The article presents the analysis of the chains of speech genres in A. P. Chekhov’s short story “Enemies”. It is argued that the speech genres which formalize the social relationships of the main characters of the story lead to an understanding of the concept of the work, as well as reveal the essence of the title of the story. It is noted that the verbal interaction of the main characters of the narrative is a system of interconnected speech genres. The article describes the speech genres that have developed in the context of the situation of verbal communication between a doctor and a patient, specifically the speech genre of a medical complaint forming the core of institutional discourse, as well as speech genres that go beyond the core area: speech genres of pleading, refusal, appeal, silence, unburdening the soul, payment for medical services. It is stated that integrating into the medical discourse, these speech genres form a functional unity with it. The author concludes that the speech genres of medical discourse reproduce the process of professional speech interaction and qualify both individual and socially conditioned qualities of the originator character and the recipient character, as well as the situation of their conversation. Invective speech genres of accusations and insults inherent in ordinary (non-institutional) discourse are characterized as conflicting speech genres indicating the status-role degradation of institutional relations. Special attention is paid to the speech genres of pleading and the speech genre of the unburdening the soul, the peculiarities of their functioning and language aspects in connection with the general cultural values and traditions of Russian spiritual culture. The article also specifies the role of the author’s remarks in the description of communicative situations.

The issue raised in this study touches upon different aspects related to expression of negative judgment in medical discourse. This issue lies within the boundaries of speech genre theory. The authors use analysis tools of such disciplines as functional linguistics, medical rhetoric and elocutary mastery. The comparison of accusation and reproach – two related to each other speech genres of expressing disapproval – is made in the introduction to the article. Lexicographic sources allowed to reveal essential semantic peculiarities of these genres. The authors have found that the most frequent means of expressing accusation and reproach is a rhetoric question which is used to express the doctor’s disapproval of the patient’s behaviour. It is pointed out that one of the reasons for the doctor’s failures in communication with patients is the improper use of rhetorical questions. This study attempts to discover communication pitfalls dealing with expressing disapproval. Understanding the structure and semantics of rhetoric questions allows to avoid common mistakes in the speech of medical students. It is necessary to view the object of the research in the context of argumentation and manipulation theories. The article also analyzes whether expressing disapproval can be regarded as a manipulative speech tactic. The authors claim that rhetoric questions should be regarded as irrelevant in the doctor’s dialog with the patient due to their negative impact on the patient’s emotional and psychological state. It is necessary to remember about risks related to iatrogenic factors.

Background: This study aims to analyze the exploration degree of popular model organisms by utilizing annotations from the UniProtKB (Swiss-Prot) knowledge base. The research focuses on understanding the genomic and post-genomic data of various organisms, particularly in relation to aging as an integral model for studying the molecular mechanisms underlying pathological processes and physiological states. Methods: Having characterized the organisms by selected parameters (numbers of gene splice variants, post-translational modifications, etc.) using previously developed information models, we calculated proteome sizes: the number of possible proteoforms for each species. Our analysis also involved searching for orthologs of human aging genes within these model species. Results: Our findings indicate that genomic and post-genomic data for more primitive species, such as bacteria and fungi, are more comprehensively characterized compared to other organisms. This is attributed to their experimental accessibility and simplicity. Additionally, we discovered that the genomes of the most studied model organisms allow for a detailed analysis of the aging process, revealing a greater number of orthologous genes related to aging. Conclusions: The results highlight the importance of annotating the genomes of less-studied species to identify orthologs of marker genes associated with complex physiological processes, including aging. Species that potentially possess unique traits associated with longevity and resilience to age-related changes require comprehensive genomic studies. [ABSTRACT FROM AUTHOR]

The article examines the using of media rumors as pragmatic influence mechanism in the modern communication. The printed and electronic messages with rumors make the material of research. The complex methods of analysis of the rumors role in the modern society are used. The inductive, descriptive and comparative, cognitive and discursive, semantic, definitional, functional and stylistic analysis is carried out. For the complex analysis of rumors in the modern media discourse, verbal means of rumors impact on the addressee in modern foreign-language and Russian mass media linguacultural, linguapragmatic, discursive approaches are applied. In the article, we realize the aim of the study. We identify the role of media rumors in the modern society. The ways of analysis are useful for development of rumorology, communication theory, linguapragmatics, sociolinguistics, cultural linguistics, media linguistics. In the article, we establish that mass media trying to realize the function of estimation and critics create and spread rumors themselves, use actively pseudo arguments and lie instead of exposure of various rumors, pseudo arguments, lies.

