The red and far-red light photoreceptor phytochrome B (phyB) transmits light signals following cytosol-to-nuclear translocation to regulate transcriptional networks therein. This necessitates changes in protein-protein interactions of phyB in the cytosol, about which little is presently known. Via introduction of a nucleus-excluding G767R mutation into the dominant, constitutively active phyBY276H (YHB) allele, we explore the functional consequences of expressing a cytosol-localized YHBG767R variant in transgenic Arabidopsis seedlings. We show that YHBG767R elicits selective constitutive photomorphogenic phenotypes in dark-grown phyABCDE null mutants, wild type and other phy-deficient genotypes. These responses include light-independent apical hook opening, cotyledon unfolding, seed germination and agravitropic hypocotyl growth with minimal suppression of hypocotyl elongation. Such phenotypes correlate with reduced PIF3 levels, which implicates cytosolic targeting of PIF3 turnover or PIF3 translational inhibition by YHBG767R. However, as expected for a cytoplasm-tethered phyB, YHBG767R elicits reduced light-mediated signaling activity compared with similarly expressed wild-type phyB in phyABCDE mutant backgrounds. YHBG767R also interferes with wild-type phyB light signaling, presumably by formation of cytosol-retained and/or otherwise inactivated heterodimers. Our results suggest that cytosolic interactions with PIFs play an important role in phyB signaling even under physiological conditions.

SUMMARY Tomato (Solanum lycopersicum L.) is an important model plant species in photomorphogenesis research. Ultraviolet B (UV‐B) induces the dissociation of homodimers of the photoreceptor UV RESISTANCE LOCUS8 (UVR8) into monomers, which translocate into the nucleus. Nuclear accumulation of UVR8 is a prerequisite for its signaling function. Previous studies have reported that SUPPRESSOR OF PHYTOCHROME A‐105 (SPA) family members may regulate UV‐B signaling in Arabidopsis (Arabidopsis thaliana); however, the underlying mechanism is unknown. Here, we show that the tomato genome encodes four SPA (SlSPA) orthologs. Genome‐edited Slspa3 mutants exhibited enhanced photomorphogenic responses in white light, suggesting that SlSPA3 inhibits general photomorphogenesis. By contrast, UVR8‐mediated gene expression in response to UV‐B was compromised in Slspa3 mutants, suggesting that SlSPA3 promotes UV‐B signaling. UV‐B‐induced nuclear accumulation of UVR8, which is essential for UV‐B signaling, was reduced in the Slspa3 mutants. Moreover, UV‐B‐induced nuclear accumulation of UVR8 was also reduced in the Arabidopsis spa1 spa2 spa3 and spa1 spa2 spa4 triple mutants, indicating a conserved mechanism in these two species. Notably, spa1 spa2 spa4 exhibited normal UV‐B‐induced interaction between UVR8 and the plant morphogenesis repressor CONSTITUTIVE PHOTOMORPHOGENIC 1 (COP1). This suggests that the well‐established mechanisms of UVR8 nuclear retention remained unaffected in spa1 spa2 spa4. Thus, our work uncovered a potentially unrecognized mechanism by which SPA proteins regulate UV‐B signaling through the promotion of UVR8 nuclear abundance in land plants. [ABSTRACT FROM AUTHOR]

ABSCISIC ACID‐INSENSITIVE 4 (ABI4) is a pivotal transcription factor which coordinates multiple aspects of plant growth and development as well as plant responses to environmental stresses. ABI4 has been shown to be involved in regulating seedling photomorphogenesis; however, the underlying mechanism remains elusive. Here, we show that the role of ABI4 in regulating photomorphogenesis is generally regulated by sucrose, but ABI4 promotes hypocotyl elongation of Arabidopsis seedlings under blue (B) light under all tested sucrose concentrations. We further show that ABI4 physically interacts with PHYTOCHROME INTERACTING FACTOR 4 (PIF4), a well‐characterized growth‐promoting transcription factor, and post‐translationally promotes PIF4 protein accumulation under B light. Further analyses indicate that ABI4 directly interacts with the B light photoreceptors cryptochromes (CRYs) and inhibits the interactions between CRYs and PIF4, thus relieving CRY‐mediated repression of PIF4 protein accumulation. In addition, while ABI4 could directly activate its own expression, CRYs enhance, whereas PIF4 inhibits, ABI4‐mediated activation of the ABI4 promoter. This study aimed to enhance androgenesis efficiency protocols by identifying critical factors using artificial neural networks and multivariate sensitivity analysis. Androgenesis studies were conducted on donor tomato plants grown under different lighting regimes (LEDs and natural light). Results indicated that LED lighting significantly enhanced photomorphogenesis, photosynthesis, but also subsequent anther development being crucial for successful haploid regeneration. An androgenesis model was developed using 60 different treatments, including as inputs: genotype, lighting regime, anther stage, and medium composition. Artificial Neural Networks (ANNs) and multivariate sensitivity analysis successfully ranked the importance of those four factors, identifying plant growth regulators (PGRs) media formulation as the most critical factors. The study also confirmed spontaneous haploid genome duplication in regenerants and validated the use of flow cytometry and cytogenetic analysis for ploidy determination. These findings provide valuable and practical insights into the factors driving androgenesis, facilitating the development of more efficient, targeted and cost-effective breeding strategies, such as the production of doubled haploid tomato.Key message: Our results, utilizing ANNs and multivariate analysis, identify key factors controlling androgenesis in tomato, leading to the development of innovative procedures for a rapid, cost-effective, and efficient production of doubled haploids. Light affects almost every aspect of plant development. It is perceived by photoreceptors, among which phytochromes (PHY) are responsible for monitoring the red and far-red spectrum. Arabidopsis thaliana possesses five phytochrome genes (phyA–phyE). Whereas functions of phyA and phyB are extensively studied, our knowledge of other phytochromes is still rudimentary. To analyze phyD function, we expressed it at high levels in different phytochrome-deficient genetic backgrounds. Overexpressed phyD-YFP can govern effective light signaling but only at low temperatures and in cooperation with functional phyC. Under these conditions, phyD-YFP accumulates to high levels, and opposite to phyB, this pool is stable in light. By comparing the photoconvertible phyD-YFP and phyB levels and their signaling in continuous and pulsed irradiation, we showed that phyD-YFP is a less efficient photoreceptor than phyB. This conclusion is supported by the facts that only a part of the phyD-YFP pool is photoconvertible and that thermal reversion of phyD-YFP is faster than that of phyB. Our data suggest that the temperature-dependent function of phyD is based on the amount of phyD protein and not on its Pfr stability, as described for phyB. We also found that phyD-YFP and phyB-GFP are associated with strongly overlapping genomic locations and are able to mediate similar changes in gene expression; however, the efficiency of phyD-YFP is lower. Based on these data, we propose that under certain conditions, synergistic interaction of phyD and phyC can substitute phyB function in seedlings and in adult plants and thus increases the ability of plants to respond more flexibly to environmental changes. ELONGATED HYPOCOTOYL5 (HY5) and PHYTOCHROME INTERACTING FACTORs (PIFs) are two types of important light-related regulators of plant growth, however, their interplay remains elusive. Here, we report that the activated tomato (Solanum lycopersicum) HY5 (SlHY5) triggers the transcription of a Calcium-dependent Protein Kinase SlCPK27. SlCPK27 interacts with and phosphorylates SlPIF4 at Ser-252 and Ser-308 phosphosites to promote its degradation. SlPIF4 promotes hypocotyl elongation mainly by activating the transcription of SlDWF, a key gene in brassinosteroid (BR) biosynthesis. Such a SlHY5-SlCPK27-SlPIF4-BR cascade not only plays a crucial role in photomorphogenesis but also regulates thermomorphogenesis. Our results uncover a previously unidentified mechanism that integrates Ca2+ signaling with the light signaling pathways to regulate plant growth by modulating BR biosynthesis in response to changes in ambient light and temperature. Light is one of the most essential environmental factors that tightly and precisely control various physiological and developmental processes in plants. B‐box CONTAINING PROTEINs (BBXs) play central roles in the regulation of light‐dependent development. In this study, we report that BBX9 is a positive regulator of light signaling. BBX9 interacts with the red light photoreceptor PHYTOCHROME B (phyB) and transcription factors PHYTOCHROME‐INTERACTING FACTORs (PIFs). phyB promotes the stabilization of BBX9 in light, while BBX9 inhibits the transcriptional activation activity of PIFs. In turn, PIFs directly bind to the promoter of BBX9 to repress its transcription. On the other hand, BBX9 associates with the positive regulator of light signaling, BBX21, and enhances its biochemical activity. BBX21 associates with the promoter regions of BBX9 and transcriptionally up‐regulates its expression. Collectively, this study unveiled that BBX9 forms a negative feedback loop with PIFs and a positive one with BBX21 to ensure that plants adapt to fluctuating light conditions. [ABSTRACT FROM AUTHOR]

