Abstract Background To assess the prevalence and epidemic pattern of multidrug-resistant tuberculosis in Hangzhou City, Zhejiang Province, China during 2012–2022. Methods All the tuberculosis cases undergoing drug susceptibility testing during 2012–2022 were included in this study. De-identified information was extracted from the electronic database Tuberculosis Information Management System for analysis of drug resistance prevalence in Hangzhou and ecdemic multidrug-resistant tuberculosis which originated from other regions. Chi-square tests were used to compare drug resistance rates between different groups, while Chi-square tests for trend were used to evaluate the change of drug resistance rates over the years of 2012–2022. The sources and destinations of ecdemic multidrug-resistant tuberculosis were illustrated using a Sankey diagram. Results Of 21,127 cases included in this study, 1119 (5.3%) were multidrug-resistant tuberculosis. A significant decline in multidrug-resistant tuberculosis rates was observed during 2012–2022. There was a significant difference in multidrug-resistant tuberculosis rates among immigrant population and local residents in Hangzhou City. Of 1119 multidrug-resistant tuberculosis cases, 515(46%) were ecdemic multidrug-resistant tuberculosis cases, of which 277(53.8%) were from other parts of Zhejiang Province and 238(46.2%) were from other provinces in China. Anhui, Jiangxi and Sichuan were among top three provinces which were the source of ecdemic multidrug-resistant tuberculosis cases. Three districts including Xiaoshan, Shangcheng and Linping districts had the most cases in Hangzhou. The proportion of ecdemic multidrug-resistant tuberculosis cases in Binjiang, Xiaoshan, Qiantang and Linping districtalso exceeded 30% of total cases. Conclusions Multidrug-resistant tuberculosis prevalence has been declining in Hangzhou. Migrant population contributed to a significant potion of cases in Hangzhou. Interventions should be tailed to local and migrant residents.

Early embryonic development relies on intricately regulated gene expression, and miRNAs influence zygotic genome activation (ZGA), cleavage, and cell fate determination through post-transcriptional regulatory mechanisms. miR-192 is expressed in early pig embryos and participates in various reproductive processes. However, its role in pre-implantation pig embryo development remains poorly understood. In this study, we microinjected the miR-192 agonist (miR-192 agomir) into parthenogenetically activated pig embryos to evaluate its effects on early pig embryo development. Our findings indicate that compared to the control group (agomir NC), miR-192 agomir impairs the developmental capacity of parthenogenetic pig embryos to reach the 2-cell, 4-cell, and blastocyst stages. This impairment leads to imbalances in the oxidative–reductive system and abnormalities in mitochondrial function during the 4-cell stage, resulting in the significant accumulation of ROS, notable decreases in the expression of antioxidant enzymes CAT and SOD1 mRNA, reduction in mitochondrial membrane potential, and induction of apoptosis in pig blastocysts. Additionally, the overexpression of miR-192 inhibits the expression of its target genes YY1 and the pluripotency factor NANOG mRNA. In conclusion, this study reveals that the overexpression of miR-192 adversely affects early pig embryo development, providing new evidence for understanding the role miR-192 plays in reproduction.

This study proposes a new strategy for achieving rapid volumetric heating of moldable low-dielectric-loss glass over a wide temperature range using only microwave sources. As a proof of concept, both multi-physics modeling and experimental demonstration of microwave heating (MWH) of Pyrex 7740 glass are performed. The results reveal that there is an activation threshold of 400 °C for this glass, i.e., below this threshold the glass is mainly heated by SiC molds through interfacial heat transfer, while above it the glass can be directly heated by 2.45-GHz MWs through volumetric dielectric loss. As a result, the glass preform can attain an internal temperature of 727 °C (well above its transition point) within a short MWH process (130 s), at a low level of specific energy consumption (6.43 kW h/kg). The average volumetric heating rate of the glass is estimated as 5.44 °C/s, which outperforms the reported rates (2.00−3.60 °C/s) in previous precision glass molding (PGM) studies. Furthermore, the temperature difference in the entire glass can be minimized through MWH combined with post-annealing. With the demonstrated capabilities of instantaneous, volumetric, and selective heating, the proposed MWH strategy holds great promise in PGM and many other glass thermoforming sectors.

Abstract Background Inflammatory myofibroblastic tumor (IMT) is a unique, rarely metastatic tumor composed of myofibroblasts and fibrous spindle cells with inflammatory cell infiltration that can affect any organ in the human body. By reviewing the relevant literature on PubMed, we found that this is the first case report of IMT with both gastric and cardiac involvement. Case presentation A 57-year-old male patient was admitted to the hospital with complaints of malaise, poor appetite, and epigastric pain with black stools. We found a mass in the patient’s stomach and left atrium by contrast-enhanced computed tomography, 18 F-fluorodeoxyglucose positron emission tomography/computed tomography, and other tests. The patient underwent laparoscopic Billroth II subtotal gastrectomy and Braun’s gastrointestinal reconstruction under general anesthesia. On the 46th day following stomach surgery, the cardiac tumor was removed under general anesthesia. The patient has treated with doxorubicin 70 mg of D1 chemotherapy two months after cardiac surgery. Postoperative pathological immunohistochemistry of the mass confirmed the diagnosis of an IMT. His review three months after the cardiac surgery suggested the progression of the left atrial mass, but he declined further treatment and finally died one month after the review. Conclusions As a unique class of tumors that rarely metastasize, IMTs have an unknown etiology and pathogenesis, and distant metastasis is primarily observed in patients with negative activin receptor-like kinase (ALK) expression. The preferred treatment for IMT is complete surgical resection, and the effectiveness of adjuvant therapy for patients with distant metastases is still being determined. The clinical presentation of IMT lacks specificity and is often related to the location of tumor growth, which poses a diagnostic challenge. Pathological immunohistochemistry is the only way to confirm the diagnosis at present. Our case report reminds clinicians that a category of ALK-negative IMT with a tendency toward distant metastasis should not be ignored.

