A new exponentially fitted three term method is developed for the numerical treatment of a class of linear second order singularly perturbed differential-difference equations (SPDDEs) which involves the small delay in un-differentiated term. The solution of such equations with the interval and boundary conditions exhibits twin layer or oscillatory behaviour. The method uses the Taylor's series expansion for constructing an equivalent valid version of the original problem first and then, to derive a new three term finite difference recurrence relationship/scheme. The non-uniformity in the solution is resolved by the introduction of a suitable fitting parameter in the derived new scheme. Finally the resulting system of algebraic equations is solved by the well known "discrete invariant algorithm." Method is analyzed for the stability and convergence, and the theory is illustrated by solving several test example problems. Computational results are tabulated and compared to show the applicability, accuracy and efficiency of the method. Background: The new zoonotic viral infection, monkeypox, is a global health issue. Our study aimed at studying the epidemiology, clinical presentation, complications, case fatality rate, and transmission among the present cases of monkeypox infection. Methods: Articles were searched in PubMed, Google Scholar, and Science Direct databases using the keywords "Monkeypox" [MeSH] or "Monkeypox virus" (MeSH). Narrative reviews, conference abstracts, commentaries, and articles in languages other than English were excluded. Results: From three databases, 1442 studies were identified. Seven hundred ten articles were excluded because they included data before 2022, leaving 732 items for screening. After filtering 320 data due to data duplication, 412 remained. Due to the inclusion of systematic reviews, meta-analyses, reviews, comments, and articles in languages other than English, 257 were excluded. Eligibility based on full-text review was applied to the remaining 155, excluding 129. So, the study covered a total of remaining 26 articles. We studied 2352 confirmed cases from published literature, accounting for approximately 4% of infected cases worldwide. Around 81.71% of patients have a bisexual or men having sex with men (MSM) preference. Approximately 30.18% of confirmed cases were HIV positive. Male sex was also identified as a risk factor in our review. Conclusion: Monkeypox human-to-human and human-to-animal transmission are rising. Thus, it is essential to do research on the prevention, clinicodemographic trends, and treatment of monkeypox. Background: Blood gas analysis is integral to assessing emergency department (ED) patients with acute respiratory or metabolic disease. Arterial blood gas (ABG) is the gold standard for oxygenation, ventilation, and acid–base status but is painful to obtain. Peripheral venous blood gas (VBG) is a valuable alternative as it is less painful and easy to collect. The comparability of ABG and VBG was studied in various conditions. But in hypotension, previous findings were inconsistent. So, we studied the correlation and agreement between ABG and VBG in hypotensive patients. Methodology: The study was conducted at the emergency department of a tertiary healthcare center in Northern India. Patients with hypotension above 18 years who satisfied the inclusion criteria were clinically evaluated. Patients who require ABG as a part of routine care were sampled. ABG was collected from the radial artery. VBG was obtained from the cubital or dorsal hand veins. Both samples were collected within 10 min and were analyzed. All ABG and VBG variables were entered in premade proforma. The patient was then treated and disposed of according to institutional protocol. Results: A total of 250 patients were enrolled. The mean age was 53.25 ± 15.71 years. 56.8% were male. The study included 45.6% septic, 34.4% hypovolemic, 18% cardiogenic, and 2% obstructive shock patients. The study found a strong correlation and agreement for ABG and VBG pH, pCO2, HCO3, lactate, sodium, potassium, chloride, ionized calcium, blood urea nitrogen, base excess, and arterial/alveolar oxygen ratio. Hence, regression equations were made for the aforementioned. There was no correlation observed between ABG and VBG pO2 and SpO2. Our study concluded that VBG could be a reasonable alternative for ABG in hypotensive patients. We can also mathematically predict values of ABG from VBG using regression equations derived. Conclusions: ABG sampling causes most unpleasant experiences to patients and is associated with complications like arterial injury, thrombosis, air or clotted-blood embolism, arterial occlusion, hematoma, aneurysm formation, and reflex sympathetic dystrophy. The study has shown strong correlations and agreements for most ABG and VBG parameters and can predict ABG mathematically using regression formulas formulated from VBG. As the title of the present article shows, it highlights the three philosophically integrated areas – (1) pramāṇa-epistemology (theory of comprehensive knowledge involving both perception and inference), (2) logic (although a part of pramāṇa-epistemology, it has two modes, namely, inductive reasoning and deductive reasoning), and (3) language (or semantics, i.e. the double negation theory of meaning, which falls under inference). These are interconnected as well as overlapping within the Buddhist mainstream tradition of the process philosophy as opposed to the substantialist philosophy. The same is the case with the three celebrated Buddhist thinkers – Vasubandhu, Dignāga (also spelt as Diññāṇa), and Dharmakīrti – who develop their radical and critical views focusing on these areas in historical-cum-philosophical order. It is worth noting that within the same mainstream Buddhist tradition, each one of the three thinkers picks up the problematic issues from their predecessors – from the Buddha to their immediate predecessors respectively – for their solutions against the backdrop of the two conflicting mainstream traditions – Buddhist and non-Buddhist. The central focus of these thinkers is first to identify the crucial issues, doctrinal principles, terminology, and methodology in their own ways and conceptual frameworks, which generate not only the mutual conflicts in the course of dialogues but also strengthen their positions by means of their new radical ideas, innovations, terminologies, methodologies, and doctrinal principles. As a result, the three selected areas and their crucial issues are explained, elaborated, and interpretated