Infection with Fasciola hepatica (liver fluke) causes fasciolosis (or fascioliasis) and poses a considerable economic as well as welfare burden to both the agricultural and animal health sectors. Here, we explore the ex vivo anthelmintic potential of synthetic derivatives of hederagenin, isolated in bulk from Hedera helix. Thirty-six compounds were initially screened against F. hepatica newly excysted juveniles (NEJs) of the Italian strain. Eleven of these compounds were active against NEJs and were selected for further study, using adult F. hepatica derived from a local abattoir (provenance unknown). From these eleven compounds, six demonstrated activity and were further assessed against immature liver flukes of the Italian strain. Subsequently, the most active compounds (n = 5) were further evaluated in ex vivo dose response experiments against adult Italian strain liver flukes. Overall, MC042 was identified as the most active molecule and the EC50 obtained from immature and adult liver fluke assays (at 24 h post co-culture) are estimated as 1.07 μM and 13.02 μM, respectively. When compared to the in vitro cytotoxicity of MDBK bovine cell line, MC042 demonstrated the highest anthelmintic selectivity (44.37 for immature and 3.64 for adult flukes). These data indicate that modified hederagenins display properties suitable for further investigations as candidate flukicides.

This second edition guideline was prepared to assist in the planning, conduct and interpretation of studies to assess the efficacy of parasiticides against ectoparasites of ruminants. It provides updated information on the selection of animals, dosage determination, dosage confirmation and field studies, record keeping and result interpretation. This guideline is intended to assist investigators on how to conduct specific studies, to provide specific information for registration authorities involved in the decision-making process, to assist in the approval and registration of new ectoparasiticides, and to facilitate the worldwide adoption of standard procedures.

National audience; Le présent bilan de mandature 2016-2021 de la CPCN (Conférence des présidentes et présidents de sections et de commissions interdisciplinaires du Comité national de la recherche scientifique) a une triple finalité. Il constitue d’abord une forme d’autoévaluation de l’action de la CPCN et, à travers elle, de l’action coordonnée des sections et commissions interdisciplinaires (CID), au cours de cette mandature, en rappelant autant que de besoin le contexte dans lequel cette action s’est inscrite. Il vise également à rendre compte de cette action auprès de la communauté scientifique française à travers une diffusion la plus large possible. Il doit enfin permettre un passage de relais avec les sections et CID et avec la CPCN qui seront mises en place à l’automne 2021.

Control of liver fluke infections remains a significant challenge in the livestock sector due to widespread distribution of drug resistant parasite populations. In particular, increasing prevalence and economic losses due to infection with Fasciola hepatica is a direct result of drug resistance to the gold standard flukicide, triclabendazole. Sustainable control of this significant zoonotic pathogen, therefore, urgently requires the identification of new anthelmintics. Plants represent a source of molecules with potential flukicidal effects and, amongst their secondary metabolites, the diterpenoid abietic acids can be isolated in large quantities. In this study, nineteen (19) chemically modified abietic acid analogues (MC_X) were first evaluated for their anthelmintic activities against F. hepatica newly excysted juveniles (NEJs, from the laboratory-derived Italian strain); from this, 6 analogues were secondly evaluated for their anthelmintic activities against adult wild strain flukes. One analogue, MC010, was progressed further against 8-week immature- and 12-week mature Italian strain flukes. These guidelines are intended to provide an in-depth review of current knowledge and assist the planning and implementation of studies for evaluating the efficacy of parasiticides in reducing transmission of vector-borne pathogens (VBPs) to dogs and cats. At present, the prevention of VBP transmission in companion animals is generally achieved through the administration of products that can repel or rapidly kill arthropods, thus preventing or interrupting feeding before transmission occurs. The present guidelines complement existing guidelines, which focus on efficacy assessment of parasiticides for the treatment, prevention and control of flea and tick infestations, but also give guidance for studies focused on other vectors (i.e. mosquitoes and phlebotomine sand flies). The efficacy of parasiticides in reducing VBP transmission can be evaluated through laboratory or field studies. As such, the present guidelines provide recommendations for these studies, representing a tool for researchers, pharmaceutical companies and authorities involved in the research, development and registration of products with claims for reducing VBP transmission in dogs and cats, respecting the overall principles of the 3Rs (replacement, reduction and refinement). Gaps in our current understanding of VBP transmission times are herein highlighted and the need for further basic research on related topics is briefly discussed.

