Pulsed Electromagnetic Fields (PEMF) are widely used, with excellent clinical outcomes. However, their mechanism of action has not yet been completely understood. The purpose of this review is to describe current observations on the mechanisms of PEMF, together with its clinical efficacy. Osteoblast responsiveness to PEMF is described on several scales, from the cell membrane to clinically relevant bone formation. PEMF has been shown to activate membrane adenosine receptors. The role of adenosine receptors in activating intracellular second messenger pathways, such as the canonical Wnt/β-catenin pathway and the mitogen-activated protein kinases (MAPK) pathway, is described. The responsiveness of osteoblasts and the synthesis of structural and signaling proteins constitute the role of PEMFs in promoting osteogenesis and bone matrix synthesis, and they are described. Multiple studies, ranging from observational and randomized to meta-analyses that investigate the clinical efficacy of PEMF, are described. This review presents a favorable conclusion on the clinical effects of PEMF while unlocking the "black box" of PEMF's mechanism of action, thus improving confidence in the clinical utility of PEMF in bone repair. Pulsed electromagnetic field (PEMF) therapy is a potential non-invasive treatment to modulate immune responses and inhibit tumor growth. Cervical cancer (CC) is influenced by IL-37-mediated immune regulation, making PEMF therapy a potential strategy to impede CC progression. This study aimed to elucidate the effects of PEMF on IL-37 regulation and its molecular mechanisms in CC. CC cell-xenografted mouse models, including IL-37 transgenic (IL-37tg) mice, were used to assess tumor growth through in vivo fluorescence imaging and analyze CC cell apoptosis via flow cytometry. TCGA-CESC transcriptome and clinical data were analyzed to identify key inflammation and immune-related genes. CD8+ T cell models were stimulated with PEMF, and apoptosis, oxidative stress, and inflammatory factor expression were analyzed through RT-qPCR, Western blot, and flow cytometry. PEMF treatment significantly inhibited IL-37 expression (p < 0.05), promoted inflammatory factor release (TNF-α and IL-6), and activated oxidative stress, leading to increased CC cell apoptosis (p < 0.05). IL-37 interaction with SMAD3 impacted the p38/NF-κB signaling pathway, modulating CD8+ T cell activity and cytotoxicity. Co-culture of Hela cells with CD8+ T cells under PEMF treatment showed reduced proliferation (by 40%), migration, and invasion (p < 0.05). In vivo experiments with CC-bearing mice demonstrated that PEMF treatment downregulated IL-37 expression (p < 0.05), enhanced CD8+ T cell function, and inhibited tumor growth (p < 0.05). These molecular mechanisms were validated through RT-qPCR, Western blot, and immunohistochemistry. BACKGROUND: Pulsed electromagnetic fields, as an important physical therapy, are exactly effective in the treatment of osteoarthritis, but the mechanism has not been fully clarified. OBJECTIVE: To observe the effect of pulsed electromagnetic field on the degeneration of knee joint cartilage in aged rats. METHODS: Eight 6-month-old Sprague-Dawley rats were selected as the young group and were subjected to normal diet with no treatment. Sixteen 22-monthold Sprague-Dawley rats were randomly divided into old group (n=8) and pulsed electromagnetic field group (n=8). The rats in the pulsed electromagnetic field group were subjected to a pulsed electromagnetic field intervention, once a day, 5 days per week for continuous 8 weeks. The rats in the old group were given no treatment. All rats were anesthetized and executed after 8 weeks for the detection of relevant indexes. RESULTS AND CONCLUSION: Compared with the young group, serum type II collagen C-terminal peptide level was increased in the old group (P < 0.05); compared with the old group, serum type II collagen C-terminal peptide level was decreased in the pulsed electromagnetic field group (P < 0.05). Micro-CT showed that the bone volume fraction, bone mineral density, and number of bone trabeculae decreased (P < 0.05) and the trabecular separation increased (P < 0.05) in the tibia of rats in the aged group compared with the young group; and the bone volume fraction, bone density, and number of trabeculae increased (P < 0.05) and the trabecular separation decreased (P < 0.05) in the tibia of rats in the pulsed electromagnetic field group compared with the aged group. The tibial plateau Safranin O-fast green staining showed that the articular cartilage structure of rats in the aged group was disorganized, and the number of chondrocytes was obviously reduced, and the tidal line could not be distinguished. The above results were improved in the pulsed electromagnetic field group. RT-qPCR and western blot assay showed that the mRNA and protein expression levels of matrix metalloproteinase 1, matrix metalloproteinase 13, P53 and P21 in the articular cartilage and subchondral bone of rats were elevated in the aged group compared with the young group (P < 0.05) and decreased in the pulsed electromagnetic field group compared with the old group (P < 0.05). To conclude, pulsed electromagnetic fields may improve osteoarthritis in aged rats by inhibiting chondrocyte senescence, alleviating articular cartilage degradation and inhibiting subchondral bone osteoporosis through suppressing the expression of P53/P21. Diabetic foot syndrome is a syndrome of specific conditions affecting the foot. It is a complication of diabetes. It occurs in 12–25% of patients with diabetes. Untreated, it leads to irreversible deformities and necrosis of the foot, often resulting in amputation. In this study the statistics and consequences of diabetic foot syndrome are described. Patients with diabetes need to take care of their lower limbs. Proper footwear can prevent foot wounds. Available solutions for the prevention and treatment of diabetic foot syndrome are presented herein: footwear, insoles and requirements for footwear materials. Appropriate equipment for a person with diabetes, especially one who has been diagnosed with diabetic foot syndrome or is at risk of such a condition, includes footwear and replaceable insoles. The parameters of footwear, insoles and footwear materials that are most optimal for patients with diabetes and diabetic foot syndrome were defined. Alveolar bone remodeling during the retention phase is essential for successful orthodontic treatment. Pulsed electromagnetic field (PEMF) therapy is an adjunctive therapy for bone-related diseases that induces osteogenesis and prevents bone loss. This study aimed to examine the role of PEMF exposure during the retention phase of orthodontic treatment in alveolar bone remodeling. A total of 36 male Wistar rats were divided into control, PEMF 7, and PEMF 14 groups; a 50 g force nickel–titanium closed-coil spring was inserted to create mesial movement in the first molar for 21 d. Furthermore, the spring was removed, and the interdental space was filled with glass ionomer cement. Concurrently, rats were exposed to a PEMF at 15 Hz with a maximum intensity of 2.0 mT 2 h daily, for 7 and 14 days. Afterwards, the cements were removed and the rats were euthanized on days 1, 3, 7, and 14 to evaluate the expression of Wnt5a mRNA and the levels of RANKL, OPG, ALP, and Runx2 on the tension side. The data were analyzed with ANOVA and post hoc tests, with p < 0.05 declared statistically significant. PEMF exposure significantly upregulated Wnt5a mRNA expression, OPG and ALP levels, and Runx2 expression, and decreased RANKL levels in the PEMF 7 and 14 groups compared to the control group (p < 0.05). This study showed that PEMF exposure promotes alveolar bone remodeling during the orthodontic retention phase on the tension side by increasing alveolar bone formation and inhibiting resorption. Purpose: PEMF (pulsed electromagnetic fields) founds application in several medical fields to accelerate bone wounds healing and to reduce inflammation. The aim of our study was to evaluate the effectiveness of PEMF in reducing postoperative swelling and pain in patients undergoing orthognathic surgery. Methods: A prospective observational monocentric study was conducted on a sample of 30 patients undergone to orthognathic surgery in Maxillofacial Surgery Unit of University of Naples Federico II. The patients who followed these inclusion criteria were enrolled in the study: age ≥ 18 years, Class III malocclusion, Surgical procedure of Le Fort I osteotomy + Bilateral Sagittal Split Osteotomy (BSSO), Written informed consent. Patients were divided into two groups: Group SD) postoperative standard treatment with medical therapy and cryotherapy, Group SD + PEMF) postoperative standard therapy + PEMF. Each patient underwent a 3D facial scan, at one (1d) and four (4d) days after surgery to compare the swelling reduction. The pain score was assessed through VAS score and analgesics administration amount. Results: In SD + PEMF group, the facial volume reduction between 1d and 4d scan was on average 56.2 ml (6.23%), while in SD group, it was 23.6 ml (2.63%). The difference between the two groups was 3.6% (p = 0.0168). VAS pain values were significantly higher in SD group compared to SD + PEMF group in the second day after surgery (P = 0.021) and in the total 4 days (P = 0.008). Conclusions: Our data suggest that PEMF is valid tool to promote faster postoperative swelling and pain reduction in patients undergoing orthognathic surgery. [ABSTRACT FROM AUTHOR]

Abstract Pulsed electromagnetic field (PEMF) therapy has been extensively investigated in clinical studies for the treatment of angiogenesis-related diseases. However, there is a lack of research on the impact of PEMFs on energy metabolism and mitochondrial dynamics during angiogenesis. The present study included tube formation and CCK-8 assays. A Seahorse assay was conducted to analyze energy metabolism, and mitochondrial membrane potential assays, mitochondrial imaging, and reactive oxygen species assays were used to measure changes in mitochondrial structure and function in human umbilical vein endothelial cells (HUVECs) exposed to PEMFs. Real-time polymerase chain reaction was used to analyze the mRNA expression levels of antioxidants, glycolytic pathway-related genes, and genes associated with mitochondrial fission and fusion. The tube formation assay demonstrated a significantly greater tube network in the PEMF group compared to the control group. The glycolysis and mitochondrial stress tests revealed that PEMFs promoted a shift in the energy metabolism pattern of HUVECs from oxidative phosphorylation to aerobic glycolysis. Mitochondrial imaging revealed a wire-like mitochondrial morphology in the control group, and treatment with PEMFs led to shorter and more granular mitochondria. Our major findings indicate that exposure to PEMFs accelerates angiogenesis in HUVECs, likely by inducing energy metabolism reprogramming and mitochondrial fission.