for better understanding. All of which are rooted in the Buddha's path of wisdom, ethics, and liberation from the human predicament (duḥkha-nivṛtti). In this grand project of the deepest concerns, the Buddha utilized multiple strategies like understanding and controlling the problematic nature of the mind (Pāli citta, manasa) and its concomitance (Pāli cetasika, dhammā) by means of the concentrative meditation (Pāli jhāna, Sanskrit (hereafter Skt., dhyāna), cultivation of knowledge (Pāli vijjā, Skt. vidyā) and conduct/moral purity (Pāli caraṇa, Skt. ācaraṇa), destruction of afflictions/defilements (Pāli kilesa, Skt. kleśa), critical and logical thinking with valid arguments, and so on. His disciples also treat him as the possessor of valid method, arguments, meaning, practice, and purpose (Skt. pramāṇabhūta, the term used by Dignāga). He believed in the common humanity as the community of sufferers and the autonomy of every human being (Pāli attakāra), but strongly rejected the hierarchy of humanity on the basis of caste, birth, and dogmatic religious identity. For these reasons, following the Buddha and his celebrated followers like Vasubandhu, Dignāga, and Dharmakīrti, my task in this article is how to clearly and elaborately discuss the above identified issues and theories, first to understand them for myself and then logically prove the whole process of knowledge and the designed purpose through communication to those who have the intention to hear and understand the framework of common language for their benefits. I wish the readers like students and young teachers benefit from my research work. Further, since my learning of the Tibetan language is zero, but comfortable in Sanskrit and Pāli, I have been heavily dependent on three great modern thinkers who have widely written independently and also translated the Buddhist Tibetan texts, which were translated from the original Sanskrit texts now lost, into English in the areas of Buddhist epistemology, logic, and semantics. These modern scholars are Masaaki Hattori, Shoryo Katsura, and Richard Hayes. Besides them, I have also little benefitted from some other scholars who have worked in the same areas. Misregulation of gut function and homeostasis impinges on the overall well‐being of the entire organism. Diarrheal disease is the second leading cause of death in children under 5 years of age, and globally, 1.7 billion cases of childhood diarrhea are reported every year. Accompanying diarrheal episodes are a number of secondary effects in gut physiology and structure, such as erosion of the mucosal barrier that lines the gut, facilitating further inflammation of the gut in response to the normal microbiome. Here, we focus on pathogenic bacteria‐mediated diarrhea, emphasizing the role of cyclic adenosine 3',5'‐monophosphate and cyclic guanosine 3′,5′‐monophosphate in driving signaling outputs that result in the secretion of water and ions from the epithelial cells of the gut. We also speculate on how this aberrant efflux and influx of ions could modulate inflammasome signaling, and therefore cell survival and maintenance of gut architecture and function. To date, insect scale robots capable of controlled flight have used flapping-wings for generating lift, but this requires a complex and failure-prone mechanism. A simpler alternative is electrohydrodynamic (EHD) thrust, which requires no moving mechanical parts. In EHD, corona discharge generates a flow of ions in an electric field between two electrodes; the high-velocity ions transfer their kinetic energy to neutral air molecules through collisions, accelerating the gas and creating thrust. We introduce a fabrication process for EHD thruster based on 355 nm laser micromachining, which potentially allows for greater materials selection, such as fiber-based composites, than is possible with semiconductor-based lithographic processing. Our four-thruster device measures 1.8 × 2.5 cm and is composed of steel emitters and a lightweight carbon fiber mesh. We measured the electrical current and thrust of each thruster of our four-thruster design, showing agreement with the Townsend relation. The peak thrust of our device, at 5.2 kV, was measured to be 3.03 times its 37 mg (363.0 μN) mass using a precision balance. In free flight, we demonstrated liftoff at 4.6 kV. [ABSTRACT FROM AUTHOR]

Autosomal dominant polycystic kidney disease (ADPKD) is caused by mutations in PKD1 and PKD2 encoding polycystin‐1 (PC1) and polycystin‐2 (PC2), respectively. The molecular pathways linking polycystins to cyst development in ADPKD are still unclear. Intracystic fluid secretion via ion transporters and channels plays a crucial role in cyst expansion in ADPKD. Unexpectedly, we observed significant and selective up‐regulation of NHA2, a member of the SLC9B family of Na+/H+ exchangers, that correlated with cyst size and disease severity in ADPKD patients. Using three‐dimensional cultures of MDCK cells to model cystogenesis in vitro, we showed that ectopic expression of NHA2 is causal to increased cyst size. Induction of PC1 in MDCK cells inhibited NHA2 expression with concordant inhibition of Ca2+ influx through store‐dependent and ‐independent pathways, whereas reciprocal activation of Ca2+ influx by the dominant negative membrane‐anchored C‐terminal tail fragment of PC1 elevated NHA2. We showed that NHA2 is a target of Ca2+/NFAT signalling and is transcriptionally induced by methylxanthine drugs such as caffeine and theophylline, which are contraindicated in ADPKD patients. Finally, we observed robust induction of NHA2 by vasopressin, which is physiologically consistent with increased levels of circulating vasopressin and up‐regulation of vasopressin V2 receptors in ADPKD. Our findings have mechanistic implications on the emerging use of vasopressin V2 receptor antagonists such as tolvaptan as safe and effective therapy for polycystic kidney disease and reveal a potential new regulator of transepithelial salt and water transport in the kidney. Land use changes in the wake of urbanization have arranged the urban residential sites to be encircled by anthropogenic activities, resulting in greater exposure to air pollution. There are limited studies carried out on source apportionment of urban landscape-induced air pollution and their spatial representativeness for urban residential sites, and the present study addressed this issue. Characterization, dissemination analysis, and source apportionment of particulate matter (PM)2.5 in a residential area, Lucknow have been carried out. Samples of PM2.5 were