Magnetic iron oxide nanocrystals (MIONs) are established as potent theranostic nanoplatforms due to their biocompatibility and the multifunctionality of their spin-active atomic framework. Recent insights have also unveiled their attractive near-infrared photothermal properties, which are, however, limited by their low near-infrared absorbance, resulting in noncompetitive photothermal conversion efficiencies (PCEs). Herein, we report on the dramatically improved photothermal conversion of condensed clustered MIONs, reaching an ultrahigh PCE of 71% at 808 nm, surpassing the so-far MION-based photothermal agents and even benchmark near-infrared photothermal nanomaterials. Moreover, their surface passivation is achieved through a simple self-assembly process, securing high colloidal stability and structural integrity in complex biological media. The bifunctional polymeric canopy simultaneously provided binding sites for anchoring additional cargo, such as a strong near-infrared-absorbing and fluorescent dye, enabling in vivo optical and photoacoustic imaging in deep tissues, while the iron oxide core ensures detection by magnetic resonance imaging. In vitro studies also highlighted a synergy-amplified photothermal effect that significantly reduces the viability of A549 cancer cells upon 808 nm laser irradiation. Integration of such-previously elusive-photophysical properties with simple and cost-effective nanoengineering through self-assembly represents a significant step toward sophisticated nanotheranostics, with great potential in the field of nanomedicine.

Hypericin (Hyp) is a hydrophobic pigment found in plants of the genus Hypericum which exhibits low levels of solubility in water. This work shows that the solubility of Hyp can be significantly increased through the addition of cromolyn disodium salt (DSCG). Performed studies using UV-VIS absorption and fluorescence spectroscopies demonstrate that Hyp remains in a predominantly biologically photodynamic active monomeric form in the presence of DSCG at concentrations ranging from 4.6×10-3 to 1.2×10-1mol·L-1. The low association constant between Hyp and DSCG (Ka=71.7±2M-1), and the polarity value of 0.3 determined for Hyp in a DSCG-water solution, lead to a suggestion that the monomerization of Hyp in aqueous solution can be explained as a result of the hydrotropic effect of DSCG. This hydrotropic effect is most likely a result of interactions between two relative rigid aromatic rings of DSCG and a delocalized charge on the surface of the Hyp molecule. The triplet-triplet (T-T) electronic transition observed in is Hyp in the presence of DSCG suggests a possible production of reactive oxygen species once Hyp is irradiated with visible light in a DSCG aqueous solution.

In this work we established three from altogether six proton dissociation constants (p K a ) of hydroxyl groups of hypericin in its monomeric form. The monomeric state of hypericin (5.0 × 10 −6 mol·L −1 ) in aqueous solution was stabilised by the presence of hydrotropic drug Cromolyn disodium salt (6.0 × 10 −2 mol·L −1 ). Data show that one acid-base transition occurs with the p K a of 7.8 and the other two are characterised by the apparent single p K a of 11.5. The spectral changes of hypericin above pH 13 indicate that the last two hydroxyls are deporotonized at this high pH values.

A rapid separation of the ten nuclearly-encoded subunits of mitochondrial cytochrome c oxidase, and ten out of the eleven subunits of cytochrome bc 1 , was achieved using a short, 50 mm C 18 -reversed-phase column. The short column decreased the elution time 4–7 fold while maintaining the same resolution quality. Elution was similar to a previously published protocol, i.e., a water/acetonitrile elution gradient containing trifluoroacetic acid. Isolated subunits were identified by MALDI-TOF. The rapidity of the described method makes it extremely useful for determining the subunit composition of isolated mitochondrial complexes. The method can be used for both analytical and micro-preparative purposes.

A composite material of graphite/polystyrene sulfonate was prepared by solvothermal method. The dissolution of polystyrene sulfonate in water ensures homogeneous distributions of graphite and creates composition material at room temperatures. Water removing by the drying, permit to prepare films. Structure of the film was analyzed by scanning electron microscopy. The influence of different ratio between graphite and polystyrene sulfonate is discussed.