Abstract Objective: To investigate the involvement of HOX transcript antisense RNA (HOTAIR) in the injury of renal tubular epithelial cells induced by high glucose. Results: In high glucose‐induced HK‐2 cells, the expression of HOTAIR was upregulated, resulting in suppressed cell proliferation. Meanwhile, HOTAIR upregulates the expression of pro‐apoptotic proteins Bax and cleaved caspase‐3, while downregulating the expression of the anti‐apoptotic protein Bcl‐2. Luciferase reporter assays revealed that HOTAIR could target miR‐126‐5p. Additionally, it was found that the PI3K/Akt signaling pathway serves as a downstream target of miR‐126‐5p. Knockdown of HOTAIR relieved apoptosis, whereas further inhibition of miR‐126‐5p led to apoptosis in HK‐2 cells. Conclusions: HOTAIR plays a regulatory role in mediating high glucose‐induced injuries in HK‐2 cells, specifically affecting apoptosis and cell viability, via the miR‐126‐5p/PI3K/Akt signaling pathway.

Objective The study was conducted to screen differentially expressed miRNAs in sows at early pregnancy by high-throughput sequencing and explore its mechanism of action on embryo implantation. Methods The blood serum of pregnant and non-pregnant Landrace×Yorkshire sows were collected 14 days after artificial insemination, and exosomal miRNAs were purified for high throughput miRNA sequencing. The expression patterns of 10 differentially expressed (DE) miRNAs were validated by quantitative reverse transcription-polymerase chain reaction (qRT-PCR). The qRT-PCR quantified the abundance of serum exosomal miR-192 in pregnant and control sows, and the diagnostic power was assessed by receiver operating characteristic (ROC) analysis. The target genes of DE miRNAs were predicted with bioinformatics software, and the functional and pathway enrichment analysis was performed on gene ontology and the Kyoto encyclopedia of genes and genomes terms. Furthermore, a luciferase reporter system was used to identify the target relation between miR-192 and integrin alpha 4 (ITGA4), a gene influencing embryo implantation in pigs. Finally, the expression levels of miRNAs and the target gene ITGA4 were analyzed by qRT-PCR, and western blot, with the proliferation of BeWo cells detected by cell counting kit-8 (CCK-8). Results A total of 221 known miRNAs were detected in the libraries of the pregnant and non-pregnant sows, of which 55 were up-regulated and 67 were down-regulated in the pregnant individuals compared with the non-pregnant controls. From these, the expression patterns of 10 DE miRNAs were validated. The qRT-PCR analysis further confirmed a significantly higher expression of miR-192 in the serum exosomes extracted from pregnant sows, when compared to controls. The ROC analysis revealed that miR-192 provided excellent diagnostic accuracy for pregnancy (area under the ROC curve [AUC] = 0.843; p>0.001). The dual-luciferase reporter assay indicated that miR-192 directly targeted ITGA4. The protein expression of ITGA4 was reduced in cells that overexpressed miR-192. Overexpression of miR-192 resulted in the decreased proliferation of BeWo cells and regulated the expression of cell cycle-related genes. Conclusion Serum exosomal miR-192 could serve as a potential biomarker for early pregnancy in pigs. miR-192 targeted ITGA4 gene directly, and miR-192 can regulate cellular proliferation.

Tuberculosis (TB) is a major global health threat despite its virtual elimination in developed countries. Issues such as drug accessibility, emergence of multidrug-resistant strains, and limitations of the current BCG vaccine highlight the urgent need for more effective TB control measures. This study constructed BCG strains overexpressing Rv1002c and found that the rBCG-Rv1002c strain secreted more glycosylated proteins, significantly enhancing macrophage activation and immune protection against Mycobacterium tuberculosis (M. tb). These results indicate that Rv1002c overexpression promotes elevated levels of O-glycosylation in BCG bacteriophages, enhancing their phagocytic and antigenic presentation functions. Moreover, rBCG-Rv1002c significantly upregulated immune regulatory molecules on the macrophage surface, activated the NF-κB pathway, and facilitated the release of large amounts of NO and H2O2, thereby enhancing bacterial control. In mice, rBCG-Rv1002c immunization induced greater innate and adaptive immune responses, including increased production of multifunctional and long-term memory T cells. Furthermore, rBCG-Rv1002c-immunized mice exhibited reduced lung bacterial load and histological damage upon M. tb infection. This result shows that it has the potential to be an excellent candidate for a preventive vaccine against TB.

