the indirect immunofluorescence test, applied to acetone-ethanol fixed sections of colon from germ-free rats. Nine of these sera were also positive in triple layer tests in which the second layer consisted of rabbit antibodies to human immunoglobulin chains and the third layer of fluorescein conjugated sheep anti-rabbit immunoglobulin. All sera contained anti-colon antibodies having light chains of both κ- and &lamda;-type. Heavy chains of both γ-, μ- and α-type occurred at almost equal frequency and only three of the nine positive sera contained antibodies of a single immunoglobulin class. One out of four tested sera which contained antibody of the IgA-class also gave positive reactions for secretory-piece determinants. The staining pattern obtained with antibodies of different classes was similar, Anti-colon antibodies belonging to the IgD or IgE classes were not detected. Cryostat sections of colon from germ-free rats were incubated with sera from patients with ulcerative colitis or healthy controls. With peroxidase-conjugated antibody from sheep to human immunoglobulin, a distinct staining of goblet cell surface was observed with the patients' sera but not with the controls. The staining compares well with that obtained by indirect immunofluorescence. It could easily be distinguished from staining due to endogenous peroxidase. [ABSTRACT FROM AUTHOR]

Repeat sequences from the Plasmodium falciparum blood stage antigen Pf332 frequently comprise the pentapeptide VTEEI, an epitope recognized by certain parasite neutralizing antibodies. This B-cell epitope was assembled in an octavalent multiple antigen peptide (MAP) system either as trimers (VTEEI)3 (MAP1) or as an integral part of a naturally occurring Pf332 undecamer repeat sequence SVTEEIAEEDK (MAP2). Characteristics of the immunogenicity of these subunit constructs were evaluated in H-2 congenic mice. MAP1 generated antibody responses in mice of the H-2d, H-2k and H-2q haplotypes, but not in H-2b or H-2s mice, whereas MAP2 only induced antibodies in mice of H-2k haplotype. When analysing T-cell responses induced by the MAP, lymph node cells from responder strains primed in vivo with MAPI proliferated in response to restimulation with both MAPI and the peptide (VTEEI)3. MAP2, however, did not induce a detectable T-cell proliferation. Additionally, the lack of antibody response to MAP1 in H-2b mice could be circumvented by combining the MAP1 peptide and a H-2b-restricted T-cell epitope in a diepitope MAP construct. Despite the fact that the motif VTEEI has not been identified in Pf332 sequences in the form of a trimer, MAP1 did induce Pf332 protein-reactive antibodies. Antigenic diversity in held populations of Plasmodium falciparum parasites may delay the acquisition of protective immunity to malaria, the development of which may thus require repeated exposure to infection over a prolonged period of time. In this study we show that P. falciparum parasites may vary in their sensitivity to antibody-mediated invasion/growth inhibition in vitro. Wild isolates of P. falciparum from children living in an endemic area of Burkina Faso were tested for their sensitivity to the growth inhibitory effects of antibodies originating from the same (autologous) and front other donors (heterologous). A significantly lower invasion inhibition activity was obtained when the isolates and antibodies were tested in autologous compared with heterologous combinations. The lower sensitivity to growth inhibition by autologous antibodies may be due to immune pressure in vivo, selecting from a heterogeneous parasite population those with a low expression of the antigens recognized by the host's antibodies. Alternatively, the parasites cultured from each child might represent expanding parasite populations. mainly constituting strains not earlier seen by the immune system of that specific host. Sera from patients with Plasmodium falciparum, P. vivax or P. ovale malaria were selected according to their high levels of antibodies against human erythrocyle membranes as measured in a microELISA. The specificity of the anti-erythrocyte antibodies in these sera and two normal sera was investigated by means of an immunoblotting technique in combination with SDS-polyacrylamide gel electrophoresis. All the patients' sera as well as the control sera contained antibodies against several erythrocyte polypeptides. As compared with normal sera, most malaria sera showed elevated levels of antibodies against polypeptides of 80K, 70K, 40K and 28K molecular weights. Two sera reacted strongly against a polypeptide with an electrophoretic mobility similar to the α subunit of spectrin. One serum showed strong reaction and several other sera, including normal sera, showed weak reaction against a 45K molecular weight polypeptide corresponding to actin. No pervading differences