Abstract Background Enhancers are important gene regulatory elements that promote the expression of critical genes in development and disease. Aberrant enhancer can modulate cancer risk and activate oncogenes that lead to the occurrence of various cancers. However, the underlying mechanism of most enhancers in cancer remains unclear. Here, we aim to explore the function and mechanism of a crucial enhancer in melanoma. Methods Multi-omics data were applied to identify an enhancer (enh17) involved in melanoma progression. To evaluate the function of enh17, CRISPR/Cas9 technology were applied to knockout enh17 in melanoma cell line A375. RNA-seq, ChIP-seq and Hi-C data analysis integrated with luciferase reporter assay were performed to identify the potential target gene of enh17. Functional experiments were conducted to further validate the function of the target gene ETV4. Multi-omics data integrated with CUT&Tag sequencing were performed to validate the binding profile of the inferred transcription factor STAT3. Results An enhancer, named enh17 here, was found to be aberrantly activated and involved in melanoma progression. CRISPR/Cas9-mediated deletion of enh17 inhibited cell proliferation, migration, and tumor growth of melanoma both in vitro and in vivo. Mechanistically, we identified ETV4 as a target gene regulated by enh17, and functional experiments further support ETV4 as a target gene that is involved in cancer-associated phenotypes. In addition, STAT3 acts as a transcription factor binding with enh17 to regulate the transcription of ETV4. Conclusions Our findings revealed that enh17 plays an oncogenic role and promotes tumor progression in melanoma, and its transcriptional regulatory mechanisms were fully elucidated, which may open a promising window for melanoma prevention and treatment.

Microalgae biofilm has garnered significant attention from researchers in the field of sewage treatment due to its advantages such as ease of collection and stable sewage treatment capabilities. Using agricultural waste as biofilm carriers has become a hotspot in reducing costs for this method. This study first combined Tetradesmus obliquus with loofah to form a microalgae biofilm for the study of periodic nitrogen and phosphorus removal from municipal wastewater. The biofilm could stably treat 7 batches of wastewater within one month. The removal rate of TP almost reached 100 %, while the removal rates of NH4+ and TN both reached or exceeded 80 %. The average biomass yield over 25 days was 102.04 mg/L/day. The polysaccharide content increased from 8.61 % to 16.98 % during the cyclic cultivation. The lipid content gradually decreased from 40.91 to 26.1 %. The protein content increased from 32.93 % in the initial stage to 41.18 % and then decreased to 36.31 % in the later stage. During the mid-stage of culturing, the richness of anaerobic bacteria decreased, while the richness of aerobic and facultative bacteria increased, which was conducive to the construction of the microalgae-bacteria symbiotic system and steadily improved the effect of nitrogen and phosphorus removal. As the culturing progressed, the Rotifers that emerged during the mid-stage gradually damaged the biofilm over time, leading to a decline in the effectiveness of sewage treatment in the later stages. This study offers technical support for carrier selection in microalgae biofilm methods and for the periodic removal of nitrogen and phosphorus from wastewater.

Climate change is driving global endeavours to achieve carbon neutrality and renewable energy expansion. Sludge, a nutrient-rich waste, holds energy potential yet poses environmental challenges that need proper management. We conducted a comprehensive life cycle assessment to evaluate the energy balance and environmental footprint of the most commonly used sludge management scenarios in BRICS countries, namely Brazil, Russia, India, China, and South Africa. Technologies such as incineration and anaerobic digestion with energy recovery units (i.e., cogeneration unit) maximize energy balance and minimize the environmental footprint, with incineration showing a superior performance. Shifting sludge management scenarios from the worst to the best can boost energy production by 1.4–98.4 times and cut the environmental footprint by 1.5–21.4 times. In 2050, these improvements could lead to a 98-fold boost in energy generation and a 25-fold drop in carbon emissions, according to the Announced Pledges Scenarios. Optimizing parameters such as volatile solids and anaerobic digestion efficiency further boosts energy output and minimizes the environmental footprint. This study offers robust evidence to support sustainable sludge management and thus promote energy recovery and carbon neutrality goals, guide technological transitions, and inform policymaking for sustainable development.

The detection of endogenous hormones is of great significance for disease diagnosis, treatment guidance, and the development of personalized healthcare. By using biosensors, real-time monitoring and quantitative analysis of endogenous hormone levels can be achieved, providing scientific basis for clinical decision-making. The continuous innovation in this field will bring new breakthroughs in medical diagnosis and treatment, which are expected to accelerate the early detection of diseases and the implementation of personalized treatment plans. In this review, we mainly review the research progress of endogenous hormone biosensors based on different biological recognition elements, namely enzyme-based biosensors, immunobiosensors, aptamer-based biosensors, and molecularly imprinted polymer (MIP) based biosensors. Meanwhile, this paper also analyzes the detection efficiency and clinical application of these biosensors. Finally, we summarize the current challenges and future development directions of the different types of biosensors involved in the discussion.

Abstract Background There has been a significant surge in the global prevalence of diabetes mellitus (DM), which increases the susceptibility of individuals to ovarian cancer (OC). However, the relationship between DM and OC remains largely unexplored. The objective of this study is to provide preliminary insights into the shared molecular regulatory mechanisms and potential biomarkers between DM and OC. Methods Multiple datasets from the GEO database were utilized for bioinformatics analysis. Single cell datasets from the GEO database were analysed. Subsequently, immune cell infiltration analysis was performed on mRNA expression data. The intersection of these datasets yielded a set of common genes associated with both OC and DM. Using these overlapping genes and Cytoscape, a protein‒protein interaction (PPI) network was constructed, and 10 core targets were selected. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were then conducted on these core targets. Additionally, advanced bioinformatics analyses were conducted to construct a TF-mRNA-miRNA coregulatory network based on identified core targets. Furthermore, immunohistochemistry staining (IHC) and real-time quantitative PCR (RT-qPCR) were employed for the validation of the expression and biological functions of core proteins, including HSPAA1, HSPA8, SOD1, and transcription factors SREBF2 and GTAT2, in ovarian tumors. Results The immune cell infiltration analysis based on mRNA expression data for both DM and OC, as well as analysis using single-cell datasets, reveals significant differences in mononuclear cell levels. By intersecting the single-cell datasets, a total of 119 targets related to mononuclear cells in both OC and DM were identified. PPI network analysis further identified 10 hub genesincludingHSP90AA1, HSPA8, SNRPD2, UBA52, SOD1, RPL13A, RPSA, ITGAM, PPP1CC, and PSMA5, as potential targets of OC and DM. Enrichment analysis indicated that these genes are primarily associated with neutrophil degranulation, GDP-dissociation inhibitor activity, and the IL-17 signaling pathway, suggesting their involvement in the regulation of the tumor microenvironment. Furthermore, the TF-gene and miRNA-gene regulatory networks were validated using NetworkAnalyst. The identified TFs included SREBF2, GATA2, and SRF, while the miRNAs included miR-320a, miR-378a-3p, and miR-26a-5p. Simultaneously, IHC and RT-qPCR reveal differential expression of core targets in ovarian tumors after the onset of diabetes. RT-qPCR further revealed that SREBF2 and GATA2 may influence the expression of core proteins, including HSP90AA1, HSPA8, and SOD1. Conclusion This study revealed the shared gene interaction network between OC and DM and predicted the TFs and miRNAs associated with core genes in monocytes. Our research findings contribute to identifying potential biological mechanisms underlying the relationship between OC and DM.