The fast food supply chain is facing increasing operating costs due to volatile food and energy prices. Based on a case study of a major fast food logistics operator, this paper quantifies the potential for fuel generation from the waste generated by quick-service restaurants in Britain. Several fuel pathways and supply chains were mapped to understand the carbon intensity of the various waste-to-fuel opportunities, the number of heavy goods vehicles that might be powered and the key factors that could help companies make better informed decisions related to fuel generation from waste.

Scholars have often assumed that the Upper Canadian social class system was shaped by a hierarchical and landed patronage system known as the Family Compact. Based on the views of Bishop John Strachan and Lieutenant Governor John Graves Simcoe, this Family Compact is viewed as a post-War of 1812 development and is said to replace the oligarchy that was in place in 1791. An examination of the Loyalist settlement townships, in particular Ernestown Township and the Cataraqui Townships, suggests instead that a mercantile aristocracy of patronage and wealth existed by 1791, including Richard Cartwright Jr. of Kingston, along with rural leaders such as the Fairfields and Parrotts of Ernestown. This study of a key and complex time and place challenges prevailing views on class and class consciousness in Upper Canada and refines our understanding of this society. Such an investigation is timely given both the seeming unwillingness of historians to fully challenge existing depictions of the Upper Canadian class system, despite their noticeable flaws, and the impending commemorations of the War of 1812. Using archival documents ( accounts and letters) relating to two Loyalist/merchant families (the Parrotts and the Fairfields) along with a re-interpretation of secondary sources, a new view of a “Merchant Compact” is explored. This approach encompasses the changing relations of the settlements in question (Ernestown/Bath and Kingston) and shows the importance of previously neglected figures such as James Parrott. More broadly, it contributes new layers of analysis to the discussion of class consciousness in Upper Canada.

Increasing threats from anthelmintic resistant nematode populations warrant and motivate a reappraisal of chemotherapeutic strategies for nematode control in ruminant livestock and horses. The objective of this paper is to present a guideline for the evaluation of products containing two or more constituent anthelmintic actives in a single dosage form for the treatment of nematode infections in these animals. At present, regulatory policies on the approval of such products vary across jurisdictions, and this World Association for the Advancement of Veterinary Parasitology (W.A.A.V.P.) guideline should enable the harmonization of the requirements. This guideline makes clear recommendations on the minimal standards needed, but stresses that registration dossiers for combination anthelmintic products submitted for approval must conform to the standards and practices already established in existing guidelines for anthelmintics.