The objective of this research is to explore the temporal importance of community-scale human activity features for rapid assessment of flood impacts. Ultimate flood impact data, such as flood inundation maps and insurance claims, becomes available only weeks and months after the floods have receded. Crisis response managers, however, need near-real-time data to prioritize emergency response. This time lag creates a need for rapid flood impact assessment. Accordingly, community-scale big data (such as satellite imagery) has been utilized for the early estimation of end flood impacts. Some recent studies have shown promising results for using human activity fluctuations as indicators of flood impacts. Existing studies, however, used mainly a single community-scale activity feature for the estimation of flood impacts and have not investigated their temporal importance for indicating flood impacts. Hence, in this study, we examined the importance of heterogeneous human activity features (such as human mobility, visits to points-of-interest, and social media posts) in different flood event stages. Using four community-scale big data categories we derived ten features related to the variations in human activity (e.g., travel, credit card transactions, and online communications) and evaluated their temporal importance for rapid assessment of flood impacts. Using multiple random forest models, we examined the temporal importance of each feature in indicating the extent of flood impacts (measured by the flood insurance claims and flood inundations) in the context of the 2017 Hurricane Harvey in Harris County, Texas. Our findings reveal that 1) fluctuations in human activity index and percentage of congested roads are the most important indicators for rapid flood impact assessment during response and recovery stages; 2) variations in credit card transactions assumed a middle ranking in both response and recovery stages; and 3) patterns of geolocated social media posts (Twitter) were of low importance across flood stages. Insights derived from data analysis reveal the potential for harnessing community-scale data characterizing human activity fluctuations for rapid assessment of flood impacts. The results of this research could rapidly forge a multi-tool enabling crisis managers to identify hotspots with severe flood impacts at various stages then to plan and prioritize effective response strategies.

Abstract Understanding actor collaboration networks and their evolution is essential to promoting collective action in resilience planning and management of interdependent infrastructure systems. Local interactions and choice homophily are two important network evolution mechanisms. Network motifs encode the information of network formation, configuration, and the local structure. Homophily effects, on the other hand, capture whether the network configurations have significant correlations with node properties. The objective of this paper is to explore the extent to which local interactions and homophily effects influence actor collaboration in resilience planning and management of interdependent infrastructure systems. We mapped bipartite actor collaboration network based on a post-Hurricane Harvey stakeholder survey that revealed actor collaborations for hazard mitigation. We examined seven bipartite network motifs for the mapped collaboration network and compared the mapped network to simulated random models with same degree distributions. Then we examined whether the network configurations had significant statistics for node properties using exponential random graph models. The results provide insights about the two mechanisms—local interactions and homophily effect—influencing the formation of actor collaboration in resilience planning and management of interdependent urban systems. The findings have implications for improving network cohesion and actor collaborations from diverse urban sectors.

Abstract The objective of this study was to investigate the importance of multiple county-level features in the trajectory of COVID-19. We examined feature importance across 2787 counties in the United States using data-driven machine learning models. Existing mathematical models of disease spread usually focused on the case prediction with different infection rates without incorporating multiple heterogeneous features that could impact the spatial and temporal trajectory of COVID-19. Recognizing this, we trained a data-driven model using 23 features representing six key influencing factors affecting the pandemic spread: social demographics of counties, population activities, mobility within the counties, movement across counties, disease attributes, and social network structure. Also, we categorized counties into multiple groups according to their population densities, and we divided the trajectory of COVID-19 into three stages: the outbreak stage, the social distancing stage, and the reopening stage. The study aimed to answer two research questions: (1) The extent to which the importance of heterogeneous features evolved at different stages; (2) The extent to which the importance of heterogeneous features varied across counties with different characteristics. We fitted a set of random forest models to determine weekly feature importance. The results showed that: (1) Social demographic features, such as gross domestic product, population density, and minority status maintained high-importance features throughout stages of COVID-19 across 2787 studied counties; (2) Within-county mobility features had the highest importance in counties with higher population densities; (3) The feature reflecting the social network structure (Facebook, social connectedness index), had higher importance for counties with higher population densities. The results showed that the data-driven machine learning models could provide important insights to inform policymakers regarding feature importance for counties with various population densities and at different stages of a pandemic life cycle.

Objectives: The impact of tuberculosis (TB) history on the risk of multidrug-resistant tuberculosis (MDR-TB) is not yet fully understood. We aimed to identify the impact of different TB history at the onset of future MDR-TB. Methods: A large, retrospective, population-based cohort study was performed between 2005 and 2019. A multivariable Cox model was used to evaluate independent risk factors for MDR-TB for individuals with different previous TB history (PTBH), such as newly diagnosed TB history (NDTH) and re-treated TB history (RTH). Results: Overall, 12 172 individuals with PTBH were included in this study. The main impacts of different PTBH at the onset of future MDR-TB were as follows: (a) low family income, high-risk occupation, TB patients with severe infection, extended or shortened treatment course, 2H3R3Z3E3/4H3R3 and frequency of sputum culture were significantly linked to incident MDR-TB only in individuals with NDTH (P < 0.05); (b) passive mode of TB case finding, individualised treatment regimens, 3HRZES/6HRE, duration of pulmonary cavities, excellent frequency of chest X-ray examination and duration of negative sputum smear were significantly associated with incident MDR-TB only in individuals with RTH (P < 0.05); (c) age

