The shear strength deterioration of bedding planes between different rock types induced by cyclic loading is vital to reasonably evaluate the stability of soft and hard interbedded bedding rock slopes under earthquake; however, rare work has been devoted to this subject due to lack of attention. In this study, experimental investigations on shear strength weakening of discontinuities with different joint wall material (DDJM) under cyclic loading were conducted by taking the interface between siltstone and mudstone in the Shaba slope of Yunnan Province, China as research objects. A total of 99 pairs of similar material samples of DDJM (81 pairs) and discontinuities with identical joint wall material (DIJM) (18 pairs) were fabricated by inserting plates, engraved with typical surface morphology obtained by performing three-dimensional laser scanning on natural DDJMs sampled from field, into mold boxes. Cyclic shear tests were conducted on these samples to study their shear strength changes with the cyclic number considering the effects of normal stress, joint surface morphology, shear displacement amplitude and shear rate. The results indicate that the shear stress vs. shear displacement curves under each shear cycle and the peak shear strength vs. cyclic number curves of the studied DDJMs are between those of DIJMs with siltstone and mudstone, while closer to those of DIJMs with mudstone. The peak shear strengths of DDJMs exhibit an initial rapid decline followed by a gradual decrease with the cyclic number and the decrease rate varies from 6% to 55.9% for samples with varied surface morphology under different testing conditions. The normal stress, joint surface morphology, shear displacement amplitude and shear rate collectively influence the shear strength deterioration of DDJM under cyclic shear loading, with the degree of influence being greater for larger normal stress, rougher surface morphology, larger shear displacement amplitude and faster shear rate.

Abstract Super-enhancers are a class of DNA cis-regulatory elements that can regulate cell identity, cell fate, stem cell pluripotency, and even tumorigenesis. Increasing evidence shows that epigenetic modifications play an important role in the pathogenesis of various types of cancer. However, the current research is far from enough to reveal the complex mechanism behind it. This study found a super-enhancer enriched with abnormally active histone modifications in pancreatic ductal adenocarcinoma (PDAC), called DKK1-super-enhancer (DKK1-SE). The major active component of DKK1-SE is component enhancer e1. Mechanistically, AP1 induces chromatin remodeling in component enhancer e1 and activates the transcriptional activity of DKK1. Moreover, DKK1 was closely related to the malignant clinical features of PDAC. Deletion or knockdown of DKK1-SE significantly inhibited the proliferation, colony formation, motility, migration, and invasion of PDAC cells in vitro, and these phenomena were partly mitigated upon rescuing DKK1 expression. In vivo, DKK1-SE deficiency not only inhibited tumor proliferation but also reduced the complexity of the tumor microenvironment. This study identifies that DKK1-SE drives DKK1 expression by recruiting AP1 transcription factors, exerting oncogenic effects in PDAC, and enhancing the complexity of the tumor microenvironment.

Abstract Respiratory syncytial virus (RSV) is the major cause of bronchiolitis and pneumonia in young children and the elderly. There are currently no approved RSV-specific therapeutic small molecules available. Using high-throughput antiviral screening, we identified an oral drug, the prenylation inhibitor lonafarnib, which showed potent inhibition of the RSV fusion process. Lonafarnib exhibited antiviral activity against both the RSV A and B genotypes and showed low cytotoxicity in HEp-2 and human primary bronchial epithelial cells (HBEC). Time-of-addition and pseudovirus assays demonstrated that lonafarnib inhibits RSV entry, but has farnesyltransferase-independent antiviral efficacy. Cryo-electron microscopy revealed that lonafarnib binds to a triple-symmetric pocket within the central cavity of the RSV F metastable pre-fusion conformation. Mutants at the RSV F sites interacting with lonafarnib showed resistance to lonafarnib but remained fully sensitive to the neutralizing monoclonal antibody palivizumab. Furthermore, lonafarnib dose-dependently reduced the replication of RSV in BALB/c mice. Collectively, lonafarnib could be a potential fusion inhibitor for RSV infection.

With over 9 million fatalities per year expected by 2030, infectious diseases will remain a significant burden on the world economy and cause high mortality rates. An excellent method to increase the bioactivity of levofloxacin (LEV) in pediatric abdominal wound repair is the finding of a stimuli-based drug delivery system (DDS). We designed and developed an LEV incorporated with zeolite imidazole framework-8 (ZIF-8) as a promising nanocarrier for wound healing applications. The spectral analysis and morphological analysis confirm the formation of our newly fabricated composites. Mouse embryonic fibroblast NIH3T3 cells, the cytotoxicity, cytocompatibility, and cell proliferation characteristics of LEV@ZIF-8 were evaluated in vitro. LEV@ZIF-8 composite considerably improved the biocompatibility against NIH3T3 cells after 72-h of exposure, according to in vitro experiments. Under acidic circumstances, the pH-responsive drug release studies exhibit superior LEV release, and in physiological circumstances, there is no unintended drug release. The LEV@ZIF-8 composite-treated cells demonstrate the most remarkable cell growth and migration method in a very short time, according to the results of the wound scratch experiment. The composite exposure concentration depended on inhibition against various microorganisms in the antibacterial activity testing. According to the study, LEV@ZIF-8 are appropriate and effective DDS for stimuli-based pediatric abdominal wound repair.

Abstract Although protein subunit vaccines generally have acceptable safety profiles with precise antigenic content, limited immunogenicity can lead to unsatisfactory humoral and cellular immunity and the need for vaccine adjuvants and delivery system. Herein, we assess a vaccine adjuvant system comprising Quillaja Saponaria-21(QS-21) and cobalt porphyrin polymeric micelles that enabling the display of His-tagged antigen on its surface. The nanoscale micelles promote antigen uptake and dendritic cell activation to induce robust cytotoxic T lymphocyte response and germinal center formation. Using the recombinant protein antigens from influenza A and rabies virus, the micelle adjuvant system elicited robust antiviral responses and protected mice from lethal challenge. In addition, this system could be combined with other antigens to induce high titers of neutralizing antibodies in models of three highly pathogenic viral pathogens: Ebola virus, Marburg virus, and Nipah virus. Collectively, our results demonstrate this polymeric micelle adjuvant system can be used as a potent nanoplatform for developing antiviral vaccine countermeasures that promote humoral and cellular immunity.

Abstract Background Insulin like growth factor II mRNA binding protein 3 (IGF2BP3) has been implicated in numerous inflammatory and cancerous conditions. However, its precise molecular mechanisms in endometriosis (EMs) remains unclear. The aim of this study is to examine the influence of IGF2BP3 on the occurrence and progression of EMs and to elucidate its underlying molecular mechanism. Methods Efects of IGF2BP3 on endometriosis were confrmed in vitro and in vivo. Based on bioinformatics analysis, RNA immunoprecipitation (RIP), RNA pull-down assays and Fluorescent in situ hybridization (FISH) were used to show the association between IGF2BP3 and UCA1. Single-cell spatial transcriptomics analysis shows the expression distribution of glutaminase 1 (GLS1) mRNA in EMs. Study the effect on glutamine metabolism after ectopic endometriotic stromal cells (eESCs) were transfected with Sh-IGF2BP3 and Sh-UCA1 lentivirus. Results Immunohistochemical staining have revealed that IGF2BP3 was upregulated in ectopic endometriotic lesions (EC) compared to normal endometrial tissues (EN). The proliferation and migration ability of eESCs were greatly reduced by downregulating IGF2BP3. Additionally, IGF2BP3 has been observed to interact with urothelial carcinoma associated 1 (UCA1), leading to increased stability of GLS1 mRNA and subsequently enhancing glutamine metabolism. Results also demonstrated that IGF2BP3 directly interacts with the 3’ UTR region of GLS1 mRNA, influencing its expression and stability. Furthermore, UCA1 was able to bind with c-MYC protein, stabilizing c-MYC mRNA and consequently enhancing GLS1 expression through transcriptional promotion. Conclusion These discoveries underscored the critical involvement of IGF2BP3 in the elevation and stability of GLS1 mRNA in the context of glutamine metabolism by interacting with UCA1 in EMs. The implications of our study extended to the identification of possible therapeutic targets for individuals with EMs.