The Beganchik locality is a stratigraphic sequence of loessic deposits, pedogenic horizons and Paleolithic occupations located at the Kama-Volga confluence. The sequence is exposed on a bluff formed on the west side of an erosional remnant between the Kuybyshev Reservoir and the former channel of the Aktay River. Although the site is known for its Terminal Paleolithic-Mesolithic occupations of the Pleistocene-Holocene transition, evidence of older occupations and remains of fauna has been identified. Our research team identified evidence of human presence associated with a pedogenic horizon of MIS 3 age. Two AMS radiocarbon ages from a hearth produced ages around 47 000 years BP. Pollen and phytoliths from two soils horizons, including the one associated with the hearths indicate a steppe environment coincident with the formation of correlative soils elsewhere in the Russian Plain.

The article examines the materials of the beginning of the Migration period from the Biklyan’ settlement, located in the Lower Kama region. The area of 100 m2 was excavated in 2018, where a rectangular structure slightly deepened into subsoil loam with a large pit in the central part was revealed. The finds are mainly represented by fragments of ceramics, as well as an iron sickle, spinning whorls, a fragment of a crucible and ceramic objects of unknown purpose. The artifacts from the Biklyan’ settlement have close analogies in the materials of the sites dated to the beginning of the Migration period in the Middle Volga region (3rd–4th centuries CE) and reflect the traditions of several different population groups. The bulk of the ceramics collection is similar to the vessels of the Middle Volga variant of the Kiev culture; there are also fragments that find analogies in the assemblages of the forest-steppe peripheral sites of the Late Sarmatian world and in the simultaneous cultures of the Kama region. Stratigraphic records and radiocarbon dating testify in favor of the relatively short-term functioning of the site in the late 3rd – early 4th centuries.

