Abstract Optical vortices, featured with an infinite number of orthogonal channels of orbital angular momentum, have demonstrated marvelous potentials in optical multiplexing and associated applications. However, conventional vortex beams with global phase modulation approach usually possess a single topological charge (TC) and a uniform radial distance with the donut-shaped intensity, leaving unlimited spatial intensity information unexplored. Here, to break the spatial capacity limitation, we introduce an entirely new concept of a spatial-frequency patching metasurface by patching the field distribution piece-by-piece in the spatial-frequency domain, thereby breaking the symmetry of the beam morphology and allowing for local manipulation of spatial intensity and TC distributions. Moreover, by superimposing two orthogonal circular polarized perfect VBs, our breakthrough offers a super-capacity with at least 13 channels across a 3D parametric space, including morphology, polarization azimuth and ellipticity angle, namely super-capacity perfect vector vortex beams (SC-PVVBs). Furthermore, we have designed an optimized Dammann grating to facilitate an array of SC-PVVBs, thereby unleashing the full potentials across 13 channels/bits for multi-dimensional complex information communications. Our findings promise dense data transmission in an ultra-secure manner using VBs, opening up new avenues in super-capacity optical information technology in an integrated metasurface platform.

Photothermal therapy is a promising tumor treatment approach that selectively eliminates cancer cells while assuring the survival of normal cells. It transforms light energy into thermal energy, making it gentle, targeted, and devoid of radiation. However, the efficacy of treatment is hampered by the absence of accurate and noninvasive temperature measurement method in the therapy. Therefore, there is a pressing demand for a noninvasive temperature measurement method that is real-time and accurate. This article presents one such attempt based on thermal strain photoacoustic (PA) temperature measurement. The method was first modelled, and a circular array-based photoacoustic photothermal system was developed. Experiments with Indian ink as tumor simulants suggest that the temperature monitoring in this work achieves a precision of down to 0.3 °C. Furthermore, it is possible to accomplish real-time temperature imaging, providing accurate two-dimensional temperature mapping for photothermal therapy. Experiments were also conducted on human fingers and nude mice, validating promising potentials of the proposed method for practical implementations.

Optical encryption is pivotal in information security, offering parallel processing, speed, and robust security. The simplicity and compatibility of speckle‐based cryptosystems have garnered considerable attention. Yet, the predictable statistical distribution of speckle optical fields’ characteristics can invite statistical attacks, undermining these encryption methods. The proposed solution, a deep adversarial learning‐based speckle modulation network (DeepSLM), disrupts the strong intercorrelation of speckle grains. Utilizing the unique encoding properties of speckle patterns, DeepSLM facilitates license editing within the modulation phase, pioneering a layered authentication encryption system. Our empirical studies confirm DeepSLM's superior performance on key metrics. Notably, the testing dataset reveals an average Pearson correlation coefficient above 0.97 between decrypted images and their original counterparts for intricate subjects like human faces, attesting to the method's high fidelity. This innovation marries adjustable modification, optical encryption, and deep learning to enforce tiered data access control, charting new paths for creating user‐specific access protocols.

Abstract Artificial intelligence has gained significant attention for exploiting optical scattering for optical encryption. Conventional scattering media are inevitably influenced by instability or perturbations, and hence unsuitable for long-term scenarios. Additionally, the plaintext can be easily compromised due to the single channel within the medium and one-to-one mapping between input and output. To mitigate these issues, a stable spin-multiplexing disordered metasurface (DM) with numerous polarized transmission channels serves as the scattering medium, and a double-secure procedure with superposition of plaintext and security key achieves two-to-one mapping between input and output. In attack analysis, when the ciphertext, security key, and incident polarization are all correct, the plaintext can be decrypted. This system demonstrates excellent decryption efficiency over extended periods in noisy environments. The DM, functioning as an ultra-stable and active speckle generator, coupled with the double-secure approach, creates a highly secure speckle-based cryptosystem with immense potentials for practical applications.