This thesis examines the properties of the interface between two phases in phase separated systems. We are interested in how finite size effects modify the statistical properties of these interfaces, in particular how the dependence of the free energy on the system size gives rise to long range critical Casimir forces close to thecritical point. Often the interfaces in phase separated systems are described by simplified or coarsegrained models whose only degrees of freedom are the interface height. We review how the statics and dynamics of these interface models can be derived from microscopic spin models and statistical field theories. We then examine finite size effects for continuous interface models such as the Edwards Wilkinson model and discrete models such as the Solid-On-Solid model and discuss their relevance to the critical Casimir effect. In the second part of the thesis we examine models of driven interfaces which have nonequilibrium steady states. We develop a model C type model of an interface which shows a nonequlibrium steady state even with constant driving. The resulting nonequlibrium steady state shows properties seen in experiments on sheared colloidal systems, notably the suppression of height fluctuations but an increase in the fluctuations’correlation length. Finally we propose a new model for one dimensional interfaces which is a modification of the solid on-solid model and containing an extra entropic term ,whose correspondance with physical systems is yet to be found.; Cette thèse examine les propriétés de l’interface entre deux phases dans un système de phases séparées. Nous regardons comment les effets de taille finies modifient les propriétés statistiques de ces interfaces,en particulier comment la dépendance de l’énergie libre par rapport à la taille du système donne lieu à des interactions de Casimir critique à longue portée proche du point critique. Souvent, les interfaces sont décrites par des modèles simplifiés ou coarse-grained dont les seuls degrés de libertés ont les hauteurs de l’interface. Nous rappelons comment les propriétés statiques et dynamiques de ces interfaces sont retrouvées à partir de modèles microscopiques de spins et de la théorie statistique des champs. Nous étudions ensuite les effets de taille finie pour les interfaces continues comme le modèle Edwards-Wilkinson ou discrètes comme le modèle Solid-On-Solid,et discutons leur pertinence dans le cadre de l’effet Casimir critique. Dans la seconde partie de la thèse, nous examinons des modèles d’interfaces sous écoulement possédant des états stationnaires hors-équilibre. Nous développons ces équations dans le cadre du modèle C d’une interface,ayant un état stationnaire hors-équilibre lorsque soumis à un écoulement uniforme. L’état stationnaire hors-équilibre résultant exhibe des propriétés retrouvées dans les expériences sur des colloïdes sous cisaillement ,notamment la suppression des fluctuations de la hauteur de l’interface et une augmentation de la longueur de corrélation des fluctuations. Finalement,nous proposons un nouveau modèle pour des interfaces uni-dimensionnelles qui est une modification du modèle Solid-on-Solid contenant un terme supplémentaire d’entropie, dont la correspondance à des systèmes physiques reste à être trouvée.

Spin ices, such as Dy2Ti2O7 and Ho2Ti2O7, are materials exhibiting exotic magnetic properties. They were the first frustrated ferromagnetic crystalline compounds to be discovered. The frustration leads to the fractionnalisation of the spin degrees of freedom and the emergence of magnetic monopoles, whose physics is formalised in the dumbbell model. In this thesis, we study the full phase diagram of this model in analogy with theS=2 Blume-Capel model. We identify in this diagram the fragmented phase observed experimentally in Ho2Ir2O7, and we localise the critical point of the transition between the spin ice phase and the fragmented phase.In a second part, we show numerically that the dynamics of this system at thecritical point belongs to the 3D Ising university class. We use for this two tools :the Kibble-Zurek scaling law and the fluctuation-dissipation ratio. For the latter, ithas been necessary to introduce a novel method to measure response functions. Wealso emphasize that these tools are specifically interesting for spin ice materials, as the unusually long microscopic time scale (1 μs) should make it possible to experimentallyobserve out-of-equilibrium phenomena related to critical slowing down.In a third part, we use the violation of the fluctuation-dissipation theorem to characterise a strongly out-of-equilibrium regime of spin ice - a thermal quench from high to low temperature, where degrees of freedom are kinetically blocked because ofthe Coulombic attraction between the monopoles.; Les glaces de spin, comme Dy2Ti2O7 et Ho2Ti2O7, sont des matériaux présentant un magnétisme particulièrement exotique. Ils constituent les premiers composés cristallins ferromagnétiques frustrés à avoir été découverts. Cette frustration permet la fractionnalisation des degrés de liberté de spin et l’émergence de monopôles magné-tiques, dont la physique est formalisée par le modèle des haltères.Dans cette thèse, nous étudions dans un premier temps le diagramme de phase de ce modèle grâce à un parallèle avec le modèle de Blume-Capel S = 2. On identifie dans ce diagramme la phase fragmentée observée expérimentalement dans Ho2Ir2O7,et on localise le point critique de la transition entre la phase glace de spin et la phase fragmentée.Dans un second temps, on montre numériquement que la dynamique du système autour de ce point critique appartient à la classe d’universalité du modèle d’Ising 3D. On utilise pour cela deux outils : les lois d’échelle de Kibble-Zurek et le rapport de fluctuation-dissipation. L’obtention de ce dernier a nécessité l’introduction d’une méthode novatrice pour le calcul des fonctions de réponse. Nous soulignons également que ces outils sont spécifiquement intéressants dans le cas des glaces de spin où les temps microscopiques sont de l’ordre de 1 μs, rendant le ralentissement critique observable expérimentalement.Dans un troisième temps, nous employons à nouveau la violation du théorème de fluctuation-dissipation pour caractériser un régime fortement hors équilibre de la phase glace de spin, où les degrés de liberté sont cinétiquement bloqués du fait de l’attraction coulombienne entre les monopôles.