This paper investigates the integration of augmented reality (AR) technology into English translation teaching for college students, emphasizing the pivotal role of innovative teaching methods in enhancing students' translation skills and learning experiences. To address the issue of insufficient interest in English translation teaching, the paper initially assesses the purpose and significance of learning English translation through a questionnaire survey, elucidating challenges encountered in English language acquisition. Subsequently, adhering to AR principles, a teaching demonstration platform rooted in AR technology is conceived and developed, intricately aligned with English translation instruction. The platform serves as a solution to issues in English learning, such as inadequate course comprehension, low utilization of teaching resources, and instructors' lack of experience. The research culminates in the analysis of survey results, wherein the quantitative disparities in translation ability between students utilizing the research platform and those subjected to traditional teaching methods are examined. The findings underscore the positive impact of the AR-based research platform on improving students' translation proficiency. The AR platform heightens learners' engagement in the learning process, contributes to constructing a robust knowledge framework, and enhances overall learning outcomes. The platform offers educators opportunities to optimize experimental courses and elevate teaching standards. The paper's outcomes present novel pedagogical scenarios for learners, propose technical solutions for other technical disciplines and furnish a theoretical foundation and application model for a new generation of experimental demonstration platforms.

Abstract Artificial intelligence (AI) plays a key role in the distribution automation system (DAS). By using artificial intelligence technology, it is possible to intelligently verify and monitor distribution automation terminals, improve their safety and reliability, and reduce power system operating and maintenance costs. At present, researchers are exploring a variety of application methods and algorithms of the distribution automation terminal intelligent acceptance system based on artificial intelligence, such as machine learning, deep learning and expert systems, and have made significant progress. This paper comprehensively reviews the existing research on the application of artificial intelligence technology in distribution automation systems, including fault detection, network reconfiguration, load forecasting, and network security. It undertakes a thorough examination and summarization of the major research achievements in the field of distribution automation systems over the past few years, while also analyzing the challenges that this field confronts. Moreover, this study elaborates extensively on the diverse applications of AI technology within distribution automation systems, providing a detailed comparative analysis of various algorithms and methodologies from multiple classification perspectives. The primary aim of this endeavor is to furnish valuable insights for researchers and practitioners in this domain, thereby fostering the advancement and innovation of distribution automation systems.

Abstract Betweenness centrality is one of the key measures of the node importance in a network. However, it is computationally intractable to calculate the exact betweenness centrality of nodes in large-scale networks. To solve this problem, we present an efficient CBCA (Centroids based Betweenness Centrality Approximation) algorithm based on progressive sampling and shortest paths approximation. Our algorithm firstly approximates the shortest paths by generating the network centroids according to the adjacency information entropy of the nodes; then constructs an efficient error estimator using the Monte Carlo Empirical Rademacher averages to determine the sample size which can achieve a balance with accuracy; finally, we present a novel centroid updating strategy based on network density and clustering coefficient, which can effectively reduce the computation burden of updating shortest paths in dynamic networks. The experimental results show that our CBCA algorithm can efficiently output high-quality approximations of the betweenness centrality of a node in large-scale complex networks.

Aiming at the issues of poor hydration activity, long setting time, and low early strength in industrial by-product fluorogypsum (FG), 1,2,3-propenyltricarboxylic acid (PA) was utilized to enhance its reactivity. The mechanism was investigated using modern analytical techniques such as XRD, 1H low-field NMR, and FTIR spectroscopy. The results indicate that the incorporation of PA can significantly augment the rate of FG hydration and enhance its hydration activity. The optimal concentration of PA was determined to be 0.008%. The FG paste exhibited a reduction in setting time by approximately 500 min compared to the control sample. Furthermore, the flexural and compressive strengths increased by 29.6% (reaching 3.5 MPa) and 40.2% (reaching 40.1 MPa), respectively, after a curing period of 28 d. Through the process of physical adsorption, PA facilitates the transformation from anhydrite (CaSO4-II) to dihydrate gypsum (CaSO4·2H2O). Subsequently, PA promotes the growth of dihydrate gypsum crystals along each crystal axis, resulting in the formation of regular hexagonal prisms. The prismatic crystals of dihydrate gypsum are arranged in a closely packed, layered structure that results in reduced pore and porosity content, and enhanced mechanical strength.

Abstract Although the coronavirus disease (COVID-19) emergency status is easing, the COVID-19 pandemic continues to affect healthcare systems globally. It is crucial to have a reliable and population-wide prediction tool for estimating COVID-19-induced hospital admissions. We evaluated the feasibility of using wastewater-based epidemiology (WBE) to predict COVID-19-induced weekly new hospitalizations in 159 counties across 45 states in the United States of America (USA), covering a population of nearly 100 million. Using county-level weekly wastewater surveillance data (over 20 months), WBE-based models were established through the random forest algorithm. WBE-based models accurately predicted the county-level weekly new admissions, allowing a preparation window of 1-4 weeks. In real applications, periodically updated WBE-based models showed good accuracy and transferability, with mean absolute error within 4-6 patients/100k population for upcoming weekly new hospitalization numbers. Our study demonstrated the potential of using WBE as an effective method to provide early warnings for healthcare systems.

Abstract Background Thrombosis of one of the umbilical arteries is a rare complication of pregnancy and is associated with adverse pregnancy outcomes, including stillbirth and intrauterine growth restriction. Although extremely rare, umbilical artery thrombosis (UAT) in monochorionic diamniotic twins is difficult to diagnose prenatally and manage. UAT has a poor prognosis and is associated with an increased perinatal mortality rate. In most previous cases, emergency cesarean section was performed or intrauterine fetal death occurred at the time of UAT diagnosis. Case presentation: Herein, we report an extremely rare case of sequential UAT in monochorionic diamniotic twins diagnosed via ultrasound at 29+ 5 weeks of gestation in a 34-year-old woman. Following expectant management with intensive monitoring for 16 days, two healthy infants were delivered through an emergency cesarean section. UAT in both fetuses was confirmed by pathological examination. The mother and twins described in this case underwent long-term follow-up and are currently in good health without any complications. Conclusions Based on our experience, we suggest that expectant management should be undertaken as long as the mother and infants are stable on ultrasonographic scans and are closely monitored. When UAT is suspected, we believe that the best delivery time should be determined by considering complaints of unusual fetal movements, non-stress test evidence, gestational age, amniotic fluid volume, and blood flow in the umbilical artery, middle cerebral artery, and ductus venosus. Obstetricians should ensure that the patients and their families are clearly informed about all potential risks of expectant management for UAT.

Phenazine-based redox-active centers are capable of averting chemical bond rearrangements by coupling during the reaction process, leading to enhanced stabilization of the material. When introduced into a high-performance polymer with excellent physicochemical properties, they can be endowed with electrochemical properties and related prospective applications while maintaining the capabilities of the materials. In this study, a facile C-N coupling method was chosen for the synthesis of serial poly(aryl ether sulfone) materials containing phenazine-based redox-active centers and to explore their electrochemical properties. As expected, the cyclic voltammetry curves of PAS-DPPZ-60, which basically overlap after thousands of cycles, indicate the stability of the electrochemical properties. As an electrochromic material, the transmittance change in PAS-DPPZ-60 exhibits only a slight attenuation after as long as 600 cycles. Meanwhile, as an organic battery cathode material, PAS-DPPZ has a theoretical specific capacity of 126 mAh g−1, and the capacity retention rate is 82.6% after 100 cycles at a 0.1 C current density. The perfect combination of advantageous features between phenazine and poly(aryl ether sulfone) is considered to be the reason for the favorable electrochemical performance of the material series.

