Objective: In this study, the thermodynamic protonation constant ((_s^s)〖pK〗_a ) of antifungal azole derivative miconazole and tioconazole were investigated by the reversed-phase liquid chromatography (RPLC) method. Although classical stationary phases are the most commonly used column-packed materials in the analyses of these compounds by the RPLC method, the determination of the pKa value of miconazole and tioconazole with a cyano-based column has not been reported so far.Material and Method: Chromatographic behavior of the selected compounds was determined with Ultra Cyano column (Restek®, 150×3.0 mm I.D, 3µm) which is compatible with hydrophobic compounds. The dependence of the retention times on the acetonitrile content of the mobile phase and the effect of the mobile phase pH on the chromatographic retention was determined for each compound by changing the pH in the range of 3.0-7.50.Result and Discussion: The (_s^s)〖pK〗_a values were determined by a nonlinear regression program. The thermodynamic aqueous pKa ((_w^w)〖pK〗_a ) value of the studied compounds was calculated from the (_s^s)〖pK〗_a value by using the macroscopic parameters (dielectric constant and mole fraction) that play an important role in the solvent properties. The calculated (_w^w)〖pK〗_a values were compared with literature values., Amaç: Bu çalışmada, antifungal azol türevi mikonazol ve tiokonazolün termodinamik protonasyon sabiti ((_s^s)〖pK〗_a ) ters faz sıvı kromatografisi (RPLC) yöntemi ile araştırılmıştır. Bu bileşiklerin RPLC yöntemi ile analizlerinde klasik sabit fazlar en yaygın olarak kullanılan kolon dolgulu malzemeler olmasına rağmen, siyano bazlı bir kolonla şimdiye kadar mikonazol ve tiokonazolün pKa değerinin tayini rapor edilmemiştir.Gereç ve Yöntem: Seçilen bileşiklerin kromatografik davranışları hidrofobik bileşiklerle uyumlu Ultra siyano kolon (Restek®, 150×3.0 mm I.D, 3µm) ile belirlenmiştir. Alıkonma sürelerinin mobil fazın asetonitril içeriğine bağımlılığı ve mobil faz pH'ının kromatografik alıkonma üzerindeki etkisi, pH 3.0-7.50 aralığında değiştirilerek her bir bileşik için belirlenmiştir.Sonuç ve Tartışma: (_s^s)〖pK〗_a değerleri, doğrusal olmayan bir regresyon programı ile belirlenmiştir. Çalışılan bileşiklerin termodinamik suda pKa ((_w^w)〖pK〗_a ) değeri, çözücü özelliklerinde önemli bir rol oynayan makroskopik parametreler (dielektrik sabiti ve mol fraksiyonu) kullanılarak (_s^s)〖pK〗_a değerinden hesaplanmıştır. Hesaplanan (_w^w)〖pK〗_a değerleri literatür değerleri ile karşılaştırılmıştır.

The solubility values were fitted to various cosolvency equations. The model accuracies were studied with the computation of the mean relative deviations. The thermodynamic behavior was investigated according to the van't Hoff and Gibbs equations for clotrimazole in the propylene glycol + water system. Furthermore, the density data for clotrimazole were determined in mixtures of propylene glycol + water and fitted to the Jouyban-Acree equation.

