Abstract

International audience; Cell morphology is altered in the migration process, and the underlying cytoskeleton remodeling is highly dependent of intracellular Ca2+ concentration. Many calcium channels are known to be involved in migration. Inositol 1,4,5-trisphosphate receptor (IP3R) was demonstrated to be implicated in breast cancer cells migration, but its involvement in morphological changes during the migration process remains unclear. In the present work, we showed that IP(3)R3 expression was correlated to cell morphology. IP(3)R3 silencing induced rounding shape and decreased adhesion in invasive breast cancer cell lines. Moreover, IP(3)R3 silencing decreased ARHGAP18 expression, RhoA activity, Cdc42 expression and (Y861)FAK phosphorylation. Interestingly, IP(3)R3 was able to regulate profilin remodeling, without inducing any myosin II reorganization. IP3R3 silencing revealed an oscillatory calcium signature, with a predominant oscillating profile occurring in early wound repair. To summarize, we demonstrated that IP3R3 is able to modulate intracellular Ca2+ availability and to coordinate the remodeling of profilin cytoskeleton organization through the ARHGAP18/RhoA/mDia1/PAK pathway.

Abstract

International audience; Apoptosis is a regulated process, leading to cell death, which is involved in several pathologies including neurodegenerative diseases and stroke. Caspase-3 is a key enzyme of the apoptotic pathway and is considered as a major target for the treatment of abnormal cell death. Sensitive and non-invasive methods to monitor caspase-3 activity in cells and in the brain of living animals are needed to test the efficiency of novel therapeutic strategies. In the present study, we have biochemically characterized a caspase-3 far-red fluorescent probe, QCASP3.2, that can be used to detect apoptosis in vivo. The specificity of cleavage of QCASP3.2 was demonstrated using recombinant caspases and protease inhibitors. The functionality of the probe was also established in cere-bellar neurons cultured in apoptotic conditions. QCASP3.2 did not exhibit any toxicity and appeared to accurately reflect the induction and inhibition of caspase activity by H 2 O 2 and PACAP, respectively, both in cell lysates and in cultured neurons. Finally, intravenous injection of the probe after cere-bral ischemia revealed activation of caspase-3 in the infarcted hemisphere. Thus, the present study demonstrates that QCASP3.2 is a suitable probe to monitor apoptosis both in vitro and in vivo and illustrates some of the possible applications of this caspase-3 fluorescent probe.

Abstract

The precision limit for the considered setups are depicted below. For TD and ITD fDOT, we note that (i) the accuracy of the depth estimation decreases with the depth of the fluorescent volume and (ii) this accuracy substatially depends on the fluorescence life-time τ. The longer the lifetime, the poorer the precision limit. Moreover, for realistic lifetime, the CW and the ITD setups in reflection geometry achieve comparable precision limits.

Abstract

Luminescence diffuse optical tomography has become a valuable tool in optical molecular imaging. As an actor of this recent field, the French company Quidd has developed a luminescence reflectance imaging system for small animal in vivo imaging. In previous studies, we have determined the potential and the limits of such approaches for the determination of the depth of fluorophores/bioluminescent substrates in tissues [1,2]. In the present work, we demonstrate the first reconstructions on test samples. For this study, we use a prototype of the Quidd Optical System (QOS) as well as calibrated phantom (cf. Fig. 1) and source. The optical detection set-up is composed of a cooled CCD camera with a telecentric lens. The camera can be translated along the z and x axes and rotated with an angular amplitude of 120°. The phantom is a diffusive epoxy resin hemisphere whose optical properties are given in Fig. 1 for 635 nm. An optical fiber inserted inside the phantomand connected to an hallogen lamp with a 635 nm filter is used to modelized a point-like isotropic bioluminescent source. The output measured power of the fiber is 5 mW. The light-transport (forward) model relies on the diffusion equation and the inverse problem is solved using a least-squares criterion. The optimization method [3] is easy and fast to implement, and does not use an explicit Tikhonov regularization. Our aim is to localize the bioluminescent point-like source inside the phantom, and to reconstruct its power. Three experiments were performed for three positions of the fiber. In each case, a single reflectance image was taken with the camera right above the phantom. The results given by the algorithm are gathered in the following figure: the sources are located with sub-millimiter precision and their power is estimated with a fairly good accuracy

