The present study was conducted on four healthy breeding Sahiwal bulls with the aim to evaluate fresh seminal characteristics of Sahiwal bull along with their methodology. Physio-morphological seminal attributes of ejaculated semen were evaluated and recorded. Overall mean seminal volume irrespective of the bull was recorded as 5.83 ± 0.15. Overall mean seminal pH in the freshly collected semen of Sahiwal bull was 6.71±0.02. Overall mean mass motility of spermatozoa in the freshly collected semen of Sahiwal bull was 4.78 ± 0.05. Seminal mass motility did not differ significantly amongst different bulls. Overall mean sperm concentration in the freshly collected semen of Sahiwal bull was 1434.72 ± 276.42. Mean value of bull number S-109 was significantly higher compared to other bulls. Overall mean percentage of live spermatozoa in freshly collected semen of Sahiwal bull was 92.91 ± 0.59. Mean value of bull number S-103 and S-109 was significantly higher as compared to other bulls. The overall mean percentage of progressively motile spermatozoa in freshly collected semen of Sahiwal bull was 84.06 ± 0.97. Adding endogenous antioxidants to the diluent is significantly associated with semen quality during the freezing process. This study aimed to investigate the effects of adding glutathione to AndroMed diluent on the preservation of crucial sperm attributes, namely, intact plasma membrane (IPM), intact acrosome membrane (IAM), and progressive motility of Bali cattle spermatozoa. A completely randomized design was used, and spermatozoa samples were obtained from a Bali cattle and divided into two diluent treatment groups (36 diluent samples in each group with six replications), namely pure AndroMed as the control and a group with the addition of glutathione (1 mmol) to AndroMed. Each treatment was replicated six times and evaluated at three freezing stages, including postdilution, post-equilibration, and post-thawing, for crucial sperm properties. The results indicated that fresh Bali cattle spermatozoa had progressive motility, IAM, and IPM of 75%, 89%, and 88%, respectively. During the freezing process, there was a significant decrease in semen quality, including progressive motility, IAM, and IPM of spermatozoa after dilution to post-equilibration and post-equilibration to post-thawing in both treatment groups. Meanwhile, the addition of 1 mmol of glutathione to AndroMed diluent had a significant difference in increasing progressive motility, IAM, and IPM of Bali cattle spermatozoa at each stage of semen freezing, including postdilution, post-equilibration, and post thawing when compared with controls. Based on the results, it can be concluded that adding 1 mmol of glutathione to the AndroMed diluent enhanced the quality and integrity of Bali cattle semen, including progressive motility, IAM, and IPM during the freezing process. Body weight, growth rate and testicular biometry of Black Bengal bucks (19) belonging to different age groups were studied. Further, this study was extended to delineate the histological changes in the testes with advancement of ages and to evaluate the seasonal variations in the important semen parameters. The body weight differed significantly across the age groups but the body weight gain (g/d) was higher in the age groups of 0–4 and 17–20 months. The mean scrotal circumference increased with the advancement of age and attained maximum circumference at the age of 12 months. The mean testicular diameter increased significantly till 10th month of age. All these biometrical parameters were highly correlated with each other, and with age and body weight. The histological examination of testes revealed an effect of age on the seminiferous tubules and stages of the spermiogenesis. At 3.5 months of age, lumen of seminiferous tubules became distinct with the abundance of spermatozoa. These observations along with testicular biometry indicated the age at which bucks attained puberty. Significant effect of the seasons (e.g., spring and summer) was observed on the semen parameters with the improvement in the progressive motility, percentage of live spermatozoa, chromatin integrity and a drop in membrane integrity during summer. These results will provide an important impetus in selection of breeding bucks of Black Bengal breed.

This study researched the impact of bacterial contamination of Lohmann brown rooster semen on the biochemical parameters of seminal plasma to evaluate its potential consequences on the sperm progressive motility. Semen was collected from 27 stud roosters, and the sperm concentration and progressive motility were measured using computer-assisted semen analysis (CASA). Seminal plasma was separated, and selected biochemical parameters were measured using commercially available assays. An aliquot of each semen sample was cultured, the colonies were counted and the MALDI Biotyper was used for bacterial identification. The samples were divided into three categories based on their sperm progressive motility and the data were compared and statistically evaluated. Moreover, Pearson’s correlation analysis was performed. The results showed that the lower the sperm progressive motility, the higher the level of colony-forming units. Moreover, sperm concentration was significantly higher (p < 0.05) in the group with the highest bacterial occurrence and the lowest proportion of progressively motile spermatozoa. Calcium, magnesium, creatinine, uric acid, alkaline phosphatase, and total proteins significantly changed in semen samples with the lowest proportion of progressive motility. In conclusion, seminal plasma biochemistry may mirror changes occurring in semen as a result of bacterial presence in the reproductive tract of poultry.

Interactions between spermatozoa and bacterial species in ejaculates comes along with deleterious effects on the structure and function of male gametes. However, the effect of bacterial load on the seminal plasma remains unexplored. Therefore, this study was focused on the identification of bacterial species present in Duroc boar semen (n=18), quantification of the bacterial load (CFU) and alterations to the biochemical parameters of seminal plasma. The computer-assisted semen analysis (CASA) analysed the progressive motility (PRO; motility=25µm/s) of spermatozoa, their concentration and kinematic parameters. The MALDI Biotyper was used for the bacterial characterization of fresh ejaculates. Seminal plasma was separated and biochemical analysis was carried out using the Randox RX Monza analyzer. Calcium (Ca), magnesium (Mg), total proteins (TP), aspartate transaminase (AST), alanine transaminase (ALT), cholesterol (CHOL), bilirubin (BR), albumins (ALB) and uric acid (UA) were subjected to the Pearson correlation with CFU and PRO. The bacterial identification showed the prevalence of Escherichia coli and Pseudomonas aeruginosa. The correlation analysis showed that the progressive motility of spermatozoa was strongly dependent (-0.799; P<0.01) on the seminal bacterial load. Moreover, a negative significant correlation (-0.810; P<0.001) was detected between CFU and Mg concentration. Also ALT (-0.647; P<0.01) and ALB (-0.484; P<0.05) were negatively affected by the bacterial load. Altogether, the presence of bacterial species may adversely affect the progressive motility as well as the biochemical composition of seminal plasma and thereby create unfavorable conditions for the sperm survival. In recent years, the demand for effective and low-toxic stimulators of rhizogenesis, which are used in microclonal propagation of plants, has been increasing in Ukraine. One of the promising directions in the search for effective compounds is molecular modeling based on known natural and synthetic compounds. The development of new highly effective and low-toxic biologically active compounds is largely based on derivatives of nitrogen-containing heterocycles, and quinoline occupies a significant place among them. Modern methods of chemometric analysis make it possible to find certain regularities in the "chemical structure – biological activity" and to select the most promising compounds for experimental research. The values of lipophilicity log P for neutral forms and the value of the distribution coefficient log D at pH = 7 were obtained by quantum chemical calculation. The values of log P and log D of the studied compounds are in the most favourable interval for overcoming the biological membranes of the cells of the root system, depending on the pH of the environment. According to Lipinski’s "rule of five", all studied compounds can show high biological activity. The toxicity of compounds of 2-((6-R-quinolin-4-yl)thio)acetic acid derivatives was evaluated by computer programs and experimentally. Among the derivatives of 2-((6-R-quinolin-4-yl)thio)acetic acid, the most toxic compounds were those that did not have alkoxy substituents in the 6th position of the quinoline ring. Sodium salts are more toxic than the corresponding acids. This is due to an increase in the bioavailability of ionized compounds. Derivatives of 2-((6-R-quinolin-4-yl)thio)acetic acid (sodium salt of 2-((quinolin-4-yl)thio)acetic acid (QAC-5) showed the greatest toxic effect on the model of the study of progressive sperm motility) and 2-((quinolin-4-yl)thio)acetic acid (QAC-1), which will reduce this indicator by 15–20% compared to intact. The toxicity assessment of the studied compounds made it possible to determine a number of factors of the structure of molecules which affect the level of toxic action of 2-((6-R-quinolin-4-yl)thio)acetic acid derivatives and the directions of creation of non-toxic growth stimulants in this series. The impact on rhizogenesis during microclonal reproduction in vitro in explants Paulownia clone 112 and further adaptation of microplants in vivo hybrid molecules of quinoline and acetic acid, which are analogues of known growth stimulants, was studied. A number of factors influencing the level of influence on rhizogenesis of the action of derivatives of 2-((6-R-quinolin-4- yl)thio)acetic acid and directions of creation of highly active substances in this series was defined. The studied compounds showed a high stimulating effect on rhizogenesis in vitro in Paulownia explants. It was established that the sodium salt of 2-((quinolin-4-yl)thio)acetic acid was the greatest stimulator of rhizogenesis compared to the corresponding original acid. The presence of alkoxy groups in the 6th position and methyl in the 2nd position of the quinoline ring of 2-((6-R-quinolin-4-yl)thio)acetic acid reduced the activity of the compounds. The selection of new effective, low-toxic, less expensive substances was carried out for further testing as potential stimulators of rhizogenesis for microclonal propagation of plants.