were seen in the pattern of specificities of the anti-erythrocyte ghost antibodies between sera from patients with P. falciparum, P. vivax or P. ovale infections. An enzyme linked immunosorbent assay (ELISA) has been developed to estimate disease related antibodies in sera from malaria patients or individuals living in malaria endemic areas. As antigen, Percoll enriched fractions (mainly late trophozoites, schizonts) from Plasmodium falciparum in vitro cultures were used. An ELISA with ghosts from normal human red blood cells (RBC) was performed in parallel. One hundred and seventy-five sera were tested for their reactivity with either one of the two antigens. Seven sera from patients with acute P.falciparum infection were negative. Most of these had been taken very early in infection and consecutive samples taken later usually were positive. The antibodies reacting with the P. falciparum antigen had a high parasite specificity as indicated by inhibition and absorption experiments. Many sera also had elevated levels of antibodies specific for RBC antigens. The IgG subclass levels of anti-Plasmodium falciparum antibodies in human sera were determined in ELISA with monoclonal mouse antibodies specific for the human IgG subclasses as analytical reagents. The parasite antigen was a trophozoite/schizont enriched preparation of in vitro cultivated P. falciparum. Serum samples were from Swedish malaria patients and adult Liberians. Parasite specific antibodies were found in all four subclasses. Relatively elevated levels of IgGl antibodies were found both in Swedish patients and in the Liberians. Relative to the Liberians, high IgG2 antibody levels were seen in most Swedish patients. In Liberian sera but not in those from Swedish patients elevated IgG3 levels were found. In two Swedish patients followed consecutively for a period of 15 weeks, elevated levels of IgG1 and IgG3 antibodies were seen after relapse. No differences between the groups in regard to the levels origG4 antibodies were noticed. Blood lymphocytes from thirteen patients with CLL were studied for surface-bound Ig (SIg), Fc receptors (EA rosettes), receptors for sheep erythrocytes (E rosettes) and receptors for Helix pomatia A haemagglutinin (FTP), a carbohydrate-binding protein with specificity for N-acetyl-D- galactosamine and related sugars. Fluorescein-labelled HP binds to subpopulations of human peripheral blood lymphocytes (PBL) treated with neuraminidase. In normal peripheral blood, HP binds to the T-lymphocytes while the majority of the B cells bearing surface-bound immunoglobulin do not have receptors for HP. In untreated CLL, HP binds to 90-100% of the neur-aminidase-treated PBL. Almost all of the SIg-positive cells in CLL patients also have receptors for HP. Two groups of patients were found: in one the total fraction of SIg+ cells was ⩽50% and about 30% of these lost their Ig during incubation at 37° C. No such loss of Slig was revealed in the remaining patients where total SIg+fraction was ≈70%. These patients usually had higher blood lymphocyte counts, probably reflecting a more advanced disease. CLL patients in remission with low numbers of leukaemic cells also had low numbers of blood lymphocytes carrying both SIg and HP-receptors. Human lymphocytes, activated by PHA or PPD, damage homologous cells in tissue culture, as shown by release of radioactivity from target cells labelled with 51Cr-chromate. Cytotoxic effects of supernatants from activated lymphocytes are difficult to demonstrate by this method. However, such supernatants inhibit the incorporation of radioactive amino acids into homologous and heterologous target cells. In this investigation the cytopathic effects of supernatants from human lymphocytes, stimulated with PHA or PPD, and that of the stimulated lymphocytes themselves were quantitatively assessed by four different methods. Established cell lines of human (Chang and HeLa cells) or mouse origin (L-cells) were used as target cells. Cytotoxic effects of supernatants were demonstrated in a micro modification of the cell counting assay. Supernatants also inhibited incorporation of 14C-leucine or 14C-thymidine into target cells. However, when assayed by release of 51Cr, this cytotoxic effect was usually weak or absent. In contrast, stimulated lymphocytes showed strong cytotoxicity in the chromium assay. The results suggest that stimulated human lymphocytes release factors which are not primarily cytolytic but which inhibit target cell metabolism and proliferation, eventually leading to cell death. The occurrence of immunoglobulin determinants on the surface of lymphocytes from human blood was assessed by indirect immunofluorescent staining of living cells after cultivation with phytohaemaggtutinin or other stimulants. While antisera to γ, μ or α-determinants only stained a few cells, antisera