1. Seismic magnitude clustering is prevalent in field and laboratory catalogs

Abstract: Abstract Clustering of earthquake magnitudes is still actively debated, compared to well-established spatial and temporal clustering. Magnitude clustering is not currently implemented in earthquake forecasting but would be important if larger magnitude events are more likely to be followed by similar sized events. Here we show statistically significant magnitude clustering present in many different field and laboratory catalogs at a wide range of spatial scales (mm to 1000 km). It is universal in field catalogs across fault types and tectonic/induced settings, while laboratory results are unaffected by loading protocol or rock types and show temporal stability. The absence of clustering can be imposed by a global tensile stress, although clustering still occurs when isolating to triggered event pairs or spatial patches where shear stress dominates. Magnitude clustering is most prominent at short time and distance scales and modeling indicates >20% repeating magnitudes in some cases, implying it can help to narrow physical mechanisms for seismogenesis.

Abstract: Indoor navigation is increasingly widespread in complex indoor environments, and indoor path planning is the most important part of indoor navigation. Path planning generally refers to finding the most suitable path connecting two locations, while avoiding collision with obstacles. However, it is a fundamental problem, especially for 3D complex building model. A common way to solve the issue in some applications has been approached in a number of relevant literature, which primarily operates on 2D drawings or building layouts, possibly with few attached attributes for obstacles. Although several digital building models in the format of 3D CAD have been used for path planning, they usually contain only geometric information while losing abundant semantic information of building components (e.g. types and attributes of building components and their simple relationships). Therefore, it becomes important to develop a reliable method that can enhance application of path planning by combining both geometric and semantic information of building components. This paper introduces a method that support 3D indoor path planning with semantic information.

3. A Compact Narrow-Band Bandstop Filter Using Spiral-Shaped Defected Microstrip Structure

Abstract: A novel compact narrow-band bandstop filter is implemented by using the proposed spiral-shaped defected microstrip structure (SDMS) in this paper. Compared with other DMSs, the presented SDMS exhibits the advantage of compact size and narrow stopband. Meanwhile, an approximate design rule of the SDMS is achieved and the effects of the dimensions on the resonant frequency and 3 dB fractional bandwidth (FBW) are analyzed in detail. Both the simulation and measurement results of the fabricated bandstop filter show that it has a 10 dB stopband from 3.4 GHz to 3.6 GHz with more than 45 dB rejection at the center frequency.

4. A 3D Model Based Imdoor Navigation System for Hubei Provincial Museum

Abstract: 3D models are more powerful than 2D maps for indoor navigation in a complicate space like Hubei Provincial Museum because they can provide accurate descriptions of locations of indoor objects (e.g., doors, windows, tables) and context information of these objects. In addition, the 3D model is the preferred navigation environment by the user according to the survey. Therefore a 3D model based indoor navigation system is developed for Hubei Provincial Museum to guide the visitors of museum. The system consists of three layers: application, web service and navigation, which is built to support localization, navigation and visualization functions of the system. There are three main strengths of this system: it stores all data needed in one database and processes most calculations on the webserver which make the mobile client very lightweight, the network used for navigation is extracted semi-automatically and renewable, the graphic user interface (GUI), which is based on a game engine, has high performance of visualizing 3D model on a mobile display.

5. Multi-dimensional Indoor Location Information Model

Abstract: Aiming at the increasing requirements of seamless indoor and outdoor navigation and location service, a Chinese standard of Multidimensional Indoor Location Information Model is being developed, which defines ontology of indoor location. The model is complementary to 3D concepts like CityGML and IndoorGML. The goal of the model is to provide an exchange GML-based format for location needed for indoor routing and navigation. An elaborated user requirements analysis and investigation of state-of-the-art technology in expressing indoor location at home and abroad was completed to identify the manner humans specify location. The ultimate goal is to provide an ontology that will allow absolute and relative specification of location such as "in room 321", "on the second floor", as well as, "two meters from the second window", "12 steps from the door".

Abstract: Abstract Two-dimensional (2D) materials offer a prospect of membranes that combine negligible gas permeability with high proton conductivity and could outperform the existing proton exchange membranes used in various applications including fuel cells. Graphene oxide (GO), a well-known 2D material, facilitates rapid proton transport along its basal plane but proton conductivity across it remains unknown. It is also often presumed that individual GO monolayers contain a large density of nanoscale pinholes that lead to considerable gas leakage across the GO basal plane. Here we show that relatively large, micrometer-scale areas of monolayer GO are impermeable to gases, including helium, while exhibiting proton conductivity through the basal plane which is nearly two orders of magnitude higher than that of graphene. These findings provide insights into the key properties of GO and demonstrate that chemical functionalization of 2D crystals can be utilized to enhance their proton transparency without compromising gas impermeability.

