Acute pancreatitis (AP) is a potentially fatal condition with no targeted treatment options. Although inhibiting xanthine oxidase (XO) in the treatment of AP has been studied in several experimental models and clinical trials, whether XO is a target of AP and what its the main mechanism of action is remains unclear. Here, we aimed to re-evaluate whether XO is a target aggravating AP other than merely generating reactive oxygen species that trigger AP. We first revealed that XO expression and enzyme activity were significantly elevated in the serum and pancreas of necrotizing AP models. We also found that allopurinol and febuxostat, as purine-like and non-purine XO inhibitors, respectively, exhibited protective effects against pancreatic acinar cell death in vitro and pancreatic damage in vivo at different doses and treatment time points. Moreover, we observed that conditional Xdh overexpression aggravated pancreatic necrosis and severity. Further mechanism analysis showed that XO inhibition restored the hypoxia-inducible factor 1-alpha (HIF-1α)-regulated lactate dehydrogenase A (LDHA) and NOD-like receptor family pyrin domain containing 3 (NLRP3) signaling pathways and reduced the enrichment of 13C6-glucose to 13C3-lactate. Lastly, we observed that clinical circulatory XO activity was significantly elevated in severe cases and correlated with C-reactive protein levels, while pancreatic XO and urate were also increased in severe AP patients. These results together indicated that proper inhibition of XO might be a promising therapeutic strategy for alleviating pancreatic necrosis and preventing progression of severe AP by downregulating HIF-1α-mediated LDHA and NLRP3 signaling pathways.

Abstract Depressive symptoms occur commonly in Alzheimer’s disease (AD). Although abnormalities in the amygdala-frontal circuit have been linked to emotional dysregulation and cognitive impairment, the neurological basis underlying these associations in AD patients with depressive symptoms (ADD) is unclear. We aimed to investigate the relationship between the amygdala-frontal circuit and depressive symptoms and cognitive function in ADD. We recruited 60 ADD, 60 AD patients without depressive symptoms (ADND), and 60 healthy controls (HC). Functional connectivity (FC) maps of the bilateral amygdala were compared. Fractional anisotropy (FA) of the amygdala-frontal circuit connected by the uncinate fasciculus (UF) was calculated using automated fiber quantification (AFQ). In addition, mediation analysis was performed to explore the effects of the amygdala-frontal circuit on the relationship between depressive symptoms and cognitive function. We found decreased bilateral amygdala FC with the inferior frontal gyrus (IFG) in the ADD group compared to the ADND and HC groups. Moreover, FA in the left frontal UF (nodes 64–97) was significantly lower in the ADD group than ADND group. Notably, amygdala-based FC with IFG and the left frontal UF FA mediated the relationship between depressive symptoms and cognitive function in ADD, with mediating effects ranging between 15 and 18%. Our study is the first to demonstrate the mediating effect of functional and microstructural abnormalities in the amygdala-frontal circuit in ADD. The findings suggest that the amygdala-frontal circuit may underlie emotional dysregulation in ADD, providing potential targets for treatment strategies.

Abstract How disruptions to normal cell differentiation link to tumorigenesis remains incompletely understood. Wilms tumor, an embryonal tumor associated with disrupted organogenesis, often harbors mutations in epigenetic regulators, but their role in kidney development remains unexplored. Here, we show at single-cell resolution that a Wilms tumor-associated mutation in the histone acetylation reader ENL disrupts kidney differentiation in mice by rewiring the gene regulatory landscape. Mutant ENL promotes nephron progenitor commitment while restricting their differentiation by dysregulating transcription factors such as Hox clusters. It also induces abnormal progenitors that lose kidney-associated chromatin identity. Furthermore, mutant ENL alters the transcriptome and chromatin accessibility of stromal progenitors, resulting in hyperactivation of Wnt signaling. The impacts of mutant ENL on both nephron and stroma lineages lead to profound kidney developmental defects and postnatal mortality in mice. Notably, a small molecule inhibiting mutant ENL’s histone acetylation binding activity largely reverses these defects. This study provides insights into how mutations in epigenetic regulators disrupt kidney development and suggests a potential therapeutic approach.

Introduction: The level of atmospheric CO2 has continuously been increasing and the resulting greenhouse effects are receiving attention globally. Carbon removal from the atmosphere occurs naturally in various ecosystems. Among them, saline environments contribute significantly to the global carbon cycle. Carbonate deposits in the sediments of salt lakes are omnipresent, and the biological effects, especially driven by halophilic microalgae and bacteria, on carbonate formation remain to be elucidated. Objectives: The present study aims to characterize the carbonates formed in saline environments and demonstrate the mechanisms underlying biological-driven CO2 removal via microalgal-bacterial consortium. Methods: The carbonates naturally formed in saline environments were collected and analyzed. Two saline representative organisms, the photosynthetic microalga Dunaliella salina and its mutualistic halophilic bacteria Nesterenkonia sp. were isolated from the inhabiting saline environment and co-cultivated to study their biological effects on carbonates precipitation and isotopic composition. During this process, electrochemical parameters and Ca2+ flux, and expression of genes related to CaCO3 formation were analyzed. Genome sequencing and metagenomic analysis were conducted to provide molecular evidence. Results: The results showed that natural saline sediments are enriched with CaCO3 and enrichment of genes related to photosynthesis and ureolysis. The co-cultivation stimulated 54.54% increase in CaCO3 precipitation and significantly promoted the absorption of external CO2 by 49.63%. A pH gradient was formed between the bacteria and algae culture, creating 150.22 mV of electronic potential, which might promote Ca2+ movement toward D. salina cells. Based on the results of lab-scale induction and 13C analysis, a theoretical calculation indicates a non-negligible amount of 0.16 and 2.3 Tg C/year carbon sequestration in China and global saline lakes, respectively. Conclusion: The combined effects of these two typical representative species have contributed to the carbon sequestration in saline environments, by promoting Ca2+ influx and increase of pH via microalgal and bacterial metabolic processes.

Glial fibrillary acidic protein (GFAP) and neurofilament light (NfL) are putative non-amyloid biomarkers indicative of ongoing inflammatory and neurodegenerative disease processes. Hence, this study aimed to demonstrate the relationship between plasma biomarkers (GFAP and NfL) and 18F-AV-1451 tau PET images, and to explore their effects on cognitive function. Ninety-one participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI) database and 20 participants from the Shanghai Action of Prevention Dementia for the Elderly (SHAPE) cohort underwent plasma biomarker testing, 18F-AV-1451 tau PET scans and cognitive function assessments. Within the ADNI, there were 42 cognitively normal (CN) individuals and 49 with mild cognitive impairment (MCI). Similarly, in the SHAPE, we had 10 CN and 10 MCI participants. We calculated the standardized uptake value ratios (SUVRs) for the regions of interest (ROIs) in the 18F-AV-1451 PET scans. Using plasma biomarkers and regional SUVRs, we trained machine learning models to differentiate between MCI and CN subjects with ADNI database and validated in SHAPE.Results showed that eight selected variables (including left amygdala SUVR, right amygdala SUVR, left entorhinal cortex SUVR, age, education, plasma NfL, plasma GFAP, plasma GFAP/ NfL) identified by LASSO could differentiate between the MCI and CN individuals, with AUC ranging from 0.783 to 0.926. Additionally, cognitive function was negatively associated with the plasma biomarkers and tau deposition in amygdala and left entorhinal cortex. Increased tau deposition in amygdala and left entorhinal cortex were related to increased plasma biomarkers. Moreover, tau pathology mediated the effect of plasma biomarkers level on the cognitive decline. The present study provides valuable insights into the association among plasma markers (GFAP and NfL), regional tau deposition and cognitive function. This study reports the mediation effect of brain regions tau deposition on the plasma biomarkers level and cognitive function, indicating the significance of tau pathology in the MCI patients.

Photovoltaic power generation and energy storage technology are current hotspots in the clean energy industry. As a core piece of equipment, an inverter is subjected to higher demands for its voltage regulation range and output performance. A differential boost inverter features a boosting function not found in traditional inverters, effectively widening the input voltage range, with characteristics of a single-stage structure and simple modulation. However, the differential boost inverter faces the issue of high common-mode voltage. Considering that the magnitude of the common-mode voltage changes with different modulation methods, a novel modulation strategy with a flexible common-mode voltage is proposed, which can effectively suppress the common-mode voltage under the same gain without affecting the output power quality. This article first introduces the topology, working principle, and traditional modulation methods of the differential boost inverter. Then, the new modulation strategy is defined, and the gain equation is derived. The new modulation method’s applicable scenarios are discussed and compared to other modulation methods based on the gain curve, concluding that the new method has advantages in terms of common-mode voltage amplitude and power quality under low boost gain. Finally, the effectiveness of the proposed method is verified through simulation and experiments.

