Abstract

International audience; The photochemical behaviour of rivastigmine has been investigated under UV-B irradiation by HPLC and NMR. Kinetic data have been obtained irradiating aqueous solutions (5 x 10-5 M). For mechanistic purposes and products studies irradiation has also been carried out on concentrated solutions (1 x 10-3 M) in water/acetonitrile, acetonitrile, methanol, methanol/acetonitrile, in the presence and absence of oxygen. Photoproducts, isolated by chromatographic processes, have been identified by spectroscopic means. Photochemical cleavage of the benzyl-nitrogen bond occurs and leads to radical-derived and, mainly, ion-derived products. Mechanistic pathways have been hypothesised on the basis of both steady-state irradiation and laser flash photolysis experiments.

Abstract

α- Asarone, 9-hydroxylinolenic acid and stigmasta-4,22-diene-3,6-dione isolated from Pistia stratiotes and showing in vitro antialgal activity, have been found in the water in which the plant was cultivated. Of other antialgal compounds such as 9-hydroxy and 13-hydroxy-linoleic acids, coniferyl and sinapyl alcohols in the growth medium have also been identified. New phenyl propanoid glucosides and C13 norterpene glucosides have been isolated from the plant.

Abstract

The presence of potential algicides in Juncus effusus was studied. Sequential Et2O and MeOH exts. were made of dried plants that had been collected in spring and summer, and phenanthrenes were purified from the exts. All the free dihydrophenanthrenes and tetrahydropyrenes inhibited the test alga, Selenastrum capricornutum. One substance (I) inhibited several algae in eutrophic ecosystems with activity similar to that of CuSO4. Glycosylation severely inhibited algicidal activity of all compds. examd.

Abstract

The abiotic degradation of iodosulfuron-methyl-ester was investigated under both alkaline and acidic pH conditions in the dark, and results showed it to be a rather stable molecule in neutral or slightly alkaline environments. Photochemical reactions were studied using a high-pressure mercury arc lamp, and results showed that direct phototransformation is possible under normal environmental conditions (lambda > 290 nm). High-performance liquid chromatography (HPLC-UV and HPLC-MS) analyses were used to identify the degradates and to study the kinetics of photodecomposition and hydrolysis. Five main products of iodosulfuron-methyl-ester degradation were tentatively identified, and one of them (4-methoxy-6-methyl-1,3,5-triazin-2-amine) was confirmed using an authentic standard. Among the phototransformation mechanisms, photosubstitution of the iodide atom by a hydroxyl group, photodissociation of the N-S bond, and photoassisted hydrolysis were observed. The quantum efficiencies (multiwavelength quantum yield) of the photodegradation under different conditions were determined, and values of 0.054 +/- 0.02 (pH 9.6), 0.08 +/- 0.02 (pH 7), and 0.044 +/- 0.008 (pH 5.3) were obtained.

35. Solid-state photodimerization of steroid enones

Abstract

Androst-4-ene-3,17-dione and 17a-methyltestosterone are dimerized in the solid-state by UV radiation. These substances were selected by a search in the Cambridge Structural Database among the steroid enones presenting in the crystalline state an intermolecular short contact between a hydrogen alpha to a carbonyl group and the oxygen of an enone system. Dimerization occurs by transfer of the hydrogen to the oxygen and connection between the two involved carbons. Androst-4-ene-3,17-dione affords dimer and trimer, both formed by connection of the C16 of a molecule with the C3 of a near one. Irradiation of 17a-methyltestosterone gives two isomeric trienones. These compounds are reasonably formed by dehydration of unisolated intermediate products obtained by coupling of two molecules through a linkage between the C2 and the C3' carbons. The formation mechanisms of the photoproducts are satisfactory explained on the basis of the molecular arrangement of the monomers in the crystal state. Modelling of the dimeric molecules was done using molecular mechanics calculations. Single crystal X-ray analysis of the dimer of androst-4-ene-3,17-dione confirms the structural interpretation of spectral data. The conformer found in the solid state well agrees with the results of molecular mechanics calculations.

Abstract

Silybin is the major flavonolignan of silymarin and it displays a plethora of biological effects, generally ascribed to its antioxidant properties. Herein we shall describe an efficient synthetic strategy to obtain a variety of new and more water-soluble silybin and 2,3-dehydrosilybin (DHS) derivatives in which the 23-hydroxyl group was converted to a sulfate, phosphodiester, or amine group, using a solution-phase approach. Furthermore a new and efficient method for the preparation of DHS from silybin was developed and optimised. The antioxidant properties of the new compounds were evaluated in a cellular model in vivo and they displayed an antioxidant activity comparable to or higher than silybin and DHS, being able to prevent H(2)O(2)-induced generation of intracellular reactive oxygen species (ROS). Most of the derivatives also displayed a better hydrophilicity while retaining the biological activities of silybin and they might broaden the in vivo applications of this class of natural compounds.
