The poorly studied leatherleaf slug genus Valiguna in Thailand was carefully investigated. Members of this genus are phenotypically similar, making their identification very challenging. This study clarifies the taxonomic status of all Valiguna species in Thailand by combining morphological and anatomical studies with DNA barcoding. Monophyly of all Valiguna species was confirmed by analysis of the mitochondrial COI data and that all Valiguna species have the acropleurocaulis type of penis. Currently, three Valiguna species are recognised: V. siamensis, V. semicerina Mitchueachart & Panha, sp. nov., and V. crispa Mitchueachart & Panha, sp. nov. that are new to science. For distinct characteristics, V. siamensis is characterised by having a cylindrical penis and honeycomb-like glans, V. semicerina sp. nov. has a lanceolate penis with half honeycomb-like glans, and V. crispa sp. nov. has a cylindrical penis with wavy-like glans. In addition, more detailed descriptions of the radula and genitalia of all three species and their distribution are also carefully presented, enhancing the understanding of this leatherleaf slug genus in Thailand. Terrestrial gastropods have a lens‐bearing eye on the tip of their tentacles. There are two morphologically distinct photoreceptors, called Type‐I and Type‐II photoreceptors, in the retina. Type‐I photoreceptors are equipped with highly developed photoreceptive microvilli in their outer rhabdomeric segment, whereas Type‐II photoreceptors have short and fewer microvilli. Although both types of photoreceptors send afferent projections directly to the brain, their destinations in the brain, called optic neuropiles, have not been sufficiently investigated. Our recent studies revealed that there are commissural fibers in the cerebral ganglia that transmit photic information acquired by bilateral eyes. Moreover, some of the retinal photoreceptors are connected by gap junctions to the photosensitive brain neurons, suggesting the functional interaction of the photic information between the eye and brain photoreceptors, as well as between bilateral eyes. However, it has not been clarified which type of retinal photoreceptors send commissural projections to the contralateral hemiganglion nor interact with the brain photoreceptors. In the present study, we demonstrated by molecular histological analyses and tracer injections that (1) Type‐I and Type‐II photoreceptors send glutamatergic afferent projections to the medial and lateral lobes of the ipsilateral optic neuropile, respectively, (2) direct synaptic interaction between bilateral optic nerves occurs in the medial lobe of the optic neuropile, and (3) brain photosensory neurons form gap junctions with the medial lobe of the contralateral optic neuropile. Despite being simultaneously male and female, hermaphrodites may still need to assume the male or female sexual role in a mating encounter, with the option to swap roles afterwards. For the great pond snail, Lymnaea stagnalis, deciding which sexual role to perform has important consequences, since sperm transfer and male reproductive success can be decreased. We hypothesised that detecting cues that indicate a possible mating encounter could help them to adapt their mating behaviour. Therefore, we experimentally assessed whether signalling the presence of a conspecific with an odour can affect the sexual role of Lymnaea stagnalis. The results showed that learning resulted in either an increased ability to mate as a male or in faster mating compared to the control group. These findings reveal that learning shapes the mating dynamics of Lymnaea stagnalis, thus showing that cognitive processes not only affect mating in separate-sexed species but also in hermaphrodites. Ullung Island is an oceanic island in the Sea of Japan about 130 km distant from the Korean peninsula. The biota of this oceanic island is thought to be derived from the closest continental region while the importance of long-distance dispersal from other areas for the formation of the Ullung Island biota was considered negligible. In this study, we have examined the origin of the Ullung Island-endemic land snail Karaftohelix adamsi (Kuroda & Hukuda, 1944) using a molecular phylogenetic approach with two mitochondrial (COI and 16S rRNA) and nuclear (ITS regions) DNA sequences. Our results revealed that K. adamsi has originated from the northernmost region of the Japanese archipelago or Sakhalin Island, most likely from Rebun Island of Hokkaido, by long-range dispersal across a distance of 1200 km. This finding reveals that the biota of the oceanic Ullung Island has not exclusively originated from the Asian mainland, but that some elements have originated from the more distant archipelago of Japan and the Far East of Russia. Abstract Background Spiral cleavage is a feature of non-ecdysozoan protostomes, in which left-right reversal frequently evolved in gastropod molluscs. In pulmonate gastropods, maternal molecules are