The Junggar Basin is rich in oil and gas resources, with huge exploration potential, which is one of the main arenas of increasing oil and gas reserve and producing in China. In recent years, CNPC has made major breakthroughs in exploration in the Junggar Basin, revealing the orderly symbionic distribution pattern of conventional and conventional oil and gas. In order to further learn from the oil and gas achievements and exploration and development experience in neighboring areas and the new geological theories, for formulating SINOPEC's oil and gas exploration strategy in the Junggar Basin, the potential and distribution characteristics of the oil and gas resources in the Junggar Basin are systematically analyzed in this paper and it is pointed out that no enough attention is paid to natural gas and unconventional petroleum. The key breakthrough direction and exploration strategy of SINOPEC in the Junggar Basin are put forward in the paper after reviewing the major exploration breakthroughs in multiple fields, multiple strata, and multiple types in recent years and combining with the oil and gas geolo-gical conditions and exploration practice of SINOPEC's mining rights area. The research reveals that the Junggar Basin has entered a new period of exploration dominated by the middle and lower assemblage, and the exploration ideas must be adjusted accordingly. We must insist on the transformation from early outside-source exploration to intra-source and near-source exploration, and from early conventional petroleum exploration to both conventional and unconventional petroleum exploration. In order to make big exploration breakthrough of SINOPEC and realize the target of increasing reserves and production in the near and medium term in the Junggar Basin as soon as possible, four exploration strategies are proposed in the paper, including further figuring out the resources, establishing an integrated research team of conventional and unconventional petroleum, strengthening the joint research of geological and engineering integration, and increasing the support of risk exploration.

Abstract To understand the epidemiological and genetic characteristics of B19V, a multiple-province surveillance of patients with febrile rash illnesses (FRIs) were conducted in China during 2009 ~ 2021. The clinical specimens of 3,820 FRI patients were collected and tested for B19V DNA. A total of 99 (2.59%) patients were positive for B19V, and 49 (49.49%) were children under 5 years old. B19V infections occurred throughout the year without obvious seasonal pattern. Ten NS1-VP1u sequences and seven genome sequences were obtained in this study, identified as subgenotype 1a. Combined with the globally representative genome sequences, no temporal and geographic clustering trends of B19V were observed, and there was no significant correlation between B19V sequences and clinical manifestations. The evolutionary rate of the B19V genome was 2.30 × 10–4 substitutions/site/year. The number of negative selection sites was higher than that of positive selection sites. It was the first to comprehensively describe the prevalence patterns and evolutionary characteristics of B19V in FRI patients in China. B19V played the role in FRI patients. Children under 5 years old were the main population of B19V infection. Subgenotype 1a was prevalent in FRI patients in China. B19V showed a high mutation rate, while negative selection acted on the genome.

Abstract Background Obesity has been confirmed to be associated with infertility. However, the association between metabolically healthy obesity (MHO), a subset of obesity with no metabolic abnormalities, and female infertility has not yet been investigated. This study aimed to examine the association between MHO and the risk of female infertility among United States. Methods This study utilized a cross-sectional design and included 3542 women aged 20–45 years who were selected from the National Health and Nutrition Examination Survey (NHANES) 2013–2020 database. The association between MHO and the risk of infertility was evaluated using risk factor–adjusted logistic regression models. Results Higher BMI and WC were associated with increased infertility risk after adjusting for potential confounding factors (OR (95% CI): 1.04(1.02, 1.06), P = 0.001; OR (95% CI): 1.02 (1.01, 1.03), P

The lower play of the Junggar Basin has huge oil and gas potential, which is the most important strategic succession field for oil and gas exploration. Large-scale delta sand bodies are developed in Permian and Triassic in the four sags in the hinterland, in which new discoveries of oil and gas have been made in succession. In order to reveal the sedimentary characteristics, sedimentary model, and distribution patterns of sand bodies in the hinterland, the studies on basin prototype, sequence framework, paleogeomorphic restoration and sedimentary system were systematically carried out based on a large number of newly drilled cores, logging and geophysical data. The results show that: the development background of large depression lake basin can be identified in the development period of the Upper Permian-Lower Triassic shallow water braided river delta in the hinterland of the basin, and the study area has the characteristics of flat terrain, small slope, sufficient material supply, extremely shallow and frequently turbulent water bodies, alternating oxidation and reduction environments, and overall oxidation environment during the sedimentation period. The shallow braided river delta is characterized by coarse grain size, low impurity content, medium texture maturity, long distance transportation, strong hydrodynamic scouring, cross bedding and parallel bedding. The advantageous water system in the sedimentation period of the Upper Wuerhe and Baikouquan formations in the hinterland mainly comes from the northwest and northeast, and extends from north to south as a whole, with three provenance systems developed, namely Wuerhe provenance, Karamay provenance and Kelameily provenance. The terrain within the basin has obvious zoning characteristics, forming four facies belts namely fan delta area, braided river delta plain area, front area and lake area, thus forming a shallow water braided river delta sedimentary pattern of "large plain, small front" in the hinterland, where plain area and front area are favorable sand body development zones.

The lower play (Permian to Triassic) in the hinterland is the most important strategic succession field for oil and gas exploration in the Junggar Basin. Multiple wells have been drilled into high-quality clastic rock reservoirs below 6 000 m, greatly breaking through the deadline of buried depth of traditional clastic rock effective reservoirs. It has been made clear that the development status and pore evolution process are the key issues that determine whether oil and gas can be accumulated. Taking typical drilling wells in the hinterland as an example, this paper comprehensively analyzed the petrology, physical properties and pore structure characteristics of deep and ultra-deep clastic reservoirs in the hinterland of the Junggar Basin from a qualitative and quantitative perspective, and quantitatively restored the pore evolution process by integrating the microscopic analysis of rock thin sections, porosity and permeability tests, image analysis technology, quantitative characterization of porosity evolution, temperature measurement of inclusions, basin modeling and other methods. The results show that the clastic rock in the lower play of the hinterland is mainly developed in the Permian Upper Wuerhe Formation, Triassic Baikouquan Formation and Triassic Kelamayi Formation, of which the sand in the Triassic Baikouquan Formation is the most developed, followed by the Permian Upper Wuerhe Formation and Triassic Kalamayi Formation. There is little difference in rock types among different layers, which mainly composed of lithic sandstone with a small amount of feldspar lithic sandstone. The composition of rock debris is mainly medium-basic volcanic rock debris, with low content of feldspar and quartz, and the sum of them is generally less than 20%. The Kelamayi Formation is dominated by primary pores with the best reservoir property and the highest porosity of 13.18%. The Upper Wuerhe Formation and Baikouquan Formation are dominated by secondary corrosion pores, and the corrosion materials are mainly medium-basic volcanic debris, laumontite cement and a small amount of feldspar. The reservoir properties of them are not as good as those of the Kelamayi Formation. The pore evolution of the Kelamayi Formation has experienced weak compaction (21.08% pore reduction by compaction), weak cementation (2.88% pore reduction by cementation), and weak corrosion (1.4% pore increase by corrosion). Today's high porosity is mainly due to the large amount of preservation of primary pores under weak compaction and late weak cementation. The Baikouquan Formation and Upper Wuerhe Formation have undergone strong compaction (26.60% and 26.43% pore reduction by compaction, respectively), strong cementation (7.43% and 11% pore reduction by cementation, respectively), and strong corrosion (6.32% and 4.21% pore increase by corrosion, respectively). Secondary corrosion is the main way for them to increase porosity, but it is insufficient to compensate for the porosity reduction effect of strong compaction and cementation, resulting in lower porosity in the two formations.