Introduction: Refractory lupus nephritis (LN) causes kidney disease progression and increases the risk of loss of renal function. Due to the high specificity and few side effects of biological agents, they are recommended for the treatment of systemic lupus erythematosus. There are few data on telitacicept for the treatment of refractory LN. Case Presentation: Here, we report the efficacy and safety of telitacicept in the treatment of refractory LN in a 25-year-old female patient. This patient with refractory lupus developed Pneumocystis jirovecii pneumonia while using multitargeted therapy, and the patient’s urine protein was rapidly relieved after telitacicept combination with low-dose mycophenolate mofetil (MMF). Conclusion: This result suggests that telitacicept has a positive effect on refractory LN with no significant side effects. Further reports and a registry are necessary to confirm that telitacicept with low-dose MMF should be preferred in refractory LN.

The Megalyridae are a small family of parasitoid wasps comprising eight extant genera (71 species) and six extinct genera (13 species). Here, we report eight new species from Late Cretaceous Kachin (Myanmar) and Taimyr (Russia) ambers; the family is recorded for the first time from the latter. †Cretolyra noijebumensis gen. et sp. nov., †Cretolyra shawi gen. et sp. nov., †Genkyhag innebula gen. et sp. nov., †Megacoxa chandrahrasa gen. et sp. nov., †Megacoxa janzeni gen. et sp. nov., and †Megacoxa synchrotron gen. et sp. nov., are described from late Albian – early Cenomanian Kachin amber; †Kamyristi exfrigore gen. et sp. nov. and †Kamyristi yantardakhensis gen. et sp. nov. from Taimyr amber (Baikura, late Albian – early Cenomanian Ognevka Formation and Yantardakh, Santonian Kheta Formation, respectively). Phylogenetic analyses of the family are presented and the classification of the Megalyridae is revised accordingly. A new tribe, †Megalavini trib. nov., is erected to accommodate †Cretolyra gen. nov., †Genkyhag gen. nov., †Megacoxa gen. nov. together with †Megalava Perrichot, 2009; †Megalavini trib. nov. is characterized by the vein M fully pigmented and veins Rs+M and M+Cu aligned, the crenulate mesometapectal sulcus, and the pronotal spiracle not posteriorly surrounded by cuticle. This spiracular condition was previously only observed in †Megazar Perrichot, 2009, thus the subfamily †Megazarinae Perrichot, 2009 stat. nov. is proposed for the clade (†Megalavini + †Megazar), as sister to the remaining Megalyridae. The latter is defined as the Megalyrinae, with †Kamyristi gen. nov. being retrieved as sister to all other genera except Rigel Shaw, 1987 under parsimony analyses. To align phylogeny with classification, three new tribes are erected (†Kamyristini trib. nov., †Megallicini trib. nov., and †Valaaini trib. nov.) and the †Cretodinapsini is synonymized under Megalyrini. A grouping [Rigel + Megalyridia] is supported under Bayesian analyses, which is the only specific conflict with the parsimony analyses, suggesting some degree of caution with respect to the internal relationships of the Megalyrinae. Finally, a revised key to the Megalyridae genera is provided.

Abstract The placenta is a unique organ for ensuring normal embryonic growth in the uterine. Here, we found that maternal RNA transcription in Dlk1-Dio3 imprinted domain is essential for placentation. PolyA signals were inserted into Gtl2 to establish a mouse model to prevent the expression of maternal RNAs in the domain. The maternal allele knock-in (MKI) and homozygous (HOMO) placentas showed an expanded junctional zone, reduced labyrinth and poor vasculature impacting both fetal and maternal blood spaces. The MKI and HOMO models displayed dysregulated gene expression in the Dlk1-Dio3 domain. In situ hybridization detected Dlk1, Gtl2, Rtl1, miR-127 and Rian dysregulated in the labyrinth vasculature. MKI and HOMO induced Dlk1 to lose imprinting, and DNA methylation changes of IG-DMR and Gtl2-DMR, leading to abnormal gene expression, while the above changes didn’t occur in paternal allele knock-in placentas. These findings demonstrate that maternal RNAs in the Dlk1-Dio3 domain are involved in placental vasculature, regulating gene expression, imprinting status and DNA methylation.

Abstract The rainy season in South China is divided into two phases, the pre- and postrainy seasons, according to the seasonal progression of the East Asian summer monsoon. The precipitation prediction skills for the two rainy seasons are investigated using subseasonal-to-seasonal (S2S) hindcast data from the European Centre for Medium-Range Weather Forecasts (ECMWF) for 2001–2019. The precipitation prediction skills and biases differ between the two rainy seasons, although some similar characteristics exist regarding circulation patterns and their influence on precipitation. During the two rainy seasons, the prediction ability of circulation at 850 hPa in key areas is relatively high, and the influence of circulation on precipitation is well captured; additionally, the relationship between circulation in key areas at 500 hPa and precipitation is less accurately constrained. Moreover, the precipitation prediction skill in the prerainy season is higher than that in the postrainy season. The main bias is that the 200 hPa westerly winds provide favorable divergence conditions for prerainy season precipitation (preprecipitation), while the postrainy season precipitation (postprecipitation) displays almost no correlation with the circulation in the reanalysis product; however, the simulated circulation at 200 hPa is closely connected to the precipitation in both rainy seasons; therefore, the lower prediction skill in the postrainy season is likely associated with overestimation of the complex physical mechanism of the upper-level circulation in the model.

The epithelial mesenchymal transition (EMT) plays significant roles in the progression of cancer and fibrotic disease. Moreover, this process is reversible, resulting in mesenchymal epithelial transition (MET), which plays an important role in cancer metastasis. There is a lack of methods to trace and target EMT cells using synthetic biology circuits, which makes it difficult to study the cell fate or develop targeted treatments. In this study, we introduced responsive EMT sensing circuits, which sense the EMT using specific promoters that respond to transcription factors typical of EMT activation (EMT-TFs). The transcriptional strength of EMT-sensing promoters decreased more than 13-fold in response to the overexpression of the EMT-TF. Then, the NOT gate circuits were built by placing the tetR transcription repressor under the control of EMT sensing promoters and expressed an output signal using the constitutive CMV promoter modified with tetO sites This circuit is named EMT sensing and responding circuits .When the EMT transcription factors was present, we observed a 5.8-fold signal increase in the system. Then, we successfully distinguished mesenchymal breast cancer cells from epithelial cancer cells and repressed the proliferation of EMT tumor cells using our circuits. The EMT sensing and responding circuits are promising tools for the identification of EMT cells, which is crucial for EMT-related disease therapy and investigating the mechanisms underlying the reversible EMT process.