Abstract To deal with intra‐abdominal sepsis, one of the major global causes of death in hospitalized patients, efficient abscess drainage is crucial. Despite decades of advances, traditional catheters have demonstrated poor drainage and absorption properties due to their simple tubular structures and their dense nonporous surface. Herein, inspired by porous sponges and fractal roots, a multifaceted hydrogel catheter with effective drainage, absorptive, and robust properties, is presented. Its unique fractal structures provide extensive internal branching and a high specific surface area for effective drainage, while the hierarchical porous structures provide a wide range of absorption capabilities. Additionally, its distinctive multi‐interpenetration network maintains robust and appropriate mechanical properties, even after absorption multiple times of liquid and mechanical disturbance, allowing for intact removal from the abdominal cavity without harm to the animal in vivo. Besides, the loaded antimicrobial peptides are capable of being released in situ to inhibit the potential for infections. In vivo experiments have demonstrated that this hydrogel catheter efficiently removes lethal abscesses and improves survival. It is believed that this innovative and practical catheter will create a future precedent for hydrogel drainage devices for more effective management of intra‐abdominal sepsis.

Wound healing introduces a series of interesting challenges in healthcare, such as open wounds, burn wounds, and chronic infection wounds. Hence more accurate and effective biomedical materials are expected, for which emerging hydrogels with unique and excellent properties have demonstrated great potential. When they are applied as wound dressings, tissue patches, plugging agent, drug carriers, and biosensors, etc., there are urgent requirements for adhesive capabilities. The existence of interfacial water, soft tissue and moving surfaces, safety, and harmlessness, however, constitutes major constraints for medical adhesive hydrogels. In nature, organisms often adhere through unique structures or specific chemical components, which stimulates inspirations for medical hydrogels to overcome the above challenges. In this review, we will classify adhesive hydrogels into glue-like adhesives and tape-like patches according to their form. First, this review will introduce the principles and design ideas of biomedical adhesive hydrogels. Then, as biomedical materials, the applications and effects of their interactions with organisms through adhesion will be summarized. Finally, this review will summarize the achievements, challenges, and future development of bio-inspired adhesive hydrogel towards wound healing.

Abstract A communication channel should be built to transmit information from one place to another. Imaging is 2 or higher dimensional information communication. Conventionally, an imaging channel comprises a lens with free space at its both sides, whose transfer function is usually known and hence the response of the imaging channel can be well defined. Replacing the lens with a thin scattering medium, the image can still be extracted from the detected optical field, suggesting that the scattering medium retains or reconstructs not only energy but also information transmission channels. Aided by deep learning, we find that unlike the lens system, there are different channels in a scattering medium: the same scattering medium can construct different channels to match the manners of source coding. Moreover, it is found that without a valid channel, the convolution law for a spatial shift-invariant system (the output is the convolution of the point spread function and the input object) is broken, and in this scenario, information cannot be transmitted onto the detection plane. Therefore, valid channels are essential to transmit information through even a spatial shift-invariant system. These findings may intrigue new adventures in imaging through scattering media and reevaluation of the known spatial shift-invariance in various areas.

Abstract Many hydrogel patches are developed to solve the pervasive and severe challenge of complex wound healing, while most of them still lack satisfactory controllability and comprehensive functionality. Herein, inspired by multiple creatures, including octopuses and snails, a novel muti‐functional hydrogel patch is presented with controlled adhesion, antibacterial, drug release features, and multiple monitoring functions for intelligent wound healing management. The patch with micro suction‐cup actuator array and a tensile backing layer is composed of tannin grafted gelatin, Ag‐tannin nanoparticles, polyacrylamide (PAAm) and poly(N‐isopropylacrylamide) (PNIPAm). In virtue of the photothermal gel‐sol transition of tannin grafted gelatin and Ag‐tannin nanoparticles, the patches exert a dual anti‐microbial effect and temperature‐sensitive snail mucus‐like features. In addition, as the “suction‐cups” consisting of thermal responsive PNIPAm can undergo a contract‐relax transformation, the medical patches can adhere to the objects reversibly and responsively, and release their loaded vascular endothelial growth factor (VEGF) controllably for wound healing. More attractively, benefiting from their fatigue resistance, self‐healing ability of the tensile double network hydrogel, and electrical conductivity of Ag‐tannin nanoparticles, the proposed patches can report multiple wound physiology parameters sensitively and continuously. Thus, it is believed that this multi‐bioinspired patch has immense potential for future wound healing management.

