3. Gut microorganisms of Locusta migratoria in various life stages and its possible influence on cellulose digestibility

ABSTRACT Locusta migratoria is an important phytophagous pest, and its gut microbial communities play an important role in cellulose degradation. In this study, the gut microbial and cellulose digestibility dynamics of Locusta migratoria were jointly analyzed using high-throughput sequencing and anthrone colorimetry. The results showed that the gut microbial diversity and cellulose digestibility across life stages were dynamically changing. The species richness of gut bacteria was significantly higher in eggs than in larvae and imago, the species richness and cellulose digestibility of gut bacteria were significantly higher in early larvae (first and second instars) than in late larvae (third to fifth instars), and the diversity of gut bacteria and cellulose digestibility were significantly higher in imago than in late larvae. There is a correlation between the dynamics of gut bacterial communities and cellulose digestibility. Enterobacter, Lactococcus, and Pseudomonas are the most abundant genera throughout all life stages. Six strains of highly efficient cellulolytic bacteria were screened, which were dominant gut bacteria. Carboxymethyl cellulase activity (CMCA) and filter paper activity (FPA) experiments revealed that Pseudomonas had the highest cellulase enzyme activity. This study provides a new way for the screening of cellulolytic bacteria and lays the foundation for developing insects with significant biomass into cellulose-degrading bioreactors.IMPORTANCECellulose is the most abundant and cheapest renewable resource in nature, but its degradation is difficult, so finding efficient cellulose degradation methods is an urgent challenge. Locusta migratoria is a large group of agricultural pests, and the large number of microorganisms that inhabit their intestinal tracts play an important role in cellulose degradation. We analyzed the dynamics of Locusta migratoria gut microbial communities and cellulose digestibility using a combination of high-throughput sequencing technology and anthrone colorimetry. The results revealed that the gut microbial diversity and cellulose digestibility were dynamically changed at different life stages. In addition, we explored the intestinal bacterial community of Locusta migratoria across life stages and its correlation with cellulose digestibility. The dominant bacterial genera at different life stages of Locusta migratoria were uncovered and their carboxymethyl cellulase activity (CMCA) and filter paper activity (FPA) were determined. This study provides a new avenue for screening cellulolytic bacteria and lays the foundation for developing insects with significant biomass into cellulose-degrading bioreactors.

Sheep body size can directly reflect the growth rates and fattening rates of sheep and is also an important index for measuring the growth performance of meat sheep.Inner Mongolia Cashmere Goat is a local excellent breed of cashmere and meat dual-purpose, which is a typical heterogeneous indumentum. The hair follicles cycle through periods of vigorous growth (anagen), a regression caused by apoptosis (catagen), and relative rest (telogen). At present, it is not clear which genes affect the cycle transformation of hair follicles and unclear how proteins impact the creation and expansion of hair follicles.we using multi-omics joint analysis methodologies to investigated the possible pathways of transformation and apoptosis in goat hair follicles. The results showed that 917,1,187, and 716 proteins were specifically expressed in anagen, catagen andtelogen. The result of gene ontology (GO) annotation showed that differentially expressed proteins (DEPs) are in different growth cycle periods, and enriched GO items are mostly related to the transformation of cells and proteins. The Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment result indicated that the apoptosis process has a great impact on hair follicle’s growth cycle. The results of the protein interaction network of differential proteins showed that the ribosomal protein family (RPL4, RPL8, RPS16, RPS18, RPS2, RPS27A, RPS3) was the core protein in the network. The results of combined transcriptome and proteomics analysis showed that there were 16,34, and 26 overlapped DEGs and DEPs in the comparison of anagen VS catagen, catagen VS telogen and anagen VS telogen, of which API5 plays an important role in regulating protein and gene expression levels. We focused on API5 and Ribosomal protein and found that API5 affected the apoptosis process of hair follicles, and ribosomal protein was highly expressed in the resting stage of hair follicles. They are both useful as molecular marker candidate genes to study hair follicle growth and apoptosis,and they both have an essential function in the cycle transition process of hair follicles. The results of this study may provide a theoretical basis for further research on the growth and development of hair follicles in Inner Mongolian Cashmere goats.