Background and purpose: Combined hyperlipidemia is associated with an increased risk of cardiovascular events. This clinical trial investigated phospholipovit (essential phospholipids, Institute of Biomedical Chemistry, Moscow, Russia), an ultra-small phospholipid nanoparticle (micelles), targeted to phospholipids of HDL in lowering non-HDL-cholesterol (non-HDL-C) and triglycerides (TG) levels in patients with combined hyperlipidemia and moderate cardiovascular risk. Experimental approach: A randomized, double-blinded, placebo-controlled phase II trial was conducted on 100 patients. Phospholipovit or placebo was randomly administered orally (500 mg) 2 times a day for 12 weeks. The primary endpoint was the percent change of non-HDL-C from baseline to 12 weeks of exposure. Findings/Results: Treatment with phospholipovit resulted in a mean non-HDL-C reduction of 13.2% versus 4.3% compared with placebo. The absolute decrease in non-HDL-C was -23.2 (-48.7 - 7.0) mg/dL versus -7.3 (-17.0 - 12.0) mg/dL, significantly. The therapeutic target of non-HDL-C less than 130 mg/dL (3.4 mmol) was achieved in 15 of 39 patients (38.5%) in the phospholipovit group versus 2 of 41 patients (4.9%) in the placebo group OR 11.8 (2.4 - 116). Significant reduction in TG, apolipoprotein B, total cholesterol, and very low-density lipoprotein cholesterol levels was also observed. There were no changes in the liver and kidney functions, vital signs, or electrocardiography. There were no serious adverse events. Conclusion and implications: Phospholipovit significantly reduced non-HDL-C, TG, and atherogenic lipoproteins in patients with combined hyperlipidemia and moderate cardiovascular risk. This article provides a systematic review of research conducted on the proteomic composition of blood as part of a complex biological age estimation. We performed a comprehensive analysis of 17 publicly available datasets and compiled an integral list of proteins. These proteins were sorted based on their detection probability using mass spectrometry in human plasma. We propose this list as a basis for creating a panel of peptides and quantifying the content of selected proteins in the format of a proteomic aging clock. The selected proteins are especially notable for their roles in inflammatory processes and lipid metabolism. Our findings suggest, for the first time, that proteins associated with systemic disorders, including those approved by the FDA for clinical use, could serve as potential markers of aging. Gene expression is a tightly regulated process that involves multiple layers of control, including transcriptional, post-transcriptional, and translational regulation. To gain a comprehensive understanding of gene expression dynamics and its functional implications, it is crucial to compare translatomic, transcriptomic, and proteomic data. The two most common analysis methods, Ribo-seq and RNC-Seq, were used to analyze the translatome of the same sample, whose datasets were downloaded from the TranslatomeDB database. The resulting translatome maps obtained for three cell lines (HBE, A549, and MCF-7) using these two methods were comparatively analyzed. The two methods of translatome analysis were shown to provide comparable results and can be used interchangeably. The obtained mRNA translation patterns were annotated in the transcriptome and proteome context for the same sample, which may become the basis for the reconstruction of the molecular mechanisms of pathological process development in the future. Limit of detection (LoD) is a term that is used to characterize the sensitivity of an analytical method. The existing limitation of the sensitivity of analysis using modern mass spectrometry methods has been experimentally shown to be a limiting factor in the application of proteomic technologies in medicine. This article proposes a concept of a new technology that will set a new vector of development in the development of systems for solving problems of medical diagnostics and deals with theoretical and practical aspects of creating a new technology for the detection of single biomacromolecules (in particular, proteins) in biological samples. Such technology should be based on the principle of signal registration similar to that used in a Geiger counter (also known as a Geiger–Müller counter or G-M counter), a device that automatically counts the number of ionizing particles that hit it. This counter is free from probabilistic components; it registers a signal if there is at least one target molecule in the analysis chamber. Predictive medical diagnostics require technology based on methods where sensitivity allows for the detection of single marker molecules in a biological sample volume of 1–10 µL, the smallest volume of biomaterial used in laboratory diagnostics. Creation of a detector with a sensitivity of 10−18 M would allow for the detection of one molecule in 1 µL of the sample, which fundamentally makes this approach analogous to a G-M counter for solutions. To date, bioanalytical methods are limited to a sensitivity of 10−12 M (which is approximately 1 million molecules per 1 μL), which is insufficient to capture the early stages of pathological processes. The article is devoted to the development and substantiation of proposals for deepening the integration processes of the transport and logistics market of Russia and China. The trends in the development of the world market of transport and logistics services are analyzed with an emphasis on the border regions of Russia and China; identified its key problems and possible directions for their solution to deepen the subsequent integration between the border regions; an assessment of the effectiveness of the development of the transport infrastructure of the border regions of Russia, as well as the prospects for integrating Russia into the system of world economic relations in the Asia-Pacific region (APR) is given. Simple Summary: Obesity is a significant health concern associated with fat accumulation and complications like inflammation, cancer, and diabetes. Understanding its molecular roots is crucial, especially for young individuals who are capable of lifestyle changes. Our study examined underweight, lean, overweight, and obese individuals by analyzing blood samples using metabolomics, proteomics, and genomics. Using metabolomics, we identified 313 substances; proteomics revealed 708 proteins, and genomics explored 647,250 point mutations. Models predicting body mass index showed that, individually, proteomics provided more value for prediction, followed by metabolomics and genomics. Combining proteomics and metabolomics in a multiomic approach yielded the best results, surpassing single-factor analyses. This pioneering study is the first to focus on obesity in young people, integrating genomic, proteomic, and metabolomic data. Our findings provide valuable insights into the molecular mechanisms of obesity, offering potential for more targeted interventions. Obesity is a socially significant disease that is characterized by a disproportionate accumulation of fat. It is also associated with chronic inflammation, cancer, diabetes, and other comorbidities. Investigating biomarkers and pathological processes linked to obesity is especially vital for young individuals, given their increased potential for lifestyle modifications. By comparing the genetic, proteomic, and metabolomic profiles of individuals categorized as underweight, normal, overweight, and obese, we aimed to determine which omics layer most accurately reflects the phenotypic changes in an organism that result from obesity. We profiled blood plasma samples by employing three omics methodologies. The untargeted GC×GC–MS metabolomics approach identified 313 metabolites. To augment the metabolomic dataset, we integrated a label-free HPLC–MS/MS proteomics method, leading to the identification of 708 proteins. The genomic layer encompassed the genotyping of 647,250 SNPs. Utilizing omics data, we trained sparse Partial Least Squares models to predict body mass index. Molecular features exhibiting frequently non-zero coefficients were selected as potential biomarkers, and we further explored enriched biological pathways. Proteomics was the most effective in single-omics analyses, with a median absolute error (MAE) of 5.44 ± 0.31 kg/m2, incorporating an average of 24 proteins per model. Metabolomics showed slightly lower performance (MAE = 6.06 ± 0.33 kg/m2), followed by genomics (MAE = 6.20 ± 0.34 kg/m2). As expected, multiomic models demonstrated better accuracy, particularly the combination of proteomics and metabolomics (MAE = 4.77 ± 0.33 kg/m2), while including genomics data did not enhance the results. This manuscript is the first multiomics study of obesity in a gender-balanced cohort of young adults profiled by genomic, proteomic, and metabolomic methods. This work demonstrates the use of a modified mica to concentrate proteins, which is required for proteomic profiling of blood plasma by mass spectrometry (MS). The surface of mica substrates, which are routinely used in atomic force microscopy (AFM), was modified with a photocrosslinker to allow "irreversible" binding of proteins via covalent bond formation. This modified substrate was called the AFM chip. This study aimed to determine the role of the surface and crosslinker in the efficient concentration of various types of proteins in plasma over a wide concentration range. The substrate surface was modified with a 4-benzoylbenzoic acid N-succinimidyl ester (SuccBB) photocrosslinker, activated by UV irradiation. AFM chips were incubated with plasma samples from a healthy volunteer at various dilution ratios (102X, 104X, and 106X). Control experiments were performed without UV irradiation to evaluate the contribution of physical protein adsorption to the concentration efficiency. AFM imaging confirmed the presence of protein layers on the chip surface after incubation with the samples. MS analysis of different samples indicated that the proteomic profile of the AFM-visualized layers contained common and unique proteins. In the working series of experiments, 228 proteins were identified on the chip surface for all samples, and 21 proteins were not identified in the control series. In the control series, a total of 220 proteins were identified on the chip surface, seven of which were not found in the working series. In plasma samples at various dilution ratios, a total of 146 proteins were identified without the concentration step, while 17 proteins were not detected in the series using AFM chips. The introduction of a concentration step using AFM chips allowed us to identify more proteins than in plasma samples without this step. We found that AFM chips with a modified surface facilitate the efficient concentration of proteins owing to the adsorption factor and the formation of covalent bonds between the proteins and the chip surface. International Theoretical and Practical Workshop on Application of Paleo-Ecological Methods in Archaeology took place in Bolgar International Center for Archaeological Studies from 9 to 19 August 2016. The workshop was organized by Institute of Archaeology named after A.Kh. Khalikov of Tatarstan Academy of Sciences, supported by the Russian Fundamental Research Fund and the Fund for Rehabilitation of Historical and Cultural Sites of the Republic of Tatarstan.