Abstract Triboelectric nanogenerators (TENGs) are new energy collection devices that have the characteristics of high efficiency, low cost, miniaturization capability, and convenient manufacture. TENGs mainly utilize the triboelectric effect to obtain mechanical energy from organisms or the environment, and this mechanical energy is then converted into and output as electrical energy. Bioelectricity is a phenomenon that widely exists in various cellular processes, including cell proliferation, senescence, apoptosis, as well as adjacent cells’ communication and coordination. Therefore, based on these features, TENGs can be applied in organisms to collect energy and output electrical stimulation to act on cells, changing their activities and thereby playing a role in regulating cellular function and interfering with cellular fate, which can further develop into new methods of health care and disease intervention. In this review, we first introduce the working principle of TENGs and their working modes, and then summarize the current research status of cellular function regulation and fate determination stimulated by TENGs, and also analyze their application prospects for changing various processes of cell activity. Finally, we discuss the opportunities and challenges of TENGs in the fields of life science and biomedical engineering, and propose a variety of possibilities for their potential development direction.

Abstract Early diagnosis can effectively improve the survival of glioblastoma multiforme (GBM). A specific imaging technique that is simultaneously deep penetrating and sensitive to small tissue changes is desired to identify GBM. Due to its excellent features in signal contrast, detection sensitivity, and none or little attenuation in tissue, magnetic particle imaging (MPI) possesses great potential in cancer diagnosis, especially when the imaging modality is equipped with specifically targeted nanoprobes. However, when gliomas are small, the blood–brain barrier (BBB) is complete and prevents nanoprobes from entering the brain, which negates the theranostic effect. This study proposes a biomimetic nanoplatform that assist the MPI tracers in breaking through the BBB and then demonstrate a targeted and sensitive diagnosis of GBM. Afterward, the photothermal therapy and immune regulation show an excellent therapeutic effect on the GBM. It is experimentally confirmed that the MPI signal does not decay with tissue depth and shows excellent sensitivity for thousands‐cells. Only small animals are conducted in this study due to the limitations of the current commercial MPI scanner, however, this research theoretically enables large animal and human studies, which encourages a promising pathway toward the noninvasive diagnosis of early‐stage GBM in clinics.

Abstract High-resolution optical imaging through or within thick scattering media is a long sought after yet unreached goal. In the past decade, the thriving technique developments in wavefront measurement and manipulation do not significantly push the boundary forward. The optical diffusion limit is still a ceiling. In this work, we propose that a scattering medium can be conceptualized as an assembly of randomly packed pinhole cameras and the corresponding speckle pattern as a superposition of randomly shifted pinhole images. The concept is demonstrated through both simulation and experiments, confirming the new perspective to interpret the mechanism of information transmission through scattering media under incoherent illumination. We also analyze the efficiency of single-pinhole and dual-pinhole channels. While in infancy, the proposed method reveals a new perspective to understand imaging and information transmission through scattering media.

Abstract The adhesiveness of hydrogels is urgently required in various biomedical applications such as medical patches, tissue sealants, and flexible electronic devices. However, biological tissues are often wet, soft, movable, and easily damaged. These features pose difficulties for the construction of adhesive hydrogels for medical use. In nature, organisms adhere to unique strategies, such as reversible sucker adhesion in octopuses and nontoxic and firm catechol chemistry in mussels, which provide many inspirations for medical hydrogels to overcome the above challenges. In this review, we systematically classify bioadhesion strategies into structure‐related and molecular‐related ones, which cover almost all known bioadhesion paradigms. We outline the principles of these strategies and summarize the corresponding designs of medical adhesive hydrogels inspired by them. Finally, conclusions and perspectives concerning the development of this field are provided. For the booming bio‐inspired adhesive hydrogels, this review aims to summarize and analyze the various existing theories and provide systematic guidance for future research from an innovative perspective.