The Shawan Sag is one of the important exploration areas in the hinterland of the Junggar Basin. In order to better clarify the source of crude oil from the Permian, Jurassic and Cretaceous reservoirs in the sag and identify hydrocarbon accumulation processes and enrichment pattern, the authors conducted a carbon isotope and biomarker analysis on the typical crude oil samples from the eastern Shawan Sag in this study, and further carried out crude oil classification and oil-source correlation based on the analysis of geochemical characteristics of crude oil. The results indicate that the crude oil from different strata in the eastern Shawan Sag can be divided into three types. The first type mainly occurs in the Permian and Jurassic reservoirs, and its whole oil δ13C value and Pr/Ph ratio range from -31.0‰ to -29.0‰ and from 1.0 to 2.0, respectively. The relative contents of αααR regular sterane display the characteristics of C27 < C28 < C29, and the terpanes are mainly characterized by C20 < C21>C23 TT and C24TeT/C26TT < 1, with the gammacerane index lower than 0.30. This type of crude oil is mainly derived from the source rocks in the Middle Permian Lower Wuerhe Formation. In the oil samples from the Upper Permian Upper Wuerhe Formation, Ts content is below the detection limit, the β-carotane/C30 hopane ratio is greater than 1, and the tricyclic terpane distribution displays the characteristics of C20>C21>C23TT. The distribution fraction of methyl phenanthrene reflects that the crude oil is in the stage of high-maturity evolution. All these indicate the mixing of crude oil generated in the Lower Permian source rocks. The second type occurs in the Middle Jurassic reservoirs. The whole oil δ13C value and individual n-alkanes δ13C value are higher than -29.0‰. The Pr/Ph ratio is relatively higher, ranging from 2.0 to 2.5, with C24TeT/C26TT>1, which indicate that this type of crude oil is mainly derived from the Jurassic source rocks. The third type occurs in the Lower Cretaceous reservoirs, and its whole oil δ13C value and Pr/Ph ratio are lower than -31.0‰ and 1.0, respectively. The αααR regular sterane contents display the characteristics of C27≈C28

Vaccination with the primary two-dose series of SARS-CoV-2 mRNA protects against infection with the ancestral strain, and limits the presentation of severe disease after re-infection by multiple variants of concern (VOC), including Omicron, despite the lack of a strong neutralizing response to these variants. We compared antibody responses in serum samples collected from mRNA-1273 (Moderna) vaccinated subjects to identify mechanisms of immune escape and cross-protection. Using pseudovirus constructs containing domain-specific amino acid changes representative of Omicron BA.1, combined with domain competition and RBD-antibody depletion, we showed that RBD antibodies were primarily responsible for virus neutralization and variant escape. Antibodies to NTD played a less significant role in antibody neutralization but acted along with RBD to enhance neutralization. S2 of Omicron BA.1 had no impact on neutralization escape, suggesting it is a less critical domain for antibody neutralization; however, it was as capable as S1 at eliciting IgG3 responses and NK-cell mediated, antibody-dependent cell cytotoxicity (ADCC). Antibody neutralization and ADCC activities to RBD, NTD, and S1 were all prone to BA.1 escape. In contrast, ADCC activities to S2 resisted BA.1 escape. In conclusion, S2 antibodies showed potent ADCC function and resisted Omicron BA.1 escape, suggesting that S2 contributes to cross-protection against Omicron BA.1. In line with its conserved nature, S2 may hold promise as a vaccine target against future variants of SARS-CoV-2.

Abstract Background In seed plants, the ovule is the precursor to the seed. The process of ovule development and differentiation is regulated by multiple factors, including starch metabolism and endogenous hormones. Castanea henryi produces nuts with high nutritional value. However, the high proportion of empty buds restricts the commercial use of the tree. Previous studies have shown that the empty bud phenotype is closely related to ovule abortion. If none of the ovules in the ovary expand rapidly and develop in 7–8 weeks after pollination, an empty bud will form. Therefore, we studied the development and molecular mechanisms underlying single seed formation in C. henryi. Results We found that 49 days after pollination (DAP) is a critical period for the formation of fertile and abortive ovules. The morphology and starch distribution of the fertile and abortive ovules differed significantly at 49 DAP. The fertile ovules were smooth and round in appearance, with a large amount of starch. In contrast, abortive ovules were smaller with only a small amount of starch. The embryo sac of the abortive ovule proceeded to develop abnormally, and the entire ovule lacked starch. We identified 37 candidate genes involved in metabolism with potential roles in the regulation of starch levels. Three ADP-glucose pyrophosphorylase (AGPase) genes, one granule-bound starch synthase (GBSS) gene, and two beta-amylase genes could affect starch accumulation. The levels of auxin, cytokinins, gibberellins, and jasmonic acid in fertile ovules were higher than those in abortive ovules. In addition, the levels of endogenous abscisic acid and salicylic acid in abortive ovules were higher than those in fertile ovules of the same age, consistent with the expression patterns of genes related to the synthesis of abscisic and salicylic acid and signal transduction. We identified and mapped the differentially expressed genes associated with hormone synthesis and signal transduction. Conclusions These results improve our general understanding of the molecular mechanisms underlying single seed development in C. henryi and the phenomenon of empty buds, providing directions for future research.

A bandwidth-tunable multiband microwave photonic filter (MPF) using a strongly coupled microring resonator (MRR) with optical phase modulation is demonstrated theoretically and experimentally. Two notches of the MRR adjacent to the optical carrier break the intrinsic balance of the phase-modulated signal and then two sub-MPFs are formed thanks to the conversion from phase modulation to intensity modulation (PM-IM). The frequency responses of the two sub-MPFs are superimposed to form a passband with a fixed center frequency. Due to the periodicity of the MRR transmission spectrum, multiple passbands separated by a certain spacing appear in the frequency domain and combine to form a multiband MPF, whose bandwidth can be adjusted by tuning the wavelength of the carrier. The tunable bandwidth and shape factors ranging from 0.73 GHz to 2.73 GHz and 5.06 to 1.38 are experimentally performed. The number of passbands can be reconfigured. The proposed multiband MPF has the potential to be employed in modern multi-standard wireless communication systems.

Optical beamforming in microwave photonics is promising for supporting broadband wireless communications. However, the current optical beamforming lacks freedom because of the fixed connection between radio frequency (RF) signal and antenna elements (AEs). This manuscript tackles this challenge by proposing a dynamical optical beamforming architecture that reconfigures the antenna subarray for signal transmission depending on the number of signals to be transmitted. The proposed architecture employs an optical switching and distributing network (SDN) to realize a flexible connection between signals and AEs. An instance of the proposed architecture in photonic integrated circuits, which enables three working modes and transmits four RF signals through sixteen AEs, was presented and numerically simulated. The optical field distribution and beam pattern plots illustrated the operational principle and validated the feasibility of the proposed SDN architecture. Furthermore, the impact of the introduced architecture on the signal amplitude–phase consistency and the comparison of the proposed dynamic architecture and conventional fixe architectures are analyzed and discussed. The results indicate that the proposed architecture exhibits variable beamforming gain with lower hardware complexity.

Castanea henryi is a monoecious woody food tree species whose yield and industrialization potential are limited by its low female-to-male flower ratio. Here, the male flowers on the male inflorescence of C. henryi were converted to female flowers by triple applications of exogenous cytokinin (CK) (N-(2-chloro-4-pyridyl)-N’-phenylurea, CPPU). To study the role of exogenous CK in flower sex determination, cytological and transcriptomic analyses were performed on samples from the five stages after CK treatment. Cytological analysis showed that stage 3 (nine days after the last CK treatment) was the critical stage in the differential development of the pistil primordium and stamen primordium. On this basis, one key module and two modules with significant positive correlations with stage 3 were identified by weighted gene co-expression network analysis (WGCNA), combined with transcriptome data. The CK and GA biosynthesis- and signaling-related genes, three transcription factor (TF) families, and 11 floral organ identity genes were identified in the related modules. In particular, the TFs WRKY47, ERF021, and MYB4, and floral organ identity genes AGL11/15, DEF, and SEP1 with large differences are considered to be critical regulators of sex determination in C. henryi. Based on these results, a genetic regulatory network for exogenous CK in the sex determination of flowers in C. henryi is proposed. This study contributes to the understanding of the role of CK in the sex regulation of flowers and provides new insights into the regulatory network of sex determination in C. henryi.