collected during mid-winter 2021–2022 at near-road-locations (viz. L1, L2, L3, and L4) of the study area. Samples of PM2.5 chemical speciation were done by inductively coupled plasma mass spectrometry and ion chromatography for 26-elements and 5-ions, respectively. Mean PM2.5 was recorded highest at L3 in the daytime and lowest at L4 in the nighttime with the values 166 μg/m3 and 76 μg/m3 respectively. ArcGIS-inverse distance weighing (IDW) simulations identified the representativeness of PM2.5 for the entire residential site. IDW mapping identified the PM2.5 propagation up to 200 m–500 m over the study area. The diversity in the concentration of PM2.5 observed within the residential area due to the influence of the respective day and night times, multiple sources mix and their spatial distribution. Chemical speciation data of PM2.5 applied to Positive Matrix Factorization version 5.0—as receptor model (PMF v5.0) run for individual sites separately and pool all four sites' data together as a single dataset, which ultimately confirmed seven factors. The PMF model outputs ascertained the mean apportionment of PM2.5 by the determined seven local sources that is, vehicular exhaust—21%, resuspended road dust—15%, cooking fuel emission—12%, waste burning—12%, diesel generator sets exhaust—10%, secondary aerosols—10%, and Construction and Demolition—7%. The remaining 13% mean contribution to PM2.5 was evidenced by unaccounted sources. The present study outcomes qualify the significance to undertake source apportionment studies in connection with urban landscape patterns for air pollution abatement specifically for rapidly changing cities. Endosomes have emerged as a central hub and pathogenic driver of Alzheimer's disease (AD). The earliest brain cytopathology in neurodegeneration, occurring decades before amyloid plaques and cognitive decline, is an expansion in the size and number of endosomal compartments. The strongest genetic risk factor for sporadic AD is the e4 allele of Apolipoprotein E (ApoE4). Previous studies have shown that ApoE4 potentiates presymptomatic endosomal dysfunction and defective endocytic clearance of amyloid beta (Aβ), although how these two pathways are linked at a cellular and mechanistic level has been unclear. Here, we show that aberrant endosomal acidification in ApoE4 astrocytes traps the low-density lipoprotein receptor-related protein (LRP1) within intracellular compartments, leading to loss of surface expression and Aβ clearance. Pathological endosome acidification is caused by e4 risk allele-selective down-regulation of the Na+/H+ exchanger isoform NHE6, which functions as a critical leak pathway for endosomal protons. In vivo, the NHE6 knockout (NHE6KO) mouse model showed elevated Aβ in the brain, consistent with a causal effect. Increased nuclear translocation of histone deacetylase 4 (HDAC4) in ApoE4 astrocytes, compared with the nonpathogenic ApoE3 allele, suggested a mechanistic basis for transcriptional downregulation of NHE6. HDAC inhibitors that restored NHE6 expression normalized ApoE4-specific defects in endosomal pH, LRP1 trafficking, and amyloid clearance. Thus, NHE6 is a downstream effector of ApoE4 and emerges as a promising therapeutic target in AD. These observations have prognostic implications for patients who have Christianson syndrome with loss of function mutations in NHE6 and exhibit prominent glial pathology and progressive hallmarks of neurodegeneration. The pH of the endolysosomal system is tightly regulated by a balance of proton pump and leak mechanisms that are critical for storage, recycling, turnover, and signaling functions in the cell. Dysregulation of endolysosomal pH has been linked to aging, amyloidogenesis, synaptic dysfunction, and various neurodegenerative disorders, including Alzheimer's disease. Therefore, understanding the mechanisms that regulate luminal pH may be key to identifying new targets for managing these disorders. Meta-analysis of yeast microarray databases revealed that nutrient-limiting conditions inhibited the histone deacetylase (HDAC) Rpd3 and thereby up-regulated transcription of the endosomal Na+/H+ exchanger Nhx1, resulting in vacuolar alkalinization. Consistent with these findings, Rpd3 inhibition by the HDAC inhibitor and antifungal drug trichostatin A induced Nhx1 expression and vacuolar alkalinization. Bioinformatics analysis of Drosophila and mouse databases revealed that caloric control of the Nhx1 orthologs DmNHE3 and NHE6, respectively, is also mediated by HDACs. We show that NHE6 is a target of the transcription factor cAMP-response element- binding protein (CREB), a known regulator of cellular responses to low-nutrient conditions, providing a molecular mechanism for nutrient- and HDAC-dependent regulation of endosomal pH. Of note, pharmacological targeting of the CREB pathway to increase NHE6 expression helped regulate endosomal pH and correct defective clearance of amyloid Aβ in an apoE4 astrocyte model of Alzheimer's disease. These observations from yeast, fly, mouse, and cell culture models point to an evolutionarily conserved mechanism for HDAC-mediated regulation of endosomalNHEexpression. Background and objective: Bleeding from the upper gastrointestinal (GI) tract is one of the common medical emergencies. In this study, we assessed patients' socio-demographic and clinical characteristics and the association of clinical characteristics with treatment outcomes among patients with suspected upper gastrointestinal bleed (UGIB) presenting to the emergency department (ED). At present, there is a scarcity of data on UGIB in Northern part of India. Material and method: The study was a single-center, prospective observational study conducted at an urban tertiary care center. Consecutive patients with suspected UGIB were enrolled in the study from August 2020 to February 2022. A detailed history was obtained, including demographic data such as age and sex, presenting complaints, history of presenting illness, history related to co-morbidities, addiction, and drug history. Pre-endoscopic Rockall and Glasgow-Blatchford Score were calculated for each patient. The patients were subsequently followed up till discharge from the hospital. The final outcomes with regard to mortality, need for blood transfusion, length of emergency department stay, and discharge were noted. Result: 141 patients were included in the study. The mean age of the patients with suspected UGIB was 48 ± 14 years. 