1. Genomic tailoring of autogenous poultry vaccines to reduce Campylobacter from farm to fork

Abstract

Abstract Campylobacter is a leading cause of food-borne gastroenteritis worldwide, linked to the consumption of contaminated poultry meat. Targeting this pathogen at source, vaccines for poultry can provide short-term caecal reductions in Campylobacter numbers in the chicken intestine. However, this approach is unlikely to reduce Campylobacter in the food chain or human incidence. This is likely as vaccines typically target only a subset of the high genomic strain diversity circulating among chicken flocks, and rapid evolution diminishes vaccine efficacy over time. To address this, we used a genomic approach to develop a whole-cell autogenous vaccine targeting isolates harbouring genes linked to survival outside of the host. We hyper-immunised a whole major UK breeder farm to passively target offspring colonisation using maternally-derived antibody. Monitoring progeny, broiler flocks revealed a near-complete shift in the post-vaccination Campylobacter population with an ~50% reduction in isolates harbouring extra-intestinal survival genes and a significant reduction of Campylobacter cells surviving on the surface of meat. Based on these findings, we developed a logistic regression model that predicted that vaccine efficacy could be extended to target 65% of a population of clinically relevant strains. Immuno-manipulation of poultry microbiomes towards less harmful commensal isolates by competitive exclusion, has major potential for reducing pathogens in the food production chain.

2. Alternative method for assessment of southwestern Atlantic humpback whale population status.

Abstract

The population of humpback whales (Megaptera novaeangliae) wintering off eastern South America was exploited by commercial whaling almost to the point of extinction in the mid-twentieth century. Since cessation of whaling in the 1970s it is recovering, but the timing and level of recovery is uncertain. We implemented a Bayesian population dynamics model describing the population's trajectory from 1901 and projecting it to 2040 to revise a previous population status assessment that used Sampling-Importance-Resampling in a Bayesian framework. Using our alternative method for model fitting (Markov chain Monte Carlo), which is more widely accessible to ecologists, we replicate a "base case scenario" to verify the effect on model results, and introduce additional data to update the status assessment. Our approach allowed us to widen the previous informative prior on carrying capacity to better reflect scientific uncertainty around historical population levels. The updated model provided more precise estimates for population sizes over the period considered (1901-2040) and suggests that carrying capacity (K: median 22,882, mean 22,948, 95% credible interval [CI] 22,711-23,545) and minimum population size (N1958: median 305, mean 319, 95% CI 271-444) might be lower than previously estimated (K: median 24,558, mean 25,110, 95% CI 22,791-31,118; N1958: median 503, mean 850, 95% CI 159-3,943). However, posterior 95% credible intervals of parameters in the updated model overlap those of the previous study. Our approach provides an accessible framework for investigating the status of depleted animal populations for which information is available on historical mortality (e.g., catches) and intermittent estimates of population size and/or trend.

3. Oceanic Drivers of Sei Whale Distribution in the North Atlantic

Abstract

This study investigated the oceanic drivers of sei whale (Balaenoptera borealis) distribution in the central and eastern North Atlantic, and explored how distribution may have changed over almost three decades. Cetacean sightings data were available from Icelandic, Faroese and Norwegian surveys conducted throughout the central and eastern North Atlantic during summer between 1987 and 2015. Effective strip half width was estimated from the data to take account of variation in detection probability. Spatially-referenced environmental variables used as predictors in generalised additive models of sei whale relative density included: relief-related variables seabed depth, slope and aspect; monthly-varying physical oceanographic variables sea surface temperature (SST), mixed layer depth, bottom temperature, salinity, and sea surface height anomaly (SSH); and monthly-varying biological oceanographic variables chlorophyll-a concentration and primary productivity. Preliminary analysis considered which month (March-August) in the dynamic oceanographic variables explained most variability in sei whale density. Models including all variables (“full models”) could only be run for 1998-2015 because data for several variables were missing in earlier years. “Simple models" including only relief-related variables and SST were therefore run for 1987-89, and also for 1998-2015 for comparison. The best-fitting full model for 1998-2015 retained the covariates depth, May SST, May bottom temperature, July salinity, July SSH and July primary productivity. Of these, depth, May SST and July SSH were the strongest predictors of sei whale density. In the simple models for both 1987-89 and 1998-2015, depth (especially), May SST and seabed slope were the strongest predictors of sei whale density. The highest densities of sei whales were predicted in the Irminger Sea and over the Charles-Gibbs Fracture Zone; a pattern driven by large negative SSH, deep water (>1500m) and polar-temperate SST (5-12oC). There was some inter-annual variability in predicted distribution and there appears to be a northward expansion in distribution consistent with prey species responding to ocean warming. The models could be used to predict future distribution of sei whales based on future environmental conditions predicted by climate models.