6. Mathematical Limitations of Gravity Model in Constructing Regional Association Networks: A Case Study

This study evaluates the limitations of gravity models in constructing regional association networks, using China’s interprovincial economic connections as a case study. Comparison between a gravity-model-based simulated network and an actual network reveals significant topological differences. The gravity model overestimates the influence of larger, inward-oriented provinces and fails to accurately represent external connections. Attempts to refine the model with additional variables proved ineffective. Further theoretical analysis attributes these deficiencies to measurement bias from the model’s simplified binary perspective and information loss due to dimensional mismatch between pairwise predictions and complex network structures. These findings underscore the need for cautious application of gravity models and the development of more comprehensive analytical frameworks in regional network analysis.

Abstract Electrocatalytic CO2‐to‐formate conversion is considered an economically viable process. In general, Zn‐based nanomaterials are well‐known to be highly efficient electrocatalysts for the conversion of CO2 to CO, but seldom do they exhibit excellent selectivity toward formate. In this article, we demonstrate that a heterointerface catalyst ZnO/ZnSnO3 with nanosheet morphology shows enhanced selectivity with a maximum Faradaic efficiency (FE) of 86% at −0.9 V versus reversible hydrogen electrode and larger current density for the conversion of CO2 to formate than pristine ZnO and ZnSnO3. In particular, the FEs of the C1 products (CO + HCOO−) exceed 98% over the potential window. The experimental measurements combined with theoretical calculations revealed that the ZnO in ZnO/ZnSnO3 heterojunction delivers the valence electron depletion and accordingly optimizes Zn d‐band center, which results in moderate Zn–O hybridization of HCOO* and weakened Zn–C hybridization of competing COOH*, thus greatly boosting the HCOOH generation. Our study highlights the importance of charge redistribution in catalysts on the selectivity of electrochemical CO2 reduction.

Sheep body size can directly reflect the growth rates and fattening rates of sheep and is also an important index for measuring the growth performance of meat sheep. In this study, high-resolution resequencing data from four sheep breeds (Dorper sheep, Suffolk sheep, Ouessant sheep, and Shetland sheep) were analyzed. The nonsynonymous single nucleotide polymorphisms of three candidate genes (KIAA1217, SNTA1, and LTBP1) were also genotyped in 642 healthy Ujumqin sheep using MALDI-TOFMS and the genotyping results were associated with growth traits. The results showed that different genotypes of the KIAA1217 g.24429511T>C locus had significant effects on the chest circumferences of Ujumqin sheep. The SNTA1 g.62222626C>A locus had different effects on the chest depths, shoulder widths and rump widths of Ujumqin sheep. This study showed that these two sites can be used for marker-assisted selection, which will be beneficial for future precision molecular breeding.

10. Nur77 mitigates endothelial dysfunction through activation of both nitric oxide production and anti-oxidant pathways

Background: Nur77 belongs to the member of orphan nuclear receptor 4A family that plays critical roles in maintaining vascular homeostasis. This study aims to determine whether Nur77 plays a role in attenuating vascular dysfunction, and if so, to determine the molecular mechanisms involved. Methods: Both Nur77 knockout (Nur77 KO) and Nur77 endothelial specific transgenic mice (Nur77-Tg) were employed to examine the functional significance of Nur77 in vascular endothelium in vivo. Endothelium-dependent vasodilatation to acetylcholine (Ach) and reactive oxygen species (ROS) production was determined under inflammatory and high glucose conditions. Expression of genes was determined by real-time PCR and western blot analysis. Results: In response to tumor necrosis factor alpha (TNF-α) treatment and diabetes, the endothelium-dependent vasodilatation to Ach was significantly impaired in aorta from Nur77 KO as compared with those from the wild-type (WT) mice. Endothelial specific overexpression of Nur77 markedly prevented both TNF-α- and high glucose-induced endothelial dysfunction. Compared with WT mice, after TNF-α and high glucose treatment, ROS production in aorta was significantly increased in Nur77 KO mice, but it was inhibited in Nur77-Tg mice, as determined by dihydroethidium (DHE) staining. Furthermore, we demonstrated that Nur77 overexpression substantially increased the expression of several key enzymes involved in nitric oxide (NO) production and ROS scavenging, including endothelial nitric oxide synthase (eNOS), guanosine triphosphate cyclohydrolase 1 (GCH-1), glutathione peroxidase-1 (GPx-1), and superoxide dismutases (SODs). Mechanistically, we found that Nur77 increased GCH1 mRNA stability by inhibiting the expression of microRNA-133a, while Nur77 upregulated SOD1 expression through directly binding to the human SOD1 promoter in vascular endothelial cells. Conclusion: Our results suggest that Nur77 plays an essential role in attenuating endothelial dysfunction through activating NO production and anti-oxidant pathways in vascular endothelium. Targeted activation of Nur77 may provide a novel therapeutic approach for the treatment of cardiovascular diseases associated with endothelial dysfunction.