responsible for chirality patterning, on which the polarities of visceral and coiling asymmetries depend. In the pond snail, Lymnaea stagnalis (the clade Hygrophila), a frame-shift mutation of one of tandem-duplicated, diaphanous-related formin genes (diaph) resulted in incomplete reversal from dextral to sinistral cleavage. Is this mechanism of chirality regulation common to, or shared with other pulmonates? To answer this question, we examined genes involved in chirality patterning in the land snail, Bradybaena similaris which belongs to the clade Stylommatophora. Results Both dextral and sinistral siblings develop from progeny of a racemic mutant of B. similaris. Differences in maternal mRNAs between the two strains were searched by transcriptome analyses. We found fifty maternal transcripts that exhibited less expression in early embryos of the mutant strain. The most conspicuous was a homolog of diaph. The diaph gene was duplicated in the stylommatophoran ancestor (diaph-a and diaph-b), as in the case of the ancestor of Lymnaea (Lsdiaph1 and Lsdiaph2). The quantity of maternal diaph-b mRNA was drastically reduced in early embryos of the racemic mutant compared to wild-type, while diaph-a expression was at nearly the same level in both strains. Unlike the case of Lsdiaph2, which is frame-shifted to produce truncated peptides in the mutant of L. stagnalis, however, Bsdiaph-b mRNA in the mutant strain is not frame-shifted and most probably produces normal Diaph-b protein. These results suggest the presence of regulatory mechanisms of gene expression for chirality patterning in pulmonate gastropods, although genomic analyses are required for confirmation. Conclusions Heterochirality resulting from the loss of polarity control in spiral cleavage does not require mutation of the diaph gene in B. similaris. The determination of left-right polarity instead depends on the expression of this diaph gene, which is duplicated in stylommatophoran Bradybaena, as well as in hygrophilan Lymnaea. Our results provide an avenue to identifying a regulatory mechanism that controls the direction of spiral cleavage in gastropods.

Optimizing reproductive success is an essential part of evolution for both sexes. Females can optimize mating by avoiding superfluous mating advances and insemination, since both take time away from other activities and may incur costs related to sperm receipt. While many separate-sexed organisms are known to exhibit mate avoidance, much less is known about this for simultaneous hermaphrodites. We addressed this here, using a simultaneously hermaphroditic species that can choose to mate in either of the two sex roles during each mating interaction. Recently, avoidance behaviours in response to natural matings were observed in the hermaphroditic pond snail Lymnaea stagnalis , potentially deterring insemination. To disentangle whether such behaviours are mediated by the mechanical act of mating or the receipt of accessory gland proteins and/or sperm, we intravaginally injected individuals with control or test fluids. Our results show that the avoidance behaviours, crawl-out and biting, were more frequently expressed when individuals were inseminated with accessory gland proteins and/or sperm. These behavioural components of the recipient increased time in courtship prior to insemination, which is concordant with the hypothesis that the partner tries to discourage the potential sperm donor from inseminating. Understanding the mechanism underlying the effects that molluscan accessory gland proteins induce contributes to our understanding of the molecular basis of the recipient's (behavioural) responses as well as how such biochemical postcopulatory strategies evolve. • Reproduction can be optimized by avoiding surplus mating advances and inseminations. • Being inseminated takes time and can incur costs related to sperm receipt. • Simultaneous hermaphrodites cope with this while serving both reproductive interests. • Accessory gland proteins and/or sperm are found to induce crawl-out and biting. • Earlier reported avoidance behaviours seem to be used to discourage new sperm donors. Although the eye is the best-studied photoreceptive organ in animals, the presence of non-ocular photosensing systems has been reported in numerous animal species. However, most of the roles that nonocular photosensory systems play remain elusive. We found that the terrestrial slug Limax valentianus avoids light and escapes into dark areas even if it is blinded by the removal of the bilateral superior tentacle. The escape behaviourwasmore evident for short-wavelength light. Illumination to the head with blue but not red light elicited avoidance behaviour in the blinded slugs. Illumination to the tail was ineffective. The light-avoidance behaviour of the blinded slugs was not affected by the removal of the penis, which lies on the brain in the head, suggesting that the penis is dispensable for sensing light in the blinded slug. mRNA of