1. Membrane Nanodomains Homeostasis During Propofol Anesthesia as Function of Dosage and Temperature

Abstract

Some anesthetics bind and potentiate gamma-aminobutyric-acid-type receptors, but no universal mechanism for general anesthesia is known. Furthermore, often encountered complications such as anesthesia induced amnesia are not understood. General anesthetics are hydrophobic molecules easily dissolving into lipid bilayers. Recently, it was shown that general anesthetics perturb phase separation in vesicles extracted from fixed cells. Unclear is whether under physiological conditions general anesthetics induce perturbation of the lipid bilayer, and whether this contributes to the transient loss of consciousness or anesthesia side effects. Here we show that propofol perturbs lipid nanodomains in the outer and inner leaflet of the plasma membrane in intact cells, affecting membrane nanodomains in a concentration dependent manner: 1 {\mu}M to 5 {\mu}M propofol destabilize nanodomains; however, propofol concentrations higher than 5 {\mu}M stabilize nanodomains with time. Stabilization occurs only at physiological temperature and in intact cells. This process requires ARP2/3 mediated actin nucleation and Myosin II activity. The rate of nanodomain stabilization is potentiated by GABA receptor activity. Our results show that active nanodomain homeostasis counteracts the initial disruption causing large changes in cortical actin.

Abstract

The importance of specific neurons to a network’s function is best studied by precisely timed, reversible silencing of these neurons. Previously, we showed that alternating magnetic field mediated heating of magnetic nanoparticles bound to neurons expressing temperature-sensitive cation channels TRPV1, stimulates these neurons to fire and affects animal behavior in vivo (Munshi et al., 2017). Here, we demonstrate how to apply magnetic nanoparticle mediated heating to silence target neurons. Rat hippocampal neuron cultures are transfected to express the temperature gated chloride Anoctamin1 (TMEM16A) channels. Within seconds the heating of the membrane opens the Anoctamin1 (TMEM16A) channels, suppressing action potential firing. Five seconds of magnetic field application leads to about 12 seconds of silencing, with a latency of about 2 seconds and an average suppression ratio of more than 80%. The method provides a promising avenue for tether free, remote, transient neuronal silencing in vivo for both scientific and therapeutic applications.

A description is provided about the fluorescent probes for hydrogen peroxide and the reagents make use of the boronate deprotection mechanism to give increased selectivity and optical dynamic range to identify H(sub 2)O(sub 2) in the water soluble solution. Thus, it specifies that hydrogen peroxide is an important ROS (reactive oxygen species) consequence in living beings and a regular marker for oxidative stress.

Abstract

The synthesis and properties of Peroxyflour-1, a new water-soluble, turn-on optical probe for H2O2 that exhibits high selectivity and dynamic range for the small molecule over other reactive oxygen species (ROS) is reported. The fluorescein-based reagent features excellent selectivity for H2O2 over competing cellular ROS, a large dynamic response owing to its dual colorimetric /fluorometric detection mechanism and long-wavelength visible excitation and emission profiles to minimize cell and tissue damage while avoiding interfering autofluoresecence from native cellular species.