115 (81.6%) patients were male. The most common co-morbidity was chronic liver disease (40;28.4%). The most frequent presenting complaint in this study was hematemesis (96; 68.1%), followed by melena (76;53.9%). The mean (Standard Deviation, SD) of the Rockall Score was 2.46 ± 1.75. The mean (SD) of the Glasgow Blatchford Score was 12.46 ± 3.15 in patients with UGIB. Conclusion: In our study, hematemesis was the most prevalent symptom of suspected UGIB, followed by melena. Portal hypertension was the most common cause of UGIB. Most frequent comorbidities in patients suspected of UGIB were alcohol intake, Nonsteriodal Antiinflammatory Drugs (NSAIDs) abuse, and co-morbidities such as underlying chronic liver disease, hypertension, and diabetes. PURPOSE: The purpose of the study was to evaluate the long-term efficacy and safety of intravitreal bevacizumab (IVB) in the treatment of choroidal neovascularization (CNV) secondary to angioid streaks (AS). MATERIALS AND METHODS: Noncomparative, interventional retrospective case series involving ten patients (15 eyes) with a minimum follow-up of 25 months following IVB for AS-associated CNV. Demographic and clinical details at baseline and during follow-up were collected from patient records. Detailed clinical examination was followed by fundus fluorescein angiography and optical coherence tomography to confirm CNV. Both primary and recurrent CNVs were treated with monthly injections of IVB till the lesion stabilized. Primary outcome measures were the percentage of patients with stable or improved visual acuity (VA) and with stable or decreased central retinal thickness (CRT) at the last visit. Secondary outcome measure was the incidence of ocular and systemic complications. RESULTS: Recurrence was observed in 11 eyes (73.33%) over a mean follow-up of 57.33 months (range: 25-100). A mean number of injections administered was 5.60. VA improved or stabilized in 73.33% and deteriorated in 26.67% of eyes at the final visit. Mean CRT improved from 324.40 µm at baseline to 265.53 µm at final visit, which was statistically significant. Complications observed were ocular hypertension in one patient and thromboembolic event in another patient. CONCLUSIONS: IVB appears to be a safe and effective option to treat CNV and to preserve vision over a prolonged period. It cannot eliminate the risk of recurrent CNV indicating the need for more effective treatments to arrest this visually debilitating condition. [ABSTRACT FROM AUTHOR]

The present research aims to investigate the effect of oxide dispersion on the synthesis of High-Entropy (HE) alloy and its properties. For this FeNiCoCrAlMn High-Entropy (HE) alloy dispersed with 2 wt percentage alumina (Al 2 O 3 ) was synthesized by mechanical activated synthesis. The mechanical activation was carried out by milling the elemental powder mixture of aluminum, cobalt, chromium, iron, nickel and manganese in an equiatomic ratio up to 20 h 2 wt% of Al 2 O 3 was added during mechanical alloying to obtain oxide dispersed HE alloy. Upon pyrolysis at high temperature FCC emerged as major phase in both alloys (with and without oxide dispersion). The alumina dispersed HE alloy shows increase in size of AlNi and CrFeMn rich phase after sintering, relatively less thermal expansion coefficient and better oxidation resistance than HE alloy without oxide dispersion. Hardness was found to be doubled after adding 2% alumina. The obtained result indicate that oxide dispersed HE alloy can be useful material for high temperature coating application. Early intervention may be key to safe and effective therapies in patients with Alzheimer disease. Endosomal dysfunction is an early step in neurodegeneration. Endosomes are a major site of production of Aβ peptide from the processing of amyloid precursor protein (APP) by clipping enzymes (β- and γ-secretases). The β-secretase enzyme BACE1 requires acidic lumen pH for optimum function, and acid pH promotes Aβ aggregation. The Na+/H+ exchanger NHE6 provides a leak pathway for protons, limiting luminal acidification by proton pumps. Like APP, NHE6 expression was induced upon differentiation of SH-SY5Y neuroblastoma cells and localized to an endosomal compartment. Therefore, we investigated whether NHE6 expression altered APP localization and processing in a stably transfected cell culture model of human APP expression. We show that coexpression with NHE6 or treatment with the Na+/H+ ionophore monensin shifted APP away from the trans-Golgi network into early and recycling endosomes in HEK293 cells. BACKGROUND: Acute myocardial infarction (AMI), often known as a heart attack, is a medical emergency that is the top cause of mortality for both men and women worldwide. Recent evidence suggests that it is caused by the sudden blockage of a coronary artery by a blood clot, also known as coronary thrombosis. Due to the regional impending of blood supply, heart muscles become damaged. Uric acid has been shown in large cohort studies to be an important independent risk factor for cardiovascular mortality. As serum uric acid (SUA) is an early predictor hence study will help to devise a better remedy for AMI. AIM: This study aims to study the levels of SUA in patients with AMI as compared to the control. OBJECTIVE: The primary goal of this study is to examine SUA levels in patients with AMI and to correlate uric acid levels with associated mortality in AMI. The secondary goal is to predict AMI early diagnosis, better management and treatment, and prognosis. METHODS: The current study was conducted at the department of biochemistry and the central investigation laboratory at our institute, in partnership with the department of medicine, intense cardiac care units, medical intensive care units, and emergency and private hospitals. A total of 100 patients were investigated. There were 50 instances of AMI and 50 age-matched healthy controls. The Microlab 300 analyzers (semi-autoanalyzer) were used to assess SUA concentrations, and the comparisons between the two groups were examined using an unpaired t-test. A P < 0.05 was deemed statistically significant. RESULTS: There was a substantial rise in the mean level of SUA in cases on the 0, 3rd, and 7th days were 8.503 + 2.976 mg/dl, 5.472 + 2.683 mg/dl, and 4.992 + 1.707 mg/dl day, respectively, which were higher than controls group (3.976 + 0.947 mg/dl). Comparison between control and AMI SUA on 0 and 3rd days was found to be more significant than the 7th day. There was a substantial rise in blood uric acid levels in cases when compared to healthy controls (P < 0.005). Segmentation plays an essential role in the design of the automated karyotyping system (AKS). It is