Abstract

International audience; The distribution and density of pituitary adenylate cyclase-activating polypeptide (PACAP) binding sites have been investigated in the brain of the primates Jacchus callithrix (marmoset) and Macaca fascicularis (macaque) using [(125)I]-PACAP27 as a radioligand. PACAP binding sites were widely expressed in the brain of these two species with particularly high densities in the septum, hypothalamus and habenula. A moderate density of recognition sites was seen in all subdivisions of the cerebral cortex with a heterogenous distribution, the highest concentrations occurring in layers I and VI while the underlying white matter was almost devoid of binding sites. Reverse transcriptase-polymerase chain reaction (RT-PCR) analysis revealed intense expression of the mRNAs encoding the short and hop-1 variants of pituitary adenylate cyclase-activating polypeptide-specific receptor (PAC1-R) in the cortex of both marmoset and macaque, whereas vasoactive intestinal polypeptide/pituitary adenylate cyclase-activating polypeptide mutual receptor, subtype 1 (VPAC1-R) and vasoactive intestinal polypeptide/pituitary adenylate cyclase-activating polypeptide mutual receptor, subtype 2 (VPAC2-R) mRNAs were expressed at a much lower level. In situ hybridization histochemistry showed intense expression of PAC1-R and weak expression of VPAC1-R mRNAs in layer IV of the cerebral cortex. Incubation of cortical tissue slices with PACAP induced a dose-dependent stimulation of cyclic AMP formation, indicating that PACAP binding sites correspond to functional receptors. Moreover, treatment of primate cortical slices with 100 nM PACAP significantly reduced the activity of caspase-3, a key enzyme of the apoptotic cascade. The present results indicate that PACAP should exert the same neuroprotective effect in the brain of primates as in rodents and suggest that PAC1-R agonists may have a therapeutic value to prevent neuronal cell death after stroke or in specific neurodegenerative diseases.

Abstract

The development of phenyl-dithioethyloxycarbonyl (Phdec) and 2-pyridyldithioethyloxycarbonyl (Pydec) protecting groups, which are thiollabile urethanes, is described. These new disulfide-based protecting groups were introduced onto the epsilon-amino group of L-lysine; the resulting amino acid derivatives were easily converted into Nalpha-Fmoc building blocks suitable for both solid- and solution-phase peptide synthesis. Model dipeptide-(Ardec)s were prepared by using classical peptide couplings followed by standard deprotection protocols. They were used to optimize the conditions for complete thiolytic removal of the Ardec groups both in aqueous and organic media. Phdec and Pydec were found to be cleaved within 15 to 30 min under mild reducing conditions: i) by treatment with dithiothreitol or beta-mercaptoethanol in Tris-HCl buffer (pH 8.5-9.0) for deprotection in water and ii) by treatment with beta-mercapto-ethanol and 1,8-diazobicyclo-[5.4.0]undec-7-ene (DBU) in N-methyl-pyrrolidinone for deprotection in an or ganic medium. Successful solid-phase synthesis of hexapeptides Ac-Lys-Asp-Glu-Val-Asp-Lys(Ardec)-NH 2 has clearly demonstrated the full orthogonality of these new amino protecting groups with Fmoc and Boc protections. The utility of the Ardec orthogonal deprotection strategy for site-specific chemical modification of peptides bearing several amino groups was illustrated firstly by the preparation of a fluorogenic substrate for caspase-3 protease containing the cyanine dyes Cy 3.0 and Cy 5.0 as FRET donor/acceptor pair, and by solid-phase synthesis of an hexapeptide bearing a single biotin reporter group. ©Wiley-VCH Verlag GmbH & Co.