This paper presents the findings of a comprehensive study exploring the synergistic effects arising from the combination of microwave ablation and pulsed electromagnetic field (PEMF) therapy on prostate cancer cells. The research encompassed five distinct experimental groups, with continuous electric field measurements conducted during the entire treatment process. Group 1 and Group 2, subjected to microwave power below 350 W, exhibited specific electric field values of 72,800 V/m and 56,600 V/m, respectively. In contrast, Group 3 and Group 4, exposed to 80 W microwave power, displayed electric field levels of approximately 1450 V/m, while remaining free from any observable electrical discharges. The migratory and invasive capacities of PC3 cells were assessed through a scratch test in all groups. Notably, cells in Group 3 and Group 4, subjected to the combined treatment of microwave ablation and PEMF, demonstrated significantly accelerated migration in comparison to those in Groups 1 and 2. Additionally, Group 5 cells, receiving PEMF treatment in isolation, exhibited decreased migratory ability. These results strongly suggest that the combined approach of microwave ablation and PEMF holds promise as a potential therapeutic intervention for prostate cancer, as it effectively reduced cell viability, induced apoptosis, and impeded migration ability in PC3 cells. Moreover, the isolated use of PEMF demonstrated potential in limiting migratory capacity, which could hold critical implications in the fight against cancer metastasis. In this study, a new approach to treat prostate cancer by combining microwave ablation (MWA) and pulsed electromagnetic field (PEMF) therapy is explored. We used specific devices like rectangular waveguides for MWA and circular coils for PEMF. The energy sources utilized in the study comprised a magnetron tube system, similar to the microwave source found in a microwave oven, for generating microwaves, and a signal generator for producing PEMF. We used specialized equipment for MWA and PEMF to maintain controlled conditions, ensuring precise and reliable results. The research included testing various groups of prostate cancer cells exposed to different intensities of microwave power and magnetic flux density. The movement of cancer cells in different groups was examined through a wound healing assay, where cancer cells were placed on a flat surface, and we observed whether they filled the gap created by their movement. Interestingly, cells treated with both MWA and PEMF demonstrated faster movement compared to cells treated with MWA alone or PEMF alone. This combined treatment not only effectively decreased cell movement but also showed the potential cell death. The results showed that the combination of MWA and PEMF suggest a promising therapeutic strategy. The findings contribute to the development of precise and effective therapies that could enhance patient outcomes and quality of life. A pulsed electromagnetic field (PEMF) enhances the efficacy of several anticancer drugs. Doxorubicin (DOX) is an anticancer agent used to treat various malignancies, including breast cancer. This study examined whether a PEMF increases the anticancer effect of DOX on MCF-7 human breast cancer cells and elucidated the underlying mechanisms affected by PEMF stimulation in DOX-treated MCF-7 human breast cancer cells. A cotreatment with DOX and a PEMF potentiated the reduction in MCF-7 cell viability compared to the treatment with DOX alone. The PEMF elevated DOX-induced G1 arrest by affecting cyclin-dependent kinase 2 phosphorylation and the expression of G1 arrest-related molecules, including p53, p21, cyclin E2, and polo like kinase 1. In addition, PEMF increased the DOX-induced upregulation of proapoptotic proteins, such as Fas and Bcl-2-associated X, and the downregulation of antiapoptotic proteins, including myeloid leukemia 1 and survivin. PEMF promoted the DOX-induced activation of caspases-8, -9, and -7 and poly (adenosine diphosphateribose) polymerase cleavage in MCF-7 cells. In conclusion, PEMF enhances the anticancer activity in DOX-treated MCF-7 breast cancer cells by increasing G1 cell cycle arrest and caspase-dependent apoptosis. These findings highlight the potential use of a PEMF as an adjuvant treatment for DOX-based chemotherapy against breast cancer.

