Abstract Background Pseudomyxoma peritonei (PMP) is a rare malignant peritoneal tumor that readily recurs and metastasizes. Studies have shown that cancer stem cells (CSCs) play an important role in tumor recurrence, metastasis, and prognosis. Objective In this study, our aim was to isolate CSCs from various tissues of PMP patients and compare their proliferation, migration, and anti-inflammatory abilities. Methods We identified CSCs subsets with markers CD133+, CD166+, and CD133+/CD166+ at the gene level using single-cell mRNA sequencing (scRNA-seq). Appendiceal CSCs (AC), peritoneal CSCs (PC), and mucous CSCs (MC) were obtained using MACSQuant Tyto sorting technology and FlowSight imaging flow cytometry. The cells were cultured and markers were identified. Finally, the functional phenotypes of the three cell types were compared. Results CSCs content was highest in the appendiceal tumor tissue and lowest in the mucous tissue. The cell viability rate of the sorted CSCs was above 98%, and the positive rate of CD133+ and CD166+ was 70–80%, and CD133+/CD166+ was about 30%. Among the three types of CSCs, MC had the highest proliferation ability, and TNF-α has the greatest inhibitory effect on AC migration. Conclusion AC in patients was more inert and anti-inflammatory, whereas abdominal cavity MC and PC were more active. This study revealed the biological characteristics of CSCs in different tumor tissues of patients with PMP, providing a reference for future targeted CSCs therapy.

Abstract Background Evidence suggests that PTSD symptoms following public health emergencies are influenced by many factors. The aim of this study was to investigate the relationship between perceived social support and Post-traumatic stress disorder symptoms and to explore the chain-mediated role of resilience and positive coping style, among medical staff in Hubei Province, China, during a public health emergency. Methods Convenience sampling was used to select medical staff from two general hospitals in Hubei Province in July 2022 for this study. A total of 2,751 medical staff were included in the study. Multidimensional Scale of Perceived Social Support, 10- itemConnor- Davidson resilience scale, Simplified Coping Style Questionnaire and The Post-traumatic stress disorder Checklist for DSM-5 were used to assess the levels of perceived social support, resilience, coping style and Post-traumatic stress disorder symptoms of medical staff two years after the public health emergency. Statistical descriptions were conducted using SPSS, and a structural equation model was established using AMOS to analyze the chain-mediated roles of resilience and positive coping style between perceived social support and Post-traumatic stress disorder symptoms. Abstract Intravesical therapy (IT) is widely used to tackle various urological diseases. However, its clinical efficacy is decreased by the impermeability of various barriers presented on the bladder luminal surface, including the urinary mucus layer and the densely packed tissue barrier. In this study, we report a mucoadhesive-to-penetrating nanomotors-in-hydrogel system for urothelium-oriented intravesical drug delivery. Upon intravesical instillation, its poloxamer 407 (PLX) hydrogel gelated and adhered to the urothelium to prolong its intravesical retention. The urea afterwards diffused into the hydrogel, thus generating a concentration gradient. Urease-powered membrane nanomotors (UMN) without asymmetric surface engineering could catalyze the urea and migrate down this concentration gradient to deeply and unidirectionally penetrate the urothelial barrier. Moreover, the intravesical hybrid system-delivered gemcitabine could effectively inhibit the bladder tumor growth without inducing any side effect. Therefore, our mucoadhesive-to-penetrating nanomotors-in-hydrogel system could serve as an alternative to IT to meet the clinical need for more efficacious therapeutics for urological diseases. Abstract With increases in life expectancy, the number of patients requiring joint replacement therapy and experiencing periprosthetic osteolysis, the most common complication leading to implant failure, is growing or underestimated. In this study, we found that osteolysis progression and osteoclast differentiation in the surface of the skull bone of adult mice were accompanied by significant expansion of lymphatic vessels within bones. Using recombinant VEGF-C protein to activate VEGFR3 and promote proliferation of lymphatic vessels in bone, we counteracted excessive differentiation of osteoclasts and osteolysis caused by titanium alloy particles or inflammatory cytokines LPS/TNF-α. However, this effect was not observed in aged mice because adipogenically differentiated mesenchymal stem cells (MSCs) inhibited the response of lymphatic endothelial cells to agonist proteins. The addition of the JAK inhibitor ruxolitinib restored the response of lymphatic vessels to external stimuli in aged mice to protect against osteolysis progression. These findings suggest that inhibiting SASP secretion by adipogenically differentiated MSCs while activating lymphatic vessels in bone offers a new method to prevent periprosthetic osteolysis during joint replacement follow-up.

Hydrogels, comprising 3D hydrophilic polymer networks, have emerged as promising biomaterial candidates for emulating the structure of biological tissues and delivering drugs through topical administration with good biocompatibility. Nanozymes can catalyze endogenous biomolecules, thereby initiating or inhibiting in vivo biological processes. A nanozyme-hydrogel composite inherits the biological functions of hydrogels and nanozymes, where the nanozyme serves as the catalytic core and the hydrogel forms the structural scaffold. Moreover, the composite can concentrate nanozymes in targeted lesions and catalyze the binding of a specific group of substrates, resulting in pathological microenvironment remodeling and drug-penetrating barrier impairment. The composite also shields nanozymes to prevent burst release during catalytic production and reduce related toxicity. Currently, the application of these composites has been extended to antibacterial, anti-inflammatory, anticancer, and tissue repair applications. In this review, we elucidate the preparation methods for nanozyme-hydrogel composites, provide compelling evidence of their advantages in drug delivery and provide a comprehensive overview of their biological application.

Abstract Peptide‐based drugs hold great potential for cancer treatment, and their effectiveness is driven by mechanisms on how peptides target cancer cells and escape from potential lysosomal entrapment post‐endocytosis. Yet, the mechanisms remain elusive, which hinder the design of peptide‐based drugs. Here hendeca‐arginine peptides (R11) are synthesized for targeted delivery in bladder carcinoma (BC), investigated the targeting efficiency and elucidated the mechanism of peptide‐based delivery, with the aim of refining the design and efficacy of peptide‐based therapeutics. It is demonstrated that the over‐activated Piezo1/integrin β1 (ITGB1) signaling axis significantly facilitates tumor‐targeted delivery of R11 peptides via macropinocytosis. Furthermore, R11 peptides formed hydrogen bonds with integrin β1, facilitating targeting and penetration into tumor cells. Additionally, R11 peptides protected integrin β1 from lysosome degradation, promoting its recycling from cytoplasm to membrane. Moreover, this findings establish a positive feedback loop wherein R11 peptides activate Piezo1 by increasing membrane fusion, promoting Ca2+ releasing and resulting in enhanced integrin β1‐mediated endocytosis in both orthotopic models and clinical tissues, demonstrating effective tumor‐targeted delivery. Eventually, the Piezo1/integrin β1 signaling axis promoted cellular uptake and transport of peptides, establishing a positive feedback loop, promoting mechanical delivery to cancer and offering possibilities for drug modification in cancer therapy.

