In Western France, rapeseed (Brassica napus) cultivation faces substantial yield losses due to the root holoparasitic plant Phelipanche ramosa. However, recent observations have shown a reduction in parasitism within previously heavily infested fields. This study investigates two neighboring rapeseed soils with distinct levels of parasitic infestation, considered suppressive and conducive. Using a cocultivation system of Brassica napus and P. ramosa, we comprehensively examined rhizosphere exudates, parasitic plant attachment, and rhizosphere soil microbiota. Our findings revealed that the suppressive soil effectively reduced parasitism by impeding broomrape attachment and development, as well as inducing necrosis of tubercles. This suppressive effect was specific to postattachment stages, leaving germination and haustoriogenesis preattachment stages unaffected. Analysis of microbial structures suggested that the suppression of parasitism is predominantly of fungal rather than bacterial origin. Correlation network analyses identified three groups of amplicon sequence variants (ASVs) associated with suppression. Notably, seven ASVs were inversely correlated with parasitic attachments, and only one ASV, identified as Berkeleyomyces, a necrotrophic fungus responsible for black root rot, was positively correlated with necrosis and was more abundant in the suppressive soil. This study demonstrates the contrasting parasitic plant development on two physicochemically similar soils, highlighting the central role of fungal dynamics in the rhizosphere. These results provide valuable insights into the mechanisms underlying soil-mediated suppression of P. ramosa, offering potential strategies for mitigating the impact of this root holoparasite on rapeseed yields in the region.

The specific identification of three major morphotypes of the tropical holopelagic Sargassum species causing massive strandings on the African and Caribbean coastlines was attempted by morphological characterisation as well as quantitative and qualitative analyses of several metabolites. Of the 25 morphological variables studied on 208 samples from the North Atlantic Ocean, 22 were used to establish a dichotomous identification key, allowing without any doubt the identification of each morphotype based on their morphological criteria alone. We also attempted to differentiate morphotypes using chemical fingerprintings (HR-MAS NMR) and markers by analysing pigment level and composition using High Pressure Liquid Chromatography, terpene profiles by Thin Layer Chromatography, phenolic compound levels by the Folin-Ciocalteu assay and structures by 2D Nuclear Magnetic Resonance spectroscopy, and fatty acid composition by Gas Chromatography. While pigment level and composition, terpene profiles, and phenolic contents were not discriminating, quantification of eight fatty acids enabled the differentiation of the three morphotypes. Furthermore, phlorotannin purification permitted their structural characterisation allowing discrimination between the three morphotypes. Our study highlights the potential of the free fatty acid profile and phlorotannin structure as good chemomarkers in order to discriminate between the three morphotypes of holopelagic Sargassum.

Near-infrared spectroscopy (NIRS) provides a high-throughput phenotyping technique to assist breeding for improved faba bean seed quality. We combined chemical analysis of protein, oil content (and composition) with NIRS through chemometrics, employing Partial Least Squares (PLS), Elastic Net (EN), Memory-based Learning (MBL), and Bayes B (BB) as prediction models. Protein was the most reliably predicted trait (R2 = 0.96–0.98) across field trials, followed by oil (R2 = 0.82–0.86) and oleic acid (R2 = 0.31–0.68). Samples for training the models were selected using K-means clustering. The optimal statistical approach for prediction was compound-specific: PLS for protein (Root Mean Squared Error - RMSE = 0.46), BB for oil (RMSE = 0.067), and EN for oleic acid content (RMSE = 2.83). Reduced training set simulations revealed different effects on prediction accuracy depending on the model and compound. Several NIR regions were pinpointed as highly informative for the compounds, using the shrinkage and variable selection capabilities of EN and BB.