Abstract The objective of this study is to examine the transmission risk of COVID-19 based on cross-county population co-location data from Facebook. The rapid spread of COVID-19 in the United States has imposed a major threat to public health, the real economy, and human well-being. With the absence of effective vaccines, the preventive actions of social distancing, travel reduction and stay-at-home orders are recognized as essential non-pharmacologic approaches to control the infection and spatial spread of COVID-19. Prior studies demonstrated that human movement and mobility drove the spatiotemporal distribution of COVID-19 in China. Little is known, however, about the patterns and effects of co-location reduction on cross-county transmission risk of COVID-19. This study utilizes Facebook co-location data for all counties in the United States from March to early May 2020 for conducting spatial network analysis where nodes represent counties and edge weights are associated with the co-location probability of populations of the counties. The analysis examines the synchronicity and time lag between travel reduction and pandemic growth trajectory to evaluate the efficacy of social distancing in ceasing the population co-location probabilities, and subsequently the growth in weekly new cases across counties. The results show that the mitigation effects of co-location reduction appear in the growth of weekly new confirmed cases with one week of delay. The analysis categorizes counties based on the number of confirmed COVID-19 cases and examines co-location patterns within and across groups. Significant segregation is found among different county groups. The results suggest that within-group co-location probabilities (e.g., co-location probabilities among counties with high numbers of cases) remain stable, and social distancing policies primarily resulted in reduced cross-group co-location probabilities (due to travel reduction from counties with large number of cases to counties with low numbers of cases). These findings could have important practical implications for local governments to inform their intervention measures for monitoring and reducing the spread of COVID-19, as well as for adoption in future pandemics. Public policy, economic forecasting, and epidemic modeling need to account for population co-location patterns in evaluating transmission risk of COVID-19 across counties.

The objective of this paper is to examine population response to COVID-19 and associated policy interventions through detecting early-warning signals in time series of visits to points of interest (POIs). Complex systems, such as cities, would demonstrate early-warning signals (e.g., increased autocorrelation and standard deviation) when they approach phase transitions responding to external perturbation, such as crises, policy changes, and human behavior changes. In urban systems, population visits to POIs, such as restaurants, museums, and hospitals, represent a state of cities as complex systems. These states may undergo phase transitions due to population response to pandemic risks and intervention policies (e.g., social distancing and shelter-in-place orders). In this study, we conducted early-warning signal detection on population visits to POIs to examine population response to pandemic risks, and we evaluated time lags between detected early-warning dates and dates of first cases and policy interventions. We examined two early-warning signals, the increase of autocorrelation at-lag-1 and standard deviation, in time series of population visits to POIs in 17 metropolitan cities in the United States of America. We examined visits to grouped POIs according to two categories of essential services and non-essential services. The results show that: (1) early-warning signals for population response to COVID-19 were detected between February 14 and March 11, 2020 in 17 cities; (2) detected population response had started prior to shelter-in-place orders in 17 cities; (3) early-warning signals detected from the essential POIs visits appeared earlier than those from non-essential POIs; and 4) longer time lags between detected population response and shelter-in-place orders led to a less decrease in POI visits. The results show the importance of detecting early-warning signals during crises in cities as complex systems. Early-warning signals could provide important insights regarding the timing and extent of population response to crises to inform policymakers.

The Qianjiadian uranium deposit is located in the Kailu Depression of Songliao Basin. It is a large-scale in-situ leachable uranium deposit of sandstone type and provides a typical case of the uranium deposits in the Songliao Basin. Here we analyze the impact of oil and gas on reduction alteration, and factors including low grade mineralization. The Qianjiadian uranium deposit is characterized as a typical roll-type deposit with interlayers in oxidized zone. The occurrence of reductive low-permeability sandstone in uranium reservoirs controls the morphology of the uranium-rich orebody. We propose a metallogenic model involving multi-stage superposition characterized by “primary sediment enrichment - interlayer oxidation - superimposed transformation”. Our model would be helpful for formulating guidelines in the exploration for sandstone-type uranium deposits in the Songliao Basin.

Abstract Background Circular RNAs (circRNAs) are a new group of non-coding RNAs that play vital roles in cancer occurrence, including gastric cancer (GC). Nevertheless, the role and underlying regulatory mechanisms of circHIPK3 in GC remain unclear. Methods The expression levels of circHIPK3, miR-876-5p, and phosphoinositide-3-kinase regulatory subunit 1 (PIK3R1) were estimated by real-time quantitative polymerase chain reaction (RT-qPCR) assay. The proliferation, migration, and invasion of GC cells were determined by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl-2H-tetrazol-3-ium bromide (MTT) and transwell assay. Glutaminolysis of GC cells was assessed by measuring glutamine, glutamate, and α-ketoglutarate levels. The western blot was employed to examine the related-protein expression. The association between miR-876-5p and circHIPK3 or PIK3R1 was predicted and affirmed by bioinformatics database starBase v2.0 and dual-luciferase reporter assay, respectively. Eventually, the xenograft experiment was used to assess the role of circHIPK3 silencing in vivo. Results CircHIPK3 was upregulated in GC tissues and cells compared with controls, and circHIPK3 was more resistance to RNase R than linear homeodomain interacting protein kinase 3 (HIPK3) mRNA. Silencing of circHIPK3 inhibited GC cells proliferation, migration, invasion, and glutaminolysis as well as tumor tumorigenic ability. Moreover, we also found that miR-876-5p, interacted with PIK3R1, was a target gene of circHIPK3. CircHIPK3 silencing induced effects on GC cells were abolished by silencing of miR-876-5p. In addition, upregulation of PIK3R1 inversed miR-876-5p overexpression-induced effects on GC cells. Conclusion The circHIPK3 mediated the proliferation, migration, invasion, and glutaminolysis of GC cells partly through regulation of miR-876-5p/PIK3R1 axis by the mechanism of competing endogenous RNAs (ceRNA), indicating circHIPK3 was a GC-associated circRNA that promoted GC development.