Henan encountered a multi-stage severe convective weather event (hereinafter referred to as "6.13" event) accompanied by local tornado on 13 June 2022. Using conventional observation data, regional automatic weather station data, satellite cloud images, Doppler weather radar data, ERA5 reanalysis data and other relevant data, we conducted an analysis on atmospheric circulation environment and the evolution characteristics of mesoscale systems for the "6.13" event. In addition, we discussed the initiation and maintenance mechanisms of convection in the different periods of this event and the radar characteristics of the local tornado in Puyang of Henan. Results are as follows. (1) The "6.13" event occurs under the northwest airflow behind the northeast cold vortex trough, and it is characterized by wide impact range, long duration and multiple types of disaster weather. (2) Radar detection results show that the severe convective systems in the "6.13" event passed through Henan in three periods one after another, and strong echoes in each period lasts for 8-9 hours, with the moving speed of 30-50 km·h-1. Most of the severe convective systems are multi-cells that all moved from northwest to southeast, with some of their areas overlapping under the guidance of northwest airflow. (3) Maintenance of the strong conditional instability and the moderate to strong vertical wind shear over Henan is an important cause for multi-stage severe convective weather in the "6.13" event lasting for a long time. The first period of local severe convection in Henan is mainly triggered by the convergence line or convergence center in the boundary layer formed by the combined effect of daily changes of wind field, local cold air activity, topographic distribution, while the second and third periods of severe convection are triggered by the gust front or outflow boundary accompanied by the surface mesoscale thunderstorm high formed by the strong developing convection systems in the surrounding area in the previous period or the same period. (4) The local tornado in Puyang is generated by a rapidly developing supercell storm. The hook echo and mesoscale cyclonic vortex appear 12 minutes after this supercell storm, and the tornado vortex characteristics (TVS) appear 18 minutes later. The hook echo and mesoscale cyclonic vortex are 6 minutes ahead of the occurrence of tornado, which can provide a reference information for early warning local tornadoes.

Abstract Background Benign uretero‐ileal anastomotic stricture (UIAS) is a potentially serious complication that can arise after radical cystectomy (RC) and subsequent urinary diversion. To preserve residual renal function and improve prognosis, it is crucial to derive insights from experience and tailor individualized treatment strategies for different patients. Patients and Methods From October 2014 to June 2021, a total of 47 patients with benign UIAS underwent endoscopic management (n = 19) or reimplantation surgery (n = 28). The basic data, perioperative conditions, and postoperative outcomes of the two groups were compared and analyzed to evaluate efficacy. Results Comparing preoperative and postoperative clinical efficacy within the same group, the endoscopic group showed no significant differences in creatinine and blood urea nitrogen (BUN) levels before surgery or after extubation (p > 0.05). However, significant differences were observed in glomerular filtration rate (GFR) levels on the affected side before surgery and after extubation (p 0.05). Postoperative clinical efficacy showed no significant difference in creatinine and BUN levels between the two groups (p > 0.05). BackgroundThe role of focal amplifications and extrachromosomal circular DNA (eccDNA) is still uncertain in prostate adenocarcinoma (PRAD). Here, we first mapped the global characterizations of eccDNA and then investigate the characterization of eccDNA-amplified key differentially expressed encoded genes (eKDEGs) in the progression, immune response and immunotherapy of PRAD.MethodsCircular_seq was used in conjunction with the TCGA-PRAD transcriptome dataset to sequence, annotate, and filter for eccDNA-amplified differentially expressed coding genes (eDEGs) in PRAD and para-cancerous normal prostate tissues. Afterwards, risk models were created and eKDEGs linked to the PRAD prognosis were identified using Cox and Lasso regression analysis. The immune microenvironment of the risk model was quantified using a variety of immunological algorithms, which also identified its characteristics with regard to immunotherapy, immune response, and immune infiltration.ResultsIn this research, there was no significant difference in the size, type, and chromosomal distribution of eccDNA in PRAD and para-cancerous normal prostate tissues. However, 4,290 differentially expressed eccDNAs were identified and 1,981 coding genes were amplified. Following that, 499 eDEGs were tested in conjunction with the transcriptome dataset from TCGA-PRAD. By using Cox and Lasso regression techniques, ZNF330 and PITPNM3 were identified as eKDEGs of PRAD, and a new PRAD risk model was conducted based on this. Survival analysis showed that the high-risk group of this model was associated with poor prognosis and validated in external data. Immune infiltration analysis showed that the model risks affected immune cell infiltration in PRAD, not only mediating changes in immune cell function, but also correlating with immunophenotyping. Furthermore, the high-risk group was negatively associated with anti-CTLA-4/anti-PD-1 response and mutational burden. In addition, Tumor Immune Dysfunction and Exclusion analyses showed that high-risk group was more prone to immune escape. Drug sensitivity analyses identified 10 drugs, which were instructive for PRAD treatment.ConclusionZNF330 and PITPNM are the eKDEGs for PRAD, which can be used as potential new prognostic markers. The two-factor combined risk model can effectively assess the survival and prognosis of PRAD patients, but also can predict the different responses of immunotherapy to PRAD patients, which may provide new ideas for PRAD immunotherapy.

Understanding the spatial pattern of vegetation in the Gobi area is of paramount importance for preserving its vegetation and the overall stability of the ecosystem. Here, we investigated the populations of the predominant species, Ephedra przewalskii, at various locations within the alluvial fan situated in the southern foothills of the Tianshan Mountains in the Gobi area. The location of the E. przewalskii distribution in the sample plot was extracted from unmanned aerial vehicle (UAV) images. This article analyzes the distribution patterns of the predominant species based on different null models, tests whether the distribution patterns are affected by habitat heterogeneity, and examines whether other ecological processes affect these patterns. Additionally, using the variance decomposition method, we analyze the explanatory power of habitat factors on the distribution patterns of the predominant species. The results show that: (1) Using UAVs to locate the distribution of vegetation information for sparse vegetation pattern analysis in the Gobi region has good applicability and prospects; (2) the frequency, density, and coverage of E. przewalskii decrease sharply with decreasing altitude. At a small scale (0∼10 m), the E. przewalskii population is mainly aggregated, whereas on a large scale (>10 m), it primarily exhibits uniform or random distributions; (3) the E. przewalskii population distribution pattern is significantly affected by habitat heterogeneity, mainly on a large scale. The species distribution in the study area strongly correlates with the altitude, soil moisture, and topographic wetness index. Habitat factors can explain 64.2 % of the reasons for species distribution; and (4) The clustering of E. przewalskii population at the apex of the alluvial fan is primarily driven by facilitation, whereas at the middle and bottom sections, it is predominantly influenced by habitat heterogeneity and restricted diffusion. The results enhance our comprehension of the spatial patterns and environmental adaptation mechanisms exhibited by the predominant species in the study area. Furthermore, they provide a scientific foundation for ecological conservation and biodiversity protection in the environmentally vulnerable Gobi region.

