Osteoporosis can affect a significant part of the population and fractures are the most common complications associated with this disease, leading to high public health costs. Thus, the prevention of fractures is relevant to individuals with signs and symptoms as well as to the health system. Postmenopausal osteoporosis has been associated with oxidative stress, emphasizing the importance of an efficient defense system to maintain bone health. Lycopene is a carotenoid with antioxidant properties that may stimulate osteoblastogenesis and inhibit osteoclastogenesis. The purpose of this investigation was to analyze the influence of lycopene in the bone neoformation of calvaria defects in ovariectomized rats utilizing the concentration of 45 mg/kg. Wistar Hannover female rats were divided into ovariectomized and sham groups. The ovariectomized animals received 45 mg/kg lycopene (OvxL) or water (Ovx) by daily gavage the day after ovariectomy/sham surgery for 16 weeks. Twelve weeks after ovariectomy, there were performed 5-mm calvaria defects followed by euthanasia after 4 weeks. Samples of bone tissue were collected to perform morphological and morphometrical analysis of the neoformed bone area, and percentage with Software Image J. Morphological evaluation showed mature bone with more osteocytes in the group OVxL when compared to the other groups. SUMMARY: Impairing osteoporosis progression is a challenge, and recently the role of antioxidants has been associated to bone metabolism. Green tea extract is rich in catechins, especially epigallocatechin gallate (EGCG), which may help control osteoporosis damage in bone tissue. This investigation evaluated the efficacy of green tea ingestion containing different concentrations of EGCG in calvaria bone repair of ovariectomized rats. Wistar rats (n=15) were ovariectomized and divided into 3 groups: ovariectomized (OVX), ovariectomized + GTE 15 % EGCG (OVX/GTE15), and ovariectomized + GTE 94 % EGCG (OVX/GTE94). OBJETIVO: descrever a caracterização histológica e radiográfica do método de indução da degeneração do disco intervertebral da cauda de ratos Wistar induzida por meio de punção. MÉTODOS: ratos Wistar machos adultos foram anestesiados, radiografados e submetidos à punção dos discos intervertebrais localizados entre a sexta e a sétima e a oitava e nona vértebras coccígeas. Para a punção foi utilizada agulha de 20G, que foi introduzida até o ânulo fibroso, e foi realizada dupla rotação de 360º, mantendo-se a mesma posição durante 30 segundos antes da retirada. O disco intermediário aos segmentos lesados (7-8) não foi puncionado e foi utilizado como controle. Foi selecionado o período pós-lesão de 30 dias (n=9) para sacrifício e análise dos discos intervertebrais. Os animais foram radiografados 30 dias após a lesão para análise da altura do disco intervertebral. Os segmentos da cauda foram removidos, fixados e desmineralizados, processados e corados com Hematoxilina-Eosina para avaliação histológica. RESULTADOS: a análise radiográfica revelou a redução significativa da altura dos discos lesados em relação ao controle. A avaliação histológica revelou alterações no núcleo pulposo e ânulo fibroso dos discos lesados em relação ao controle. Não foram observadas diferenças na intensidade de lesão entre os discos proximal e distal. CONCLUSÃO: a degeneração do disco intervertebral da cauda de ratos Wistar induzida por meio de punção mostrou ser método reprodutível para estudo da degeneração do disco intervertebral. Esse modelo mostrou validade para avaliação experimental de novas intervenções terapêuticas nos processos de degeneração do disco intervertebral. OBJECTIVE: to report the induction of intervertebral disc degeneration of the rat caudal spine by needle puncture and its radiographic and histologic characterization. METHODS: adult male Wistar rats were anesthetized, submitted to the X-Ray and then to the needle puncture (20G) of intervertebral disc between the sixth and seventh (proximal segment) and the eighth and ninth (distal segment) coccygeal vertebrae. Radiographies were taken 30 days after lesion for analysis of intervertebral disc height. The intermediate disc (7-8) to injured segments was not punctured and was considered as control. All segments were removed, fixed and demineralized, processed and stained with Hematoxylin-Eosin for histological evaluation. RESULTS: radiographic analysis revealed significant reduction in disc height of lesioned discs compared to control. Similarly, histological analysis revealed significant changes in the nucleus pulposus and annulus fibrosus of the lesioned discs (proximal and distal) relative to the control. There was no difference in the intensity of injury between the proximal and distal discs. CONCLUSION: the experimental model of tail intervertebral disc degeneration by needle puncture reproduced the steps of the intervertebral disc degeneration, assessed by different instruments, and it can be used for experimental evaluation of new therapeutic interventions for intervertebral disc degeneration process. MÉTODOS: ratas Wistar machos adultos fueron anestesiados, radiografiados y sometidos a la punción de los discos intervertebrales localizados, entre la sexta y la séptima; y la octava y novena vértebras coccígeas. Para la punción, fue utilizada una aguja de 20G, que fue introducida hasta el ánulo fibroso, y fue realizada una dupla rotación de 360º, manteniendo esta posición durante 30 segundos, previamente a la retirada. El disco intermediario a los segmentos lesionados (7-8) no fue puncionado y fue considerado como control. Fue seleccionado el periodo post-lesión de 30 días (n=9) para sacrificio y análisis de los discos intervertebrales. Los animales fueron radiografiados 30 días después de la lesión para análisis de la altura del disco intervertebral. Los segmentos de la cola fueron removidos, fijados y desmineralizados, procesados y coloreados con hematoxilina-eosina para evaluación histológica. RESULTADOS: el análisis radiográfico mostró una reducción significativa de la altura de los discos lesionados en relación al control. La evaluación histológica mostró alteraciones en el núcleo pulposo y el ánulo fibroso de los discos lesionados en relación al control. No fueron observadas diferencias en la intensidad de la lesión entre los discos proximal y distal. CONCLUSIÓNES: la degeneración del disco intervertebral de la cola de ratas Wistar inducida por medio de punción mostró ser un método reproducible para el estudio de la degeneración del disco intervertebral. Ese modelo mostró validez para la evaluación experimental de nuevas intervenciones terapéuticas en los procesos de la degeneración del disco intervertebral.