Approaching a critical point, the divergence of the correlation length of fluctuations can be cut-off by a confinement of the system. This truncation fosters finite size effects with universal features in a class of continuous phase transitions. We are particularly interested in the Ising universality class, regrouping transitions such as the ferromagnetic/paramagnetic transition for uniaxial magnetic systems, the liquid/gas tran- sition and the demixing of binary mixtures. We will first present an introduction to critical phenomena, universality, finite-size scaling and Monte Carlo simulations of the Ising model, on which a major part of this work relies.A finite size effect that has particularly drawn attention in the past decades is the critical Casimir force. On the one hand, theoretical and numerical works on the subject have almost systematically been performed in magnetic model systems, such as the Ising or XY models. On the other hand, experimental approaches were all realized in fluid systems, such as binary mixtures or helium IV close to the superfluid transition.A motivation of this work was to bridge this gap by proposing, firstly, an experimental protocol for measuring the critical Casimir force in a magnetic layer and, secondly, a numerical approach in a Lennard-Jones binary mixture. The latter is of particular interest as it could lead the way to studying fluctuations of the Casimir force or out-of-equilibrium phenomena., À l'approche d'un point critique, la divergence de la longueur de corrélation des fluctuations peut être tronquée par le confinement du système. Cette troncature engendre des effets de taille finie présentant des caractères universels au sein d'un classe de transitions de phases.Nous nous sommes intéressés particulièrement à la classe d'universalité du modèle d'Ising, regroupant notamment les transitions de phase ferro/paramagnétique pour les systèmes magnétiques uniaxiaux, la transition liquide/gaz et encore la démixtion de mélanges binaires. Nous présentons tout d'abord une introduction aux phénomènes critiques, à l'universalité, au « finite-size scaling » et aux simulations Monte Carlo du modèle d'Ising, sur lesquelles se fondent la majeur partie de ce travail.Un effet de taille finie ayant attiré une grande attention durant les dernières dizaines d'années est la force de Casimir critique. Les travaux théoriques et numériques concernant cette force ont, dans leur quasi totalité, été menés dans des systèmes magnétiques modèles, tel que les modèles d'Ising ou XY. Par contre, les approches expérimentales ont toutes été réalisées dans des systèmes fluides, tels que des mélanges binaires ou de l'hélium IV proche de la transition superfluide.Une motivation de ce travail a été de chercher a résoudre cette situation paradoxale en proposant, d'une part, un protocole expérimental pour la mesure de la force de Casimir dans une couche mince magnétique et, d'autre part, une approche numérique dans un mélange binaire de type Lennard-Jones. Cette dernière approche présente l'avantage d'ouvrir la porte à des études des fluctuations de la force de Casimir ou encore hors-équilibre.

