Metabolic syndrome (MetS) is a cluster of metabolic abnormalities that occur concurrently and increase the risk of cardiovascular disease. 3-mercaptopyruvate sulfurtransferase (MPST) is a cysteine-catabolizing enzyme that yields pyruvate and hydrogen sulfide (H2S) and plays a central role in the regulation of energy homeostasis. Herein, we seek to investigate the role of MPST/H2S in MetS and its cardiovascular consequences using a mouse model of the disease. Mice were fed a high-fat diet (HFD) for 15 weeks to induce obesity and hyperglycemia and administrated a nitric oxide synthase inhibitor, during the last 5 weeks to induce hypertension and MetS. This model caused a mild left ventricular (LV) diastolic dysfunction and vascular endothelial dysfunction. Free H2S and sulfane-sulfur levels were decreased in the aorta, but unaltered in the heart. Also, downregulation of MPST and thiosulfate sulfuretransferase (TST) were observed in the aorta. Global deletion of Mpst (Mpst-/-) resulted in increased body weight and greater glucose intolerance in mice with MetS, without affecting their blood pressure, and caused an upregulation of genes involved in immune responses in the vasculature suggestive of T-cell infiltration and activation. Pharmacological restoration of H2S levels ameliorated the comorbidities of MetS; GYY4137 administration reduced body weight and blood pressure, attenuated cardiac fibrosis and improved glucose handling and endothelium-dependent relaxation. In conclusion, this study found that reduced MPST/H2S exacerbates the pathological changes associated with MetS and contributes to vascular inflammation. H2S supplementation emerges as a potential therapeutic approach to treat the abnormalities associated with MetS.

The diagnosis of pediatric tuberculosis (TB) poses a challenge for clinical teams worldwide. TB-mediated changes in the expression of host genes in the peripheral blood can serve as diagnostic biomarkers and can provide better insights into the host immune mechanisms of childhood TB. Peripheral blood mononuclear cells (PBMCs) from children (n=102) with microbiologically confirmed TB disease, TB infection (TBI), pneumonia, and healthy controls (HC) were stimulated with either the Purified Protein Derivative (PPD) or the Early Secretory Antigen 6kDa-Culture Filtrate Protein 10 (ESAT6-CFP10) complex of Mycobacterium tuberculosis (Mtb). RNA was extracted and quantified using gene expression microarrays. Differential expression analysis was performed comparing microbiologically confirmed TB to the other diagnostic groups for the stimulated and unstimulated samples. Using variable selection, we identified sparse diagnostic gene signatures; one gene (PID1) was able to distinguish TB from pneumonia after ESAT6-CFP10 stimulation with an AUC of 100% in the test set, while a combination of two genes (STAT1 and IFI44) achieved an AUC of 91.7% (CI95% 75.0%-100%) in the test set after PPD stimulation. The number of significantly differentially expressed (SDE) genes was higher when contrasting TB to pneumonia or HC in stimulated samples, compared to unstimulated ones, leading to a larger pool of candidate diagnostic biomarkers. Our approach provides enlightened aspects of peripheral TB-specific responses and can form the basis for a point of care test meeting the World Health Organization (WHO) Target Product Profile (TPP) for pediatric TB.

ABSTRACT Objectives This study aimed to investigate possible differences of functional occlusal variables between a group of migraine patients (MG) and a control group (CG). Materials and Methods Each group included 50 individuals. Instrumental functional analysis and digital occlusal analysis were performed. Variables examined were condylar displacement from a reference position to maximum intercuspation; angular difference between the steepness of the articular eminence and the contra‐lateral canine guidance; and angular difference between the steepness of the articular eminence and the ipsilateral central incisor guidance and occlusal plane inclination. Self‐reported grinding and occlusal index were also investigated. Results There were statistically significant differences in the extent of retral condylar displacement assessed both clinically [MG: 0.49 mm (SD 0.67 mm), CG: 0.29 mm (SD 0.27 mm), p = 0.012] and digital‐mechanically [MG: 1.53 mm (SD 0.95 mm), CG: 0.9 mm (SD 0.66 mm), p = 0.001], the angular difference between the steepness of the articular eminence and the contra‐lateral canine guidance [MG: 13.11° (SD 8.33°), CG: 9.47° (SD 7.08°), p = 0.021 and MG: 12.94° (SD 8.71°), CG: 9.44° (SD 8.70°), p = 0.017], and the occlusal plane inclination [MG: 11.16° (SD 4.66°), CG: 9.09° (SD 4.37°), p = 0.024]. Stromal and immune cells and their interactions have gained the attention of cardiology researchers and clinicians in recent years as their contribution in cardiac repair is increasingly recognized. The repair process in the heart is a particularly critical constellation of complex molecular and cellular events and interactions that characteristically fail to ensure adequate recovery following injury, insult, or exposure to stress conditions in this regeneration-hostile organ. The tremendous consequence of this pronounced inability to maintain homeostatic states is being translated in numerous ways promoting progress into heart failure, a deadly, irreversible condition requiring organ transplantation. Fibrosis is in fact a repair response eventually promoting cardiac dysfunction and cardiac fibroblasts are the major cellular players in this process, overproducing collagens and other extracellular matrix components when activated. On the other hand, macrophages may differentially affect fibroblasts and cardiac repair depending on their status and subsets. The opposite interaction is also probable. We discuss here the multifaceted aspects and crosstalk of this cell dipole and the opportunities it may offer for beneficial manipulation approaches that will hopefully lead to progress in heart disease interventions.