Abstract: IntroductionStaphylococcus aureus bacteremia (SAB) is a life-threatening infection particularly involving methicillin-resistant S. aureus (MRSA). In contrast to resolving MRSA bacteremia (RB), persistent MRSA bacteremia (PB) blood cultures remain positive despite appropriate antibiotic treatment. Host immune responses distinguishing PB vs. RB outcomes are poorly understood. Here, integrated transcriptomic, IL-10 cytokine levels, and genomic analyses sought to identify signatures differentiating PB vs. RB outcomes.MethodsWhole-blood transcriptomes of propensity-matched PB (n=28) versus RB (n=30) patients treated with vancomycin were compared in one independent training patient cohort. Gene expression (GE) modules were analyzed and prioritized relative to host IL-10 cytokine levels and DNA methyltransferase-3A (DNMT3A) genotype.ResultsDifferential expression of T and B lymphocyte gene expression early in MRSA bacteremia discriminated RB from PB outcomes. Significant increases in effector T and B cell signaling pathways correlated with RB, lower IL-10 cytokine levels and DNMT3A heterozygous A/C genotype. Importantly, a second PB and RB patient cohort analyzed in a masked manner demonstrated high predictive accuracy of differential signatures.DiscussionCollectively, the present findings indicate that human PB involves dysregulated immunity characterized by impaired T and B cell responses associated with excessive IL-10 expression in context of the DNMT3A A/A genotype. These findings reveal distinct immunologic programs in PB vs. RB outcomes, enable future studies to define mechanisms by which host and/or pathogen drive differential signatures and may accelerate prediction of PB outcomes. Such prognostic assessment of host risk could significantly enhance early anti-infective interventions to avert PB and improve patient outcomes.

Abstract: Invasive methicillin-resistant Staphylococcus aureus (MRSA) infections are leading causes of morbidity and mortality that are complicated by increasing resistance to conventional antibiotics. Thus, minimizing virulence and enhancing antibiotic efficacy against MRSA is a public health imperative. We originally demonstrated that diflunisal (DIF; [2-hydroxy-5-(2,4-difluorophenyl) benzoic acid]) inhibits S. aureus virulence factor expression. To investigate pharmacophores that are active in this function, we evaluated a library of structural analogues for their efficacy to modulate virulence phenotypes in a panel of clinically relevant S. aureus isolates in vitro. Overall, the positions of the phenyl, hydroxyl, and carboxylic moieties and the presence or type of halogen (F vs. Cl) influenced the efficacy of compounds in suppressing hemolysis, proteolysis, and biofilm virulence phenotypes. Analogues lacking halogens inhibited proteolysis to an extent similar to DIF but were ineffective at reducing hemolysis or biofilm production. In contrast, most analogues lacking the hydroxyl or carboxylic acid groups did not suppress proteolysis but did mitigate hemolysis and biofilm production to an extent similar to DIF. Interestingly, chirality and the substitution of fluorine with chlorine resulted in a differential reduction in virulence phenotypes. Together, this pattern of data suggests virulence-suppressing pharmacophores of DIF and structural analogues integrate halogen, hydroxyl, and carboxylic acid moiety stereochemistry. The anti-virulence effects of DIF were achieved using concentrations that are safe in humans, do not impair platelet antimicrobial functions, do not affect S. aureus growth, and do not alter the efficacy of conventional antibiotics. These results offer proof of concept for using novel anti-virulence strategies as adjuvants to antibiotic therapy to address the challenge of MRSA infection.