Abstract

This article is a book review of Beyond Confrontation: Globalists, Nationalists and Their Discontents by Phil Mullan. Like other writers, Mullan warns of the danger of rising global tensions, but what marks out his contribution is a demystifying historical analysis which identifies the underlying sources of conflict and offers an optimistic and forward-looking vision. The first part of the book examines the emergence of the current rules-based international order in the aftermath of the Second World War, uncovers its inherent flaws, and draws out how it has now become a source of conflict rather than stability. Part two is devoted to debunking myths about trade and trade agreements—particularly the idea that these are central to economic dynamism. Arguing that the relationship between trade and economic growth has been turned upside down, Mullan aims to put it back on its feet. The concluding part sets out some positive proposals for how to reimagine economic policy approaches in ways that could enable root-and-branch renewal and to think afresh about what type of political culture would facilitate such necessary changes.

Abstract

This chapter places the concept of sustainability in the context of recent debates about the environment, particularly climate change, as a ‘post-political’ issue. Exploring this concept, especially in relation to contemporary discussions of climate communication, the chapter suggests that the broad acceptance of sustainability can be understood as exemplifying the post-political condition identified by critics as foreclosing debate about possible alternative futures. The chapter argues that at the core of the problem of the post-political is a failure of agency – that the apparent difficulty of imagining large-scale social and political transformation results from a diminished view of the human subject. Ecological perspectives, whether advanced by conservatives or radicals, are hostile to attempts to master or control nature rather than respecting natural limits, and therefore tend to give expression to this anti-humanist viewpoint rather than challenging it.

Abstract

Summary Background Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. Methods We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung's disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. Findings We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung's disease) from 264 hospitals (89 in high-income countries, 166 in middle-income countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). This article examines the problem of how to interpret competing, clashing or contradictory news frames in coverage of war and conflict, focusing on the reporting of the 1992–1995 Bosnian war. ‘Ethnic war’ and ‘genocide’ featured as competing news frames in news coverage of Bosnia and several subsequent conflicts, and are often understood to be contradictory in terms of their implied explanations, moral evaluations and policy prescriptions. The author questions the assumptions that many journalists and academics have made about these frames and the relationship between them. He asks how we can make sense of clashing or contradictory scholarly analyses of these competing frames and considers a number of broader issues for framing analysis: the significance of historical context for understanding the meaning of particular framing devices, the importance of quantification in framing analysis and the role of influential sources in prompting journalists to adopt particular frames.

Abstract

Congenital diaphragmatic hernia (CDH) is typified morphologically by failure of diaphragmatic development with accompanying lung hypoplasia and persistent pulmonary hypertension of the newborn (PPHN). Patients who have labile physiology and low preductal saturations despite optimal ventilatory and inotropic support may be considered for extracorporeal membrane oxygenation (ECMO). Systematic reviews into the benefits of ECMO in CDH concluded that any benefit is unclear. Few randomised trials exist to demonstrate clear benefit and guide management. However, ECMO may have its uses in those that have reversibility of their respiratory disease. A few centres and networks have demonstrated an increase in survival rate by post hoc analysis (based on a difference in referral patterns with the availability of ECMO) in their series. One issue may be that of careful patient selection with regard to reversibility of pathophysiology. At present, there is no single test or prognostication that predicts reversibility of PPHN and criteria for referral for ECMO is undergoing continued refinement. Overall survival is similar between cannulation modes. There is no consensus on the time limit for ECMO runs. The optimal timing of surgery for patients on ECMO is difficult to definitively establish, but it seems that repair at an early stage (with careful perioperative management) is becoming less of a taboo, and may improve outcome and help with either coming off ECMO or decisions on withdrawal later. The provision of ECMO will continue to be evaluated, and prospective randomised trial are needed to help answer question of patient selection and management.

Many of today's most commercially successful videogames, from Call of Duty to Company of Heroes, are war-themed titles that play out in what are framed as authentic real-world settings inspired by recent news headlines or drawn from history. While such games are marketed as authentic representations of war, they often provide a selective form of realism that eschews problematic, yet salient aspects of war. In addition, changes in the way Western states wage and frame actual wars makes contemporary conflicts increasingly resemble videogames when perceived from the vantage point of Western audiences.This interdisciplinary volume brings together scholars from games studies, media and cultural studies, politics and international relations, and related fields to examine the complex relationships between military-themed videogames and real-world conflict, and to consider how videogames might deal with history, memory, and conflict in alternative ways. It asks: What is the role of videogames in the formation and negotiation of cultural memory of past wars? How do game narratives and designs position the gaming subject in relation to history, war and militarism? Abstract

Despite claims of continuity, contemporary data journalism is quite different from the earlier tradition of computer-assisted reporting. Although it echoes earlier claims about being scientific and democratic, these qualities are understood as resulting from better data access rather than as being something achieved by the journalist. In the context of Big Data in particular, human subjectivity tends to be downgraded in importance, even understood as getting in the way if it means hubristically theorising about causation rather than working with correlation and allowing the data to speak. Increasing ‘datafication’ is not what is driving changes in the profession, however. Rather, the impact of Big Data tends to be understood in ways that are consonant with pre-existing expectations, which are shaped by the broader contemporary post-humanist political context. The same is true in academic analysis, where actor–network theory seems to be emerging as the dominant paradigm for understanding data journalism, but in largely uncritical ways.