Since 2011, massive new strandings of holopelagic Sargassum have been reported on the coasts of the Caribbean, northern Brazil, Guiana, and West Africa, causing severe economic and ecological damage. Three common morphotypes (S. fluitans III, S. natans I, and S. natans VIII) were identified as responsible for these catastrophic events, with dominance shifts between them over time. However, the taxonomic status of these holopelagic Sargassum morphotypes remains unclear. Using an integrative taxonomy framework, combining a morphological study and molecular analyses, this study aimed to clarify their taxonomic status. Morphological analyses of 54 characters revealed no intermediate form between the three morphotypes, with the overall shape, nature of the axis, and size and shape of blades and vesicles being the most discriminating. An analysis of mitochondrial (IGS, cox2, cox3, mt16S rRNA, and nad6) and plastid (rbcL) markers confirmed the genetic divergence among the three morphotypes, with a lower level of divergence between the two S. natans morphotypes. Without additional molecular characterization, these morphotypes cannot be classified as three distinct species. However, due to their distinct morphological characteristics and sympatry within drifting aggregations, a revision of holopelagic species names is proposed, with Sargassum fluitans var. fluitans (for S. fluitans III), Sargassum natans var. natans (for S. natans I), and S. natans var. wingei (for S. natans VIII). This revision provides necessary clarity on the species involved in inundations of the tropical Atlantic.

Faba bean (Vicia faba L.) is a valuable ingredient in plant-based foods such as meat and dairy analogues. However, its typical taste and aroma are considered off-flavours in these food applications, representing a bottleneck during processing. Breeding is needed to develop varieties with minimal off-flavours and high protein content. The genetic regulation of these traits is underexplored. To dissect their genetic architecture, we performed a genome-wide association study (GWAS). A total of 245 faba bean accessions (the CGN population) were genotyped using the 90K-SPET targeted assay. These accessions were phenotyped in 2021 and 2022 in the Netherlands for protein, oil, fatty acids, lipid-derived products, phenolic acids, flavonoids, and tannins. The CGN population showed large phenotypic variation and moderate-to-high narrow-sense heritability for most traits. The growing environment significantly affected all traits, with trait-specific genotype-by-year (GxY) interactions. Condensed tannins and fatty acids were the most stable across the two years and had the highest heritability estimates (h2 > 0.6). GWAS identified a total of 148 single nucleotide polymorphisms (SNPs) loci in 2021 and 167 in 2022. Key candidate regulators included genes involved in lipid biosynthesis (ATS2, KAS, LPP), amino acid transport (CAT4) for protein storage, zero tannins locus-1 (zt-1), and regulators of the phenylpropanoid pathway, such as a shikimate kinase gene and transcription factors bHLH137-like and MYB. These results pave the way for validation studies and biotechnological applications to improve the quality of faba bean-based foods.

This study focuses on developing innovative and eco-friendly purification methods for the isolation of bioactive compounds derived from Padina pavonica, a brown abundant macroalga in Djibouti. Three distinct fractions, obtained via liquid-liquid extraction (LLE_FAE), solid-phase extraction (SPE_WE50), and flash chromatography (FC_EtOH20), were selected based on their high phenolic content and antioxidant activities. All fractions were also evaluated for their anti-ageing potential by assessing their ability to inhibit two vital skin-ageing enzymes, tyrosinase and elastase. Structural analysis by 1H-13C HMBC NMR and LC-MS revealed a selectivity of phlorotannins depending on the purification methods. The LLE_FAE fraction exhibited greater structural complexity, including compounds such as phloroglucinol, diphlorethol/difucol, fucophlorethol and bifuhalol, which likely contribute to its enhanced bioactivity compared to the fractions obtained by FC_EtOH20 and SPE_WE50, which were also active and enriched only in phloroglucinol and fucophlorethol. These findings highlight the impact of purification techniques on the selective enrichment of specific bioactive compounds and demonstrated the interest of FC or SPE in producing active phlorotannin-enriched fractions. These two purification methods hold strong potential for innovative cosmeceutical applications. Results are discussed regarding the use of P. pavonica as a promising marine resource in Djibouti to be used for the development of cosmetic industry.