This comprehensive study was carried out to investigate the effects of taxifolin in the freezing medium on post-thaw semen quality, and fertility potential of buffalo bull spermatozoa. Taxifolin was also evaluated for radical scavenging activity through DPPH (2, 2-diphenyl-1-picrylhydrazyl), and Nitric oxide (NO) inhibition (%) during in vitro conditions. Collected semen samples from four buffalo bulls were initially evaluated (consistency, volume, motility, and concentrations); the accepted samples were pooled, and diluted in extenders containing different doses of taxifolin (0 μM [control], 2 μM, 5 μM, 10 μM, and 20 μM). Diluted semen was gradually cooled (2 h, 4 °C), equilibrated (4 h, 4 °C), and then frozen in liquid nitrogen (−196 °C). Data analysis revealed that taxifolin supplementation (10 μM) displayed the highest DPPH-scavenging ability, and NO inhibition compared to the control (% DPPH, 67.31 vs. 13.50; and % NO, 78.25 vs. 21.25) respectively. Moreover, taxifolin supplementation (10 μM) significantly (P < 0.05) improved progressive motility (%), average path velocity (μm/sec), straight-line velocity (μm/sec), and seminal plasma glutathione peroxidase (μM), and superoxide dismutase (U/mL) of buffalo bull spermatozoa than control. Sperm plasma membrane integrity, mitochondrial transmembrane potential, acrosome integrity (%) and seminal plasma adenosine triphosphate (nmol/million), and total antioxidant capacity (μM/L) were enhanced significantly with taxifolin (10 and 20 μM) supplementing group. Lastly, taxifolin supplementation significantly improved the fertility rate (%, 69.09 vs. 40.38) compared to the control. Further studies to assess mechanisms by which taxifolin improves semen quality and fertility of buffalo spermatozoa are warranted. [ABSTRACT FROM AUTHOR]