Abstract Background Currently, multiple circular RNAs (circRNAs) have been verified to act as essential regulators in the progression of gastric cancer (GC). We aimed to investigate the role of circ_0008035 in GC progression. Methods Quantitative real-time polymerase chain reaction (qRT-PCR) was utilized to measure the expression of circ_0008035 and miR-599. 3-(4,5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-H-tetrazolium bromide (MTT) assay was employed to evaluate cell proliferation and ferroptosis. Western blot assay was performed to measure the levels of cyclin D1, proliferating cell nuclear antigen (PCNA) and eukaryotic initiation factor 4A1 (EIF4A1). Flow cytometry analysis was conducted to assess cell apoptosis. The iron accumulation, lipid peroxidation and mitochondrial membrane potential were examined by relevant kits. Dual-luciferase reporter assay was conducted to determine the targeting relationship between miR-599 and circ_0008035 or EIF4A1. A murine xenograft model was established to investigate the function of circ_0008035 in vivo. Results Circ_0008035 was up-regulated in GC tissues and cells. Silencing of circ_0008035 repressed cell proliferation and induced cell apoptosis and ferroptosis in GC cells. Circ_0008035 acted as a sponge of miR-599. The effects of circ_0008035 knockdown on GC cell proliferation, apoptosis and ferroptosis were abolished by miR-599 inhibition. EIF4A1 was confirmed to be a target gene of miR-599. Circ_0008035 knockdown inhibited EIF4A1 expression by targeting miR-599. Moreover, the suppressive role of circ_0008035 deficiency in GC progression could be restored by EIF4A1. Additionally, circ-0008035 knockdown hampered tumorigenesis in vivo. Conclusion Circ_0008035 promoted GC cell growth and repressed apoptosis and ferroptosis by up-regulating EIF4A1 through sponging miR-599.

New York has become one of the worst-affected COVID-19 hotspots and a pandemic epicenter due to the ongoing crisis. This paper identifies the impact of the pandemic and the effectiveness of government policies on human mobility by analyzing multiple datasets available at both macro and micro levels for New York City. Using data sources related to population density, aggregated population mobility, public rail transit use, vehicle use, hotspot and non-hotspot movement patterns, and human activity agglomeration, we analyzed the inter-borough and intra-borough movement for New York City by aggregating the data at the borough level. We also assessed the internodal population movement amongst hotspot and non-hotspot points of interest for the month of March and April 2020. Results indicate a drop of about 80% in people’s mobility in the city, beginning in mid-March. The movement to and from Manhattan showed the most disruption for both public transit and road traffic. The city saw its first case on March 1, 2020, but disruptions in mobility can be seen only after the second week of March when the shelter in place orders was put in effect. Owing to people working from home and adhering to stay-at-home orders, Manhattan saw the largest disruption to both inter- and intra-borough movement. But the risk of spread of infection in Manhattan turned out to be high because of higher hotspot-linked movements. The stay-at-home restrictions also led to an increased population density in Brooklyn and Queens as people were not commuting to Manhattan. Insights obtained from this study would help policymakers better understand human behavior and their response to the news and governmental policies.

Preparedness for adverse events is critical to building urban resilience to climate-related risks. While most extant studies investigate preparedness patterns based on survey data, this study explores the potential of big digital footprint data (i.e. population visits to points of interest (POI)) to investigate preparedness patterns in the real case of Hurricane Ida (2021). We further investigate income and racial inequality in preparedness by combining the digital footprint data with demographic and socioeconomic data. A clear pattern of preparedness was seen in Louisiana with aggregated visits to grocery stores, gasoline stations, and construction supply dealers increasing by nearly 9%, 12%, and 10% respectively, representing three types of preparedness: survival, mobility planning, and hazard mitigation. Preparedness for Hurricane Ida was not seen in New York and New Jersey states. Inequality analyses for Louisiana across census block groups (CBGs) demonstrate that CBGs with higher income have more (nearly 8% greater) preparedness in visiting gasoline stations, while CBGs with a larger percentage of the white population have more preparedness in visiting grocery stores (nearly 12% more) in the lowest income groups. The results indicate that income and racial inequality differ across different preparedness in terms of visiting different POIs.