This thesis addresses the emergent electrostatics of two-dimensional, toroidal magnetic models that possess XY symmetry, providing a platform for novel investigations into the Berezinskii-Kosterlitz-Thouless (BKT) phase transition.The BKT transition drives the thermal dissociation of bound pairs of topological defects in many two-dimensional systems, including the two-dimensional XY model of magnetism. The XY model is closely analogous to the two-dimensional Coulomb gas, but can be simulated without computing the long-range interactions of the Coulombic system. This thesis elucidates this paradox by showing that Villain's approximation to the XY model is strictly equivalent to the Maggs-Rossetto (MR) electrostatic model when applied to the two-dimensional Coulomb gas.The mapping is used to probe the BKT transition through the application of the MR algorithm to the two-dimensional Coulomb gas. By simulating the Coulombic system, fluctuations in the winding of charges around the torus are shown to turn on at the BKT transition temperature. These topological-sector fluctuations in the electric field therefore signal the high-temperature phase of the transition.It is then shown that the effective critical exponent of Bramwell-Holdsworth (BH) theory can be measured in superfluid 4He films, which correspond to effective Coulomb gases in the limit of large but finite system size. With the Coulombic system taken as the base BKT system, it is inferred that BH theory is a general property of BKT systems., Cette thèse s'intéresse aux phénomènes électrostatiques émergents dans les modèles magnétiques toroïdaux bi-dimensionnels à symétrie XY, fournissant ainsi un support pour de plus amples recherches dans le domaine de la transition de phase Berezinskii-Kosterlitz-Thouless (BKT).Dans de nombreux systèmes bi-dimensionnels, dont le modèle bi-dimensionnel XY du magnétisme, la transition BKT contrôle la dissociation thermique de paires de défauts topologiques liés. Le modèle XY est analogue au gaz de Coulomb bi-dimensionnel, à ceci près qu'il peut être simulé sans avoir à modéliser les interactions à longue distance du système Coulombien. Cette thèse élucide ce paradoxe en démontrant que l'approximation de Villain appliquée au modèle XY est strictement équivalente au modèle électrostatique de Maggs-Rossetto (MR) appliqué au système Coulombien bi-dimensionnel.Cette équivalence est utilisée pour sonder la transition BKT par l'application de l'algorithme MR au gaz de Coulomb bi-dimensionel. En simulant le système Coulombien, il est prouvé que les fluctuations dans l'organisation des charges autour du tore sont activées à la température de transition BKT. Ces fluctuations du champ électrique indiquent ainsi la phase de haute température de la transition.Il est ensuite montré que l'exposant critique effectif de la théorie de Bramwell-Holdsworth (BH) peut être mesuré dans les films d'hélium 4 superfluide, qui correspondent à des gaz de Coulomb effectifs dans la limite de systèmes de grandes tailles finies.

A Coulomb gas or fluid comprises charged particles that interact via the Coulomb interaction. Examples of a Coulombic systems include simple and complex electrolytes together with magnetic monopoles in spin ice. The long-range nature of the Coulomb interaction leads to a rich array of phenomena.This thesis is devoted to the study of the non-equilibrium behaviour of lattice based Coulomb gases and of the quasi-particle excitations in the materials known as spin ice which constitute a Coulomb gas of magnetic charges. At the centre of this study lies the second Wien effect which describes the linear increase in conductivity when an electric field is applied to a weak electrolyte. The conductivity increases due to the generation of additional mobile charges via a field-enhanced dissociation from Coulombically bound pairs.The seminal theory of Onsager gave a detailed analysis of the Wien effect. We use numerical simulations not only to confirm its validity in a lattice Coulomb gas for the first time but mainly to study its extensions due to the role of the ionic atmosphere and field-dependent mobility. The simulations also allow us to observe the microscopic correlations underlying the Wien effect.Finally, we look more closely at the emergent gas of monopoles in spin ice—the magnetolyte. The magnetic behaviour of spin ice reflects the properties of the Coulomb gas contained within. We verify the presence of the Wien effect in model spin ice and in the process predict the non-linear response when exposed to a periodic driving field, or to a field quench using Wien effect theory. We use a straightforward extension of the lattice Coulomb gas simulations to refine our predictions. It is a highly unusual result to find an analytic theory for the non-equilibrium behaviour of a highly frustrated system beyond linear response., Les gaz ou fluides de Coulomb sont composés de particules chargées couplées entre elles par interaction coulombienne à longue portée. De part la nature de ces interactions, la physique du gaz de Coulomb est très riche, comme par exemple dans des électrolytes plus ou moins complexes, mais aussi à travers l'émergence de monopôles magnétiques dans la glace de spin. Dans cette thèse nous nous intéressons au comportement hors d'équilibre des gaz de Coulomb et de la glace de spin. Au centre de cette étude se trouve le deuxième effet de Wien, qui est une croissance linéaire de la conductivité en fonction du champ électrique appliqué à un électrolyte faible. Ce phénomène est une conséquence directe de l'interaction coulombienne qui pousse les charges à se lier par paires ; le champ électrique va alors aider à dissocier ces paires et créer des charges mobiles qui amplifient la conductivité. Le deuxième effet de Wien est un processus hors-équilibre non-linéaire, remarquablement décrit par la théorie de Onsager. Nos simulations sur réseau permettent de découvrir le rôle de l'environnement ionique qui agit contre le deuxième effet de Wien, ainsi que de caractériser la mobilité du système et sa dépendance en fonction du champ externe. Les simulations nous ont aussi donné accès aux corrélations de charges qui décrivent le processus microscopique à la base de l'effet Wien. Enfin, nous regardons plus précisément le gaz émergent de monopôles dans la glace de spin, aussi appelé « magnétolyte », capable de décrire de manière remarquable les propriétés magnétiques de glace de spin. Nous décrivons la dynamique complète hors-équilibre de cette magnétolyte soumise à une forçage périodique ou une trempe dans un champ magnétique en incluant à la fois le deuxième effet de Wien et la réponse du réseau de spins qui est à la base de l'émergence des monopôles magnétiques. Tout au long, nous utilisons une simple extension des simulations de gaz de Coulomb sur réseau pour préciser nos prédictions. Il est très rare de trouver une théorie analytique du comportement hors-équilibre d'un système hautement frustré au-delà de la réponse linéaire.