Simple Summary: While previous studies showed that sperm quality has decreased over the last decades, others did not report such reductions, suggesting that the region where the population is evaluated has an influence. The current research, which was conducted on a regional scale, did not find variations in sperm count but did observe a drop in sperm motility and morphology. Semen quality has a direct relation to male fertility. Whether sperm variables in humans have decreased over the last years is still uncertain, with some studies showing a decline and others reporting no changes. In this regard, previous research has suggested that lifestyle and environmental conditions may contribute to this variability, calling for regional studies. The present work is a retrospective, unicentric study that includes semen samples analyzed between 1997 and 2017 at the Parc Taulí Hospital (Barcelona metropolitan area). First, a multivariate analysis including the age as a confounding factor showed a statistically significant decrease in semen volume, pH, progressive motility, morphology and total motile sperm over time. Contrarily, no significant variation in sperm count or concentration was observed. Mean reductions per year were −0.02 mL for volume, −0.57% for progressively motile sperm and −0.72% for sperm with normal morphology. Interestingly, the average annual temperature registered by the Spanish Meteorology Agency negatively correlated to sperm morphology and sperm count (Rs = −0.642; p = 0.002 and Rs = −0.435; p = 0.049, respectively). In conclusion, the present study based on infertile patients from the Barcelona area found a decline in sperm motility and morphology, without effects on sperm count. Simple Summary: Both in human medicine and in the veterinary field, lipidomic techniques have been applied extensively in order to evaluate health status. In equine species where the subject of focus is the fertility and sperm quality of the equids, lipidomic assessment can be a useful tool to better understand the metabolic transformation of the fatty acids of the spermatozoa as well as the erythrocyte membrane and their role on spermiogenesis. The purpose of this study is to evaluate the composition of the spermatozoa's membranes of 26 healthy male Martina Franca donkeys and the possible correlation with the sperm parameters and the erythrocytes' membrane. The results demonstrate that the membrane of erythrocytes and spermatozoa are different, which provides a deeper understanding of the distribution and the role of these fatty acids in sperm parameters that are highly associated with fertility. Fatty acid-based lipidomic analysis has been widely used to evaluate health status in human medicine as well as in the veterinary field. In equine species, there has been a developing interest in fertility and sperm quality. Fatty acids, being the principal components of the membranes, play an active role in the regulation of the metabolic activities, and their role on spermiogenesis seems to be of great importance for the resulting quality of the sperm and, thus, fertility. With the application of widely used lipidomic techniques, the aim of this study was to evaluate: (a) the fatty acid content of the spermatozoa's membranes of 26 healthy male Martina Franca donkeys and its possible correlation with sperm parameters, and (b) the evaluation of the composition of the red blood cells' membrane. PUFA omega-6 are the principal components (40.38%) of the total PUFA content (47.79%) in both types of cells; however, DPA is the predominant one on the spermatozoa's membrane (27.57%) but is not present in the erythrocyte's membrane. Spermatozoa's motility (%) is positively correlated with stearic acid and EPA, and progressive motility (%), with oleic acid. These findings offer information on the composition of both types of cells' membranes in healthy male MF donkeys and reflect the metabolic transformations of the spermatozoa's membrane during the maturation period, providing a better perception of the role of fatty acids in sperm parameters and fertility. Previous studies have indicated that outdoor air pollution has a negative impact on semen quality; however, few studies have examined whether living in a recently renovated residence is one of the factors influencing semen parameters. We aimed to examine the association between household renovation and semen parameters among infertile men. Our study was conducted at the Reproductive Medicine Center, The First Hospital of Jilin University (Changchun, China) from July 2018 to April 2020. A total of 2267 participants were enrolled in the study. The participants completed the questionnaire and provided a semen sample. Univariate and multiple logistic regression models were used to estimate the association between household renovations and semen parameters. Of the participants, about one-fifth (n = 523, 23.1%) had undergone renovations in the last 24 months. The median progressive motility was 34.50%. There was a significant difference between participants whose residences had been renovated in the last 24 months and those whose residences had not been recently renovated (z = –2.114, p =.035). Compared with participants whose residences were not recently renovated, participants who moved into the residence within 3 months after renovation had a higher risk of abnormal progressive motility after adjusting for age and abstinence time (odds ratio [OR] = 1.537, 95% confidence interval [CI]: 1.088–2.172). Testis‐specifically expressed genes are important for male reproduction according to their unique expression patterns. However, the functions of most of these genes in reproduction are unclear. Here, we showed that mouse 4930590J08Rik was a testis‐specifically expressed gene. 4930590J08Rik knockout mice exhibited a delay in the first wave of spermatogenesis and a reduction of cauda epididymal sperm. Furthermore, knockout spermatozoa exhibited defective acrosome reactions and decreased progressive motility, which led to impaired in vivo fertilization. Transcriptome analysis of testes revealed that most of the differentially expressed genes in knockout testes were associated with metabolic processes. 4930590J08Rik knockout sperm exhibited oxidative phosphorylation deficiency and were highly dependent on increased anaerobic glycolysis to compensate for ATP demands. Taken together, the 4930590J08Rik‐disrupted mouse partially mimics the phenotypes of human asthenospermia and oligozoospermia, which provides a new model for further understanding the pathogenesis of idiopathic male infertility. The presence of bacterial contamination in bovine insemination doses may lead to unexpected economic loss. Currently used antimicrobial supplements are often ineffective due to antibiotic resistance of the common bacterial contaminants. Staphylococcus aureus belongs to the frequent inhabitant of bovine semen with a deleterious effect on spermatozoa quality. Bioactive substances including naringenin, curcumin and isoquercitrin were previously studied with various beneficial effects on spermatozoa. Therefore, the study aimed to simulate bacteriospermia in vitro using S. aureus, which was previously isolated from the bovine ejaculate, and to test if the bioactive substances mentioned above would bring some protective properties against the bacterial damage. Density gradient centrifugation was used to avoid contamination by naturally present bacteria. S. aureus was inoculated in BGM-3 medium, and co-incubated with pre-washed spermatozoa and bioactive substances. At times of 0, 2, and 4 h, progressive motility, the membrane potential of mitochondria, reactive oxygen species (ROS) formation, and sperm DNA damage were evaluated. The results showed that S. aureus significantly increased ROS production, sperm cells with damaged mitochondrial membrane potential, and sperm DNA integrity which led to a decrease in the progressive movement of spermatozoa. On the other hand, all three bioactive substances significantly scavenged elevated ROS. Especially, the groups treated with naringenin and isoquercitrin showed preserved mitochondrial membrane, DNA integrity inside the sperm head, and improved sperm progressive movement under bacteriospermia in vitro. Several gene expressions are related to the success of spermatogenesis. Protamine plays an important role in sperm DNA packaging, spermatogenesis, and sperm quality. This study aimed to isolate RNA from spermatozoa and determine the gene expression profiles of protamine 1 (PRM1) and protamine 2 (PRM2). Six Pasundan bulls aged 5-8 years with a body weight of 380 kg-430 kg were used for this study. The Pasundan bulls were classified into group A (70%-79%) and group B (80%-89%) based on their sperm motility in fresh semen. In this study, correlation analysis was performed between fresh semen characteristics (volume, motility, concentration, abnormality, viability, intact plasma membrane (IPM), DNA integrity) and frozen semen characteristics after thawing (motility, viability, IPM, and DNA integrity). Total RNA was isolated from the frozen sperm pellet, and cDNA was synthesized. Specific PCR primers were used for the transcription of PRM1 and PRM2 from sperm. PRM1 and PRM2 gene expressions were evaluated by qRT-PCR, and ACTB was used as a control. The results showed that progressive sperm motility in the fresh sperm positively correlated with sperm viability and IPM. PRM1 and PRM2 were higher (p<0.05) in group B than in group A. This condition indicates the existence and influence of protamine on the parameters of progressive sperm motility. Objective: To evaluate the impact of seasonal and geographical temperature on sperm parameters in Indian men who were partners in subfertile couples. Methods: A retrospective review of the records of semen analysis from January 2009 to December 2017 that was performed as part of primary evaluation of infertile couple was undertaken. Mean values of various semen parameters were compared between seasons using one-way analysis of variance test followed by Bonferroni pair wise post-hoc comparison test. Bivariate Pearson correlation coefficient was applied to measure the correlation between age, sperm parameters and annual temperature/seasons. The logistic regression analysis was further carried out to identify significant variables contributing to normal progressive motility of sperm. AIM: The aim of this study, was to check the effect of variation in media incubation time on progressive motility of spermatozoa of men attending Acharya Vinoba Bhave Rural Hospital, Wardha Testube Baby Center. METHOD: A total 55 semen samples of patients were used in this study. Semen Analysis was performed according to WHO guidelines 2010 and observations for Sperm Count and motility was recorded as Pre-wash data. Sperm Processing was performed using density gradient method in which samples randomly divided into 3 group with media of variant incubation time at 37oC-36oC (i.e. 18 samples in ±15 min; 19 samples in ±30 min; 18 samples in ±60 min), and observations for Sperm Count and motility was recorded as Post-wash data. Retrieval rate of progressive motility of sperm in all 3 groups were evaluated and compared via Pre and Post wash data. RESULT: A timeline of individual sample in all three groups was recorded to trace the lower limit, upper limit and average retrieval rate of the group. For group with media incubation time ±15Min, upper limit value was 24%; lower limit value was 0.56% and average of progressively motile sperm retrieval rate was 8.26%. For group with media incubation time ±30Min, upper limit value was 23.3%; lower limit value was 2.0% and average of progressively motile sperm retrieval rate was 8.22%. For group with media incubation time ±60Min, upper limit value was 17.6%; lower limit value was 2.1% and average of progressively motile sperm retrieval rate was 8.14%. Variation in incubation time of sperm preparation media affects the retrieval rate of progressive motility sperms from the native semen sample which subsequently may influence the assisted reproductive technology (ART) outcomes. Male fertility, as manifest by the quantity and progressive motility of spermatozoa, is negatively impacted by obesity, dyslipidaemia and metabolic disease. However, the relative distribution of lipids in spermatozoa and the two compartments which supply lipids for spermatogenesis (seminal fluid and blood serum) has not been studied. We hypothesised that altered availability of lipids in blood serum and seminal fluid may affect the lipid composition and progressive motility of sperm. 60 men of age 35 years (median (range 20–45) and BMI 30.4 kg/m2 (24–36.5) under preliminary investigation for subfertility were recruited at an NHS clinic. Men provided samples of serum and semen, subject to strict acceptance criteria, for analysis of spermatozoa count and motility. Blood serum (n = 60), spermatozoa (n = 26) and seminal fluid (n = 60) were frozen for batch lipidomics analysis. Spermatozoa and seminal fluid had comparable lipid composition but showed marked differences with the serum lipidome. Spermatozoa demonstrated high abundance of ceramides, very-long-chain fatty acids (C20-22), and certain phospholipids (sphingomyelins, plasmalogens, phosphatidylethanolamines) with low abundance of phosphatidylcholines, cholesterol and triglycerides. Men with spermatozoa of low progressive motility had evidence of fewer concentration gradients for many lipid species between blood serum and spermatozoa compartments. Spermatozoa are abundant in multiple lipid species which are likely to contribute to key cellular functions. Summary: Sperm motility and viability of cryopreserved semen vary between boars and straws, which influences the outcomes of in vitro embryo production (IVEP). However, progressive motility is usually not considered during IVEP. The aim of this study was to assess fertilization with a 500:1 and 250:1 'progressively motile sperm to oocyte' ratio on IVEP outcomes using semen from three Duroc and three Landrace boars. Frozen–thawed sperm was centrifuged through a 45/90% Percoll® density gradient and sperm quality parameters were assessed. In vitro matured oocytes were fertilized at the two ratios, a portion was stained 10–12 h after start of fertilization to analyze fertilization and polyspermy, while the remaining zygotes were cultured up to day 7. The 500:1 ratio resulted in a higher fertilization and blastocyst yield on day 6 compared with the 250:1 ratio, but no effect of ratio was observed for polyspermy, cleavage rate or blastocyst cell number. Individual differences between boars were observed for fertilization, cleavage and blastocyst rates, but not for the other IVEP outcomes. In conclusion, a higher fertilization and blastocyst yield was obtained with the 500:1 ratio compared with the 250:1 ratio, while polyspermy level was consistent across ratios. Differences in IVEP outcomes were still observed between the individual boars although adjusted for progressive motility. Background and objective: Protein phosphatase 3 catalytic subunit gamma (PPP3CC) and protein phosphatase 3 regulatory subunit B, beta (PPP3R2) are respectively the catalytic and regulatory subunits of calcineurin in sperm. Deficiency in either protein causes impaired sperm motility leading to male infertility. Many cases of sterility are attributed to asthenozoospermia (AZS); however, it remains unknown whether PPP3CC and PPP3R2 are related to AZS. Material and methods: Quantitative PCR and Western blotting were used to investigate the expression levels of PPP3CC and PPP3R2 in the spermatozoa of patients with AZS and to explore the clinical significance. Results: Two calcineurin inhibitors cyclosporine A (CsA) and tacrolimus (FK506) markedly impaired the total motility and progressive motility of human sperm, indicating that PPP3CC or PPP3R2 might be involved in AZS. PPP3CC mRNA and protein expression was lower in the ejaculated spermatozoa of patients with AZS than in normal sperm (NS). Correlation analysis showed that PPP3CC protein expression correlated positively with progressive motility (r = 0.2592, P < 0.05); however, there were no significant differences in PPP3R2 mRNA and protein levels between AZS and NS. Conclusion: These findings suggest that the abnormal expression of PPP3CC rather than PPP3R2 might be a pathological factor or indicator in AZS. Thus, PPP3CC may be a potential therapeutic or diagnostic target for some cases of male infertility.