Heat stress inhibits plant growth and productivity. Among the main regulators, B-box zinc-finger (BBX) proteins are well-known for their contribution to plant photomorphogenesis and responses to abiotic stress. Our research pinpoints that SlBBX31, a BBX protein harboring a conserved B-box domain, serves as a suppressor of plant growth and heat tolerance in tomato (Solanum lycopersicum L.). Overexpressing (OE) SlBBX31 in tomato exhibited yellowing leaves due to notable reduction in chlorophyll content and net photosynthetic rate (Pn). Furthermore, the pollen viability of OE lines obviously decreased and fruit bearing was delayed. This not only affected the fruit setting rate and the number of plump seeds but also influenced the size of the fruit. These results indicate that SlBBX31 may be involved in the growth process of tomato, specifically in terms of photosynthesis, flowering, and the fruiting process. Conversely, under heat-stress treatment, SlBBX31 knockout (KO) plants displayed superior heat tolerance, evidenced by their improved membrane stability, heightened antioxidant enzyme activities, and reduced accumulation of reactive oxygen species (ROS). Further transcriptome analysis between OE lines and KO lines under heat stress revealed the impact of SlBBX31 on the expression of genes linked to photosynthesis, heat-stress signaling, ROS scavenging, and hormone regulation. These findings underscore the essential role of SlBBX31 in regulating tomato growth and heat-stress resistance and will provide valuable insights for improving heat-tolerant tomato varieties. Red and blue light have significant effects on plant growth; however, most of the current studies have focused on common horticultural crops such as cucumber and tomato, and there are fewer studies on how red and blue light affect the growth of melon seedlings. Therefore, in this study, we used melon (Cucumis melo L.) as the experimental material to investigate the effects of red and blue light ratios on the photomorphogenesis and photosynthesis of melon seedlings. Five red and blue light ratios were set at a fixed light intensity 200 μmol·m−2·s−1, including R:B = 1:9, R:B = 3:7, R:B = 1:1, R:B = 7:3, and R:B = 9:1. The results showed that with the increase in red light ratios, melon seedling height, stem diameter, total leaf area and stomatal pore size of melon seedlings increased, while the upper epidermis, palisade tissue, spongy tissue, leaf thickness, and stomatal density showed a decreasing trend. Under the R:B = 7:3 treatment, melon seedlings were optimized in all morphological indexes and had higher photosynthetic efficiency; these results indicated that the growth of melon seedlings could be regulated by adjusting the ratio of red to blue light, thus promoting the morphogenesis of melon seedlings. The circadian clock is an endogenous oscillator, and its importance lies in its ability to impart rhythmicity on downstream biological processes, or outputs. Our knowledge of output regulation, however, is often limited to an understanding of transcriptional connections between the clock and outputs. For instance, the clock is linked to plant growth through the gating of photoreceptors via rhythmic transcription of the nodal growth regulators, PHYTOCHROME-INTERACTING FACTORs (PIFs), but the clock's role in PIF protein stability is less clear. Here, we identified a clock-regulated, F-box type E3 ubiquitin ligase, CLOCK-REGULATED F-BOX WITH A LONG HYPOCOTYL 1 (CFH1), that specifically interacts with and degrades PIF3 during the daytime. Additionally, genetic evidence indicates that CFH1 functions primarily in monochromatic red light, yet CFH1 confers PIF3 degradation independent of the prominent red-light photoreceptor phytochrome B (phyB). This work reveals a clock-mediated growth regulation mechanism in which circadian expression of CFH1 promotes sustained, daytime PIF3 degradation in parallel with phyB signaling. Transcription factor ELONGATED HYPOCOTYL5 (HY5) is the central hub for seedling photomorphogenesis. E3 ubiquitin (Ub) ligase CONSTITUTIVE PHOTOMORPHOGENIC 1 (COP1) inhibits HY5 protein accumulation through ubiquitination. However, the process of HY5 deubiquitination, which antagonizes E3 ligase-mediated ubiquitination to maintain HY5 homeostasis has never been studied. Here, we identified that Arabidopsis thaliana deubiquitinating enzyme, Ub-SPECIFIC PROTEASE 14 (UBP14) physically interacts with HY5 and enhances its protein stability by deubiquitination. The da3-1 mutant lacking UBP14 function exhibited a long hypocotyl phenotype, and UBP14 deficiency led to the failure of rapid accumulation of HY5 during dark to light. In addition, UBP14 preferred to stabilize nonphosphorylated form of HY5 which is more readily bound to downstream target genes. HY5 promoted the expression and protein accumulation of UBP14 for positive feedback to facilitate photomorphogenesis. Cyanobacteria play a key role in primary production in both oceans and fresh waters and hold great potential for sustainable production of a large number of commodities. During their life, cyanobacteria cells need to acclimate to a multitude of challenges, including shifts in intensity and quality of incident light. Despite our increasing understanding of metabolic regulation under various light regimes, detailed insight into fitness advantages and limitations under shifting light quality remains underexplored. Here, we study photo-physiological acclimation in the cyanobacterium Synechocystis sp. PCC 6803 throughout the photosynthetically active radiation (PAR) range. Using light emitting diodes (LEDs) with qualitatively different narrow spectra, we describe wavelength dependence of light capture, electron transport and energy transduction to main cellular pools. In addition, we describe processes that fine-tune light capture, such as state transitions, or the efficiency of energy transfer from phycobilisomes to photosystems (PS). We show that growth was the most limited under blue light due to inefficient light harvesting, and that many cellular processes are tightly linked to the redox state of the plastoquinone (PQ) pool, which was the most reduced under red light. The PSI-to-PSII ratio was low under blue photons, however, it was not the main growth-limiting factor, since it was even more reduced under violet and near far-red lights, where Synechocystis grew faster compared to blue light. Our results provide insight into the spectral dependence of phototrophic growth and can provide the foundation for future studies of molecular mechanisms underlying light acclimation in cyanobacteria, leading to light optimization in controlled cultivations. Light is one of the most important factors regulating plant gene expression patterns, metabolism, physiology, growth, and development. To explore how light may induce or alter transcript splicing, we conducted RNA-Seq-based transcriptome analyses by comparing the samples harvested as etiolated seedlings grown under continuous dark conditions vs. the light-treated green seedlings. The study aims to reveal differentially regulated protein-coding genes and novel long noncoding RNAs (lncRNAs), their light-induced alternative splicing, and their association with biological pathways. We identified 14,766 differentially expressed genes, of which 4369 genes showed alternative splicing. We observed that genes mapped to the plastid-localized methyl-erythritol-phosphate (MEP) pathway were light-upregulated compared to the cytosolic mevalonate (MVA) pathway genes. Many of these genes also undergo splicing. These pathways provide crucial metabolite precursors for the biosynthesis of secondary metabolic compounds needed for chloroplast biogenesis, the establishment of a successful photosynthetic apparatus, and photomorphogenesis. In the chromosome-wide survey of the light-induced transcriptome, we observed intron retention as the most predominant splicing event. In addition, we identified 1709 novel lncRNA transcripts in our transcriptome data. This study provides insights on light-regulated gene expression and alternative splicing in rice. Light quality can be stated to be the identity of an artificial light source, and although the response of light quality may vary depending on the crop, the effect is clearly visible and can produce various results depending on the combination of an artificial light source. This study investigated the spectral reflectance, photosynthetic performance, and chlorophyll fluorescence of mini green romaine lettuce based on light quality. Most parameters related to spectral reflectance showed the best results under blue light, and photosynthetic performance was more effective with mixed light than with single-colored light, among which blue + red (BR)-LED was the most suitable. Red light was ineffective, showing mostly low results in parameters of spectral reflectance and photosynthetic performance. In the case of chlorophyll fluorescence, the light quality influenced photomorphogenesis, resulting in increased leaf length and width with R- and quantum dot (QD)-LED, which expanded the leaf area and allowed for more external light to be captured (ABS/RC and TRo/RC). However, the ratio of electronized energy (ETo/RC) was low, and the amount of energy dissipated as heat (DIo/RC) was high. Consequently, the degree of electron acceptor reduction and overall photosynthetic performance (PIABS and PItotal) were lower compared to other light qualities. Additionally, the contrasting results of QD-LED and BR-LED were attributed to the form of red light and the presence or absence of far-red light when comparing spectra. Principal component analysis also clearly distinguished light qualities for photosynthesis and growth. Dye-sensitized solar cells (DSSCs) can be used as greenhouse-glazing materials because of their translucent properties, varied colors, and ability to produce electricity. To assess their effect on plant growth and physiological responses, Petunia ×hybrida 'Madness Red' was investigated under four different light treatments: transparent polycarbonate panels (T) as the control, T + 50% shade (S), red-colored DSSCs (D), and D + cool-white light-emitting diodes (LED) (D + L). Compared to T, S increased the leaf area and the number of days to flowering and decreased the number of flowers and relative chlorophyll content (SPAD). D increased the plant height, leaf area, fresh weight, and the number of days to flowering and reduced the SPAD and number of leaves and flowers due to a lower daily light integral (DLI) and blue light than T. Compared to S, D increased the plant height, leaf area, and fresh weight while reducing the number of leaves and flowers due to the lower blue light. D + L increased the number of leaves and flowers, flowering rate, dry weight, and SPAD but inhibited stem and leaf elongation compared to D, due to an increase in DLI and blue light. The deployment of red DSSCs in greenhouses could potentially induce unfavorable morphological and developmental responses in light-sensitive plants. Light-regulated developmental processes such as photomorphogenesis and flowering play important roles in the plant life cycle, from seedling emergence to reproduction. Three members of the Arabidopsis thaliana B-BOX DOMAIN PROTEIN (BBX) family, BBX14, BBX15, and BBX16 (hereafter BBX14/15/16), redundantly regulate flowering time, but whether this genetic redundancy also affects the regulation of photomorphogenesis remains unclear. Here, we show that light induces BBX14/15/16 expression primarily in the hypocotyl, where BBX14/15/16 redundantly repress hypocotyl elongation. PHYTOCHROME-INTERACTING FACTORs (PIFs) negatively regulate BBX14/15/16 expression mainly through GOLDEN-LIKE proteins (GLKs); however, analyses of ChIP-seq data showed that PIFs are recruited to the BBX14/15/16 loci and can also regulate these genes independently of GLKs. ELONGATED HYPOCOTYL 5 (HY5), a major regulator of photomorphogenesis, also directly binds to the BBX14/15/16 loci and regulates their expression. Simultaneous knockdown of BBX14/15/16 resulted in significant downregulation of HY5 and upregulation of PIFs, suggesting that these factors participate in a feedback regulatory loop. Indeed, BBX14/15/16 induced HY5 promoter activity by binding to the HY5 promoter. The brassinosteroid-responsive gene TOUCH4 (TCH4) and several auxin-responsive SMALL AUXIN UPREGULATED RNA (SAUR) genes were upregulated in the BBX14/15/16 knockdown plants, suggesting that auxin and brassinosteroids might participate in BBX14/15/16-mediated hypocotyl regulation. Mutating the predicted BBX-binding sites in SAUR4 and TCH4 impaired their regulation by BBX14/15/16. We propose that BBX14/15/16, together with HY5 and PIFs, form a feedback loop that regulates the expression of auxin- and brassinosteroid-related genes to modulate hypocotyl elongation.