During normal cellular development, there is a complex system of checks and balances to ensure regulation of the cells as they proliferate and specialize to perform their physiological functions. Various genes cooperate, concomitantly and/or sequentially with others, to activate and direct the developmental mechanisms in a developing foetus. After their target is achieved, these pathways are either kept dormant or are used elsewhere in a different context (growth, repair, etc.). Reactivation of these genes, by various mutations and/or carcinogens, can reinstate these developmental pathways. If these pathways remain active incessantly and do not obey the normal regulatory mechanisms, cell proliferation becomes independent of the growth stimulus, and this produces a mass—cancer [1–3].

Since 2011, the international media have done much to highlight the suffering of civilians in the on-going war in Syria, including through innovative forms of reporting such as VR journalism and news games. However, by the end of 2016, questions were being raised about a number of high-profile news stories, such as the use of chemical weapons, the role of the ‘White Helmets’ relief workers, and the bombing of Aleppo and other cities. Amid the claims and counter-claims of propaganda and ‘fake news’, news audiences glimpsed shifting and clashing explanatory framings of the Syrian war. A conflict that had initially been understood against the background of the ‘Arab Spring’ began to be seen as complicated by sectarian religious tensions, the rise of Islamic State, opaque factional and regional alliances, and international tensions reminiscent of the Cold War. An imperative to establish a moral framework for the story seemed to preclude more complex and searching questions about the motivations and actions of local and international actors.

Abstract

Since the end of the Cold War there have been many competing ideas about how to explain contemporary conflicts, and about how the West should respond to them. This study examines how the media interpret conflicts and international interventions, testing the sometimes contradictory claims that have been made about recent coverage of war. Framing post-Cold War conflicts takes a comparative approach, examining UK press coverage across six different crises. Through detailed analysis of news content, it seeks to identify the dominant themes in explaining the post-Cold War international order, and to discover how far the patterns established prior to 11 September 2001 have subsequently changed. Based on extensive original research, the book includes case studies of two ‘humanitarian military interventions'(in Somalia and Kosovo), two instances where Western governments were condemned for not intervening enough (Bosnia and Rwanda), and the post-9/11 interventions in Afghanistan and Iraq.

Introduction Superior mesenteric artery syndrome is a rare condition characterised by nausea, vomiting, postprandial pain, anorexia and early satiety. Conservative management is tried initially, but if this fails, surgery is indicated. There are few reports in the literature concerning superior mesenteric artery syndrome in children, and fewer still managed surgically by minimally invasive means. Case presentation A 12-year-old girl presented with weight loss, early satiety and vomiting after corrective scoliosis surgery. After upper gastrointestinal endoscopy, contrast study and computed tomography imaging, a diagnosis of superior mesenteric artery syndrome was made. Conservative management by nasojejunal feeding failed; therefore, a laparoscopic duodeno-jejunostomy was undertaken. At follow-up, her symptoms had improved. Conclusion This report describes the youngest child to undergo laparoscopic duodeno-jejunostomy for superior mesenteric artery syndrome. Laparoscopic duodeno-jejunostomy appears to be the most widely employed and reliable minimally invasive approach to superior mesenteric artery syndrome with a high success rate and acceptably low complication rate.

This themed issue presents a selection of contributions from the Post-Screen Cultures/Prac- tices symposium held at London South Bank University in June 2016. It inaugurates what will become a new tradition of publishing an edition of the journal based on the annual MeCCSA Practice Network/JMP Practice Symposium. The 2016 symposium sought to explore the changing nature of our contemporary engagement with emergent screen- oriented technologies. The overarching theme to emerge was that the current moment is in some ways similar to the earliest days of cinema – a time of vibrant experimentation in which, while rejecting the fetishisation of technology and technological determinism, artists and media practitioners are grappling with emerging media forms and new tech- nological contexts in order to harness their potential.

For many years, the objective of environmental campaigners was to push climate change on to the agenda of political leaders and to encourage media attention to the issue. By the first decade of the twenty-first century, it appeared that their efforts had been spectacularly successful. This article examines the emotional rhetorical strategies of 3 films—The Day After Tomorrow (2004), An Inconvenient Truth (2006), and The Age of Stupid (2009)—that attempt to create engagements with the “postpolitical” problem of climate change. In all 3 films the experience of personal loss, the potential for future loss, and the emotions associated with loss are fundamental to affective engagement. The emotional loading of representations of environmental problems derives partly from concerns about human political agency and subjectivity. It is not so much that emotional or moral appeals are simply added on in order to bolster a political message, but rather that autobiographical narratives of loss and morality occupy the space once dominated by modernist forms of politics.
