23. Effect of bioaugmentation on long-term biodegradation of diesel/biodiesel blends in soil microcosms

Abstract

We studied long-term (64.5 weeks) biodegradation of diesel fuel, diesel/biodiesel blends (B10-B90) and biodiesel fuels in urban soil microcosms containing indigenous microorganisms, or indigenous microorganisms augmented with a hydrocarbon-degrading bacterial community. Mineralization extent (mmol of CO2 per day) of B10-B30 blends was smaller compared with diesel fuel at both short- (28 days) and long-term (109 days), and increased with biodiesel content. Priming with hydrocarbon degraders accelerated mineralization in the short-term (by up to 140%), with highest influence using blends with lower biodiesel content, but did not significantly influence kinetics and mineralization extent in the long-term. Although the biodiesel fraction was degraded completely within 64.5 weeks, 3–12% of the total aromatic and aliphatic hydrocarbons remained in the microcosms. Barcoded 16S rRNA gene MiSeq sequencing analysis revealed a significant effect of blend type on the community structure, with a marked enrichment of Sphingobacteriia and Actinobacteria classes. However, no significant influence was determined in the long-term, suggesting that the inoculated bacterial community may not have survived. Our findings show that biodiesel is preferentially degraded in urban soil and suggest that the value of bioaugmentation for bioremediating biodiesel fuels with hydrocarbon-degrading bacteria is limited to short-term exposures to lower (B10-B30) blends.

Abstract

Little is known about the effect of ionic liquids (ILs) on the structure of soil microbial communities and resulting biodiversity. Therefore, we studied the influence of six trihexyl(tetradecyl)phosphonium ILs (with either bromide or various organic anions) at sublethal concentrations on the structure of microbial community present in an urban park soil in 100-day microcosm experiments. The biodiversity decreased in all samples (Shannon's index decreased from 1.75 down to 0.74 and OTU's number decreased from 1399 down to 965) with the largest decrease observed in the microcosms spiked with ILs where biodegradation extent was higher than 80%. (i.e. [P66614][Br] and [P66614][2,4,4]). Despite this general decrease in biodiversity, which can be explained by ecotoxic effect of the ILs, the microbial community in the microcosms was enriched with Gram-negative hydrocarbon-degrading genera e.g. Sphingomonas. It is hypothesized that, in addition to toxicity, the observed decrease in biodiversity and change in the microbial community structure may be explained by the primary biodegradation of the ILs or their metabolites by the mentioned genera, which outcompeted other microorganisms unable to degrade ILs or their metabolites. Thus, the introduction of phosphonium-based ILs into soils at sub-lethal concentrations may result not only in a decrease in biodiversity due to toxic effects, but also in enrichment with ILs-degrading bacteria.

Abstract

Knowledge about biodegradability of ionic liquids (ILs) in terrestrial systems is limited. Here, using urban park soil microcosms spiked with either ammonium- or phosphonium-based ILs [didecyldimethylammonium 3-amino-1,2,4-triazolate, benzalkonium 3-amino-1,2,4-triazolate, trihexyl(tetradecyl)phosphonium chloride, or trihexyl(tetradecyl)phosphonium 1,2,4-triazolate], we studied their (i) 300-day primary biodegradation, and (ii) influence on CO2 evolution from the microcosms. The primary biodegradation ranged from 21 to 33% of total compound in the dissolved phase. The evolution of CO2 from spiked microcosms was either lower or within the range of background soil respiration, indicating no or small mineralization of the parent compounds and/or their metabolites, and their negligible or small toxicity to soil microorganisms. Our results suggest the potential for persistence of the four studied ILs in urban park soils. •Primary, 300-day biodegradation ranged from 21 to 33%.•CO2 evolution from the spiked soils was within the range of background respiration.•The studied ILs show potential for long-term persistence in urban park soils.

Abstract

BACKGROUND Knowledge regarding microaerophilic and anaerobic specific spoilage organisms (SSOs) is crucial for an appropriate evaluation of vacuum-packed ham. The objective of this study was to characterize the SSO community in vacuum-packed ham by a culture-dependent technique and MiSeq next-generation sequencing (NGS) platform. The relation between changes among the SSO group in the ham and changes in sensory characteristics of the product was also assessed. RESULTS In the study, conventional microbiological analyses were employed in order to establish the participation of several groups of microorganisms in the deterioration of vacuum-packed ham. The diversity of the SSO group in the product was further assessed with the use of MiSeq NGS technology. The bacteria identified in sliced cooked ham belonged mostly to four phyla, namely Actinobacteria, Proteobacteria, Firmicutes and Bacteroidetes. A temperature of 4 °C favoured the development of mesophilic and psychrophilic/psychrotrophic flora, mainly Lactobacillaceae, Enterobacteriaceae and Micrococcaceae families. A high ratio of Brochothrix thermosphacta species and new, cold-tolerant Clostridium spp. was also observed. The growth of these microorganisms facilitated changes in the pH value and organoleptic characteristics of the product. 42. Screening and Identification of TrichodermaStrains Isolated from Natural Habitats with Potential to Cellulose and Xylan Degrading Enzymes Production

Abstract

A total of 123 Trichodermastrains were isolated from different habitats and tested for their ability to degrade cellulose and xylan by simple plate screening method. Among strains, more than 34 and 45% respectively, exhibited higher cellulolytic and xylanolytic activity, compared to the reference strain T. reeseiQM 9414. For strains efficiently degrading cellulose, a highest enzyme activity was confirmed using filter paper test, and it resulted in a range from 1.01 to 7.15 FPU/ml. Based on morphological and molecular analysis, the isolates were identified as Trichoderma. The most frequently identified strains belonged to Trichoderma harzianumspecies. Among all strains, the most effective in degradation of cellulose and xylose was T. harzianumand T. virens, especially those isolated from forest wood, forest soil or garden and mushroom compost. The results of this work confirmed that numerous strains from the Trichodermaspecies have high cellulose and xylan degradation potential and could be useful for lignocellulose biomass conversion e.g. for biofuel production.

Abstract

The aim of the study was to optimize a saline brine denitrification by halophilic microorganisms Haloferax mediterranei. To define the optimum parameters of the process, the Latin square method was used (Statistica 6.0). The highest specific nitrate reduction rate was observed at a temperature of 37 °C and pH 7.0. There was no significant effect of NaCl on nitrate ion reduction at the concentration range from 2.5 to 3.5 M. During brine denitrification in a batch bioreactor the rates of specific nitrate reduction and nitrate reduction reached 100.5 mg N-NO3- per 1 gram of dry mass during 24 h and 135 mg N-NO3/dm3·day, respectively.