Although far-red (FR) radiation (700–750 nm) is beyond the traditional photosynthetically active radiation (PAR) waveband (400–700 nm), recent findings suggest equivalent or similar photosynthetic action when applied along with shorter PAR wavelengths. Potential interactive effects of FR with other environmental factors that affect photosynthesis, such as CO2, have not been widely established. In the present study, the effects of four FR photon flux densities (0, 20, 40, or 60 µmol·m−2·s−1) substituting for red (R; 600–700 nm) in combination with three CO2 concentrations (400, 800, or 1200 µmol·mol−1) were investigated at three distinctive stages of young red lettuce production. We tested whether the photomorphogenic response of young plant leaves to FR would result in a higher light interception area, as well as whether interactive effects of CO2 with FR might modify the effects of FR alone. The total photon flux density (TPFD) was maintained at a low limiting intensity of 200 µmol·m−2·s−1 (daily light integral: 11.5 mol·m−2·d−1), consistent with indoor commercial setpoints for sole-source lighting. Although blue (B; 400–500 nm) was held constant at 20 µmol·m−2·s−1 for all treatments, different combinations of R and FR were adjusted. Red oakleaf lettuce (Lactuca sativa ‘Rouxai’) used as a model test crop was harvested at 14 days (small baby greens), 18 days (standard baby greens), and 22 days (teen greens) after sowing. Crop productivity was highest at 800 µmol·mol−1 CO2 for small and standard baby stages. At the teen green stage, plant productivity was similar for the two elevated CO2 concentrations. The FR substitution for R did not affect dry biomass accumulation at all CO2 concentrations tested, thus supporting equivalent photosynthetic action. A photomorphogenic effect of FR that elongated but did not increase the area of leaves persisted from the earliest to the most mature stage of crop development tested. The FR inclusion in the light recipe resulted in longer, thinner leaves with lower biomass compared to those without FR. At higher FR fluxes, purple pigmentation reduction occurred. However, the interaction of elevated CO2 with FR radiation counteracted purple pigment reduction caused by FR alone. Substitution of FR in the light recipe for indoor production of young ‘Rouxai’ lettuce at low limiting light intensity did not improve productivity, even in combination with elevated CO2.