to light chain determinants stained a larger proportion of the cells. Positive staining was recognized as 'ring' staining comprising smaller or larger parts of the cell surface. The specificity of staining was ascertained by several types of controls. After 48 hours of cultivation, anti-κ serum, applied at dilutions of 1:10-1:16 stained about 35-50 per cent and anti-λ serum about 15-20 per cent of the cells in the PHA cultures but only 3-5 per cent in the cultures incubated without PHA. When the antisera were applied at higher concentrations, positive light chain staining was also seen in the unstimulated cultures. At the highest concentrations, which could be used without increasing the non-specific background, the maximum number of κ-positive cells in the unstimulated cultures was approximately 25 per cent. Antiserum titrations showed that about 5-10 times less antiserum was needed to stain the optimal fraction of PHA treated cells. No increased staining of heavy chain determinants was achieved by increasing antiserum concentrations under the present conditions. Similar results were obtained with lymphocytes stimulated by 3 days of incubation with concanavalin A, or by 6-7 days of incubation under mixed culture conditions. Lymphocytes of a tuberculin positive donor also gave increased staining for light chain determinants after incubation for 6-7 days with antigen (PPD). The results indicate that lymphocyte stimulation is accompanied by increased amounts of surface bound immunoglobulins, At the present stage of knowledge, several explanations may account for the fact that light chain determinants are primarily accessible for staining. The above results were obtained under conditions in which no protein was present in the washings performed during processing for immunofluorescence. In the presence of low concentrations of protein more than 60 per cent of both unstimulated andstimulated cells stained for light chain determinants, while staining for heavy chain determinants remained unchanged and at a low level. Iron deficiency has been reported to affect both malaria pathogenesis and cell-mediated immune responses; however, it is unclear whether the protection afforded by iron deficiency is mediated through direct effects on the parasite, through immune effector functions or through both. We have determined cytokine mRNA expression levels in 59 children living in a malaria endemic area on the coast of Kenya who we selected on the basis of their biochemical iron status. Real-time quantitative reverse transcriptase polymerase chain reaction analysis of cytokine mRNA levels of peripheral blood mononuclear cells (PBMC) obtained from these children showed an association between interleukin-4 (IL-4) mRNA levels and all the biochemical indices of iron that we measured. Furthermore, IL-10 mRNA was higher in parasite blood smear-positive children than in blood smear-negative children irrespective of their iron status. Human basophils are potent producers of IL-4 following cross-linking of the high affinity receptor for IgE (FcϵR1). Elevated levels of both total- and malaria-specific IgE have been demonstrated in sera from people living in malaria-endemic regions. Whether or not these IgE antibodies are pathogenic is unclear. Serum containing high IgE levels obtained from malaria individuals was used to establish whether IgE-immune complexes could induce IL-4 production in human basophils. The basophils, obtained from healthy donors, were primed with 10 ng/ml of IL-3 before being transferred to wells containing goat anti-human IgE or human antimalarial IgE-immune complexes. IL-4 was induced upon stimulation of human basophils by plate bound IgE-containing immune complexes. Basophils treated similarly but with goat anti-IgG/human antimalarial- IgG-immune complexes did not secrete IL-4. Similarly mononuclear cells depleted of basophils in parallel culture did not secrete IL-4. Thus, human basophils may contribute to the polarization of T-helper type 2 in the (Th2) responses in malaria hosts via IgE-induced IL-4 production. [Copyright &y& Elsevier]

This paper investigates major histocompatibility complex (MHC) regulation of the class of the immune response given in vitro and in vivo following immunization of the congenic BALB/k (H-2k) and BALB/c (H-2d) mice with the hapten trinitrophenyl (TNP). TNP-immune lymph node cells from BALB/k mice produced high levels of interferon-γ (IFN-γ), interleukin-5 (IL-5) and IL-2 when stimulated with TNP-antigen-presenting cells (APC) in vitro, while TNP-immune lymph node cells from BALB/c mice produced very low levels of these cytokines. No significant difference was found in antigen-specific production of IL-3, IL-4 and tumour necrosis factor-α (TNF-α). There was a strong correlation between the pattern of cytokine production in vitro and the secondary