Optical techniques offer a wide variety of applications as light-matter interactions provide extremely sensitive mechanisms to probe or treat target media. Most of these implementations rely on the usage of ballistic or quasi-ballistic photons to achieve high spatial resolution. However, the inherent scattering nature of light in biological tissues or tissue-like scattering media constitutes a critical obstacle that has restricted the penetration depth of non-scattered photons and hence limited the implementation of most optical techniques for wider applications. In addition, the components of an optical system are usually designed and manufactured for a fixed function or performance. Recent advances in wavefront shaping have demonstrated that scattering- or component-induced phase distortions can be compensated by optimizing the wavefront of the input light pattern through iteration or by conjugating the transmission matrix of the scattering medium. This offers unprecedented opportunities in many applications to achieve controllable optical delivery or detection at depths or dynamically configurable functionalities by using scattering media to substitute conventional optical components. In this article, the recent progress of wavefront shaping in multidisciplinary fields is reviewed, from optical focusing and imaging with scattering media, functionalized devices, modulation of mode coupling, and nonlinearity in multimode fiber to multimode fiber-based applications. Apart from insights into the underlying principles and recent advances in wavefront shaping implementations, practical limitations and roadmap for future development are discussed in depth. Looking back and looking forward, it is believed that wavefront shaping holds a bright future that will open new avenues for noninvasive or minimally invasive optical interactions and arbitrary control inside deep tissues. The high degree of freedom with multiple scattering will also provide unprecedented opportunities to develop novel optical devices based on a single scattering medium (generic or customized) that can outperform traditional optical components.

Abstract Face recognition has become ubiquitous for authentication or security purposes. Meanwhile, there are increasing concerns about the privacy of face images, which are sensitive biometric data and should be protected. Software‐based cryptosystems are widely adopted to encrypt face images, but the security level is limited by insufficient digital secret key length or computing power. Hardware‐based optical cryptosystems can generate enormously longer secret keys and enable encryption at light speed, but most reported optical methods, such as double random phase encryption, are less compatible with other systems due to system complexity. In this study, a plain yet highly efficient speckle‐based optical cryptosystem is proposed and implemented. A scattering ground glass is exploited to generate physical secret keys of 17.2 gigabit length and encrypt face images via seemingly random optical speckles at light speed. Face images can then be decrypted from random speckles by a well‐trained decryption neural network, such that face recognition can be realized with up to 98% accuracy. Furthermore, attack analyses are carried out to show the cryptosystem's security. Due to its high security, fast speed, and low cost, the speckle‐based optical cryptosystem is suitable for practical applications and can inspire other high‐security cryptosystems.

Iterative wavefront shaping is a powerful tool to overcome optical scattering and enable the focusing of diffusive light, which has exciting potential in many applications that desire localized light delivery at depths in tissue-like complex media. Unsatisfactory performance and efficiency, however, have been a long-standing problem, and the large discrepancy between theoretical and experimental results has hindered the wide applications of the technology. Currently, most algorithms guiding the iterative search for optimum phase compensation rely heavily on randomness to achieve solution diversity. It is similar to black-box optimization, in which the mechanism for arriving at a good solution is unclear. The lack of clear guidance on the new solution generation process considerably affects the efficiency of optimization. Therefore, we propose a probability-based iterative algorithm that combines the genetic algorithm and ant colony optimization to develop new solutions based on a probability map. Thanks to the clearer guidance provided by the probability map and the reduced involvement of randomness, we can obtain optimization results with optimal efficiency for single and multiple focuses behind scattering media. In addition, with the proposed algorithm, we also demonstrate higher adaptability in an unstable scattering environment and more spatially uniform optical focusing in the field of view. This study advances the state-of-the-art in the practice of iterative wavefront shaping. More importantly, the significant improvement in optimization efficiency and adaptability, if further engineered, can potentially inspire or open up wide applications that desire localized and enhanced optical delivery in situ.