Opn5A, xenopsin, retinochrome and, to a lesser extent, rhodopsin was expressed in the brain according to RT-PCR. Lightevoked neural responses were recorded from the left cerebro-pleural connective of the isolated suboesophageal ganglia of the brain, revealing that the brain is sensitive to short wavelengths of light (400-480 nm). This result is largely consistent with the wavelength dependency of the light-avoidance behaviour of the blinded slugs that we observed in the present study. This morphological description of Onchidium reevesii (J.E. Gray, 1850), a common species on the coast of China, previously called Onchidium struma (Qiu, 1991), includes the external features and the digestive, nervous and reproductive systems. The radula and the penis are illustrated using electron microscopy. The surface of the notum is granular and hundreds of dorsal papillae are present, each with an eye. The position of the pneumostome is mid posterior on the hyponotum. The radular formula varies from 62:1:62 to 110:1:110. A convolute tubular rectal gland is present. Unusual features of the reproductive system include the penis having a proximal long, spiral stalk followed by a distal region covered with hooks, and the flagellum of the penial gland being long and heavily coiled. Two additional new members of the onchidione family, 16- epi -onchidione ( 1 ) and 4- epi -onchidione ( 2 ), co-occurring with six previously reported bis-γ-pyrone polypropionates including onchidione ( 3 ), were isolated from the marine pulmonate Onchidium sp. Their structures were determined by extensive spectroscopic analysis and by comparison with 3 and onchidione-related derivatives. The absolute configuration of 1 was established by X-ray diffraction analysis employing graphite monochromated Cu K α radiation (λ = 0.71073 Å) with small Flack parameter 0.08. In addition, the absolute stereochemistry of previously reported onchidionol ( 6 ) was confirmed by the X-ray diffraction analysis. Abstract: Aim: We assess biogeographical patterns, population structure and the range of species in the pulmonate genus Siphonaria across the sub‐Antarctic. We hypothesized that locally endemic cryptic species will be found across the distribution of these direct‐developing limpets in the sub‐Antarctic. Location: The sub‐Antarctic coasts of the Southern Ocean including South America, the Falkland/Malvinas, South Georgia, Kerguelen and Macquarie Islands. Methods: Multi‐locus phylogenetic reconstructions, mtDNA time‐calibrated divergence time estimations and population‐based analyses of Siphonaria populations were used at the scale of the Southern Ocean. Results: We resolve two widely distributed lineages of Siphonaria (S. lateralis and S. fuegiensis) across the sub‐Antarctic. MtDNA divergence time estimates suggest that they were separated around 4.0 Ma (3.0 to 8.0 Ma). Subsequently both species followed different evolutionary pathways across their distributions. Low levels of genetic diversity characterize the populations of both species, reflecting the role of Quaternary glacial cycles during their respective demographic histories, suggesting high levels of dispersal among geographically distant localities. Main conclusions: Siphonaria lateralis and S. fuegiensis constitute sister and broadly co‐distributed species across the sub‐Antarctic. Unexpected transoceanic similarities and low levels of genetic diversity in both these direct‐developing species imply recurrent recolonization processes through long‐distance dispersal to isolated sub‐Antarctic islands. For such groups of Southern Ocean invertebrates, rafting may be more effective for long‐distance dispersal than a free‐living planktotrophic larval stage. This biogeographical model may explain why many marine species lacking a dispersal phase exhibit broad distributions, low genetic diversity and low population structure over thousands of kilometres. Sexual selection is generally predicted to act more strongly on males than on females. The Darwin-Bateman paradigm predicts that this should also hold for hermaphrodites. However, measuring this strength of selection is less straightforward when both sexual functions are performed throughout the organism’s lifetime. Besides, quantifications of sexual selection are usually done during a short time window, while many animals store sperm and are long-lived. To explore whether the chosen time frame affects estimated measures of sexual selection, we recorded mating success and reproductive success over time, using a simultaneous hermaphrodite. Our results show that male sexual selection gradients are consistently positive. However, an individual’s female mating success seems to negatively affect its own male reproductive success, an effect that only becomes visible several weeks into the experiment, highlighting that the time frame is crucial for the quantification and interpretation of sexual selection measures, an insight that applies to any iteroparous mating system.