The ability to recognize the body sizes of sheep is significantly influenced by posture, especially without artificial fixation, leading to more noticeable changes. This study presents a recognition model using the Mask R-CNN convolutional neural network to identify the sides and backs of sheep. The proposed approach includes an algorithm for extracting key frames through mask calculation and specific algorithms for head-down, head-up, and jumping postures of Ujumqin sheep. The study reported an accuracy of 94.70% in posture classification. We measured the body size parameters of Ujumqin sheep of different sexes and in different walking states, including observations of head-down and head-up. The errors for the head-down position of rams, in terms of body slanting length, withers height, hip height, and chest depth, were recorded as 0.08 ± 0.06, 0.09 ± 0.07, 0.07 ± 0.05, and 0.12 ± 0.09, respectively. For rams in the head-up position, the corresponding errors were 0.06 ± 0.05, 0.06 ± 0.05, 0.07 ± 0.05, and 0.13 ± 0.07, respectively. The errors for the head-down position of ewes, in terms of body slanting length, withers height, hip height, and chest depth, were recorded as 0.06 ± 0.05, 0.09 ± 0.08, 0.07 ± 0.06, and 0.13 ± 0.10, respectively. For ewes in the head-up position, the corresponding errors were 0.06 ± 0.05, 0.08 ± 0.06, 0.06 ± 0.04, and 0.16 ± 0.12, respectively. The study observed that sheep walking through a passage exhibited a more curved knee posture compared to normal measurements, often with a lowered head. This research presents a cost-effective data collection scheme for studying multiple postures in animal husbandry.

Abstract Background Cryptocaryon irritans, a common parasite in tropical and subtropical marine teleost fish, has caused serious harm to the marine aquaculture industry. Honokiol was proven to induce C. irritans tomont cytoplasm shrinkage and death in our previous study, but the mechanism by which it works remains unknown. Methods In this study, the changes of apoptotic morphology and apoptotic ratio were detected by microscopic observation and AnnexinV-FITC/PI staining. The effects of honokiol on intracellular calcium ([Ca2+]i) concentration, mitochondrial membrane potential (ΔΨm), reactive oxygen species (ROS), quantity of DNA fragmentations (QDF) and caspase activities were detected by Fluo-3 staining, JC-1 staining, DCFH-DA staining, Tunel method and caspase activity assay kit. The effects of honokiol on mRNA expression levels of 61 apoptosis-related genes in tomonts of C. irritans were detected by real-time PCR. Results The results of the study on the effects of honokiol concentration on C. irritans tomont apoptosis-like death showed that the highest levels of prophase apoptosis-like death rate (PADR), [Ca2+]i concentration, ROS, the activities of caspase-3/9 and the lowest necrosis ratio (NER) were obtained at a concentration of 1 μg/ml, which was considered the most suitable for inducing C. irritans tomont apoptosis-like death. When C. irritans tomonts were treated with 1 μg/ml honokiol, the [Ca2+]i concentration began to increase significantly at 1 h. Following this, the ROS, QDF and activities of caspase-3/9 began to increase significantly, and the ΔΨm began to decrease significantly at 2 h; the highest PADR was obtained at 4 h. The mRNA expression of 14 genes was significantly upregulated during honokiol treatment. Of these genes, itpr2, capn1, mc, actg1, actb, parp2, traf2 and fos were enriched in the pathway related to apoptosis induced by endoplasmic reticulum (ER) stress. Conclusions This article shows that honokiol can induce C. irritans tomont apoptosis-like death. These results suggest that honokiol may disrupt [Ca2+]i homeostasis in ER and then induce C. irritans tomont apoptosis-like death by caspase cascade or mitochondrial pathway, which might represent a novel therapeutic intervention for C. irritans infection. Natural products are a rich resource in drug discovery. In the report, series novel rutaecarpine-derived compounds with potent anti-inflammatory activity and therapeutic efficacy against dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) were designed and synthesized. Thirty-two target compounds inhibiting on LPS-induced NO production in RAW264.7 cells were screened. Compound 7mA with the most potent inhibitory activity and low cytotoxicity was selected as representative compound for further studies on the anti-inflammatory efficacy and mechanism. The results indicated that 7mA could significantly inhibit the transcription or expression of iNOS, IL-6 and IL-1β via regulating the TLR4/MAPK/NF-κB signal pathway. What's more, 7mA also could alleviate UC in DSS-induced mice, and down-regulated the release of IL-6, IL-1β, inhibited activation of TLR4/MAPK/NF-κB pathway in mice model as well, suggesting that this new rutaecarpine-derived scaffold might serve as therapeutic candidate for further development to anti-inflammatory agent.