Abstract: Virulence factor expression is integral to pathogenicity of Staphylococcus aureus. We previously demonstrated that aspirin, through its major metabolite, salicylic acid (SAL), modulates S. aureus virulence phenotypes in vitro and in vivo. We compared salicylate metabolites and a structural analogue for their ability to modulate S. aureus virulence factor expression and phenotypes: (i) acetylsalicylic acid (ASA, aspirin); (ii) ASA metabolites, salicylic acid (SAL), gentisic acid (GTA) and salicyluric acid (SUA); or (iii) diflunisal (DIF), a SAL structural analogue. None of these compounds altered the growth rate of any strain tested. ASA and its metabolites SAL, GTA and SUA moderately impaired hemolysis and proteolysis phenotypes in multiple S. aureus strain backgrounds and their respective deletion mutants. Only DIF significantly inhibited these virulence phenotypes in all strains. The kinetic profiles of ASA, SAL or DIF on expression of hla (alpha hemolysin), sspA (V8 protease) and their regulators (sigB, sarA, agr (RNAIII)) were assessed in two prototypic strain backgrounds: SH1000 (methicillin-sensitive S. aureus; MSSA) and LAC-USA300 (methicillin-resistant S. aureus; MRSA). DIF induced sigB expression which is coincident with the significant inhibition of RNAIII expression in both strains and precedes significant reductions in hla and sspA expression. The inhibited expression of these genes within 2 h resulted in the durable suppression of hemolysis and proteolysis phenotypes. These results indicate that DIF modulates the expression of key virulence factors in S. aureus via a coordinated impact on their relevant regulons and target effector genes. This strategy may hold opportunities to develop novel antivirulence strategies to address the ongoing challenge of antibiotic-resistant S. aureus.

15. Impact of the Novel Prophage ϕSA169 on Persistent Methicillin-Resistant Staphylococcus aureus Endovascular Infection

Abstract: ABSTRACT Persistent methicillin-resistant Staphylococcus aureus (MRSA) endovascular infections are life-threatening syndromes with few therapeutic options. The potential impact of bacteriophages on the persistent outcome has not been well studied. In this study, we investigated the role of a novel prophage (ϕSA169) in MRSA persistence by using a lysogen-free clinically resolving bacteremia (RB) isolate and comparing it to a derivative which was obtained by infecting the RB strain with ϕSA169, which has been lysogenized in a clinical persistent MRSA bacteremia (PB) isolate. Similar to the PB isolate, the ϕSA169-lysogenized RB strain exhibited well-defined in vitro and in vivo phenotypic and genotypic signatures related to the persistent outcome, including earlier activation of global regulators (i.e., sigB, sarA, agr RNAIII, and sae); higher expression of a critical purine biosynthesis gene, purF; and higher growth rates accompanied by lower ATP levels and vancomycin (VAN) susceptibility and stronger δ-hemolysin and biofilm formation versus its isogenic parental RB isolate. Notably, the contribution of ϕSA169 in persistent outcome with VAN treatment was confirmed in an experimental infective endocarditis model. Taken together, these results indicate the critical role of the prophage ϕSA169 in persistent MRSA endovascular infections. Further studies are needed to identify the mechanisms of ϕSA169 in mediating the persistence, as well as establishing the scope of impact, of this prophage in other PB strains. IMPORTANCE Bacteriophages are viruses that invade the bacterial host, disrupt bacterial metabolism, and cause the bacterium to lyse. Because of its remarkable antibacterial activity and unique advantages over antibiotics, for instance, bacteriophage is specific for one species of bacteria and resistance to phage is less common than resistance to antibiotics. Indeed, bacteriophage therapy for treating infections due to multidrug-resistant pathogens in humans has become a research hot spot. However, it is also worth considering that bacteriophages are transferable and could cotransfer host chromosomal genes, e.g., virulence and antimicrobial resistance genes, while lysogenizing and integrating into the bacterial chromosome (prophage), thus playing a role in bacterial evolution and virulence. In the current study, we identified a novel prophage, ϕSA169, from a clinical persistent MRSA bacteremia isolate, and we determined that ϕSA169 mediated well-defined in vitro and in vivo phenotypic and genotypic signatures related to the persistent outcome, which may represent a unique and important persistent mechanism(s).

17. Identification of Methicillin-Resistant Staphylococcus aureus (MRSA) Genetic Factors Involved in Human Endothelial Cells Damage, an Important Phenotype Correlated with Persistent Endovascular Infection

Abstract: Methicillin-resistant Staphylococcus aureus (MRSA) is a leading cause of life-threatening endovascular infections. Endothelial cell (EC) damage is a key factor in the pathogenesis of these syndromes. However, genetic factors related to the EC damage have not been well studied. This study aims to identify genetic determinants that impact human EC damage by screening the genome-wide Nebraska Transposon Mutant Library (NTML). A well-established MTT assay was used to test the in vitro damage of human EC cell line (HMEC-1) caused by each mutant strain in the NTML. We first confirmed some global regulators and genes positively impact the EC damage, which is consistent with published results. These data support the utility of the high-throughput approach. Importantly, we demonstrated 317 mutants significantly decreased the EC damage, while only 6 mutants enhanced the EC damage vs. parental JE2 strain. The majority of these genes have not been previously defined to affect human EC damage. Interestingly, many of these newly identified genes are involved in metabolism, genetic and environmental information processing, and cellular processes. These results advance our knowledge of staphylococcal genetic factors related to human EC damage which may provide novel targets for the development of effective agents against MRSA endovascular infection.