The present investigation was carried out to study the semen characteristics of Frieswal bulls. In the present investigation a total of 181 ejaculates seminal collected from six bulls were studied and out of these 82.30 per cent were creamy in colour. The density of Frieswal bulls’ semen was found to be thick (93.92%). The overall mean ejaculate volume (ml), individual motility (%), sperm concentration (millions/ml), live sperms (%) and Hypo-osmotic swelling (HOS) positive spermatozoa in neat semen of Frieswal bulls were 5.60±0.16, 60.39±0.04, 1076.1±32.56, 78.93±0.004 and 73.76±0.47, respectively. The differences in mean values of ejaculated semen volume, sperm concentration, live sperm count and progressive motility of semen were highly significant (P<0.05) among the bulls. It was concluded that the, Frieswal bull produced better quality semen in comparison to another breed. Background: Fertil Aid for Men is a non-prescription dietary supplement, it's a mixture of multiple antioxidants, vitamins, a blend of herbal ingredients, and amino acid, as L-Carnitine. This research aims to appraise the effectiveness of a mixture of vitamins and antioxidants on seminal fluid parameters in males with idiopathic infertility. Method: Thirty-three men with idiopathic infertility were involved inthis prospective clinical study. The baseline semen specimens were taken from the subjects following three to seven days' abstinence. Semen parameters including (volume, pH, concentration, and motility) were measured. Abnormal sperm sample subjects were given a FertilAid capsule three times daily for 90 days then final semen analysis was estimated. The outcomes of this study demonstrated consumption of fertilAid has significantly improved sperm count/ml (p < 0.05), also significant amelioration in the sperm motility including progressive and non-progressive motility (p < 0.05). The rate of total sperm motility was significantly improved (p < 0.05). The concentration of immotile sperm has considerablyreduced (p < 0.05). Conclusion: The administration of a combination of L-Carnitine, Zinc, Maca Root, Asian Ginseng, Vitamin C, E, Beta Carotene, and Selenium PLUS CoQ10 in idiopathic male infertility has notably improved sperm parameters. High levels of polyunsaturated fatty acids in avian sperm cause more susceptibility to lipid peroxidation. Aging in roosters reduces the antioxidant capacity of sperm and thus fertility. The purpose of this study was to investigate the effects of different levels of alpha-lipoic acid (ALA) as a feed supplement to improve the semen quality and fertility parameters of aged broiler breeder roosters and identification of its most effective level. A total of forty-two roosters at 45 wk of age were randomly assigned to 7 treatments (0, 15, 40, 70, 95, 120, and 145 mg ALA/bird per day) for 8 wk. Semen parameters and body weight were assessed biweekly, and testosterone plasma levels were determined in the 8th wk of the experimental period. Artificial insemination was performed at the end of the experiment to evaluate the fertility potential. The dietary administration of ALA had no significant effects on body weight, semen volume, average path velocity, linearity, straightness, wobble, the amplitude of lateral head displacement, beat-cross frequency, sperm concentration, morphology, plasma testosterone level, fertility, or hatchability (P > 0.05). Aims: The aim of the current study was to develop a sperm motility index (SMI) to test the sperm toxicity of ultrasound gels and vaginal lubricants used in reproductive medicine. Settings and Design: Two ultrasound gels (Aquasonic® and Kefus®) and five vaginal lubricants (Vaginesil™, Velastisa®, K-Y Jelly®, Control®, and Durex®) were studied. Abstract Background and aims It has been reported that Helicobacter pylori (HP) infection was more prevalent in infertile populations. HP infection could lead to decreased sperm parameters, and treating the HP infection could improve the quality of sperm. However, studies investigating the relationship between infertility and HP infection are still limited, and more evidence is required. Therefore, we performed the present study to investigate the impact of HP infection on sperm quality in males and on ovarian reserve in females. Methods A total of 16,522 patients who visited the Second Hospital of Zhejiang University from January 2016 to June 2019 due to abdominal discomfort and underwent a 13/14C-urea breath HP test were included in this retrospective cross-sectional study. Among them, 565 had performed sperm analysis or ovarian reserve tests in the past three months and were involved for further analyses. Sperm parameters were examined with a computer-assisted sperm analysis system, and serum anti-Müllerian hormone (AMH) and sex hormones were tested with an electrochemiluminescence method. Results Among 363 patients who underwent the sperm test, 136 (37.47%) had HP infection. Among 202 patients who underwent the AMH test, 55 (27.23%) had HP infection. There was no difference in sperm concentration and motility between the HP+ and HP− groups (P > 0.05). Further subgroup analyses stratified into 5-year age groups confirmed that there was no significant difference in sperm parameters (P > 0.05). When pooled with previously published data, no significant difference in sperm concentration or motility was found (P > 0.05). Meanwhile, this study found that the serum AMH level was similar between the HP+ and HP− groups (P > 0.05). Further subgroup analyses confirmed that there was no significant difference in serum AMH level (P > 0.05). Conclusions There were no differences in sperm parameters and AMH levels based on history of HP infection among Chinese patients.