Branched broomrape (Phelipanche ramosa (L.) Pomel), an obligate parasitic weed with a wide host range, is known for its devasting effects on many crops worldwide. Soil fungi, notably Fusarium sp., are described as pathogenic to broomrape, while the hypothesis of the phytotoxicity of fusaric acid produced by F. verticillioides for parasitic weeds of the genus Orobanche has been proposed. Using image analysis and untargeted metabolomics, this study investigated fungal metabolites phytotoxic for P. ramosa and produced by the F. venenatum MIAE02836 strain, isolated from symptomatic broomrapes and identified as a promising candidate for broomrape biocontrol. Phytotoxicity tests of crude extracts from the fungus alone or in interaction with broomrape on P. ramosa microcalli and quantification of necrosis by image analysis confirmed the phytotoxic potential of F. venenatum MIAE02836 metabolites towards the early developmental stages of P. ramosa. Data analysis of a non-targeted metabolomics approach revealed numerous metabolites produced by F. venenatum MIAE02836. Four of them, accumulated during interaction with the parasitic plant, are known for their phytotoxic potential: maculosin, cyclo(Leu-Phe), phenylalanyl-D-histidine and anguidine. These results suggest that combining image acquisition of the microcalli screening test and untargeted metabolomic approach is an interesting and relevant method to characterize phytotoxic fungal metabolites.

In this data paper, the sea level time series at Socoa (Saint-Jean-de-Luz, southwestern France) is extended through a data archaeology exercise. We conducted a comprehensive study of national and local archives to catalogue water level records stored in ledgers (handwritten record books) and charts (marigrams from mechanical float gauges), along with other associated documents (metadata). A dedicated effort was undertaken to preserve more than 2000 documents by archiving them in digital formats. Using this large set of rescued documents, the Socoa time series has been extended back to 1875, with more than 58 station-years of additional data. The final time series has hourly sampling, while the raw dataset has a finer sampling frequency of up to 5 min. By analysing precise levelling information, we assessed the continuity of the vertical datum. We also compared the new century-long time series to nearby tide gauge data to ensure its datum consistency. While the overall quality of the time series is generally good, siltation of the stilling well has occasionally affected certain parts of the record. We have successfully identified these impacted periods and flagged the corresponding data as doubtful. This extended high-resolution sea level time series at Socoa, spanning more than 100 years, will be valuable for advancing climate research, particularly when studying the decadal-scale variations in the North Atlantic and investigating the storminess and extreme events along the French Basque coast. The screening of 166 extracts from tropical marine organisms (invertebrates, macroalgae) and 3 cyclolipopeptides from microorganisms against yeast prions highlighted the potential of Verongiida sponges to prevent the propagation of prions. We isolated the known compounds purealidin Q (1), aplysamine-2 (2), pseudoceratinine A (3), aerophobin-2 (4), aplysamine-1 (5), and pseudoceratinine B (6) for the first time from the Wallisian sponge Suberea laboutei. We then tested compounds 1–6 and sixteen other bromotyrosine and bromophenol derivatives previously isolated from Verongiida sponges against yeast prions, demonstrating the potential of 1–3, 5, 6, aplyzanzine C (7), purealidin A (10), psammaplysenes D (11) and F (12), anomoian F (14), and N,N-dimethyldibromotyramine (15). Following biological tests on mammalian cells, we report here the identification of the hitherto unknown ability of the six bromotyrosine derivatives 1, 2, 5, 7, 11, and 14 of marine origin to reduce the spread of the PrPSc prion and the ability of compounds 1 and 2 to reduce endoplasmic reticulum stress. These two biological activities of these bromotyrosine derivatives are, to our knowledge, described here for the first time, offering a new therapeutic perspective for patients suffering from prion diseases that are presently untreatable and consequently fatal.

Abstract Coastal water level measurements represent one of the earliest geophysical measurements and allow an assessment of historical sea level rise and trends in tides, river flow and storm surge. However, recovery and digitization of archival tidal records have been much less widespread and systematic than, for example meteorological records. In this contribution, we discuss data rescue efforts and lessons learned in France, the United States and the United Kingdom, countries with early and extensive tide gauge networks by the mid‐19th century. We highlight the importance of (a) cataloguing the historical gauge records, as a first step towards locating them; (b) locating data in archives, and then recovering and saving data by any means necessary, including photographs and scanning; (c) obtaining metadata, including both quantitative survey records, gauge checks and clock data, but also qualitative records such as gauge notes, letters and reports; and (d) quantitative statistical analysis of data and datum quality, using both standard data‐entry checks but also tools that leverage the unique predictability of tide measurements. Methods for digitizing original analogue records are also discussed, including semi‐automatic, computer‐based methods of digitizing tidal charts (marigrams). Although the current best practice is described, future improvements are desirable and needed to make the more than estimated 10,000 station years of unused, undigitized records available to the scientific community.