Les systèmes de spin antiferromagnétiques en une dimension présentent des caractéristiques remarquables. Le théorème de Mermin-Wagner montre que même dans l'état fondamental de leur Hamiltonien, ces systèmes ne possèdent pas d'ordre à longue distance dans leurs fonctions de corrélation spin-spin s'ils possèdent une symétrie continue. Néanmoins, un quasi-ordre à longue distance peut exister, avec des fonctions de corrélation spin-spin décroissant en loi de puissance avec la distance, comme par exemple dans la chaîne de spin-1/2. Ce cas est décrit par la théorie des liquides de Luttinger. Même dans le cas d'un ordre à courte distance où les fonctions de corrélations spin-spin décroissent exponentiellement, un ordre topologique peut être présent. Il peut être mis en évidence par un paramètre d'ordre dépendant des opérateurs de spin de façon non-locale ou par la présence d'excitations de bord, comme par exemple dans le cas de la chaînes de spin-1. D'autre part, il existe une équivalence formelle entre les opérateurs de spin-1/2 et les bosons de coeur dur, qui permet de traduire les propriétés des systèmes magnétiques dans le langage des bosons en interaction. Après une rapide revue des méthodes théoriques utilisées pour la description des systèmes de basse dimensionnalité, je décrirais mes travaux sur les systèmes échelles de spin antiferromagnétiques et les applications aux systèmes de bosons en interaction.

Nous regardons comment les effets de taille finies modifient les propriétés statistiques de ces interfaces,en particulier comment la dépendance de l’énergie libre par rapport à la taille du système donne lieu à des interactions de Casimir critique à longue portée proche du point critique. Souvent, les interfaces sont décrites par des modèles simplifiés ou coarse-grained dont les seuls degrés de libertés ont les hauteurs de l’interface. Nous rappelons comment les propriétés statiques et dynamiques de ces interfaces sont retrouvées à partir de modèles microscopiques de spins et de la théorie statistique des champs. Nous étudions ensuite les effets de taille finie pour les interfaces continues comme le modèle Edwards-Wilkinson ou discrètes comme le modèle Solid-On-Solid,et discutons leur pertinence dans le cadre de l’effet Casimir critique. Dans la seconde partie de la thèse, nous examinons des modèles d’interfaces sous écoulement possédant des états stationnaires hors-équilibre. Nous développons ces équations dans le cadre du modèle C d’une interface,ayant un état stationnaire hors-équilibre lorsque soumis à un écoulement uniforme. L’état stationnaire hors-équilibre résultant exhibe des propriétés retrouvées dans les expériences sur des colloïdes sous cisaillement ,notamment la suppression des fluctuations de la hauteur de l’interface et une augmentation de la longueur de corrélation des fluctuations. Finalement,nous proposons un nouveau modèle pour des interfaces uni-dimensionnelles qui est une modification du modèle Solid-on-Solid contenant un terme supplémentaire d’entropie, dont la correspondance à des systèmes physiques reste à être trouvée.