Background: To date, too little attention has been paid to monitoring and estimating the risk of incident multidrug-resistant tuberculosis (MDR-TB) among individuals with a previous tuberculosis history (PTBH). The purpose of this study was to assess the incidence of and risk factors for MDR-TB in those individuals.Methods: Between 2005 and 2020, a large, retrospective, population-based cohort study was performed in Hangzhou, China. A multivariable Cox regression model was used to evaluate independent predictors of incident MDR-TB among individuals with PTBH.Results: The incidence density of MDR-TB was 22.6 per 1,000 person-years (95% confidence level and an interval of 20.9–24.3) for individuals with PTBH. The incidence of MDR-TB increased significantly in individuals who• were under 60 years old.• were male.• had a history of direct contact.• came from low-income families.• worked in high-risk occupations.• lived in rural areas.• had a retreatment TB history.• had an unfavorable outcome in their previous treatment (P < 0.05).In addition, we found that the following factors were significantly linked to the MDR-TB risk among individuals with PTBH (P < 0.05):• sociodemographic factors such as the 21–30 and 31–40 year age groups, or a history of direct contact.• clinical factors like passive modes of TB case finding (PMTCF), human immunodeficiency virus infection, unfavorable treatment outcomes, retreated TB history, non-standardized treatment regimens of retreatment TB patients, and duration of pulmonary cavities (DPC).• microbiological factors, such as duration of positive sputum culture.We also found that the 21–30 year age group, low family income, and PMTCF were significantly linked to incident MDR-TB only in males with PTBH, whilst the 41–50 year age group, extended treatment course, and DPC were significantly associated with female MDR-TB only.Conclusion: The incidence of MDR-TB was high, with a higher rate among subjects with a history of direct contact and unfavorable treatment outcomes. There was a gender difference in the incidence density and risk factors of MDR-TB among individuals with PTBH. Long-term monitoring and gender-specific risk-factor modifications should be given to individuals with PTBH.

The spread of pandemics such as COVID-19 is strongly linked to human activities. The objective of this article is to specify and examine early indicators of disease spread risk in cities during the initial stages of outbreak based on patterns of human activities obtained from digital trace data. In this study, the Venables distance (Dv) and the activity density (Da) are used to quantify and evaluate human activities for 193 United States counties, whose cumulative number of confirmed cases was greater than 100 as of March 31, 2020. Venables distance provides a measure of the agglomeration of the level of human activities based on the average distance of human activities across a city or a county (less distance could lead to a greater contact risk). Activity density provides a measure of level of overall activity level in a county or a city (more activity could lead to a greater risk). Accordingly, Pearson correlation analysis is used to examine the relationship between the two human activity indicators and the basic reproduction number in the following weeks. The results show statistically significant correlations between the indicators of human activities and the basic reproduction number in all counties, as well as a significant leader-follower relationship (time lag) between them. The results also show one to two weeks’ lag between the change in activity indicators and the decrease in the basic reproduction number. This result implies that the human activity indicators provide effective early indicators for the spread risk of the pandemic during the early stages of the outbreak. Hence, the results could be used by the authorities to proactively assess the risk of disease spread by monitoring the daily Venables distance and activity density in a proactive manner.

We examined the effect of social distancing on changes in visits to urban hotspot points of interest. In a pandemic situation, urban hotspots could be potential superspreader areas as visits to urban hotspots can increase the risk of contact and transmission of a disease among a population. We mapped census-block-group to point-of-interest (POI) movement networks in 16 cities in the United States. We adopted a modified coarse-grain approach to examine patterns of visits to POIs among hotspots and non-hotspots from January to May 2020. Also, we conducted chi-square tests to identify POIs with significant flux-in changes during the analysis period. The results showed disparate patterns across cities in terms of reduction in hotspot POI visitors. Sixteen cities were divided into two categories using a time series clustering method. In one category, which includes the cities of San Francisco, Seattle and Chicago, we observed a considerable decrease in hotspot POI visitors, while in another category, including the cities of Austin, Houston and San Diego, the visitors to hotspots did not greatly decrease. While all the cities exhibited overall decreased visitors to POIs, one category maintained the proportion of visitors to hotspot POIs. The proportion of visitors to some POIs (e.g. restaurants) remained stable during the social distancing period, while some POIs had an increased proportion of visitors (e.g. grocery stores). We also identified POIs with significant flux-in changes, indicating that related businesses were greatly affected by social distancing. The study was limited to 16 metropolitan cities in the United States. The proposed methodology could be applied to digital trace data in other cities and countries to study the patterns of movements to POIs during the COVID-19 pandemic.

Abstract Heat wave is serious natural disaster that can harm human health and affect social economy, transportation and ecological environment. This paper investigates the long term trends of high temperature events in three major cities (Beijing, Tianjin and Shijiazhuang) of northern China during 1970–2019, and quantifies the contributions of main influencing factors to the variability of high temperature days. High temperature events in Beijing–Tianjin–Shijiazhuang cities mainly occur from June to July and account for 60–65% of the annual total, showing a significant upward trend in the interannual change. There is a trend of high temperature events starting early and ending late in recent 50 years, and this trend is intensified by the increasing urbanization. Due to geographical location and city scale, the frequency and intensity of high temperature events show difference among Beijing, Tianjin and Shijiazhuang cities. Variance analysis shows that climate warming contributes 20.2–25.5% to the variability of high temperature days, while urban heat island accounts for 14.7–24.2%, equal to the sum of the contributions from atmospheric circulation and solar activity.