Abstract Background Human papillomavirus (HPV) has been considered as one of the most common sexually transmitted viruses that may be linked to unexplained infertility in men. The possible mechanisms underlying correlation between HPV infection and infertility could be related to the altered sperm parameters. Current studies have investigated the effect of HPV seminal infection on sperm quality in infertile men, but have shown inconsistent results. Methods We systematically searched PubMed, Embase, Web of Science and CNKI for studies that examined the association between HPV seminal infection and sperm progressive motility. Data were pooled using a random-effects model. Outcomes were the sperm progressive motility rate. Results are expressed as standardised mean difference (SMD) with 95% confidence interval (CI). Heterogeneity was evaluated by the I-square (I 2 ) statistic. Results Ten studies were identified, including 616 infertile patients with HPV seminal infection and 2029 infertile controls without HPV seminal infection. Our meta-analysis results indicated that sperm progressive motility was significantly reduced in HPV-infected semen samples compared with non-infected groups [SMD:-0.88, 95% CI:-1.17 ~ − 0.59]. There existed statistical heterogeneity (I 2 value: 86%) and the subgroup analysis suggested that study region might be the causes of heterogeneity. Conclusions HPV semen infection could significantly reduce sperm progressive motility in infertile individuals. There were some limitations in the study such as the differences in age, sample sizes and the number of HPV genotypes detected. Further evidences are needed to better elucidate the relationship between HPV seminal infection and sperm quality.

Objective: To summarize the currently available phase I and II clinical trials of the effects of nonoxynol-9 (N-9) on human sperm structure and functions. Methods: A systematic review and meta-analysis aiming to evaluate the spermicidal activity of N-9 on motility, was conducted in PubMed, EMBASE, and Cochrane databases by 10 March 2021. The counted numbers of progressive motile (PR) sperm in cervical mucus and the vanguard sperm penetration distances were analyzed. Other effects on sperm structures and physiological activities were reviewed as well. Results: In the pooled results, percentages or counted numbers of PR sperm decreased after the treatment of N-9. Vanguard sperm penetration distance was shortened in treated groups. N-9 has been confirmed to damage the structures of sperm, as well as other organelles like acrosome and mitochondria. The physiological activities such as generation of reactive oxygen species, superoxide dismutase activity, acrosin activity, and hemizona binding were all inhibited in the reviewed studies. Conclusions: N-9 has several impacts on sperm owing to its potency in reducing sperm motility and cervical mucus penetration, as well as other functional competencies.

Male germ cells have the potential to generate reactive oxygen species (ROS) during different stages of differentiation, laboratory manipulation and during their interaction with dissolved oxygen in extender. These ROS and its subset reactive nitrogen species (RNS) perform a concentration specific bimodal role, favouring oxidative eustress at physiological concentrations. The present study was aimed to investigate the dose and time dependent effect of the most potent RNS, i.e. nitric oxide (NO) compounds, specifically SNP (Sodium nitroprusside) and L-NAME (N-Nitro-L-Arginine Methyl Ester) supplementation in extender on progressive motility. The semen samples were divided into 8 aliquots comprising of various concentrations of SNP (Control, 0.1, 1 and 10 μmol/L) and L-NAME (Control, 1, 10 and 100 μmol/L) and refrigerated for 0, 6, 24 and 48 h. The progressive motility (%) improved significantly particularly with the addition of 1 μM SNP and 10 μM L-NAME as compared to control during all time intervals. Subsequently semen sample was cryopreserved with optimum level of SNP (1μmol/L) and L-NAME (10 μmol/L) from these results by adding it in extender and assessed for progressive motility before cryopreservation and after 24 h, 1 month and 2 months of cryopreservation. It is concluded that the addition of SNP and L NAME in cryopreservation media at 1 μM and 10 μM respectively improved the sperm motility significantly. On further evaluation a significant increase in progressive motility was observed for L-NAME treated extender at 24 h and SNP treated extender at 1 and 2 month interval in comparison to control.

Assisted reproductive technologies have been used in zoos and breeding centres to preserve rare and endangered species of wild animals over the past three decades. The most common way to preserve the genetic material of farm and wildlife animals is to create cryobanks of semen producers and embryos. Studies of domestic species under controlled conditions provide an excellent opportunity to develop effective semen handling techniques for application to wild species of the genus Equus. The All-Russian Research Institute for Horse Breeding (ARRIH) began studies on the cryopreservation of genetic resources in the early 1950s. In 1954 the ARRIH got the world's first foal from the artificial insemination by cryopreserved sperm. The collection of genetic resources of ARRIH has been successfully working since 1972 and contains nowadays cryopreserved sperm from 56 stallions of 16 breeds of domestic and foreign selections. Freezing and subsequent thawing of stallion sperm leads to a decrease in progressive sperm motility and an increase in the number of sperm with structural pathologies. Electron microscopy is one of the most accurate methods used to assess the structural integrity of sperm. We conducted an electron microscopic study of native and cryopreserved sperm of 35 stallions of riding and trotting breeds, aged 4 to 22 years (mean age 11.3 ± 0.9 years). We found that organoids with a denser structure are more resistant to freezing and subsequent thawing of sperm. The use of stallion sperm cryopreservation according to the standard technology of ARRIH leads to a decrease in the number of sperms with intact heads by an average of 19.7%, and the share of sperm with intact heads in cryopreserved sperm is 49.1 ± 2.7% (p < 0.001). The most negatively cryopreservation of semen affects an acrosome of sperms. The main pathologies of acrosomes in sperm cryopreservation are acrosome hypoplasia and its degradation (secondary absence of acrosome). The number of spermatozoa with acrosome hypoplasia and the absence of internal content after cryopreservation increases by 20.9%, and the share of such sperms is 31.7% in thawed sperm (p < 0.001). The number of sperm with acrosome degradation during sperm freezing and thawing increases by 10.4%, and is 18.6 ± 2.4% in average. The share of spermatozoa with normal mitochondrial structure in native sperm is 89.3 ± 2.0% in average, in cryopreserved sperm – decreases by 6.5%. The share of spermatozoa with normal axoneme after cryopreservation decreases slowly, averaging by 4.4% and is 81.4 ± 2.0%. Some of the available assisted reproduction technologies will need to be further optimised in domestic horses before they can be applied to the endangered Przewalski's horse Equus ferus przewalskii.

