Diffraction-limited imaging in epi-fluorescence microscopy remains a challenge when sample aberrations are present or when the region of interest rests deep within an inhomogeneous medium. Adaptive optics is an attractive solution albeit with limited field of view and requiring relatively complicated systems. Alternatively, reconstruction algorithms have been developed over the years to correct for aberrations. Unfortunately, purely postprocessing techniques tend to be ill-posed and provide only incremental improvements in image quality. Here, we report a computational optical approach using unknown speckle illumination and matched reconstruction algorithm to correct for aberrations and reach or surpass diffraction limited resolution. The data acquisition is performed by shifting an unknown speckle pattern with respect to the fluorescent object. Confocal and multiphoton microscopy are effective techniques to obtain high-contrast images of 2-D sections within bulk tissue. However, scattering limits their application to depths only up to ~1 millimeter. Multimode fibers make excellent ultrathin endoscopes that can penetrate deep inside the tissue with minimal damage. Here, we present Multiview Scattering Scanning Imaging Confocal (MUSSIC) Microscopy that enables high signal-to-noise ratio (SNR) imaging through a multimode fiber, hence combining the optical sectioning and resolution gain of confocal microscopy with the minimally invasive penetration capability of multimode fibers. The key advance presented here is the high SNR image reconstruction enabled by employing multiple coplanar virtual pinholes to capture multiple perspectives of the object, re-shifting them appropriately and combining them to obtain a high-contrast and high-resolution confocal image. The last decade has seen the development of a wide set of tools, such as wavefront shaping, computational or fundamental methods, that allow to understand and control light propagation in a complex medium, such as biological tissues or multimode fibers. A vibrant and diverse community is now working on this field, that has revolutionized the prospect of diffraction-limited imaging at depth in tissues. There are several methods to focus light behind a scattering medium, but very few use fluorescence light as feedback or can be used without access to the distal side of the scatterer. Among all the wave-front shaping techniques, retrieving the transmission matrix of a scattering material is the only one that allows for focusing on multiple spots after a single set of measurements. Here we propose a method to retrieve the transmission matrix of a scatterer using fluorescence light as feedback without access to the distal side. We present an ultra-thin hybrid imaging system based on an optical multimode fiber (MMF) and an optical fiber hydrophone that combines optical resolution photoacoustic and fluorescence microscopy. To control the illumination at the distal tip of the MMF, a digital micromirror device modulates the amplitude of the optical wavefront which is coupled into the MMF. A set of pre-calibrated speckle illuminations combined with a reconstruction algorithm enables photoacoustic and fluorescence imaging of samples located at the distal tip of the fiber with optical resolution determined by the numerical aperture.

Controlling the propagation and interaction of light in complex media has sparked major interest in the last few years. Unfortunately, spatial light modulation devices suffer from limited speed that precludes real-time applications such as imaging in live tissue. To address this critical problem we introduce a phase-control technique to characterize complex media based on the use of fast 1D spatial light modulators and a 1D-to-2D transformation performed by the same medium being analyzed. We implement the concept using a micro-electro-mechanical grating light valve (GLV) with 1088 degrees of freedom modulated at 350 KHz, enabling unprecedented high-speed wavefront measurements. We continuously measure the transmission matrix, calculate the optimal wavefront and project a focus through various dynamic scattering samples in real-time, all within 2.4 ms per cycle. These results improve prior wavefront shaping modulation speed by more than an order of magnitude and open new opportunities for optical processing using 1D-to-2D transformations., Comment: 8 pages, 5 figures

A major open challenge in neuroscience is the ability to measure and perturb neural activity in vivo from well-defined neural sub-populations at cellular resolution anywhere in the brain. However, limitations posed by scattering and absorption prohibit non-invasive (surface) multiphoton approaches for deep (>2mm) structures, while Gradient Refreactive Index (GRIN) endoscopes are thick and cause significant damage upon insertion. Here, we demonstrate a novel microendoscope to image neural activity at arbitrary depths via an ultrathin multimode optical fiber (MMF) probe that is 5-10X thinner than commercially available microendoscopes. We demonstrate micron-scale resolution, multispectral and volumetric imaging. In contrast to previous approaches, we show that this method has an improved acquisition speed that is sufficient to capture rapid neuronal dynamics in-vivo in rodents expressing a genetically encoded calcium indicator. Our results emphasize the potential of this technology in neuroscience applications and open up possibilities for cellular resolution imaging in previously unreachable brain regions.