Summary: Background: Mental symptoms have been shown to be associated with dementia. As the most common neuropsychiatric disorder, it is unclear whether and why anxiety increases the risk of cognitive progression in elderly. Methods: The aim of this study was to investigate the longitudinal effects of anxiety on cognitive impairment in non-dementia elderly and to explore the underlying biological processes using multi-omics including microarray-based transcriptomics, mass spectrometry-based proteomics and metabolomics, cerebrospinal fluid (CSF) biochemical markers, and brain diffusion tensor imaging (DTI). The Alzheimer's Disease Neuroimaging Initiative (ADNI), Chinese Longitudinal Healthy Longevity Survey (CLHLS) and Shanghai Mental Health Centre (SMHC) cohorts were included. Findings: Anxiety was found to increase the risk of subsequent cognitive progression in the ADNI, and a similar result was observed in the CLHLS cohort. Enrichment analysis indicated activated axon/synapse pathways and suppressed mitochondrial pathways in anxiety, the former confirmed by deviations in frontolimbic tract morphology and altered levels of axon/synapse markers, and the latter supported by decreased levels of carnitine metabolites. Mediation analysis revealed that anxiety's effect on the longitudinal cognition was mediated by brain tau burden. Correlations of mitochondria-related expressed genes with axon/synapse proteins, carnitine metabolites, and cognitive changes were found. Interpretation: This study provides cross-validated epidemiological and biological evidence that anxiety is a risk factor for cognitive progression in non-dementia elderly, and that axon/synapse damage in the context of energy metabolism imbalance may contribute to this phenomenon. Hand, foot and mouth disease, caused by enterovirus A71 (EV-A71) infection, is common in children in China. Here, the authors estimate EV-A71 incidence and seroprevalence using data from two longitudinal cohorts and find that, despite high infection rates, a large proportion of children under 6 are susceptible.

Abstract Background A major hand-foot-and-mouth disease (HFMD) pathogen, coxsackievirus A16 (CVA16), has predominated in several of the last 10 years and caused the largest number of HFMD outbreaks between 2011 and 2018 in China. We evaluated the efficacy of maternal anti-CVA16 antibody transfer via the placenta and explored the dynamics of maternal and natural infection-induced neutralizing antibodies in children. Methods Two population-based longitudinal cohorts in southern China were studied during 2013–2018. Participants were enrolled in autumn 2013, including 2475 children aged 1–9 years old and 1066 mother-neonate pairs, and followed for 3 years. Blood/cord samples were collected for CVA16-neutralizing antibody detection. The maternal antibody transfer efficacy, age-specific seroprevalence, geometric mean titre (GMT) and immune response kinetics were estimated. Results The average maternal antibody transfer ratio was 0.88 (95% CI 0.80–0.96). Transferred maternal antibody levels declined rapidly (half-life: 2.0 months, 95% CI 1.9–2.2 months). The GMT decayed below the positive threshold (8) by 1.5 months of age. Due to natural infections, it increased above 8 after 1.4 years and reached 32 by 5 years of age, thereafter dropping slightly. Although the average duration of maternal antibody-mediated protection was < 3 months, the duration extended to 6 months on average for mothers with titres ≥ 64. Conclusions Anti-CVA16 maternal antibodies are efficiently transferred to neonates, but their levels decline quickly. Children aged 0–5 years are the main susceptible population and should be protected by CVA16 vaccination, with the optimal vaccination time between 1.5 months and 1 year of age.

The receptor-binding domain (RBD) of the SARS-CoV-2 spike is a primary target of neutralizing antibodies and a key component of licensed vaccines. Substantial mutations in RBD, however, enable current variants to escape immunogenicity generated by vaccination with the ancestral (WA1) strain. Here, we produce and assess self-assembling nanoparticles displaying RBDs from WA1 and BA.5 strains by using the SpyTag:SpyCatcher system for coupling. We observed both WA1- and BA.5-RBD nanoparticles to degrade substantially after a few days at 37 °C. Incorporation of nine RBD-stabilizing mutations, however, increased yield ~five-fold and stability such that more than 50% of either the WA1- or BA.5-RBD nanoparticle was retained after one week at 37 °C. Murine immunizations revealed that the stabilized RBD-nanoparticles induced ~100-fold higher autologous neutralization titers than the prefusion-stabilized (S2P) spike at a 2 μg dose. Even at a 25-fold lower dose where S2P-induced neutralization titers were below the detection limit, the stabilized BA.5-RBD nanoparticle induced homologous titers of 12,795 ID50 and heterologous titers against WA1 of 1767 ID50. Assessment against a panel of β-coronavirus variants revealed both the stabilized BA.5-RBD nanoparticle and the stabilized WA1-BA.5-(mosaic)-RBD nanoparticle to elicit much higher neutralization breadth than the stabilized WA1-RBD nanoparticle. The extraordinary titer and high neutralization breadth elicited by stabilized RBD nanoparticles from strain BA.5 make them strong candidates for next-generation COVID-19 vaccines.

Membrane fouling is a key factor limiting the application of a membrane bioreactor (MBR), and membrane-surface modification holds the potential to control membrane fouling and solves this problem. In the research, novel nanocomposite membranes were designed and fabricated using antimicrobial copper nanoparticles (CuNPs) coupled with powdered active carbon (PAC) to mitigate membrane fouling. The successful coating was confirmed by SEM, XRD, and FTIR analysis. Compared with a pristine membrane, the functionalization of CuNPs and PAC improved the hydrophilicity of the modified membrane but led to a lower permeate flux. The result of antimicrobial adhesion experiments showed that the modified M-CuOC displayed high antibacterial activity with the bacteria count decreased by 72%. In MBR operation, the modified M-CuOC leads to the removal efficiency of chemical oxygen demand (COD) increasing to 93%, with better filtration performance under a lower TMP rise. The fouling-resistance analysis demonstrated that, although the intrinsic membrane resistance of modified M-CuOC slightly increased, the reversible and irreversible fouling resistances obviously decreased by 45% and 90%. Moreover, the membrane flux recovery efficiency of the modified M-CuOC also increased by 35%. Overall, these results indicated that, in addition to an improvement in antifouling performance, the functionalization of CuNPs and PAC also enhanced the membrane flux recovery efficiency, revealing a good antifouling potential in a practical application.

IntroductionPrevious literatures have mainly explored the impact of the experience of power on impulsive buying, but have ignored the impacts of the expectations of power. The purpose of this research is to delineates a two-facet portrait of power in the role of affecting purchase impulsiveness by proposing a theoretical extension from the experience of power to the expectations of power.MethodsFour laboratory experiments were developed that used ANOVA to verify the hypothesis. A moderated mediation path model was established including the experience of power, product attribute, the expectations of power, deservingness, and purchasing impulsiveness as observed variables.ResultsThe results revealed that powerless consumers are more likely to impulsively buy hedonic products; while powerful consumers prefer to impulsively buy utilitarian products. However, when focusing on the expectations of power, powerless consumers feel a lower perception of deservingness, which reduces their impulse to buy hedonic products. In contrast, when powerful consumers imagine how powerful people should behave in consumption, they will experience a higher sense of deservingness and increase purchasing impulsiveness for hedonic products. The underlying mechanism is that deservingness plays a mediation role in the three-way interaction impacts of the experience of power, product attribute, and the expectations of power on purchasing impulsiveness.ConclusionThe current research formulates a new theoretical perspective on the relationship between power and purchasing impulsiveness. An experience-expectation model of power is presented that proposes consumers’ purchasing impulsiveness can be affected both by the experience of power and the expectations of power.

Alzheimer’s disease (AD) is the most common form of dementia and a significant social and economic burden. Estrogens can exert neuroprotective effects and may contribute to the prevention, attenuation, or even delay in the onset of AD; however, long-term estrogen therapy is associated with harmful side effects. Thus, estrogen alternatives are of interest for countering AD. Naringin, a phytoestrogen, is a key active ingredient in the traditional Chinese medicine Drynaria. Naringin is known to protect against nerve injury induced by amyloid beta-protein (Aβ) 25–35, but the underlying mechanisms of this protection are unclear. To investigate the mechanisms of naringin neuroprotection, we observed the protective effect on Aβ25–35-injured C57BL/6J mice’s learning and memory ability and hippocampal neurons. Then, an Aβ25–35 injury model was established with adrenal phaeochromocytoma (PC12) cells. We examined the effect of naringin treatment on Aβ25–35-injured PC12 cells and its relationship with estrogen receptor (ER), phosphatidylinositol 3-kinase/protein kinase B (PI3K/AKT), and glycogen synthase kinase (GSK)-3β signaling pathways. Estradiol (E2) was used as a positive control for neuroprotection. Naringin treatment resulted in improved learning and memory ability, the morphology of hippocampal neurons, increased cell viability, and reduced apoptosis. We next examined the expression of ERβ, p-AKT (Ser473, Thr308), AKT, p-GSK-3β (Ser9), GSK-3β, p-Tau (Thr231, Ser396), and Tau in PC12 cells treated with Aβ25–35 and either naringin or E2, with and without inhibitors of the ER, PI3K/AKT, and GSK-3β pathways. Our results demonstrated that naringin inhibits Aβ25–35-induced Tau hyperphosphorylation by modulating the ER, PI3K/AKT, and GSK-3β signaling pathways. Furthermore, the neuroprotective effects of naringin were comparable to those of E2 in all treatment groups. Thus, our results have furthered our understanding of naringin’s neuroprotective mechanisms and indicate that naringin may comprise a viable alternative to estrogen therapy.