The clinical application of cancer immunotherapy is unsatisfied due to low response rates and systemic immune-related adverse events. Microwave hyperthermia can be used as a synergistic immunotherapy to amplify the antitumor effect. Herein, we designed a Gd-based metal-organic framework (Gd-MOF) nanosystem for MRI-guided thermotherapy and synergistic immunotherapy, which featured high performance in drug loading and tumor tissue penetration. The PD-1 inhibitor (aPD-1) was initially loaded in the porous Gd-MOF (Gd/M) nanosystem. Then, the phase change material (PCM) and the cancer cell membrane were further sequentially modified on the surface of Gd/MP to obtain Gd-MOF@aPD-1@CM (Gd/MPC). When entering the tumor microenvironment (TME), Gd/MPC induces immunogenic death of tumor cells through microwave thermal responsiveness, improves tumor suppressive immune microenvironment and further enhances anti-tumor ability of T cells by releasing aPD-1. Meanwhile, Gd/MPC can be used for contrast-enhanced MRI. Transcriptomics data revealed that the downregulation of MSK2 in cancer cells leads to the downregulation of c-fos and c-jun, and ultimately leads to the apoptosis of cancer cells after treatment. In general, Gd/MPC nanosystem not only solves the problem of system side effect, but also achieves the controlled drug release via PCM, providing a promising theranostic nanoplatform for development of cancer combination immunotherapy.

Porcine circovirus type 2 (PCV2) infection leads to multi-system inflammation in pigs, and this effect can be achieved by upregulating host miR-21. The underlying mechanism of miR-21 regulates PCV2-induced inflammation is already known, however, how PCV2 regulates miR-21 levels and function using both autonomic and host factors remains to be further revealed. Here we present the first evidence that PCV2 ORF5 induces an inflammatory response by up-regulating miR-21 level through targeting nuclear miR-30d. In this study, we found that overexpression of ORF5 significantly increased miR-21 level and promoted the expression of inflammatory cytokines and activation of the NF-κB pathway, while ORF5 mutation had the opposite effect. Moreover, the differential expression of miR-21 could significantly change the pro-inflammatory effect of ORF5, indicating that ORF5 promotes inflammatory response by up-regulating miR-21. Bioinformatics analysis and clinical detection found that nuclear miR-30d was significantly down-regulated after ORF5 overexpression and PCV2 infection, and targeted pri-miR-21 and PCV2 ORF5. Functionally, we found that miR-30d inhibited the levels of miR-21 and inflammatory cytokines in cells. Mechanistically, we demonstrated that ORF5 inhibits miR-30d expression levels through direct binding but not via the circRNA pathway, and miR-30d inhibits miR-21 levels by targeting pri-miR-21. In summary, the present study revealed the molecular mechanism of ORF5 upregulation of miR-21, further refined the molecular chain of PCV2-induced inflammatory response and elucidated the role of miRNAs in it.

The increasing ocean warming due to climate change significantly threatens regional marine ecosystems by raising the frequency and severity of extreme temperature events. This study examines patterns and trends of maximum annual sea surface temperature (Tmax) in the Eastern China Seas from 1985 to 2022. The results show a significant warming trend in Tmax, exceeding the global average, with notable differences between southern and northern regions. The northern Tmax warming rate is faster, with occurrence times significantly advancing, while the southern Tmax warming rate is slower, with occurrence times significantly delayed. The southern Tmax and its timing are closely correlated with the annual maximum air temperature and its timing. In the north, Tmax timing is influenced by latent heat flux (QLH); a significant increase in August QLH inhibits the continued rise of SST, causing Tmax to advance. The study also highlights a significant increase in marine heatwaves at Tmax timing, with higher Tmax indicating a higher occurrence probability. By elucidating these Tmax trends and dynamics, our study enhances understanding of regional climate impacts, supporting targeted conservation efforts and adaptive ecosystem management strategies in the Eastern China Seas.

Abstract Extremely low‐frequency (ELF) electromagnetic (EM) waves adeptly propagate in harsh cross‐medium environments, overcoming rapid decay that hinders high‐frequency counterparts. Traditional antennas, however, encounter challenges concerning size, efficiency, and power. Here, drawing inspiration from nature, we present a groundbreaking piezo‐actuated, bionic flapping‐wing magnetic‐dipole resonator (BFW‐MDR), operating in the electro‐mechano‐magnetic coupling mechanism, designed for efficient ELF EM wave transmission. The unique rigid‐flexible hybrid flapping‐wing structure magnifies rotation angles of anti‐phase magnetic dipoles by tenfold, leading to constructive superposition of emitted magnetic fields. Consequently, the BFW‐MDR exhibits a remarkable quality factor of 288 and an enhanced magnetic field emission of 514 fT at 100 meters with only 6.9 mW power consumption, surpassing traditional coil antennas by three orders of magnitude. The communication rate is doubled by the ASK+PSK modulation method. Its robust performance in cross‐medium communication, even amidst various interferences, underscores its potential as a highly efficient antenna for underwater and underground applications.

We used daily sea surface temperature (SST) data and hourly drifter data to investigate ocean responses to tropical cyclone (TC) intensity and outer size (wind radius of 34 kt, or R34) in the Northwest Pacific. Results showed that SST cooling is more sensitive to TC R34 than to TC intensity; namely, TCs with a larger R34 cause stronger SST cooling regardless of their intensity. TCs with an R34 ≥125 nmi could cool SST 0.9°C more than TCs with an R34

There is an effective way to enhance the spatial resolution of hyperspectral remote sensing images by fusing them with multispectral remote sensing images. However, most of the existing deep fusion techniques adopt discretized explicit models to approximate the complex continuous nonlinear mapping in the fusion process, leading to limitations in enhancing the fidelity of spatial details. Additionally, existing algorithms commonly utilize discrete methods such as bilinear or bicubic interpolation during the hyperspectral upsampling process, leading to the loss of crucial spatial-spectral features. To this end, this study proposes a novel Implicit Transformer Fusion Generative Adversarial Network (ITF-GAN), which incorporates the continuity perception mechanism of implicit neural representation with the powerful self-attention mechanism of the Transformer architecture, which uses point-to-point implicit functions aiming to efficiently process information in both spatial and spectral dimensions. Besides, a guided implicit neural sampling module is introduced in the hyperspectral image up-sampling process to enhance the coordinated expression of features in the spatial and spectral domains, which improves the spatial resolution and spectral fidelity of the fused image during the upsampling process. A series of fusion experiments including 4x, 8x, and 16x scale factors have shown that ITF-GAN has significant advantages over current popular fusion algorithms in both objective evaluation indicators and subjective visual evaluation.

Rebar lap splicing is a widely used method of reinforcement connection in precast concrete structures, and the lap-slip model serves as an important prerequisite for conducting detailed mechanical calculations of reinforced concrete lap splice members. Most traditional bond-slip mechanical models are semi-empirical and semi-theoretical formulas derived from experimental data; they do not consider the effect of reinforcement lap spacing, which prevents them from accurately reflecting the force transfer characteristics of lap reinforcement in concrete. In this paper, a Back Propagation (BP) neural network based on a Genetic Algorithm (GA) is used to establish a lap-slip model of reinforced concrete considering rebar spacing, which improves on the current bond-slip mechanical model. Several sets of rebar lap tests based on RC/ECC/UHPC were designed, and full-sample parameter sensitivity analyses were carried out using the lap length, the thickness of the concrete cover, the rebar spacing and the stirrup ratio as test parameters. The results showed that rebar lap spacing was an important influencing factor in the proposed lap-slip model, with a sensitivity coefficient reaching 0.471. The lap-slip model, based on the GA-BP neural network, was developed and compared with existing models; the predictive ability of the model was verified by the ten-fold crossover method. The generalizability of the GA-BP model was verified by randomly selecting non-training test data as well as lap test data of rebar in different types of concrete. The results indicated that the model exhibits high predictive and generalization abilities, with most correlation coefficients exceeding 0.95. The proposed model was compared with a mechanical model established using the traditional fitting formula method, and the results showed that the prediction accuracy of the lap-slip model established based on the GA-BP neural network is slightly lower than that of the mechanical model obtained using the fitting formula method, but it still offers high prediction accuracy, and the prediction results are more objective and realistic, which is of guiding significance for practical applications and related research.