Background: Mitochondrial (mt) genome sequences are widely used for species identification and to study the phylogenetic relationships among Gastropoda. However, to date, limited data are available as taxon sampling is narrow. In this study we sequenced the complete mt genomes of the freshwater gastropods Radix swinhoei (Lymnaeidae) and Planorbarius corneus (Planorbidae). Based on these sequences, we investigated the gene rearrangement in these two species and the relationships with respect to the ancestral gene order and assessed their phylogenetic relationships. Methods: The complete mt genomes of R. swinhoei and P. corneus were sequenced using Illumina-based paired-end sequencing and annotated by comparing the sequence information with that of related gastropod species. Putative models of mitochondrial gene rearrangements were predicted for both R. swinhoei and P. corneus, using Reishia clavigera mtDNA structure as the ancestral gene order. The phylogenetic relationships were inferred using thirteen protein sequences based on Maximum likelihood and Bayesian inference analyses. Results: The complete circular mt genome sequences of R. swinhoei and P. corneus were 14,241 bp and 13,687 bp in length, respectively. Comparison of the gene order demonstrated complex rearrangement events in Gastropoda, both for tRNA genes and protein-coding genes. The phylogenetic analyses showed that the family Lymnaeidae was more closely related to the family Planorbidae, consistent with previous classification. Nevertheless, due to the position recovered for R. swinhoei, the family Lymnaeidae was not monophyletic. Conclusion: This study provides the complete mt genomes of two freshwater snails, which will aid the development of useful molecular markers for epidemiological, ecological and phylogenetic studies. We describe a new stylommatophoran land snail of the family Partulidae from Palau. The new species has a combination of morphological and ecological characters that do not allow its placement in any existing partulid genus, so we describe a new genus for it. The new genus is characterized by a large (18-23 mm) obese-pupoid shell; smooth protoconch; teleoconch with weak and inconsistent, progressively stronger, striae; last half of body whorl not extending beyond the penultimate whorl; widely expanded and reflexed peristome; relatively long penis, with longitudinal pilasters that fuse apically into a fleshy ridge that divides the main chamber from a small apical chamber; and vas deferens entering and penial-retractor muscle attaching at the apex of the penis. Unlike all other partulids, the new species is strictly associated with rocks in contact with the ground. Comparing the other three Palauan species - currently assigned to Partula - to our new genus and to other partulids makes it clear that they require their own genus because their morphology is quite different from that of true Partula and from that of all other genera. The effects of multiple stressors on estuarine organisms are not well understood. Using cage experiments we measured the survival and growth of the pulmonate gastropod Amphibola crenata at five locations which differed contaminant levels. Water nutrients came from a nearby sewage treatment works and the sediment contained low levels of trace metals. Over 6 weeks of exposure, sediment surface chlorophyll levels varied amongst locations. The Chl a values were positively correlated with sediment N and P and trace metals As, Cd, Cu, Pb and Zn. Pulmonate survival depended on location, highest mortality was from a site close to the treatment plant and mortality rate of large individuals decreased significantly with distance away from it. For four locations, medium A. crenata had higher survival than small (juveniles) or adults. Growth rates of small individuals exceeded those for medium and large A. crenata . The mean length increment/week for medium gastropods ranged between 0.49 and 1.11 mm and was negatively correlated with the amount of Chl a in the surface sediment, suggesting the negative effects of eutrophication on gastropod growth. Growth rate of the pulmonate was not correlated with nutrient concentration or trace metal concentrations in the sediment. The dry weight condition index (CI) did not correlate with the growth rate, and for medium individuals, was unaffected by any of the environmental variables. The CI of small individuals was negatively affected by increasing water nutrient levels and the CI of large individuals negatively affected by increasing sediment nutrients and trace metal concentrations. The results from this study suggest that gastropod growth and survival could be used as tools to monitor the effects of changing nutrient levels and recovery from eutrophication within temperate estuaries. Abstract: Reproductive success is a critical fitness attribute that is directly influenced by resource availability. Here, we investigate the effects of diet‐based resource availability on three interrelated aspects of reproductive success: a change in mating system based on mate availability, consequent inbreeding depression, and the deterioration of reproductive efficiency with age (senescence). We employed a factorial experimental design using 22 full‐sib families of the hermaphroditic freshwater snail Physa acuta to explore these interactions. Individual snails were reared in one of two mate‐availability treatments (isolated [selfing] or occasionally paired [outcrossing]) and one of two diet treatments (boiled lettuce or Spirulina, an algae that is rich in protein, vitamins, and minerals). Spirulina‐fed snails initiated reproduction at a 13% earlier age and 7% larger size than lettuce‐fed snails. Spirulina also resulted in a 30% reduction in the time delay before selfing. Compared to lettuce, a diet of Spirulina increased inbreeding depression by 52% for egg hatching rate and 64% for posthatching juvenile survival. Furthermore, Spirulina led to a 15‐fold increase in the rate of reproductive senescence compared with a diet of lettuce. These transgenerational, interactive effects of diet on inbreeding depression and reproductive senescence are discussed in the context of diet‐induced phenotypic plasticity. [ABSTRACT FROM AUTHOR]