Light is a fundamental environmental factor for living organisms on earth—not only as a primary energy source but also as an informational signal. In fungi, light can be used as an indicator for both time and space to control important physiological and morphological responses. Botrytis cinerea (B. cinerea) is a devastating phytopathogenic fungus that exploits light cues to optimize virulence and the balance between conidiation and sclerotia development, thereby improving its dispersal and survival in ecosystems. However, the components and mechanisms underlying these processes remain obscure. Here, we identify a novel light-signaling component in B. cinerea, BcCfaS, which encodes a putative cyclopropane fatty-acyl-phospholipid synthase. BcCfaS is strongly induced by light at the transcriptional level and plays a crucial role in regulating photomorphogenesis. Deletion of BcCfaS results in reduced vegetative growth, altered colony morphology, impaired sclerotial development, and enhanced conidiation in a light-dependent manner. Moreover, the mutant exhibits serious defects in stress response and virulence on the host. Based on a lipidomics analysis, a number of previously unknown fungal lipids and many BcCfaS-regulated lipids are identified in B. cinerea, including several novel phospholipids and fatty acids. Importantly, we find that BcCfaS controls conidiation and sclerotial development by positively regulating methyl jasmonate (MeJA) synthesis to activate the transcription of light-signaling components, revealing for the first time the metabolic base of photomorphogenesis in fungi. Thus, we propose that BcCfaS serves as an integration node for light and lipid metabolism, thereby providing a regulatory mechanism by which fungi adapt their development to a changing light environment. These new findings provide an important target for antifungal design to prevent and control fungal disease.