Recent remarkable progress in wave-front shaping has enabled control of light propagation inside linear media to focus and image through scattering objects. In particular, light propagation in multimode fibers comprises complex intermodal interactions and rich spatiotemporal dynamics. Control of physical phenomena in multimode fibers and its applications is in its infancy, opening opportunities to take advantage of complex mode interactions. In this work, we demonstrate a wave-front shaping approach for controlling nonlinear phenomena in multimode fibers. Using a spatial light modulator at the fiber input and a genetic algorithm optimization, we control a highly nonlinear stimulated Raman scattering cascade and its interplay with four wave mixing via a flexible implicit control on the superposition of modes that are coupled into the fiber. We show for the first time versatile spectrum manipulations including shifts, suppression, and enhancement of Stokes and anti-Stokes peaks. We report a method to design at will the spatial profile of transmitted coherent light after propagation through a scattering sample. We compute an operator based on the experimentally measured transmission matrix, obtained by numerically adding an arbitrary mask in the Fourier domain prior to focusing. We demonstrate the strength of the technique through several examples: propagating Bessel beams, thus generating foci smaller than the diffraction limited speckle grain, donut beams, and helical beams. We characterize the 3D profile of the achieved foci and analyze the fundamental limitations of the technique. Our approach generalizes Fourier optics concepts for random media, and opens in particular interesting perspectives for super-resolution imaging through turbid media., Comment: 6 pages, 5 figures

Imaging through opaque, highly scattering walls is a long sought after capability with potential applications in a variety of fields. The use of wavefront shaping to compensate for scattering has brought a renewed interest as a potential solution to this problem. A key to the practicality of any imaging technique is the capability to focus light without direct access behind the scattering wall. Here, we address this problem using photoacoustic feedback for wavefront optimization. By combining the spatially non-uniform sensitivity of the ultrasound transducer to the generated photoacoustic waves with an evolutionary competition among optical modes, the speckle field develops a single, high intensity focus significantly smaller than the acoustic focus used for feedback. Notably, this method is not limited by the size of the absorber to form a sub-acoustic optical focus. We demonstrate imaging behind a scattering medium with up to ten times improvement in signal-to-noise ratio (SNR) and five to six times sub-acoustic resolution. The results presented here open up opportunities for 3D imaging applications in the diffusive regime beyond the sound wave diffraction-limited resolution, including medical imaging, photothermal therapy, neuroscience, and optogenetics., Comment: 9 pages, 4 figures

When a single molecule is detected in a wide-field microscope, the image approximates the point spread function of the system. However, as the distribution of molecules becomes denser and their images begin to coincide, existing solutions to determine the number of molecules present and their precise three-dimensional locations can tolerate little to no overlap. A solution to this problem involving matched optical and digital techniques, as here proposed, is critical to increase the allowable labeling density and to accelerate single-molecule localization microscopy., Comment: 4 pages, 4 figures

We introduce a system capable of focusing light through a multimode fiber in 37ms, one order of magnitude faster than demonstrated in previous reports. As a result, the focus spot can be maintained during significant bending of the fiber, opening numerous opportunities for endoscopic imaging and energy delivery applications. We measure the transmission matrix of the fiber by projecting binary-amplitude computer generated holograms using a digital micromirror device and a field programmable gate array controller. The system shows two orders of magnitude enhancements of the focus spot relative to the background.