Trace elements of metallic minerals are important to constrain the formation processes and genetic types of ore deposits. Trace elements have been mainly focused on pyrite, magnetite, and sphalerite but have rarely been applied to chalcopyrite. To reveal the relationships between chalcopyrite trace elements and ore deposit types, ore petrography and LA-ICP-MS trace element of chalcopyrite in collected copper concentrate samples from porphyry copper deposits (PCD), magmatic copper-nickel sulfide deposits (MSD), sedimentary rock-hosted stratiform copperdeposits (SSC), iron oxide copper-gold deposits (IOCG), sedimentary exhalative deposits (SEDEX) and volcanogenic massive sulfide deposits (VMS) have been carried out. In chalcopyrite, Mn, Co, Ni, Se, Ag, Sn, Pb, and Bi contents are more than 1 000×10-6, and Ga, Ge, Mo, Cd, In, Sb, Te, Au and Tl contents are up to 100×10-6, which together indicate chalcopyrite is an important carrier for many trace elements. Antimony-Tl, In-Sn, Pb-Bi, and Mn-Ni in the chalcopyrite are positively correlated.Meanwhile, in chalcopyrite, Sb, Tl, In, and Sn mainly occur in the form of solid solution, Pb and Bi in the form of galena inclusions, and Mn, Co, As, Te, Ag, and Ni are both developed. Trace elements of chalcopyrite from PCD and VMS are variable. The concentrations of Ni and In in chalcopyrite from MSD are high and low, and Ge and Sn from SSC are higher and lower than other types, respectively. Moreover, the concentration of Se in chalcopyrite is higher from MSD and VMS, but is lower from SEDEX and SSC. Different concentrations of Ni, In, and Sn in the chalcopyrite are mainly related to different magmatism, and Se is principally controlled by temperature. The high concentrations of Ge in chalcopyrite from SSC may be related to ore-forming temperature and host rocks.Therefore, based on the above trace elements characteristics, Ni-Co and Ni-In diagrams can distinguish MSD from other deposit types, the diagram of Ni-Se can differentiate SEDEX, SSC from VMS, and the diagram of Ge-Sn is used to isolate SSC from SEDEX. In addition, the diagram of Co/Ni-Ag/Bi can differentiate between MSD and PCD, while Zn-Sn/In can discriminate IOCG from others to some extent.These first systematically proposed diagrams will provide a new reference for distinguishing the genetic types of deposits.

To achieve sustainable development, the energy transition from lignite burning to renewable energy resources for electric power generation is essential for Greece. Wind and solar energy have emerged as significant sources in this transition. Surprisingly, numerous studies have examined the potential for onshore wind based on land eligibility, while few studies on open-field photovoltaic (PV) installations have been conducted. Therefore, based on the Specific Framework for Spatial Planning and Sustainable Development for Renewable Energy Sources (SFSPSD-RES), along with insights from previous relevant studies, this work conducts a land eligibility analysis of onshore wind and open-field PV installations in Greece using the software Geospatial Land Availability for Energy Systems (GLAES 1.2.1) and ArcGIS 10.2. Additionally, through an in-depth exploration of wind and solar PV energy potential in decommissioned lignite mines integrated with wind power density (WPD) and global horizontal irradiation (GHI) maps, this study compares the suitability of wind versus solar as energy sources for the decarbonization of Greece. Overall, despite the greater spatial eligibility for onshore wind turbines compared to open-field PV power plants, the relatively lower wind energy potential and operational limitations of wind turbines lead to the study’s conclusion that solar energy (PV) is more suitable for the decarbonization of Greece.

The development and deployment of diverse resistance sources in new wheat cultivars underpin the durable control of stripe rust. In the present study, two loci for adult plant resistance (APR), QYrSM155.1 and QYrSM155.2, were identified in the Chinese wheat breeding line Shaanmai 155. QYrSM155.1 was mapped to a 3.0-cM interval between the single-nucleotide polymorphism (SNP) markers AX-109583610 and AX-110907562 on chromosome arm 2BL. QYrSM155.2 was mapped to a 2.1-cM interval flanked by the SNP markers AX-110378556 and AX-86173526 on chromosome arm 7AS. A genome-wide association study was used to identify markers associated with APR in a panel of 411 spring wheat lines. Thirteen and 11 SNPs were significantly associated with QYrSM155.1 and QYrSM155.2, respectively, corresponding to physical intervals of 653.75–655.52 Mb on 2BL and 81.63–83.93 Mb on 7AS. To characterize the haplotype variation and the distribution of these QTL, haplotype analysis was performed based on these SNPs in an independent panel of 1101 worldwide wheat accessions. Three major haplotypes (2B_h1, 2B_h2, and 2B_h3) for QYrSM155.1 and four major haplotypes (7A_h1, 7A_h2, 7A_h3, and 7A_h4) for QYrSM155.2 were identified. Accessions individually harboring QYrSM155.1_h1 and QYrSM155.2_h1 haplotypes and their combination displayed resistance. Additional assays of 1306 current Chinese cultivars and breeding lines using markers flanking QYrSM155.1 and QYrSM155.2 indicated that the resistance haplotypes of the two QTL were present in respectively 1.45% and 14.16% of lines. Increasing resistance haplotype frequencies at these two loci using marker-assisted selection should benefit wheat production in China.

In this paper, we study the optimality conditions for set optimization problems with set criterion. Firstly, we establish a few important properties of the Minkowski difference for sets. Then, we introduce the generalized second-order lower radial epiderivative for a set-valued maps by Minkowski difference, and discuss some of its properties. Finally, by virtue of the generalized second-order lower radial epiderivatives and the generalized second-order radial epiderivatives, we establish the necessary optimality conditions and sufficient optimality conditions of approximate Benson proper efficient solutions and approximate weakly minimal solutions of unconstrained set optimization problems without convexity conditions, respectively. Some examples are provided to illustrate the main results obtained.

Bones are constantly exposed to mechanical forces from both muscles and Earth’s gravity to maintain bone homeostasis by stimulating bone formation. Mechanotransduction transforms external mechanical signals such as force, fluid flow shear, and gravity into intracellular responses to achieve force adaptation. However, the underlying molecular mechanisms on the conversion from mechanical signals into bone formation has not been completely defined yet. In the present review, we provide a comprehensive and systematic description of the mechanotransduction signaling pathways induced by mechanical stimuli during osteogenesis and address the different layers of interconnections between different signaling pathways. Further exploration of mechanotransduction would benefit patients with osteoporosis, including the aging population and postmenopausal women.

A novel carbon quantum dots decorated C-doped α-Bi2O3 photocatalyst (CBO/CQDs) was synthesized by solvothermal method. The synergistic effect of adsorption and photocatalysis highly improved contaminants removal efficiencies. The ceftriaxone sodium degradation rate constant (k) of CBO/CQDs was 11.4 and 3.2 times that of pure α-Bi2O3 and C-doped α-Bi2O3, respectively. The interstitial carbon doping generated localized states above the valence band, which enhanced the utilization of visible light and facilitated the separation of photogenerated electrons and holes; the loading of CQDs improved the charge carrier separation and extended the visible light response; the reduced particle size of CBO/CQDs accelerated the migration of photogenerated carriers. The •O2− and h+ were identified as the dominant reactive species in ceftriaxone sodium degradation, and the key role of •O2− was further investigated by NBT transformation experiments. The Fukui index was applied to ascertain the molecular bonds of ceftriaxone sodium susceptible to radical attack, and intermediates analysis was conducted to explore the possible degradation pathways. The toxicity evaluation revealed that some degradation intermediates possessed high toxicity, thus the contaminants require sufficient mineralization to ensure safe discharge. The present study makes new insights into synchronous carbon dopping and CQDs decoration on modification of α-Bi2O3, which provides references for future studies.