Using an AZ91D magnesium alloy as the research object, a three-dimensional (3D) numerical model was created and simulated using simulation analysis software. The effects of a rotating electromagnetic field (REMF), a pulsed electromagnetic field (PEMF) and a rotating pulsed electromagnetic field (R-PEMF) on the magnetic field and the flow field of a metal melt were investigated. The simulation results show that during the REMF treatment a rotational force is generated on the cross-section of the melt, and the PEMF exerts a magnetic pressure on the cross-section of the melt. Under the combined effect of these two forces, the Lorentz force increases parallel to the melt axis, and a secondary flow directed towards the core of the melt is generated, which contributes to the homogenization of the melt. By analyzing the solidification structure and elemental distribution of the metal, R-PEMF was shown to effectively improve the solidification structure and macrosegregation of Al in AZ91D magnesium alloy. BACKGROUND: Pulsed electromagnetic field is a non-invasive and non-radiative treatment method. Clinical use of pulsed electromagnetic fields in the treatment of orthopedic diseases has achieved certain results. OBJECTIVE: To review the current clinical application of the pulsed electromagnetic field in the treatment of orthopedic diseases, providing a scientific theoretical basis for the clinical treatment of orthopedic diseases. METHODS: The first author used a computer to search PubMed, CBM, Cochrane Library, CNKI, and WanFang Data for related studies on the pulsed electromagnetic field in the treatment of orthopedic diseases, using the keywords of “pulsed electromagnetic field, orthopedics, osteoarthritis, osteoporosis, bone healing, electromagnetic navigation” in English and Chinese. For the literature related to the same content, recent publications were selected. A total of 69 articles were selected from the search results for review. RESULTS AND CONCLUSION: Pulsed electromagnetic field has a definite curative effect on fracture healing. It can be used in the treatment of osteomyelitis by antibacterial, bactericidal, anti-inflammatory and promoting bone healing, and can inhibit osteoporosis and its progress. In addition, the treatment of early osteoarthritis, femoral head necrosis and postoperative rehabilitation of late joint replacement through various ways can become a treatment for orthopedic diseases. However, the therapeutic mechanism of the pulsed electromagnetic field for a variety of orthopedic diseases is still unclear, and most of the research is still in the primary stage. In the future, it is still necessary to obtain more reliable evidence from high-quality research and clinical trials to provide a more perfect basis for the clinical treatment of orthopedic diseases. Background: Temporomandibular joint osteoarthritis (TMJOA) is characterized by articular cartilage degeneration and progressive synovitis. How to effectively inhibit TMJOA in the early stage has been a hot topic in the biomedical field. As a non‐invasive physiotherapy, pulsed electromagnetic field (PEMF) treatment has shown great potential in the treatment of osteoarthritis (OA) in extremity joints. Objective: This study aims to investigate the biological effect of PEMF intervention on TMJ cartilage degeneration and synovium inflammation at the early stage of TMJOA. Methods: PEMF (2.0 mT, 15 Hz, 2 h/day) treatment was given to rats in which TMJOA was induced by applying the unilateral anterior crossbite (UAC). Histological and immunohistochemical staining, TUNEL assay, real‐time PCR and western blotting assay were performed to detect the changes of the morphology and the expression of pro‐inflammatory and degradative factors in condylar cartilage and synovium. Results: Obvious condylar cartilage degeneration, characterized by decreased cartilage thickness, degraded cartilage extracellular matrix, increased expression of pro‐inflammatory and degradative factors (TNF‐α, IL‐1β, MMP‐13, ADAMTS‐5, IL‐6, MMP‐3, MMP‐9 and COL‐X) and increased chondrocytes death, was observed in UAC group, accompanied by synovium hyperplasia and up‐regulation of pro‐inflammatory and degradative factors in synovium. PEMF intervention reversed the decreased cartilage thickness at 3 weeks and degraded cartilage extracellular matrix at 6 weeks. Moreover, the up‐regulation of pro‐inflammatory, degradative and hypertrophyic factors and chondrocytes death in condylar cartilage induced by UAC were inhibited to some extent. In addition, the synovium hyperplasia and the up‐regulation of pro‐inflammatory and degradative factors in synovium were inhibited at 3 weeks and 6 weeks. Conclusions: Appropriate PEMF stimulation can reverse the loss of cartilage extracellular matrix, the chondrocytes death, the increased expression of pro‐inflammatory and degradative factors in cartilage, the decreased cartilage thickness and synovium inflammation induced by UAC at the early stage of TMJOA to some extent. Introduction: The purpose of this study is to evaluate whether using a Fracture Healing Patch (FHP) device that generates pulsed electromagnetic fields (PEMF), applied at the fracture site immediately after open reduction and internal fixation surgery, can accelerate healing of acute distal radius fractures. Methods: In a prospective, double-blind, randomized, and sham-controlled study, thirty-two patients with DRFs treated with ORIF were included. Patients were allocated to a PEMF (active) group (n = 15) or a control (sham) group (n = 17). All patients were assessed with regard to functional Patient-Rated Wrist Evaluation (PRWE), SF12, and radiological union outcomes (X-rays and computed tomography (CT) scans) at 2, 4, 6, and 12 weeks postoperatively. Results: Patients treated with the FHP demonstrated significantly bone bridging at 4 weeks as assessed by CT (70% vs 54%, p = 0.05). Mean grip strength in the active group was significantly higher as compared to control (16 ± 9 kg vs 7 ± 3.5 kg, respectively, p = 0. 02). The function subscale of the PRWE was significantly better in PEMF-treated group at 6 weeks after surgery (27.2 VS 35.5, p = 0.04). No statistically significant differences were found in SF12. Conclusion: PEMF application after ORIF of DRFs is safe, may accelerate bone healing which could lead to an earlier return to daily life activities and work. Pulsed electromagnetic field (PEMF) stimulation is a non-invasive therapeutic modality that has been employed in a variety of applications, including skin wound healing, tissue regeneration, blood vessel regeneration, repair of nonunion bone fractures, and dental treatment. Currently, the potential side effects of electromagnetic fields on tissues have not been comprehensively explored, despite their established therapeutic benefits. In this study, we examined the side effects of PEMF devices on whole-body tissues using simulations. The electromagnetic field simulation software CST Studio Suite was utilized, and an RF coil that could output different frequencies (100 Hz, 100 kHz, and 27 MHz) and magnetic fields (0.25 mT, 0.5, and 1 mT) was designed for the purpose of simulations. A realistic rat model, available from the CST library, was employed. Using the Debye equations according to the relevant frequency value, the electrical and thermal properties of the rat were calculated, and the simulation was carried out in two stages: EM and thermal analysis. The specific absorption rate in the tissues with EM simulation and the temperature distribution in the tissues with thermal analysis were examined. The effects created by the PEMF device at different frequencies and magnetic field intensities were presented comparatively. According to the results, the highest SAR value was observed for three different magnetic field intensities at a frequency of 27 MHz, resulting in a temperature difference of 0.7 °C. This study aims to determine the EM effects on rats in a simulated environment according to different frequencies and magnetic field values, as well as to determine whether the thermal effects created can produce a side response. Osteoporotic osteoarthritis (OPOA) is a specific phenotype of OA with high incidence and severe cartilage damage. This study aimed to explore the protective efficacy of PEMF on the progression of OPOA and observed the effects of PEMF on PPARγ, autophagy- and apoptosis-related proteins in OPOA rats. Rats were randomly divided into three groups: control group, OPOA group, and PEMF group (n = 6). One week after surgery, the rats in PEMF group were subjected to PEMF (3.82 mT, 8 Hz, 40 min/day and 5 day/week) for 12 weeks. Results showed that PEMF retarded cartilage degeneration and bone loss, as evidenced by pathological staining image, decreased MMP-13 expression and increased bone mineral density. PEMF inhibited the serum levels of inflammatory cytokines, and the expressions of caspase-3 and caspase-8, while upregulated the expression of PPARγ. Moreover, PEMF significantly improved the autophagy disorders, represented by decrease expressions of Beclin-1, P62, and LC3B. The research demonstrates that PEMF can effectively prevent cartilage and subchondral bone destruction in OPOA rats. The