Neste trabalho utilizou-se seis tartarugas Phrynops hilarii, originárias de ilhas do estuário do rio Guaíba Porto Alegre (RS), que estavam ambientadas em nosso biotério. A coleta de sangue foi realizada em todos os períodos sazonais, por punção de vasos laterais do pescoço e coletados em tubos de ensaio heparinizados. Foram realizados esfregaços sanguíneos, corados com Leishmann e Giemsa, contando-se quinhentas células de cada animal e após a obtenção dos dados, foi aplicado o teste estatístico de Bonferroni. Para a análise autorradiográfica foi injetado 1000μCi / kg de thymidine-H A. Para microscopia eletrônica processamos a nata leucocitária obtida por meio de centrifugação do sangue, para a analise citoquímica incubamos com citidina-5'-monofosfato, betaglicerofosfato de sódio, Trimetafosfatase, para averiguar a resposta fagocitária utilizamos 0,01% de carvão coloidal.Os resultados mostram que os leucócitos de Phrynops hilarii tem descrição de leucócitos semelhantes às outras espécies, somente os basófilos e linfócitos não sofreram alterações em sua distribuição sazonal. Todos os leucócitos com exceção dos basófilos apresentaram renovação celular após sete dias. Caracterizamos monoblasto, promonócito, monócitos e macrófago no sangue circulante bem como a capacidade dos fagócitos mononucleares de fagocitar células mortas e materiais inerte. The aim of this study was to analyze the leukocytes in the blood using electronic and light microscopy and their seasonal distribution, also to characterize the leukocyte cells replacement and mainly to characterize the mononuclear phagocytes in the blood and their phagocytic capacity. In this study, it was used six turtles (Phrynops hilarii), caught at the Guaíba river estuary, Porto Alegre, Rio Grande do Sul, Brazil and lodged for 1 week at the Central Animal House, University of São Paulo, Ribeirão Preto, São Paulo, Brazil. Blood was obtained during the seasonal periods by puncturing the lateral vessels of the neck. The blood samples were stained by Leishmann and Giemsa, counting five hundred cells in each animal. After the obtained data, it was applied the Bonferroni test as statistical method. For the autoradiographic analysis, it was injected in the circulating blood 1000 μCi/kg of 3Hthymidine. For electronic microscopy, it was processed the leukocyte substrate by circulating blood centrifugation. For cytochemical analyses, blood smears were air dried, post-fixed in 4% formalin and submitted to the determination of the following enzyme activities: acid phosphatases (β-g1ycerophosphatase and citidine-5′-sodium monophosphatase), and trimetaphosphatase. The results showed that the leukocytes of Phrynops hilarii have the leukocytes description similar to the other species, only the basophiles and lymphocytes did not suffer alterations in their seasonal distribution. All the leukocytes, in exception of the basophiles showed cells replacement after seven days. It was characterized the monocytes and macrophages in the circulating blood as well as the phagocytes capacity.

The aim of this study was to present a possible carrier for MTA, monoolein gel, with the objective to maintain this material in the place that was inserted and verify if this procedure is able to optimize its action. The data were evaluated by histomorphometric method and submitted to statistical analysis. The histological responses observed in this study indicate that the MTA is a reliable material and should be considered effective in bone periapical defects and the monoolein gel was capable to maintain the MTA in situ El objetivo de este estudio fue presentar el gel de monoleína como un posible cargador para el MTA y verificar si es capaz de optimizar su acción. The aim of this article is to show the clinical and histochemical alterations of the first periodontal ligament, on the right side, after upper molars teeth extraction on the left side in gerbils. After two months, the periodontal ligaments were removed and processed for histochemical analysis. The data showed that TRAP reaction was able to evidence the osteoclastic activity in the hyperfunction hemimandible, right side, explaining the functional changes in the periodontal ligament after teeth extraction, and a little gingival recession and radicular exposure of teeth without function was observed at inferior molars of the left side El objetivo de este artículo es mostrar las alteraciones clínicas e histoquímicas del primer ligamento periodontal del lado derecho, después de la extracción del molar superior izquierdo en gerbiles {Meriones unguiculatus). Luego de dos meses, los ligamentos periodontales fueron retirados y procesados para el análisis histoquímico. Los resultados mostraron que la reacción de TRAP es capaz de evidenciar la actividad osteoclástica en la hiperfunción de la semimandíbula derecha, explicando los cambios funcionales del ligamento periodontal después de la extracción dental, siendo observada una pequeña recesión gingival y exposición radicular de los dientes sin función, en los molares inferiores izquierdos