pivotal to segment interphase cells and other debris usually found in the input G metaphase images. The performance of AKSs is considerably less when interphase cells and debris are present in the input images. In this article, two semantic segmentation models are proposed. For this experiment, an annotated dataset is generated from the G banded metaphase images which are prepared at Regional Cancer Centre (RCC), Thiruvananthapuram, Kerala, India. Inspired by the results of UNet, a lighter version L‐UNet is developed and experimented with. It shows the validation IoU (Intersection over Union) of 0.9809 and F1‐score of 0.9903 on the RCC dataset and the test IoU of 0.9720 and F1‐score of 0.9858 on the CRCN‐NE dataset. As backbone semantic segmentation models are state of the art, an efficient model, Eff‐UNet, is also proposed here. In this model, EfficientNetB03 acts as the backbone that extracts powerful features and UNet acts as the decoder that predicts the segmentation map. It performs with the validation IoU of 0.9842 and F1‐score of 0.9920 on the RCC dataset and the test IoU of 0.7545 and F1‐score of 0.7778 on the CRCN‐NE dataset. To derive this model, 25 encoder–decoder architectures are evaluated with various top‐performing CNNs (convolutional neural networks) as encoders and segmentation networks as decoders. The aim of this study was to investigate the healing of human extraction sockets filled with β-tricalcium phosphate and type I collagen (β-TCP/Clg) cones with or without a barrier membrane. Twenty patients were divided in two groups: (A) β-TCP/Clg non-membrane and (B) β-TCP/Clg + barrier membrane. Clinical examination and biopsies from the grafted sites were collected 9 months later. Bone samples were analyzed using histomorphometry and immunohistochemistry. The horizontal dimension of the alveolar ridge was significantly reduced 9 months after socket preservation in the non-membrane group. There was bone formation with no significant differences between the two groups in the areas occupied by new bone (A = 42.4%; B = 45.3%), marrow (A = 42.7%; B = 35.7%), or residual graft (A = 9.7%; B = 12.5%). Immunohistochemistry revealed osteonectin expression in both groups. Both groups demonstrated sufficient amounts of vital bone and socket morphology to support dental implant placement after the 9-month healing period. Efficacy of certain plant extracts in reducing Bean common mosaic potyvirus strain blackeye cowpea mosaic (BCMV-BlCM) disease in cowpea was evaluated. All the six botanicals Azadirachta indica, Boerhaavia diffusa, Bougainvillea spectabilis, Clerodendrum inerme, Psidium guajava, and Thuja occidentalis improved the germination and vigour index of cowpea. The disease incidence was reduced to 7% when 0.75% (w/v) of B. diffusa leaf extract was used as seed treatment under screen house conditions when compared to 80% in control. Under field conditions B. diffusa reduced the disease incidence up to 40% at 0.75% (w/v) concentration of extract. In spray treatment, B. diffusa and B. spectabilis reduced the disease incidence up to 13 and 12% under screenhouse conditions, whereas B. diffusa and C. inerme reduced the disease incidence up to 31 and 32% under field conditions. When plant extracts were mixed with BCMV-BlCM inoculum and young seedlings inoculated, B. spectabilis, C. inerme and M. jalapa extracts reduced the disease incidence up to 42, 40 and 48% respectively under screenhouse conditions when compared with the control. Background: The World Health Organization (WHO) reports that Asia and Africa have the highest Chronic Liver Disease (CLD) mortality rate. Cirrhosis, responsible for 22.2 fatalities per 100,000 people, is India's 10th most common cause of mortality. The increasing prevalence of chronic liver disease necessitates a study to identify predictive factors for patients who visit the emergency department. Identifying elements that enhance the predictive value of mortality in unstable patients with CLD complications is important in emergency departments. This study aims to determine Clinical and Laboratory Parameters as mortality predictors in adult chronic liver disease patients. Methodology: The study was conducted at the emergency department of a tertiary healthcare center in Northern India. Patients with chronic liver disease above 18 years of age who satisfied the inclusion criteria were clinically evaluated. Clinical and demographic details were collected, and data was analyzed. Results: Two hundred thirty-six patients were enrolled. The mean age was 50.77 ± 14.26 years. 78.4% of the participants were men. Abdominal distension, affecting 59.7% of patients, was the most common presenting ailment, followed by melena and hematemesis, affecting 41.9% and 32.6%, respectively. The mean stay in the emergency department was 10.29 ± 8.10 h. Refractory septic shock, the leading cause of mortality, accounts for 69.2% of all deaths, alongside grade 4 hepatic encephalopathy and massive Upper Gastrointestinal (UGI) bleeding, as identified in our study. Factors such as altered mental sensorium, high respiratory rate, low SpO2, increased heart rate, low systolic blood pressure, low diastolic blood pressure, and low Glasgow Coma Scale (GCS) on Emergency Department (ED) arrival are significantly associated with mortality. Conclusions: Chronic liver disease, a prevalent condition in India, most commonly seen in middle aged men and lower socioeconomic groups. The parameters independently associated with mortality in our study were presence of altered mental sensorium, Glasgow coma scale, Child Pugh class and need for ICU admission. The plywood manufacturing process involves debarking of log, peeling and pressing of ribbons of wood after glueing with proper resin, trimming, sanding and finishing. Washings of glue mixer, resin kettle and a glue spreader generate solids and various hazardous chemicals like phenol, sodium hydroxide and formaldehyde. To protect the environment and human health, it is necessary to treat and reuse the wastewater of plywood manufacturing. There are several methods available for the treatment of plywood industries wastewater and use of lime is one of them. In the present study viability of lime treatment was performed to achieve zero liquid discharge in a Plywood Industry. An optimum lime dose of 1.5 g/L was applied to the wastewater of melamine urea formaldehyde and phenol formaldehyde resin units. After treatment, removal of 28.6% and 41.2% total dissolved solid and 36.8% and 37.5% total suspended solid were observed in melamine urea formaldehyde