21. Electric Vehicle Charging Scheduling and Pricing Based on Stackelberg Game

Abstract: The disorderly charging and discharging of a large number of electric vehicles will have a serious negative impact on the safety and economic operation of the power system. In order to avoid safety problems and obtain maximum economic benefits, a master-slave game is proposed to model the maximization of profits for agents and car owners. In the upper model of the model, the optimization objective of the main body is to minimize the operating cost and minimize the load mean square error; in the lower model, the goal is to minimize the cost of electric vehicles; the particle swarm algorithm is used, and the model decoupling characteristics are used from the inside to the outside. Solve the Nash equilibrium point of the optimization model, obtain the set of optimal time-of-use electricity price and electric vehicle charging and discharging scheme, and finally obtain the global optimal pricing strategy. Various typical scenarios before and after optimization are compared and analyzed in detail through calculation examples, and the effect of the proposed model and the overall scheduling benefit are verified. Беспорядочная зарядка и разрядка большого количества электромобилей окажет серьезное негативное влияние на безопасность и экономичность работы энергосистемы. Во избежание проблем с безопасностью и получения максимальной экономической выгоды предлагается игра «хозяин-раб» для моделирования максимизации прибыли агентов и автовладельцев. В старшей модели целью оптимизации основного корпуса является минимизация эксплуатационных расходов и минимизация среднеквадратичной ошибки нагрузки; в младшей модели цель - минимизировать стоимость электромобилей; используется алгоритм роя частиц и используются характеристики развязки модели изнутри наружу. Предлагается решение точки равновесия Нэша модели оптимизации, получение набора оптимальной цены на электроэнергию во время использования и схемы зарядки и разрядки электромобиля и, наконец, получение глобальной оптимальной стратегии ценообразования. Различные типичные сценарии до и после оптимизации сравниваются и подробно анализируются с помощью примеров расчетов, а также проверяется влияние предлагаемой модели и общее преимущество планирования.

22. Optimization Design of Key Parameters of Solar Flat Panel Solar Collector’s Own Structure Based on PYTHON

Abstract: Combined with domestic and foreign literature, the research progress, key parameters and existing problems of flat-panel solar collectors are analyzed. On this basis, the thermal efficiency of flat-panel solar collectors is studied by means of experiments and numerical simulations. This paper introduces and analyzes the structural characteristics of flat-panel solar collectors. Based on the structural characteristics of flat-panel solar energy and research at home and abroad, the influence of the structural parameters of each flat-panel solar collector on the collector performance was studied. According to the heat transfer process and structural characteristics of the collector, the heat energy equation of the collector is established, and the calculation process of the heat loss of the collector is simplified according to the existing research, and the calculation of the heat loss coefficient is an empirical formula. In order to facilitate the experimental study, an efficiency factor and a heat transfer factor are introduced, and the temperature of the fluid at the inlet of the collector is used to replace the surface temperature of the heat-absorbing plate, which is difficult to measure. The calculation program is used to obtain the influence data of each key parameter of the collector on the thermal efficiency and use Origin to process the data to visualize the influence of each parameter on the thermal efficiency. On this basis, a variance analysis method is proposed to optimize the collector. Based on the PYTHON software, the calculation program is written according to the variance analysis method, and the key parameters that will greatly affect the instantaneous efficiency of the collector are combined in pairs, and the optimal collector parameter combination corresponding to the maximum instantaneous efficiency is studied. This research has a certain guiding effect on the utilization of solar energy. На основе литературных данных проанализированы ход исследований, основные параметры и существующие проблемы плоскопанельных солнечных коллекторов. Исходя из этого, тепловой КПД плоских солнечных коллекторов изучается с помощью экспериментов и численного моделирования. В статье представлены и проанализированы структурные характеристики плоских солнечных коллекторов. На основе структурных характеристик плоскопанельной солнечной энергии и исследований было изучено влияние структурных параметров каждого плоскопанельного солнечного коллектора на производительность коллектора. В соответствии с процессом теплопередачи и конструктивными характеристиками коллектора устанавливается уравнение тепловой энергии коллектора, а процесс расчета тепловых потерь коллектора упрощается в соответствии с существующими исследованиями, а расчет коэффициента тепловых потерь является эмпирической формулой. Для облегчения экспериментального исследования вводятся коэффициент полезного действия и коэффициент теплоотдачи, а температура жидкости на входе в коллектор используется вместо температуры поверхности теплопоглощающей пластины, которую трудно измерить. Программа расчета используется для получения данных о влиянии каждого ключевого параметра коллектора на тепловую эффективность и использует Origin для обработки данных для визуализации влияния каждого параметра на тепловую эффективность. На этой основе предлагается метод дисперсионного анализа для оптимизации коллектора. На основе программного обеспечения PYTHON программа расчета написана в соответствии с методом дисперсионного анализа, а ключевые параметры, которые будут сильно влиять на мгновенную эффективность коллектора объединены попарно, а оптимальная комбинация параметров коллектора, соответствующая максимальной мгновенной эффективности, изучалась. Это исследование имеет определенное направляющее влияние на использование солнечной энергии.