Objectives The efficacy of transjugular intrahepatic portosystemic shunt (TIPS) plus extrahepatic collateral embolisation (TIPS+E) in reducing rebleeding and hepatic encephalopathy (HE) post-TIPS was recently reported in a meta-analysis, but further validation is essential. This study aims to confirm the effectiveness of TIPS+E using real-world data.Methods The multicentre retrospective cohort included 2077 patients with cirrhosis who underwent TIPS±E (TIPS: 631, TIPS+E: 1446) between January 2010 and December 2022. Regression and propensity score matching (PSM) were used to adjust for baseline characteristic differences. After PSM, clinical outcomes, including rebleeding, HE, survival and further decompensation (FDC), were analysed. Baseline data from all patients contributed to the construction of prognostic models.Results After PSM, 1136 matched patients (TIPS+E: TIPS=568:568) were included. TIPS+E demonstrated a significant reduction in rebleeding (HR 0.77; 95% CI 0.59 to 0.99; p=0.04), HE (HR 0.82; 95% CI 0.68 to 0.99; p=0.04) and FDC (HR 0.85; 95% CI 0.73 to 0.99; p=0.04), comparing to TIPS. Significantly, TIPS+E also reduced rebleeding, HE and FDC in subgroup of using 8 mm diameter stents and embolising of gastric varices+spontaneous portosystemic shunts (GV+SPSS). Our paper investigates how qualified foreign institutional investors (QFII) impact stock liquidity in the Chinese A-share market using data for 2005 to 2019. Contrary to previous findings, we find that QFII enhance stock liquidity. Specifically, QFII’s participation is negatively associated with the individual stock illiquidity and positively related to stock trading volume. Moreover, using a step-by-step procedure, we provide evidence that QFII raises stock liquidity by ameliorating information asymmetry. QFII will attract more market attention and improve firm disclosure quality. We address possible endogeneity with fixed effects and instrumental variables, and our findings are robust to self-selection bias, stock market shocks, and alternative explanatory variables.

This study aimed to investigate the role of miR-558 in tumor angiogenesis by targeting heparinase (HPSE) in tongue squamous cell carcinoma (TSCC)-derived exosomes. In the present study, the role of exosome miR-558 in angiogenesis in vitro and in vivo was investigated by cell proliferation, migration, tube formation, subcutaneous tumor formation in mice, and in vivo Matrigel plug assay. The target genes of miR-558 were detected by means of dual luciferase assay. It was found that TSCC cells secrete miR-558 into the extracellular environment, with exosome as the carrier. Human umbilical vein endothelial cells (HUVEC) ingested exosomes, which not only increased the expression level of miR-558, but also enhanced their proliferation, migration, and tube formation functions. In vivo Matrigel plug assay demonstrated that TSCC cell-derived exosome miR-558 promoted neovascularization in vivo. Compared with negative control cells, TSCC cells overexpressing miR-558 formed subcutaneous tumors in nude mice, with larger volume, heavier mass, and more vascularization. Dual luciferase assay confirmed that HPSE was the direct target gene regulated by miR-558. HPSE promoted the proliferation, migration, and tube formation of HUVECs, and the knockout of HPSE could downregulate the pro-angiogenic effect of miR-558. In summary, miR-558 in TSCC exosomes promotes the proliferation, migration, and tube formation of HUVECs by targeting HPSE, and enhancing tumor angiogenesis.

Abstract As one of the major causes of antimicrobial resistance, β‐lactamase develops rapidly among bacteria. Detection of β‐lactamase in an efficient and low‐cost point‐of‐care testing (POCT) way is urgently needed. However, due to the volatile environmental factors, the quantitative measurement of current POCT is often inaccurate. Herein, we demonstrate an artificial intelligence (AI)‐assisted mobile health system that consists of a paper‐based β‐lactamase fluorogenic probe analytical device and a smartphone‐based AI cloud. An ultrafast broad‐spectrum fluorogenic probe (B1) that could respond to β‐lactamase within 20 s was first synthesized, and the detection limit was determined to be 0.13 nmol/L. Meanwhile, a three‐dimensional microfluidic paper‐based analytical device was fabricated for integration of B1. Also, a smartphone‐based AI cloud was developed to correct errors automatically and output results intelligently. This smart system could calibrate the temperature and pH in the β‐lactamase level detection in complex samples and mice infected with various bacteria, which shows the problem‐solving ability in interdisciplinary research, and demonstrates potential clinical benefits.

Hundreds of green tea products are available on the tea market and exhibit different characteristics. In the present study, seven types of green tea were processed, and their nonvolatile profiles were analyzed by liquid chromatography–mass spectrometry. Non-spreading green tea contained higher concentrations of catechins and flavonoid glycosides, but lower concentrations of amino acids, caffeine, and theaflavins. Non-rolling green teas with a straight appearance contained higher concentrations of flavonoid glycosides and theaflavins. In contrast, leaf-rolling green teas contained much lower concentrations of flavonoid glycosides and catechins. These seven green tea qualities all decreased following prolonged storage, concurrent with increasing concentrations of proanthocyanidins, catechins dimers, theaflavins, and organic acids. The leaf-rolling green teas exhibited reduced levels of deterioration during storage in terms of their nonvolatile profile and sensory quality. Findings show that moderate destruction on tea leaves during green tea processing is beneficial to both tea flavor and quality maintenance during storage.

Context: It is very necessary to delay ovarian aging and prevent age-related health problems. The active ingredient in Honghua Xiaoyao tablet (HHXYT) has the effects of anti-oxidation, anti-inflammation, immune regulation and so on.Objective: To explore the effect and mechanism of Honghua Xiaoyao tablet on aging model mice.Materials and methods: The aging model was established by intraperitoneal injection of D-galactose in model mice. The mice in the HHXYT-L,M,H group were given 0.3 g/kg, 0.6 g/kg and 1.2 g/kg Honghua Xiaoyao tablet suspension respectively, and the HHXYT-M + E2 group was given 0.6 g/kg HHXYT +0.13 mg/kg estradiol valerate for 30 days. In this study, ELISA, HE, Western blot, IH and TUNEL were used.Results: HHXYT + E2 can improve the gonadal index, estrous cycle of aging mice. In HHXYT-M + E2 group, the level of FSH and LH decreased, while E2 and AMH increased significantly. The number of growing follicles in HHXYT-M + E2 group increased, which was better than that of HHXYT alone. Western blot results showed that HHXYT-M + E2 group decreased the expression of Bax, cleaved-Parp, cleaved-Casp-3 and CytC molecules and increased the expression of Bcl-2 in ovarian tissue. FSHR expression decreased in model group and increased in HHXYT group. TUNEL staining showed that the number of apoptotic cells in HHXYT group was reduced, and the HHXYT-M + E2 group was the most significantly.Discussion and conclusion: HHXYT can improve the level of sex hormones and increase the number of growing follicles in aging mice. HHXYT-M + E2 group has the best effect, and its mechanism may be related to reducing ovarian granulosa cell apoptosis.