potential mechanism may be related to upregulation of PPARγ, inhibition of chondrocyte apoptosis and inflammation, and improvement of autophagy disorder. PEMF therapy thus shows promising application prospects in the treatment of postmenopausal OA. Osteoporotic osteoarthritis (OPOA) is a very common combination disease, that characterized by chronic pain, swollen joints and susceptibility to fractures. It is particularly common in postmenopausal women. At present, drug therapy is the main treatment method, but the adverse reactions are serious and can not stop the progression of the disease. PEMF is a safe physical therapy that has been shown to increase bone density, reduce pain, and improve joints mobility. In this study, we aimed to explore the protective effect and potential mechanism of PEMF on OPOA. We found that PEMF significantly inhibited the inflammatory response, ameliorated the damaged cartilage and subchondral bone in OPOA rats, that maybe related to the regulation of chondrocyte autophagy and apoptosis. Pulsed electromagnetic field (PEMF), as a non-invasive, safe and effective physical therapy, is first applied in treating patients with delayed healing and non-union of fractures, and is now used as a clinical adjunct in the treatment of osteoporosis. In vitro and animal studies have demonstrated that PEMF stimulates bone formation by osteoblasts and inhibits bone resorption by osteoclasts, ultimately achieving structural integrity of bone and preservation of bone mass and bone strength. A large number of clinical studies have also shown that PEMF has satisfactory therapeutic effect on increasing bone mineral density, relieving pain, and improving life quality of patients with osteoporosis. However, the certainty of the positive effects of PEMF on osteoporosis still needs to be further investigated. In this review, we summarize the relevant clinical literature on PEMF for the treatment of osteoporosis in the domestic and overseas, and discuss the possible mechanisms, aiming to provide some reference for future clinical studies. Introduction: Exposure to pulsed electromagnetic field (PEMF) has been revealed to affect the differentiation and proliferation of human mesenchymal stem cells derived from dental pulp multipotent stromal stem cells (DP-MSCs). This study aimed to investigate the differentiation effect of electromagnetic fields (EMFs) on the DP-MSC. Materials and Methods: PEMF was produced by a system comprising a multi-meter autotransformer, solenoid coils, and teslameter. This study included 10 groups of DP-MSCs which underwent different electromagnetic radiation time and beam intensity. Three samples tested for each group. The effect of PEMF with the intensity of 0.5 and 1 mT (mili Tesla) and 50 Hz on the proliferation rate of DP-MSC was evaluated at 20 and 40 minutes per day for seven days. MTT assay was applied to determine the growth and proliferation of DP-MSC. Gene expression of DMP1 for differentiation of DPSCs to odontoblasts was confirmed by Real Time PCR., ANOVA statistical analysis and Kruskal-Wallis test were used to analyze the data. Results: The survival in all exposure groups was significantly higher than that in control except in the group of 40 minutes, 1 mT (P<0.05). In 20 minutes, 0.5 mT exposure, the survival intensity is significantly more than others (P<0.05). In general, the intensity of survival was recorded, 20, 0.5 mT≥20, 1 mT≥40, 0.5 mT≥40, 1 mT respectively. Therefore, according to the obtained results, ELF-EMF increases the survival of cells except for one case (40 minutes, 1 mT), even though the effective underlying mechanisms in this process are still unclear. Conclusions: The results obtained promise that in the future, by placing an important part of the pulp next to the electromagnetic field, the lost part of the pulp can be reconstructed and the dentin barrier can be created. Functional tissue engineering strategies provide innovative approach for the repair and regeneration of damaged cartilage. Hydrogel is widely used because it could provide rapid defect filling and proper structure support, and is biocompatible for cell aggregation and matrix deposition. Efforts have been made to seek suitable scaffolds for cartilage tissue engineering. Here Alg-DA/Ac-β-CD/gelatin hydrogel was designed with the features of physical and chemical multiple crosslinking and self-healing properties. Gelation time, swelling ratio, biodegradability and biocompatibility of the hydrogels were systematically characterized, and the injectable self-healing adhesive hydrogel were demonstrated to exhibit ideal properties for cartilage repair. Furthermore, the new hydrogel design introduces a pre-gel state before photo-crosslinking, where increased viscosity and decreased fluidity allow the gel to remain in a semi-solid condition. This granted multiple administration routes to the hydrogels, which brings hydrogels the ability to adapt to complex clinical situations. Pulsed electromagnetic fields (PEMF) have been recognized as a promising solution to various health problems owing to their noninvasive properties and therapeutic potentials. PEMF treatment offers a better clinical outcome with fewer, if any, side effects, and wildly used in musculoskeletal tissue repair. Thereby we propose PEMF as an effective biophysical stimulation to be 4th key element in cartilage tissue engineering. In this study, the as-prepared Alg-DA/Ac-β-CD/gelatin hydrogels were utilized in the rat osteochondral defect model, and the potential application of PEMF in cartilage tissue engineering were investigated. PEMF treatment were proven to enhance the quality of engineered chondrogenic constructs in vitro, and facilitate chondrogenesis and cartilage repair in vivo. All of the results suggested that with the injectable self-healing adhesive hydrogel and PEMF treatment, this newly proposed tissue engineering strategy revealed superior clinical potential for cartilage defect treatment.

Myocardial infarction (MI) is a serious threat to human health. Although monotherapy with pulsed electromagnetic fields (PEMFs) or adipose-derived stem cells (ADSCs) has been reported to have positive effect on the treatment of MI, a satisfactory outcome has not yet been achieved. In recent years, combination therapy has attracted widespread interest. Herein, we explored the synergistic therapeutic effect of combination therapy with PEMFs and ADSCs on MI and found that the combination of PEMFs and ADSCs effectively reduced infarct size, inhibited cardiomyocyte apoptosis and protected the cardiac function in mice with MI. In addition, bioinformatics analysis and RT-qPCR showed that the combination therapy could affect apoptosis by regulating the expression of miR-20a-5p. A dual-luciferase reporter gene assay also confirmed that the miR-20a-5p could target E2F transcription factor 1 (E2F1) and inhibit cardiomyocyte apoptosis by regulating the E2F1/p73 signaling pathway. Therefore, our study systematically demonstrated the effectiveness of combination therapy on the inhibition of cardiomyocyte apoptosis by regulating the miR-20a-5p/E2F1/p73 signaling pathway in mice with MI. Thus, our study underscored the effectiveness of the combination of PEMFs and ADSCs and identified miR-20a-5p as a promising therapeutic target for the treatment of MI in the future. Abstract Background Temporomandibular joint osteoarthritis (TMJOA) is characterized by abnormal subchondral bone remodeling and cartilage degeneration. As a non-invasive biophysical technology, pulsed electromagnetic field (PEMF) treatment has been proven to be efficient in promoting osteogenesis. However, the potential bone protective effect and mechanism of PEMF on abnormal subchondral bone remodeling in TMJOA are unknown. Methods Unilateral anterior crossbite (UAC) was used to create TMJOA model in rats, and 17β-estradiol (E2) were injected daily to mimic patients with high-physiological levels of estrogen. Mouse osteoblast-like MC3T3-E1 cells treated with recombinant murine IL-1β was used to establish inflammatory environment in vitro. The treatment group were subjected to PEMF (2.0mT, 15 Hz, 2 h/d). Micro-CT scanning, histological staining, real-time PCR and western blotting assays were preformed to observe the changes in the subchondral bone. Results Abnormal resorption of subchondral bone induced by UAC, characterized by decreased bone mineral density, increased osteoclast activity and expression of osteoclast-related factors (RANKL) and down-regulated expression of osteogenesis-related factors (OPG, ALP, Runx2 and OCN) at the early stage, could be reversed by PEMF exposure, which was similar to the effect of estrogen. In addition, PEMF exposure and E2 supplement may have a synergistic effect to some extent. Moreover, PEMF exposure could promote the ALP activity and osteogenic mineralization ability of MC3T3-E1 cells. PEMF promoted the expression of factors related to Wnt/β-Catenin signal pathway both in vivo and in vitro. Conclusions Appropriate PEMF exposure have a protective effect on subchondral bone in TMJOA at early stage, in which canonical Wnt/β-Catenin pathway may be involved. PEMF may be a promising biophysical approach for early intervention of TMJOA in clinic.