This paper introduces a novel approach to personal risk (PR) identification using federated learning (FL) in wireless communication scenarios, leveraging generalized information. The primary focus is on harnessing the power of distributed data across various wireless devices while ensuring data privacy and security, a critical concern in PR assessment. To this end, we propose an FL-based model that effectively aggregates learning from diverse, decentralized data sources to analyze the PR factors. The proposed method involves training local models on individual devices, which are then aggregated to form a comprehensive global model. This process not only preserves data privacy by keeping sensitive information on the device but also utilizes the widespread availability and connectivity of wireless devices to enhance data richness and model robustness. To address the challenges posed by the wireless environment, such as data heterogeneity and communication constraints, we further implement advanced aggregation algorithms and optimization techniques tailored to these unique conditions. We finally evaluate the performance of our proposed method based on two primary metrics of identification accuracy and convergence rate of the federated learning process. Through extensive simulations and real-world experiments, we demonstrate that our approach not only achieves high accuracy in PR identification but also ensures rapid convergence, making it a viable solution for real-time risk assessment in wireless networks.

Adaptive gradient algorithms have been successfully used in deep learning. Previous work reveals that adaptive gradient algorithms mainly borrow the moving average idea of heavy ball acceleration to estimate the first- and second-order moments of the gradient for accelerating convergence. However, Nesterov acceleration which uses the gradient at extrapolation point can achieve a faster convergence speed than heavy ball acceleration in theory. In this article, a new optimization algorithm which combines adaptive gradient algorithm with Nesterov acceleration by using a look-ahead scheme, called NALA, is proposed for deep learning. NALA iteratively updates two sets of weights, i.e., the ‘fast weights’ in its inner loop and the ‘slow weights’ in its outer loop. Concretely, NALA first updates the fast weights k times using Adam optimizer in the inner loop, and then updates the slow weights once in the direction of Nesterov’s Accelerated Gradient (NAG) in the outer loop. We compare NALA with several popular optimization algorithms on a range of image classification tasks on public datasets. The experimental results show that NALA can achieve faster convergence and higher accuracy than other popular optimization algorithms.

To study the effect of resting on the quality of frozen dough, the changes in the moisture state of the dough during the resting process (0, 30, 60, 90, 120 min), the moisture distribution of dough and their effects on ice crystal distribution, microstructure, and creep recovery characteristics after dough freezing were investigated. The results showed that the freezable water content in the dough was decreased by 0.96% with the extension of the resting time. The pores and number of ice crystals formed in the dough during the freezing process gradually decreased and the distribution of ice crystals became more and more uniform. The average pore area decreased from 22.65 to 11.80 with the extension of the resting time from 0 min to 120 min, and the number of pores decreased from 22.65 to 11.80 with the extension of the resting time. Motor imagery (MI) based brain computer interface (BCI) has been extensively studied to improve motor recovery for stroke patients by inducing neuroplasticity. However, due to the lower spatial resolution and signal-to-noise ratio (SNR) of electroencephalograph (EEG), MI based BCI system that involves decoding hand movements within the same limb remains lower classification accuracy and poorer practicality. To overcome the limitations, an adaptive hybrid BCI system combining MI and steady-state visually evoked potential (SSVEP) is developed to improve decoding accuracy while enhancing neural engagement. On the one hand, the SSVEP evoked by visual stimuli based on action-state flickering coding approach significantly improves the recognition accuracy compared to the pure MI based BCI. On the other hand, to reduce the impact of SSVEP on MI due to the dual-task interference effect, the event-related desynchronization (ERD) based neural engagement is monitored and employed for feedback in real-time to ensure the effective execution of MI tasks. Eight healthy subjects and six post-stroke patients were recruited to verify the effectiveness of the system. The results showed that the four-class gesture recognition accuracies of healthy individuals and patients could be improved to $94.37~\pm ~4.77$ % and $79.38~\pm ~6.26$ %, respectively. Moreover, the designed hybrid BCI could maintain the same degree of neural engagement as observed when subjects solely performed MI tasks. These phenomena demonstrated the interactivity and clinical utility of the developed system for the rehabilitation of hand function in stroke patients.

Neural networks have received a lot of attention in recent years. However, neural networks are rarely used in the construction industry, especially for the risk assessment of new projects such as green buildings. This paper was mainly based on the theoretical analysis of green building evaluation and the establishment of green building comprehensive evaluation index system. Based on artificial neural network, the comprehensive evaluation system of green building projects has been studied. According to the principle of construction risk evaluation index system, a risk evaluation index system corresponding to green building projects has been established. According to the established risk index system, the corresponding BP (back propagation) neural network model was constructed. The BP neural network was learned and trained by MATLAB (matrix&laboratory) software. BIM (Building Information Modeling) model can carry the characteristics of building parameters and the characteristics of BIM technology collaborative design, information interoperability, performance simulation and other characteristics were used. Based on the information transfer between Autodesk’s Revit software and other analysis software, with DB-Link (Database Link) plug-in, Access database, Excel functions and macro commands as the medium, a green building auxiliary evaluation module based on BIM technology was constructed. According to the established comprehensive evaluation index system, the software and neural network model were used to conduct a comprehensive evaluation of green buildings for a green complex building project. The utilization rate of recyclable materials in the architectural design scheme of this project accounted for 17.66% of the total building materials. The results of the evaluation show that a green comprehensive building project has been built in a good grade, which is in line with its actual situation in the green building grade.

Face forgery detection aims to identify manipulated or altered facial images or videos created using artificial intelligence. Existing detection methods exhibit favorable performance on high-quality videos, but the videos in daily applications are commonly compressed into low-quality formats via social media. The detection difficulty is increased by the poor quality, indistinct detail features, and noises such as artifacts in these images or videos. To address this challenge, we propose a multi-scale dual-branch network for compressed face forgery, called MDCF-Net, effectively capturing cross-domain forgery features at various scales in compressed facial images. The MDCF-Net comprises two branches: an RGB domain branch utilizing Transformers to extract multi-scale fine-texture features from the original RGB images; a frequency domain branch designed to capture artifacts in low-quality videos by extracting global spectral features as a supplementary measure. Then, we introduce a feature fusion module (FFM) based on multi-head attention to merge diverse feature representations in a spatial-frequency complementary manner. Extensive comparative experiments on public datasets such as FaceForensics++, Celeb-DF, and WildDeepfake demonstrate the significant advantage of MDCF-Net in detecting highly compressed and low-quality forged images or videos, especially in achieving state-of-the-art performance on the FaceForensics++ low-quality dataset. Our approach presents a new perspective and technology for low-quality face forgery detection.

How to achieve fast and accurate small object detection holds crucial theoretical and practical significance. However, this task encounters substantial challenges due to scale differences among instances in the scene, along with the scarcity of inherent features and weak representation of small instances. To alleviate the above problems, we propose a novel attention-based cascaded adaptive feature pyramid fusion network, CA2Det, to effectively improve the small object detection performance. First, to prevent the information degradation of small-instance features during training, we introduce the efficient and lightweight Shuffle Attention mechanism to highlight the features of small instances. Second, to mitigate the information conflicts arising from the scale inconsistency among instances, we design a double-layer cascaded adaptive fusion pyramid module, CAFP, which can effectively suppress the information conflicts while enabling full information exchange across layers. Finally, we combine sparse convolution to achieve efficient high-resolution input, providing richer geometric information of the instances. Compared to the baseline network, on the COCO benchmark dataset and the popular UAV dataset VisDrone, both of which contain a large number of small instances, the proposed method improves the detection accuracy mAP values by 1.1% and 2.2%, respectively, while having a good real-time detection speed.