Exposure to ambient PM2.5 is associated with neurodegenerative disorders, in which microglia activation plays a critical role. Thus far, the underlying mechanisms for PM2.5-induced microglia activation have not been well elucidated. In this study, a human microglial cell line (HMC3) was used as the in vitro model to examine the inflammatory effect (hall marker of microglia activation) of PM2.5 and regulatory pathways. The expression of inflammatory mediators including interleukin-6 (IL-6) and cyclooxygenase-2 (COX-2) as well as the brain derived neurotrophic factor (BDNF) were determined by ELISA and/or real-time PCR, respectively. Flow cytometry was used to measure the production of intracellular reactive oxygen species (ROS). Western blot was used to measure protein levels of Toll-like receptor 4 (TLR4), NF-κB inhibitor α (IκBα) and COX-2. It was shown that PM2.5 stimulation increased IL-6 and COX-2 expression but decreased BDNF expression in a dose-dependent manner. Further studies showed that PM2.5 triggered the formation of ROS and pre-treatment with the ROS scavenger acetylcysteine (NAC) significantly suppressed PM2.5-induced IL-6 and COX-2 expression. Moreover, the nuclear factor kappa B (NF-κB) inhibitor BAY11–7085 or the TLR4 neutralizing antibody markedly blocked PM2.5-induced IL-6 and COX-2 expression. However, NAC or BAY11–7085 exhibited minimal effect on PM2.5-induced BDNF down-regulation. In addition, pre-treatment with BAY11–7085 or TLR4 neutralizing antibody reduced ROS production induced by PM2.5, and NAC pre-treatment inhibited TLR4 expression and NF-κB activation induced by PM2.5. Collectively, PM2.5 treatment induced IL-6 and COX-2 but suppressed BDNF expression. PM2.5-induced IL-6 and COX-2 expression was mediated by interactive oxidative stress and TLR4/NF-κB pathway.

Flash sintering (FS) is a novel technique for rapidly densifying silicon carbide (SiC) ceramics. This work achieved a rapid sintering of SiC ceramics by the utilization of ultra-high temperature flash sintering within 60 s. Pyrolysis carbon (PyC) “bridges” were constructed between SiC particles through the carbonisation of phenolic resin, providing a large number of current channels. The incubation time of the flash sintering process was significantly reduced, and the sintering difference between the centre and the edge regions of the ceramics was minimized, with an average particle size of the centre region and edge region being 12.31 and 9.02 μm, respectively. The results showed that the porosity of the SiC ceramics after the flash sintering was reduced to 14.79% with PyC “bridges” introduced, and the Vickers hardness reached 19.62 GPa. PyC “bridges” gradually evolved from amorphous eddy current carbon to oriented graphite carbon, indicating that the ultra-high temperature environment in which the sample was located during the flash sintering was successfully constructed. Ultra-high temperature flash sintering of SiC is expected to be applied to the local repair of matrix damage in SiC ceramic matrix composites.

Abstract The spatial distribution and data quality of curve number (CN) values determine the performance of hydrological estimations. However, existing CN datasets are constrained by universal-applicability hypothesis, medium resolution, and imbalance between specificity CN tables to generalized land use/land cover (LULC) maps, which hinder their applicability and predictive accuracy. A new annual CN dataset named CUSCN30, featuring an enhanced resolution of 30 meters and accounting for temporal variations in climate and LULC in the continental United States (CONUS) between 2008 and 2021, was developed in this study. CUSCN30 demonstrated good performance in surface runoff estimation using CN method when compared to observed surface runoff for the selected watersheds. Compared with existing CN datasets, CUSCN30 exhibits the highest accuracy in runoff estimation for both normal and extreme rainfall events. In addition, CUSCN30, with its high spatial resolution, better captures the spatial heterogeneity of watersheds. This developed CN dataset can be used as input for hydrological models or machine learning algorithms to simulate rainfall-runoff across multiple spatiotemporal scales.

Abstract The shotgun proteomic analysis is currently the most promising single-cell protein sequencing technology, however its identification level of ~1000 proteins per cell is still insufficient for practical applications. Here, we develop a pick-up single-cell proteomic analysis (PiSPA) workflow to achieve a deep identification capable of quantifying up to 3000 protein groups in a mammalian cell using the label-free quantitative method. The PiSPA workflow is specially established for single-cell samples mainly based on a nanoliter-scale microfluidic liquid handling robot, capable of achieving single-cell capture, pretreatment and injection under the pick-up operation strategy. Using this customized workflow with remarkable improvement in protein identification, 2449–3500, 2278–3257 and 1621–2904 protein groups are quantified in single A549 cells (n = 37), HeLa cells (n = 44) and U2OS cells (n = 27) under the DIA (MBR) mode, respectively. Benefiting from the flexible cell picking-up ability, we study HeLa cell migration at the single cell proteome level, demonstrating the potential in practical biological research from single-cell insight.

Abstract Objective This study investigated the clinical, imaging, and electroencephalogram (EEG) characteristics of methylmalonic acidemia (MMA) with nervous system damage as the primary manifestation. Methods From January 2017 to November 2022, patients with nervous system injury as the main clinical manifestation, diagnosed with methylmalonic acidemia by metabolic and genetic testing, were enrolled and analyzed. Their clinical, imaging, and electroencephalogram data were analyzed. Results A total of 18 patients were enrolled, including 15 males and 3 females. The clinical symptoms were convulsions, poor feeding, growth retardation, disorder of consciousness, developmental delay, hypotonia, and blood system changes. There were 6 cases (33%) of hydrocephalus, 9 (50%) of extracerebral space widened, 5 (27%) of corpus callosum thinning, 3 (17%) of ventricular dilation, 3 (17%) of abnormal signals in the brain parenchyma (frontal lobe, basal ganglia region, and brain stem), and 3 (17%) of abnormal signals in the lateral paraventricular. In addition, there were 3 cases (17%) of cerebral white matter atrophy and 1 (5%) of cytotoxic edema in the basal ganglia and cerebral peduncle. EEG data displayed 2 cases (11%) of hypsarrhythmia, 3 (17%) of voltage reduction, 12(67%) of abnormal discharge, 13 (72%) of abnormal sleep physiological waves or abnormal sleep structure, 1 (5%) of immature (delayed) EEG development, and 8 (44%) of slow background. There were 2 cases (11%) of spasms, 1 (5%) of atonic seizures, and 1 (5%) of myoclonic seizures. There were 16 patients (89%) with hyperhomocysteinemia. During follow-up, 1 patient was lost to follow-up, and 1 died. In total, 87.5% (14/16) of the children had varying developmental delays. EEG was re-examined in 11 cases, of which 8 were normal, and 3 were abnormal. Treatments included intramuscular injections of vitamin B12, L-carnitine, betaine, folic acid, and oral antiepileptic therapy. Acute treatment included anti-infective, blood transfusion, fluid replacement, and correcting acidosis. The other treatments included low-protein diets and special formula milk powder. Conclusion Methylmalonic acidemia can affect the central nervous system, leading to structural changes or abnormal signals on brain MRI. Metabolic screening and genetic testing help clarify the diagnosis. EEG can reflect changes in brain waves during the acute phase.

Abstract Resveratrol (RSV) is a natural polyphenol compound found in various plants that has been shown to have potential benefits for preventing aging and supporting cardiovascular health. However, the specific signal pathway by which RSV protects the aging heart is not yet well understood. This study aimed to explore the protective effects of RSV against age‐related heart injury and investigate the underlying mechanisms using a D‐galactose‐induced aging model. The results of the study indicated that RSV provided protection against age‐related heart impairment in mice. This was evidenced by the reduction of cardiac histopathological changes as well as the attenuation of apoptosis. RSV‐induced cardioprotection was linked to a significant increase in antioxidant activity and mitochondrial transmembrane potential, as well as a reduction in oxidative damage. Additionally, RSV inhibited the production of pro‐inflammatory cytokines such as interleukin‐1β (IL‐1β) and tumor necrosis factor‐α (TNF‐α). Furthermore, the expression of toll‐like receptor 4 (TLR4), nuclear factor kappa‐B p65 (NF‐κB p65), and notch 1 protein were inhibited by RSV, indicating that inhibiting the Notch/NF‐κB pathway played a critical role in RSV‐triggered heart protection in aging mice. Moreover, further data on intestinal function demonstrated that RSV significantly increased short‐chain fatty acids (SCFAs) in intestinal contents and reduced the pH value in the feces of aging mice. RSV alleviated aging‐induced cardiac dysfunction through the suppression of oxidative stress and inflammation via the Notch/NF‐κB pathway in heart tissue. Furthermore, this therapeutic effect was found to be associated with its protective roles in the intestine.

