Abstract

Background: Exposure to ambient air pollution is associated with a significant number of deaths. Much of the evidence associating air pollution with adverse effects is from North American and Europe, partially due to incomplete data in other regions limiting location specific examinations. The aim of the current paper is to leverage satellite derived air quality data to examine the relationship between ambient particulate matter and all-cause and cause-specific mortality in Asia. Methods: Six cohorts from the Asia Cohort Consortium provided residential information for participants, recruited between 1991 and 2008, across six countries (Bangladesh, India, Iran, Japan, South Korea, and Taiwan). Ambient particulate material (PM2·5) levels for the year of enrolment (or 1998 if enrolled earlier) were assigned utilizing satellite and sensor-based maps. Cox proportional models were used to examine the association between ambient air pollution and all-cause and cause-specific mortality (all cancer, lung cancer, cardiovascular and lung disease). Models were additionally adjusted for urbanicity (representing urban and built characteristics) and stratified by smoking status in secondary analyses. Country-specific findings were pooled via random-effects meta-analysis. Findings: More than 300,000 participants across six cohorts were included, representing more than 4-million-person years. A positive relationship was observed between a 5 µg/m (Dockery et al., 1993) increase in PM2·5 and cardiovascular mortality (HR: 1·06, 95 % CI: 0.99, 1·13). The additional adjustment for urbanicity resulted in increased associations between PM2.5 and mortality outcomes, including all-cause mortality (1·04, 95 % CI: 0·97, 1·11). Results were generally similar regardless of whether one was a current, never, or ex-smoker. Abstract

The family of pyrabactin resistance 1 (PYR1)/PYR1-like (PYL) regulatory components of ABA receptors (RCAR) play a vital role in the initial step of ABA signaling. To understand the expression mode of PYL genes in response to various abiotic stresses in Chinese cabbage (Brassica rapa ssp. pekinensis), the members of BrPYL gene family were first identified with the relevant bioinformatics software. And then, the relative expressions of identified BrPYLs after heat, cold, drought, and salt treatments for 0, 24, 48, and 72 h were determined via real-time quantitative PCR. Here, we identified 24 PYLs in the B. rapa genome. Based on the phylogenetic analysis, these BrPYL genes were divided into three classes and distributed on ten chromosomes in Chinese cabbage. Most of BrPYL genes in the same group have similar gene structures and intron numbers. There were seven genes (BrPYL5, BrPYL8, BrPYL22, BrPYL3, BrPYL18, BrPYL11, and BrPYL21) from Group A with two introns and one gene (BrPYL19) from Group D with one intron. Analysis of conserved motifs suggested that every group contained motif 2 containing the Polyketide_cyc2 domain. Subsequently, the prediction of cis-acting elements indicated that BrPYL genes had 5 stress-related elements and 5 hormone-related elements, among which the number of MYC (dehydration reaction) was the highest, suggesting that BrPYL genes could respond to hormones and abiotic stresses. Expression patterns under four abiotic stresses showed that the expressions of BrPYL4, BrPYL11, BrPYL21, and BrPYL23 responded to these stresses at different time points. To conclude, we identified the BrPYL genes and build the BrPYLs expression mode in response to various abiotic stresses. Abstract

Red tide is a major marine ecological disaster in the coastal area which indicates the change of ecological environment. Bohai Bay is an important platform for the Beijing-Tianjin-Hebei area for regional economic integration, which is an important channel for the marine biology in the Yellow Sea and Bohai Sea. Therefore, in this work, the red tide data from 2002 to 2017 were analyzed to study the spatiotemporal distribution and the changing characteristics of biological groups in this area. In the past 15 years, the occurrence of red tide has been limited from the month of June to August. Spatially, the red tide occurrence was less common in the south than north, and the most frequent red tide event recorded from Tianjin Port channel to the northern area of Jianhe kou. The main red tide biota were dinoflagellates, diatoms and Chrysophytae and the dominant red tide species were Noctiluca scintillans, Skeletonema costatum, Phaeocystis globosa and Karenia mikimotoi. Besides, the red tide caused by toxic algae was increasing year by year. However, the high seawater temperature in summer, flood, anthropogenic nutrient influx, and favorable southwest and southeast monsoon wind positively influenced the red tide events. Abstract

To investigate large structural clonal mosaicism of chromosome X, we analysed the SNP microarray intensity data of 38,303 women from cancer genome-wide association studies (20,878 cases and 17,425 controls) and detected 124 mosaic X events >2 Mb in 97 (0.25%) women. Here we show rates for X-chromosome mosaicism are four times higher than mean autosomal rates; X mosaic events more often include the entire chromosome and participants with X events more likely harbour autosomal mosaic events. X mosaicism frequency increases with age (0.11% in 50-year olds; 0.45% in 75-year olds), as reported for Y and autosomes. Methylation array analyses of 33 women with X mosaicism indicate events preferentially involve the inactive X chromosome. Our results provide further evidence that the sex chromosomes undergo mosaic events more frequently than autosomes, which could have implications for understanding the underlying mechanisms of mosaic events and their possible contribution to risk for chronic diseases.