Neuroinflammation is one of the most important pathological processes following brain ischemia. Pulsed electromagnetic fields (PEMFs) protect against brain ischemia, but their role in regulating neuroinflammation remains unclear. In the present study, we investigated the biological effects of PEMF exposure on brain ischemia-induced neuroinflammation through the astrocytic cholinergic anti-inflammatory pathway. PEMF exposure reduced the activation of astrocytes and neuroinflammation following brain ischemia by directly modulating astrocytic injury and inflammatory cytokine release. Inhibition of nicotinic acetylcholine receptor alpha 7 subunit (α7nAChR) by a specific antagonist reversed the regulatory effects of PEMF on astrocytes. Furthermore, negative regulation of signal transducer and activator of transcription 3 (STAT3) by α7nAChR was found to be an important downstream mechanism through which PEMF regulates astrocyte-related neuroinflammation. PEMF suppressed STAT3 phosphorylation and nuclear translocation by activating α7nAChR. Background: this pilot study aimed at determining whether the application of a novel new method of generating pulsed electromagnetic field (PEMF), the Fracture Healing Patch (FHP), accelerates the healing of acute distal radius fractures (DRF) when compared to a sham treatment. Methods: 41 patients with DRFs treated with cast immobilization were included. Patients were allocated to a PEMF group (n = 20) or a control (sham) group (n = 21). All patients were assessed with regard to functional and radiological outcomes (X-rays and CT scans) at 2, 4, 6 and 12 weeks. Results: fractures treated with active PEMF demonstrated significantly higher extent of union at 4 weeks as assessed by CT (76% vs. 58%, p = 0.02). SF12 mean physical score was significantly higher in PEMF treated group (47 vs. 36, p = 0.005). Time to cast removal was significantly shorter in PEMF treated patients, 33 ± 5.9 days in PEMF vs. 39.8 ± 7.4 days in sham group (p = 0.002). Conclusion: early addition of PEMF treatment may accelerate bone healing which could lead to a shorter cast immobilization, thus allowing an earlier return to daily life activities and work. AimsThe aim of this study was to develop a new class of gallium (Ga)-doped chitosan (CS) coatings fabricated by electrophoretic deposition (EPD) in staphylococcal infection therapy.Methods and resultsBiofilm formation on EPD CS/Ga coatings by Staphylococcus epidermidis and Staphylococcus aureus, which are the main strains involved in postarthroplasty infections, was assessed. The codeposition of an antibacterial agent was effective; Ga loaded into CS matrix reduces biofilm viability by up to 86% and 80% for S. epidermidis and S. aureus strains respectively. Lastly, the influence of pulsed electromagnetic field (PEMF) on the bactericidal activity of CS/Ga coatings was investigated in vitro. To this end, the coatings were incubated with S. epidermidis and S. aureus and exposed to the PEMF using two different frequencies and times. Biofilm viability for S. epidermidis was decreased by 35-40% in the presence of low-frequency (LF) and high-frequency (HF) PEMF respectively. Biofilm viability by S. aureus was not further reduced in the presence of LF PEMF, but decreased by 38% at HF PEMF.ConclusionsThis study has established that a combination of PEMFs with the antibacterial agent improves bactericidal activity of Ga against S. epidermidis strain 14990 and S. aureus strain 12600.Significance and impact of the studyThis new integrated approach could reduce the incidence of infection in orthopaedic implant applications. Darbeli Elektromanyetik Alan (PEMF) elektromanyetik alanların iyonlaştırıcı olmayan formlarından biri olup nörodejeneratif bozuklukların semptomlarının tedavisi gibi çeşitli tıbbi problemler için alternatif bir tedavi olarak kullanılmaktadır. Çalışmanın amacı, yüksek doz hidrojen peroksit (H2O2) ile muamele edilen insan nöroblastoma hücre hattında kısa süreli 75 Hz frekanslı PEMF maruziyetinin glutatyon (GSH) miktarına etkilerini araştırmaktır. Hücreler üç deneysel gruba ayrılmıştır: (I) sham-kontrol; (II) H2O2 ile muamele edilen hücreler; (III) H2O2 muamelesinin ardından PEMF'ye maruz bırakılan hücreler. Hücre canlılığı ve glutatyon miktarı sırasıyla spektrofotometrik ve Yüksek Performanslı Likit Kromatografi (HPLC) teknikleri kullanılarak ölçülmüştür. Yüksek doz H2O2 ile muamele edilen nöroblastoma hücre hattında muamele sonrası PEMF maruziyetinin oksidatif stresin zararlı etkilerine karşı sitoprotektif etkisinin, hücre canlılığında ve GSH miktarında artış ile ilişkili olduğu bulunmuştur.

Notable characteristics of the skeleton are its responsiveness to physical stimuli and its ability to remodel secondary to changing biophysical environments and thereby fulfill its physiological roles of stability and movement. Bone and cartilage cells have many mechanisms to sense physical cues and activate a variety of genes to synthesize structural molecules to remodel their extracellular matrix and soluble molecules for paracrine signaling. This review describes the response of a developmental model of endochondral bone formation which is translationally relevant to embryogenesis, growth, and repair to an externally applied pulsed electromagnetic field (PEMF). The use of a PEMF allows for the exploration of morphogenesis in the absence of distracting stimuli such as mechanical load and fluid flow. The response of the system is described in terms of the cell differentiation and extracellular matrix synthesis in chondrogenesis. Emphasis is placed upon dosimetry of the applied physical stimulus and some of the mechanisms of tissue response through a developmental process of maturation. PEMFs are used clinically for bone repair and have other potential clinical applications. Rheumatoid arthritis (RA) is an autoimmune disease of the joint synovial membranes. RA is difficult to prevent or treat; however, blocking proinflammatory cytokines is a general therapeutic strategy. Pulsed electromagnetic field (PEMF) is reported to alleviate RA's inflammatory response and is being studied as a non-invasive physical therapy. In this current study, PEMF decreased paw inflammation in a collagen-induced arthritis (CIA) murine model. PEMF treatment at 10 Hz was more effective in ameliorating arthritis than at 75 Hz. In the PEMF-treated CIA group, the gross inflammation score and cartilage destruction were lower than in the untreated CIA group. The CIA group treated with PEMF also showed lower serum levels of IL-1β but not IL-6, IL-17, or TNF-α. Serum levels of total anti-type II collagen IgG and IgG subclasses (IgG1, IgG2a, and IgG2b) remained unchanged. In contrast, tissue protein levels of IL-1β, IL-6, TNF-α, receptor activator of nuclear factor kappa-Β (RANK), RANK ligand (RANKL), IL-6 receptor (IL-6R), and TNF-α receptor1 (TNFR1) were all lower in the ankle joints of the PEMF-treated CIA group compared with the CIA group. The results of this study suggest that PEMF treatment can preserve joint morphology cartilage and delay the occurrence of CIA. BACKGROUND: Mesenchymal stem cells, with self-replication and multidirectional differentiation potential, attracted much attention in the field of bone tissue regeneration. As a non-invasive physical stimulation factor, the magnetic field can effectively regulate the biological behaviors of mesenchymal stem cells such as adhesion, proliferation, osteogenic differentiation and mineralization, which means that it has a broad application prospect in bone tissue regeneration medicine. The effect of the magnetic field depends on parameters such as magnetic field strength, frequency and exposure time. Due to the window effect, the appropriate parameter range of magnetic field promoting bone regeneration remains to be clarified. OBJECTIVE: To review the effects of different magnetic fields and different magnetic parameters on osteogenic differentiation of mesenchymal stem cells as well as the possible mechanisms, and to summarize the appropriate parameters for promoting proliferation and osteogenic differentiation. METHODS: Databases of CNKI and PubMed were searched for relevant articles published from January 2010 to September 2021 using the keywords including “magnetic field, static magnetic field, pulsed electromagnetic field, mesenchymal stem cells, proliferation, osteogenic differentiation, bone regeneration, bone repair” in Chinese and English. A few classic early articles were also included. By reading the titles and abstracts, a preliminary screening was conducted. Repetitive studies, low-quality or irrelevant articles were excluded. Finally, a total of 56 articles were retained and analyzed for this review. RESULTS AND CONCLUSION: (1) Moderate static magnetic field, low frequency or low intensity pulsed electromagnetic field, have positive effects on the proliferation and osteogenic differentiation of mesenchymal stem cells. High-frequency pulsed