Magnetic graphene (Fe3O4@G) was synthesized as a magnetic solid phase extractor for the extraction and enrichment of organophosphorus pesticides in green tea by chemical coprecipitation method. Based on this, ultra high performance liquid chromatography tandem mass spectrometry method was developed for the determination of 19 organophosphorus pesticide residues in green tea. 40 mg adsorbent and 4 g sodium chloride were used in this experiment with the pH of sample solution at 7 for 20 min extraction time, 3.0 mL acetone desorption. The result showed that the prepared Fe3O4@G material had good stability and reusability with the linear correlation coefficients greater than 0.999 in the range of 5～500 μg/L. The limits of detection (LOD) and the limits of quantitation (LOQ) were 5.0～6.0 μg/kg and 15.0～20.0 μg/kg, respectively. The recoveries were between 61.2% and 94.9% with the relative standard deviations (RSD, n=6) in the range of 2.6%～10.2% when the samples were labeled at 20.0, 40.0 and 200.0 μg/kg. This method was safe and economy with a wide application range, easier control of pretreatment, less amount of organic solvent and reusable materials, which has a good application prospect in the enrichment and separation of pesticides in tea by the magnetic dispersion solid phase extraction technology.

Faced with complex and ever-changing environmental conditions in the agricultural field, efficient agricultural information gathering is crucial for optimising agricultural output. Therefore, a new path planning algorithm combining ant colony algorithm and particle swarm optimization is proposed in this study. The aim is to achieve fast and accurate path planning for agricultural information gathering robots in diverse agricultural environments. The global search ability of particle swarm optimization algorithm in finding optimal paths and the local search advantage of ant colony algorithm in obstacle avoidance are used to optimise the movement strategy of robots in agricultural environments. The research results showed that the global path planning distance of this method was 19.328m. The execution time was 0.97s. In local path planning, the proposed algorithm had a fitness function value of 30.123 when the number of iterations reached 53. In mixed path planning, the proposed algorithm reduced the movement time by 3.2s. The conclusion shows that the algorithm proposed in this study has high applicability and efficiency in practical applications, providing an effective strategy for path planning of agricultural information gathering robots. It has important practical significance for promoting the development of intelligent agriculture.

Abstract Background Current hypertension guidelines recommend combination of an angiotensin-converting enzyme inhibitor or angiotensin-receptor blocker with a calcium-channel blocker or thiazide diuretic as initial antihypertensive therapy in patients with monotherapy uncontrolled hypertension. However, to what extent these two different combinations are comparable in blood pressure (BP)-lowering efficacy and safety remains under investigation, especially in the Chinese population. We investigated the BP-lowering efficacy and safety of the amlodipine/benazepril and benazepril/hydrochlorothiazide dual therapies in Chinese patients. Methods In a multi-center, randomized, actively controlled, parallel-group trial, we enrolled patients with stage 1 or 2 hypertension from July 2018 to June 2021 in 20 hospitals and community health centers across China. Of the 894 screened patients, 560 eligible patients were randomly assigned to amlodipine/benazepril 5/10 mg (n = 282) or benazepril/hydrochlorothiazide 10/12.5 mg (n = 278), with 213 and 212 patients, respectively, who completed the study and had a valid repeat ambulatory BP recording during follow-up and were included in the efficacy analysis. The primary outcome was the change from baseline to 24 weeks of treatment in 24-h ambulatory systolic BP. Adverse events including symptoms and clinically significant changes in physical examinations and laboratory findings were recorded for safety analysis. Results In the efficacy analysis (n = 425), the primary outcome, 24-h ambulatory systolic BP reduction, was − 13.8 ± 1.2 mmHg in the amlodipine/benazepril group and − 12.3 ± 1.2 mmHg in the benazepril/hydrochlorothiazide group, with a between-group difference of − 1.51 (p = 0.36) mmHg. The between-group differences for major secondary outcomes were − 1.47 (p = 0.18) in 24-h diastolic BP, − 2.86 (p = 0.13) and − 2.74 (p = 0.03) in daytime systolic and diastolic BP, and − 0.45 (p = 0.82) and − 0.93 (p = 0.44) in nighttime systolic and diastolic BP. In the safety analysis (n = 560), the incidence rate of dry cough was significantly lower in the amlodipine/benazepril group than in the benazepril/hydrochlorothiazide group (5.3% vs 10.1%, p = 0.04). Conclusions The amlodipine/benazepril and benazepril/hydrochlorothiazide dual therapies were comparable in ambulatory systolic BP lowering. The former combination, compared with the latter, had a greater BP-lowering effect in the daytime and a lower incidence rate of dry cough. Trial registration ClinicalTrials.gov, NCT03682692. Abstract Intestinal fibrosis is one of the major complications of inflammatory bowel disease (IBD) and a pathological process that significantly impacts patient prognosis and treatment selection. Although current imaging assessment and clinical markers are widely used for the diagnosis and stratification of fibrosis, these methods suffer from subjectivity and limitations. In this study, we aim to develop a radiomics diagnostic model based on multi-slice computed tomography (MSCT) and clinical factors. MSCT images and relevant clinical data were collected from 218 IBD patients, and a large number of quantitative image features were extracted. Using these features, we constructed a radiomics model and transformed it into a user-friendly diagnostic nomogram. A nomogram was developed to predict fibrosis in IBD by integrating multiple factors. The nomogram exhibited favorable discriminative ability, with an AUC of 0.865 in the validation sets, surpassing both the logistic regression (LR) model (AUC = 0.821) and the clinical model (AUC = 0.602) in the test set. In the train set, the LR model achieved an AUC of 0.975, while the clinical model had an AUC of 0.735. The nomogram demonstrated superior performance with an AUC of 0.971, suggesting its potential as a valuable tool for predicting fibrosis in IBD and improving clinical decision-making. The radiomics nomogram, incorporating MSCT and clinical factors, demonstrates promise in stratifying fibrosis in IBD. The nomogram outperforms traditional clinical models and offers personalized risk assessment. However, further validation and addressing identified limitations are necessary to enhance its applicability.

Abstract Objective Iron accumulation is associated with osteoporosis. This study aims to explore the effect of chronic iron accumulation induced by hepcidin1 deficiency on aging osteoporosis. Methods Iron accumulation in hepcidin1 knockout aging mice was assessed by atomic absorption spectroscopy and Perl’s staining. Bone microarchitecture was observed using Micro-CT. Hepcidin, ferritin, oxidative stress, and markers of bone turnover in serum were detected by enzyme-linked immunosorbent assay. Bone formation and resorption markers were measured by real-time quantitative PCR. Cell aging was induced by D-galactose treatment. CCK-8, flow cytometry, EdU assays, and Alizarin red staining were performed to reveal the role of hepcidin1 knockout in cell model. Iron Colorimetric Assay Kit and western blot were applied to detect iron and ferritin levels in cells, respectively. Results In hepcidin1-knockout mice, the ferritin and iron contents in liver and tibia were significantly increased. Iron accumulation induced by hepcidin1 knockout caused a phenotype of low bone mass and deteriorated bone microarchitecture. Osteogenic marker was decreased and osteoclast marker was increased in mice, accompanied by increased oxidative stress level. The mRNA expression levels of osteoclast differentiation markers (RANKL, Mmp9, OPG, Trap, and CTSK) were up-regulated, while bone formation markers (OCN, ALP, Runx2, SP7, and Col-1) were down-regulated in model group, compared to wild type mice. In vitro, hepcidin1 knockdown inhibited proliferation and osteogenic differentiation, while promoted apoptosis, with increased levels of iron and ferritin. Conclusion Iron accumulation induced by hepcidin1 deficiency aggravates the progression of aging osteoporosis via inhibiting osteogenesis and promoting osteoclast genesis.