During the last decade, knowledge about BBX proteins has greatly increased. Genome-wide studies identified the BBX gene family in several ornamental, industry, and food crops; however, reports regarding the role of these genes as regulators of agronomically important traits are scarce. Here, by phenotyping a knockout mutant, we performed a comprehensive functional characterization of the tomato locus Solyc12g089240, hereafter called SlBBX20. The data revealed the encoded protein as a positive regulator of light signaling affecting several physiological processes during the life span of plants. Through inhibition of PHYTOCHROME INTERACTING FACTOR 4 (SlPIF4)–auxin crosstalk, SlBBX20 regulates photomorphogenesis. Later in development, it controls the balance between cell division and expansion to guarantee correct vegetative and reproductive development. In fruits, SlBBX20 is transcriptionally induced by the master transcription factor RIPENING INHIBITOR (SlRIN) and, together with ELONGATED HYPOCOTYL 5 (SlHY5), up-regulates flavonoid biosynthetic genes. Finally, SlBBX20 promotes the accumulation of steroidal glycoalkaloids and attenuates Botrytis cinerea infection. This work clearly demonstrates that BBX proteins are multilayer regulators of plant physiology because they affect not only multiple processes during plant development but they also regulate other genes at the transcriptional and post-translational levels. Coniferous forests represent one of the most valuable ecosystems worldwide, due to the ecological and economic services they provide. Pinus pseudostrobus Lindl., is one of the most promising species for its use in reforestation programs, so it was proposed in this work to evaluate the effect of different LED light spectra on the induction of photomorphogenic responses. To this end, the effects of different light sources (red LED, blue LED, red + blue LED, white LED, and fluorescent light) on the induction and characteristics of callus formed from transverse thin cell layers (tTLC) of hypocotyls of P. pseudostrobus in vitro plantlets were evaluated. The best results in terms of callus characteristics formed were obtained from hypocotyl segments grown on a WPM medium containing 1 mg L-1 of BAP and 3 mg L-1 of 2,4-D, under a blue LED light spectrum. With the increasing availability of LEDs, researchers in photobiology have easier access to customized light sources. However, the abundance of different light sources poses new challenges for the correct characterization of existing light conditions. The photobiological effect of a light source depends mainly on the number of photons involved and the spectral composition. However, light sources are mainly described by parameters such as radiant flux, dominant or peak wavelength, and correlated color temperature (CCT). Therefore, in this work, chromatic and white light sources were measured for their spectral composition, various characterization parameters were determined, and the resulting photon flux densities were calculated, focusing on dominant versus peak wavelength for chromatic LEDs and the CCT for white LEDs and fluorescent tubes. The use of the dominant wavelength is inappropriate as it is partly outside the actual spectral range. It was also shown that white light sources with the same CCT have significantly different spectral compositions and, therefore, may have different photobiological effects. The results of this work should serve as a basis for life scientists to better compare light sources, to correctly interpret existing parameters, and to describe light conditions in a standardized and comparable way. Coordinated morphogenic adaptation of growing plants is critical for their survival and propagation under fluctuating environments. Plant morphogenic responses to light and warm temperatures, termed photomorphogenesis and thermomorphogenesis, respectively, have been extensively studied in recent decades. During photomorphogenesis, plants actively reshape their growth and developmental patterns to cope with changes in light regimes. Accordingly, photomorphogenesis is closely associated with diverse growth hormonal cues. Notably, accumulating evidence indicates that light‐directed morphogenesis is profoundly affected by two recently identified phytochemicals, karrikins (KARs) and strigolactones (SLs). KARs and SLs are structurally related butenolides acting as signaling molecules during a variety of developmental steps, including seed germination. Their receptors and signaling mediators have been identified, and associated working mechanisms have been explored using gene‐deficient mutants in various plant species. Of particular interest is that the KAR and SL signaling pathways play important roles in environmental responses, among which their linkages with photomorphogenesis are most comprehensively studied during seedling establishment. In this review, we focus on how the phytochemical and light signals converge on the optimization of morphogenic fitness. Cr3+ doped phosphors with deep-red luminescence hold great promise as artificial lighting sources for plants growth. Among them, α-Al 2 O 3 :Cr3+ is featured with an ultra-intense pure zero phonon line (ZPL) emission with negligible sidebands. Nonetheless, further enhancements in its luminescence thermal stability are required to fully unlock its potential for practical applications. To tackle this challenge, we propose that Al 20 B 4 O 36 with a negative expansion coefficient could serve as a more favorable host materials for Cr3+ ion, thereby enhancing the luminescence thermal stability. Our first-principles calculations have unveiled similar electronic structures for α-Al 2 O 3 :Cr3+ and Al 20 B 4 O 36 :Cr3+, suggesting that the observed zero phonon R-line (2 E g) emission, arising from the 2 E g →4 A 2g transition in α-Al 2 O 3 :Cr3+, might also be present in Al 20 B 4 O 36 :Cr3+. Based on these findings, we prepared Al 20 B 4 O 36 :Cr3+ phosphors using a simple solid reaction method and confirmed that Al 20 B 4 O 36 :Cr3+ indeed exhibits a similar ultra-intense pure ZPL emission centered at 694 nm. Moreover, Al 20 B 4 O 36 :Cr3+ phosphors demonstrate remarkable properties including a high internal quantum efficiency (IQE) of 98.2 % and superior luminescence thermal stability compared to α-Al 2 O 3 :Cr3+. A phosphor converted light emitting diode (pc-LED) is developed by coating Al 20 B 4 O 36 :Cr3+ phosphor onto a near-ultraviolet LED chip. The lighting from this deep-red LED significantly impacts the photomorphogenesis of bean sprouts. Cryptochromes are widely dispersed flavoprotein photoreceptors that regulate numerous developmental responses to light in plants, as well as to stress and entrainment of the circadian clock in animals and humans. All cryptochromes are closely related to an ancient family of light-absorbing flavoenzymes known as photolyases, which use light as an energy source for DNA repair but themselves have no light sensing role. Here we review the means by which plant cryptochromes acquired a light sensing function. This transition involved subtle changes within the flavin binding pocket which gave rise to a visual photocycle consisting of light-inducible and dark-reversible flavin redox state transitions. In this photocycle, light first triggers flavin reduction from an initial dark-adapted resting state (FADox). The reduced state is the biologically active or 'lit' state, correlating with biological activity. Subsequently, the photoreduced flavin reoxidises back to the dark adapted or 'resting' state. Because the rate of reoxidation determines the lifetime of the signaling state, it significantly modulates biological activity. As a consequence of this redox photocycle Crys respond to both the wavelength and the intensity of light, but are in addition regulated by factors such as temperature, oxygen concentration, and cellular metabolites that alter rates of flavin reoxidation even independently of light. Mechanistically, flavin reduction is correlated with conformational change in the protein, which is thought to mediate biological activity through interaction with biological signaling partners. In addition, a second, entirely independent signaling mechanism arises from the cryptochrome photocycle in the form of reactive oxygen species (ROS). These are synthesized during flavin reoxidation, are known mediators of biotic and abiotic stress responses, and have been linked to Cry biological activity in plants and animals. Additional special properties arising from the cryptochrome photocycle include responsivity to electromagnetic fields and their applications in optogenetics. Finally, innovations in methodology such as the use of Nitrogen Vacancy (NV) diamond centers to follow cryptochrome magnetic field sensitivity in vivo are discussed, as well as the potential for a whole new technology of 'magneto-genetics' for future applications in synthetic biology and medicine. ICT1 is an Arabidopsis thaliana line that overexpresses the gene encoding the S30 ribosomal subunit, leading to tolerance to exogenous indole-3-carbinol. Indole-3-carbinol (I3C) is a protective chemical formed as a breakdown of I3M in cruciferous vegetables. The overexpression of S30 in ICT1 results in transcriptional changes that prime the plant for the I3C, or biotic insult. Emerging evidence suggests that ribosomal proteins play important extra-ribosomal roles in various biochemical and developmental processes, such as transcription and stress resistance. In an attempt to elucidate the mechanism leading to I3C and stress resistance in ICT1, and using a multi-pronged approach employing transcriptomics, metabolomics, phenomics, and physiological studies, we show that overexpression of S30 leads to specific transcriptional alterations, which lead to both changes in metabolites connected to biotic and oxidative stress tolerance and, surprisingly, to photomorphogenesis. Summary: ELONGATED HYPOCOTYL 5 (HY5), a bZIP‐type transcription factor, is a master regulator of light‐mediated responses. ELONGATED HYPOCOTYL 5 binds to the promoter of c. 3000 genes, thereby regulating various physiological and biological processes, including photomorphogenesis, flavonoid biosynthesis, root development, response to abiotic stress and nutrient homeostasis. In recent decades, it has become clear that light signaling plays a crucial role in promoting nutrient uptake and assimilation. Recent studies have revealed the molecular mechanisms underlying such encouraging effects and the crucial function of the transcription factor HY5, whose activity is regulated by many photoreceptors. The discovery that HY5 directly activates the expression of genes involved in nutrient uptake and utilization, including several nitrogen, iron, sulphur, phosphorus and copper uptake and assimilation‐related genes, enhances our understanding of how light signaling regulates uptake and utilisation of multiple nutrients in plants. Different light spectra from light-emitting diodes (LEDs) trigger species-specific adaptive responses in plants. We exposed Artemisia argyi (A. argyi) to four LED spectra: white (the control group), monochromatic red light (R), monochromatic blue light (B), or a mixture of R and B light of photon flux density ratio is 3 (RB), with equivalent photoperiod (14 h) and light intensity (160 μmol s−1 m−2). R light accelerated photomorphogenesis but decreased biomass, while B light significantly increased leaf area and short-term exposure (7 days) to B light increased total phenols and flavonoids. HPLC identified chlorogenic acid, 3,5-dicaffeoylquinic acid, gallic acid, jaceosidin, eupatilin, and taxol compounds, with RB and R light significantly accumulating chlorogenic acid, 3,5-dicaffeoylquinic acid, and gallic acid, and B light promoting jaceosidin, eupatilin, and taxol. OJIP measurements showed that B light had the least effect on the effective quantum yield ΦPSII, with higher rETR(II), Fv/Fm, qL and PIabs, followed by RB light. R light led to faster photomorphology but lower biomass than RB and B lights and produced the most inadaptability, as shown by reduced ΦPSII and enlarged ΦNPQ and ΦNO. Light is a crucial environmental factor that impacts