Abstract The precise regulation of cellular behaviors within a confined, crowded intracellular environment is highly amenable in diagnostics and therapeutics. While synthetic circuitry system through a concatenated chemical reaction network has rarely been reported to mimic dynamic self‐assembly system. Herein, a catalytic self‐defined circuit (CSC) for the hierarchically concatenated assembly of DNA domino nanostructures is engineered. By incorporating pre‐sealed symmetrical fragments into the preying hairpin reactants, the CSC system allows the hierarchical DNA self‐assembly via a microRNA (miRNA)‐powered self‐sorting catalytic hybridization reaction. With minimal strand complexity, this self‐sustainable CSC system streamlined the circuit component and achieved localization‐intensified cascaded signal amplification. Profiting from the self‐adaptively concatenated hybridization reaction, a reliable and robust method has been achieved for discriminating carcinoma tissues from the corresponding para‐carcinoma tissues. The CSC‐sustained self‐assembly strategy provides a comprehensive and smart toolbox for organizing various hierarchical DNA nanostructures, which may facilitate more insights for clinical diagnosis and therapeutic assessment.

BackgroundKidney cancer is a prevalent malignancy with an increasing incidence worldwide. Blood cell indices and inflammation-related markers have shown huge potential as biomarkers for predicting cancer incidences, but that is not clear in kidney cancer. Our study aims to investigate the correlations of blood cell indices and inflammation-related markers with kidney cancer risk.MethodsWe performed a population-based cohort prospective analysis using data from the UK Biobank. A total of 466,994 participants, free of kidney cancer at baseline, were included in the analysis. The hazard ratios (HRs) and 95% confidence intervals (CIs) for kidney cancer risk were calculated using Cox proportional hazards regression models. Restricted cubic spline models were used to investigate nonlinear longitudinal associations. Stratified analyses were used to identify high-risk populations. The results were validated through sensitivity analyses.ResultsDuring a mean follow-up of 12.4 years, 1,710 of 466,994 participants developed kidney cancer. The Cox regression models showed that 13 blood cell indices and four inflammation-related markers were associated with kidney cancer incidence. The restricted cubic spline models showed non-linear relationships with kidney cancer. Finally, combined with stratified and sensitivity analyses, we found that the mean corpuscular hemoglobin concentration (MCHC), red blood cell distribution width (RDW), platelet distribution width (PDW), systemic immune-inflammation index (SII), and product of platelet count and neutrophil count (PPN) were related to enhanced kidney cancer risk with stable results.ConclusionOur findings identified that three blood cell indices (MCHC, RDW, and PDW) and two inflammation-related markers (SII and PPN) were independent risk factors for the incidence of kidney cancer. These indexes may serve as potential predictors for kidney cancer and aid in the development of targeted screening strategies for at-risk individuals.

Extensive research has been conducted on the waste sorting behavior (WSB) of residents, while it is the first time that the classification behavior of urban and rural residents is compared under the same theoretical framework in China. Based on questionnaire data from 478 urban and rural residents, structural equation modeling (SEM) was used to investigate the internal factors influencing the WSB by integrating the Theory of Planned Behavior (TPB) and the Norm Activation Model (NAM). Hierarchical regression analysis was utilized to investigate the moderating effect of external factors on the residents' intentions and behavior. The results show that the degree of deviation between rural residents' intentions and behavior is much larger than that of urban residents. Personal norms are the key factors affecting urban residents' waste sorting. In contrast, for rural residents, attitude is the most critical factor, but the influence of subjective norms is insignificant. In addition, we found that policy restraints and economic incentives significantly moderate the association between urban residents' sorting intention and behavior, with economic incentives having a better effect than policy restraints. In contrast, the impact of policy restraints on rural residents is better than that of urban areas. However, the moderating effect of economic incentives is insignificant for rural residents. The findings furnish the government with meaningful strategies to narrow the urban-rural waste management gap.

Abstract Background Bladder cancer (BCa) is a highly malignant tumor, and if left untreated, it can develop severe hematuria and tumor metastasis, thereby endangering the patient's life. The purpose of this paper was to detect the expression of ATAD3A in BCa and research the relationship between ATAD3A and pathological features of bladder cancer and the prognosis of patients. Methods First, the expression of ATAD3A in BCa and normal bladder tissues was analyzed based on the UALCAN and Oncomine public databases. Second, 491 cases of surgically resected bladder cancer specimens and 110 cases of normal adjacent tissues were immunohistochemically stained. The expression of ATAD3A was quantified, and the value and prognosis of ATAD3A as a biomarker of BCa were evaluated. Abstract A technique is developed to quantify the ultra-trace 231Pa (35–3904 ag) concentration in seawater using multi-collector inductively coupled plasma mass spectrometry (MC-ICP-MS). The method is a modification of the process developed by Shen et al. (Anal Chem 75(5):1075–1079, 2003. https://doi.org/10.1021/ac026247r ) and extends it to the application of very low levels of actinides, and the 35 ag 231Pa can be measured with a precision of 15%. The total process blank for the water column was 0.02 ag/g, while the values of the large and small particles were ~ 30 ag/g. The ionization efficiency (ions generated/atom loaded) varies from 0.7 to 2.4%. The measurement time is 2–5 min. The amount of 231Pa needed to produce 231Pa data with an uncertainty of ± 0.8–15% is 35–3904 ag (~ 0.9 × 105 to 10 × 106 atoms). This article presents a novel and feasible approach for researching the quantity of the ceramic phase and component optimization in high-temperature titanium alloys with small trace amounts of ceramic phases. Different near-α titanium alloys with varying yttrium and boron contents were prepared through the utilization of a vacuum non-consumable arc furnace melting method. The alloy used was a Ti-5.9Al-4Sn-3.9Zr-3.8Mo-0.4Si base. Its microstructure, texture, mechanical properties, and fracture behavior were studied. The observation of the as-cast structure shows that the addition of different doses of trace Y and B elements significantly refines both the original β grains and α grains. Moreover, the addition of the B element transforms the Widmanstätten structure in the titanium alloy structure into a basketweave structure. The addition of Y can refine the grain structure, improve the uniformity of the matrix structure, and act as a strong deoxidizer, which will take away the oxygen in the matrix and purify it. The TiB whiskers generated with the addition of B promotes dynamic recrystallization behavior and leads to more equiaxed α grains being precipitated around them, resulting in a significant refinement of the microstructure of the as-cast alloy. After adding a small amount of B, the texture strength of the α phase is significantly reduced, indicating that TiB whiskers inhibit the formation of texture. After conducting performance screening and structure analysis, the study supplements the analysis of Y’s regulation of the titanium alloy structure. The regulation is primarily explained by combining the results of the analysis of boron content, phase diagram analysis, mechanical properties, and fracture analysis. The mechanical analysis introduces the unique load transfer strengthening of TiB whiskers combined with an analysis of high-temperature mechanical properties, as the threshold for addition. The optimal amounts of Y and B additions are 0.6 wt% and 0.8 wt%, respectively. The optimized alloy obtained under this condition can achieve a tensile strength of 950 Mpa at 500 °C without any plastic deformation or heat treatment.