The present study was conducted with an objective of establishing a relationship between semen evaluation parameters of freezable and non-freezable Murrah buffalo semen. Work was conducted at Frozen Semen Bank, RCDF Limited, Bassi, Jaipur, Rajasthan, on 12 apparently healthy pure bred Murrah buffalo breeding bulls. A total of 96 ejaculates (48 each freezable and non-freezable) were analysed at five different stages of processing namely neat semen evaluation, post dilution, post equilibration, post thaw and after 1 h of incubation post-thaw at 37°C for progressive motility, live dead count, reaction to hypo-osmotic solution and acrosomal integrity. Evaluation of the semen was done in two groups, i.e. freezable and non-freezable semen. There was a significant difference in terms of all the semen evaluation parameters between freezable and non-freezable semen at different stages of evaluation. Also, results of the study revealed a highly significant correlation between the various semen evaluation parameters for both the groups.

Control® and Velastisa® presented toxicity at 10% after 2 h, while K‑Y Jelly® showed toxicity at 10% from 1 h of incubation. Regarding ultrasound gels, Aquasonic® showed toxic effects after only 0.5 h (SMI = 0.70 ± 0.15), while Kefus® showed slightly toxic effects after 2 h (SMI 0.69 ± 0.07). Conclusions: SMI is an accurate tool to evaluate sperm toxicity. One of the main strengths of the article is the inclusion of representative semen samples and known products used worldwide. As a source of growth factors, platelet-rich plasma (PRP) has been widely used in the repair of various injuries due to its cytoprotective properties in regenerative medicine. The objective of the present study was to investigate the effect of autologous PRP supplementation on the quality of frozen-thawed human sperm. Twelve normozoospermic semen samples were collected, and each sample was divided into 4 aliquots and added with different proportions of PRP (0%, 2%, 5%, and 10%) separately, followed by cryopreservation. Sperm motility, viability, membrane integrity, DNA fragmentation index (DFI), reactive oxygen species (ROS) levels and mitochondrial membrane potential were measured and analyzed. The results showed that the addition of 5% PRP improved sperm progressive motility (30.3 ± 2.7 VS. 28.1 ± 2.6), viability (65.5 ± 4.2 VS. 59.6 ± 3.9), and membrane integrity (52.4 ± 3.6 VS. 49.2 ± 3.4) after cryopreservation (P < 0.05). Slight attenuation of ROS generation, restoration of mitochondrial membrane potential and reduction of DNA fragments were also observed in post-thawed sperm supplemented with PRP, but there was no significant (P > 0.05). In conclusion, autologous PRP has a partial protective effect on cryopreservation of human spermatozoa, and the combined application with other high-efficiency cryoprotectant is worthy of further study. Study Question: Is there an association between the human testis-specific gene, testis developmental related gene 1 (TDRG1) and human sperm motility?Summary Answer: TDRG1 is associated with asthenozoospermia and involved in regulating human sperm motility.What Is Known Already: Many testis-specific proteins potentially regulate spermatogenesis and sperm motility. We have identified a novel human testis-specific gene, TDRG1, which encodes a 100-amino-acid protein localized in the human sperm tail, yet little is known about its role in human spermatozoa.Study Design, Size, Duration: Sperm samples were obtained from normozoospermic men and asthenozoospermic men who visited the reproductive medical center at Jiangxi Maternal and Child Health Hospital, Nanchang, Jiangxi, China between February 2018 and January 2019. In total, 27 normozoospermic men and 25 asthenozoospermic men were recruited to participate in the study.Participants/materials, Setting, Methods: The level of TDRG1 in sperm of normozoospermic and asthenozoospermic men was examined by immunoblotting and immunofluorescence assays. Progressive motility was examined by computer-aided sperm analysis. The correlation between the TDRG1 protein level and progressive motility was analyzed by linear regression. TDRG1 was imported into the sperm of normozoospermic and asthenozoospermic men using a cell-penetrating peptide (CPP)-fused TDRG1 recombinant protein (CPP-TDRG1), and the progressive motility was examined. Also, the altered proteins associated with TDRG1 in asthenozoospermic sperm were detected using label-free quantification method-based quantitative proteomic technology. TDRG1-interacting proteins were identified by co-immunoprecipitation coupled with tandem mass spectrometry analysis.Main Results and the Role Of Chance: The mean level of TDRG1 was significantly decreased in sperm of asthenozoospermic men compared with normozoospermic men (P < 0.05) and was positively correlated with percentage of progressively motile sperm (r2 = 0.75, P = 0.0001). The introduction of TDRG1 into human sperm, using CPP, significantly increased progressive motility (P < 0.05) and improved the progressive motility of sperm from asthenozoospermic men to the normal level. Antioxidant and reactive oxygen species (ROS) has been implicated in male infertility. Antioxidants are compounds which scavenge formation or oppose of the actions ROS. Spermatozoa are particularly vulnerable to ROS because of inherent deficiencies in intracellular antioxidant enzyme protection, thus total body antioxidant capacity become more substantial to protect sperm. In this study to try to evaluate the role of serum total antioxidant capacity, uric acid, glutathione, total thiols, ferritin, albumin and malodialdehyde in male infertility. 66 men included in this study 40 were infertile and 26 were healthy fertile. Results showed a significant decrease in sperm progressive motility and a significant increase in sperm tail abnormality, also there were a significant decrease in serum total antioxidant capacity, uric acid and albumin in infertile group. Because of lack of the intracellular antioxidant enzymes sperm cannot protect plasma membrane that surrounds the tail, the protection depends on the antioxidant defense afforded by the seminal plasma and on the entire body antioxidant capacity. Decrease of serum antioxidant capacity, uric acid and albumin may cause destroy of sperm tail membrane and this may lowering of sperm progressive motility.