The oil expulsion efficiency and retention efficiency of shale affect the enrichment and preservation of shale oil. Two series of semi-closed hydrous pyrolysis experiments were performed under in situ geological conditions on a Paleogene shale sample as a comparable analog to evaluate the generation and preservation potential of shale oil in the Funing Formation shale in the Subei Basin. The results show that 1) the oil-generation capacity evolution of different lithological combination source rocks in the Funing Formation of the Subei Basin can be roughly divided into four stages: a) relatively slow oil-generating and slow gas-generating, b) relatively fast oil-generating and slow gas-generating, c) oil cracking into gas, and d) kerogen cracking into gas; 2) different lithological combinations have different hydrocarbon generation, expulsion, and retention efficiencies. The total oil generation rate and gas generation rate of pure shale are higher than those of shale with a silty interlayer, and the exchange point between the oil expulsion rate and retention rate of pure shale is earlier than that of shale with the silty interlayer, which indicates that the pure shale experienced the expulsion and retention process earlier. Oil retention mainly occurs at an EqVRo of 0.84%–1.12%, while oil is mainly discharged to the adjacent siltstone at an EqVRo of 1.12%–1.28%. Based on the simulation under geological conditions, it is recognized that for shale oil exploration in the Subei Basin, the favorable thermal maturity is at an EqVRo of 0.84%–1.12%, and the favorable lithology is the shale with the silty interlayer. On one hand, the siltstone interlayer can provide pore space for the early generated oil, and the concentration difference of hydrocarbons between the shale and the interlayer can be formed so that the generated shale can continuously enter the interlayer. On the other hand, the shale above the interlayer can be used as a cap rock to preserve shale oil. The favorable area for shale oil exploration in the Subei Basin is the area with relatively high maturity (at a VR value of about 1.1%)

Abstract Background The duration of antibodies against SARS-CoV-2 in Covid-19 patients remains uncertain. Longitudinal serological studies are needed to prevent disease and transmission of the virus. Methods In 2020, 414 blood samples were tested, obtained from 157 confirmed Covid-19 patients, in a prospective cohort study in Shanghai. Results The seropositive rate of IgM peaked at 40.5% (17/42) within 1 month after illness onset and then declined. Breakthrough of the exploration of deep shale gas in the Sichuan Basin, Southwest China, has been achieved for decades, there are still some controversies about the adsorption performance and occurrence state of shale gas in deep strata. Isothermal adsorption experiments were carried out under high temperature and high pressure with typical shale samples from deep stratum of the Lower Silurian Longmaxi Formation in Fuling and Dingshan areas in the southeastern Sichuan Basin. Results show that there is a big difference between absolute adsorption and excess adsorption, thus, it is proposed that the excess adsorption capacity should be corrected to the absolute adsorption capacity when evaluating the adsorption performance of deep shale gas. After correction, the variation of absolute shale adsorption with pressure appeared to have three stages including "rapid rise", "slow rise" and "steady rise". The absolute adsorption capacity of deep shale samples from Fuling and Dingshan areas is higher than 2.0 m3/t at 110℃ and 70 MPa, and combined with the adsorption and free gas ratios of deep shale, the total gas content is more than 4.0 m3/t, indicating that deep shale may still have good adsorption performance under high temperature and high pressure. The evaluation of deep shale adsorption capacity can provide reliable parameters for the study of shale gas content and the evaluation of resource potential and reserve for shale gas. Considering the difference of ratio of free gas to adsorbed gas between deep shale and shallow-medium shale, different exploration and production methods can be adopted to effectively improve the production capacity and production cycle of shale gas.

Impedance/admittance models (IM/AMs) have been widely used to analyze the small-signal stability of grid-tied power electronic devices, such as the voltage source converter (VSC). They can be either derived from theoretical analysis under white-box conditions, where all parameters and control structures are fully known, or measured based on experiments under black-box conditions, where the topology and parameters of the controllers are completely unknown. As the IM/AMs depend on specific operating conditions, it is highly desirable to develop fast algorithms for IM/AM prediction (or estimation) under the black-box and variable-operating-point conditions. This article extends the nearly-decoupled AM method for sequence AMs proposed recently by Liu et al to fit any unknown control structure, including not only grid-following VSC, but also grid-forming VSC. It is, therefore, referred to as the fully-decoupled IM (FDIM) method. Furthermore, a curve fitting method for the transfer function is proposed to expedite the algorithm, based on the information of a few disturbance frequencies only. Finally, the algorithm is verified by wide simulations and experiments under different situations, including the direct-drive wind turbine generator. The whole approach is expected to be broadly applicable to the stability analysis of power-electronic-based power systems under variable operating conditions.

The relationship between the type and intensities of lipids of blood and pancreas and the pathological changes in the pancreas during severe acute pancreatitis (SAP) remains unclear. In our study, we employed a rat model of SAP induced through intraperitoneal ornithine injections. We collected serum and pancreas samples at various time points (0–144 h) for histopathological and biochemical assessments, followed by lipidomic analyses using LC-MS/MS or in situ mass spectrometry imaging (MSI) To discern changes over time or at specific points, we employed time-course and univariate analyses for lipid screening, respectively. Our findings indicated that the peak inflammation in the Orn-SAP model occurred within the 24–30 h timeframe, with evident necrosis emerging from 24 h onwards, followed by regeneration starting at 48 h. Time-course analysis revealed an overall decrease in glycerophospholipids (PEs, PCs, LPEs, LPCs), while CEs exhibited an increase within the pancreas. Univariate analysis unveiled a significant reduction in serum TAGs containing 46–51 carbon atoms at 24 h, and CERs in the pancreas significantly increased at 30 h, compared with 0 h. Moreover, a substantial rise in TAGs containing 56–58 carbon atoms was observed at 144 h, both in serum and pancreas. MSI demonstrated the CERs containing saturated mono-acyl chains of 16 and 18 carbon atoms influenced pancreatic regeneration. Tracing the origin of FFAs hydrolyzed from pancreatic glycerophospholipids and serum TAGs during the early stages of inflammation, as well as FFAs utilized for CEs and CERs synthesis during the repair phase, may yield valuable strategies for diagnosing and managing SAP.

New vaccine delivery technologies, such as mRNA, have played a critical role in the rapid and efficient control of SARS-CoV-2, helping to end the COVID-19 pandemic. Enveloped virus-like particles (eVLPs) are often more immunogenic than protein subunit immunogens and could be an effective vaccine platform. Here, we investigated whether the genetic delivery of eVLPs could achieve strong immune responses in mice as previously reported with the immunization of in vitro purified eVLPs. We utilized Newcastle disease virus-like particles (NDVLPs) to display SARS-CoV-2 prefusion-stabilized spikes from the WA-1 or Beta variant (S-2P or S-2Pᵦ, respectively) and evaluated neutralizing murine immune responses achieved by a single-gene-transcript DNA construct for the WA-1 or Beta variant (which we named S-2P-NDVLP-1T and S-2Pᵦ-NDVLP-1T, respectively), by multiple-gene-transcript DNA constructs for the Beta variant (S-2Pᵦ-NDVLP-3T), and by a protein subunit–DNA construct for the WA-1 or Beta variant (S-2P-TM or S-2Pᵦ-TM, respectively). The genetic delivery of S-2P-NDVLP-1T or S-2Pᵦ-NDVLP-1T yielded modest neutralizing responses after a single immunization and high neutralizing responses after a second immunization, comparable to previously reported results in mice immunized with in vitro purified S-2P-NDVLPs. Notably, genetic delivery of S-2Pᵦ-NDVLP-3T yielded significantly higher neutralizing responses in mice after a second immunization than S-2Pᵦ-NDVLP-1T or S-2Pᵦ-TM. Genetic delivery also elicited high spike-specific T-cell responses. Collectively, these results indicate that genetic delivery can provide an effective means to immunize eVLPs and that a multiple-gene transcript eVLP platform may be especially efficacious and inform the design of improved vaccines.