Copper (II) oxide graphene composite materials were prepared on the electrode surface at the solution of CuCl2. The resulting composite material was characterized by cyclic voltammetry on surface of the glassy carbon electrode. Prepared copper (II) oxide graphene nanocomposite was scraped from the electrode surface and was measured Raman spectra by optical tweezers combined with micro Raman spectroscopy. This composite nanomaterial was used on determination of glucose and can be used to construct bio batteries too.

Several attempts have appeared recently to control optical trapping systems via touch tablets and cameras instead of a mouse and joystick. Our approach is based on a modern low-cost hardware combined with fingertips and speech recognition software. Positions of operator's hands or fingertips control the positions of trapping beams in holographic optical tweezers that provide optical manipulation with microobjects. We tested and adapted two systems for hands position detection and gestures recognition – Creative Interactive Gesture Camera and Leap Motion. We further enhanced the system of Holographic Raman tweezers (HRT) by voice commands controlling the micropositioning stage and acquisition of Raman spectra. Interface communicates with HRT either directly by which requires adaptation of HRT firmware, or indirectly by simulating mouse and keyboard messages. Its utilization in real experiments speeded up the operator’s communication with the system cca. Two times in comparison with the traditional control by the mouse and the keyboard.

We proposed the improved control software for the holographic optical tweezers (HOT) proper for simple semi-automated sorting. The controller receives data from both the human interface sensors and the HOT microscope camera and processes them. As a result, the new positions of active laser traps are calculated, packed into the network format and sent to the remote HOT. Using the photo-polymerization technique, we created a sorting container consisting of two parallel horizontal walls where one wall contains “gates” representing a place where the trapped particle enters into the container. The positions of particles and gates are obtained by image analysis technique which can be exploited to achieve the higher level of automation. Sorting is documented on computer game simulation and the real experiment.

Holographic Raman tweezers (HRT) manipulates with microobjects by controlling the positions of multiple optical traps via the mouse or joystick. Several attempts have appeared recently to exploit touch tablets, 2D cameras or Kinect game console instead. We proposed a multimodal “Natural User Interface” (NUI) approach integrating hands tracking, gestures recognition, eye tracking and speech recognition. For this purpose we exploited “Leap Motion” and “MyGaze” low-cost sensors and a simple speech recognition program “Tazti”. We developed own NUI software which processes signals from the sensors and sends the control commands to HRT which subsequently controls the positions of trapping beams, micropositioning stage and the acquisition system of Raman spectra. System allows various modes of operation proper for specific tasks. Virtual tools (called “pin” and “tweezers”) serving for the manipulation with particles are displayed on the transparent “overlay” window above the live camera image. Eye tracker identifies the position of the observed particle and uses it for the autofocus. Laser trap manipulation navigated by the dominant hand can be combined with the gestures recognition of the secondary hand. Speech commands recognition is useful if both hands are busy. Proposed methods make manual control of HRT more efficient and they are also a good platform for its future semi-automated and fully automated work.

Holographic optical tweezers provide a contactless way to trap and manipulate several microobjects independently in space using focused laser beams. Although the methods of fast and efficient generation of optical traps are well developed, their user friendly control still lags behind. Even though several attempts have appeared recently to exploit touch tablets, 2D cameras, or Kinect game consoles, they have not yet reached the level of natural human interface. Here we demonstrate a multi-modal 'natural user interface' approach that combines finger and gaze tracking with gesture and speech recognition. This allows us to select objects with an operator's gaze and voice, to trap the objects and control their positions via tracking of finger movement in space and to run semi-automatic procedures such as acquisition of Raman spectra from preselected objects. This approach takes advantage of the power of human processing of images together with smooth control of human fingertips and downscales these skills to control remotely the motion of microobjects at microscale in a natural way for the human operator.