and phenol formaldehyde resin units wastewater. Phenol and chemical oxygen demand removals were more than 38% and 40% from the wastewater of both units. Lime treated wastewater was used to prepare melamine urea formaldehyde and phenol formaldehyde glue/resin by mixing chemicals. The prepared resins exhibit characteristics within prescribed limits of plywood manufacturing. The mechanical and physical properties of plywood specimen have also found satisfactory. Polycystic ovarian syndrome (PCOS) is a commonly occurring reproductive disorder among the reproductive-aged women. Its global occurrence varies based on diagnostic guidelines, ethnicities, and locations of concern. Insulin resistance (IR) is commonly observed around 65–70% of women diagnosed with PCOS, representing a prevalent association. Consequently, the study was designed with an objective of illustrating the effect of insulin on mural and cumulus granulosa cells (GCs) of PCOS patients in comparison to normal ovulating women. This study is a case–control design, wherein a total of 80 participants were recruited meeting criterion of inclusion and exclusion, divided into 8 groups with each group consisting of 10 samples. The process involves the isolation and culturing of mural granulosa cells (MGC) and cumulus granulosa cells (CGC) with and without exposure to insulin. The proteins released by untreated GCs and insulin-treated GCs were extracted, and complex protein mixtures were digested with trypsin, followed by tandem mass spectrometry analysis and data processing using bioinformatics. We found 595 proteins in both control and PCOS samples, of which 310 were contributed by MGCs and 285 by CGCs. The PCOS MGCs expressed 20%, both the normal MGCs and CGCs have equal representation of 16% by each, whereas the PCOS CGCs proteins contributed 15% of the total of the proteomic expression. However, the poor expression observed with the Insulin exposure, the Insulin treated PCOS CGCs contributes 13%, PCOS MGCs contributes 8%. The normal MGCs upon the Insulin treatment give 8% then and there only 4% of proteins expressed by normal CGCs after Insulin treatment. The Venn analysis widened on their precise expression topographies. The examination of strings exhibited important protein–protein interaction pathways. This is a pioneering investigation aimed to establish the link between hyperinsulinemia in localized follicular GCs and PCOS mechanisms by comparing them to control group. The examination of various attributes, mechanisms, and traits shown by genes and proteins in individuals with PCOS compared to control populations, alongside the investigation of the dynamics of these genes and proteins following exposure to insulin, holds promise for the formulation of novel hypotheses and strategies in the identification of new biomarkers. Background: The reproductive system is heavily dependent on ovarian follicles, which are made up of germ cells (oocytes) and granulosa cells (GCs), including cumulus granulosa cells (CGCs) and mural granulosa cells (MGCs). Understanding their normal and steroid-induced functions is the key to understanding the pathophysiology of endocrinal diseases in women. Objective: This study investigated the differentially expressed proteins by CGCs and MGCs of patients with polycystic ovarian syndrome (PCOS) and without subsequent exposure to dehydroepiandrosterone sulfate (DHEAS) and functional differentiation. Design: The present study was observational and experimental study carried out in hospital involving 80 female patients undergoing IVF for infertility. Methods: In this study, we isolated CGCs and MGCs from the follicular fluid of both PCOS and non-PCOS patients undergoing in vitro fertilization (IVF). The cells were cultured and treated with DHEAS for 48 hours, and these cells were extracted, digested, and analyzed by tandem mass spectrometry followed by processing of the results using open-source bioinformatics tools. Results: The present investigation discovered 276 and 341 proteins in CGCs and MGCs, respectively. DHEAS reduced the number of proteins expressed by CGCs and MGCs to 34 and 57 from 91 and 94, respectively. Venn results of CGCs revealed 49, 53, 36, and 21 proteins in normal CGCs, PCOS-CGCs, post-DHEAS, and PCOS-CGCs, respectively. Venn analysis of MGCs showed 51 proteins specific to PCOS and 29 shared by normal and PCOS samples after DHEAS therapy. MGCs express the most binding and catalytic proteins, whereas CGCs express transporter-related proteins. A protein pathway study demonstrated considerable differences between normal and PCOS samples, while DHEAS-treated samples of both cell lines showed distinct pathways. String findings identified important network route components such as albumin, actin, apolipoprotein, complement component C3, and heat shock protein. Conclusion: This is the first study to show how DHEAS-induced stress affects the expression of proteins by MGCs and CGCs isolated from normal and PCOS patients. Introduction: β‐Tricalcium phosphate (β‐TCP) is a synthetic osteoconductive scaffold that is used as an alternative to autogenous bone grafts. The purpose of this case series was to examine the effectiveness of a polymer‐coated β‐TCP particulate graft in preserving alveolar ridge height after tooth extraction. Effectiveness was evaluated using radiographic measurements, together with a histomorphometric evaluation of bone formation. Case Series: Eight patients, treatment planned for extraction and dental implant placement in the posterior region, were consecutively enrolled in the case series. Study teeth were extracted, and an alloplastic bone substitute, consisting of synthetic β‐TCP granules coated with a biodegradable polymer (i.e., poly[lactic‐co‐glycolic] acid [PLGA]), was placed and adapted into the extraction socket. Patients were recalled at 1, 2, and 4 weeks and 3 months postoperatively to evaluate wound healing and at 4 to 5 months for implant placement. Bone specimens were collected at osteotomy preparation. Healing was uneventful at each graft site. Five sites were available for surgical reentry and implant placement; the sites exhibited ≤10% reduction in radiographic bone height. Histologic evidence of vital bone growth was evident at each grafted site. Conclusions: The PLGA‐coated β‐TCP bone substitute exhibited good stability and retention after socket grafting. Ring‐expansion strategies are valuable synthetic tools that take benefit of existing ring structures and evade the unfavorable enthalpic‐and entropic effects that arise with end‐to‐end cyclizations. One