Abstract: The oral gingival barrier is a constantly stimulated and dynamic environment where homeostasis is often disrupted, resulting in inflammatory periodontal diseases. Type 2 diabetes (T2D) has been reported to be associated with gingival barrier dysfunction, but the effect and underlying mechanism are inconclusive. Herein, we performed single-cell RNA sequencing (scRNA-seq) of gingiva from leptin receptor-deficient mice ( db/db) to examine the gingival heterogeneity in the context of T2D. Periodontal health of control mice is characterized by populations of Krt14+-expressing epithelial cells and Col1a1+-fibroblasts mediating immune homeostasis primarily through the enrichment of innate lymphoid cells. The db/db gingiva exhibited decreased epithelial/stromal ratio and dysfunctional barrier. We further observed stromal, particularly fibroblast immune hyperresponsiveness, linked to the recruitment of myeloid-derived cells at the db/db gingiva. Both scRNA-seq and histological analysis suggested the inflammatory signaling between fibroblasts and neutrophils as a potential driver of diabetes-induced periodontal damage. Notably, the “immune-like” stromal cells were wired toward the induction of gingival IL-17A hyperresponsiveness in db/db mice. Our work reveals that the “immune-like” fibroblasts with transcriptional diversity are involved in the innate immune homeostasis at the diabetic gingiva. It highlights a potentially significant role of these cell types in its pathogenesis.

Abstract: Staphylococcus aureus (especially methicillin-resistant S. aureus [MRSA]) is frequently associated with persistent bacteremia (PB) during vancomycin therapy despite consistent susceptibility in vitro. Strategic comparisons of PB strains versus those from vancomycin-resolving bacteremia (RB) would yield important mechanistic insights into PB outcomes. Abstract: Two novel complexes, namely [Cd(L)(NO3)2]n (1) and {[Cd(L)(NO3)2](CH3CH2OH)}n (2) (L = 1,3,5-tris[1-(4-oxidroxypyridinium-methylene)]benzene) are synthesized solvothermally in different solvents. Single crystal and powder X-ray diffraction, infrared spectroscopy, thermogravimetry, 2D fingerprint plots, and Hirshfeld surface analyses are carried out for these complexes and their topology and thermal stability are also analyzed. Two complexes exhibit the same 2D interpenetrating structure formed by the ligand coordinated with Cd(II) ions. Due to the presence of ethanol molecules in complex 2, part of π⋯π stacking in complex 1 is blocked, resulting in new C—H⋯π and hydrogen bonding. This change makes the weak force stronger and forms a more stable framework. The result is that the thermal stability of 2 is higher than that of 1.