In this study, solidification microstructure of fine primary silicon being uniformly distributed on single αAl matrix was obtained when adding Cu–9 wt%P alloy to an Al–10 wt%Si alloy melt or an Al–20 wt%Si alloy melt, stirring at 200 r/min for 12 min and cooling at the cooling rate of 16.3 K·s−1. The change law of silicon phase size with cooling rate and modified treatment parameters was determined by examining the solidification microstructures. It was found that ultrafine AlP was easily swallowed by primary silicon when the cooling rate was slow. The formation mechanism of superfine AlP particles, the growth process of αAl phase around primary silicon and the disappearance process of eutectic silicon were researched under intense stirring of the melt during the Cu–9 wt%P alloy modification. Moreover, the conditions under which fine primary silicon was uniformly distributed in single αAl matrix in hypereutectic Al-Si alloy were put forwarded. Finally, the formation mechanism of the solidification microstructure without eutectic Si in hypereutectic Al-Si alloy was analyzed.

Abstract Background To evaluate prevalence and patterns of drug resistance among pulmonary tuberculosis (TB) patients in Hangzhou City, China. Methods Sputum samples of smear positive TB patients enrolled in 2011 and 2015 were collected and tested for drug susceptibility, and demographic and medical record data were extracted from the electronic database of China Information System for Disease Control and Prevention. Chi-square test was used to compare drug resistance prevalence between new and treated patients and between male and female patients, and Chi-square test for trend was used to compare the prevalence over calendar years 2011 and 2015. Results Of 1326 patients enrolled in 2015, 22.3% had resistance to any first-line anti-TB drugs and 8.0% had multi-drug resistance (MDR); drug resistance rates among previously treated cases were significantly higher than among new cases. Significant declines of resistance to isoniazid, rifampin, ethambutol and streptomycin, and MDR from 2011 to 2015 were observed among previously treated patients, while a significant decline of resistance to rifampin was observed among new cases. Conclusions While the prevalence of acquired drug resistance decreased due to due to implementation of DOTS-Plus program, the prevalence of primary drug resistance due to transmission remained high. Greater efforts should be made to screen drug resistance for case finding and to reduce transmission through improving the treatment and management of drug-resistant patients.

The distribution and diurnal variation of short-duration heavy rainfall (SDHR) and the influence of a complex underlying surface were studied by using fine-scale hourly precipitation data in the Beijing–Tianjin–Hebei (BTH) region during the summers of 2014–2020. Areas prone to SDHR are located mainly in the southern foothills of the Yanshan Mountains, the foothills area, and the trumpet-shaped topographic entrance area north of Beijing, areas inland of the west coast of Bohai Bay, and the northern Beijing urban area. Owing to the influence of topography and the geographical location, the distribution and diurnal variation SDHR is significantly different in the western and northern mountainous areas, the foothills, and the plains. Compared to the underlying urban surface, the topography and the land–sea interface have considerable effects on the distribution of SDHR. A key finding is that the foothills of northern of Beijing, eastern slope and piedmont area of the Taihang Mountains, and the land–sea interface of Bohai Bay play important roles in the formation and propagation of SDHR.

Abstract Background Tissue engineering has emerged as a promising alternative for small-diameter vascular grafts. The aim of this study was to determine the feasibility of using decellularized aortae of fetal pigs (DAFPs) to construct tissue-engineered, small-diameter vascular grafts and to test the performance and application of DAFPs as vascular tissue-engineered scaffolds in the canine arterial system. Methods DAFPs were prepared by continuous enzymatic digestion. Canine vascular endothelial cells (ECs) were seeded onto DAFPs in vitro and then the vascular grafts were cultured in a custom-designed vascular bioreactor system for 7 days of dynamic culture following 3 days of static culture. The grafts were then transplanted into the common carotid artery of the same seven dogs from which ECs had been derived (two grafts were prepared for each dog with one as a backup; therefore, a total of 14 tissue-engineered blood vessels were prepared). At 1, 3, and 6 months post-transplantation, ultrasonography and contrast-enhanced computed tomography (CT) were used to check the patency of the grafts. Additionally, vascular grafts were sampled for histological and electron microscopic examination. Results Tissue-engineered, small-diameter vascular grafts can be successfully constructed using DAFPs and canine vascular ECs. Ultrasonographic and CT test results confirmed that implanted vascular grafts displayed good patency with no obvious thrombi. Six months after implantation, the grafts had been remodeled and exhibited a similar structure to normal arteries. Immunohistochemical staining showed that cells had evenly infiltrated the tunica media and were identified as muscular fibroblasts. Scanning electron microscopy showed that the graft possessed a complete cell layer, and the internal cells of the graft were confirmed to be ECs by transmission electron microscopy. Conclusions Tissue-engineered, small-diameter vascular grafts constructed using DAFPs and canine vascular ECs can be successfully transplanted to replace the canine common carotid artery. This investigation potentially paves the way for solving a problem of considerable clinical need, i.e., the requirement for small-diameter vascular grafts.