Optical-resolution photoacoustic microscopy (OR-PAM) enjoys superior spatial resolution and has received intense attention in recent years. The application, however, has been limited to shallow depths because of strong scattering of light in biological tissues. In this work, we propose to achieve deep-penetrating OR-PAM performance by using deep learning enabled image transformation on blurry living mouse vascular images that were acquired with an acoustic-resolution photoacoustic microscopy (AR-PAM) setup. A generative adversarial network (GAN) was trained in this study and improved the imaging lateral resolution of AR-PAM from 54.0 µm to 5.1 µm, comparable to that of a typical OR-PAM (4.7 µm). The feasibility of the network was evaluated with living mouse ear data, producing superior microvasculature images that outperforms blind deconvolution. The generalization of the network was validated with in vivo mouse brain data. Moreover, it was shown experimentally that the deep-learning method can retain high resolution at tissue depths beyond one optical transport mean free path. Whilst it can be further improved, the proposed method provides new horizons to expand the scope of OR-PAM towards deep-tissue imaging and wide applications in biomedicine.

Optogenetics is proved to be a powerful tool for exploring the connection between behavior and neural circuits. The progress greatly benefits from the advances of optical techniques that enable high spatiotemporal resolution for selective single‐neuron stimulation through modulating light. Efficient propagation of modulated light, however, is handicapped by strong optical scattering in biological tissues, which results in inherent tradeoff between penetration depth and resolution. Schemes like graded index (GRIN) lens‐based microendoscopes are developed to yield more confined delivery of light, but tissue damage caused by the insertion of the bulky components cannot be ignored. Herein, an optically selective precise neuron stimulation using an ultrathin multimode fiber (MMF) is demonstrated, which is empowered by optical wavefront shaping to achieve light focusing and rapid raster scanning without mechanical movement at the distal end of the MMF and even through a mouse skull. With this method, primary neurons expressing Chr2 can be regulated spatiotemporally in experiment. Although a lot shall be further improved, the work may open up new venues for noninvasive or minimally invasive all‐optical investigation of neural circuits in used‐to‐be optically inaccessible brain regions.

Existing acoustic-resolution photoacoustic/ultrasonic endoscopy (PA/USE) generally employs a point-focused transducer for ultrasound detection, which is only sensitive in its focal region, thus the lateral resolution and sensitivity drop dramatically when the targets move far from its focus. Even if a dynamic focusing algorithm is applied, the sensitivity out of the transducer focus is still much lower than that in the focus in ultrasonic imaging mode. In this work, we propose an acoustic-resolution PA/USE with a line-focused transducer to realize automatic focusing for the first time. In comparison to a point-focused transducer, the line-focused transducer emits a more uniform sound field, causing the original signal intensity and signal-to-noise ratio (SNR) of the front and rear targets to be closer in the radial direction, which is beneficial for improving target signal uniformity in ultrasonic imaging. Simultaneously, we improved the resolution of the defocus area by modifying a prior work of back-projection (BP) reconstruction algorithm typically used in point-focused transducer based PAE and applying it to line-focused PA/USE. This combined approach may significantly enhance the depth of field of ultrasonic imaging and the resolution of the defocus zone in PA/US imaging, compared to the conventional method. Sufficient numerical simulations and phantom experiments were performed to verify this method. The results show that our method can effectively improve the lateral resolution in the image’s defocused region to achieve automatic focusing and perfectly solve the defect of the target signal difference in the far-focus region in ultrasonic imaging, while also enhancing the image SNR and contrast. The proposed method in this paper lays foundations for the realization of photoacoustic/ultrasonic combined endoscopy with enhanced lateral resolution and depth of field, which can potentially benefit a many of biomedical applications.