Abstract: AIMS This study investigated the dose-effect of manganese (Mn) addition on wheat straw (WS) decomposition, and explored the potential mechanisms of Mn involved in the acceleration of WS decomposition in regards to the soil microbial communities and enzyme activities. METHODS AND RESULTS A 180-day incubation experiment was performed to examine the decomposition of WS under four Mn levels, that is, 0, 0.25, 1 and 2 mg g-1 . The effects of microbial communities and enzyme activities were evaluated using control (0 mg g-1 ) and Mn (0.25 mg g-1 ) treatments. Our results revealed that Mn (0.25 mg g-1 ) addition significantly increased WS decomposition, and enhanced the release of carbon and nitrogen. Optimal Mn addition (0.25 mg g-1 ) also caused significant increases in the activity of neutral xylanase (NEX), laccase (Lac), manganese peroxidase (MnP) and lignin peroxidase (LiP) within the incubation period. Mn (0.25 mg g-1 ) addition also enriched some operational taxonomic units (OTUs) that, in turn, had the potential ability to decompose crop straw, such as secreting lignocellulolytic enzymes. CONCLUSIONS Mn (0.25 mg g-1 ) could promote WS decomposition through enrichment of the microbial species involved in biomass decomposition, which enhanced the lignocellulose-degrading enzyme activity. SIGNIFICANCE AND IMPACT OF THE STUDY This study provides evidence for Mn to promote WS biodegradation after Mn application, opening new windows to improve the utilization efficiency of crop residues.

Abstract: Streptococcus gordonii and Streptococcus sanguinis are primary colonizers of tooth surfaces and are generally associated with oral health, but can also cause infective endocarditis (IE). These species express “Siglec-like” adhesins that bind sialylated glycans on host glycoproteins, which can aid the formation of infected platelet–fibrin thrombi (vegetations) on cardiac valve surfaces. We previously determined that the ability of S. gordonii to bind sialyl T-antigen (sTa) increased pathogenicity, relative to recognition of sialylated core 2 O-glycan structures, in an animal model of IE. However, it is unclear when and where the sTa structure is displayed, and which sTa-modified host factors promote valve colonization. In this study, we identified sialylated glycoproteins in the aortic valve vegetations and plasma of rat and rabbit models of this disease. Glycoproteins that display sTa vs. core 2 O-glycan structures were identified by using recombinant forms of the streptococcal Siglec-like adhesins for lectin blotting and affinity capture, and the O-linked glycans were profiled by mass spectrometry. Proteoglycan 4 (PRG4), also known as lubricin, was a major carrier of sTa in the infected vegetations. Moreover, plasma PRG4 levels were significantly higher in animals with damaged or infected valves, as compared with healthy animals. The combined results demonstrate that, in addition to platelet GPIbα, PRG4 is a highly sialylated mucin-like glycoprotein found in aortic valve vegetations and may contribute to the persistence of oral streptococci in this protected endovascular niche. Moreover, plasma PRG4 could serve as a biomarker for endocardial injury and infection.

Abstract: Persistent methicillin-resistant Staphylococcus aureus (MRSA) endovascular infections represent a significant subset of S. aureus infections and correlate with exceptionally high mortality. We have recently demonstrated that the lysogenization of prophage ϕSA169 from a clinical persistent MRSA bacteremia isolate (300-169) into a clinical resolving bacteremia MRSA isolate (301-188) resulted in the acquisition of well-defined in vitro and in vivo phenotypic and genotypic profiles related to persistent outcome. However, the underlying mechanism(s) of this impact is unknown. In the current study, we explored the genetic mechanism that may contribute to the ϕSA169-correlated persistence using RNA sequencing. Transcriptomic analyses revealed that the most significant impacts of ϕSA169 were: (i) the enhancement of fatty acid biosynthesis and purine and pyrimidine metabolic pathways; (ii) the repression of galactose metabolism and phosphotransferase system (PTS); and (iii) the down-regulation of the mutual prophage genes in both 300-169 and 301-188 strains. In addition, the influence of different genetic backgrounds between 300-169 and 301-188 might also be involved in the persistent outcome. These findings may provide targets for future studies on the persistence of MRSA.

Abstract: Persistent methicillin-resistant Staphylococcus aureus (MRSA) endovascular infections represent a significant clinical-therapeutic challenge. Of particular concern is antibiotic treatment failure in infections caused by MRSA that are “susceptible” to antibiotic in vitro. In the current study, we investigate specific purine biosynthetic pathways and stringent response mechanism(s) related to this life-threatening syndrome using genetic matched persistent and resolving MRSA clinical bacteremia isolates (PB and RB, respectively), and isogenic MRSA strain sets. We demonstrate that PB isolates (vs RB isolates) have significantly higher (p)ppGpp production, phenol-soluble-modulin expression, polymorphonuclear leukocyte lysis and survival, fibronectin/endothelial cell (EC) adherence, and EC damage. Importantly, an isogenic strain set, including JE2 parental, relP-mutant and relP-complemented strains, translated the above findings into significant outcome differences in an experimental endocarditis model. These observations indicate a significant regulation of purine biosynthesis on stringent response, and suggest the existence of a previously unknown adaptive genetic mechanism in persistent MRSA infection.