Abstract Lactic acid bacteria (LAB) are widely used in the food industry, but little is known about their population genomics, hindering our understanding of their genetic background and evolution. To better understand the evolution of LAB and their potential applications to related food production, we conducted a scoping review of the literature focusing on genomic issues related to LAB. The research progress of LAB population and functional genomics was summarized by sorting the literature of LAB genomics research in the last 10–15 years. According to the existing evidence, the formation mechanism of characteristics of LAB and their habitat adaptive evolution directed by genome decay and horizontal gene transfer were elucidated. The major evolutionary driving forces of LAB are mutations and horizontal gene transfer (HGT) events with or without the involvement of mobile genetic elements instead of recombination. We also underline the application of LAB genome in food safety and properties. Collectively, this review paper provides up‐to‐date information of the population and functional genomics of LAB, current challenges in the field, and suggestions for improving the exploitation and application of LAB resources.

Abstract Background Haemorrhage transformation (HT) is a serious complication of intravenous thrombolysis (IVT) in acute ischaemic stroke (AIS). Accurate and timely prediction of the risk of HT before IVT may change the treatment decision and improve clinical prognosis. We aimed to develop a deep learning method for predicting HT after IVT for AIS using noncontrast computed tomography (NCCT) images. Methods We retrospectively collected data from 828 AIS patients undergoing recombinant tissue plasminogen activator (rt-PA) treatment within a 4.5-h time window (n = 665) or of undergoing urokinase treatment within a 6-h time window (n = 163) and divided them into the HT group (n = 69) and non-HT group (n = 759). HT was defined based on the criteria of the European Cooperative Acute Stroke Study-II trial. To address the problems of indiscernible features and imbalanced data, a weakly supervised deep learning (WSDL) model for HT prediction was constructed based on multiple instance learning and active learning using admission NCCT images and clinical information in addition to conventional deep learning models. Threefold cross-validation and transfer learning were performed to confirm the robustness of the network. Of note, the predictive value of the commonly used scales in clinics associated with NCCT images (i.e., the HAT and SEDAN score) was also analysed and compared to measure the feasibility of our proposed DL algorithms. Results Compared to the conventional DL and ML models, the WSDL model had the highest AUC of 0.799 (95% CI 0.712–0.883). Abstract Wearable sweat sensors are becoming increasingly popular for their robust capabilities in non-invasive, dynamic, and continuous real-time monitoring of biological information. Real-time monitoring of large-scale samples is crucial for realizing intelligent health. A major bottleneck for enabling large-scale sweat elucidation is the fabrication of wearable sensors equipped with microfluidic devices and flexible electrodes in a cost-effective, homogeneous performance and rapid large-scale way. Herein, a “Screen+Wax”-printing technique was introduced to prepare these components and construct “Lego Bricks” type wearable sweat sensor sensor to monitor sweat Na+ and K+. Flexible electrode arrays and paper-based microfluidic layers (they act as building blocks) were fabricated on polyethylene terephthalate and paper surfaces, respectively, using screen printing and wax printing. Gold nanoparticles and Na+/K+ ion-selective membranes were modified on the electrode surfaces by electrodeposition and drop coating, respectively. In this work, we highlight the excellent performance of the “Lego Bricks” type wearable sweat sensor in testing the Na+ and K+ imbalance of sweat from different body regions during exercise and, more significantly, to track the physical activity during prolonged exercise under different interventions. Furthermore, the prepared “Lego Bricks” wearable sweat ion electrochemical sensor is demonstrated to be capable of large-scale samples elucidation with outstanding performance and cost-effectiveness, which is expected to deeply integrate sweat monitoring into physical activity, providing an important tool for intelligent health.

Abstract Background Approximately 40% of patients with hormone receptor (HR)‐positive and human epidermal growth factor receptor 2 (HER2)‐negative advanced breast cancer (ABC) exhibit PIK3CA mutations. Aims This study aims to evaluate the safety, tolerability, pharmacokinetics, and preliminary antitumor activity of HS‐10352, a selective PI3Kα inhibitor, in this patient population. Materials and Methods Conducted as a phase 1 dose‐escalation trial, HS‐10352 was administered orally once‐daily (QD) at dose levels of 2, 4, 6, and 8 mg. The primary endpoints were dose‐limiting toxicity (DLT) and the maximum tolerated dose (MTD). This study is registered at ClinicalTrials.gov (NCT04631835). Results Between August 2020 and March 2022, a total of 18 female patients were enrolled. DLT, manifested as hyperglycemia, occurred in two patients in the 8 mg QD group, establishing an MTD of 6 mg QD. The most common treatment‐related adverse events were hyperglycemia (88.9%) and weight loss (61.3%). In the 6 mg QD group, four patients (66.7%) had a partial response (PR), and one (16.7%) had stable disease (SD). Among the four patients with PIK3CA mutated tumors in this dosage group, three (75.0%) had PR and one (25.0%) had SD. The median progression‐free survival was not reached (95% confidence interval, 11.1‐NA). Discussion and Conclusion HS‐10352 at 6 mg QD was well‐tolerated in patients with HR‐positive, HER2‐negative ABC, and showed preliminary antitumor activity in patients with PIK3CA mutated tumors. These findings support the further clinical development of HS‐10352.

Hyperspectral remote sensing image (HSI) fusion with multispectral remote sensing images (MSI) improves data resolution. However, current fusion algorithms focus on local information and overlook long-range dependencies. The parameter of network tuning prioritizes global optimization, neglecting spatial and spectral constraints, and limiting spatial and spectral reconstruction capabilities. This study introduces SwinGAN, a fusion network combining Swin Transformer, CNN, and GAN architectures. SwinGAN's generator employs a detail injection framework to separately extract HSI and MSI features, fusing them to generate spatial residuals. These residuals are injected into the supersampled HSI to produce the final image, while a pure CNN architecture acts as the discriminator, enhancing the fusion quality. Additionally, we introduce a new adaptive loss function that improves image fusion accuracy. The loss function uses L1 loss as the content loss, and spatial and spectral gradient loss functions are introduced to improve the spatial representation and spectral fidelity of the fused images. Our experimental results on several datasets demonstrate that SwinGAN outperforms current popular algorithms in both spatial and spectral reconstruction capabilities. The ablation experiments also demonstrate the rationality of the various components of the proposed loss function.

ABSTRACT: Egg production is an important economic trait in layer ducks and understanding the genetics basis is important for their breeding. In this study, a genome-wide association study (GWAS) for egg production traits in 303 female Longyan Shan-ma ducks was performed based on a genotyping-by-sequencing strategy. Sixty-two single nucleotide polymorphisms (SNPs) associated with egg weight traits were identified (P < 9.48 × 10-5), including 8 SNPs at 5% linkage disequilibrium (LD)-based Bonferroni-corrected genome-wide significance level (P < 4.74 × 10-6). One hundred and nineteen SNPs were associated with egg number traits (P < 9.48 × 10-5), including 13 SNPs with 5% LD-based Bonferroni-corrected genome-wide significance (P < 4.74 × 10-6). These SNPs annotated 146 target genes which contained known candidate genes for egg production traits, such as prolactin and prolactin releasing hormone receptor. This study identified that these associated genes were significantly enriched in egg production-related pathways (P < 0.05), such as the oxytocin signaling, MAPK signaling, and calcium signaling pathways. It was notable that 18 genes were differentially expressed in ovarian tissues between higher and lower egg production in Shan-ma ducks. The identified potential candidate genes and pathways provide insight into the genetic basis underlying the egg production trait of layer ducks.