various aspects of plant development. Phytochromes, as light sensors, regulate myriads of downstream genes to mediate developmental reprogramming in response to changes in environmental conditions. CONSTITUTIVELY PHOTOMORPHOGENIC 1 (COP1) is an E3 ligase for a number of substrates in light signaling, acting as a central repressor of photomorphogenesis. The interplay between phytochrome B (phyB) and COP1 forms an antagonistic regulatory module that triggers extensive gene expression reprogramming when exposed to light. Here, we uncover a role of COP1 in light-dependent chromatin remodeling through the regulation of VIL1 (VIN3-LIKE 1)/VERNALIZATION 5, a Polycomb protein. VIL1 directly interacts with phyB and regulates photomorphogenesis through the formation of repressive chromatin loops at downstream growth-promoting genes in response to light. Furthermore, we reveal that COP1 governs light-dependent formation of chromatin loop and limiting a repressive histone modification to fine-tune expressions of growth-promoting genes during photomorphogenesis through VIL1. Plants rely upon a diverse range of metabolites to control growth and development, and to overcome stress that results from suboptimal conditions. Karrikins (KARs) are a class of butenolide compounds found in smoke that stimulate seed germination and regulate various developmental processes in plants. KARs are perceived via a plant α/β-hydrolase called KARRIKIN INSENSITIVE2 (KAI2), which also functions as a receptor for a postulated phytohormone, provisionally termed KAI2 ligand (KL). Considered natural analogues of KL, KARs have been extensively studied for their effects on plant growth and their crosstalk with plant hormones. The perception and response pathway for KAR–KL signalling is closely related to that of strigolactones, another class of butenolides with numerous functions in regulating plant growth. KAR–KL signalling influences seed germination, seedling photomorphogenesis, root system architecture, abiotic stress responses, and arbuscular mycorrhizal symbiosis. Here, we summarize current knowledge of KAR–KL signalling, focusing on its role in plant development, its effects on stress tolerance, and its interaction with other signalling mechanisms. PHYTOENE SYNTHASE (PSY) is a rate-limiting enzyme catalysing the first committed step of carotenoid biosynthesis, and changes in PSY gene expression and/or protein activity alter carotenoid composition and plastid differentiation in plants. Four genetic variants of PSY (psy-4 , psy-90 , psy-130 , and psy-145) were identified using a forward genetics approach that rescued leaf virescence phenotypes and plastid abnormalities displayed by the Arabidopsis CAROTENOID ISOMERASE (CRTISO) mutant ccr2 (carotenoid and chloroplast regulation 2) when grown under a shorter photoperiod. The four non-lethal mutations affected alternative splicing, enzyme–substrate interactions, and PSY:ORANGE multi-enzyme complex binding, constituting the dynamic post-transcriptional fine-tuning of PSY levels and activity without changing localization to the stroma and protothylakoid membranes. psy genetic variants did not alter total xanthophyll or β-carotene accumulation in ccr2 , yet they reduced specific acyclic linear cis -carotenes linked to the biosynthesis of a currently unidentified apocarotenoid signal regulating plastid biogenesis, chlorophyll biosynthesis, and photomorphogenic regulation. ccr2 psy variants modulated the PHYTOCHROME-INTERACTING FACTOR 3/ELONGATED HYPOCOTYL 5 (PIF3/HY5) ratio, and displayed a normal prolamellar body formation in etioplasts and chlorophyll accumulation during seedling photomorphogenesis. Thus, suppressing PSY activity and impairing PSY:ORANGE protein interactions revealed how cis -carotene abundance can be fine-tuned through holoenzyme–metabolon interactions to control plastid development. It is already known that light quality and intensity have major influences on the growth, etiolation, germination, and morphology of many plant species, but there is limited information about the effect of wavelength and light intensity on nutrient absorption by plants. Therefore, this study was established to evaluate the plant growth, stomata formation, chlorophyll index, and absorption of macro- and micronutrients by common bean plants under six light treatments. The experimental design was completely randomized and consisted of six treatments: strong blue (blue LED at high light intensity); weak blue (blue LED at low light intensity); strong red (red LED at high light intensity); weak red (red LED at low light intensity; pink (combined red + blue LED), and white (combined red + white led). The stomatal density (stomata mm−2); the SPAD index; plant height (cm); root length (cm); plant dry weight (g); root dry weight (g); and the concentrations of N, S, K, Mg, Ca, B, Zn, Mn, and Fe on leaf analysis were influenced by all treatments. We found that plant photomorphogenesis is controlled not only by the wavelength, but also by the light intensity. Etiolation was observed in bean plants under blue light at low intensity, but when the same wavelength had more intensity, the etiolation did not happen, and the plant height was the same as plants under multichromatic lights (pink and white light). The smallest plants showed the largest roots, some of the highest chlorophyll contents, and some of the highest stomatal densities, and consequently, the highest dry weight, under white LED, showing that the multichromatic light at high intensity resulted in better conditions for the plants in carbon fixation. The effect of blue light on plant morphology is intensity-dependent. Plants under multichromatic light tend to have lower concentrations of N, K, Mg, and Cu in their leaves, but the final amount of these nutrients absorbed is higher because of the higher dry weight of these plants. Plants under blue light at high intensity tended to have lower concentrations of N, Cu, B, and Zn when compared to the same wavelength at low intensity, and their dry weight was not different from plants grown under pink light. Dye-sensitized solar cells (DSSCs) can be used as greenhouse glazing materials in agrivoltaic systems because they are translucent, have different colors, and can produce electricity. However, the light quality of DSSCs differs from that of sunlight, and the visible light transmittance is low. Therefore, we compared the plant shape, growth, and leaf color of coleus, a highly photosensitive plant, under transparent glass and red-colored DSSCs. Coleus 'Highway Rose' was grown in transparent (T, the control), shaded (S), and DSSC (D) chambers maintained at 23 ± 2 °C. The DSSC chambers were additionally illuminated with blue (B), green (G), white (W), B+G, and R+B+W light-emitting diodes (LEDs) (D+L) at 60 μmol·m−2·s−1 photosynthetic photon flux density for 15 h from 05:00 to 20:00. The coleus generally exhibited good growth under the T treatment. However, the light quality of DSSCs differed from that of sunlight, and the visible light transmittance decreased. Coleus exhibited increased growth and leaf color characteristics under the supplemental B lighting treatments (D+L(RBW), D+L(B), D+L(BG), and D+L(W)). Supplemental lighting with B LEDs using DSSCs improved plant morphology growth and leaf color. On the other hand, supplemental G lighting reinforced the shade avoidance syndrome. Summary: In leaves of C4 plants, the reactions of photosynthesis become restricted between two compartments. Typically, this allows accumulation of C4 acids in mesophyll (M) cells and subsequent decarboxylation in the bundle sheath (BS). In C4 grasses, proliferation of plasmodesmata between these cell types is thought to increase cell‐to‐cell connectivity to allow efficient metabolite movement. However, it is not known whether C4 dicotyledons also show this enhanced plasmodesmal connectivity and so whether this is a general requirement for C4 photosynthesis is not clear. How M and BS cells in C4 leaves become highly connected is also not known.We investigated these questions using 3D‐ and 2D‐electron microscopy on the C4 dicotyledon Gynandropsis gynandra as well as phylogenetically close C3 relatives.The M–BS interface of C4G. gynandra showed higher plasmodesmal frequency compared with closely related C3 species. Formation of these plasmodesmata was induced by light. Pharmacological agents that perturbed photosynthesis reduced the number of plasmodesmata, but this inhibitory effect could be reversed by the provision of exogenous sucrose.We conclude that enhanced formation of plasmodesmata between M and BS cells is wired to the induction of photosynthesis in C4G. gynandra. In smart greenhouse farming, the impact of light qualities on plant growth and development is crucial but lacks systematic identification of optimal combinations. This study addresses this gap by analysing various light properties' effects (photoperiod, intensity, ratio, light–dark order) on Arabidopsis thaliana growth using days-to-flower (DTF) and hypocotyl length as proxies to measure plant growth and development. After establishing suitable ranges through a comprehensive literature review, these properties varied within those ranges. Compared to white light, a 16-h cycle of blue light reduces DTF and hypocotyl length by 12 % and 3 %, respectively. Interestingly, similar results can be achieved using a shorter photoperiod of 14-h light (composed of 8 h of a mixture of 66.7 μ mol m − 2 s − 1 red and 800 μ mol m − 2 s − 1 blue lights (i.e. blue:red ratio of 12:1) followed by 6 h of monochromatic red light and 10-h dark. Phytochromes (phy) are distributed in various plant organs, and their physiological effects influence plant germination, flowering, fruiting, and senescence, as well as regulate morphogenesis throughout the plant life cycle. Reactive oxygen species (ROS) are a key regulatory factor in plant systemic responses to environmental stimuli, with an attractive regulatory relationship with phytochromes. With the development of high-throughput sequencing technology, omics techniques have become powerful tools, and researchers have used omics techniques to facilitate the big data revolution. For an in-depth analysis of phytochrome-mediated signaling pathways, integrated multi-omics (transcriptomics, proteomics, and metabolomics) approaches may provide the answer from a global perspective. This article comprehensively elaborates on applying multi-omics techniques in studying phytochromes. We describe the current research status and future directions on transcriptome-, proteome-, and metabolome-related network components mediated by phytochromes when cells are subjected to various stimulation. We emphasize the importance of multi-omics technologies in exploring the effects of phytochromes on cells and their molecular mechanisms. Photomorphogenic responses of etiolated seedlings include the inhibition of hypocotyl elongation and opening of the apical hook. In addition, dark-grown seedlings respond to light by the formation of adventitious roots (AR) on the hypocotyl. How light signaling controls adventitious rooting is less well understood. Hereto, we analyzed adventitious rooting under different light conditions in wild type and photomorphogenesis mutants in Arabidopsis thaliana. Etiolation was not essential for AR formation but raised the competence to form AR under white and blue light. The blue light receptors CRY1 and PHOT1/PHOT2 are key elements contributing to the induction of AR formation in response to light. Furthermore, etiolation-controlled competence for AR formation depended on the COP9 signalosome, E3 ubiquitin ligase CONSTITUTIVELY PHOTOMORPHOGENIC (COP1), the COP1 interacting SUPPRESSOR OF PHYA-105 (SPA) kinase family members (SPA1,2 and 3) and Phytochrome-Interacting Factors (PIF). In contrast, ELONGATED HYPOCOTYL5 (HY5), suppressed AR formation. These findings provide a genetic framework that explains the high and low AR competence of Arabidopsis thaliana hypocotyls that were treated with dark, and light, respectively. We propose that light-induced auxin signal dissipation generates a transient auxin maximum that explains AR induction by a dark to light switch.