Boar fertility is an important factor in farm production; it is therefore of interest to determine factors which reduce the fertilising capacity of semen samples stored at 17°C for use in intrauterine insemination. This work evaluated the effect of the number of rest days between each mounting of the boar, and the number of days that the semen was stored at 17°C, on sperm motility and semen concentration. We also analysed whether the boar's age influenced the sperm concentration. The results showed that only the total motility diminished as the storage time at 17°C increased (p <.05). A low negative correlation was observed between the variables' rest days and total and progressive motility. The sperm concentration presented no relation with rest days or the boar's age. The boars' rest days had no effect on motility and sperm concentration in the males studied, allowing them to be used with the frequencies described with no effect on these parameters. Chromium is an essential element that is required for the normal physiology but can be toxic to humans above a certain level. In spite of growing interest in research on chromium exposure to human health consensus about its effect on human, semen quality has not been achieved. The aim of the present study is to evaluate the impact of chromium exposure on semen parameters. A total of 760 males attending andrology laboratory of Tongji Hospital, Wuhan, for routine semen analysis were enrolled and requested to provide semen and urine samples. The urine level of chromium was evaluated using inductively coupled plasma mass spectrometry (ICP-MS), and computer-assisted semen analysis (CASA) was applied to examine semen parameters. Associations between semen parameters and urinary chromium were analyzed by means of multivariate linear regression analysis. Multivariate analysis showed a negative association between the urinary concentrations of chromium and progressive motility (β = − 0.014, p = 0.040) and total motility (β = − 1.077, p = 0.048), while other semen parameters did not show any statistically significant changes. Bacterial contamination of ejaculates has captured the interest of the wide andrological community. Interactions between spermatozoa and bacteria are partially known. The aim of our study was to evaluate qualitative bacterial composition of fresh bovine semen and to reveal associations amongst the quantity of bacteria (Colony Forming Unit - CFU/mL) present in bovine ejaculates with progressive motility of spermatozoa and biochemical parameters of seminal plasma. Fresh semen (n=20) was collected from Holstein-Friesian bulls. Progressive motility (PROG=25µm/s) was analysed using Computer-assisted sperm analysis (CASA). Seminal plasma was separated and subjected to assessment (Randox RX Monza analyzer) of following biochemical parameters: Magnesium (Mg), Calcium (Ca), Phosphor (P), Urea, Aspartate transaminase (AST), Alanine transaminase (ALT), Alkaline phosphatase (ALP), Cholesterol, Bilirubin, Triglycerides, Albumin and Uric acid. The identification of bacteria showed the prevalence of the genus Staphylococcus. The Pearson correlation analysis showed a negative significant (P<0.001) correlation between CFU and PROG. Further, positive significant correlations were observed amongst PROG and biochemical parameters (P<0.01 in case of Mg, Urea, ALP, ALB; P<0.05 in case of Ca, ALT, Bilirubin and Uric acid). The results show negative significant correlations amongst CFU and biochemical parameters (P<0.001 in case of Mg; P<0.01 in case of ALT, ALP and P<0.05 in case of Uric acid). In conclusion, our results reveal that bacteria may affect not only on the sperm motility, but also on the composition of the seminal plasma. An association between progressive motility (PM) and spermatozoa fertility competence has been suggested. However, the mechanism that underlies PM is not clear enough. We examined physiological characteristics and fatty acid composition of fresh spermatozoa with high and low PM. Additional analysis of fatty acid composition and structural characteristics was performed on spermatozoa samples with high and low progressively motile spermatozoa’s survival (PMSS), i.e., the ratio between the proportion of progressively motile spermatozoa after and before cryopreservation. Finally, a fertility field trial was conducted to examine the association between the number of PM spermatozoa within the insemination straw post thawing and conception rate. Analysis of fresh spermatozoa revealed a higher omega-6 to omega-3 ratio in ejaculates with low PM relative to those with high PM (p < 0.01). The proportion of polyunsaturated fatty acids was higher in low-PMSS fresh samples (p < 0.05) relative to their high-PMSS counterparts. Fresh samples with high-PMSS expressed a higher mitochondrial membrane potential (p < 0.05) and a higher proportion of viable cells that expressed reactive oxygen species (ROS; p < 0.05). Post-thawing evaluation revealed a reduced proportion of progressively motile sperm, with a prominent effect in samples with high PM relative to low PM, defined before freezing (p < 0.01). No differences in spermatozoa mitochondrial membrane potential or ROS level were found post-thawing. A fertility study revealed a positive correlation between the number of progressively motile spermatozoa within a standard insemination straw and conception rate (p < 0.05). Considering these, the bull PMSS is suggested to be taken into account at the time of straw preparation.

Over the past five decades, there has been increasing evidence to indicate global declines in human semen quality. Parallel adverse trends measured in male animals indicate a potential environmental aetiology. This study evaluated the progressive motility (PM) of stallion ejaculate through a systematic review and meta-analysis. A total of 696 estimates of equine PM from 280 studies, which collected semen samples between the years 1990 and 2018, were collated for meta-analysis. The method of motility analysis, breed, season of collection, and geographical location were extracted. Simple linear regression determined temporal trends in stallion PM. Studies using microscopy estimated PM to be significantly greater compared to computer-automated methods (p ≤ 0.001). For Arabian breeds, PM was consistently higher than other breeds. Over time, there was a significant decline in PM for studies from Europe (n = 267) but a significant increase for studies from North America (n = 259). Temporal trends indicate the fertilising capacity of equine ejaculate has remained consistently high in the last three decades. That being so, variations observed suggest methodological, geographical, and individual stallion differences may significantly influence actual and reported stallion fertility potential.