This investigation provides a comprehensive analysis of genomic diversity and selection signatures in Zaobei beef cattle, an indigenous breed known for its adaptation to hot and humid climates and superior meat quality. Whole-genome resequencing was conducted on 23 Zaobei cattle, compared with 46 Simmental cattle to highlight genetic distinctions. Population structure analysis confirmed the genetic uniqueness of Zaobei cattle. Using methods such as DASDC v1.01, XPEHH, and θπ ratio, we identified 230, 232, and 221 genes through DASDC, including hard sweeps, soft sweeps, and linkage sweeps, respectively. Coincidentally, 109 genes were identified when using XPEHH and θπ ratio methods. Together, these analyses revealed eight positive selection genes (ARHGAP15, ZNF618, USH2A, PDZRN4, SPATA6, ROR2, KCNIP3, and VWA3B), which are linked to critical traits such as heat stress adaptation, fertility, and meat quality. Moreover, functional enrichment analyses showed pathways related to autophagy, immune response, energy metabolism, and muscle development. The comprehensive genomic insights gained from this study provide valuable knowledge for breeding programs aimed at enhancing the beneficial traits in Zaobei cattle.

Most space shuttle fuel tanks use a center column to hold the Propellant Management Device (PMD). This paper analyzes the gas–liquid interface state in the tanks with a central column during microgravity experiments conducted in the Chinese Space Station. It launches an extended study to investigate the gas–liquid interface state under different gravity conditions. Using the perturbation method and boundary layer theory, we numerically calculated the morphology of the gas–liquid interface under varying gravity conditions based on the Young–Laplace equation. The results were then compared to those obtained from existing commercial software and were found to be consistent. Based on this, the study develops two types of calculation procedures. The first procedure generates the corresponding shape of the liquid surface by inputting the height of the liquid surface endpoints and the gravity level. The second procedure is based on the targeting method and generates the corresponding liquid surface by inputting the volume of the liquid in the storage tank and the gravity level. The procedures were used to analyze the variation of gas–liquid interface properties under different gravity conditions. This study offers theoretical support for liquid management in aerospace engineering fuel tanks.

Agricultural non-point source pollution (ANSP) constraints on the development of the agricultural economy, grasp the interaction between the two, and develop targeted prevention and control policies in accordance with the evolution of different stages of the characteristic is essential. This study examines the economic characteristics of ANSP in Shandong Province to propose targeted countermeasures. Selected the time series data of Shandong Province from 2000 to 2020 to test the Environmental Kuznets Curve (EKC) characteristics between ANSP and agricultural economic growth (AEG), and the Vector Error Correction model (VECM) is used further to explore the dynamic relationship and mechanism. We validate the existence of an Inverted U-shaped EKC in Shandong Province, and the inflection points of pesticides, fertilizers, and agricultural films were reached in 2007 and 2008. The changes in pesticide loss and fertilizer loss have a positive impact on the dynamics of AEG in the short term, but there is an incremental inhibition in the long term. The dynamic effect of the impulse response diagram between the amount of agricultural film residues and AEG all shows a positive impact. The average explanatory contribution of pesticide loss, fertilizer loss, and agricultural film residue to AEG decreases by 12.14%, 7.7%, and 3%, respectively. Therefore, policy suggestions are proposed to realize the coordinated development of ANSP and economic development.

Abstract Efficient photocatalytic reduction of CO2 to high‐calorific‐value CH4, an ideal target product, is a blueprint for C1 industry relevance and carbon neutrality, but it also faces great challenges. Herein, we demonstrate unprecedented hybrid SiC photocatalysts modified by Fe‐based cocatalyst, which are prepared via a facile impregnation‐reduction method, featuring an optimized local electronic structure. It exhibits a superior photocatalytic carbon‐based products yield of 30.0 µmol g−1 h−1 and achieves a record CH4 selectivity of up to 94.3%, which highlights the effectiveness of electron‐rich Fe cocatalyst for boosting photocatalytic performance and selectivity. Specifically, the synergistic effects of directional migration of photogenerated electrons and strong π‐back bonding on low‐valence Fe effectively strengthen the adsorption and activation of reactants and intermediates in the CO2 → CH4 pathway. This study inspires an effective strategy for enhancing the multielectron reduction capacity of semiconductor photocatalysts with low‐cost Fe instead of noble metals as cocatalysts.

Seed size traits, including seed length (SL), seed width (SW), and seed thickness (ST), are crucial appearance parameters that determine soybean seed weight, yield, and ultimate utilization. However, there is still a large gap in the understanding of the genetic mechanism of these traits. Here, 281 soybeans were utilized to analyze the genetic architecture of seed size traits in different years through multiple (single-locus and multi-locus) genome-wide association study (GWAS) models, and candidate genes were predicted by integrating information on gene function and transcriptome sequencing data. As a result, two, seven, and three stable quantitative trait nucleotides (QTNs) controlling SL, SW, and ST were detected in multiple environments using the single-locus GWAS model, and concurrently detected by the results of the multi-locus GWAS models. These stable QTNs are located on 10 linkage disequilibrium blocks, with single genome regions ranging in size from 20 to 440 kb, and can serve as the major loci controlling soybean seed size. Furthermore, by combining gene functional annotation and transcriptome sequencing data of seeds at different developmental stages from two extreme soybean accessions, nine candidate genes, including Glyma.05G038000, Glyma.05G244100, Glyma.05G246900, Glyma.07G070200, Glyma.11G010000, Glyma.11G012400, Glyma.17G165500, Glyma.17G166500, and Glyma.20G012600 within the major loci that may regulate soybean seed size, were mined. Overall, these findings offer valuable insights for molecular improvement breeding as well as gene functional studies to unravel the mechanism of soybean seed size.

This article proposes a lightweight underwater biological target detection network based on the improvement of YOLOXs, addressing the challenges of complex and dynamic underwater environments, limited memory in underwater devices, and constrained computational capabilities. Firstly, in the backbone network, GhostConv and GhostBottleneck are introduced to replace standard convolutions and the Bottleneck1 structure in CSPBottleneck_1, significantly reducing the model’s parameter count and computational load, facilitating the construction of a lightweight network. Next, in the feature fusion network, a Contextual Transformer block replaces the 3 × 3 convolution in CSPBottleneck_2. This enhances self-attention learning by leveraging the rich context between input keys, improving the model’s representational capacity. Finally, the positioning loss function Focal_EIoU Loss is employed to replace IoU Loss, enhancing the model’s robustness and generalization ability, leading to faster and more accurate convergence during training. Our experimental results demonstrate that compared to the YOLOXs model, the proposed YOLOXs-GCE achieves a 1.1% improvement in mAP value, while reducing parameters by 24.47%, the computational load by 26.39%, and the model size by 23.87%. This effectively enhances the detection performance of the model, making it suitable for complex and dynamic underwater environments, as well as underwater devices with limited memory. The model meets the requirements of underwater target detection tasks.