Red and blue light have significant effects on plant growth; however, most of the current studies have focused on common horticultural crops such as cucumber and tomato, and there are fewer studies on how red and blue light affect the growth of melon seedlings. Therefore, in this study, we used melon (Cucumis melo L.) as the experimental material to investigate the effects of red and blue light ratios on the photomorphogenesis and photosynthesis of melon seedlings. Five red and blue light ratios were set at a fixed light intensity 200 μmol·m−2·s−1, including R:B = 1:9, R:B = 3:7, R:B = 1:1, R:B = 7:3, and R:B = 9:1. The results showed that with the increase in red light ratios, melon seedling height, stem diameter, total leaf area and stomatal pore size of melon seedlings increased, while the upper epidermis, palisade tissue, spongy tissue, leaf thickness, and stomatal density showed a decreasing trend. Under the R:B = 7:3 treatment, melon seedlings were optimized in all morphological indexes and had higher photosynthetic efficiency; these results indicated that the growth of melon seedlings could be regulated by adjusting the ratio of red to blue light, thus promoting the morphogenesis of melon seedlings.

Light quality can be stated to be the identity of an artificial light source, and although the response of light quality may vary depending on the crop, the effect is clearly visible and can produce various results depending on the combination of an artificial light source. This study investigated the spectral reflectance, photosynthetic performance, and chlorophyll fluorescence of mini green romaine lettuce based on light quality. Most parameters related to spectral reflectance showed the best results under blue light, and photosynthetic performance was more effective with mixed light than with single-colored light, among which blue + red (BR)-LED was the most suitable. Red light was ineffective, showing mostly low results in parameters of spectral reflectance and photosynthetic performance. In the case of chlorophyll fluorescence, the light quality influenced photomorphogenesis, resulting in increased leaf length and width with R- and quantum dot (QD)-LED, which expanded the leaf area and allowed for more external light to be captured (ABS/RC and TRo/RC). However, the ratio of electronized energy (ETo/RC) was low, and the amount of energy dissipated as heat (DIo/RC) was high. Consequently, the degree of electron acceptor reduction and overall photosynthetic performance (PIABS and PItotal) were lower compared to other light qualities. Additionally, the contrasting results of QD-LED and BR-LED were attributed to the form of red light and the presence or absence of far-red light when comparing spectra. Principal component analysis also clearly distinguished light qualities for photosynthesis and growth. Growth was increased by red (R)- and QD-LED, while photosynthetic performance was increased by BR- and blue (B)-LED.

Maize growth and development are regulated by light quality, intensity and photoperiod. Cryptochromes are blue/ultraviolet-A light receptors involved in stem elongation, shade avoidance, and photoperiodic flowering. To investigate the function of cryptochrome 1 (CRY1) in maize, where it is encoded by ZmCRY1, we obtained two ZmCRY1a genes (ZmCRY1a1 and ZmCRY1a2), both of which share the highest similarity with other gramineous plants, in particular rice CRY1a by phylogenetic analysis. In Arabidopsis, overexpression of ZmCRY1a genes promoted seedling de-etiolation under blue and white light, resulting in dwarfing of mature plants. In seedlings of the maize inbred line Zong 31 (ZmCRY1a-OE), overexpression of ZmCRY1a genes caused a reduction in the mesocotyl and first leaf sheath lengths due to down-regulation of genes influencing cell elongation. In mature transgenic maize plants, plant height, ear height, and internode length decreased in response to overexpression of ZmCRY1a genes. Expression of ZmCRY1a were insensitive to low blue light (LBL)-induced shade avoidance syndrome (SAS) in Arabidopsis and maize. This prompted us to investigate the regulatory role of the gibberellin and auxin metabolic pathways in the response of ZmCRY1a genes to LBL treatment. We confirmed a link between ZmCRY1a expression and hormonal influence on the growth and development of maize under LBL-induced SAS. These results reveal that ZmCRY1a has a relatively conservative function in regulating maize photomorphogenesis and may guide new strategies for breeding high density-tolerant maize cultivars.

Background. The study of tomato growth and development under light culture conditions is relevant for further technological improvement of crop cultivation on city farms. The use of closed intensive cultivation systems allows to control both the crop quality and yields. The purpose of the research was to analyze tomato physiological reactions and crop production process under different light conditions. Materials and methods. The determinant early-maturing tomato variety was grown in peat vegetative pots. In experiments, the effect of optical radiation on plants with various combinations of photoperiod and illumination intensity was studied. The daily light integral (DLI) at the installations was 9.5 and 19 mol/m2·day. Results.The DLI increase (light treatment12/440) stimulated biomass accumulation and acceleration of plant development. The light patterns of 18h-146 μkmol/ m2·s and 12h-440μmol/ m2·s contributed to earlier transition to generative development of plants. Comparing the effect of the same DLI (9.5 mol/m2·day) on plants morphogenesis it was noticed that tomatoes were more sensitive to light intensity changes that to the photoperiod. The patterns 18/146 and 12/440 may be prominent according to productivity indicators, high pigments content in green biomass and biochemical substances in fruits. Conclusions.The irradiation modes with DLI of 9.5 (light treatments12/220, 18/146) and 19 (light treatment12/440) mol/m2·day have similar effect on plant development. The best adaptation of the determinant tomato to irradiation conditions and the most efficient use of light energy were observed in 12-hour modes in comparison to other patterns.

Cryptochromes are widely dispersed flavoprotein photoreceptors that regulate numerous developmental responses to light in plants, as well as to stress and entrainment of the circadian clock in animals and humans. All cryptochromes are closely related to an ancient family of light-absorbing flavoenzymes known as photolyases, which use light as an energy source for DNA repair but themselves have no light sensing role. Here we review the means by which plant cryptochromes acquired a light sensing function. This transition involved subtle changes within the flavin binding pocket which gave rise to a visual photocycle consisting of light-inducible and dark-reversible flavin redox state transitions. In this photocycle, light first triggers flavin reduction from an initial dark-adapted resting state (FADox). The reduced state is the biologically active or ‘lit’ state, correlating with biological activity. Subsequently, the photoreduced flavin reoxidises back to the dark adapted or ‘resting’ state. Because the rate of reoxidation determines the lifetime of the signaling state, it significantly modulates biological activity. As a consequence of this redox photocycle Crys respond to both the wavelength and the intensity of light, but are in addition regulated by factors such as temperature, oxygen concentration, and cellular metabolites that alter rates of flavin reoxidation even independently of light. Mechanistically, flavin reduction is correlated with conformational change in the protein, which is thought to mediate biological activity through interaction with biological signaling partners. In addition, a second, entirely independent signaling mechanism arises from the cryptochrome photocycle in the form of reactive oxygen species (ROS). These are synthesized during flavin reoxidation, are known mediators of biotic and abiotic stress responses, and have been linked to Cry biological activity in plants and animals. Additional special properties arising from the cryptochrome photocycle include responsivity to electromagnetic fields and their applications in optogenetics. Finally, innovations in methodology such as the use of Nitrogen Vacancy (NV) diamond centers to follow cryptochrome magnetic field sensitivity in vivo are discussed, as well as the potential for a whole new technology of ‘magneto-genetics’ for future applications in synthetic biology and medicine.