27. Estimation of Cancer Burden Attributable to Infection in Asia

Abstract

BACKGROUND: Some infectious agents have been shown to be human carcinogens. The current study focused on estimation of cancer burden attributable to infection in different regions of Asia. METHODS: By systematically reviewing previous studies of the infection prevalence data of 13 countries in Asia and relative risks of specific cancers, we calculated the population attributable fraction of carcinogenic infections. Using data from GLOBOCAN 2012, the overall country-specific and gender-specific number of new cancer cases and deaths resulting from infection were estimated. RESULTS: Across 13 principal Asian countries, the average prevalence and range was 6.6% (0.5% in Japanese women to 15.0% in Vietnamese men) for hepatitis B virus (HBV), 2.6% (0.3% in Iran to 5.1% in Saudi Arabia) for hepatitis C virus (HCV), 7.9% (2.8% in Pakistan to 17.7% in China) for human papillomavirus (HPV), and 61.8% (12.8% in Indonesia to 91.7% in Bangladesh) for Helicobacter pylori (HP). The estimated total number of cancer cases and deaths caused by infection in these 13 countries were 1 212 026 (19.6% of all new cancer cases) and 908 549 (22.0% of all deaths from cancer). The fractions of cancer incidence attributable to infection were 19.7% and 19.5% in men and women, respectively. The percentages of cancer deaths attributable to infection were 21.9% and 22.1% in men and women, respectively. Among the main infectious agents, HP was responsible for 31.5% of infection-related cancer cases and 32.8% of infection-related cancer deaths, followed by HBV (28.6% of new cases and 23.8% of deaths), HPV (22.0% of new cases and 27.3% of deaths), and HCV (12.2% of new cases and 10.6% of deaths). CONCLUSIONS: Approximately one quarter of all cancer cases and deaths were infection-associated in Asia, which could be effectively prevented if appropriate long-term controls of infectious agents were applied.

Abstract

In an analysis of 31,717 cancer cases and 26,136 cancer-free controls from 13 genome-wide association studies, we observed large chromosomal abnormalities in a subset of clones in DNA obtained from blood or buccal samples. We observed mosaic abnormalities, either aneuploidy or copy-neutral loss of heterozygosity, of >2 Mb in size in autosomes of 517 individuals (0.89%), with abnormal cell proportions of between 7% and 95%. In cancer-free individuals, frequency increased with age, from 0.23% under 50 years to 1.91% between 75 and 79 years (P = 4.8 × 10(-8)). Mosaic abnormalities were more frequent in individuals with solid tumors (0.97% versus 0.74% in cancer-free individuals; odds ratio (OR) = 1.25; P = 0.016), with stronger association with cases who had DNA collected before diagnosis or treatment (OR = 1.45; P = 0.0005). Detectable mosaicism was also more common in individuals for whom DNA was collected at least 1 year before diagnosis with leukemia compared to cancer-free individuals (OR = 35.4; P = 3.8 × 10(-11)). These findings underscore the time-dependent nature of somatic events in the etiology of cancer and potentially other late-onset diseases.

Abstract

Objective: To analyze the clinicopathological features of invasive lobular carcinoma (ILC) and compare them with invasive ductal carcinoma (IDC), hoping to find the fact of ILC in China and assist the decision makers with proper individualized treatment. Materials and Methods: A nationwide multicenter retrospective study was performed. A total of 4211 primary breast cancer cases were randomly selected from 1999 to 2008 in seven regions of China. ILC cases were compared with IDC by clinicopathological features and molecular subtypes. Results: A total of 135 (3.2%) ILC and 3471 (82.4%) IDC cases were included for analysis. The age, tumor size, menopausal state, family history, nodal status, and stage of ILC were similar to that of IDC. ILC was more likely to be positive for estrogen receptor (65.5% vs. 57.7%) and progesterone receptor (64.7% vs. 58.5%), and less likely to overexpress human epidermal growth factor receptor-2 (17.3% vs. 23.6%). Even though, these differences are not significant, the proportion of luminal A type of ILC is significantly larger than that of IDC (54.8% vs. 42.7%; P < 0.05). Conclusion: ILC has a larger proportion of luminal A type compared with IDC. Larger sample size study for better known of molecular subtypes of ILC is needed in future to individualize the treatment decision. [ABSTRACT FROM AUTHOR]