antibody production in vivo. Sera from BALB/k mice had anti-TNP IgG2a, IgG2b and IgG3 levels threefold greater, and anti-TNP IgA levels eightfold greater, than BALB/c mice. The level of specific IgG1 and IgE was only marginally raised in BALB/k mice. In contrast to these strain differences in cytokine and antibody production, there was no difference in two measures of cellular immunity: contact sensitivity in vivo and antigen-specific lymphocyte response in vitro. Our results suggest that there is a good correlation between the production of cytokines in vitro and antibody response in vivo, but not with measures of cellular immunity. People living in Plasmodium falciparum-endemic areas frequently have elevated levels of total as well as P. falciparum-specific serum IgE. This study aimed at investigating whether the elevated serum IgE levels reflect a shift in the balance between CD4+ T helper 1 (Th1) and T helper 2 (Th2) cells in individuals naturally exposed to the P. falciparum parasite. To investigate the role of Thi and Th2 ceils in the human P. falciparum system we used the ELISPOT assay to determine the ratio of IEN-γ- and IL-4-producing cells after specific antigen or mitogen activation in vitro. The donors were individuals who had acquired immunity through natural exposure to the parasite. In response to the specific malaria antigens, very few IL-γ-producing cells were seen. However, in the response of individual donors to the polyclonal T cell activator, leucoagglutinin (La), the anti-malarial IgE levels in plasma were correlated with an increased ratio of IL-4/IFN-γ producing cells. Thus, donors with ratios of IL-4/IFN-γ>1 exhibited mean plasma anti-malarial IgE levels significantly greater than those with ratios <1. In individuals not living in P. fatciparum-endemic areas the ratio of IL-4/IFN-γ was always <1. Taken together, our data suggest a shift in the balance between Th1 and Th2 cells in naturally P. falciparum primed individuals, associated with elevated anti-P. fatciparum plasma IgE levels. T cells from patients with acute Plasmodium falciparum malaria were investigated for induction of immunoglobulin- or anti-malaria antibody secretion in vitro. Stimulation of autologous T/B cell mixtures (2T: IB) with low concentrations of P. falciparum antigen and cultured for 12 days gave rise to a T-dependent IgG secretion which was significantly elevated over that in medium controls. This was achieved with both a crude P. falciparum antigen and a partially purified preparation enriched in Pf 155. a merozoite-derived antigen deposited in the red cell membrane at invasion (Perlmann et al. 1984), Control antigen (RBC ghosts) induced IgG secretion only when added at high concentrations (>10 μg/ml). Neither of the antigens induced IgG secretion at concentrations of ≪ 10μg/ ml in control cultures of lymphocytes from patients with P. vita.\ malaria. Supernatants from cultures of P.falciparum patients frequently contained anti-P. falciparum antibodies when nanogram quantities (10-100 ng/ml) of either one of the two malaria antigen preparations was used for stimulation. No anti-P. falciparum antibodies were induced by the control antigen at any concentration. The induced anti-P. falciparum antibodies were directed to intracellular parasites and, at lower frequencies. to Pf 155 as detected on the surface of infected erythrocytes. The induction in vitro of anti-P. falciparum antibodies appeared to be correlated with the presence of such antibodies in the sera of the lymphocyte donors. The lymphocytes of only one out of eight P. vivax patients responded to antigen stimulation by secreting anti-P. falciparum antibodies. However, this donor (but not any of the others), was also P. falciparum seropositive. Taken together, these results indicate that the induction of anti-P. falciparum antibody secretion reflects a secondary response in vitro of cells primed in vivo. Isotypes (IgM and IgG-1 to 4) of anti-P. falciparum antibodies were investigated in sera of malarial patients or immune donors by enzyme linked immunosorbent assay (ELISA) and two indirect immunofluorescence assays (IFAs), one staining intra-erythrocytic parasites of all stages and the other a restricted number of parasite antigens deposited in the membrane of infected erythrocytes by invading merozoitcs (Perlmann & Wahlgren, 1983; Perlmann et al., 1984, Wahlgren et al., 1985 a). There was no correlation in overall antibody titre between the two IKAs. Antibodies of both IgM and all tour IgG isotypes were detected in both assays. With the IFA for intracellular parasites a brilliant fluorescence was obtained with antibodies of all isotypes. However IgG-2 antibodies often gave staining restricted to the surface of schizonts. The incidence and reactivity in individual sera of antibodies of the different isotypes did not