Chlorination disinfection has been widely used to kill the pathogenic microorganisms in wastewater sludge during the special Covid-19 period, but sludge chlorination might cause the generation of harmful disinfection byproducts (DBPs). In this work, the transformation of extracellular polymeric substance (EPS) and mechanisms of Cl-DBPs generation during sludge disinfection by sodium hypochlorite (NaClO) were investigated using multispectral analysis in combination with Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS). The microorganism Escherichia coli (E. coli) was effectively inactivated by active chlorine generated from NaClO. However, a high diversity of Cl-DBPs were produced with the addition of NaClO into sludge, causing the increase of acute toxicity on Q67 luminous bacteria of chlorinated EPS. A variety of N-containing molecular formulas were produced after chlorination, but N-containing DBPs were not detected, which might be the indicative of the dissociation of –NH2 groups after Cl-DBPs generated. Additionally, the release of N-containing compounds was increased in alkaline environment caused by NaClO addition, resulted in more Cl-DBPs generation via nucleophilic substitutions. Whereas, less N-compounds and Cl-DBPs were detected after EPS chlorination under acidic environment, leading to lower cell cytotoxicity. Therefore, N-containing compounds of lignin derivatives in sludge were the major Cl-DBPs precursors, and acidic environment could control the release of N-compounds by eliminating the dissociation of functional groups in lignin derivatives, consequently reducing the generation and cytotoxicity of Cl-DBPs. This study highlights the importance to control the alkalinity of sludge to reduce Cl-DBPs generation prior to chlorination disinfection process, and ensure the safety of subsequential disposal for wastewater sludge.

High-performance donor-acceptor (D-A) polymers, as an important class of electrochromic (EC) materials, have attracted extensive attention. In this paper, a series of novel poly (aryl amino ketone) (PAAK) and poly (aryl amino sulfone) (PAAS) type high-performance polymers (HPP) with electrochromism were prepared by a simple C-N coupling reaction and were coated on an indium tin oxide (ITO) substrate as EC films. All four polymers were prepared by a nucleophilic substitution reaction using commercially purchased amine monomers with difluoride sulfone/ketone using potassium carbonate as a catalyst. A series of tests were performed to compare and analyze the effects of the different electron-withdrawing abilities of sulfone and carbonyl groups, and the different conjugation lengths of these two TPA structures were connected to the EC properties of the polymer. The different phenyl or biphenyl of the two TPA structures mainly affected the oxidation potential of the polymer, while the sulfone group and the carbonyl group, with a different electron absorption ability, had a greater influence on the energy band and cyclic stability. The optical contrast of PAAS−BT at 850 nm was up to 58% and maintained 450 cycles, indicating that this series of materials had a broad application prospect waiting for further research. In addition to the performance, the raw materials used in this work could be directly and commercially purchased for a low price; the two aniline monomers were priced at about $0.43 /g and $0.15 /g, respectively. This method significantly reduces the cost and provides a new idea for subsequent large-scale production and practical applications.

The use of silicon (Si) as a lithium-ion battery’s (LIBs) anode active material has been a popular subject of research, due to its high theoretical specific capacity (4200 mAh g−1). However, the volume of Si undergoes a huge expansion (300%) during the charging and discharging process of the battery, resulting in the destruction of the anode’s structure and the rapid decay of the battery’s energy density, which limits the practical application of Si as the anode active material. Lithium-ion batteries’ capacity, lifespan, and safety can be increased through the efficient mitigation of Si volume expansion and the maintenance of the stability of the electrode’s structure with the employment of polymer binders. The main degradation mechanism of Si-based anodes and the methods that have been reported to effectively solve the Si volume expansion problem firstly are introduced. Then, the review demonstrates the representative research work on the design and development of new Si-based anode binders to improve the cycling stability of Si-based anode structure from the perspective of binders, and finally concludes by summarizing and outlining the progress of this research direction.

Transient stability assessment (TSA) has always been a fundamental and challenging problem for ensuring the security and operation of power systems. With more power electronic interface resources integrated into the grid and large renewable energies, the stability of the power system is jeopardized. Therefore, TSA of the power system should be considered in advance to keep the system running stable. In recent years, with the development of artificial intelligence (AI) technologies such as artificial neural network (ANN), support vector machine (SVM), and Markov decision process, TSA has improved dramatically. In this study, a sparse dictionary learning approach is proposed to improve the precision of the classification accuracy of transient stability assessment in power systems. Case studies of TSA using multi-layer support vector machine (ML-SVM) and long short-term memory network–based recurrent neural network (LSTM-RNN) are discussed as benchmarks to validate the proposed method. The stable and unstable dictionary learnings are designed based on datasets obtained by simulating thousands of different time-domain simulation (TDS) scenarios performed on the New-England 39-bus system in the PSAT (power system analysis toolbox) toolbox. Stable and unstable dictionaries are developed based on the K-SVD approach. The testing dataset contains both stable and unstable samples which steps into the sparse coding process to obtain the indexes. Compared with the indexes, the system’s final TSA is targeted. The proposed method exhibits satisfactory classification accuracy in transient stability prediction and provides the ability to reduce false alarms both in positives and negatives of the power system.

For aquaculture wastewater with low ammonium nitrogen concentration, combining the carrier adsorption and biological nitrogen removal processes can maximize their respective advantages. Functional ceramsite that has excellent ammonium nitrogen adsorption performance and excellent biocompatibility was the key to the moving bed biofilm reactor (MBBR) adsorption—shortcut simultaneous nitrification and denitrification (shortcut SND) process. Our group prepared a high-strength lightweight ceramsite that met those requirements. In this study, we applied functional ceramsite in MBBR to cope with low-concentration ammonium aquaculture wastewater. The findings show that utilizing functional ceramsite as a filler was conducive to the adhesion of microorganisms. The biofilm has a minimal effect on the adsorption capacity of ceramsite due to the existence of pores on its surface. Our study further examined the NH4+-N adsorption mechanism of bio-ceramsite. The Freundlich adsorption isotherm model and the quasi-second-order kinetic model had better fitting effects on the NH4+-N adsorption process. The adsorption of bio-ceramsite to NH4+-N was an endothermic process that included physical and chemical adsorption. Furthermore, the results of adsorption thermodynamics suggested that bio-ceramsite has an affinity for the adsorption of NH4+-N. Consequently, this functional ceramsite can be a promising option for MBBR to improve nitrogen removal from aquaculture wastewater.

Excessive discharge of ammonia nitrogen wastewater from intensive aquaculture has worsened in recent years. Therefore, there is an urgent need to develop an effective and energy-saving denitrification technology. This study intends to adopt a moving bed biofilm reactor (MBBR) to remove ammonia nitrogen through the combination of adsorption and shortcut simultaneous nitrification and denitrification (SND). The research focuses on the operational parameters and regeneration mechanism of the MBBR adsorption-shortcut SND process. The optimal operating parameters in the adsorption stage were a hydraulic retention time of 8 h and an agitation rate of 120 r/min. For the shortcut SND stage, the ideal optimal parameters were two times alkalinity and dissolved oxygen (DO) 1.0 mg/L. Under optimal operating parameters conditions, the SND rate, TN removal rate, NH4+-N removal rate and nitrite accumulation rate were 89.1%, 84.0%, 94.3%, and 86.4%, respectively. The synergetic actions of ion exchange and microorganisms were the main driving force for regenerating ceramsite zeolite components. The synergistic inhibitory effect of high-concentration free ammonia and low-level DO on nitrite-oxidizing bacteria was the key to achieving stable and efficient NO2−-N accumulation. NO2−-N produced in shortcut nitrification entered the ceramsite through complex mass transfer, and denitrifying bacteria can reduce these NO2−-N to N2.