Tetrahedrally coordinated aluminate Al(OH)4- and dialuminate Al2O(OH)62- anions are considered to be major species in aluminum-rich alkaline solutions. However, their relative abundance remains difficult to spectroscopically quantify due to local structure similarities and poorly understood effects arising from extent of polymerization and counter-cations. To help unravel these relationships here we report detailed characterization of three solid-phase analogues as structurally and compositionally well-defined reference materials. We successfully synthesized a cesium salt of the aluminate monomer, CsAl(OH)4·2H2O, for comparison to potassium and rubidium salts of the aluminate dimer, K2Al2O(OH)6, and Rb2Al2O(OH)6, respectively. Single crystal and powder X-ray diffraction methods clearly reveal the structure and purity of these materials for which a combination of 27Al MAS-NMR, Al K-edge X-ray absorption and Raman/IR spectroscopies was then used to fingerprint the two major tetrahedrally coordinated Al species. The resulting insights into the effect of Al-O-Al bridge formation between aluminate tetrahedra on spectroscopic features may also be generalized to the many materials that are based on this motif.

The growing interest in plant protein sources, such as pulses, is driven by the necessity for sustainable food production and climate change mitigation strategies. Faba bean (Vicia faba L.) is a promising protein crop for temperate climates, owing to its remarkable yield potential (up to 8 tonnes ha−1 in favourable growing conditions) and high protein content (~29% dry matter basis). Nevertheless, the adoption of faba bean protein in plant-based products that aim to resemble animal-derived counterparts is hindered by its distinctive taste and aroma, regarded as “off-flavors”. In this review, we propose to introduce off-flavor as a trait in breeding programs by identifying molecules involved in sensory perception and defining key breeding targets. We discuss the role of lipid oxidation in producing volatile and non-volatile compounds responsible for the beany aroma and bitter taste, respectively. We further investigate the contribution of saponin, tannin, and other polyphenols to bitterness and astringency. To develop faba bean varieties with diminished off-flavors, we suggest targeting genes to reduce lipid oxidation, such as lipoxygenases (lox) and fatty acid desaturases (fad), and genes involved in phenylpropanoid and saponin biosynthesis, such as zero-tannin (zt), chalcone isomerase (chi), chalcone synthase (chs), β-amyrin (bas1). Additionally, we address potential challenges, including the need for high-throughput phenotyping and possible limitations that could arise during the genetic improvement process. The breeding approach can facilitate the use of faba bean protein in plant-based food such as meat and dairy analogues more extensively, fostering a transition toward more sustainable and climate-resilient diets.

The effect of UV radiation on the accumulation of mycosporine-like amino acids (MAAs) and pigments was investigated on red macroalga Palmaria palmata cultivated for 21 days. The data were combined with the effect of NaNO3 to further investigate the synthesis of these nitrogenous compounds. A progressive decrease in both total MAA and pigment contents was observed, with a positive effect of nitrate supply. Usujirene was the only MAA exhibiting a significantly increasing content when exposed to UV radiation, changing from 9% to 24% of the total MAA’s contribution, with no variation observed with NaNO3. This suggests a specific induction or synthesis pathway of usujirene for photoprotection, while the synthesis of other MAAs could have been limited by an insufficient amount of UV radiation and/or irradiance. The photoprotective ability of some MAAs could have been impacted by nitrogen starvation over time, resulting in a limited synthesis and/or potential use of MAAs as a nitrogen source for red macroalgae. The data confirmed the multiple effects of environmental factors on the synthesis of MAAs while providing new insights into the specific synthesis of usujirene, which could find an application in the cosmetics sector as natural sunscreen or an anti-ageing agent.