electromagnetic field produces obvious inhibitory effect. (2) The magnetic field modulation effect was positively correlated with the total exposure time, but its effect did not increase with the time of single intermittent exposure. (3) Factors outside the magnetic field, such as stem cell type, differentiation stage, and cell density, also have complex effects on the biological effects of the magnetic field, which should be paid attention to in clinic. (4) Magnetic fields have effects on cellular targets such as plasma membrane, cytoskeleton and ion channel. Background: Sarcopenia has been defined as a progressive and generalized disorder of skeletal muscle that involves accelerated loss of muscle mass and function. The prevalence of sarcopenia worldwide is up to 15% in healthy elderly, approximately 76% of acutely hospitalized elderly patients, and up to 69% of patients admitted for post-acute geriatric rehabilitation. The loss of muscle mass results in serious consequences and several chronic diseases and senility, characterized by functional losses, muscle weakness, loss of independence and increased risk of falls and death. Sarcopenia has been defined as a progressive and generalized muscle disorder. Here we investigate the effects of PEMF in muscle strength and functional capabilities of elderly sarcopenic patients. Methods. Fifteen elderly people of both sexes, aged between 65 and 80 years old, classified according to the degree of muscle weakness. The participants were submitted to PEMF therapy in 12 sessions three times a week with the following parameters. In the first two sessions, only submaximal contraction was applied during 30 min with a frequency of 5 Hz at the sport mode of the equipment. In the following 10 sessions we used cycles of Submaximal contractions -- frequency of 5 Hz with 5s duration and Supramaximal contractions -- frequency of 30 Hz with 5s duration. The intensity of the equipment was adjusted according to the sensitivity of the patient and session duration was 30 min. Results. the TUG functional tests showed a significant reduction in execution time, with an initial average of 40 ± 10s to 22 ± 6s after treatment sessions. The average increase in muscle strength was 27 ± 9% after PEMF treatment. Conclusions. This case series report is the first in the world scientific literature to demonstrate the effectiveness of PEMF (Supramaximus™) therapy in gaining muscle strength and functionality in elderly sarcopenic patients. PEMF therapy, at least for the treatment of sarcopenia in the elderly, can be considered as the definitive evolution of electrical currents, with greater effectiveness and acceptance by the patient. Background: The Pulsed Electromagnetic Field (PEMF) technology has attracted increasing interest since consistent evidence of its therapeutic properties has been demonstrated to treat musculoskeletal conditions. However, this technology is not new, and has already been used in different experimental models and clinical studies for the treatment of tendinopathies, osteoarthritis, increased cell proliferation, bone consolidation, among others. Methods: In this work, we carried out an integrative review of clinical and experimental studies published in the last twenty years, on the available scientific evidence that demonstrate the effects of PEMF in different applications for health treatments. Five databases including Medline, Pubmed Central, Scopus, Lilacs and PEDro were searched for studies from 2001 to September 2022. The results were analyzed by the team of researchers and clinical professionals to assure methodological quality of the studies for the elaboration of the theoretical review on the effects of electromagnetic field stimulation on skeletal muscles, tendon or bones. Results: Sixty-two studies were included in this review, presenting evidence of the biological effects of PEMF that can suggest it´s possible use to treat different disorders. Conclusions: Pulsed electromagnetic fields (PEMF) present relevant clinical and experimental evidence of beneficial effects in the treatment of several musculoskeletal inflammatory disorders, such as tendinopathies and osteoarthritis, in addition to the treatment of urinary incontinence and abdominal diastasis. The low-frequency pulsed electromagnetic field (PEMF) may have possible cytoprotective effects against the destructive effects of oxidative stress. The goal was to investigate if shortterm low-frequency PEMF has cytoprotective effects in glioblastoma cell line following high-dose hydrogen peroxide (H2O2) treatment. U87-MG cells were divided into four groups: Sham-control group; PEMF group (cells exposed to PEMF); H2O2 group (cells treated with H2O2 at time intervals 30 min and 48 h, respectively); H2O2+PEMF group (cells exposed to PEMF after H2O2 treatment at time intervals 30 min and 48 h, respectively). The cell viability, levels of reactive oxygen species, glutathione peroxidase activity, and the amount of glutathione were measured. The cytoprotective effect of PEMF against deleterious effects of oxidative stress triggered by different time interval of H2O2 treatment might be mediated by the increase in the cell viability, the elevation in the antioxidant enzyme activity/amount, and the decrease in the reactive oxygen species level. In addition, the cytoprotective effect of PEMF varies depending on different time intervals of H2O2 treatment. In the light of these findings, further in vivo and/or in vitro studies on neurophysiological effects of PEMFs and their underlying molecular mechanisms are needed to elucidate neurotoxic or neuroprotective role against antioxidant defense mechanisms. Pulsed electromagnetic field (PEMF) therapy, a noninvasive treatment, has shown promise in mitigating nerve damage. However, unaccustomed exercises, such as eccentric contractions (ECCs), can damage both muscle and nerve tissue. This study investigated whether magnetic stimulation (MS) with PEMF could aid in nerve recovery after ECCs in the elbow flexors. Twenty participants were randomly assigned to either a control (CNT) or an MS group. Following ECCs, we measured the latency of the M-wave in the musculocutaneous nerve as an indicator of nerve function. Additionally, isometric torque, range of motion, and muscle pain were assessed for muscle function. Interestingly, only the CNT group exhibited a significant increase in latency on Day 2 (p < 0.05). The MS group, on the other hand, displayed an earlier recovery trend in isometric torque, range of motion, and muscle soreness. Notably, muscle soreness significantly decreased immediately after MS treatment compared to pretreatment levels. Objective: Relapse after orthodontic treatment remains a crucial problem. Pulsed electromagnetic fields (PEMFs) accelerate osteoblastogenesis and inhibit osteoclastogenesis. However, their effect on tooth movement during the retention phase of orthodontic treatment has not been studied. This study investigated the role of PEMF stimulation in preventing tooth relapse after orthodontic tooth movement (OTM) in rat models., Methods: Thirty-six male Wistar rats were divided into control, PEMF 7, and PEMF 14 groups. The maxillary first molar was moved mesially with a 50 g force of a Nickel Titanium closed coil spring for 21 days. Therefore, PEMF stimulations, including a frequency of 15 Hz and intensity of 2.0 mT, were applied to a retention phase for 2 h daily for 7 and 14 days. The tooth relapse distance was evaluated on days 1, 3, 7, and 14; the number of osteoblasts, osteoclasts, and fibroblasts was assessed by hematoxylin and eosin staining; and the expression of fibroblast growth factor 2 (FGF-2) and type I collagen (Col-I) was evaluated by immunohistochemistry. The data were analyzed using one-way analysis of variance and post hoc test with p < 0.05 considered statistically significant., Results: Tooth relapse distance was significantly decreased in the PEMF 7 and PEMF 14 groups compared to the control group. Aims: Distraction osteogenesis (DO) is a useful orthopaedic procedure employed to lengthen and reshape bones by stimulating bone formation through controlled slow stretching force. Despite its promising applications, difficulties are still encountered. Our previous study demonstrated that pulsed electromagnetic field (PEMF) treatment significantly enhances bone mineralization and neovascularization, suggesting its potential application. The current study compared a new, high slew rate (HSR) PEMF signal, with different treatment durations, with the standard Food and Drug Administration (FDA)-approved signal, to determine if HSR PEMF is a better alternative for bone formation augmentation. Methods: The effects of a HSR PEMF signal with three daily treatment durations (0.5, one, and three hours/day) were investigated in an established rat DO model with comparison of an FDA-approved classic signal (three hrs/day). PEMF treatments were applied to the rats daily for 35 