Background: With a diversity of pigmented shell morphotypes governed by Mendelian patterns of inheritance, the common grove snail, Cepaea nemoralis, has served as a model for evolutionary biologists and population geneticists for decades. Surprisingly, the molecular mechanisms by which C. nemoralis generates this pigmented shelled diversity, and the degree of evolutionary conservation present between molluscan shell-forming proteomes, remain unknown. Results: Here, using next generation sequencing and high throughput proteomics, we identify and characterize the major proteinaceous components of the C. nemoralis shell, the first shell-proteome for a pulmonate mollusc. The recent availability of several marine molluscan shell-proteomes, and the dataset we report here, allow us to identify 59 evolutionarily conserved and novel shell-forming proteins. While the C. nemoralis dataset is dominated by proteins that share little to no similarity with proteins in public databases, almost half of it shares similarity with proteins present in other molluscan shells. In addition, we could not find any indication that a protein (or class of proteins) is directly associated with shell pigmentation in C. nemoralis. This is in contrast to the only other partially characterized molluscan-shell pigmentation mechanism employed by the tropical abalone Haliotis asinina. Conclusions: The unique pulmonate shell-forming proteome that we report here reveals an abundance of both mollusc-specific and pulmonate-specific proteins, suggesting that novel coding sequences, and/or the extensive divergence of these sequences from ancestral sequences, supported the innovation of new shell types within the Conchifera. In addition, we report here the first evidence that molluscs use independently evolved mechanisms to pigment their shells. Background: Gastropods are among the most diverse animal clades, and have successfully colonized special habitats such as the marine sand interstitial. Specialized meiofaunal snails and slugs are tiny and worm-shaped. They combine regressive features - argued to be due to progenetic tendencies - with convergent adaptations. Microscopic size and concerted convergences make morphological examination non-trivial and hamper phylogenetic reconstructions. The enigmatic turbellarian-like Rhodopemorpha are a small group that has puzzled systematists for over a century. A preliminary molecular framework places the group far closer to the root of Heterobranchia - one of the major gastropod groups - than previously suggested. The poorly known meiofaunal Helminthope psammobionta Salvini-Plawen, 1991 from Bermuda is the most worm-shaped free-living gastropod and shows apparently aberrant aspects of anatomy. Its study may give important clues to understand the evolution of rhodopemorphs among basal heterobranchs versus their previously thought origin among 'higher' euthyneuran taxa. Results: We describe the 3D-microanatomy of H. psammobionta using three-dimensional digital reconstruction based on serial semithin histological sections. The new dataset expands upon the original description and corrects several aspects. Helminthope shows a set of typical adaptations and regressive characters present in other mesopsammic slugs (called 'meiofaunal syndrome' herein). The taxonomically important presence of five separate visceral loop ganglia is confirmed, but considerable further detail of the complex nervous system are corrected and revealed. The digestive and reproductive systems are simple and modified to the thread-like morphology of the animal; the anus is far posterior. There is no heart; the kidney resembles a protonephridium. Data on all organ systems are compiled and compared to Rhodope. Conclusions: Helminthope is related to Rhodope sharing unique apomorphies. We argue that the peculiar kidney, configuration of the visceral loop and simplicity or lack of other organs in Rhodopemorpha are results of progenesis. The posterior shift of the anus in Helminthope is interpreted as a peramorphy, i.e. hypertrophy of body length early in ontogeny. Our review of morphological and molecular evidence is consistent with an origin of Rhodopemorpha slugs among shelled 'lower Heterobranchia'. Previously thought shared 'diagnostic' features such as five visceral ganglia are either plesiomorphic or convergent, while euthyneury and a double-rooted cerebral nerve likely evolved independently in Rhodopemorpha and Euthyneura. Abstract: The development of a chronic mollusc toxicity test is a current work item on the agenda of the OECD. The freshwater pond snail Lymnaea stagnalis is one of the candidate