Zygotes without a pronuclear (0PN) or with one pronuclear (1PN) were defined as abnormal fertilization in conventional in vitro fertilization (IVF). The removal of 0PN and 1PN zygotes from conventional IVF cycles has always been controversial. This study aimed to investigate the chromosomal aneuploidy rates of 0PN- and 1PN-derived blastocysts in conventional IVF cycles and to assess the concordance rate between TE-biopsy PGT-A and miPGT-A. TE biopsies and culture media with blastocoel fluid (CM-BF) samples were whole-genome amplified by multiple annealing and looping-based amplification cycle-based single-cell ChromInst method. Next generation sequencing was performed for comprehensive chromosomal screening on a NextSeq550 sequencer using the NextSeq 500/550 High Output kit v2. The aneuploidy rates of 0PN-derived blastocysts were 19.7% for TE-biopsy PGT-A, and 36.1% for miPGT-A; the concordance rate for ploidy was 77.0%; and the sensitivity and specificity were 83.3% and 75.5%, respectively. The aneuploidy rates of 1PN-derived blastocysts were 37.5% and 37.5% by TE-biopsy PGT-A and miPGT-A, respectively; the concordance rate between TE biopsies and CM-BF samples was 83.3%; and the sensitivity and specificity were 77.8% and 86.7%, respectively. Regarding TE-biopsy PGT-A, there were no significant differences in aneuploidy rates among 0PN-, 1PN- and 2PN-derived blastocysts (PGT-M cycles) (19.7% vs. 37.5% vs. 24.3%, P = 0.226), but the aneuploidy rate of 1PN-derived blastocysts was slightly higher than the other two groups. An increase in aneuploidy rates was observed for 0PN/1PN-derived day 6 blastocysts compared to 0PN/1PN-derived day 5 blastocysts (TE-biopsy PGT-A: 35.7% vs. 19.3%, P = 0.099; miPGT-A: 39.3% vs. 35.1%, P = 0.705). The present study is the first that contributes to understanding the chromosomal aneuploidies in 0PN- and 1PN-derived blastocysts in conventional IVF cycles using TE-biopsy PGT-A and miPGT-A. The clinical application value of 0PN- and 1PN-derived blastocysts in conventional IVF should be assessed using TE-biopsy PGT-A or miPGT-A due to the existence of chromosomal aneuploidies.. In terms of consistency, the miPGT-A using blastocoel fluid enriched culture medium is promising as an alternative to TE-biopsy PGT-A for aneuploidy testing of 0PN- or 1PN-derived blastocysts in conventional IVF.

The formation of the Songliao Basin was affected by multidirectional tectonic stress and various peripheral structural belts, and has a complicated geological structure and basin evolution process. The Lishu Fault Depression in the southern Songliao Basin has the characteristics of superimposed and complex evolution of multiple tectonic activities, and is a good window to reveal the impacts of the peripheral structural belts and basin tectonic-sedimentary response. Whether strike-slip tectonic activity controls the sedimentary process of the basin is an unsolved problem. Using three-dimensional seismic data to carry out detailed interpretation, combined with fault distance statistics, structural model analysis and other research methods, the extension activities of the Sangshutai and Qinjiatun faults in the Lishu Fault Depression during the Early Cretaceous were studied. The evolution process of the Sangshutai fault was controlled by the nearly east-west extensional tectonic stress. The initial scale of the fault was large, so the Sangshutai fault was less affected by the adjustment of the extension direction. The Qinjiatun fault was controlled by the NEE-SWW extension during the Early Cretaceous Huoshiling period, and by the NE-SW extension during the Early Cretaceous Shahezi period. The regional strike-slip activity in the Early Cretaceous extension in the Lishu Fault Depression caused a change in the extension direction.

Multi-timescale interaction of power electronics devices, including voltage source converter (VSC), has made the stability and analysis of high penetrating renewable power systems very complicated. In this paper, the impedance model is used to analyze the multi-timescale characteristics and interaction of the VSC. Firstly, the multi-timescale impedance characteristics of VSC are investigated based on the Bode plots. It is found that the slow-timescale (within the DC-link voltage control scale) and fast-timescale (within the AC current control scale) models are separately consistent with the full-order model perfectly within their low- and high-frequency ranges. In addition, there exists a high impedance peak within the intermediate frequency range (roughly from 10 Hz to 100 Hz). Then, the impedance peak is theoretically estimated and explained by the slow-fast-scale impedance parallel resonance through transfer-function diagram analysis. Moreover, it is found that the impedance peak is more related to some outer controllers, such as the alternative voltage control and active power control. Specifically, larger proportional coefficients can greatly suppress the resonance peak. Finally, simulations and experiments are conducted to verify the generality of the multi-timescale characteristics and interaction of the VSC. Hence these findings are not only significant to provide a physical insight into the inner key structure of the impedance of VSC, but also expected to be helpful for controller and parameter design of the VSC.

The coronavirus disease (COVID-19) pandemic is a major challenge worldwide. However, the epidemic potential of common human coronaviruses (HCoVs) remains unclear. This study aimed to determine the epidemiological and co-infection characteristics of common HCoVs in individuals with influenza-like illness (ILI) and severe acute respiratory infection (SARI). This retrospective, observational, multicentre study used data collected from patients admitted to nine sentinel hospitals with ILI and SARI from January 2015 through December 2020 in Shanghai, China. We prospectively tested patients for a total of 22 respiratory pathogens using multi-real-time polymerase chain reaction. Of the 4541 patients tested, 40.37% (1833/4541) tested positive for respiratory pathogens and 3.59% (163/4541) tested positive for common HCoVs. HCoV infection was more common in the non-endemic season for respiratory pathogens (odds ratio: 2.33, 95% confidence interval: 1.64–3.31). HCoV-OC43 (41.72%, 68/163) was the most common type of HCoV detected. The co-infection rate was 31.29% (51/163) among 163 HCoV-positive cases, with HCoV-229E (53.13%, 17/32), the HCoV type that was most frequently associated with co-infection. Respiratory pathogens responsible for co-infections with HCoVs included parainfluenza virus, rhinovirus/enterovirus, influenza A virus, and adenovirus. Furthermore, we identified one patient co-infected with HCoV-OC43 and HCoV-NL63/HKU1. The prevalence of common HCoVs remains low in ILI/SARI cases, in Shanghai. However, the seasonal pattern of HCoVs may be opposite to that of other respiratory pathogens. Moreover, HCoVs are likely to co-exist with specific respiratory pathogens. The potential role of co-infections with HCoVs and other pathogenic microorganisms in infection and pathogenesis of ILI and SARI warrants further study.

A real-time eye diagram monitoring method for optical signals is proposed and experimentally demonstrated based on optical sampling. In the system, the optical signals under test are directly sampled by an optical sampling pulse train with a narrow pulse width and a high repetition frequency. The sampling pulse train is achieved in a Mach-Zehnder modulator (MZM), gated on-off by an electrical pulse. The sampled optical signals are then broadened and detected by a photodetector (PD). A low-speed electrical analog-to-digital converter (ADC) will then quantify the detected electrical signals. Combining with an algorithm based on the infinitesimal calculus, the quantified data is then used to achieve the eye diagram, according to which more time-domain parameters, such as period, time jitter, Q value, and bit error rate (BER) for the optical signals under test, are obtained. Thanks to the high repetition rate of the optical sampling pulse train, the eye diagram and the time-domain parameters of the optical signals are observed in real time. Experimental results show that a real time of about 350-μs eye diagram monitoring for a 2.5-Gb/s optical signal with a dynamic range from −10 to −22 dBm is achieved. In addition, time jitters are measured to range from 4.3 to 49.8 ps. Q values are estimated to range from 20.4 to 4.3, corresponding to BERs ranging from 2.3 × 10−92 to 8.5 × 10−6. The results are also verified by a commercial real-time oscilloscope.

A novel multi-beam photonic receiver with a variable beam count has been proposed. The reconfigurable characteristic of the system is enabled with the variable splitters and optical switch arrays. According to the number of beams it is due to receive, the splitter with suitable ports is chosen to split by controlling the optical switch in order to configure the optical path. After splitting, the multi-wavelength optical carriers are sent to the wavelength division multiplexer to combine and complete the multi-beamforming process. Two structures have been designed to realize the reconfigurable function, while the optimized structure is proved by using several benchmarks for comparison. The operation step of the reconfigurable multi-beam receiver is then given. The first simulations validate that the proposed architecture can dynamically receive the multi-beam with a variable number by reconfiguring the state of the optical switch. The second simulation of the influence on the received beam pattern was performed under a different amplitude and phase inconsistency random values. The results show that the amplitude and phase inconsistency will decline the quality of the multi-beam. When the amplitude and phase inconsistency are within limits, the difference in the optical path within the outputs must be balanced and compensated during the proposed system’s fabrication and deployment.