Study Question: Is there a role for lysine glutarylation (Kglu), a newly identified protein post-translational modification (PTM), in human sperm?Summary Answer: Kglu occurs in several proteins located in the tail of human sperm, and it was reduced in asthenozoospermic (A) men and positively correlated with progressive motility of human sperm, indicating its important role in maintaining sperm motility.What Is Known Already: Since mature sperm are almost transcriptionally silent, PTM is regarded as an important pathway in regulating sperm function. However, only phosphorylation has been extensively studied in mature sperm to date. Protein lysine modification (PLM), a hot spot of PTMs, was rarely studied except for a few reports on lysine methylation and acetylation. As a newly identified PLM, Kglu has not been well characterized, especially in mature sperm.Study Design, Size, Duration: Sperm samples were obtained from normozoospermic (N) men and A men who visited the reproductive medical center between February 2016 and January 2018. In total, 61 N men and 59 A men were recruited to participate in the study.Participants/materials, Setting, Methods: Kglu was examined by immunoblotting and immunofluorescence assays using a previously qualified pan-anti-glutaryllysine antibody that recognizes glutaryllysine in a wide range of sequence contexts (both in histones and non-histone substrates) but not the structurally similar malonyllysine and succinyllysine. The immunofluorescence assay was imaged using laser scanning confocal microscopy and super-resolution structured illumination microscopy. Sperm motility parameters were examined by computer-assisted sperm analysis.Main Results and the Role Of Chance: Kglu occurs in several proteins (20-150 kDa) located in the tail of human sperm, especially in the middle piece and the latter part of the principal piece. Sperm Kglu was modulated by regulatory systems (enzymes and glutaryl-CoA) similar to those in HeLa cells. The mean level of sperm Kglu was significantly reduced in A men compared with N men (P < 0.001) and was positively correlated with progressive motility (P < 0.001). The sodium glutarate-induced elevation of Kglu levels in A men with lower Kglu levels in sperm significantly improved the progressive motility (P < 0.001). A-kinase anchor protein 4 (AKAP4) is playing a central role in flagellar structure, chemotaxis, capacitation and sperm motility. In mammals, AKAP4 is expressed during spermatogenesis. AKAP4 is synthesized as a precursor, proAKAP4, which is cleaved into mature AKAP4 during fibrous sheath assembly. The proAKAP4 is a good indicator of sperm quality in humans and boars. The aims of this work were to study the expression, the localization and the concentration of proAKAP4 and AKAP4 in equine semen, and to evaluate the possible correlation between the total and progressive motility and the concentration of proAKAP4 measured by ELISA in post-thawed semen. Frozen sperm from 13 different stallions were used. Semen samples (n = 17) were prepared using the INRA Freeze medium to reach a concentration of 150 million spermatozoa/mL, packaged and frozen in 0.5 mL straws. The precursor proAKAP4 and the mature protein AKAP4 both localize to the fibrous sheath of the principle piece of equine sperm flagellum. The concentrations of proAKAP4 were determined in the post-thawed semen using ELISA method (Horse 4MID® kits, 4BioDx, France). The mean concentration of proAKAP4 was then of 7.372 ± 0.79 ng/μL and was significantly correlated with the post-thawed total motility (Pearson coefficient r = 0.66, p = 0.002) and progressive motility (Pearson coefficient r = 0.76, p = 0.0002) and the amount of proAKAP4 represent the amount of spermatozoa that expressed proAKAP4. Taken together, these preliminary results confirm the interest to use proAKAP4 concentrations as a promising marker of stallion sperm quality as close correlation was observed between the proAKAP4 concentration and sperm motility parameters. • The precursor of AKAP4, proAKAP4, was never studied in post-thaw stallion semen. • ProAKAP4 and AKAP4 are co-localized to the principle piece of equine sperm flagellum. • ProAKAP4 content is correlated with the post-thaw semen motility. • ProAKAP4 is a promising marker of stallion sperm quality. Metformin is a leading antidiabetic drug that is used worldwide in the treatment of diabetes mellitus. This biguanide exerts metabolic and pleiotropic effects in somatic cells, although its in vitro actions on human spermatozoa remain unknown. The present study investigated the effects of metformin on human sperm function. Human spermatozoa were incubated in the presence or absence of 10 mM metformin for 8 or 20 h, and motility was measured by computer-aided sperm analysis (CASA); other parameters were evaluated by flow cytometry. Metformin significantly reduced the percentage of motile, progressive and rapid spermatozoa and significantly decreased sperm velocity. Metformin did not affect viability, mitochondrial membrane potential (MMP) or mitochondrial superoxide anion generation of human spermatozoa at any time studied. However, metformin clearly inhibited the protein kinase (PK) A pathway and protein tyrosine phosphorylation at 8 and 20 h, key regulatory pathways for correct sperm function. In summary, metformin treatment of human spermatozoa had a detrimental effect on motility and inhibited essential sperm signalling pathways, namely PKA and protein tyrosine phosphorylation, without affecting physiological parameters (viability, MMP, mitochondrial superoxide anion generation). Given the growing clinical use of metformin in different pathologies in addition to diabetes, this study highlights an adverse effect of metformin on spermatozoa and its relevance in terms of human fertility in patients who potentially could be treated with metformin in the future. In addition to diabetes, the clinical use of metformin in different pathologies is growing, and late-phase clinical trials of metformin in cancer therapy are underway. This study investigates the effects of metformin on human spermatozoa, and demonstrates undesirable effects, namely inhibition of sperm motility and essential sperm signalling pathways. The aim of the study was to evaluate the influence of different thawing methods on bull’s semen characteristics. The semen was collected and processed from 8 bulls (Holstein, n = 4; Czech Fleckvieh, n = 4) kept in private Sire insemination Centre. Four different thawing methods were tested: control thawing methods (temperature = 38.5 °C, time = 30 seconds), slow thawing method (temperature = 30 °C, time = 50 seconds), moderate thawing method (temperature = 50 °C, time = 15 seconds), rapid thawing rate (temperature = 70 °C, time = 3 seconds). The percentage rate of total and progressive motile spermatozoa above head as well as movement characteristics of straight‑line velocity (VSL, µm/s) and linearity (LIN, %) were recorded using CASA system immediately after thawing and after 2 hrs. of heat incubation (±38 °C). Subsequently the differences between semen characteristics measured immediately after thawing and after 2 hours of incubation were calculated. The data were analyzed with SAS software. All the evaluated semen traits reached significantly lowest values in total motility (−8 % to −10.5 %, P < 0.05), progressive motility (−5.6 % to −10 %, P < 0.05) VSL (−3.9 µm/s to −9 µm/s, P < 0.05) and LIN (−1.5 % to −4.8 %, P < 0.05) in control thawing method immediately after the thawing compared to others. However, these differences were negligible after 2 hrs. of incubation. The highest values of progressive motility and movement characteristics after thawing and after 2 hours incubation were detected using slow thawing method. Moreover, the rapid method of thawing showed the significantly the lowest results in progressive motility (−3.6 % to −6.6 %), VSL (−3.2 to −5.6 µm/s) and LIN (−3 % to −4.6 %) characteristics assessed after 2 hrs. of incubation compared the others (P < 0.05). Control method of thawing was most stable during the incubation and showed the significantly lowest decrease of total motility (−7.6 % to −11.8 %, P < 0.05), progressive spermatozoa motility above head (−7.5 % to −9.8 %, P < 0.05) and VSL characteristics (−2.4 to −9.7 µm/s, P < 0.05) during incubation. Therefore, this thawing protocol can be recommended at this study based on the lowest spermatozoa characteristics decline during incubation for using artificial insemination.

Boar seminal plasma (SP) proteins were associated with differences on sperm resistance to cooling at 17°C. However, information about seminal plasma proteins in boars classified by capacity of semen preservation and in vivo fertility remains lacking. Thus, the objective was to evaluate the SP proteome in boars classified by capacity of semen preservation and putative biomarkers for fertility. The ejaculates from high-preservation (HP) showed higher progressive motility during all 5 days than the low-preservation (LP) boars. There was no difference for farrowing rate between ejaculates from LP (89.7%) and HP boars (88.4%). The LP boars presented lower total piglets born (14.0 ± 0.2) than HP (14.8 ± 0.2; p < 0.01). A total of 257 proteins were identified, where 184 were present in both classes of boar, and 41 and 32 were identified only in LP and HP boars, respectively. Nine proteins were differently expressed: five were more abundant in HP (SPMI, ZPBP1, FN1, HPX, and C3) and four in LP boars (B2M, COL1A1, NKX3-2, and MPZL1). The HP boars had an increased abundance of SP proteins related to sperm resistance and fecundation process which explains the better TPB. Objective: To improve the indicators of motility, survival and fertilizing ability of spermatozoa by optimizing temperature factors and the duration of exposure at unfreezing straws. Methods: Straws by volume 0.25 mL were thawed at water bath temperatures at 65 °C, 67 °C and 70 °C for 6–7 seconds and at 75 °C for 4–6 seconds. Impact of exposure time and temperature thawing in the water bath on motility and survival of spermatozoa were studied.