snail species for such a test. This paper presents a 21-day chronic toxicity test with L. stagnalis, focussing on embryonic development. Eggs were collected from freshly laid egg masses and exposed individually until hatching. The endpoints were hatching success and mean hatching time. Tributyltin (TBT), added as TBT-chloride, was chosen as model substance. The selected exposure concentrations ranged from 0.03 to 10μgTBT/L (all as nominal values) and induced the full range of responses. The embryos were sensitive to TBT (the NOEC for mean hatching time was 0.03μgTBT/L and the NOEC for hatching success was 0.1μgTBT/L). In addition, data on maximum limit concentrations of seven common solvents, recommended in OECD aquatic toxicity testing guidelines, are presented. Among the results, further findings as average embryonic growth and mean hatching time of control groups are provided. In conclusion, the test presented here could easily be standardised and is considered useful as a potential trigger to judge if further studies, e.g. a (partial) life‐cycle study with molluscs, should be conducted. To investigate the bioaccumulation patterns of microcystins (MCs) in organs of two gastropods, samples were collected in Lake Dianchi monthly from May to October, 2008, when cyanobacteria typically bloom. The average MCs concentrations for Radix swinhoei (pulmonate) and Margarya melanioides (prosobranch) tended to be similar for the different organs: the highest values in the hepatopancreas (9.33 by 3.74μg/g DW), followed by digestive tracts (1.66 by 3.03μg/g DW), gonads (0.45 by 1.34μg/g DW) and muscles (0.22 by 0.40μg/g DW). Pulmonate had higher value than prosobranch because of the stronger bioaccumulation ability in hepatopancreas. The levels in organs of R. swinhoei were correlated with environmentally dissolved MCs, but influenced by intracellular MCs for M. melanioides. The estimated MCs concentrations in edible parts of M. melanioides were beyond the WHO’s provisional tolerable daily intake (0.04μg/kg), suggesting the risk of consumption of M. melanioides from the lake. Studies of aquatic environments exposed to 17α-ethinylestradiol (EE2) have demonstrated detrimental effects on fish communities. However, much less is known about effects on macro-invertebrates and especially how long-term exposure may affect critical life stages and ultimately population dynamics. We studied the effects of EE2 on relevant endpoints for population growth in two common freshwater gastropods, Radix balthica and Bithynia tentaculata, that differ in reproductive, foraging and anti-predator strategies and endocrine systems. We quantified critical life-history parameters (mortality, somatic growth rate, days to and size at first reproduction, egg production and hatching success) in a concentration-response, life cycle experiment. The two species responded to EE2 exposure in different ways, B. tentaculata showing a significantly lower somatic growth rate and R. balthica a higher somatic growth rate. However, the magnitudes of the effects were small and EE2 exposure did not have any significant effect on estimated population growth rates for the two snail species. The significant effects of EE2 on individual endpoints, but not on population growth rate for both species, emphasise the importance of evaluating higher level effects from long-term exposure studies. Terrestrial pulmonates can learn olfactory-aversion tasks and retain them in their long-term memory. To elucidate the cellular mechanisms underlying learning and memory, researchers have focused on both the peripheral and central components of olfaction: two pairs of tentacles (the superior arid inferior tentacles) and a pair of procerebra, respectively. bata from tentacleamputation experiments showed that either pair of tentacles is sufficient for olfactory learning. Results of procerebrum lesion experiments showed that the procerebra are necessary for olfactory learning but that either one of the two procerebra, rather than both, is used for each olfactory learning event. Together, these data suggest that there is a redundancy in the structures of terrestrial pulmonates necessary for olfactory learning. Development is left-right reversed between dextral and sinistral morphs of snails. In sympatry, they share the same gene pool, including polygenes for shell shape. Nevertheless, their shell shapes are not the mirror images of each other. This triggered a debate between hypotheses that argue either for a developmental constraint or for zygotic pleiotropic effects of the polarity gene. We found that dextrals can be wider or narrower than sinistrals depending on the population, contrary to the prediction of invariable deviation under a developmental constraint. If the pleiotropy is solely responsible instead, the mean shape of each morph should change, depending on the frequency of polarity genotype. Our simulations of this mean shape change under zygotic pleiotropy, however, show that the direction of interchiral difference remains the same regardless of genotype frequency. Our results suggest the presence of genetic variation among populations that changes the maternal or zygotic pleiotropic effect of the polarity gene. [ABSTRACT FROM AUTHOR]