potentially important class of such reactions is the Dowd–Beckwith reaction, the ring‐expansion of ketones via alkoxy radicals. The exciting advancement in this research area is starting to show its potential, as demonstrated by applying this methodology in strategy‐level bond formation to synthesize complex natural products. This Review aims to provide the first comprehensive survey of the development of the Dowd–Beckwith reaction spanning three decades from the initial report to the present day, thus providing the readers with great detail about the contributions of this reaction to organic synthesis. Among all functional groups, alkynes occupy a privileged position in synthetic and medicinal chemistry, chemical biology, and materials science. Thioalkynes, in particular, are highly useful, as they combine the enhanced reactivity of the triple bond with a sulfur atom frequently encountered in bioactive compounds and materials. Nevertheless, general methods to access these compounds are lacking. In this article, we describe the mechanism and full scope of the alkynylation of thiols using ethynyl benziodoxolone (EBX) hypervalent iodine reagents. Computations led to the discovery of a new, three-atom concerted transition state with a very low energy barrier, which rationalizes the high reaction rate. On the basis of this result, the scope of the reaction was extended to the synthesis of aryl- and alkyl-substituted alkynes containing a broad range of functional groups. New sulfur nucleophiles such as thioglycosides, thioacids, and sodium hydrogen sulfide were also alkynylated successfully to lead to the most general and practical method yet reported for the synthesis of thioalkynes. Objectives: The aim of this study is to evaluate long‐term in vivo stability of dental implants stabilized at time of placement in oversized osteotomies with a novel, self‐setting, mineral–organic bone adhesive. Materials/Methods: Canine (26) mandibular teeth were removed, and three oversized osteotomies prepared bilaterally. Implants were placed with either adhesive, particulate xenograft, or with blood clot filling the implant/osteotomy gaps. Removal torque and histology were assessed. Results: The adhesive provided significant and clinically relevant immediate implant stability of 22.2 N‐cm (95% CI 5.3; 39.0), which continued throughout the early postoperative course and persisted through the nine‐ (155 N‐cm 95% CI 113; 197) and 12‐month (171 N‐cm 95% CI 134.2; 209.4) time points. This is in comparison with the blood clot of 1.4 N‐cm (95% CI 0.7; 2.1), 128.6 N‐cm (95% CI 66.8; 190.4), and 140.7 N‐cm (95% CI 78.8; 202.5) and particulate xenograft, 1.3 N‐cm (95% CI 0.6; 2.0), 132.1 N‐cm (95% CI 94.5; 169.7), and 101.5 (95% CI 59.5; 143.5), respectively. Histological examination shows the adhesive establishes intimate contact with the implant and bony walls and is replaced with new bone without compromising stability. Soft tissue does not penetrate the adhesive, and marginal bone/biomaterial level is maintained. Control sites filled with xenograft or blood clot heal with reduced bone levels, and in some cases, xenograft particles were encapsulated in connective tissue. Conclusions: Implants placed in oversized osteotomies and lacking primary stability can be stabilized at placement with a novel, highly osteoconductive, and resorbable adhesive. Gradual replacement of the biomaterial allows osseointegration without loss of stability through 12 months of follow‐up. Abstract: Electrocardiogram (ECG) is widely used as a diagnostic tool to identify atrial tachyarrhythmias such as atrial fibrillation. The ECG signal is a P-QRS-T wave representing the cardiac function. The minute variations in the durations and amplitude of these waves cannot be easily deciphered by the naked eye. Hence, there is a need for computer aided diagnosis (CAD) of cardiac healthcare. The current paper presents a methodology for ECG based pattern analysis of normal sinus rhythm and atrial fibrillation (AF) beats. The denoised and registered ECG beats were subjected to independent component analysis (ICA) for data reduction. The weights of ICA were used as features for classification using Naive Bayes and Gaussian mixture model (GMM) classifiers. The performance and the upper bound on probability of error in classification were analyzed using Chernoff and Bhattacharyya bounds. The Naive Bayes classifier provided an average sensitivity of 99.32%, specificity of 99.33% and accuracy of 99.33%, while the GMM provided an average sensitivity of 100%, specificity of 99% and accuracy of 99.42%. The probability of error during classification was less for GMM compared to Naive Bayes classifier (NBC) as GMM provided higher performance than the NBC. [Copyright &y& Elsevier]

Choroidal osteoma is an uncommon benign osseous intraocular tumor typically seen unilaterally in young women. Visual loss can occur due to choroidal neovascularization (CNV) complicating osteoma. We report a rare case of bilateral choroidal osteoma with secondary CNV in a young male and the long-term results following anti-vascular endothelial growth factor (VEGF) therapy. A 30-year-old male with history of defective vision in both eyes since several years and recent worsening in the right eye (RE) since 2 months was found to have bilateral macular osteoma with CNV in the RE based on clinical evaluation, fluorescein angiography, optical coherence tomography, and ultrasonography. Intravitreal injection of ranibizumab at monthly intervals for three doses resulted in resolution of CNV and remained stable for 5 years. Recurrent CNV detected 6 years later responded to an injection of intravitreal bevacizumab and has remained stable till date. Anti-VEGF therapy stabilized the secondary CNV in our patient for 7 years with satisfactory structural and functional outcome, demonstrating the long-term efficacy of this modality of treatment. Objectives: The purpose of this prospective, single‐center randomized pilot study was to histologically evaluate and compare vital bone development in premolar and molar‐extraction sites grafted with two different bovine‐derived xenografts. The secondary outcome of interest was implant survival in the grafted sites. Materials and Methods: Adult patients in need of at least two tooth extractions were enrolled. A paired design was used; each patient received at least one of each type of graft at different sites. Each extraction site was randomized to one of two xenograft treatment groups. A resorbable membrane was always placed, and primary intention