In recent years, donor-acceptor (D-A)-type conjugated polymers have been widely used in the field of organic solar cells (OSCs) and electrochromism (EC). Considering the poor solubility of D-A conjugated polymers, the solvents used in material processing and related device preparation are mostly toxic halogenated solvents, which have become the biggest obstacle to the future commercial process of the OSC and EC field. Herein, we designed and synthesized three novel D-A conjugated polymers, PBDT1-DTBF, PBDT2-DTBF, and PBDT3-DTBF, by introducing polar oligo (ethylene glycol) (OEG) side chains of different lengths in the donor unit benzodithiophene (BDT) as side chain modification. Studies on solubility, optics, electrochemical, photovoltaic and electrochromic properties are conducted, and the influence of the introduction of OEG side chains on its basic properties is also discussed. Studies on solubility and electrochromic properties show unusual trends that need further research. However, since PBDT-DTBF-class polymers and acceptor IT-4F failed to form proper morphology under the low-boiling point solvent THF solvent processing, the photovoltaic performance of prepared devices is not ideal. However, films with THF as processing solvent showed relatively desirable electrochromic properties and films cast from THF display higher CE than CB as the solvent. Therefore, this class of polymers has application feasibility for green solvent processing in the OSC and EC fields. The research provides an idea for the design of green solvent-processable polymer solar cell materials in the future and a meaningful exploration of the application of green solvents in the field of electrochromism.

Giardia duodenalis is the underlying cause of a significant number of outbreaks of gastrointestinal illness in humans and animals worldwide. The purpose of this study was to elucidate the prevalence and genetic diversity of G. duodenalis in captive alpine musk deer (Moschus chrysogaster) in China. A total of 202 fecal samples were collected from three farms in Gansu Province, China. Identification of G. duodenalis was conducted by nested PCR targeting the genes coding for SSU rRNA, β-giardin (bg), glutamate dehydrogenase (gdh) and triosephosphate isomerase (tpi). The overall prevalence of G. duodenalis in captive alpine musk deer in surveyed area was 19.3% (39/202). Two G. duodenalis genetic assemblages were identified, namely assemblage A and E. Mixed genotype infections (A+E) were found in 15.4% (6/39) of positive samples. Multilocus genotyping (MLG) analysis of G. duodenalis isolates revealed six novel assemblage A MLGs formed by two newly-described MLG-subtypes which belonged to sub-assemblage AI. To the best of our knowledge, this is the first report on MLG of G. duodenalis isolates in captive alpine musk deer in China. The presence of zoonotic assemblages and sub-assemblages of G. duodenalis in deer species suggests that these animals may potentially act as a reservoir of this protozoan for humans.

Abstract Background Reversible acetylation of α-tubulin has been implicated in modulating microtuble structures and functions, which may subsequently involve in cellular apoptosis and autophage. But how to trigger apoptosis or autophage at what level of acetylated α-tubulin (Ac-α-tubulin) are not known. This study aims to demonstrate the dual functions and molecular mechanisms of α-tubulin acetylation in cellular apoptosis and autophage induced by tanespimycin in Calu-1 cells simultaneously. Methods Calu-1 cells were treated with tanespimycin alone or combined administrations of different agents (including TSA, Docetaxel, Rapamycin, 3-MA and Z-vad) respectively and cell lysates were prepared to detect the given proteins by Western Blot. The cell survival was observed by inverted phase contrast microscope and estimated by SRB assay. HDAC6, TAT1 and Hsp90α/β proteins were knocked down by siRNA technique. Results By combination administration of tanespimycin with TSA or Docetaxel, the expression of Ac-α-tubulin and cellular apoptosis were enhanced markedly. While combination of tanespimycin and Rapamycin, α-tubulin acetylation and apoptosis were inhibited, but LC3B-II expression was facilitated substantially. When tanespimycin was combined with autophage inhibitor 3-MA, α-tubulin acetylation elevation was apparently, but LC3B-II was attenuated. Apoptosis inhibitor Z-vad blocked partially Caspases activation induced by tanespimycin, but failed to hinder α-tubulin acetylation elevation. According to results of RNA interference, acetyltransferase TAT1, deacetylase HDAC6 and Hsp90 modulated the expression level of α-tubulin acetylation. Conclusion We have elucidated that acetylation of α-tubulin induced by tanespimycin has dual functions in cellular apoptosis and autophage and the level of α-tubulin acetylation reaches a degree Calu-1 cells undergo cell apoptosis rather than autophage, implying that the level of acetylated α-tubulin may determine cell fate for survival or apoptosis.

Abstract Background Penile squamous cell carcinoma (PSCC) represents an important public health problem for developing countries. The major prognostic factors in PSCC are pathological subtype, perineural invasion, lymphovascular invasion, depth of invasion and grade, which are hard to obtain precisely before the operation. Besides, micro-metastases will be detected in about 30% of intermediate-risk patients with clinically non-palpable inguinal lymph nodes after inguinal lymph node dissection (ILND). It means approximately 70% of patients are unable to benefit from ILND who might suffered from the complications of surgery. We hope some biomarkers could be found which are able to predict the outcome before surgery and reflect the inguinal lymph nodes metastasis. Methods A total of 349 consecutive patients of penile cancer in Yunnan cancer hospital in China between October 2002 and December2017. Two hundred twenty-five was succeed to follow-up. The association between NLR, LMR, PLR, LDH and Overall survival (OS), progression free survival (PFS), inguinal lymph node (N stage) was analyzed with K-M analysis, univariable, multivariable logistic regression and Kendall’s tau-b correlation coefficient. Results Multivariable analysis reveal that only PLR was significant independent factor which is associated with inferior OS and PFS; Age and LDH was associated with inferior OS; Lymph node and metastatic status remained significant for OS and PFS as NCCN and EAU Guidelines indicated; the tumor type, initial treatment and NLR LMR were not significant in predicting both OS and PFS. NLR, LMR and PLR were corresponded to N stage, while LDH was not associated with the N stage based on logistic regression model analysis. NLR, LMR and PLR were found weakly related to N stage through an application of Kendall’s tau-b correlation coefficient. Conclusions PLR was significant independent factors for OS and PFS, Age and LDH was significant independent factors for OS. NLR, LMR, PLR was corresponded to N stage.

Functional polymers featuring good processability in non-halogenated, benzene-free green solvents are highly desired due to health and environmental concerns. Herein, a series of novel D-A type conjugated polymers, PBDT-IIDs, are designed and successfully prepared by “green” functionalization of the polymers with highly hydrophilic, highly polar, highly flexible, and biocompatible oligoethylene glycol (OEG) side chains in order to improve the processability. These series polymers are named PBDT-IID2, PBDT-IID3, and PBDT-IID4, respectively, according to the number of oxygen atoms in the side chain. After confirmation by structural characterization, the basic properties of PBDT-IIDs are also investigated. With the increase in the OEG side chain length, the polymer PBDT-IID4 not only has good solubility in the halogen solvent chlorobenzene, but also exhibits excellent solubility in the green halogen-free solvent methyltetrahydrofuran (Me-THF). As a result, the green solvent Me-THF can also be applied to prepare PBDT-IIDs’ electrochromic active layers, except for chlorobenzene and toluene. The electrochromism of PBDT IIDs under both positive and negative voltages has a practical application potential. The several controllable switches between dark green and khaki (0–0.6 V) are expected to show great potential in the field of military camouflage. Furthermore, according to the principle of red, green, and blue (RGB) mixing, light blue-green in the reduced state (−1.6 V) can be used in the preparation of complementary ECDs to provide one of the three primary colors (green).