Environmental calcium is a major factor affecting the distribution of freshwater gastropods. Whilst the effects on growth and morphology are fairly well understood, little is known about how calcium availability affects other aspects of gastropod biology. Lymnaea stagnalis (L.) is considered a calciphile and exhibits reduced growth and survival in environments containing less than 20 mgl-1 Ca2+. Many freshwater systems exhibit fluctuations in calcium concentration overtime: where calcium levels are normally high there may be periods of low [Ca2+], for example following periods of flooding. Here we examined the effects of acute periods of low (20 mg l-1) environmental calcium on the physiology and behaviour of L. stagnalis, specifically measuring how locomotion and respiration differ between high calcium (80 mg l-1) and low calcium (20 mg l-1) environments. We found that in a low calcium environment crawling speed is reduced, and that this coincides with an increase in cutaneous respiration, indicating that the increased metabolic demands of calcium acquisition at low [Ca2+] reduce the energy available for locomotion. Conversely we found a decrease in aerial respiration in hypoxic conditions in the low calcium relative to the high calcium environment. In conclusion, we found that acute exposure to low environmental calcium has a highly significant effect on locomotion and respiration, which may have consequences for snail fitness when no morphological effects are apparent. In metazoan animals, almost every known mutation of visceral asymmetry, which presents the polarity of primary asymmetry established in early development, reverses development in only about half or fewer of homozygotes. However, in pulmonate snails, the dextral and sinistral alleles are traditionally known to determine the polarity of offspring with complete dominance, and thus, each parent should produce either dextral or sinistral progeny. Contrary to this expectation, we found a mutant that produces both chiral morphs (enantiomorphs) within the same clutches in Bradybaena similaris. This study demonstrates that the consistent production of both enantiomorphs is determined by a maternal effect of a recessive allele, which probably randomizes the polarity. In snails that copulate simultaneously and reciprocally, a left–right reversed strain cannot usually be established or rescued from inbreeding depression by ad hoc outbreeding because a rarely found single mutant cannot reproduce due to great difficulties of mating with the wild type and selfing. Moreover, the rare recessive homozygote cannot easily be detected because it often exhibits the wild-type phenotype in maternal inheritance and breeding difficulty hampers genotyping it by phenotyping its progeny. We generated complete mitochondrial genome sequences data for 4 genera (Onchidium, Platevindex, ParaoncidiumandPeronia) in Onchidiidae to construct a phylogenetic tree in conjunction with other 9 existing data among gastropods. The topology showed that the taxa clustered into two main groups of four species, one of which includedOnchidium strumaand thePlatevindex mortoni, the otherParaoncidium reevesiiandPeronia verruculata. The process in Pulmonata from sea to land in accordance with the evolution of respiratory organs from branchial gills to pulmonary cavity has been shown. The taxonomy of the endemic arboreal snail Satsuma albida species complex from Taiwan was unclear due to the animals' highly similar morphology, and their nocturnal and strict arboreal behaviour, leading to difficulties in collecting living specimens. This article is the first comprehensive comparative study on the systematics and taxonomy of this species complex using external morphology, anatomy of the reproductive system and molecular phylogeny. Consequently, two subspecies of S. albida are raised to species status, namely S. insignis and S. mollicula. Fourteen new species are also described. Fourteen of the 17 species showed polymorphism in banding pattern amongst populations and other species retained the whitish unity as seen in S. albida. Distributions of almost all taxa are geographically limited, with the exception of S. polymorpha sp. nov. The phylogeny of these species was reconstructed using 20 morphological characters and molecular data from the partial sequences of mtDNA CO1 and 16S rRNA genes, and the complete ITS2 sequence. Diverse animals exhibit left–right asymmetry in development. However, no example of dimorphism for the left–right polarity of development (whole-body enantiomorphy) is known to persist within natural populations. In snails, whole-body enantiomorphs have repeatedly evolved as separate species. Within populations, however, snails are not expected to exhibit enantiomorphy, because of selection against the less common morph resulting from mating disadvantage. Here we present a unique example of evolutionarily stable whole-body enantiomorphy in snails. Our molecular phylogeny of South-east Asian tree snails in the genus Amphidromus indicates that enantiomorphy has likely persisted as the ancestral state over a million generations. Enantiomorphs have continuously coexisted in every population surveyed spanning a period of 10 years. Our results indicate that whole-body enantiomorphy is maintained within populations opposing the rule of directional asymmetry in animals. Four new labdane diterpenes, 1-4, with an unusual oxidation pattern have been isolated from the marine pulmonate Trimusculus peruvianus. The structure and stereochemistry of these compounds were determined by spectroscopic data and chemical transformations. The