Recognising the urgency of addressing climate change and the imperative to mitigate its effects, the European Union (EU) has embarked on a transformative journey to reshape its energy landscape, with a pronounced emphasis on harnessing renewable energy sources (RESs) and augmenting their contribution to electricity generation. To propel Europe towards sustainable development through its energy transition, EU member states are encouraged to foster collaborative efforts on a European scale, inviting neighbouring countries to participate in joint ventures aimed at leveraging RESs for electricity generation. Consequently, it becomes imperative to evaluate the potential depth of cooperation among these nations, assessing how such partnerships can align with Europe’s overarching objectives while fostering mutually beneficial conditions. This paper seeks to undertake a thorough analysis and evaluation of the potential impacts of such cooperation, both in advancing RES objectives and in promoting broader cooperation goals concerning the countries involved. The appropriate methodological framework has been developed, utilising and implementing the fuzzy PROMETHEE multicriteria decision analysis method, to address the problem’s multidimensional character, intending to implement an appropriate action plan and promote production from RESs. The methodology has been applied to assess alternative strategies in two case study countries, Morocco and Egypt, while important outcomes have emerged towards the successful implementation of cooperation mechanisms promoting RES.

The reduction in the inner diameter of the artery due to the creation of atheromatic plaque on the artery lumen, known as artery stenosis, disrupts the blood flow, leading to medical complications, which can be fatal. The angioplasty procedure aims to reopen the artery and uses a stent to keep it open. In this study, an effort is made to determine the point of the stent, the plaque and the artery during the expansion phase of the angioplasty using the in silico Finite Element Analysis method. A literature-based design was chosen for the stent geometry, whereas simplified shapes of the balloon and the two artery layers were used. Additionally, two plaque designs were the benchmark for the eight distinct artery stenosis models within the Abaqus environment. In the context of stent angioplasty simulations, failure patterns were investigated. An inverse relationship was observed between artery stenosis and pressure at the artery failure point, while an increased danger of interlaminar failure was detected in models with larger artery stenosis. This study verifies the necessity for the inclusion of interlaminar failure in future angioplasty research.

In recent years, researchers in the field of natural heritage have intensified their efforts to develop new ways to enhance the promotion and accessibility of natural content in order to attract more audiences using virtual representations of physical objects (digital twins). Therefore, they are increasingly incorporating new technologies and digital tools in their operations since their usage by the general public and in the natural heritage (NH) museums is considered particularly effective. Simultaneously, the increasing quality of the produced digitizations has opened up new opportunities for the exploitation of the outcomes of digitization beyond the initial anticipations. Responding to the growing demand of museum visitors for a personalized digital tour experience, especially amidst the recent COVID-19 pandemic, the v-PalM project aims to develop a digital platform to offer virtual guidance and education services at the Museum of Paleontology and Geology, which is hosted at the National Kapodistrian University of Athens. The development of the platform will be based on collecting data through various methods, including crowdsourcing, innovative information, and communication technologies, taking advantage of content digitization using 3D scanning devices. This paper demonstrates an enhanced methodology for the digitization of paleontological exhibits. The methodology uses photogrammetry and laser scanning methods from various devices, such as drones, laser scanners, and smartphones. These methods create digital twins that are suitable for various scenarios, including research, education, and entertainment. The proposed methodology has been applied to over fifty paleontological museum exhibits of varying sizes and complexities, and the resulting 3D models exhibit high accuracy in both their material and geometric aspects, while they also feature crucial details that assist researchers and the scientific community.