The issue topicality is due to the increasing interest in architectural traditions of the Russian regions. The research objective is to identify the main development stages and features of the Cossack stanitsa (settlement) Paris of Novolineyniy region of Orenburg gubernia and evolution of architectural forms. The leading research method was historical-architectural analysis and summarization of archive and field data. A staged analysis of development of this type of settlements was performed. Descriptions and analysis of little-known architectural monuments of the region are presented. The paper reveals an interesting stratum of regional heritage. The article attempts to analyze a wide range of issues related to the formation of the architecture of the Cossack stanitsas (settlements) in the Novolineyniy region of Orenburg gubernia in the 19th–20th centuries. The layout, composition and stylistic features of the architecture of the Cossack stanitsas (settlements) in the Novolineyniy region are considered. Numerous materials of central and local archives were researched and are published for the first time.

The dynamics of human settling Samarskaya Luka (Volga bend) area adjacent to the Zhiguli archaeological site and burial ground has been studied by analyzing both the cultural layer of the monument and the soil strata in the adjacent ravine. Seven erosion and accumulation cycles have been distinguished in the ravine development during the Holocene, correlated with periods of the water catchment area development. Each cycle used to start with forest fires, leading to an increase in runoff and ravine cutting-in, and to complete with surface stabilization as grassland or forest was reestablished. The length of periods between changes of land use varied from a few decades in the agricultural cycles of the early Middle Ages, up to several hundred years in the nomadic-pastoral cycles of the Eneolithic – the Bronze Age, the Iron Age and the late Middle Ages. Vegetation changed within each cycle, as human utilization of the watershed resulted in elimination of the understory, reduction of tree species variety, an increase in the proportion of conifers, and, due to combination of fire and grazing, led to deforestation. A lengthy deforestation period occurred in the mid-Holocene (Eneolithic – early Bronze Age). A new period of progressive deforestation and open space growth started in the middle of the 1st millennium AD. Discontinuities in human occupation lasting a few hundred years observed in the mid-Holocene. Shorter breaks (less than a century) took place between the Scythian-Gorodets period and the Imenkovo-Khazar period, later between the Khazar period and the Bulgarian period, and in that preceding Russian colonization of the Samara Luka area in the 16th century.
