Salt stress is one of the preeminent abiotic stressors capable of strongly impacting crop productivity and quality. Within the array of strategies garnering interest in safeguarding crops against abiotic stresses, the use of plant biostimulants is emerging as a noteworthy avenue. For the above, there is an increasing interest in finding new plant extracts showing biostimulating effects in crops. In the present study, the efficacy of an aqueous extract from an aquatic species, the duckweed (Lemna minor L.), was assessed in olive plants (cv. Arbequina) grown in hydroponics and exposed to severe saline stress (150 mM NaCl). Salt stress caused considerable diminutions in biomass production, leaf net photosynthesis (Pn), leaf transpiration rate (E), and stomatal conductance (gs). The application of the duckweed extract resulted in a notable plant functionality recovery and counteracted the detrimental effects of the NaCl stress. Indeed, the plants stressed with NaCl and treated with the extract showed enhanced physiological and biometric traits compared to samples treated with NaCl alone. In particular, the duckweed extract improved photosynthetic activity and stomatal conductance, reduced the intercellular CO2 concentration, and ameliorated other physiological and morphological parameters. All these benefits influenced the whole plant growth, allowing samples treated with the extract to maintain a similar performance to that exhibited by the Control plants. [ABSTRACT FROM AUTHOR]

In vitro propagation, also known as micropropagation, has become the most widely employed method for blackberry propagation, as it overcomes the limitations of the traditional asexual propagation methods (mainly layering and cutting). In this context, synthetic seed technology represents a strategy to enhance the productivity of in vitro propagation and facilitates the exchange of plant materials between laboratories, contributing to germplasm conservation. This study aimed to identify the most suitable vegetative propagule for the encapsulation of blackberry. To this end, uninodal microcuttings (nodes) and the base of clumps were used to produce synthetic seeds for the cultivars Thornfree and Chester. Forty-five days after sowing, viability (percentage of green propagules without browning or necrosis), regeneration (percentage of propagules that sprouted and rooted simultaneously), number of shoots produced, shoot length, number of roots produced, root length, and the fresh and dry weights of the plantlets were measured. The results demonstrated that both considered propagules allowed us to obtain satisfactory regeneration rates. However, plantlets originating from the encapsulated clump's base had more shoots and roots, resulting in greater fresh and dry weights than the plantlets derived from encapsulated nodes. Therefore, for achieving more robust plantlets and enhancing overall procedural efficiency, we recommend using the base of clumps as a propagule for blackberry encapsulation. [ABSTRACT FROM AUTHOR]

Selenium (Se) is not essential for plant nutrition, however, growing evidence suggests a role of this element against oxidative stress. Here, we investigated the antioxidant effect of Se in an experimental system consisting of suspensions of olive callus cells exposed to hydrogen peroxide. Due to the disruptive capacity of oxidative stress on cytosolic Ca2+ homeostasis, Se treatments were simply and effectively assessed with cytosolic Ca2+ measurements. Se did not alter cell morphology, nor interfered with fluorometric determinations, suggesting a suitable model for monitoring alterations of cytosolic Ca2+. Furthermore, we used thapsigargin (TG), a known inhibitor of the Ca2+-ATPases of the endoplasmic reticulum, to establish whether Se had a specific impact on oxidative stress. We found that Se antagonized H2O2-mediated perturbations of cytosolic Ca2+, but was unable to offset TG-mediated disruptions. We conclude that Se merits consideration in strategies designed to tackle abiotic stress leading to oxidative insults. This study deals jointly with three aspects of environmental, agricultural and energy sustainability: (a) Biogas is a booming energy source worldwide, resulting in an increasing production of digestate, its main by-product; (b) The extraction of peat, mainly used for nursery substrates, is being banned due to the destruction of natural habitats and release of GHGs; (c) Compost can represent a replacement of peat and contributes to the containment of GHGs. This study has verified how a compost obtained from digestate can be used as a substitute for peat in the nursery sector. While previous studies have evaluated compost use on just one species at a time, this study compared the same compost on two very different species: olive tree and hazelnut tree, both with growing interest for new tree plantings. Two concentrations of compost in the potting substrate of nursery seedlings were evaluated: 30% and 45% by weight, measuring the effect on some growth parameters during the growing season. The trials showed responses positive for olive and substantially negative for hazelnut: olive trees manifested better growth parameters with 45% compost, as opposed to hazelnut, where the addition of 45% compost worsened all growth parameters. A general conclusion can be drawn: in the nursery sector, compost can be used to replace peat, but this replacement can almost never be 100 percent, having instead to calibrate the percentage of replacement according to the characteristics of the compost and the individual edaphic needs of the plant species. Inhalation of olive pollen (Olea europaea L.) is one of the main causes of allergy in Mediterranean countries and some areas of North America. The response to allergens consists in the production of inflammatory cytokines which is mediated by the deregulation of Ca2+ signals. In this study, the biological activity of the material released in olive pollen hydration (PMR) was tested on Ca2+ cytosolic of PE/CA-PJ15 cells (PJ-15). Ca2+ cytosolic was determined by fluorometric assay with the cell line PE/CA-PJ15 (PJ-15) labeled with the fluorescent probe FURA 2 AM. The material released in olive pollen hydration (PMR) was analyzed by HPLC for the determination of phenolic acids. PMR was subjected to fractionation by gel filtration, and the fractions with Ca2+-chelating activity were tested with SDS-PAGE and the single bands characterized by proteomic analysis. PMR showed high Ca2+-chelating activity and is able of blocking the increase Ca2+-cytosolic produced by thapsigargin (TG). PMR then restored Ca2+ homeostasis in PJ-15 cells deregulated by the endoplasmic reticulum Ca2+-ATPases inhibitor. It is therefore possible that PMR can antagonize the effects of allergens on Ca2+ cytosolic. The analytical characterization of the material released by the pollen highlighted in the pollen allergen Ole e 3 and in the p-coumaric acid the possible culprits of the Ca2+-antagonist activity of PMR. Furthermore, the sequence of Ole e 3 could provide information for the possible construction of a synthetic peptide to be used in an allergy-targeted Ca2+-antagonist therapy. [ABSTRACT FROM AUTHOR]