In this paper, we introduce a second-order strong subdifferential of set-valued maps, and discuss some properties, such as convexity, sum rule and so on. By the new subdifferential and its properties, we establish a necessary and sufficient optimality condition of set-based robust efficient solutions for the uncertain set-valued optimization problem. We also introduce a Wolfe type dual problem of the uncertain set-valued optimization problem. Finally, we establish the robust weak duality theorem and the robust strong duality theorem between the uncertain set-valued optimization problem and its robust dual problem. Several main results extend to the corresponding ones in the literature.

Ticks are dangerous ectoparasites of humans and animals, as they are important disease vectors and serve as hosts for various microorganisms (including a variety of pathogenic microorganisms). Diverse microbial populations coexist within the tick body. Metagenomic next-generation sequencing (mNGS) has been suggested to be useful for rapidly and accurately obtaining microorganism abundance and diversity data. In this study, we performed mNGS to analyze the microbial diversity of Haemaphysalis longicornis from Baoji, Shaanxi, China, with the Illumina HiSeq platform. We identified 189 microbial genera (and 284 species) from ticks in the region; the identified taxa included Anaplasma spp., Rickettsia spp., Ehrlichia spp., and other important tick-borne pathogens at the genus level as well as symbiotic microorganisms such as Wolbachia spp., and Candidatus Entotheonella. The results of this study provide insights into possible tick-borne diseases and reveal new tick-borne pathogens in this region. Additionally, valuable information for the biological control of ticks is provided. In conclusion, this study provides reference data for guiding the development of prevention and control strategies targeting ticks and tick-borne diseases in the region, which can improve the effectiveness of tick and tick-borne disease control.

MicroRNAs (miRNAs), which are small, noncoding RNA molecules, play an important regulatory role in gene expression at the posttranscriptional level. Relatively limited knowledge exists on miRNAs in Rhipicephalus microplus ticks in China; however, understanding the physiology of miRNA functions and expression at different developmental stages is important. In this study, three small RNA libraries were constructed for R. microplus eggs, larvae, and female adults; miRNAs were detected during these developmental stages by high-throughput sequencing, with 18,162,337, 8,090,736, and 11,807,326 clean reads, respectively. A total of 5132 known miRNAs and 31 novel miRNAs were identified. A total of 1736 differentially expressed miRNAs were significantly different at a p-value of R. microplus and their potential targets, which has significance for understanding the biology of ticks and lays the foundation for improved understanding of miRNA functioning in the regulation of R. microplus development. These results can assist future miRNA studies in other tick species that have great significance for human and animal health.

Pyroptosis is a type of programmed cell death (PCD) accompanied by an inflammatory reaction and the rupture of a membrane. Pyroptosis is divided into a canonical pathway triggered by caspase-1, and a non-canonical pathway independent of caspase-1. More and more pyroptosis-related participants, pathways, and regulatory mechanisms have been exploited in recent years. Pyroptosis plays crucial roles in the initiation, progression, and metastasis of cancer and it affects the immunotherapeutic outcome by influencing immune cell infiltration as well. Extensive studies are required to elucidate the molecular mechanisms between pyroptosis and cancer. In this review, we introduce the discovery history of pyroptosis, delineate the signaling pathways of pyroptosis, and then make comparisons between pyroptosis and other types of PCD. Finally, we provide an overview of pyroptosis in different cancer types. With the progression in the field of pyroptosis, new therapeutic targets and strategies can be explored to combat cancer.

The complete mitochondrial genome of Hemibarbus labeo BML (Cypriniformes: Cyprinidae) is 16,612 bases in length. It consists of 22 tRNA genes, 13 protein-coding genes, 2 rRNA genes, and 1 non-coding region. Its overall nucleotide composition is A: 29.85%, G: 17.07%, T: 25.86%, and C: 27.23%, respectively, with an A + T rich feature (55.71%). The gene arrangement and organization of the mitogenome from H. labeo BML were very similar to other Cyprinidae fishes. The phylogenetic analysis showed that H. labeo BML clustered in genus Hemibarbus. These results will contribute to the taxonomy and conservation biology studies of H. labeo.

Abstract Background Bread wheat is one of the most important and broadly studied crops. However, due to the complexity of its genome and incomplete genome collection of wild populations, the bread wheat genome landscape and domestication history remain elusive. Results By investigating the whole-genome resequencing data of 93 accessions from worldwide populations of bread wheat and its diploid and tetraploid progenitors, together with 90 published exome-capture data, we find that the B subgenome has more variations than A and D subgenomes, including SNPs and deletions. Population genetics analyses support a monophyletic origin of domesticated wheat from wild emmer in northern Levant, with substantial introgressed genomic fragments from southern Levant. Southern Levant contributes more than 676 Mb in AB subgenomes and enriched in the pericentromeric regions. The AB subgenome introgression happens at the early stage of wheat speciation and partially contributes to their greater genetic diversity. Furthermore, we detect massive alien introgressions that originated from distant species through natural and artificial hybridizations, resulting in the reintroduction of ~ 709 Mb and ~ 1577 Mb sequences into bread wheat landraces and varieties, respectively. A large fraction of these intra- and inter-introgression fragments are associated with quantitative trait loci of important traits, and selection events are also identified. Conclusion We reveal the significance of multiple introgressions from distant wild populations and alien species in shaping the genetic components of bread wheat, and provide important resources and new perspectives for future wheat breeding.

Wheat crops in China are constantly challenged by stripe rust. Deployment of cultivars with diverse resistances is the best strategy to control the disease. A recombinant inbred line (RIL) population derived from a cross between the resistant cultivar Chakwal 86 and susceptible landrace Mingxian 169 was studied in multiple environments to examine the underlying genetics and to identify quantitative trait loci (QTL) for stripe rust resistance. One hundred and twenty-eight RILs were genotyped with wheat 35K SNP array and a genome-wide linkage map with 1480 polymorphic SNP markers, or bins, was constructed. Two major QTL on chromosomes 1BL and 3BS, and one minor QTL on 6BS had significant effects in reducing stripe rust severity. The QTL were validated using composite interval mapping (CIM) and inclusive composite interval mapping (ICIM). These methods explained 59.0%–74.1% of the phenotype variation in disease response. The QTL on chromosome 1BL was confirmed to be Yr29/Lr46 and the one on 3BS was the resistance allele identified in CIMMYT germplasm but was not Yr30/Sr2. The QTL on 6BS probably corresponded to previously known QTL. RILs with combined QTL were more resistant than those with single or no QTL. Kompetitive allele-specific PCR (KASP) assays for the QTL with largest effect QTL on chromosome 3BS were performed on a subset of RILs and 150 unrelated wheat lines. The QTL on 3BS with its linked KASP markers can be used in marker-assisted selection to improve stripe rust resistance in breeding programs. The purposes are to monitor the nitrogen utilization efficiency of crops and intelligently evaluate the absorption of nutrients by crops during the production process. The research object is Chinese cabbage. The Chinese cabbage population with different agricultural parameters is constructed through different densities and nitrogen fertilizer application rates based on digital image processing technology, and an estimation NC (Nitrogen Content) model is established. The population is classified through the K-Means Clustering algorithm using the feature extraction method, and the Chinese cabbage population quality BPNN (Backpropagation Neural Network) model is constructed. The nonlinear mapping relationship between different agricultural parameters and population quality, and the contribution rate of each indicator, are studied. The nitrogen utilization of Chinese cabbage is monitored effectively. Results demonstrate that the proposed NC estimation model has correlation coefficients above 0.70 in different growth stages. This model can accurately estimate the NC of the Chinese cabbage population. The results of the Chinese cabbage population quality BPNN model show that the population planting density based on the seedling number is reasonable. The constructed population quality evaluation model has a high R2 value and a comparatively low RMSE (Root Mean Square Error) value for the quality evaluation of Chinese cabbage in different periods, showing that it applies to evaluate the population quality of Chinese cabbage in different growth stages. The constructed nitrogen utilization model and quality evaluation model can monitor the nutrient utilization of crops in different growth stages, ascertain the agricultural characteristics of other yield groups in different growth stages, and clarify the performance of agricultural parameters in different growth stages. The above results can provide some ideas for crop growth intelligent detection.