ABSTRACT: In the spring of 2023, 10 to 21-day-old chicks in a broiler duck farm in Shandong Province, China, developed swelling of the head and neck, moist eyes with mucous discharge, difficulty in walking, shrinking of the neck, and loose and disorganized coat. Anatomical observation revealed hemorrhages in the esophageal mucosa, myocardium, and liver, and severe hemorrhages in the trachea with copious inflammatory secretions. Soon after, similar symptoms appeared in a large number of ducks in the flock, which eventually led to the elimination of all the 20,000-odd newly introduced ducklings on the farm, resulting in huge economic losses. We detected duck plague virus in the tissues of liver, spleen and lungs of diseased and dead ducks, and successfully isolated the pathogenic strain, named SD423, by inoculating duck embryos and inoculating duck embryo fibroblasts. We successfully conducted animal regression experiments with the isolated strain, and the experimental animals in the 1 d of age group showed symptoms of swollen eyes and tearing, shrinking of the neck, crouching, and hemorrhage in organs such as the liver and intestines successively from the 3rd d. We sequenced the whole genome of the isolated duck plague strain, and by comparing the homology with the published duck plague virus whole sequences in Genbank, the virus strain obtained in this study had the highest homology with the Chinese virulent strain SD (MN518864.1), with nucleotide (nt) homology of about 99.90% and amino acid (aa) homology of about 99.75%, which indicated that the isolate is a virulent strain. Previously, it was reported that the natural infection of duck plague virus mainly occurs above 30 d of age, but the duck plague virus found in this study can naturally infect ducklings up to 20 d of age, and the mortality rate is as high as 100%. In this study, the pathogenicity test and whole genome sequence analysis of this isolate provided data support and theoretical basis for further research on pathogenicity and virulence-related gene analysis of duck plague virus.

AimsIn the year 2021, human brucellosis ranked fifth in terms of the number of cases among all statutorily notifiable infectious diseases in China, thus remaining a significant concern for public health. This study aims to provide insights into the financial burden of human brucellosis by examining hospital stays and associated costs for affected individuals.MethodsIn this retrospective study, we gathered updated data from 467 inpatient cases primarily diagnosed with human brucellosis at eight major tertiary hospitals in Beijing, China, spanning from 2013 to 2023. To comprehensively explore the economic impact on individuals, we not only analyzed the duration of hospital stays and total costs but also examined various charge types, including drug, lab test, medical imaging, medical treatment, surgical procedures, medical supplies and consumables, inpatient bed care, nursing services, and other services costs. Statistical analysis was employed to compare differences among gender, age, ethnicity, type of health insurance, condition at admission, comorbidity index, the performance of surgery, and the site of infection.ResultsBoth the length of stay and total cost exhibited significant variations among insurance, surgery, and infection site groups. Utilization categories demonstrated significant differences between patients who underwent surgery and those who did not, as well as across different infection sites. Furthermore, multiple linear regression analysis revealed that the condition at admission, Elixhauser comorbidity index, infection site, and surgery influenced both hospital stay and total cost. In addition, age and insurance type were associated with total costs.ConclusionBy delving into various utilization categories, we have addressed a significant gap in the literature. Our findings provide valuable insights for optimizing the allocation and management of health resources based on the influencing factors identified in this study.

Brucellosis is a zoonotic disease caused by a Gram-negative coccus a facultative intracellular pathogen. Neurobrucellosis has an incidence rate of 3-7% among all patients with brucellosis, while spinal cord involvement is rare and carries a significant mortality risk. This report describes a case of brucellosis myelitis in a 55-year-old male patient who presented with recurrent paralysis, incontinence, and damage to the visual and auditory nerves. The diagnosis of neurobrucellosis involves a serum tube agglutination test, cerebrospinal fluid analysis, a physical examination of the nervous system, and a comprehensive review of the patient’s medical history. The presence of brucellosis was confirmed in cerebrospinal fluid using MetaCAP™ sequencing. Treatment with a combination of rifampicin, doxycycline, ceftriaxone sodium, amikacin, compound brain peptide ganglioside, and dexamethasone resulted in significant improvement of the patient’s clinical symptoms and a decrease in the brucellosis sequence count in cerebrospinal fluid. For the first time, MetaCAP™ sequencing has been used to treat pathogenic microbial nucleic acids, which could be a valuable tool for early diagnosis and treatment of neurobrucellosis.

The purpose of this study is to undertake a comparative study of higher education indices in China and between the 31 administrative divisions of China with the international evaluation system. The analysis includes a comparison of higher education indices between China and six continents and several key countries in the world, as well as a comparison between internal regions in China and Malaysia, both of which are in Asia. The numbers and rankings of higher education institutes (HEIs) of various countries and regions listed in the QS World University Rankings (QS) over 13 years from 2010 to 2023 form the basis of corresponding higher education indices obtained through several calculation formulas. These indices are the QS Higher Education Density Index (QSHEDI), QS Higher Education Quality Index (QSHEQI), QS Higher Education Composite Index (QSHEI), and Relative QS Higher Education Composite Index (RQSHEI), and they reflect the index calculation results of the density, quality, and composite of higher education relative to the regional population. Based on the comparative analysis of the indices from multiple perspectives, this study found that there is a large gap between higher education in China and other countries, and there is also a significant imbalance in the geographical distribution of higher education resources among Chinese provinces based on the population proportion and economic impact. These disparities will hurt the sustainable development of higher education in China. Therefore, paying more attention to the international evaluation system and recognition standards like the QS Rankings becomes necessary to guide the balanced development of higher education among provinces according to the population proportion. It is also vital to improve the international influence of HEIs in China according to internationally recognized requirements so that China's higher education can develop sustainably in the process of global internationalization.

New energy vehicles (NEVs) are crucial way to solving environmental and energy dilemmas. However, the transition from traditional energy vehicles (TEVs) to NEVs is extremely intricate. If the process for TEVs switching to NEVs falls into out order, the automobile industry will be shaken. This paper takes electric vehicles (EVs), representing NEVs, and internal combustion engine vehicles (ICEVs), representing TEVs, as research objects, and constructs a dynamic Gounod duopoly model for the production decisions of two oligopolistic automakers under the dual credit policy. Combining the nonlinear dynamic theory and Cournot game, the influence of policy, market, technology, and cost on the stability of duopoly automakers’ dynamic production game equilibrium and the nonlinear evolution characteristics of Cournot-Nash equilibrium are analyzed systematically. Results show that: (1) In the transition from ICEVs to EVs, if the speed of production adjustment is too fast, the automobile system would trap into chaos and shakeouts, and the social welfare would show volatility. (2) The revised dual credit policy increases the credit ratio and decreases the credit coefficient which enhances the stability of automobile system. This revision may accelerate the transformation from ICEVs to EVs in the automobile industry. Especially, the credit coefficient plays a more significant role. (3) The consumer acceptance for vehicle prices and the range of EV are positively correlated with the production of EVs and the social welfare, and negatively correlated with the production of ICEVs. Nevertheless, the cost of the two types of vehicle had the opposite effect compared price acceptance and range of EVs.