BackgroundLate-life depression (LLD) and amnestic mild cognitive impairment (aMCI) are two different diseases associated with a high risk of developing Alzheimer’s disease (AD). Both diseases are accompanied by dysregulation of inflammation. However, the differences and similarities of peripheral inflammatory parameters in these two diseases are not well understood.MethodsWe used Luminex assays to measure 29 cytokines simultaneously in the plasma of two large cohorts of subjects at high risk for AD (23 LLD and 23 aMCI) and 23 normal controls (NCs) in the community. Demographics and lifestyle factors were also collected. Cognitive function was evaluated with the Chinese versions of the Montreal Cognitive Assessment (C-MoCA) and neuropsychological test battery (NTB).ResultsWe observed a remarkably increased level of IL-6 in the plasma and reduced levels of chemokines (CXCL11 and CCL13) in the LLD group compared with the aMCI group. The LLD group also showed lower levels of CXCL16 than the NC group. Furthermore, altered cytokine levels were associated with abnormal results in neuropsychological testing and Geriatric Depression Scale scores in both the LLD and aMCI groups. Notably, combinations of cytokines (IL-6 and CCL13) and two subitems of C-MoCA (orientation and short-term memory) generated the best area under the receiver operating characteristic curve (AUROC = 0.974).ConclusionA novel model based on proinflammatory cytokines and brief screening tests performs with fair accuracy in the discrimination between LLD and aMCI. These findings will give clues to provide new therapeutic targets for interventions or markers for two diseases with similar predementia syndromes.

Background: Late-onset Alzheimer’s disease (LOAD) and early-onset Alzheimer’s disease (EOAD) are different subtypes of AD. This study aimed to build and validate radiomics models of the hippocampus for EOAD and young controls (YCs), LOAD and old controls (OCs), as well as EOAD and LOAD.Methods: Thirty-six EOAD patients, 36 LOAD patients, 36 YCs, and 36 OCs from the Alzheimer’s Disease Neuroimaging Initiative (ADNI) database were enrolled and allocated to training and test sets of the EOAD-YC groups, LOAD-OC groups, and EOAD-LOAD groups. Independent external validation sets including 15 EOAD patients, 15 LOAD patients, 15 YCs, and 15 OCs from Shanghai Mental Health Center were constructed, respectively. Bilateral hippocampal segmentation and feature extraction were performed for each subject, and the least absolute shrinkage and selection operator (LASSO) method was used to select radiomic features. Support vector machine (SVM) models were constructed based on the identified features to distinguish EOAD from YC subjects, LOAD from OC subjects, and EOAD from LOAD subjects. The areas under the receiver operating characteristic curves (AUCs) were used to evaluate the performance of the models.Results: Three, three, and four features were selected for EOAD and YC subjects, LOAD and OC subjects, and EOAD and LOAD subjects, respectively. The AUC and accuracy of the SVM model were 0.90 and 0.77 in the test set and 0.91 and 0.87 in the validation set for EOAD and YC subjects, respectively; for LOAD and OC subjects, the AUC and accuracy were 0.94 and 0.86 in the test set and 0.92 and 0.78 in the validation set, respectively. For the SVM model of EOAD and LOAD subjects, the AUC was 0.87 and the accuracy was 0.79 in the test set; additionally, the AUC was 0.86 and the accuracy was 0.77 in the validation set.Conclusion: The findings of this study provide insights into the potential of hippocampal radiomic features as biomarkers to diagnose EOAD and LOAD. This study is the first to show that SVM classification analysis based on hippocampal radiomic features is a valuable method for clinical applications in EOAD.

We have engineered a Human Immune System (HIS)-reconstituted mouse strain (DRAGA mouse: HLA-A2. HLA-DR4. Rag1 KO. IL-2Rγc KO. NOD) in which the murine immune system has been replaced by a long-term, functional HIS via infusion of CD34+ hematopoietic stem cells (HSC) from cord blood. Herein, we report that the DRAGA mice can sustain inducible and transmissible H1N1 and H3N2 influenza A viral (IAV) infections. DRAGA female mice were significantly more resilient than the males to the H3N2/Aichi infection, but not to H3N2/Hong Kong, H3N2/Victoria, or H1N1/PR8 sub-lethal infections. Consistently associated with large pulmonary hemorrhagic areas, both human and murine Factor 8 mRNA transcripts were undetectable in the damaged lung tissues but not in livers of DRAGA mice advancing to severe H1N1/PR8 infection. Infected DRAGA mice mounted a neutralizing anti-viral antibody response and developed lung-resident CD103 T cells. These results indicate that the DRAGA mouse model for IAV infections can more closely approximate the human lung pathology and anti-viral immune responses compared to non-HIS mice. This mouse model may also allow further investigations into gender-based resilience to IAV infections, and may potentially be used to evaluate the efficacy of IAV vaccine regimens for humans.

Strawberry growers face rising production costs combined with competition from foreign imports. Relay cropping vegetables with strawberries is a unique approach that can diversify income and reduce the risk associated with strawberry production. Planting vegetable transplants on the same bed before strawberry crop termination enables continued berry harvesting while the new vegetable transplants become established. Relay cropping techniques of strawberry with eggplants were evaluated during the 2016–17 and 2017–18 seasons in Balm, FL. The strawberry crop was planted in September, and eggplant was transplanted into the beds either as a sole crop or with strawberry plants. Two experiments were conducted to optimize the planting date of strawberries and the termination date of strawberries. The objective of the research was to examine the competitive relationship between strawberry and eggplant crops and to define the optimal planting date for the eggplant and termination date of strawberries to minimize the competitive interaction and maximize the yield of both crops. Strawberry yields were unaffected (P = 0.938) by relay cropping or by the planting date of the eggplant. Eggplants grown without strawberries had 27% to 32% higher yields (P = 0.004) compared with relay-cropped eggplants, and eggplant yield decreased with later planting dates (P < 0.001). A partial budget analysis showed that transplant dates of 4 and 18 Jan. for eggplants with strawberries resulted in increased profits of $7320 and $3461 per ha, respectively, over the baseline treatment of strawberries alone, but later planting dates resulted in an overall economic loss ($7800–$16,000/ha). Strawberry termination dates did not affect eggplant yields. In conclusion, relay cropping eggplants with strawberries resulted in no effect on strawberry yields, reduced eggplant yields, but increased overall profits when eggplant were transplanted in early to mid-January. Relay cropping of strawberries with eggplants in early February to early March is less profitable than a monocrop of strawberries.

The normal development of anthers and the formation of functional pollen are the prerequisites for successful pollination and fertilization. In this study, we observed dynamic changes in inflorescence and anther development in the chinquapin (Castanea henryi) using stereomicroscopy, light microscopy, and transmission electron microscopy. We found that cytokinesis during meiosis in microsporocytes was of the simultaneous type, and that the tetrads were mainly tetrahedral. Mature pollen grains contained two cells with three germ pores. The anther wall was of the basic type and composed of epidermis, endothecium, middle layers, and tapetum. Mature anthers had no middle layer and tapetum. The tapetum was of the glandular type. At the early microspore stage, a large number of starch granules appeared in the endothecium, which was deformed at the late microspore stage. Lipid droplets appeared in tapetum during the early microspore stage, and a few lipid droplets were still found during tapetum degeneration. The mature pollen accumulated a large amount of starch and lipids. These findings demonstrated that the anther wall provides nutrients and protection for pollen development. There is relatively stable correspondence between the external morphological characteristics of male flowers and internal structure of anther development.

The shale, interlayers and adjacent carbonate rocks and siltstones in the second and fourth members of the Paleogene Funing Formation in the Subei Basin were studied using multi-temperature pyrolysis and NMR. The oil-bearing capacity of shale, interlayers and adjacent tight reservoirs in the Funing Formation and the movability of tight oil were evaluated in combination with with shale oil production data. In the second and fourth members of the Funing Formation, the content of tight oil is generally less than 5.0 mg/g in shale. Adsorption-miscible oil is dominant, and the ratio of which to total retained oil decreases with the increase of burial depth. Light free oil generally has a low content, and the actual movable oil ratio is less than 3%. However, the tight oil content is generally greater than 5 mg/g in mud shale interlayers and adjacent sandstones (oily), dominated by free oil. The light free oil content is generally more than 0.25 mg/g, and the actual movable oil ratio is generally between 4% and 7%, which is close to the 5% recovery ratio in the tight sandstone layers before reconstruction in the middle of Bakken Formation in North America. The movable tight oil mainly exists in fractures in the Funing Formation in the Subei Basin. That is, tectonic fractures are the main host of movable shale oil, and some micro pores (with ramp radius greater than 0.18 μm) work as secondary occurrence porosity. Free oil exceeds adsorbed oil. The initial production of tight oil is high in fissured lacustrine carbonates in the first and third members of the Funing Formation, but the production decreases rapidly with a short production cycle, and the economic recoverability is limited. In contrast, the initial production is lower in the tight interlayer sandstones of the source in the second and fourth members of the Funing Formation, which has a slower decline rate, longer production cycle and better economical recoverability.