Summary: The canine transmissible venereal tumor (CTVT) is a clonal cell-mediated cancer with a long evolutionary history and extensive karyotype rearrangements in its genome. However, little is known about its genetic similarity to human tumors. Here, using multi-omics data we identified 11 germline gene fusions (GGFs) in CTVT, which showed higher genetic susceptibility than others. Additionally, we illustrate a mechanism of a complex gene fusion of three gene segments (HSD17B4-DMXL1-TNFAIP8) that we refer to “greedy fusion”. Our findings also provided evidence that expressions of GGFs are downregulated during the tumor regressive phase, which is associated with DNA methylation level. This study presents a comprehensive landscape of gene fusions (GFs) in CTVT, which offers a valuable genetic resource for exploring potential genetic mechanisms underlying the development of cancers in both dogs and humans.

Background Cardiometabolic index (CMI) is a simple index to measure blood lipid, which is closely related to diabetes and stroke. Metabolically obese normal weight (MONW) individuals have higher risks of morbidity and mortality of diabetes and cardiovascular and cerebrovascular diseases. Correctly identifying individuals with MONW phenotype is essential for the prevention and control of metabolism-related diseases. However, there are few studies on the predictive value of CMI for MONW phenotype. Objective To investigate the association between CMI and MONW phenotype, and to evaluate the predictive value of CMI for MONW phenotype. Methods The multistage stratified cluster sampling method was used to select permanent residents aged ≥18 years as subjects from Nanjing. The investigation time was from January 1, 2017 to June 30, 2018. The basic data of subjects were collected and multivariate robust Poisson regression model was used to evaluate the RR value with 95%CI of CMI for MONW phenotype. The receiver operating characteristic (ROC) curve was used to evaluate the predictive ability of CMI, waist-to-height ratio (WHtR) , triglyceride/ high-density lipoprotein cholesterol (TG/HDL-C) ratio, waist circumference (WC) and body mass index (BMI) for MONW phenotype. DeLong test was used to compare the area under the ROC curve (AUC) of the above-mentioned five indicators, and to further explore the value of CMI in predicting MONW phenotype in different gender and age groups. Results A total of 30 408 people were included, including 13 213 males and 17 195 females, 23 691 cases of MHNW and 6 717 cases of MONW. There was statistically significant difference in age, education, occupation, current smoking, alcohol consumption, physical activity, duration of static behavior, high red meat intake, disease history, medication history, height, waist circumference (WC) , body mass index (BMI) , total cholesterol (TC) , triglyceride (TG) , high density lipoprotein cholesterol (HDL-C) , low density lipoprotein cholesterol (LDL-C) , systolic blood pressure (SBP) , diastolic blood pressure (DBP) , fasting blood glucose (FPG) , TG/HDL-C, waist-height ratio (WHtR) and CMI (P

17. Calculation of Characteristic Point Parameters for Restoring Model of Corroded Short-Pier RC Shear Walls

Based on the quasi-static tests of 12 corroded RC (reinforced concrete) shear walls, it was found that reinforcement corrosion has a great influence on the skeleton curve of RC shear walls. With an increase in the degree of corrosion, the bearing capacity of specimens decreases, and the deformation capacity worsens. Increasing the diameter of longitudinal reinforcements can significantly improve the bearing capacity of corroded RC shear walls, while the deformation capacity of corroded specimens can be improved by increasing the lateral distributed reinforcement or the transverse reinforcement in the embedded column. In order to accurately evaluate the seismic performance of corroded RC shear walls, we considered descent segments of four broken-line models to estimate the skeleton curve. After considering the influence of corrosion on the parameters of the characteristic point for the skeleton curve, the calculation formulas of the characteristic point parameters of the skeleton curve for the corroded RC shear wall were determined based on the test data fitting. It was proven that the formula for the characteristic point parameters for the skeleton curve of corroded RC shear walls has good applicability. This study lays a theoretical foundation for the seismic performance evaluation of an RC shear wall structure in a salt fog environment. It provides a theoretical basis for further improving the life-cycle seismic capacity evaluation system for RC structures.