absolute stereochemistry of 4 was assigned by application of the modified Mosher method. These compounds exhibit in vitro moderate cytotoxic activity against human colon carcinoma cell lines. The VD and RPD neurons of Lymnaea stagnalis innervate other central neurons, certain skin areas, the pneumostome area, and the auricle of the heart. Recently, a set of four (λ, ω, α, β) neuropeptides produced by these giant neurons and by certain other central neurons has been characterized. Although alternative splicing of the preprohormone of these neurons yields at least 10 different α neuropeptides, an affinity-purified antiserum directed against a domain common to all α neuropeptides has previously been shown to be highly selective in staining VD, RPD and other neurons that produce the preprohormone. Since the gene encoding the neuropeptides is structurally similar to that expressed in R15 of the marine opisthobranch Aplysia californica, we have used the affinity purified antiserum as a marker for VD/RPD-related systems in other molluscs. Immunopositive neurons and fibers are observed in the central nervous systems of all species studied ( Achatina fulica, Anodonta sp., Aplysia brasiliana, A. californica, Bulinus truncatus, Cepea sp., Eobania vermiculata, Helix aspersa, H. pomatia, Limax maximus, Mytilus edulis, Nassarius reticulatus, Viviparus viviparus). Several medium-sized and small neurons and 1-4 giant neurons are found in the pulmonates and opisthobranchs. The giant neurons in pulmonates have locations in the subesophageal ganglion, axonal branching patterns, and terminal arborizations in the auricle of the heart; all these characteristics are similar to those of VD and RPD. Double-labelling (Lucifer yellow injection, immunocytochemistry) confirms that the two giant neurons in Helix pomatia are Br and Br. The immunoreactive cells in A. fulica appear to include the VIN and PON neurons. The antiserum also stains cells that appear to be the R15 neurons in two Aplysia species. The small and medium-sized neurons are distributed widely over the central ganglia of opisthobranchs and pulmonates. In prosobranchs and bivalves, small neurons are found in the cerebral and abdominal ganglia. No evidence has been found for innervation of the heart in these latter groups but in M. edulis, immunoreactive terminals can be observed in the gill. The results suggest the evolutionary conservation of immunoreactive peptides and the neurons that produce them, and thus support and extend previous hypotheses regarding the homology of certain giant neurons across molluscan species. The ultrastructure of the cells involved in the formation of the periostracum in Helisoma duryi duryi is described. Two distinct types of periostracal cells can be recognized: light and dark. The Golgi complexes of the light cell are involved in the biogenesis of periostracal units which are extruded at the base of the periostracal groove. Here they algin into a single row so as to form the membrane-like lamellar covering of the periostracum. While the secretions of the dark periostracal cells contribute to the initial components of the dark layer of the periostracum, the mantle edge gland cells are responsible for its thickening. Background: Spiral cleavage is a feature of non-ecdysozoan protostomes, in which left-right reversal frequently evolved in gastropod molluscs. In pulmonate gastropods, maternal molecules are responsible for chirality patterning, on which the polarities of visceral and coiling asymmetries depend. In the pond snail, Lymnaea stagnalis (the clade Hygrophila), a frame-shift mutation of one of tandem-duplicated, diaphanous-related formin genes (diaph) resulted in incomplete reversal from dextral to sinistral cleavage. Is this mechanism of chirality regulation common to, or shared with other pulmonates? To answer this question, we examined genes involved in chirality patterning in the land snail, Bradybaena similaris which belongs to the clade Stylommatophora. Results: Both dextral and sinistral siblings develop from progeny of a racemic mutant of B. similaris. Differences in maternal mRNAs between the two strains were searched by transcriptome analyses. We found fifty maternal transcripts that exhibited less expression in early embryos of the mutant strain. The most conspicuous was a homolog of diaph. The diaph gene was duplicated in the stylommatophoran ancestor (diaph-a and diaph-b), as in the case of the ancestor of Lymnaea (Lsdiaph1 and Lsdiaph2). The quantity of maternal diaph-b mRNA was drastically reduced in early embryos of the racemic mutant compared to wild-type, while diaph-a expression was at nearly the same level in both strains. Unlike the case of Lsdiaph2, which is frame-shifted to produce truncated peptides in the mutant of L. stagnalis, however, Bsdiaph-b mRNA in the mutant strain is not frame-shifted and most probably produces normal Diaph-b protein. These results suggest the presence of regulatory mechanisms of gene expression for chirality patterning in pulmonate gastropods, although genomic analyses are required for confirmation. Conclusions: Heterochirality resulting from the loss of polarity control in spiral cleavage does not require mutation of the diaph gene in B. similaris. The determination of left-right polarity instead depends on the expression of this diaph gene, which is duplicated in stylommatophoran Bradybaena, as well as in hygrophilan Lymnaea.