42. Laboratory Inspirational Information on the 'Burst-Like' and 'Swarm-Like' Energy Releases from Anthropogenic Rock Fracture Processes

Abstract: ABSTRACT: Laboratory experimental tests can provide inspirational information for geologists and/or geophysicists to understand field-scale phenomena and processes. Field-scale anthropogenic seismicity catalogs are usually dominated by swarm-like features, while the natural seismicity of structural faults (i.e., the San Andreas fault) clearly exhibits the co-existence of both swarm-like and burst-like energy releases. This paper compares the energy release features from the laboratory anthropogenic (i.e., loading induced) rock fracture processes from different types of laboratory tests. We use a comparatively objective model to inspect the topology structure of the laboratory energy releases, i.e., an acoustic emission (AE) network, and to inspect/characterize the energy release features. The model we have used demands only the energy releases obey the scaling-law. Thus scale-invariance on these energy releases has been inspected and verified. By combining the known different laboratory settings with their consequentially induced different energy release features, we postulate an inspirational understanding on the mechanisms that distinguish the burst-like and swarm-like energy releases. The spatial discontinuity may be one influential factor for differentiating the catalog into dominant swarm-like or mixed burst- and swarm-like behavior. Further investigations on identifying and quantifying other possible influential factors that differentiate the catalogs into different types are warranted. 1. INTRODUCTION Induced seismicity from hydraulic fracturing (Kozłowska et al., 2018; Skoumal et al., 2015b, 2018a; Skoumal et al., 2018b) or waste water disposal (Skoumal et al., 2020a; Skoumal et al., 2015a; Skoumal et al., 2016; Skoumal et al., 2020b) is dominated by swarm-like features. This swarm-like feature describes the energy releases of the stressed underground rock mass as having spatial-temporal affinity, for instance, characterized by narrow aftershock/seismic-active zones (Karimi and Davidsen, 2021). Natural seismicity, on the contrary, display clearly different mixed burst-like and swarm-like features (Zaliapin and Ben-Zion, 2013a, b), with some regions of seismicity reported as more swarm-like or more burst-like (Brodsky et al., 1998; Sturtevant et al., 1996; Zaliapin and Ben-Zion, 2013b). Multiple mechanisms can cause the different regional observations of the burst-like or the swarm-like seismicity features, e.g., the brittle and creeping features due to lithology differences, geothermal flows (Zaliapin and Ben-Zion, 2013b), or the "rectified diffusion" process because of the bubble release from the porous fluid (Sturtevant et al., 1996), etc. A further comprehensive and quantitative decoupling of the possible mechanisms involved is challenging for field-scale studies. This is partially due to the limited accessibility to various types of in-situ measurements and the limited temporal coverage of scientific observations on the seismic activity.

43. Optimally Extracting the Asymptotic Features of Rock Fracture Process from Mode-I and Mixed-Mode Tests Under Three-Point Bending

Abstract: ABSTRACT: This paper consists of a series of attempts and discussions on the data analysis approaches to investigate and extract the asymptotic features in the rock fracture process. The rock fracture discussed here were under mode-I and mixed-mode loadings. We examine the process of creating the rock fracture from the artificial notch, as well as the following rock fracture propagation processes. The analysis consists of the spatial-sequential feature, and the topological triggering-triggered feature of the energy releases from the rock fracture process, as well as the change of data variance on the delineated rock fracture path through the DIC (digital image correlation) observation. The spatial-sequential feature shows a comparatively stable behavior during the rock fracture propagation stage. The growth of the rock fracture within this stable stage is produced from the naturally induced rock fracture; It can be fundamentally different from that induced from an artificial notch, especially in a mode-I test. The topological feature cross-validates this difference. The change of the data variance of the DIC delineated rock fracture path suggests similar conclusions, i.e., an erratic feature at the initial rock fracture path followed with stable propagation. The combination of the analyses may provide an approach to investigate the fracture toughness in rock materials more naturally/spontaneously, i.e., the influence of artificial notch is reasonably minimized or qualitatively characterized, and the rock fracture is induced from the preceding induced rock fracture. 1. INTRODUCTION The behavior of rock mass under stress are of broad interesting for a variety of issues, for instance, the natural disasters like earthquake, tsunami, debris flows, and geological and civil engineering projects. As the rock mass behavior is strongly related to the natural discontinuities within, the theory of fracture mechanics has been extensively used for rock mechanics studies. Within fracture mechanics, the crack length and the fracture toughness are two key elements for the fracture propagation criterion, under the simplification of LEFM (linear elastic fracture mechanics (Asem et al., 2021)). The materialization of LEFM into the rock material requires the size-effect law so that the laboratory observations can be extrapolated to the prediction of large-scale structures. The laboratory observations of three-point bending tests on the beam samples with central or eccentric notch settings are usually used for measuring the parameter of fracture toughness. Such theoretical structure of LEFM requires as assumption that the non-linear region surrounding the crack tip, i.e., the rock fracture process zone (FPZ), to be small and surrounded by linear behavior. Many studies have reported the rock FPZ can be of non-negligible size (Nolen-Hoeksema and Gordon, 1987; Petit and Barquins, 1988; Wong and Xiong, 2018) and can show spatial temporal and evolution features (Xiong et al., 2021).