Inner Mongolia Cashmere Goat fleece is derived from a heterogeneous coat. The guard hair is produced by primary hair follicles, and the down hair is produced by secondary hair follicles. Reports have focused on research related to the different hair follicle types, but no related study has addressed the difference in the proteins of down hair (DH) and guard hair (GH), and whether the protein composition of these materials differs remains to be studied. The protein composition of DH and GH was studied. A total of 108 proteins were identified in DH, and 116 proteins were identified in GH; 39 proteins were differentially expressed, of which 18 proteins were upregulated and 21 proteins were downregulated in DH versus GH. In this study, through the measurement of DH and GH samples from Inner Mongolia Cashmere Goats, the diameter of DH relative to GH was downregulated (P < 0.05), a property that may be related to the KRT38 protein. Moreover, the strength of GH was significantly higher than that of DH (P < 0.05), a property that may be affected by the KRTAP8-1 protein. GO analysis showed that the different traits of DH and GH from Inner Mongolia Cashmere Goats are affected by keratin, actin, and calcium-binding proteins. This study uses a nonlabelled quantitative proteomics method to study the proteins in DH and GH, aiming to identify the keratin family in Inner Mongolia Cashmere Goats and to provide a new direction for studying DH and GH traits at the protein level.

Abstract Background The current study set out to identify the miRNA-mRNA regulatory networks that influence the radiosensitivity in esophageal cancer based on the The Cancer Genome Atlas (TCGA) and the Gene Expression Omnibus (GEO) databases. Methods Firstly, esophageal cancer-related miRNA-seq and mRNA-seq data were retrieved from the TCGA database, and the mRNA dataset of esophageal cancer radiotherapy was downloaded from the GEO database to analyze the differential expressed miRNAs (DEmiRNAs) and mRNAs (DEmRNAs) in radiosensitive and radioresistant samples, followed by the construction of the miRNA-mRNA regulatory network and Gene Ontology and KEGG enrichment analysis. Additionally, a prognostic risk model was constructed, and its accuracy was evaluated by means of receiver operating characteristic analysis. Results A total of 125 DEmiRNAs and 42 DEmRNAs were closely related to the radiosensitivity in patients with esophageal cancer. Based on 47 miRNA-mRNA interactions, including 21 miRNAs and 21 mRNAs, the miRNA-mRNA regulatory network was constructed. The prognostic risk model based on 2 miRNAs (miR-132-3p and miR-576-5p) and 4 mRNAs (CAND1, ZDHHC23, AHR, and MTMR4) could accurately predict the prognosis of esophageal cancer patients. Finally, it was verified that miR-132-3p/CAND1/ZDHHC23 and miR-576-5p/AHR could affect the radiosensitivity in esophageal cancer. Conclusion Our study demonstrated that miR-132-3p/CAND1/ZDHHC23 and miR-576-5p/AHR were critical molecular pathways related to the radiosensitivity of esophageal cancer.

Abstract Background The amino acid transporter SLC6A14, which transports 18 of the 20 proteinogenic amino acids, is too low to be detected in healthy normal tissues but is significantly increased in some solid cancers. However, little is known about the roles of SLC6A14 in colorectal cancer (CRC). Methods The mRNA and protein levels of SLC6A14 were detected using TCGA database, real-time polymerase chain reaction, western blot, and tissue microarrays, respectively. Amino acids concentration was determined by LC-MS/MS. Cell proliferation and apoptosis were determined using MTT assay and flow cytometry. Transwell invasion assay and wound healing assay were employed to analyze cell migration and invasion. The protein levels of Akt-mTOR signaling pathway and MMPs proteins were detected by western blot. Results Both of the mRNA and protein levels of SLC6A14 were upregulated in CRC tissues, and the protein levels of SLC6A14 were closely related to the tumor cells differentiation: the higher the expression of SLC6A14 was, the poorer the differentiation of the tumor cells was. Further knockdown SLC6A14 with siRNA or treatment with α-MT in CRC cell lines reduced cell proliferation and migration in vitro and inhibited xenograft tumor growth in vivo. Mechanistically, SLC6A14 was demonstrated to regulate the expression and phosphorylation of Akt-mTOR, which mediates the promoting tumor growth function of SLC6A14. Blockade of SLC6A14 with α-MT inhibited the activation of mTOR signaling. Conclusion SLC6A14 was upregulated in CRC and could promote tumor progression by activating the Akt-mTOR signaling pathway, which may serve as an effective molecular target for the treatment of CRC.