Urothelial carcinoma (UC) is a common malignancy of the lower and upper urinary tract. Recurrent UC has poor prognosis due to delayed diagnosis and a lack of clinical management guidance, especially for upper urinary tract UC. Patients with germline or somatic BRCA1/2 mutations are a special population in UC. No evidence is available so far on the effectiveness of poly ADP-ribose polymerase inhibitor (PARPi) in this population. Here, we report a 60-year-old female patient diagnosed with left ureter high-grade UC. Recurrent lesions were found 20 months after radical surgery. Computed tomography (CT) examination showed a slightly high-density soft tissue mass (3.2 × 3.1 cm) on the left posterior wall of the abdomen (waist), soft tissue mass adjacent to the left inner wall of the pelvis (3.2 × 4.2 cm), and multiple enlarged lymph nodes to the left of abdominal aorta. A next-generation sequencing (NGS)-based 605-gene panel detected a novel BRCA2 pathogenic germline mutation c.1670T>A (p.L557*), and a series of somatic insertion and deletion (INDEL) mutations of BRCA1, RB1, and JAK2, and single nucleotide variation (SNV) mutations of TP53, KMT2D, MET, ROS1, and IL7R. The above lesions were reduced significantly or disappeared (partial response, PR) after a 3-month Olaparib treatment, and the patient’s general condition remained well. In conclusion, this study proved for the first time that PARPi was effective for UC treatment in patients carrying germline BRCA2 pathogenic mutations, providing new treatment options for such patients. In addition, the circulating tumor DNA (ctDNA) test can be used for drug selection and response monitoring in UC treatment.

Natural protein-based hydrogels possess excellent biocompatibility; however, most of them are weak or brittle. In the present work, high strength hybrid dual-crosslinking BSA gels (BSA DC gels), which have both chemical cross-linking and physical cross-linking, were fabricated by a facile photoreaction-heating process. BSA DC gels showed high transparency (light transmittance of ~90%) and high strength. At optimal conditions, BSA DC gel exhibited high compressive strength (σc,f) of 37.81 ± 2.61 MPa and tensile strength (σt,f) of 0.62 ± 0.078 MPa, showing it to be much stronger than physically cross-linked BSA gel (BSA PC gel) and chemically cross-linked BSA gel (BSA CC gel). More importantly, BSA DC gel displayed non-swelling properties while maintaining high strength in DI water, pH = 3.0, and pH = 10.0. Moreover, BSA DC gel also demonstrated large hysteresis, rapid self-recovery, and excellent fatigue resistance properties. It is believed that our BSA DC gel can potentially be applied in biomedical fields.

Integrating denitrifying anaerobic methane oxidation (DAMO) with Anammox provides alternative solutions to simultaneously remove nitrogen and mitigate methane emission from wastewater treatment. However, the practical application of DAMO has been greatly limited by slow-growing DAMO microorganisms living on low-solubility gaseous methane. In this work, DAMO and Anammox co-cultures were fast enriched using high concentration of mixed sludges from various environments, and achieved nitrogen removal rate of 76.7 mg NH4+-N L−1 d−1 and 87.9 mg NO3−-N L−1 d−1 on Day 178. Subsequently, nitrogen removal rate significantly decreased but recovered quickly through increasing methane flushing frequency, indicating methane availability could be the limiting factor of DAMO activity. Thus, this work developed a novel Membrane Aerated Membrane Bioreactor (MAMBR), which equipped with gas permeable membrane for efficient methane delivery and ultrafiltration membrane for complete biomass retention. After inoculated with enriched sludge, nitrogen removal rates of MAMBR were significantly enhanced to 126.9 mg NH4+-N L−1 d−1 and 158.8 mg NO3−-N L−1 d−1 by membrane aeration in batch test. Finally, the MAMBR was continuously fed with synthetic wastewater containing ammonium and nitrite to mimic the effluent from partial nitritation. When steady state with nitrogen loading rate of 2500 mg N L−1 d−1 was reached, the MAMBR achieved total nitrogen removal of 2496.7 mg N L−1 d−1, with negligible nitrate in effluent (~6.5 mg NO3−-N L−1). 16S rRNA amplicon sequencing and fluorescence in situ hybridization revealed the microbial community dynamics during enrichment and application. The high performance of nitrogen removal (2.5 kg N m−3 d−1) within 200 days operation and excellent biomass retention capacity (8.67 kg VSS m−3) makes the MAMBR promising for practical application of DAMO and Anammox in wastewater treatment. Aerobic granular sludge (AGS) is promising in wastewater treatment. However, the formation and existence of AGS under low organic loading rate (OLR) is still not fully understood due to a knowledge gap in the variations and correlations of N-acyl-homoserine lactones (AHLs), the microbial community, extracellular polymeric substances (EPS) and other physiochemical granule properties. This study comprehensively investigated the AHL-mediated quorum sensing (QS) and microbial community characters in the AGS fed with ammonium-rich wastewater under a low OLR of 0.15 kg COD (m3 d)−1. The results showed that the AGS appeared within 90 days, and the size of mature granules was over 700 μm with strong settleability and ammonium removal performance. More tightly-bound extracellular polysaccharide and tightly-bound extracelluar protein were produced in the larger AGS. C10-HSL and C12-HSL gradually became dominant in sludge, and short-chain AHLs dominated in water. EPS producers and autotrophic nitrifiers were successfully retained in the AGS under low OLR. AHL-mediated QS utilized C10-HSL, C12-HSL and 3OC6-HSL as the critical AHLs to regulate the TB-EPS in aerobic granulation, and autotrophic nitrifiers may perform interspecific communication with C10-HSL. The correlations of bacterial genera with AGS properties and AHLs were complex due to the dynamic fluctuations of microbial composition and other variable factors in the mixed-culture system. These findings confirmed the participation of AHL-mediated QS in the regulation of microbial community characters and AGS properties under low OLR, which may provide guidance for the operation of AGS systems under low OLR from a microbiological viewpoint. Abstract Wet age‐related macular degeneration (wet‐AMD) is a leading cause of irreversible blindness. Current treatment of AMD requires monthly intravitreal injection, which is difficult to be implemented in many parts of the world. In recent years, controlled release of anti‐vascular endothelial growth factor (VEGF) therapeutics has attracted intense research interest aiming to reduce the injection frequency to one or two times per year. In this study, we evaluated the in vivo pharmacokinetics and the long‐term therapeutic efficacy of an in situ hydrogel encapsulating an anti‐VEGF antibody in nonhuman primates. We show that after a single injection of anti‐VEGF controlled release hydrogel, a relatively constant concentration of drug can be maintained in the monkey eye for at least 5 months and the dose was sufficient for the treatment of recurrent choroidal neovascularization induced by repeat laser photocoagulation in monkeys. Our result suggested that when formulated into a controlled release formulation, a single dose of anti‐VEGF may be sufficient for a half‐year treatment and controlled release may be a suitable strategy to reduce the injection frequency in the treatment of AMD in human.