relate to the immune status of the donors (acute infection or clinically immune) but related well to the degree of malarial exposure as reflected by the overall antibody titres. This, in all three assays, high titred sera frequently contained antibodies of all isotypes white low titred sera usually only contained antibodies of IgM, IgG-1 and IgG-3 isotype. On average, the overall expression of antibodies of different isotypes in individual sera appears to reflect a sequential downstream (5' to 3') activation of the corresponding Igh-C genes in P. falciparum specific B cell clones. The antigen-induced DNA synthesis in vitro in lymphocytes from patients with acute Plasmodium falciparum malaria was investigated. The patients and healthy controls from Sweden or Colombia were the same as those studied in the accompanying paper (Troye-Blomberg el al., 1983). The malarial antigens used were sonicated membrane preparations or purified and concentrated supernatants from in vitro cultures of P. facliparum, similar preparations derived from normal human erythrocytes served as control antigen. In the patients' lymphocytes P. falciparum antigens induced a weak or moderate but significant stimulation of DNA synthesis, peaking after 3-4 days of incubation. This early response was specific for P. faleiparum since it was not obtained with lymphocytes from healthy donors nor with those from patients with acute P. vivax or P. ovale malaria. No antigen-induced response was seen in about half of the P.falciparum patients. However in a few negative eases, available for consecutive testing, positive reactions were seen with lymphocytes taken 2 weeks after infection when the blood of these patients was free of parasites. The early response induced in patients' lymphocytes to the P. falciparum antigens was not obtained with RBC antigen, however, these preparations frequently induced a response rising to significant levels later during incubation (day 5-6)- Similar delayed responses were obtained when either- patients' or control donors' lymphocytes were exposed to the P. falciparum antigens. This indicates that both the RBC and the parasite preparations contained mitogenic substances affecting human lymphocytes in general and easily obscuring the P, falciparum specific responses seen only in the patients. This latter response was relatively low and short lived, suggesting that it reflected a secondary in vitro stimulation of in vivo primed lymphocytes and that it was regulated by suppressor mechanisms. The mitogen-induced DNA synthesis in vitro in lymphocytes from 20 patients acutely ill with Plasmodium falciparum malaria was compared with that of 16 healthy donors. Within both groups part of the donors were individuals who had only experienced short exposure or none at all to the parasite (Sweden) while the other part were donors living in a malaria endemic area (Colombia), The proliferative response to the T cell mitogen La (leucoagglutinin from PHA) of the patients was significantly reduced as compared with that of the controls. With pokeweed mitogen which stimulates T cells and induces a T cell-dependent activation of B cells, no difference between patients or controls was seen. The results were similar for the donors of different geographical origin and malaria background. Lymphocytes and monocytes from the peripheral blood of these donors were also studied for surface marker distribution by means of monoclonal antibodies. Both the absolute and the relative frequencies of T cells in the blood of the malaria patients were significantly reduced as compared with the controls. Furthermore, in almost all eight patients tested, the ratio between T4+ T cells (including the helper/inducer subsets) and T8+ T cells (including the suppressor and cytotoxic subsets) were below 1: 1 while they were close to 2:1 in the controls. The results indicate that the relative frequency of T8+ T cells, expressed as percentage total T cells (T.3+ ) was significantly elevated in the P. falciparum patients. The cytotoxic functions of highly purified blood lymphocytes from patients with breast cancer were studied before and after radiotherapy. Addition of PHA or of rabbit antibodies to target cells (chicken erythrocytes) were chosen as two means of inducing lymphocyte cytotoxicity in vitro. The proportion of T and non-T lymphocytes was determined by means of E and EAC rosette tests. The antibody-induced cytotoxicity of lymphocytes decreased following radiotherapy while that mediated by PHA remained unchanged. There was some reduction in the percentage of EAC rosette-forming cells. These results, as well as our earlier observations, suggest that the decrease in the peripheral blood of the proportion of lymphocytes with receptors for activated complement is responsible for changes in the antibody-mediated lymphocyte cytotoxicity.