Intravenous injection is a rapid, low-cost, and direct method that is commonly used to deliver multifarious biotherapeutics and vaccines. However, intravenous injection often causes trauma or tissue injury that requires professional operation. Transdermal drug delivery overcomes the aforementioned defects, and the microneedles (MNs) array is one of the most promising transdermal drug delivery platforms. Timely, precise, and non-invasive monitoring and evaluation of the effects of MNs in transdermal administration is significant to the research of drug efficiency response to specific diseases. In this sense, photoacoustic computed tomography (PACT), which provides wavelength-selective and deep-penetrating optical contrast, could be a promising imaging tool for in situ evaluation of the treatment effects. In this work, we propose the use of PACT to non-invasively assess the effects of real-time drug delivery in glioma tumors through transdermal administration with degradable indocyanine green-loaded hyaluronic acid MNs (ICG-HA-MNs). The outcome is systematically and quantitatively compared with that via intravenous injection. It is found that the photoacoustic signals of ICG in the tumor site express a faster elevation and shorter duration time in the intravenous injection group; by contrast, the photoacoustic signals demonstrate a lower intensity but prolonged duration time in the MNs group. The observed phenomenon indicates faster response but shorter drug duration for intravenous injection, which is in contrast with the lower loading but prolonged performance for transdermal drug delivery with MNs. These results exhibit good consistency with the earlier, common-sense findings reported from other aspects, confirming that PACT can serve as a potential imaging tool to precisely, non-invasively, and quickly evaluate in situ drug delivery effects and provide constructive guidance for the design and fabrication of microneedles.

Abstract Time-gated reflection matrix (RM) has been successfully used for optical imaging deep inside scattering media. Recently, this method was extended to enhance the spatiotemporal focusing of light ultra-deep inside scattering media. This is achieved by calibrating the decomposition of the RM with the Tikhonov regularization parameter to convert multiply scattered photons that share the same time of flight with the singly scattered photons into singly scattered photons. Such a capability suggests a reshaping to the interaction mechanism between light and scattering media, which may benefit or inspire wide optical applications that desire enhanced spatiotemporal focusing of light at depths inside scattering media.

Graphene derivatives, possessing strong Raman scattering and near-infrared absorption intrinsically, have boosted many exciting biosensing applications. The tunability of the absorption characteristics, however, remains largely unexplored to date. Here, we proposed a multilayer configuration constructed by a graphene monolayer sandwiched between a buffer layer and one-dimensional photonic crystal (1DPC) to achieve tunable graphene absorption under total internal reflection (TIR). It is interesting that the unique optical properties of the buffer-graphene-1DPC multilayer structure, the electromagnetically induced transparency (EIT)-like and Fano-like absorptions, can be achieved with pre-determined resonance wavelengths, and furtherly be tuned by adjusting either the structure parameters or the incident angle of light. Theoretical analyses demonstrate that such EIT- and Fano-like absorptions are due to the interference of light in the multilayer structure and the complete transmission produced by the evanescent wave resonance in the configuration. The enhanced absorptions and the huge electrical field enhancement effect exhibit potentials for broad applications, such as photoacoustic imaging and Raman imaging.

Array-based sensing is a powerful tool for identifying analytes in complex environments with unknown interferences. In array-based sensing, the sensors, which transduce binding details to signal outputs, are of crucial importance for identifying analytes. Aggregation-induced emission luminogens (AIEgens) enjoy the advantages of easy synthesis and high sensitivity, which enable them to facilely form a sensor pool through structural modifications and sensitively reflect the subtle changes associated with binding events. All these features make AIEgens excellent candidates for array-based sensing, and attempts have been made by several research groups to explore their potentials in array-based sensing. In this review, we introduce the recent progresses of employing AIEgens as sensors in sensing assays and in building up sensor arrays for identification of varied biological analytes, including biomolecules and bacteria. Examples are selected to illustrate the working mechanism, probe design and selection, capability of the sensor array, and implications of these sensing methods.

Light scattering inside disordered media poses a significant challenge to achieve deep depth and high resolution simultaneously in biomedical optical imaging. Wavefront shaping emerged recently as one of the most potential methods to tackle this problem. So far, numerous algorithms have been reported, while each has its own pros and cons. In this article, we exploit a new thought that one algorithm can be reinforced by another complementary algorithm since they effectively compensate each other’s weaknesses, resulting in a more efficient hybrid algorithm. Herein, we introduce a systematical approach named GeneNN (Genetic Neural Network) as a proof of concept. Preliminary light focusing has been achieved by a deep neural network, whose results are fed to a genetic algorithm as an initial condition. The genetic algorithm furthers the optimization, evolving to converge into the global optimum. Experimental results demonstrate that with the proposed GeneNN, optimization speed is almost doubled and wavefront shaping performance can be improved up to 40% over conventional methods. The reinforced hybrid algorithm shows great potential in facilitating various biomedical and optical imaging techniques.