AbstractA promising paradigm for drug administration that has garnered increasing attention in recent years is the direct transfer (DT) of nanoparticles for transcellular drug delivery. DT requires direct cell-cell contact and facilitates unidirectional and bidirectional matter exchange between neighboring cells. Consequently, DT enables fast and deep penetration of drugs into the targeted tissues. This comprehensive review discusses the direct transfer concept, which can be delineated into the following three distinct modalities: membrane contact-direct transfer, gap junction-mediated direct transfer (GJ-DT), and tunneling nanotubes-mediated direct transfer (TNTs-DT). Further, the intercellular structures for each modality of direct transfer and their respective merits and demerits are summarized. The review also discusses the recent progress on the drugs or drug delivery systems that could activate DT.

Atypical lobular endocervical glandular hyperplasia (ALEGH) is considered a precancerous lesion of gastric-type adenocarcinoma (GAS)/minimal deviation adenocarcinoma (MDA) characterized by an insidious onset, atypical symptoms, and often negative human papillomavirus (HPV) screening. Early screening for this disease is challenging, leading to a high rate of missed clinical diagnoses and the development of malignant tumors at the onset. Increased vaginal discharge and the presence of imaging cystic masses at the internal cervical ostium are often observed in patients with ALEGH. Therefore, we reviewed the clinical data of two cases of ALEGH that were identified and diagnosed in the early stages at our hospital. Through a comprehensive analysis of the medical history and diagnosis plan, combined with a review of relevant literature, to improve the early recognition and diagnosis of ALEGH, as well as strengthen the management of cervical precancerous lesions.

ObjectiveThe aim of this study is to investigate the value of ultrasound combined with computed tomography (CT) in identifying early low-grade appendiceal mucinous neoplasm and appendicitis.MethodsPatients with early low-grade appendiceal mucinous neoplasm and appendicitis from September 2017 to September 2021, including 40 patients with low-grade appendiceal mucinous neoplasm and 40 patients with appendicitis, were collected in this study. Clinical data as well as ultrasound and CT findings of all patients were retrospectively analyzed. Univariate and multivariate logistic regression analyses were applied to establish the ultrasound model, the CT model, and the combined model.ResultsThe nomogram showed that specific characteristics of CT were dilated appendiceal diameter and clear surrounding fat space in the low-grade appendiceal mucinous neoplasm and that specific characteristics of ultrasound were thin or clear layer appendix wall and flocculent echo in the appendix cavity. These four features were used to construct a nomogram for predicting early low-grade appendiceal mucinous neoplasm, and the area under the curve value was 0.839.ConclusionUltrasound combined with CT for diagnosis of early low-grade appendiceal mucinous neoplasm has a significant value; when found significantly dilated appendix in the lower right abdomen, with thin wall, wall calcification, clear surrounding fat space, and progressive enhancement, especially non-specific symptoms similar to appendicitis, the physician should timely consider the possibility of low-grade appendiceal mucinous neoplasm.

Objective: To investigate the clinical value of laparoscopic cytoreductive surgery (CRS) in treating of appendiceal pseudomyxoma peritonei with limited disease and low tumor burden. Methods: The clinical data of patients with appendiceal pseudomyxoma peritonei treated by surgery with CRS at the Aerospace Center Hospital from January 2018 to December 2021 were retrospectively analyzed. The patients were divided into laparoscopic or open CRS groups according to the operation method. A propensity score-matched (PSM) analysis (1:1) was performed, the related clinical variables were compared between the two groups, and the effect on progression-free survival (PFS) was also analyzed. We theoretically investigate the tunable dual- and multi-channel filter in one-dimensional aperiodic photonic crystals (PCs) embedded with graphene. The dielectrics slabs arrayed alternately submit to the Cantor sequence rule and graphene sheets are local at the interfaces of dielectrics layers. Cantor PCs are fractal structures and support a series of discrete resonant channels in transmission spectra, viz. fractal states. Consequently, dual- and multi-filtering channels could be achieved in the compound systems. The central wavelength of each channel and its value of transmission could be modulating by the chemical potential of grapheme and the incident angle as well. It shows that the dual-channel amplitude of transmission can be varied by over 100% and the multiple channels could be switched on and off by the chemical potential of graphene. This study will be very helpful in designing tunable optical filters.

Abstract Background The efficacy of vitamin C in sepsis remains controversial. Whether vitamin C can alleviate lipopolysaccharide (LPS)-induced myocardial injury by inhibiting pyroptosis has not been studied. This study aimed to evaluate the effects of vitamin C on LPS-induced myocardial injury in vitro. Methods H9C2 cells were treated with indicated concentrations of LPS, and the cell viability was then assessed by CCK-8 assay. The levels of lactate dehydrogenase (LDH), CK-MB, IL-18 and IL-1β were examined by enzyme-linked immunosorbent assay (ELISA). The levels of intracellular reactive oxygen species (ROS) were measured using the fluorescent probe dichlorodihydrofluorescein diacetate (DCFH-DA). Western blot assays were conducted to determine the levels of the ROS-associated protein nicotinamide adenine dinucleotide phosphate oxidase 4 (Nox4) and pyroptosis-associated proteins, such as NOD-like receptor (NLR) family pyrin domain containing 3 (NLRP3), caspase-1 and gasdermin D (GSDMD). The AKT inhibitor MK-2206 was then applied to explore the signalling pathway. Finally, H9C2 cells were divided into the control group, LPS group, vitamin C + LPS group, and N-acetyl-l-cysteine (NAC) + LPS group. The intracellular ROS, levels of associated proteins, cell viability, and release of LDH, CK-MB, IL-18 and IL-1β were examined. Results LPS decreased cell viability and induced ROS and pyroptosis in H9C2 cells in a dose-dependent manner. Moreover, LPS activated the AKT/mTOR pathway in H9C2 cells. The AKT inhibitor MK-2206 protected H9C2 cells from LPS-induced death by suppressing pyroptosis, without changing intracellular ROS level. Vitamin C significantly inhibited intracellular ROS and cell pyroptosis in LPS-treated H9C2 cells. Moreover, vitamin C suppressed the activation of the AKT/mTOR pathway. Conclusions Our data suggest that vitamin C alleviates LPS-induced myocardial injury by inhibiting pyroptosis via the ROS-AKT/mTOR signalling pathway and thus provide novel insights into the prevention of sepsis-induced myocardial dysfunction.

Abstract Aiming at evaluating the lifetime of the neutron, we introduce a novel statistical method to analyse the updated compilation of precise measurements including the 2022 dataset of Particle Data Group (PDG). Based on the minimization for the information loss principle, unlike the median statistics method, we apply the most frequent value (MFV) procedure to estimate the neutron lifetime, irrespective of the Gaussian or non-Gaussian distributions. Providing a more robust way, the calculated result of the MFV is $$\tau _n=881.16^{+2.25}_{-2.35}$$ τ n = 881 . 16 - 2.35 + 2.25 s with statistical bootstrap errors, while the result of median statistics is $$\tau _n=881.5^{+5.5}_{-3}$$ τ n = 881 . 5 - 3 + 5.5 s according to the binomial distribution. Using the different central estimates, we also construct the error distributions of neutron lifetime measurements and find the non-Gaussianity, which is still meaningful.