Abstract: Persistent methicillin-resistant Staphylococcus aureus (MRSA) endovascular infections represent a significant clinically challenging subset of invasive, life-threatening S. aureus infections. We have recently demonstrated that purine biosynthesis plays an important role in such persistent infections. Cyclic di-AMP (c-di-AMP) is an essential and ubiquitous second messenger that regulates many cellular pathways in bacteria. However, whether there is a regulatory connection between the purine biosynthesis pathway and c-di-AMP impacting persistent outcomes was not known. Here, we demonstrated that the purine biosynthesis mutant MRSA strain, the ΔpurF strain (compared to its isogenic parental strain), exhibited the following significant differences in vitro: (i) lower ADP, ATP, and c-di-AMP levels; (ii) less biofilm formation with decreased extracellular DNA (eDNA) levels and Triton X-100-induced autolysis paralleling enhanced expressions of the biofilm formation-related two-component regulatory system lytSR and its downstream gene lrgB; (iii) increased vancomycin (VAN)-binding and VAN-induced lysis; and (iv) decreased wall teichoic acid (WTA) levels and expression of the WTA biosynthesis-related gene, tarH. Substantiating these data, the dacA (encoding diadenylate cyclase enzyme required for c-di-AMP synthesis) mutant strain (dacAG206S strain versus its isogenic wild-type MRSA and dacA-complemented strains) showed significantly decreased c-di-AMP levels, similar in vitro effects as seen above for the purF mutant and hypersusceptible to VAN treatment in an experimental biofilm-related MRSA endovascular infection model. These results reveal an important intersection between purine biosynthesis and c-di-AMP that contributes to biofilm-associated persistence in MRSA endovascular infections. This signaling pathway represents a logical therapeutic target against persistent MRSA infections. IMPORTANCE Persistent endovascular infections caused by MRSA, including vascular graft infection syndromes and infective endocarditis, are significant and growing public health threats. A particularly worrisome trend is that most MRSA isolates from these patients are “susceptible” in vitro to conventional anti-MRSA antibiotics, such as VAN and daptomycin (DAP), based on Clinical and Laboratory Standards Institute breakpoints. Yet, these antibiotics frequently fail to eliminate these infections in vivo. Therefore, the persistent outcomes in MRSA infections represent a unique and important variant of classic “antibiotic resistance” that is only disclosed during in vivo antibiotic treatment. Given the high morbidity and mortality associated with the persistent infection, there is an urgent need to understand the specific mechanism(s) of this syndrome. In the current study, we demonstrate that a functional intersection between purine biosynthesis and the second messenger c-di-AMP plays an important role in VAN persistence in experimental MRSA endocarditis. Targeting this pathway may represent a potentially novel and effective strategy for treating these life-threatening infections.

Abstract: Sichuan sika deer and livestock are sympatric species, the uncontrolled number of livestock increases the life pressure of Sichuan sika deer. This work investigated microhabitat characteristics at 173 sites from May to September 2011 to improve the understanding of microhabitat utilize relationship between the two species at the Tiebu Nature Reserve. The current study indicated that Sichuan sika deer preferred to hide in the forest habitat and forage in pasture and shrub patch, while livestock use pasture more frequently. Compared to Sichuan sika deer, the pasture faraway from forest edge being used more frequently by livestock. However. The overlap of forage habitats is still evident, especially in pastures that close to the forests. Based on these findings, the current investigation provided a method to manage livestock, which could relieve competition and could protect Sichuan sika deer efficiently.