Summary: The development of a seedling into a photosynthetically active plant is a crucial process. Despite its importance, we do not fully understand the regulatory mechanisms behind the establishment of functional chloroplasts.We herein provide new insight into the early light response by identifying the function of three basic region/leucine zipper (bZIP) transcription factors: bZIP16, bZIP68, and GBF1. These proteins are involved in the regulation of key components required for the establishment of photosynthetically active chloroplasts. The activity of these bZIPs is dependent on the redox status of a conserved cysteine residue, which provides a mechanism to finetune light‐responsive gene expression.The blue light cryptochrome (CRY) photoreceptors provide one of the major light‐signaling pathways, and bZIP target genes overlap with one‐third of CRY‐regulated genes with an enrichment for photosynthesis/chloroplast‐associated genes. bZIP16, bZIP68, and GBF1 were demonstrated as novel interaction partners of CRY1. The interaction between CRY1 and bZIP16 was stimulated by blue light. Furthermore, we demonstrate a genetic link between the bZIP proteins and cryptochromes as the cry1cry2 mutant is epistatic to the cry1cry2bzip16bzip68gbf1 mutant.bZIP16, bZIP68, and GBF1 regulate a subset of photosynthesis associated genes in response to blue light critical for a proper greening process in Arabidopsis. Introduction: Plant growth and greening in response to light require the synthesis of photosynthetic pigments such as chlorophylls and carotenoids, which are derived from isoprenoid precursors. In Arabidopsis, the pseudo-etiolated-in-light phenotype is caused by the overexpression of repressor of photosynthetic genes 2 (RPGE2), which regulates chlorophyll synthesis and photosynthetic genes. Methods: We investigated a homologous protein in the Russian dandelion (Taraxacum koksaghyz) to determine its influence on the rich isoprenoid network in this species, using a combination of in silico analysis, gene overexpression, transcriptomics and metabolic profiling. Results: Homology-based screening revealed a gene designated pseudo- etiolated-in-light-like (TkPEL-like), and in silico analysis identified a light-responsive G-box element in its promoter. TkPEL-like overexpression in dandelion plants and other systems reduced the levels of chlorophylls and carotenoids, but this was ameliorated by the mutation of one or both conserved cysteine residues. Comparative transcriptomics in dandelions overexpressing TkPEL-like showed that genes responsible for the synthesis of isoprenoid precursors and chlorophyll were downregulated, probably explaining the observed pale green leaf phenotype. In contrast, genes responsible for carotenoid synthesis were upregulated, possibly in response to feedback signaling. The evaluation of additional differentially expressed genes revealed interactions between pathways. Discussion: We propose that TkPEL-like negatively regulates chlorophyll- and photosynthesis-related genes in a light-dependent manner, which appears to be conserved across species. Our data will inform future studies addressing the regulation of leaf isoprenoid biosynthesis and photomorphogenesis and could be used in future breeding strategies to optimize selected plant isoprenoid profiles and generate suitable plant-based production platforms. Light-induced changes in miRNAs, morphogenesis, and photosynthetic processes in phytochrome-deficient mutant plants grown under different light qualities were studied. miRNA activity in many processes is regulated by phytochromes and phytochrome-interacting factors (PIFs). The reduced content of photoreceptors in phytochrome mutants affects the PIF-microRNA interaction. In plants grown under red light (RL) and white light (WL), the phenotype of phyb mutant was distorted; however, under blue light (BL) conditions, the phyb phenotype was normalized. The photosynthetic rates of both the mutants and wild type were higher under BL than under RL and WL. The expression of most studied miRNAs increased in phyaphyb mutants under BL conditions, which is probably one of the reasons for the normalization of the phenotype, the increase in PSII activity, and the photosynthetic rate. MicroRNAs under BL can partially improve photosynthesis and phenotype of the mutants, which indicates the conjugation of the functioning of phytochromes in miRNA formation.

With the increasing availability of LEDs, researchers in photobiology have easier access to customized light sources. However, the abundance of different light sources poses new challenges for the correct characterization of existing light conditions. The photobiological effect of a light source depends mainly on the number of photons involved and the spectral composition. However, light sources are mainly described by parameters such as radiant flux, dominant or peak wavelength, and correlated color temperature (CCT). Therefore, in this work, chromatic and white light sources were measured for their spectral composition, various characterization parameters were determined, and the resulting photon flux densities were calculated, focusing on dominant versus peak wavelength for chromatic LEDs and the CCT for white LEDs and fluorescent tubes. The use of the dominant wavelength is inappropriate as it is partly outside the actual spectral range. It was also shown that white light sources with the same CCT have significantly different spectral compositions and, therefore, may have different photobiological effects. The results of this work should serve as a basis for life scientists to better compare light sources, to correctly interpret existing parameters, and to describe light conditions in a standardized and comparable way.

Introducing plants in the design of biophilic indoor environments is fundamental for improving human health, well-being, and performance. Previous studies showed that the phenotype of the model plant Arabidopsis thaliana grown under LED-sourced CoeLux® lighting systems was characterized by low biomass production rates, a small leaf area, and a low lamina-to-petiole length ratio, suggesting the onset of a strong shade avoidance syndrome. Therefore, it is essential to identify new strategies to improve plant growth under these peculiar light conditions. In the present work, we investigated the effects of two growing media (i.e., low-fertility soil and soil-less substrate), solid and liquid fertilizers, manure, biochar, perlite, mirror reflection of light, and a 24 h photoperiod on A. thaliana plants growing under CoeLux® lighting systems at a light intensity of 30 μmol m−2s−1. We found that the biochar soil amendment to low-fertility soil increases both the above-ground plant biomass and leaf area. Furthermore, the application of a mirror behind the plants and a continuous photoperiod improves not only the biomass and the leaf area but also the lamina-to-petiole length ratio. The combination of different beneficial treatments can further boost plant growth in the low-intensity light environment characterizing the CoeLux® biophilic lighting systems. [ABSTRACT FROM AUTHOR]

In Arabidopsis, the photoreceptors phytochrome B (PhyB) and UV-B resistance 8 (UVR8) mediate light responses that play a major role in regulating photomorphogenic hypocotyl growth, but how they crosstalk to coordinate this process is not well understood. Here we report map-based cloning and functional characterization of an ultraviolet (UV)-B-insensitive, long-hypocotyl mutant, lh1 , and a wild-type-like mutant, lh2 , in cucumber (Cucumis sativus), which show defective CsPhyB and GA oxidase2 (CsGA20ox-2), a key gibberellic acid (GA) biosynthesis enzyme, respectively. The lh2 mutation was epistatic to lh1 and partly suppressed the long-hypocotyl phenotype in the lh1lh2 double mutant. We identified phytochrome interacting factor (PIF) CsPIF3 as playing a critical role in integrating the red/far-red and UV-B light responses for hypocotyl growth. We show that two modules, CsPhyB–CsPIF3–CsGA20ox-2–DELLA and CsPIF3–auxin response factor 18 (CsARF18), mediate CsPhyB-regulated hypocotyl elongation through GA and auxin pathways, respectively, in which CsPIF3 binds to the G/E-box motifs in the promoters of CsGA20ox-2 and CsARF18 to regulate their expression. We also identified a new physical interaction between CsPIF3 and CsUVR8 mediating CsPhyB-dependent, UV-B-induced hypocotyl growth inhibition. Our work suggests that hypocotyl growth in cucumber involves a complex interplay of multiple photoreceptor- and phytohormone-mediated signaling pathways that show both conservation with and divergence from those in Arabidopsis.