The Kuanchuanpu biota (ca. 535 Ma) from the basal Cambrian in South China yields various phosphatized, three-dimensionally preserved microscopic fossils, such as animal embryos, cyanobacteria, and algae. This provides an exceptional window for investigating the rapid divergent evolution of animal phyla in the early Cambrian ocean. However, the fossil eukaryotic algae are poorly documented and are not properly incorporated into the conceptual food web of the Cambrian ocean ecosystem. In this study, we report a new taxon of calathiform alga, Calathophycus irregulatus Tang gen. et sp. nov., and an indeterminate alga, from the Kuanchuanpu biota. Calathophycus irregulatus can be distinguished from other reported phosphatized multicellular algal fossils from the Kuanchuanpu biota and the Ediacaran Weng’an biota (Guizhou Province, China), based on the external morphology and features of cell clump aggregation. The observation of C. irregulatus and the indeterminate alga indicates a certain diversity of algae at the beginning of the Cambrian. Concerning the regularly or irregularly shaped multicellular algal fossils from the Kuanchuanpu Formation, co-occurring spherical fossils, previously interpreted as “cleavage stage” of animal embryos, should be reconsidered in affinity.

The theory about the brittle failures of multiple locked patches in a seismogenic fault system developed by us since 2010 is introduced in the present study. It is stated by the theory that the progressive failures of locked patch result in the occurrence of earthquakes due to fault movement, where the major earthquakes occurred at its volume expansion and peak strength points are referred to as characteristic ones. We analyze the seismogenic law of characteristic earthquakes in the Islamabad-Kathmandu seismic zone that extends along Himalayan seismic belt, and assess the seismicity trend of this zone by this theory. The results indicate that a MW 8.4 ~ 8.8 characteristic earthquake will strike the central Himalaya and the magnitude parameters of the 2015 Nepal earthquakes have a great impact on judging its occurrence time. When adopting the parameters by CEDC, the zone has almost reached the critical state and the expected characteristic one will take place in a short term (most likely within 10 years), probably with some less than or equal to MW 7.0 foreshocks. When adopting the parameters by NEIC, the occurrence time of the expected characteristic one cannot be judged at present until the critical state after the occurrence of some expected less than or equal to MW 8.1 preshocks is reached. Abstract Current methods used for diagnosis of acute infection of pathogens rely on detection of nucleic acids, antigens, or certain classes of antibodies such as IgM. Here we report a virus enzyme assay as an alternative to these methods for detection of acute viral infection. In this method, we used a luciferin derivative as the substrate for detection of the enzyme activity of influenza viral neuraminidase as a means for diagnosis of influenza. The resulting commercial test, the qFLU Dx Test, uses a different supply chain that does not compete with those for the current tests. The assay reagents were formulated as a master mix that accommodated both the neuraminidase and luciferase reactions, thereby enabling rapid and prolonged production of stable light signal in the presence of influenza virus in the sample. The assay was evaluated using depository throat swab specimens. As expected, the assay exhibited similar detection rates for all influenza types and subtypes except for A(H7N9), which exhibited lower detection rate due to lower viral titer in the specimens. When throat swab specimens were diluted with the sample buffer of the test kit and tested with the qFLU Dx Test. The sensitivity and specificity were 82.41% (95% confidence interval: 79.66–85.84%) and 95.39% (95% confidence interval: 94.32–96.46%), respectively, for these diluted specimens in comparison to a real-time polymerase chain reaction assay. The uniqueness of the qFLU Dx Test as an enzymatic assay makes it highly complementary with currently available methods.

Non-Hermitian systems associated with exceptional points (EPs) are expected to demonstrate a giant response enhancement for various sensors. The widely investigated enhancement mechanism based on diverging from an EP should destroy the EP and further limits its applications for multiple sensing scenarios in a time sequence. To break the above limit, here, we proposed a new enhanced sensing mechanism based on shifting an EP. Different from the mechanism of diverging from an EP, our scheme is an EP nondemolition and the giant enhancement of response is acquired by a slight shift of the EP along the parameter axis induced by perturbation. The new sensing mechanism can promise the most effective response enhancement for all sensors in the case of multiple sensing in a time sequence. To verify our sensing mechanism, we construct a mass sensor and a gyroscope with concrete physical implementations. Our work will deepen the understanding of EP-based sensing and inspire designing various high-sensitivity sensors in different physical systems.