Cancer has long been a hot research topic, and recent years have witnessed the incidence of cancer trending toward younger individuals with great socioeconomic burden. Even with surgery, therapeutic agents serve as the mainstay to combat cancer in the clinic. Intensive research on nanomaterials can overcome the shortcomings of conventional drug delivery approaches, such as the lack of selectivity for targeted regions, poor stability against degradation, and uncontrolled drug release behavior. Over the years, different types of drug carriers have been developed for cancer therapy. One of these is liposome-in-gel (LP–Gel), which has combined the merits of both liposomes and hydrogels, and has emerged as a versatile carrier for cancer therapy. LP–Gel hybrids have addressed the lack of stability of conventional liposomes against pH and ionic strength while displaying higher efficiency of delivery hydrophilic drugs as compared to conventional gels. They can be classified into three types according to their assembled structure, are characterized by their nontoxicity, biodegradability, and flexibility for clinical use, and can be mainly categorized based on their controlled release, transmucosal delivery, and transdermal delivery properties for anticancer therapy. This review covers the recent progress on the applications of LP–Gel hybrids for anticancer therapy.

Abstract Background The COVID-19 pandemic has become a huge threat to human health, infecting millions of people worldwide and causing enormous economic losses. Many novel small molecule drugs have been developed to treat patients with COVID-19, including Paxlovid, which block the synthesis of virus-related proteins and replication of viral RNA, respectively. Despite satisfactory clinical trial results, attention is now being paid to the long-term side effects of these antiviral drugs on the musculoskeletal system. To date, no study has reported the possible side effects, such as osteoarthritis, of Paxlovid. This study explored the effects of antiviral drug, Paxlovid, on chondrocyte proliferation and differentiation. Methods In this study, both in vitro and in vivo studies were performed to determine the effect of Paxlovid on chondrocyte degeneration and senescence. Furthermore, we explored the possible mechanism behind Paxlovid-induced acceleration of cartilage degeneration using transcriptome sequencing and related inhibitors were adopted to verify the downstream pathways behind such phenomenon. Results Paxlovid significantly inhibited chondrocyte extracellular matrix protein secretion. Additionally, Paxlovid significantly induced endoplasmic reticulum stress, oxidative stress, and downstream ferroptosis, thus accelerating the senescence and degeneration of chondrocytes. In vivo experiments showed that intraperitoneal injection of Paxlovid for 1 week exacerbated cartilage abrasion and accelerated the development of osteoarthritis in a mouse model. Conclusions Paxlovid accelerated cartilage degeneration and osteoarthritis development, potentially by inducing endoplasmic reticulum stress and oxidative stress. Long-term follow-up is needed with special attention to the occurrence and development of osteoarthritis in patients treated with Paxlovid.

For the first time, the ultrasonic surface rolling processing (USRP) technology was applied to a real bearing raceway, and a finite element simulation model for USRP on a bearing raceway was established. Practical tests were conducted to evaluate the surface integrity of the bearing raceway before and after USRP treatment. The USRP parameters were optimized using Taguchi-gray correlation analysis. The results show that the optimal process parameters for the 7224 bearing raceway are: an amplitude of 10 μm, a static pressure of 2400 N, a spindle speed of 45 rad/min, and rolling times of 7. In the process of USRP, residual compressive stress results from inhomogeneous elastic and plastic deformation. Adjacent tempered martensitic laths merge and deflect at a certain angle along the rolling direction to form directional grain stacks, resulting in a flocculent plastic deformation layer. The USRP based on the finishing mechanism of “peak-cutting and valley-filling” can improve the surface micro-morphology, reduce roughness value Ra to approximately 0.16 μm, and effectively eliminate microscopic cracks caused by grinding. The micro-hardness of the bearing raceway surface increased from 760 HV to 840 HV under the dual action of surface grain refinement and work hardening effect.

The production of cement, which is the key ingredient of concrete, leads to environmental pollution by releasing massive amounts of CO _2 and using significant natural resources. Therefore, shifting towards sustainable and greener materials is essential for mitigating these challenges. In this study, recycled concrete powder (RCP) was used as a cement replacement (0%, 5.0%, 10%, and 15%), solving the waste dumps issue and promoting sustainability. Furthermore, the concrete is also reinforced with steel fibers which were obtained from waste rubber tires to improve concrete tensile strength. The concrete properties were evaluated through slump cone test, compressive strength, failure patterns, tensile strength, scanning electronic microscopy, and FTIR analysis. The results indicate that the concrete strength properties improved with the substitution of RCP. The compressive and tensile strength of the optimum mix (10% RCP and 2.0% addition of steel fibers) are 15.8% and 23% more than those of reference concrete. However, the concrete flow is adversely impacted due to RCP angular particle shapes. Failure patterns indicate that RCP and steel fibers improved concrete ductility. SEM and FTIR analysis indicate microstructural improvement with RCP and steel fibers. Finally, the analysis concluded that the developed concrete showed better performance, solved waste dumps issues, and promoted sustainability.

However, the association between inflammation-related indexes and sex hormones in the general population remained uncertain.MethodsWe incorporated data from the NHANES 2013-2016 survey of American adults. On the basis of distribution and comparison analysis, we chose to undertake separate analyses of men and women (including premenopausal and postmenopausal groups). Multivariable weighted linear regression models, eXtreme Gradient Boosting (XGBoost) models, generalized linear analysis, stratified models, logistic regression models and sensitivity analysis were utilized to assess the relationships between inflammation-related indexes and sex hormones.ResultsTotal 9372 participants out of 20146 were fitted into our research. We conducted separate gender analysis due to different distribution. Multivariable weighted linear regression indicated every component of the inflammation-related index was negatively correlated with at least one component of the male hormone indexes. However, SII, NLR, PPN, and NC were associated positively with female estradiol. XGBoost identify SII, PLR and NLR were the critical indexes on sex hormones. Inflammation-related indexes was associated with Testosterone deficiency in male and postmenstrual group and associated with Excessive Estradiol in premenstrual group. Finally, the subgroup analysis revealed that the association between sex hormones and inflammatory indicators was prominent in American adults over the age of 60 or those with BMI (>28 kg/m2).ConclusionIn all, inflammation-related indexes act as independent risks associated with sex hormone alterations and metabolic disorder in both genders. Using multiple models, we revealed the relative importance of inflammation-related indexes. Subgroup analysis also identified the high-risk population. More prospective and experimental research should be conducted to validate the results.