Despite numerous assessments of the impact of land-use change (LUC) on terrestrial evapotranspiration (ET) that have been conducted using land surface models (LSMs), no attempts have been made to evaluate their performance in this regard globally. Errors in simulating LUC impacts on ET largely stem from LUC data interpretation (LI, i.e. mapping of gridded LUC data into annual plant function types) and model structure (MS, i.e. parameterization of land-surface processes). The objective of this study was to benchmark ET estimates from four LSMs using the Zhang-curve, a prototype of the Budyko framework that has been validated against global hydrological observations and used widely to quantify the impacts of LUC on ET. A framework was further proposed to quantify and attribute errors in estimated ET changes induced by LI or MS. Results showed that all LSMs underestimated ET changes by about 55%–78%, and 37%–48% of the error was attributable to LI, but only 11%–32% of the error was attributable to MS across the four LSMs. From a hydrological perspective, our analysis provided insights about the errors in estimated impacts of LUC on ET by LSMs. The results demonstrated that LUC data interpretation accounted for a larger fraction of errors than LSM structure. Therefore, there is an urgent need for the defining and development of consistent protocols for interpreting global LUC data for future assessments.

Excess sludge reduction has been a research hotspot for a long time. Ultrasonic treatment of excess sludge was an efficient and green pretreatment method, and also can be combined with the addition of oxidants. To improve the effect of ultrasound treatment on sludge destruction, hydrogen peroxide (H2O2) was added to examine the combined results in the current study. The effects of the ultrasound/hydrogen peroxide system on the release of sludge organic matter during the destruction process were studied. Single-factor experiments were carried out to determine the optimal operating conditions. With the initial pH of 11.0, H2O2 concentration of 0.5 mmol/L, initial sludge concentration of 17 g/L, and 15 min ultrasonic treatment, the maximum soluble chemical oxygen demand (ΔSCOD) in the sludge supernatant after destruction was achieved at 3662.78 ± 239.21 mg/L, with a disintegration degree of 28.61 ± 2.14%, sludge reduction rate of 19.47 ± 0.82%, and the change of mixture sludge concentration (ΔMLSS) of 3.31 ± 0.06 g/L. Meanwhile, the release of nitrogen and phosphorus were greatly improved. Under the optimal conditions, the release of total nitrogen (TN), ammonia nitrogen (NH3-N) and total phosphorus (TP) were 282.30 ± 24.06 mg/L, 25.68 ± 2.09 mg/L, and 105.69 ± 7.84 mg/L, respectively. The current work had provided solid evidence showing the addition of hydrogen peroxide can effectively strengthen the treatment effects of ultrasound on sludge destruction.

Miscible and near-miscible flooding are used to improve the performance of carbon-dioxide-enhanced oil recovery in heterogeneous porous media. However, knowledge of the effects of heterogeneous pore structure on CO2/oil flow behavior under these two flooding conditions is insufficient. In this study, we construct pore-scale CO2/oil flooding models for various flooding methods and comparatively analyze CO2/oil flow behavior and oil recovery efficiency in heterogeneous porous media. The simulation results indicate that compared to immiscible flooding, near-miscible flooding can increase the CO2 sweep area to some extent, but it is still inefficient to displace oil in small pore throats. For miscible flooding, although CO2 still preferentially displaces oil through big throats, it may subsequently invade small pore throats. In order to substantially increase oil recovery efficiency, miscible flooding is the priority choice; however, the increase of CO2 diffusivity has little effect on oil recovery enhancement. For immiscible and near-miscible flooding, CO2 injection velocity needs to be optimized. High CO2 injection velocity can speed up the oil recovery process while maintaining equivalent oil recovery efficiency for immiscible flooding, and low CO2 injection velocity may be beneficial to further enhancing oil recovery efficiency under near-miscible conditions.

In this work, artificial oxygenation treatment (pure oxygen aeration or oxygen enriched water injection) combined with the introduction of exogenous microorganisms was employed to purify urban river sediment for the first time. Results showed that the developed in situ remediation strategy could increase the dissolved oxygen (DO) concentration and oxidation-reduction potential (ORP) value of the sediments. Benefiting from the increase of DO concentration, the bacterial diversity was enhanced. The highest removal efficiencies of organic matter were 18.4% and 22.3% through pure oxygen aeration and oxygen enriched water injection, respectively. More importantly, overlying water quality was not affected. By comparison, oxygen enriched water injection treatment could achieve better performance on sediment purification. Introducing exogenous microorganisms further reduced the organic matter content of the sediment. In short, the current work not only proposed a promising strategy for controlling urban river sediment pollution, but also provided novel insight for the understanding of river sediment containing highly concentrated organic matter.

Stripe rust or yellow rust (YR), caused by Puccinia striiformis f. sp. tritici (Pst), is one of the most important diseases of wheat (Triticum aestivum L.). Widespread deployment of resistant cultivars is the best means of achieving durable disease control. The red grain, spring wheat cultivar Napo 63 produced by CIMMYT in the 1960s shows a high level of adult-plant resistance to stripe rust in the field. To elucidate the genetic basis of resistance in this cultivar we evaluated 224 F2:3 lines and 175 F2:6 recombinant inbred lines (RILs) derived from a cross between Napo 63 and the Pst-susceptible line Avocet S. The maximum disease severity (MDS) data of F2:3 lines and the relative area under the disease progress curve (rAUDPC) data of RILs were collected during the 2014–2015 and 2015–2016 wheat growing seasons, respectively. Combined bulked segregant analysis and 90K single nucleotide polymorphism (SNP) arrays placed 275 of 511 polymorphic SNPs on chromosome 2B. Sixty four KASP markers selected from the 275 SNPs and 76 SSR markers on 2B were used to identify a chromosome region associated with rust response. A major effect QTL, named Qyrnap.nwafu-2BS, was identified by inclusive composite interval mapping and was preliminarily mapped to a 5.46 cM interval flanked by KASP markers 90K-AN34 and 90K-AN36 in chromosome 2BS. Fourteen KASP markers more closely linked to the locus were developed following a 660K SNP array analysis. The QTL region was finally narrowed to a 0.9 cM interval flanked by KASP markers 660K-AN21 and 660K-AN57 in bin region 2BS-1-0.53. The resistance of Napo 63 was stable across all environments, and as a QTL, explained an average 66.1% of the phenotypic variance in MDS of F2:3 lines and 55.7% of the phenotypic variance in rAUDPC of F5:6 RILs. The short genetic interval and flanking KASP markers developed in the study will facilitate marker-assisted selection, gene pyramiding, and eventual positional cloning of Qyrnap.nwafu-2BS.

We sought to assess light characteristics and user acceptability of a prototype Bright Classroom (BC), designed to prevent children's myopia by exposing them to light conditions resembling the outdoors. Conditions were measured throughout the school year in the glass-constructed BC, a traditional classroom (TC) and outdoors. Teachers and children completed user questionnaires, and children rated reading comfort at different light intensities. A total of 230 children (mean age 10.2 years, 57.4% boys) and 13 teachers (36.8 years, 15.4% men) completed questionnaires. The median (Inter Quartile Range) light intensity in the BC (2,540 [1,330-4,060] lux) was greater than the TC (477 [245-738] lux, P < 0.001), though less than outdoors (19,500 [8,960-36,000] lux, P < 0.001). A prominent spectral peak at 490-560 nm was present in the BC and outdoors, but less so in the TC. Teachers and children gave higher overall ratings to the BC than TC, and light intensity in the BC in summer and on sunny days (>5,000 lux) was at the upper limit of children's comfort for reading. In summary, light intensity in the BC exceeds TC, and is at the practical upper limit for routine use. Children and teachers prefer the BC.
