Abstract

Although the production of NO within rodent phagocytes is well-characterized, its production and function within human phagocytes are less clear. We show here that neutrophils within human buffy coat preparations stimulated with a mixture of interleukin 1, tumor necrosis factor [Alpha], and interferon [Gamma] contain inducible NO synthase mRNA and protein, one of the enzymes responsible for NO production. The protein colocalizes with myeloperoxidase within neutrophil primary granules. Using an inhibitor of NO synthase, L-N-monomethyl arginine, we show that activity of this enzyme is required for the formation of nitrotyrosine around phagocytosed bacteria, most likely through the intermediate production of peroxynitrite, a reaction product of NO and superoxide anions.

Abstract

Immunocytochemistry and Northern blotting during bidirectional regulation of osteoclasts function by nitric oxide, shows the presence of the constitutive calcium-sensitive nitric oxide synthase (cNOS) isoform in normal rat osteoclasts, and in human preosteoclast cell line. The presence of inducible (I) NOS isoform was also demonstrated. A basal level of transcript was observed in human preosteoclast line. Osteoclasts function may require continuous increases in NO production by cNOS against a basal inhibitory background activity of the iNOS isoform.

Abstract

An experiment was conducted to study the pattern of expression of the tissue-specific splicing protein SmN in the heart and brain of rodents. Analysis of tissue samples showed increased levels of SmN during brain development but not during development of the heart. In a subsequent experiment, it was shown that the high level of SmN corresponds to the abundance of AmN mRNA in the brain tissue. Abstract

Oncogenes are genes that make proteins (oncoproteins) that have the potential for causing cancer. An oncogene may come from a normal gene that has been damaged by X-rays, ultraviolet light, certain toxic chemicals, or other source of genetic mutation. Many different types of human tumors have been found to contain oncogenes and their associated oncoproteins. In some cases, prognosis has been related to the presence of oncoproteins in certain types of tumors. The oncogene called c-erbB-2 has been identified in some breast tumors, as well as other tumors, but has not been identified previously in endocrine tumors (tumors in the adrenal gland, pancreas, thyroid gland and lung). To determine if oncogenes are present in endocrine tumors, a special staining procedure using antibodies (immunoreactivity) was undertaken to screen tumors for several different types of oncoproteins (c-erbB-2, c-myc, L-myc, and N-myc). Seventy-one lung tumors (44 small cell carcinomas, 15 atypical and 12 bronchial) 28 thyroid tumors (medullary thyroid carcinoma), 27 adrenal gland tumors (pheochromocytomas), and 17 pancreas tumors (insulinomas) were tested. Oncoproteins from the c-erbB-2 oncogene were found in 15 percent of the lung tumors and in 22 percent of the adrenal tumors. Oncoproteins from the c-myc oncogene were found only in the lung tumors (32 percent), N-myc in thyroid tumors (21 percent), and L-myc in adrenal tumors (15 percent). It is concluded that different types of oncoproteins may be associated with different types of tumors, and that this may be useful for identifying different subtypes of endocrine tumors. Abstract

C-terminal alpha-amidation is a post-translational modification necessary for the biological activity of many regulatory peptides produced in the respiratory tract. This modification is a two-step process catalyzed by two separate enzyme activities, both derived from the peptidyl-glycine alpha-amidating mono-oxygenase (PAM) precursor. The distribution of these two enzymes, peptidyl-glycine alpha-hydroxylating monoxygenase (PHM) and peptidyl-alpha-hydroxyglycine a amidating lyase (PAL), was studied in the normal lung and in lung tumors using immunocytochemical methods and in situ hybridization. In normal lung the enzymes were located in some cells of the airway epithelium and glands, the endothelium of blood vessels, some chondrocytes of the bronchial cartilage, the alveolar macrophages, smooth muscle cells, neurons of the intrinsic ganglia, and in myelinated nerves. A total of 24 lung tumors of seven different histological types were studied. All cases contained PAM-immunoreactive cells with various patterns of distribution. All immunoreactive cells were positive for the PHM antiserum but only some of them for the PAL antiserum. The distribution of PAM co-localizes with some other previously described amidated peptides, suggesting that amidation is an important physiological process taking place in the normal and malignant human lung tissue.

Abstract

Endothelin-1-like immunoreactivity has been found in endocrine cells of the midgut of the desert locust Locusta migratoria. Several antisera have been directed against the whole molecule and its C-terminal sequence. Endothelin-1-immunoreactive cells are present in the main region of the midgut (ventriculus) and in the midgut caeca but not in the ampullae through which the malpighian tubules drain. Endothelin-1-like immunoreactivity colocalizes with FMRFa immunoreactivity in the cells of the main region of the midgut but not in those in the midgut caeca. Endothelin-1-immunoreactive cells are present not only in adults but also throughout the five instars of posthatching development.