Abstract Chemoresistance is the main obstacle in osteosarcoma (OS) treatment; however, the underlying mechanism remains unclear. In this study, it is discovered that DDRGK domain‐containing protein 1 (DDRGK1) plays a fundamental role in chemoresistance induced in OS. Bioinformatic and tissue analyses indicate that higher expression of DDRGK1 correlates with advanced tumor stage and poor clinical prognosis of OS. Quantitative proteomic analyses suggest that DDRGK1 plays a critical role in mitochondrial oxidative phosphorylation. DDRGK1 knockout trigger the accumulation of reactive oxygen species (ROS) and attenuate the stability of nuclear factor erythroid‐2‐related factor 2 (NRF2), a major antioxidant response element. Furthermore, DDRGK1 inhibits ubiquitin‐proteasome‐mediated degradation of NRF2 via competitive binding to the Kelch‐like ECH‐associated protein 1 (KEAP1) protein, which recruits NRF2 to CULLIN(CUL3). DDRGK1 knockout attenuates NRF2 stability, contributing to ROS accumulation, which promotes apoptosis and enhanced chemosensitivity to doxorubicin (DOX) and etoposide in cancer cells. Indeed, DDRGK1 knockout significantly enhances osteosarcoma chemosensitivity to DOX in vivo. The combination of DDRGK1 knockdown and DOX treatment provides a promising new avenue for the effective treatment of OS.

Abstract Intervertebral disc degeneration (IVDD)‐induced lower back pain (LBP) is a common problem worldwide. The underlying mechanism is partially accredited to ferroptosis, based on sequencing analyses of IVDD patients from the gene expression omnibus (GEO) databases. In this study, it is shown that polydopamine nanoparticles (PDA NPs) inhibit oxidative stress‐induced ferroptosis in nucleus pulposus (NP) cells in vitro. PDA NPs scavenge reactive oxygen species (ROS), chelate Fe2+ to mitigate iron overload, and regulate the expression of iron storage proteins such as ferritin heavy chain (FHC), ferritin, and transferrin receptor (TFR). More importantly, PDA NPs co‐localize with glutathione peroxidase 4 (GPX4) around the mitochondria and suppress ubiquitin‐mediated degradation, which in turn exerts a protective function via the transformation and clearance of phospholipid hydroperoxides. PDA NPs further down‐regulate malondialdehyde (MDA) and lipid peroxide (LPO) production; thus, antagonizing ferroptosis in NP cells. Moreover, PDA NPs effectively rescue puncture‐induced degeneration in vivo by targeting ferroptosis and inhibiting GPX4 ubiquitination, resulting in the upregulation of antioxidant pathways. The findings offer a new tool to explore the underlying mechanisms and a novel treatment strategy for IVDD‐induced LBP.

ObjectiveTo systematically evaluate the clinical efficacy and safety of Shenkang injection (SKI) combined with alprostadil in the treatment of chronic renal failure (CRF).MethodRandomized controlled trials (RCTs) of Shenkang injection combined with alprostadil in CRF treatment were investigated by retrieving a total of 7 databases including CNKI, Wanfang database, VIP, CBM, PubMed, Embase and Cochrane Library, with the search time ranging from 2012 to now. Revman 5.2 software was used for data analysis, and Cochrane bias risk tool was used to evaluate the quality of the included literature. The final results were represented by relative risk (RR), mean difference (MD) and 95% confidence interval (95% CI).ResultsA total of 20 RCTs and 1,573 patients were included in this study. Meta-analysis showed that the overall response rate (ORR) of the treatment group was superior to the control group [RR = 0.20, 95% CI (0.16, 0.25), P < 0.00001]. Compared with the control group, the treatment group achieved favorable improvement in terms of the creatinine clearance rate (Ccr) [MD = 9.48, 95% CI (8.73, 10.24), P < 0.00001], serum creatinine (Scr) [MD = −55.12, 95% CI (−63.42, −46.82), P < 0.00001], quantitative urine protein (Upro) [MD = −0.48, 95% CI (−0.53, −0.43), P < 0.00001], and blood urea nitrogen (BUN) [MD=-3.73, 95% CI (−4.08, −3.3) 7, P < 0.00001]. There was no statistical difference in the incidence of adverse reactions in each group.ConclusionCurrently, Shenkang injection combined with alprostadil has been widely used in clinical treatment of CRF due to the certain effect superior to other methods. However, its specific efficacy and safety need to be further verified through numerous large-scale clinical trials.

Abstract Background Prostatic cancer (PCa) is one of the most common malignant tumors in men worldwide. Emerging evidence indicates significance of hypoxia and immunity in PCa invasion and metastasis. This study aimed to develop a hypoxia- and immune-related gene risk signature and explore the molecular mechanisms to formulate a better prognostic tool for PCa patients. Methods The hypoxia and immune scores of all PCa patients in The Cancer Genome Atlas (TCGA) dataset were calculated via the maximally selected rank statistics method and the ESTIMATE algorithm. From common genes identified overlapping hypoxia- and immune-related differentially expressed genes (DE-HRGs and DE-IRGs), a hypoxia- and immune-related gene risk signature was developed utilizing univariate and multivariate Cox regression analyses, and validated in the Memorial Sloan Kettering Cancer Centre (MSKCC) database. The immune cell infiltration level of PCa samples were evaluated with ssGSEA algorithm. Differential expression of prognostic genes was evidenced by immunohistochemistry and western blot (WB) in paired PCa samples. Expression levels of these genes and their variations under regular and hypoxic conditions were examined in cell lines. The functional effects of the prognostic gene on PCa cells were examined by wound healing and transwell assays. Results A hypoxia- and immune-related gene risk signature constructed by ISG15 and ZFP36 displays significant predictive potency, with higher risk score representing worse survival. A nomogram based on independent prognostic factors including the risk score and Gleason score exhibited excellent clinical value in the survival prediction of PCa. Infiltration levels of eosinophils, neutrophils, Tcm, Tem, TFH, Th1 cells, and Th17 cells were significantly lower in the high-risk group. Conversely, aDC, pDC, T helper cells, and Tregs were significantly higher. Additionally, the two prognostic genes were closely correlated with the tumor-infiltrating immune cell subset in PCa progression. RT-qPCR and WB presented higher and lower expression of ISG15 and ZFP36 in PCa cells, respectively. They were correspondingly increased and decreased in PCa cells under hypoxic conditions. Wound healing and transwell assays showed that over-expression of ISG15 promoted the migration and invasion of PCa cells. Conclusion Our study identified a novel hypoxia- and immune-related gene signature, contributing a new perspective to the treatment of PCa

Classical Swine Fever (CSF) is a highly infectious and fatal disease caused by Classical Swine Fever Virus (CSFV). It is recognized as one of the most serious viral diseases in pig industry. Since CSF was first reported in 1810, CSF has caused significant economic losses to the pig industry in the world and continues to threaten global pork production and human food security. CSF was listed as one of the most important notifiable infectious diseases by the World Organization for Animal Health (OIE), and it was listed as "Class I infectious diseases" in China. At present, CSF is clinically manifested complicated and changeably as acute form with high mortality or subacute type, chronic type, recessive type and continuous infection type with variable mortality, and often infected combined with various diseases. Vaccination is still one of the main means to prevent and control CSF in most countries, especially in developing countries. The development and application of various laboratory diagnostic methods and clinical detection techniques for CSFV antigens and antibodies play a very important role in the prevention and control of CSF. With the rapid development of biotechnology, more diagnostic techniques and new-type vaccines for CSFV have been developed and approved. CSFV antigen and antibody diagnostic techniques, and the development and prospects of live attenuated and new-type CSFV vaccines such as subunit and vector vaccines are reviewed in order to provide references for better prevention and control of CSF.