Thebenzimidazole ring in the structure of B12 vitamin and the imidazole ring inthe structure of histidine are heterocyclic structures with importantbiological and clinical applications in medicine, pharmacy and pharmaceuticalchemistry. These structures havemany different biological activities such as anthelmintic, antifungal,antiviral and anti-HIV and have recently been used as an anticancer agent,draws attention to these structures. In this study, the ionization/protonation constants of six drugmolecules which have containing benzimidazole/imidazole moiety were determinedin mobile phase media containing 40% (v/v) ACN:H2O by reversed phaseliquid chromatographic method. Gemini NX C18 (250 mm× 3.0 mm I.D., 5 µm) column was used inexperimental studies and chromatographic analyzes were performed at a flow rateof 1 mL/min and at 30°C. The lipophilicity and solubility values of these sixdrug molecules were calculated using the Abraham solute descriptor values and theretention data obtained by the reversed phase liquid chromatographic method, B12vitamini yapısında bulunan benzimidazol halkası ve histidinin yapısında bulunanimidazol halkası; tıpta, eczacılıkta, ilaç kimyasında önemli biyolojik veklinik uygulamalara sahip heterosiklik yapılardır. Bu yapıların anthelmintik,antifungal, antiviral ve anti-HIV gibi birçok farklı biyolojik aktiviteye sahipolması ve son zamanlarda antikanser ajan olarak kullanılması dikkatleri buyapılar üzerine çekmektedir. Bu çalışmada benzimidazol/imidazol halkası içerenaltı ilaç molekülünün ters faz sıvı kromatografik yöntemle %40 (v/v) ACN:H2Oiçeren mobil faz ortamında iyonlaşma/protonasyon sabitleribelirlenmiştir. Deneysel çalışmalarda Gemini NX C18 (250 x 3.0 mm I.D., 5 µm) kolon kullanılmıştır ve kromatografik analizler 1 mL/dakikaakış hızında, 30°C’de gerçekleştirilmiştir. Abraham çözünentanımlayıcıları ve ters faz sıvı kromatografik yöntemle elde edilen bu alıkonmaverileri kullanılarak bu altı ilaç molekülünün lipofilisite ve çözünürlükdeğerleri hesaplanmıştır.

Bileşiklerin oral absorpsiyonunun değerlendirilmesinde, hesaplanmış ve deneysel olarak ölçülen fizikokimyasal parametreleri içeren çalışmalara ihtiyaç duyulmaktadır. İyonlaşma/protonasyon sabiti (pKa), çözünürlük ve lipofilisite (l〖ogP〗_(o/w)) iyi oral absorpsiyona sahip ilaçları tanımlayan en önemli fizikokimyasal parametreler arasındadır. Bu parametreler ilaç adayı moleküllerin absorpsiyon, dağılım, metabolize edilme ve atılım (ADME) davranışının tahminine de izin vermektedir.Bu tez çalışmasında benzimidazol ve imidazol fonksiyonel grubu içeren anthelmintik ve antifungal özellik taşıyan (albendazol, astemizol, mebendazol, tiyabendazol, klotrimazol, metimazol (tiyamazol) ve metronidazol) ilaç moleküllerinin ters faz sıvı kromatografik yöntemle alıkonma davranışı incelenmiştir. Kullanılan bu yöntemde altı bileşik için Kinetex Core-Shell C8 (Phenomenex, 150mm×2,6 µm, 4,6 mm) kolon; metimazol için Kinetex Core-Shell EVO C18 (Phenomenex, 250mm × 4,6 mm, 5 µm) kolon ile 25°C'de 0,8 mL/dakika akış hızında, analizler gerçekleştirilmiştir. Bu kromatografik analizlerden elde edilen veriler kullanılarak bileşiklerin farklı asetonitril-su derişimlerinde pKa değerleri deneysel olarak belirlenmiştir.Diğer iki fizikokimyasal özellik olan çözünürlük ve lipofilisite değerleri Abraham çözünen tanımlayıcıları kullanılarak hesaplanmıştır. Bu tanımlayıcılar kullanılarak ilaç moleküllerinin kan beyin bariyerinden geçişi de hesaplanmıştır.Geliştirilen kromatografik metotta bileşiklerin mümkün olan en kısa sürede analizini gerçekleştirmek ve bileşiklerin simetrik piklerini elde etmek amaçlanmıştır. Bunun için optimum ayırma koşulları belirlenmiştir. Geliştirilen RPLC metodunun geçerliliğinin kanıtlanması amacıyla metot validasyonu gerçekleştirilmiş ve Uluslararası Uyum Konferansı (International Conference on Harmonization, ICH) tarafından önerilen