soft tissue closure was achieved. Four months later, implants were placed and a trephine drill was used to remove bone cores for histologic and histomorphometric analysis. Results: Sixteen patients with 40 extraction sites were enrolled; 20 sites were grafted with one type of xenograft and 20 with another. Mean patient age was 53.5 years, and 65% of patients were male. Evaluation of bone core samples taken from grafted sites showed no significant difference in the mean value of percentage of new bone formation between the different grafted sites (33.4% and 32.4%, p =.76). Cumulative implant survival was 97.5% at the 24‐month follow‐up visit. Conclusion: Within the limitations of this pilot study, no statistically significant differences in new bone growth between sites grafted with two different types of xenograft were found. Abstract: Background: Recombinant human bone morphogenetic protein‐2 in an absorbable collagen sponge carrier (rhBMP‐2/ACS) has been shown to support significant bone formation when used to augment the maxillary sinus for implant dentistry. Nevertheless, bone biomaterials have been suggested to extend rhBMP‐2/ACS with limited support of the merits of such approaches. Objectives: To evaluate local bone formation/dental implant osseointegration following implantation of rhBMP‐2/ACS combined with a ceramic bone biomaterial using a mini‐pig sinus augmentation model. Methods: Twelve adult Göttingen mini‐pigs received rhBMP‐2/ACS (rhBMP‐2 adjusted to 0.43 mg/cc) alone or combined with an off‐the‐shelf biphasic ceramic (15%/85% HA/ß‐TCP) biomaterial at 3:1, 1:1 and 1:3 ratios randomized to contra‐lateral maxillary sinus sites yielding rhBMP‐2/ACS fractions of 100%, 75%, 50% and 25%, respectively. A 4‐cc implant volume was used for all sites. Two threaded dental implants (ø4.0 × 11.5 mm) were placed at each site. The animals were euthanized at 8 weeks for histologic analysis. Results: Surgical execution and healing were generally uneventful, infraorbital local swelling was observed in all animals until suture removal. rhBMP‐2/ACS combined with the ceramic biomaterial did not significantly enhance local bone formation (range 9.0 ± 1.5 to 9.7 ± 2.1 mm) compared with rhBMP‐2/ACS alone (8.6 ± 1.1 mm; p > 0.05). Variations in rhBMP‐2/ACS to ceramic matrix ratios yielding rhBMP‐2 doses approximating 0.4, 0.9, 1.3 and 1.7 mg/sinus did not appreciably influence bone formation/osseointegration. Conclusions: Whereas rhBMP‐2/ACS supports significant bone formation/osseointegration in the mini‐pig sinus augmentation model and thus appears an effective alternative for sinus augmentation procedures, adding a ceramic biomaterial to rhBMP‐2/ACS does not produce meaningful biological advantages. Understanding what determines species’ geographic distributions is crucial for assessing global change threats to biodiversity. Measuring limits on distributions is usually, and necessarily, done with data at large geographic extents and coarse spatial resolution. However, survival of individuals is determined by processes that happen at small spatial scales. The relative abundance of coexisting species (i.e. ‘community structure’) reflects assembly processes occurring at small scales, and are often available for relatively extensive areas, so could be useful for explaining species distributions. We demonstrate that Bayesian Network Inference (BNI) can overcome several challenges to including community structure into studies of species distributions, despite having been little used to date. We hypothesized that the relative abundance of coexisting species can improve predictions of species distributions. In 1570 assemblages of 68 Mediterranean woody plant species we used BNI to incorporate community structure into Species Distribution Models (SDMs), alongside environmental information. Information on species associations improved SDM predictions of community structure and species distributions moderately, though for some habitat specialists the deviance explained increased by up to 15%. We demonstrate that most species associations (95%) were positive and occurred between species with ecologically similar traits. This suggests that SDM improvement could be because species co-occurrences are a proxy for local ecological processes. Our study shows that Bayesian Networks, when interpreted carefully, can be used to include local conditions into measurements of species’ large-scale distributions, and this information can improve the predictions of species distributions. Methods Nine adult Göttingen mini-pigs were used for evaluation of a biphasic ceramic (15%/85% HA/ß-TCP) and an allogeneic mineralized bone biomaterial. Treatments randomized to contralateral sinus sites included sham-surgery (control) and biomaterials. Two threaded dental implants (ø4.0 × 11.5 mm) were placed at each sinus site. The animals were euthanized at 8 weeks for histologic analysis. Results Execution of the surgical protocol and healing was unremarkable. Limited infraorbital swelling was observed until suture removal. The biphasic ceramic and allogeneic bone biomaterials produced significantly increased bone formation (5.2 ± 1.9 mm and 4.9 ± 1.6 mm vs. 2.6 ± 0.5 mm, p < 0.05) and osseointegration (18.0 ± 6.0% and 25.1 ± 18.2% vs. 10.1 ± 8.0%, p < 0.05) over the sham-surgery control. No significant differences were observed between biomaterials. Conclusions Implantation of biphasic ceramic or allogeneic bone biomaterials enhances bone formation in the mini-pig maxillary sinus, however, dental implant bone support is incomplete resulting in overall limited osseointegration. In this study, we present a highly efficient method for proteomic profiling of cysteine residues in complex proteomes and in living cells. Our method is based on alkynylation of cysteines in complex proteomes using a 'clickable' alkynyl benziodoxolone bearing an azide group. This reaction proceeds fast, under mild physiological conditions, and with a very high degree of chemoselectivity. The formed azide-capped alkynyl-cysteine adducts are readily detectable by LC-MS/MS, and can be further functionalized with TAMRA or biotin alkyne via CuAAC. We demonstrate the utility of alkynyl benziodoxolones for chemical proteomics applications by identifying the proteomic targets of curcumin, a diarylheptanoid natural product that was and still is part of multiple human clinical trials as anticancer agent. Our results demonstrate that curcumin covalently modifies several key players of cellular signaling and metabolism, most notably the enzyme casein kinase I gamma.