Dichroism is a material property that causes anisotropic light-matter interactions for different optical polarizations. Dichroism relates to molecular types and material morphology and thus can be used to distinguish different dichroic tissues. In this paper, we present single-shot dichroism photoacoustic microscopy that can image tissue structure, linear dichroism, and polarization angle with a single raster scanning. We develop a fiber-based laser system to split one laser pulse into three with different polarization angles, sub-microseconds time delay, and identical pulse energy. A dual-fiber optical-resolution photoacoustic microscopy system is developed to acquire three A-lines per scanning step. In such a way, dichroism imaging can achieve the same speed as single-wavelength photoacoustic microscopy. Moreover, the three polarized pulses originate from one laser pulse, which decreases pulse energy fluctuations and reduces dichroism measurement noise by ∼35 %. The new dichroism photoacoustic imaging technique can be used to image endogenous or exogenous polarization-dependent absorption contrasts, such as dichroic tumor or molecule-labeled tissue. Wavefront shaping (WFS) techniques have been used as a powerful tool to control light propagation in complex media, including multimode fibers. In this paper, we propose a new application of WFS for multimode fiber-based sensors. The use of a single multimode fiber alone, without any special fabrication, as a sensor based on the light intensity variations is not an easy task. The twist effect on multimode fiber is used as an example herein. Experimental results show that light intensity through the multimode fiber shows no direct relationship with the twist angle, but the correlation coefficient (CC) of speckle patterns does. Moreover, if WFS is applied to transform the spatially seemingly random light pattern at the exit of the multimode fiber into an optical focus. The focal pattern correlation and intensity both can serve to gauge the twist angle, with doubled measurement range and allowance of using a fast point detector to provide the feedback. With further development, WFS may find potentials to facilitate the development of multimode fiber-based sensors in a variety of scenarios.

Coherent optical control within or through scattering media via wavefront shaping has seen broad applications since its invention around 2007. Wavefront shaping is aimed at overcoming the strong scattering, featured by random interference, namely speckle patterns. This randomness occurs due to the refractive index inhomogeneity in complex media like biological tissue or the modal dispersion in multimode fiber, yet this randomness is actually deterministic and potentially can be time reversal or precompensated. Various wavefront shaping approaches, such as optical phase conjugation, iterative optimization, and transmission matrix measurement, have been developed to generate tight and intense optical delivery or high-resolution image of an optical object behind or within a scattering medium. The performance of these modulations, however, is far from satisfaction. Most recently, artificial intelligence has brought new inspirations to this field, providing exciting hopes to tackle the challenges by mapping the input and output optical patterns and building a neuron network that inherently links them. In this paper, we survey the developments to date on this topic and briefly discuss our views on how to harness machine learning (deep learning in particular) for further advancements in the field.

Multiple light scattering has been regarded as a barrier in imaging through complex media such as biological tissues. Owing to recent advances in wavefront shaping techniques, optical imaging through intact biological tissues without invasive procedures can now be used for direct experimental studies, presenting promising application opportunities in in vivo imaging and diagnosis. Although most of the recent proof of principle breakthroughs have been achieved in the laboratory setting with specialties in physics and engineering, we anticipate that these technologies can be translated to biological laboratories and clinical settings, which will revolutionize how we diagnose and treat a disease. To provide insight into the physical principle that enables the control of multiple light scattering in biological tissues and how recently developed techniques can improve bioimaging through thick tissues, we summarize recent progress on wavefront shaping techniques for controlling multiple light scattering in biological tissues.