parametreler test edilerek metodun performansı ve güvenilirliği değerlendirilmiştir. In the evaluation of oral absorption of compounds, there is a need for studies including calculated and experimentally measured physicochemical parameters. Ionization/protonation constant (pKa), solubility and lipophilicity (l〖ogP〗_(o/w)) are among the most important physicochemical parameters that define drugs with good oral absorption. These parameters also define prediction of ADME behaviour of drug candidates.In this thesis, retention behaviour of benzimidazole and imidazole functional group carrying anthelmintic and antifungal drug molecules (albendazole, astemizole, mebendazole, thiabendazole, clotrimazole, methimazole (thiamazole) and metronidazole) was investigated with reversed phase liquid chromatographic method. Analyses were carried out with Kinetex Core-Shell C8 (Phenomenex, 150mm×2,6 µm, 4,6 mm) column for six compounds and Kinetex Core-Shell EVO C18 (Phenomenex, 250mm × 4,6 mm, 5 µm) column for methimazole with 0,8 mL/minute flow rate at 25 °C. By using the data obtained from these chromatographic analyses, pKa values of the compounds were determined experimentally at different acetonitrile-water concentrations.Two other physicochemical properties, the solubility and lipophilicity values, were calculated using Abraham solute descriptors. Using these solute descriptors, the transport of of drug molecules through the blood brain barrier was also calculated.In the developed chromatographic method, it is aimed to perform the analysis of the compounds as soon as possible and to obtain symmetrical peaks of the compounds. For this purpose, optimum separation condition was determined. Method validation has been performed to prove the validity of the developed RPLC method and the parameters proposed by ICH were tested and the performance and reliability of the method were evaluated. 286

Bu tez çalışmasında amaç yeni termofilik mikroorganizmalardan pektin liyaz enzimini üretmek ve üretilen bu enzimi saflaştırıp karakterize etmektir.Bu amaçla termofilik Aspergillus niger Ocak 4 fungusu farklı besi ortamlarında yetiştirilmiştir. En yüksek enzim elde edilen besi ortamı belirlenmiştir. Termofilik fungustan pektin liyaz enzimi üretimli için, amonyum sülfat ile kısmi saflaştırılma yapılmış ve jel filtrasyon ve iyon değişim kromotografisi ile de saflaştırma işlemlerine kademeli olarak devam edilmiştir.Saflaştırılan enzimin optimum pH ve sıcaklık çalışmaları yapılmıştır. Böylece saflaştırılan enzim için en uygun pH ve sıcaklık değerleri elde edilmiştir. SDS-PAGE yöntemi ile de enzimin molekül ağırlığı tayini yapılmıştır.Saflaştırılan pektin liyaz enzimi için optimum pH 8.0; optimum sıcaklık ve süre sırasıyla 40 ºC, 60 dk olarak belirlenmiştir. Bu enzim için jel filtrasyon kromotografisi ve iyon değişim kromotografisi ile sırasıyla 4,2 kat ve 76,5 kat saflaştırma katsayısı elde edilmiştir. The aim of this study is produce pectin lyase enzyme from thermophilic microorganisms and purify and characterize this enzyme.For this purpose thermophilic Aspergillus niger Ocak 4 fungi was growth different submerged conditions. Submerged conditions obtaining maximum enzyme was determined. Pectin lyase enzyme was produced from thermophilic microorganisms, partial purification with ammonia sulphate was done. Purification studies were also resumed with gel filtration and ion-exchange chromatography.Optimum pH and temperature studies of purified enzyme was done. Thereby optimum pH and temperature values were obtained for purified enzyme. Molecular weight determination of purified enzyme was done with SDS-PAGE.For purified pectin lyase enzyme, optimum pH 8.0; optimum temperature and time 40 ºC, 60 min were determined. For this enzyme 4,2 and 76,5 purification fold were obtained with ion-exchange chromatography and gel filtration chromatography respectively. 73