days, starting from the distraction phase until termination. Radiography, micro-CT (μCT), biomechanical tests, and histological examinations were employed to evaluate the quality of bone formation. Results: All rats tolerated the treatment well and no obvious adverse effects were found. By comparison, the HSR signal (three hrs/day) treatment group achieved the best healing outcome, in that endochondral ossification and bone consolidation were enhanced. In addition, HSR signal treatment (one one hr/day) had similar effects to treatment using the classic signal (three three hrs/day), indicating that treatment duration could be significantly shortened with the HSR signal. Conclusion: HSR signal may significantly enhance bone formation and shorten daily treatment duration in DO, making it a potential candidate for a new clinical protocol for patients undergoing DO treatments. Background: Temporomandibular joint osteoarthritis (TMJOA) is characterized by abnormal subchondral bone remodeling and cartilage degeneration. As a non-invasive biophysical technology, pulsed electromagnetic field (PEMF) treatment has been proven to be efficient in promoting osteogenesis. However, the potential bone protective effect and mechanism of PEMF on abnormal subchondral bone remodeling in TMJOA are unknown.Methods: Unilateral anterior crossbite (UAC) was used to create TMJOA model in rats, and 17β-estradiol (E2) were injected daily to mimic patients with high-physiological levels of estrogen. Mouse osteoblast-like MC3T3-E1 cells treated with recombinant murine IL-1β was used to establish inflammatory environment in vitro. The treatment group were subjected to PEMF (2.0mT, 15 Hz, 2 h/d). Micro-CT scanning, histological staining, real-time PCR and western blotting assays were preformed to observe the changes in the subchondral bone.Results: Abnormal resorption of subchondral bone induced by UAC, characterized by decreased bone mineral density, increased osteoclast activity and expression of osteoclast-related factors (RANKL) and down-regulated expression of osteogenesis-related factors (OPG, ALP, Runx2 and OCN) at the early stage, could be reversed by PEMF exposure, which was similar to the effect of estrogen. In addition, PEMF exposure and E2 supplement may have a synergistic effect to some extent. Moreover, PEMF exposure could promote the ALP activity and osteogenic mineralization ability of MC3T3-E1 cells. PEMF promoted the expression of factors related to Wnt/β-Catenin signal pathway both in vivo and in vitro.Conclusions: Appropriate PEMF exposure have a protective effect on subchondral bone in TMJOA at early stage, in which canonical Wnt/β-Catenin pathway may be involved. Objective Physical factors have been used to address disuse osteoporosis, but their effects and mechanism remain unclear. The purpose of this study was to determine the effects of pulsed electromagnetic field (PEMF) and whole-body vibration (WBV) on disuse osteoporosis to increase knowledge about treating osteoporosis. Methods A disuse osteoporosis rat model was developed by hind-limb unloading (HU) for 6 weeks. Forty 4-month-old female Sprague–Dawley rats were divided into 5 groups and given the following interventions: HU, HU treated with PEMF (HUP), HU treated with WBV (HUW), HU treated with both PEMF and WBV (HUPW), and no intervention (controls). After 8 weeks of intervention, measurements were taken. Results HU induced a decrease in bone mineral density (BMD), whereas HUP, HUW, and HUPW increased it. Moreover, the bone resorption markers tartrate-resistant acid phosphatase (TRAP) and C-terminal peptide of type 1 collagen in the HU group significantly increased, whereas the osteogenesis markers osteocalcin and N-terminal propeptide of type 1 procollagen significantly decreased. The markers osteocalcin and N-terminal propeptide of type 1 procollagen significantly increased, but TRAP and C-terminal peptide of type 1 collagen significantly decreased in the HUPW, HUP, and HUW groups compared with the HU group. In particular, HUPW effectively increased osteocalcin and decreased TRAP compared with HUP and WBV. Microcomputed tomography analysis of the femur indicated that HUPW improved trabecular number, bone volume over total volume, bone surface over bone volume, trabecular separation, and the structure model index compared with HUP and that it improved bone surface over bone volume, trabecular separation, and structure model index compared with HUW. The HUPW group showed a significant increase in maximum load compared with the HUW group and a significant increase in elastic modulus compared with the HUP group. Conclusion PEMF, WBV, and their combination all attenuated bone resorption and enhanced osteogenesis. WBV and the combination of treatments have great potential to improve osteogenesis compared with PEMF. In addition, HUPW significantly attenuated bone resorption compared with HUW and HUP. Impact The results of this study indicated that HUPW could effectively improve disuse osteoporosis compared with HUP, given that trabecular number and bone volume over total volume are associated with disuse osteoporosis. Moreover, BMD recovered well with HUP, HUW, and HUPW but the bone structure—especially mechanical performance—did not, indicating that osteoporosis should be evaluated with BMD and mechanical performance, not with BMD in isolation. To use contemporary welded transport structures made of alloys based on aluminum, it is necessary to develop new promising methods aimed at extending their service life. As one of these methods, we can mention the procedure of electrodynamic treatment (EDT) aimed at increasing the durability of structures by optimizing the stress-strain state in the zone of expected fatigue fracture. By applying the microprocessor control in the process of two-channel electrodynamic treatment, we can increase the efficiency of treatment due to the controlled synchronization of the components of electrodynamic actions as compared with the efficiency of single-channel treatment. In the course of fatigue testing of the experimental welded samples of AMg6 aluminum alloy for a cyclic amplitude 2σa = 80–160 MPa, both electrodynamic treatments make their durability N 2.0–2.4 and 2.2–2.7 times higher, respectively, despite the fact that the two-channel treatment is realized with a threefold lower power consumption than the single-channel treatment. Due to the fact that the passage of current pulses through an experimental sample in the course of two-channel electrodynamic treatment becomes impossible, the parameter N becomes 1.5 times lower than in the case of electrodynamic treatment performed with the passage of current; this is connected with the absence of the influence of pulsed electric component, which intensifies the relaxation of residual welding stresses caused by electroplasticity. The two-channel electrodynamic treatment of the weld and the line of fusion doubles the value of N as compared with the corresponding value obtained for EDT solely of the weld. This is explained by the propagation of the zone of compressive stresses to the line of fusion, which results in a smaller range of cyclic stresses than in the case of EDT solely of the weld. Objective: This study investigated nonablative/noncoagulative multipolar radiofrequency and pulsed electromagnetic field (RF/PEMF) treatment for vaginal laxity (VL) and its impact on sexual function in parous women. Methods: This prospective, open-label single-center study enrolled 34 female subjects, 23?59 years of age, with ?1 vaginal delivery and self-reported VL. Three monthly intravaginal treatments with RF/PEMF energy were performed. Treatment and follow-up assessments included the vaginal health index (VHI), vaginal pH, female sexual function index (FSFI), and VL/sexual satisfaction (SS) and subject satisfaction scores. Mean score and percent improvement over baseline were reported. Subject discomfort/pain was assessed after each treatment. Results: Total and each individual domain scores of the VHI improved significantly, while vaginal pH levels decreased from baseline to both 1 and 4 months (p?0.01) after the last treatment. FSFI (<0.001), VL (<0.001), and SS (<0.001), including overall satisfaction scores (<0.01), improved post-treatment, with positive effects further sustained until at least 4 months post-treatment. Pain/discomfort post-treatment was reported as none to mild. No noticeable adverse events (AEs) or unanticipated side effects were reported. Conclusions: Nonablative/noncoagulative multipolar RF/PEMF is safe and is associated with significant 1- and 4-month post-treatment improvements in symptoms associated with VL and sexual dysfunction, as assessed by the VHI, vaginal pH, FSFI, and VL subject satisfaction score. SS and overall satisfaction scores also improved. The treatment was well tolerated with no or little pain, and no adverse events were reported. Clinical Trial Registration number: NCT04607798.