35. On the Multiscale Oceanic Heat Transports Toward the Bases of the Antarctic Ice Shelves

The mass balance of the Antarctic Ice Sheet (AIS) is important to global sea-level change. The AIS loses mass mainly through basal melting and subsequent calving of the Antarctic ice shelves. However, the simulated basal melting rates are very uncertain in ice sheet models, partially resulting from the poor understanding of oceanic heat transports. In this article, we review the recent progress in understanding and simulating such heat transports. Regulated by major circulation features, Circumpolar Deep Water (CDW) is much closer to the Bellingshausen–Amundsen Seas and the Cooperation Sea (60°E to 90°E) and the sector further east to 160°E. The ice shelves within these sectors are experiencing enhanced basal melting resulting from tropical forcing and intensified westerlies. Around West Antarctica, the isopycnal structure favors the delivery of CDW across slopes and shelves, while around East Antarctica, the persistent and strong westward Antarctic Slope Current (Front) acts to prevent warm-water intrusion. Both eddies and troughs favor heat transport to the fronts of the ice shelves and even into the cavities. The sharp contrast between the water column thicknesses on both sides of ice shelf fronts blocks the barotropic inflows and can excite topographic Rossby waves. Inside the cavities, the heat fluxes to the bases of the ice shelves are controlled by the cavity geometry, the circulations in the cavities, and the properties of the water masses beneath the ice shelves. Limited direct observations of cavities have promoted the development of various models. To improve basal melting simulations, meltwater plume models have been developed to study meltwater-laden mixed layer dynamics by increasing the vertical resolution, with recent advanced studies considering the vertical structures of frazil ice concentration and velocity. To reduce the uncertainties in the simulated and projected basal mass loss of the Antarctic ice shelves, future efforts should be devoted to improving the bathymetry and cavity geometry, investigating small-scale processes and parameterizing these processes in coupled climate–ice sheet models, and quantifying the feedback from the mass loss of the AIS.

The measurements of body size data not only reflect the physical fitness, carcass structure, excellent growth condition, and developmental relationship among tissues and organs of animals but are also critical indicators to measure the growth and development of sheep. Computer vision-based body size identification is a non-contact and stress-free method. In this study, we analyzed different body size traits (height at wither, body slanting length, chest depth, chest circumference, shank circumference, hip height, shoulder width, and rump width) and the body weight of 332 Ujumqin sheep and significant correlations (P < 0.05) were obtained among all traits in Ujumqin sheep. Except for shoulder width, rump width, and shank circumference, all were positively correlated, and the effect of sex on Ujumqin sheep was highly significant. The main body size indexes affecting the body weight of rams and ewes were obtained through stepwise regression analysis of body size on body weight, in order of chest circumference, body slanting length, rump width, hip height, height at wither, and shoulder width for rams and body slanting length, chest circumference, rump width, hip height, height at wither and shoulder width for ewes. The body slanting length, chest circumference, and hip height of ewes were used to construct prediction equations for the body weight of Ujumqin sheep of different sexes. The model's prediction accuracy was 83.9% for the rams and 79.4% for ewes. Combined with a Mask R-CNN and machine vision methods, recognition models of important body size parameters of Ujumqin sheep were constructed. The prediction errors of body slanting length, height at wither, hip height, and chest circumference were ~5%, chest depth error was 9.63%, and shoulder width, rump width, and shank circumference errors were 14.95, 12.05, and 19.71%, respectively. The results show that the proposed method is effective and has great potential in precision management.

The growth of secondary hair follicles in cashmere goats follows a seasonal cycle. Melatonin can regulate the cycle of cashmere growth. In this study, melatonin was implanted into live cashmere goats. After skin samples were collected, transcriptome sequencing and histological section observation were performed, and weighted gene co-expression network analysis (WGCNA) was used to identify key genes and establish an interaction network. A total of 14 co-expression modules were defined by WGCNA, and combined with previous analysis results, it was found that the blue module was related to the cycle of cashmere growth after melatonin implantation. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis showed that the first initiation of exogenous melatonin-mediated cashmere development was related mainly to the signaling pathway regulating stem cell pluripotency and to the Hippo, TGF-beta and MAPK signaling pathways. Via combined differential gene expression analyses, 6 hub genes were identified: PDGFRA, WNT5A, PPP2R1A, BMPR2, BMPR1A, and SMAD1. This study provides a foundation for further research on the mechanism by which melatonin regulates cashmere growth.