Oxygen level, including blood oxygen saturation (sO2) and tissue oxygen partial pressure (pO2), are crucial physiological parameters in life science. This paper reviews the importance of these two parameters and the detection methods for them, focusing on the application of photoacoustic imaging in this scenario. sO2 is traditionally detected with optical spectra-based methods, and has recently been proven uniquely efficient by using photoacoustic methods. pO2, on the other hand, is typically detected by PET, MRI, or pure optical approaches, yet with limited spatial resolution, imaging frame rate, or penetration depth. Great potential has also been demonstrated by employing photoacoustic imaging to overcome the existing limitations of the aforementioned techniques.

Since its introduction to the field in mid-1990s, photoacoustic imaging has become a fast-developing biomedical imaging modality with many promising potentials. By converting absorbed diffused light energy into not-so-diffused ultrasonic waves, the reconstruction of the ultrasonic waves from the targeted area in photoacoustic imaging leads to a high-contrast sensing of optical absorption with ultrasonic resolution in deep tissue, overcoming the optical diffusion limit from the signal detection perspective. The generation of photoacoustic signals, however, is still throttled by the attenuation of photon flux due to the strong diffusion effect of light in tissue. Recently, optical wavefront shaping has demonstrated that multiply scattered light could be manipulated so as to refocus inside a complex medium, opening up new hope to tackle the fundamental limitation. In this paper, the principle and recent development of photoacoustic imaging and optical wavefront shaping are briefly introduced. Then we describe how photoacoustic signals can be used as a guide star for in-tissue optical focusing, and how such focusing can be exploited for further enhancing photoacoustic imaging in terms of sensitivity and penetration depth. Finally, the existing challenges and further directions towards in vivo applications are discussed.

Significance: In nonballistic regime, optical scattering impedes high-resolution imaging through/inside complex media, such as milky liquid, fog, multimode fiber, and biological tissues, where confocal and multiphoton modalities fail. The significant tissue inhomogeneity-induced distortions need to be overcome and a technique referred as optical wavefront shaping (WFS), first proposed in 2007, has been becoming a promising solution, allowing for flexible and powerful light control. Understanding the principle and development of WFS may inspire exciting innovations for effective optical manipulation, imaging, stimulation, and therapy at depths in tissue or tissue-like complex media. Aim: We aim to provide insights about what limits the WFS towards biomedical applications, and how recent efforts advance the performance of WFS among different trade-offs. Approach: By differentiating the two implementation directions in the field, i.e., precompensation WFS and optical phase conjugation (OPC), improvement strategies are summarized and discussed. Results: For biomedical applications, improving the speed of WFS is most essential in both directions, and a system-compatible wavefront modulator driven by fast apparatus is desired. In addition to that, algorithm efficiency and adaptability to perturbations/noise is of concern in precompensation WFS, while for OPC significant improvements rely heavily on integrating physical mechanisms and delicate system design for faster response and higher energy gain. Conclusions: Substantial improvements in WFS implementations, from the aspects of physics, engineering, and computing, have inspired many novel and exciting optical applications that used to be optically inaccessible. It is envisioned that continuous efforts in the field can further advance WFS towards biomedical applications and guide our vision into deep biological tissues. [ABSTRACT FROM AUTHOR]

Three-dimensional single-pixel imaging (3D SPI) has become an attractive imaging modality for both biomedical research and optical sensing. 3D-SPI techniques generally depend on time-of-flight or stereovision principle to extract depth information from backscattered light. However, existing implementations for these two optical schemes are limited to surface mapping of 3D objects at depth resolutions, at best, at the millimeter level. Here, we report 3D light-field illumination single-pixel microscopy (3D-LFI-SPM) that enables volumetric imaging of microscopic objects with a near-diffraction-limit 3D optical resolution. Aimed at 3D space reconstruction, 3D-LFI-SPM optically samples the 3D Fourier spectrum by combining 3D structured light-field illumination with single-element intensity detection. We build a 3D-LFI-SPM prototype that provides an imaging volume of ~390 × 390 × 3,800 µm³ and achieves 2.7-µm lateral resolution and better than 37-µm axial resolution. Its capability of 3D visualization of label-free optical absorption contrast is demonstrated by imaging single algal cells in vivo.