Aiming to calculate the winding parasitic capacitance for high-frequency high-voltage application, this paper proposes an improved energy method by dividing the electrostatic field into several energy regions. Analytical models of the divided energy regions are established based on the assumed electric field distribution. The stored electrostatic energy in the specified regions covers not only the energy between adjacent turns but also the non-adjacent turns, and a compensation factor is introduced to correct the layer misalignment problem of the standard winding, which makes the analytical energy fairly accurate. Following closely, a calculation procedure of parasitic capacitance is demonstrated. In the end, the accuracy and effectiveness of this method are verified by finite element simulations and measurements.

Background: The Mini-Mental State Examination (MMSE) and Montreal Cognitive Assessment (MoCA) are the most commonly used tools for cognitive impairment screening. The present study aimed to investigate the ability of the MOCA and MMSE to differentiate between cognitively normal elderly individuals, MCI patients and dementia patients at different ages and education levels and to establish the optimal cutoff scores of the MoCA and MMSE for MCI and dementia in the Chinese elderly.Methods: A total of 2,954 Chinese elderly individuals, including 1,746 normal controls, 599 MCI patients and 249 dementia patients, were consecutively recruited in the study. The optimal cutoffs for MoCA and MMSE were determined using receiver operating characteristic (ROC) analysis among the different age and education levels in the three groups. Furthermore, comparison of ROC curves were made to evaluate the performances of the two tests.Results: The area under the curve(AUC) of the MoCA (0.82) for detecting MCI was significantly higher than that of the MMSE (0.75) (P < 0.001). When the sample was divided according to age and education level, the AUC of the MoCA (0.84) was higher than those of the MMSE (0.71) for MCI (P < 0.001) in the younger and more highly-educated groups. The optimal cutoff scores of the MoCA for the groups aged ≤ 75 years old and education ≤ 6 years, aged > 75 years old and education ≤ 6 years, aged ≤ 75 years old and education > 6 years, aged > 75 years old and education > 6 years in screening for MCI were identified as 19.5, 15.5, 24.5 and 24.5, respectively, and the optimal cutoff scores for dementia were 18.5, 10.5, 18.5 and 20.5, respectively. For MMSE in the above four groups, the cutoff scores to detect MCI were 26.5, 22.5, 28.5 and 26.5, respectively, and the optimal cutoff scores for dementia were 23.5, 19.5, 23.5 and 23.5, respectively.Conclusion: Compared to MMSE, the MoCA is more suitable for discriminating MCI in younger and more highly educated elderly Chinese individuals. However, the MMSE has advantage over MoCA in screening MCI in individuals with lower education levels and the older groups of Chinese elderly.

Abstract Impaired visual cognition in residents of hypoxic environment has been widely reported; however, the underlying electrophysiological mechanisms remain unclear. In this study, 23 college students underwent three sessions of a Clock task test before a 30‐day high‐altitude exposure (Test 1) and 1 week (Test 2) and 3 months (Test 3) after they returned to lowlands. The Clock task consists of a visual spatial angle and a visual non‐spatial color discrimination subtask. Simultaneously, electroencephalography (EEG) was recorded during the Clock task. The behavioral results showed that, compared with Test 1, accuracy in Test 2 was significantly decreased in both the Angle and Color tasks, and reaction time (RT) was significantly increased in the Angle task. The event‐related potentials results showed that, during both tasks amplitudes of the occipital N1 and P3 components during both tasks were significantly decreased in Test 2, compared with Test 1. Moreover, N1 amplitude was negatively correlated with RT and positively correlated with accuracy. Further time–frequency EEG analysis showed that theta power at occipital sites was significantly decreased in both tasks in Test 2, compared with Test 1, and was negatively correlated with RT in the Angle task. In Test 3, both the behavioral performance and EEG activity recovered to the baseline level in Test 1. These findings suggested that hypoxia impairs both visual spatial and visual non‐spatial discriminations, and these impairments can recover after subjects return to lowlands. Inhibition of brain electrophysiological activity in the visual cortex may explain the deficits in visual cognition.

Background: Hand, foot, and mouth disease (HFMD) caused by enterovirus A71 (EV-A71) poses a serious threat to children's health. Kinetics of the neutralising antibody (NAb) response in EV-A71 infected HFMD patients remains unclear. The ideal sampling time of paired serum samples for serological diagnosis of EV-A71 infection is not well defined. Methods: HFMD inpatients admitted to Henan Children's Hospital between February 15, 2017 and February 15, 2018 were enrolled. Serial serum samples collected during hospitalisation and up to 1.5 years after discharge were tested for NAb against EV-A71. Random intercept modelling with B-spline was conducted to characterize the kinetics of the EV-A71 NAb response over time after illness onset. Findings: A total of 524 serum samples from 264 EV-A71 RNA positive HFMD inpatients were collected. NAb titres of EV-A71 infected patients were estimated to increase from 40 (95% CI: 9-180) at the day of onset to the peak of 2417 (95% CI: 1859-3143) at day 13, then remained above 1240 until 26 months. For serological diagnosis of EV-A71 infection, if at least a 4-fold rise in titre was used as the criteria, the acute phase serum should be collected at 0-4 days, the corresponding convalescent serum should be collected 14.9 days (95% CI: 9.1-23.8) after illness onset. Interpretation: EV-A71 infection induced a strong and persistent humoral immune response in HFMD patients. The findings provide a scientific support for determining the collection time of paired serum samples for serological diagnosis of EV-A71 infected HFMD patients. Abstract. Background:. There is no absolute consensus for the best time for triggering. The aim of this study was to investigate the effect of different proportion of dominant follicles (PDF) on the human chorionic gonadotropin (HCG) day for the clinical outcomes in patients with polycystic ovary syndrome (PCOS) of different ovarian stimulation protocols. Methods:. A total of 371 cycles of the gonadotropin-releasing hormone (GnRH) agonist long protocol and 347 cycles of GnRH antagonist protocol from January 2014 to December 2016 were included in this retrospective study. Based on the PDF on the day of the HCG administration, the included patients were divided into three groups: Group A (low PDF), PDF 40%. The measurements regarding ovarian stimulation characteristics, fertilization rate, top quality embryo rate, clinical pregnancy rate, and ovarian hyperstimualtion syndrome (OHSS) rate were compared in different PDF groups with different protocols. Results:. In both the GnRH antagonist protocol and GnRH agonist long protocol, the characteristics such as mean age, anti-Mullerian hormone, antral follicle count (AFC), and body mass index were comparable between groups. The number of oocytes retrieved decreased statistically significantly as the PDF and rate of matured oocytes increased. In the GnRH agonist long protocol, the rate of normally fertilized oocytes was highest in Group A (59.74 ± 31.21 vs. 49.70 ± 37.95, 49.67 ± 36.62; F = 3.743, P = 0.025). There were no significant differences in the rate of top-quality embryos and the clinical pregnancy rate between the groups. The clinical pregnancy rate was similar in the three groups (63.6%, 62.5%, 67.5%, respectively, χ2 = 0.989, P = 0.911). The moderate and severe OHSS rate increased statistically significantly when the PDF increased, which was highest in group C (1.4%, 3.1%, 6.7%, respectively, χ2 = 12.014, P = 0.017). In the GnRH antagonist protocol, there were no significant differences in the rate of top-quality embryos, the rate of normally fertilized oocytes, the clinical pregnancy rate, and the moderate and severe OHSS rate between the groups. The clinical pregnancy rate in Group C was higher than that in Group A (57.9% vs. 46.6%, χ2 = 10.850, P = 0.093). Conclusions:. In the GnRH antagonist protocol, PDF on the HCG day of less than 20% may be unfavorable to the clinical pregnancy rate in PCOS. In the GnRH agonist long protocol, delaying the HCG trigger timing has no good effect on clinical pregnancy and the risk of OHSS might increase in patients with PCOS.

REF6 is a H3K27me3 histone demethylase that targets regions of active chromatin in the Arabidopsis genome. Here Qiu et al. show that REF6 preferentially binds unmethylated DNA via its zinc-finger domains suggesting that DNA methylation might deter REF6 binding in heterochromatin.