43. Identification of the key proteins associated with different hair types in sheep and goats

Animal-derived fiber has the characteristics of being light, soft, strong, elastic and a good thermal insulator, and it is widely used in many industries and traditional products, so it plays an important role in the economy of some countries. Variations in phenotypes of wool fibers among different species and breeds are important for industry. We found that the mean fiber diameter of cashmere was significantly smaller than that of sheep wool (p < 0.01), and sheep wool was significantly smaller than goat wool (p < 0.01). Compared with traditional proteomics technology, we analyzed cashmere, guard hair, and wool by Laber-free proteomics technology and detected 159, 204, and 70 proteins, respectively. Through the sequential windowed acquisition of all theoretical fragmentations (SWATH), 41 and 54 differentially expressed proteins were successfully detected in the cashmere vs. wool group and guard hair vs. wool group. Protein‒protein interaction network analysis of differentially expressed proteins revealed many strong interactions related to KRT85, KRTAP15-1 and KRTAP3-1. The final analysis showed that the proportion of KRT85, KRTAP15-1 and KRTAP3-1 might be the key to the difference in fiber diameter and could be used as a potential molecular marker for distinguishing different fiber types.

Wireless energy harvesting enables the conversion of ambient energy into electrical power for small wireless electronic devices. This technology offers numerous advantages, including availability, ease of implementation, wireless functionality, and cost-effectiveness. Radio frequency energy harvesting (RFEH) is a specific type of wireless energy harvesting that enables wireless power transfer by utilizing RF signals. RFEH holds immense potential for extending the lifespan of wireless sensors and wearable electronics that require low-power operation. However, despite significant advancements in RFEH technology for self-sustainable wearable devices, numerous challenges persist. This literature review focuses on three key areas: materials, antenna design, and power management, to delve into the research challenges of RFEH comprehensively. By providing an up-to-date review of research findings on RFEH, this review aims to shed light on the critical challenges, potential opportunities, and existing limitations. Moreover, it emphasizes the importance of further research and development in RFEH to advance its state-of-the-art and offer a vision for future trends in this technology.

49. On the influences of the continental shelf bathymetry correction in Prydz Bay, East Antarctica

The bathymetry around Antarctica can govern the shelf sea circulations and play a key role in conditioning water masses. In Prydz Bay, the Prydz Bay Gyre and coastal currents are also determined by the continental shelf topography. However, due to the paucity of beam echo sounding data, the bathymetric datasets in Prydz Bay still have large uncertainties. With the aid of in situ hydrographic observations, this study focuses on the correction of an up-to-date bathymetric dataset and the resultant influences on the shelf circulation and the basal melting of the ice shelves. The corrected bathymetry mainly improves the biased shallow representations in the uncorrected bathymetric data set, with a maximum change of ~500 m deepening in the eastern flank of Prydz Bay. Sensitivity numerical experiments show that the bathymetric corrections in Prydz Bay have a significant impact on the circulation pattern and onshore warm water intrusions. In addition, the corrected bathymetry markedly decreases the heat transport towards the calving front of the Amery Ice Shelf. The onshore heat transport reduces by ~22.18% from ~5.23×1013 J s-1 to ~4.07×1013 J s-1 over the outer shelf. Over the inner shelf, the heat transport towards the Amery Ice Shelf reduces by ~18.15% from ~5.95×1013 J s-1 to ~4.87×1013 J s-1. Consequently, the temporally and spatially averaged basal melting rate of the Amery Ice Shelf reduces by ~13.04% from ~0.69 m yr-1 to ~0.60 m yr-1.

Electrochemical CO2 reduction reaction (ECRR) from CO2 and H2O as reactants is one of the most promising candidates for the cycle and utilization of carbon resources. The expanding of product scope and industrial application of such technique are still inhibited by its activity and selectivity limitations, which can be improved by the rational design of catalysts. Therefore, heterostructured materials with novel interfaces which may modify the behavior of reactants and intermediates, can in principle improve ECRR performance. In this perspective, the advantages of heterostructured materials as electrocatalysts are introduced, and recent research toward ECRR based on them is highlighted according to the number of coupled carbon atoms. The electrosynthesis of CO, HCOOH, CH3OH, CH4, C2H4, CH3COOH and CH3COCH3 are detailly emphasized and evaluated as representative examples from the viewpoints of catalyst structure, activity, product selectivity and mechanism. Finally, the major challenges and opportunities of heterostructured materials in ECRR are also proposed. This paper aims to guide future investigations of designing diverse heterostructured materials, upgrading carbon-based products, and further promote the scale-up application of ECRR.