Development of procedures which accelerate osseointegration of dental implants, reduce the period of healing, and lead to an early rehabilitation of the patient are required for successful oral rehabilitation. Pulsed electromagnetic field (PEMF) is a noninvasive, therapeutic form of low field magnetic stimulation that has been used for healing bone non unions and various fractures. It acts on osteoblasts and bone, affecting their metabolism, therefore, increasing the tissue integration of the implanted devices and their clinical success. A broad range of settings that includes magnetic field intensity, frequency and duration of application, etc. used for PEMFs stimulation is a hurdle to properly define treatment protocols and extensive research is needed to overcome this issue. The present review includes studies that investigated the effects of PEMFs on the response of bone cells to different classes of biomaterials and the reports that focused on in vivo and in vitro investigations of biomaterials implanted in bone. This study is expected to serve as a guide for researchers and clinicians to bring into their clinical use these strategies to improve implant osseointegration in deficient and osteoporotic bone. Rotator cuff repair failure remains common due to poor tendon healing, particularly at the enthesis. We previously showed that pulsed electromagnetic field (PEMF) therapy improved the mechanical properties of the rat supraspinatus tendon postoperatively. However, little is known about the mechanisms behind PEMF‐dependent contributions to improved healing in this injury model. The objective of this study was to determine the influence of PEMF treatment on tendon gene expression and cell composition, as well as bone microarchitecture and dynamic bone metabolism during early stages of healing. We hypothesized that PEMF treatment would amplify tendon‐healing related signaling pathways while mitigating inflammation and improve bone metabolism at the repair site. Rats underwent rotator cuff injury and repair followed by assignment to either control (non‐PEMF) or PEMF treatment groups. Gene and protein expression as well as tendon and bone histological assessments were performed 3, 7, 14, 21, and 28 days after injury. Gene expression data demonstrated an upregulation in the bone morphogenetic protein 2 signaling pathway and increases in pro‐osteogenic genes at the insertion, supporting important processes to re‐establish the tendon‐bone interface. PEMF also downregulated genes related to a fibrotic healing response. Anti‐inflammatory effects were demonstrated by both gene expression and macrophage phenotype. PEMF significantly increased the rate of kinetic bone formation directly adjacent to the tendon enthesis as well as the number of cuboidal surface osteoblasts (active osteoblasts) in the humeral head. Microbial infection is a major barrier to wound care and repair process of the cells. Various US-FDA-approved drugs and surgical procedures have been used for wound healing progression. However, the use of these pharmaceutical products has an associated toxicity and may produce harmful side effects. To overcome these problems, researchers worldwide are trying to develop non-invasive wound healing processes for wound care. We have developed a unique, tunable and Arduino microcontroller-based extremely low-frequency (ELF) pulsed electromagnetic field (PEMF) device for accelerating the wound healing process by reducing the microbial infection. The layout and detailed design of the device are presented here. Simulations carried out for the fabrication of the Helmholtz coil used in the PEMF device are also presented. This system can provide the flexibility of dose substance by selecting the type and frequency of the waveform, exposure duration, and magnetic field strength. To test the efficacy of the device in the biological system, we have designed colony-forming unit assay in Staphylococcus aureus with an exposure to PEMF in the frequency range of 5–30 Hz for 90 min. The experiments have been conducted in controlled EMF environments as per the international standard exposure guidelines. The results show a 60% reduction in S. aureus CFU after 90 minutes of PEMF exposure, and hence, show that the designed system is very effective for wound care by reducing the microbial growth. BACKGROUND: Equine superficial digital flexor (SDF) tendonitis is a very common cause of lameness in athletic horses inducing long lameness and prolonged recovery. Multiple treatment protocols have been proposed for this injury none of which have been able to solve the problem completely. Therefore, an affordable, widely available, and non-invasive thera-peutic protocol is sought in the horse industry. OBJECTIVES: The present study aimed to compare the use of topical dimethyl sulfoxide (DMSO) with a combination of topical DMSO and pulsed electromagnetic field (PEMF). METHODS: The subjects were divided into two groups of five polo ponies. The animals in the control group were treated by topical DMSO and controlled exercise, while the ponies in the experiment group received the same protocol plus PEMF on the injured tendon. RESULTS: The ultrasonographic factors evaluated in the current study were mostly similar between the two groups except for fiber alignment and echogenicity. The latter variables were shown to have a slightly better improvement in the test group indicating the minimal positive effects of PEMF. In clinical evaluation, both groups were almost identical sug-gesting no impacts for PEMF. CONCLUSIONS: In conclusion, applying PEMF using different protocols, such as more extended application at differ-ent frequencies is deemed necessary to elicit a favorable outcome of PEMF for the treatment of SDF tendonitis.

Skeletal muscle tissue engineering (SMTE) aims at the in vitro generation of 3D skeletal muscle engineered constructs which mimic the native muscle structure and function. Although native skeletal muscle is a highly dynamic tissue, most research approaches still focus on static cell culture methods, while research on stimulation protocols indicates a positive effect, especially on myogenesis. A more mature muscle construct may be needed especially for the potential applications for regenerative medicine purposes, disease or drug disposition models. Most efforts towards dynamic cell or tissue culture methods have been geared towards mechanical or electrical stimulation or a combination of those. In the context of dynamic methods, pulsed electromagnetic field (PEMF) stimulation has been extensively used in bone tissue engineering, but the impact of PEMF on skeletal muscle development is poorly explored. Here, we evaluated the effects of PEMF stimulation on human skeletal muscle cells both in 2D and 3D experiments. First, PEMF was applied on 2D cultures of human myoblasts during differentiation. In 2D, enhanced myogenesis was observed, as evidenced by an increased myotube diameter and fusion index. Second, 2D results were translated towards 3D bioartificial muscles (BAMs). BAMs were subjected to PEMF for varying exposure times, where a 2-h daily stimulation was found to be effective in enhancing 3D myotube formation. Third, applying this protocol for the entire 16-days culture period was compared to a stimulation starting at day 8, once the myotubes were formed. The latter was found to result in significantly higher myotube diameter, fusion index, and increased myosin heavy chain 1 expression. This work shows the potential of electromagnetic stimulation for enhancing myotube formation both in 2D and 3D, warranting its further consideration in dynamic culturing techniques.

Orthopedic fractures have a significant impact on patients in the form of economic loss and functional impairment. Beyond the standard methods of reduction and fixation, one adjunct that has been explored since the late 1970s is electrical stimulation. Despite robust evidence for efficacy in the preclinical arena, human trials have mixed results, and this technology is not widely accepted. The purpose of this review is to examine the body of literature supporting electrical stimulation for the purpose of fracture healing in humans with an emphasis on device specifications and stimulation protocols and delineate a minimum reporting checklist for future studies of this type. We have isolated 12 studies that pertain to the administration of electrical stimulation for the purpose of augmenting fracture healing in humans. Of these, one was a direct current electrical stimulation study. Six studies utilized pulsed electromagnetic field therapy and five used capacitive coupling. When examining these studies, the device specifications were heterogenous and often incomplete in what they reported, which rendered studies unrepeatable. The stimulation protocols also varied greatly study to study. To demonstrate efficacy of electrical stimulation for fractures, the authors recommend isolating a fracture type that is prone to nonunion to maximize the electrical stimulation effect, a homogenous study population so as to not dilute the effect of electrical stimulation, and increasing scientific rigor in the form of pre-registration, blinding, and sham controls. Finally, we introduce the critical components of minimum device specification reporting for repeatability of studies of this type.