Background: The objective of this work was to investigate the association between the function of the stomatognathic system and migraine presence through an instrumental functional analysis in a group of diagnosed migraine patients and a control group. Methods: This study included 50 individuals in each group. A jaw-tracking analysis was performed using Cadiax 4. Tracings of the following movements were recorded: open/close, protrusion/retrusion, mediotrusion, speech, bruxing, and mastication. The tracings were evaluated for their quantity, quality, transversal characteristics, speed, curvature pattern, and condylar stability. Results: Statistically significant differences between the groups were established for several aspects of the evaluation. Migraineurs presented with (a) higher values of mandibular lateral translation in protrusion/retrusion (p = 0.001), open/close (p = 0.031), and mastication (p = 0.016); (b) transient velocity losses in open/close (p = 0.001) and protrusive movements (p = 0.018); (c) a compromised condylar stability for protrusion/retrusion (p = 0.001) and mediotrusion (p = 0.003); (d) a compromised quality for protrusion/retrusion (p < 0.001) and mediotrusion (p = 0.003); and (e) a more frequent “figure-eight” curvature in open/close (p = 0.012). Conclusions: The importance of the stomatognathic function in migraine pathogenesis and treatment should be considered by using a patient-centered and interdisciplinary approach.

Since its conceptualization in 2008, blockchain technology has advanced rapidly and been applied in multiple domains. In higher education, blockchain can be applied to develop ICT systems that can revolutionize student accreditation through certificate verification and micro-accreditations, which represent skills and other learning outcomes, in the form of digital/smart badges. While there are multiple studies that highlight the significance of blockchain in higher education and propose digital systems, few of those studies include the evaluation of such proposed systems by real users. As such, the research question of how useful a higher education blockchain system would be for its relevant stakeholders remains largely unanswered. In the research publication at hand, a blockchain-powered higher education platform was applied in the School of Electrical and Computer Engineering of the National Technical University of Athens, where it was used and evaluated by students and professors at the school. The evaluation of the platform was positive, and participants found that the smart badge functionality was among the most useful. Finally, the execution and evaluation of the pilot led to several lessons learned and policy recommendations towards dealing with existing barriers and further promoting blockchain in higher education.

Three neutral nickel(II) complexes of 3,5–dibromo–salicylaldehyde (3,5–diBr–saloH) were synthesized in the presence or absence of 1,10–phenanthroline (phen) or its derivative 2,9–dimethyl–1,10–phenanthroline (neoc) as co–ligands, namely [Ni(3,5–diBr–salo)2(neoc)] (complex 1), [Ni(3,5–diBr–salo)2(phen)] (complex 2) and [Ni(3,5–diBr–salo)2(H2O)2] (complex 3), and were characterized by various techniques. The crystal structure of [Ni(3,5–diBr–salo)2(neoc)] was determined by single-crystal X-ray crystallography. According to employed studying techniques, the complexes interact tightly with calf-thymus DNA by an intercalative fashion. Furthermore, compounds 1–3 bind tightly and reversibly to human and bovine serum albumin.

The deployment of carbon management strategies like carbon capture and storage (CCS) and carbon dioxide removal (CDR) at scale will require significant investments in transport infrastructure to deliver CO2 to reliable storage. While pipeline transport has dominated the conversation due to economic reasons, there is increasing evidence that other modes may become viable alternatives when considering scale, regional opportunities, and social acceptance. This paper assesses the viability of rail for CO2 transport in the United States using market analysis, techno-economic assessment and geographic information systems mapping. We believe rail presents many advantages, notably in existing infrastructure with established right-of-ways, but also as an instrument to address unwanted effects of our energy transition, particularly in coal communities. We find that the strategic replacement of coal as a freight commodity could translate into 100 Mt/yr of CO2 movement by rail by 2050, and support up to 60,000 jobs in that industry. Further, we find that while rail pricing is notoriously volatile, there is strong support for rail being the least cost option over pipeline for volumes under 2 Mt CO2 per year, which aligns well with smaller, more risk-averse, and distributed carbon management projects that are scheduled to deploy over the next decade. Rail can also be an alternative in regions where CO2 pipeline projects have had limited success, like in the Midwest, where CO2 is captured from ethanol plants that are already serviced by rail networks. Likewise, rail can service roughly 25% of point-source CCS opportunities that are not proximal to projected trunk pipeline networks, of which 94% are located 1-mile from railroad. Finally, rail may be an integral part of CDR development in regions that are not coterminous with geologic storage, particularly in the Western and Northern US.

Clean energy technological innovations are widely acknowledged as a prerequisite to achieving ambitious long-term energy and climate targets. However, the optimal speed of their adoption has been parsimoniously studied in the literature. This study seeks to identify the optimal intensity of moving to a green hydrogen electricity sector in Greece, using the OSeMOSYS energy modeling framework. Green hydrogen policies are evaluated, first, on the basis of their robustness against uncertainty and, afterwards, against conflicting performance criteria and for different decision-making profiles towards risk, by applying the VIKOR and TOPSIS multi-criteria decision aid methods. Although our analysis focuses exclusively on the power sector and compares different rates of hydrogen penetration compared to a business-as-usual case without considering other game-changing innovations (such as other types of storage or carbon capture and storage), we find that a national transition to a green hydrogen economy can support Greece in potentially cutting at least 16 MtCO2 while stimulating investments of EUR 10–13 bn. over 2030–2050.