This paper proposes and tests a multilayer framework for simulating the network dynamics of inter-organizational coordination among interdependent infrastructure systems (IISs) in resilience planning. Inter-organizational coordination among IISs (such as transportation, flood control, and emergency management) would greatly affect the effectiveness of resilience planning. Hence, it is important to examine and understand the dynamics of coordination in networks of organizations within and across various systems in resilience planning. To capture the dynamic nature of coordination frequency and the heterogeneity of organizations, this paper proposes a multilayer network simulation framework enabling the characterization of inter-organizational coordination dynamics within and across IISs. In the proposed framework, coordination probabilities are utilized to approximate the varying levels of collaboration among organizations. Based on these derived collaborations, the simulation process perturbs intra-layer or inter-layer links and unveils the level of inter-organizational coordination within and across IISs. To test the proposed framework, the study examined a multilayer collaboration network of 35 organizations from five infrastructure systems within Harris County, Texas, based on the data gathered from a survey in the aftermath of Hurricane Harvey. The results indicate that prior to Hurricane Harvey: (1) coordination among organizations across different infrastructure systems is less than the coordination within the individual systems; (2) organizations from the community development system had a low level of coordination for hazard mitigation with organizations in flood control and transportation systems; (3) achieving a greater level of coordination among organizations across infrastructure systems is more difficult and would require a greater frequency of interaction (compared to within-system coordination). The results show the capability of the proposed multilayer network simulation framework to examine inter-organizational coordination dynamics at the system level (e.g., within and across IISs). The assessment of inter-organizational coordination within and across IISs sheds light on important organizational interdependencies in IISs and leads to recommendations for improving the resilience planning process.

In order to better understand the effect of social media in the dissemination of scholarly articles, employing the daily updated referral data of 110 PeerJ articles collected over a period of 345 days, we analyze the relationship between social media attention and article visitors directed by social media. Our results show that social media presence of PeerJ articles is high. About 68.18% of the papers receive at least one tweet from Twitter accounts other than @PeerJ, the official account of the journal. Social media attention increases the dissemination of scholarly articles. Altmetrics could not only act as the complement of traditional citation measures but also play an important role in increasing the article downloads and promoting the impacts of scholarly articles. There also exists a significant correlation among the online attention from different social media platforms. Articles with more Facebook shares tend to get more tweets. The temporal trends show that social attention comes immediately following publication but does not last long, so do the social media directed article views.

Studying precipitation diurnal variation characteristics is crucial to better understand their formation mechanism. Moreover, it is fundamental in assessing regional climate variability and to validate the effectiveness of cloud and precipitation parameterization schemes in weather and climate models. Based on observational data from 2008 to 2017, this manuscript objective is to assess the aerosol effects on summer precipitation on an hourly-scale diurnal variation basis in the Beijing metropolitan area. The results put in evidence that the precipitation frequency and duration on polluted days were 25.0% and 14.8% lower with respect to clean days. No significant differences were observed in total daily accumulated rainfall between clear and polluted days. While heavy precipitation mean intensity on polluted days increased by 13.5%, which is potentially linked with aerosol microphysical effects. Note that precipitation occurs earlier on polluted days, with peak time of 1 ∼ 2-hour in advance compared with clean days over the urban areas of Beijing, which may be primarily ascribed to the influence of advanced turning local circulation of mountain-valley breezes and urban heat island circulation due to aerosol-radiation effects.

Oxidative stress has been demonstrated to be involved in the etiology of alcoholic fatty liver disease (AFLD). Previous studies had demonstrated that resveratrol (RES) could reduce oxidative stress by different mechanisms. However, the effect of RES on alcohol-induced fatty liver remains unclear. In the present study, a total of 48 male SD rats were divided into three groups: Control, AFLD, and RES groups. Rats were administered with either nothing or 65% vol/vol alcohol (5 ml/kg/day in the first three days, and then 10 ml/kg/day in the following days) with or without RES supplementation (250 mg/kg/day) for 4 weeks. Blood and liver tissue samples were collected and subjected to biochemical assays, histological examination, Western blot, and mitochondrial radical oxygen species (ROS) assays. China’s scientific output has risen precipitously over the past decade; it is now the world’s second-largest producer of scientific papers, behind only the United States. The quality of China’s research is also on the rise (Van Noorden, 2016). The online visibility and impact of China’s research are also important issues worth exploring. In this study, we investigate the altmetric performance of publications in the field of Biotechnology and Applied Microbiology and published by authors from Chinese affiliations. We find that papers published by those authors from Chinese affiliations have much lower visibility on the social web than articles from other countries, when there is no significant difference for the citations. Fewer of China’s publications get tweeted, and those tweeted publications attract less social attention. A geographical analysis of tweeters shows that scholarly articles get most of their social attention from the authors’ home countries, a finding that is also confirmed by correlation and regression analysis. This situation, which is unfavorable for researchers from Chinese affiliations, is caused, in part, by the inaccessibility of mainstream social networking platforms in mainland China.

The positive definite solutions of the nonlinear matrix equation are discussed. A necessary and sufficient condition for the existence of positive definite solutions for this equation is derived. Then, the uniqueness of the Hermitian positive definite solution is studied based on an iterative method proposed in this paper. Lastly the perturbation analysis for this equation is discussed.

Mitochondria play diverse roles in mammalian physiology. The architecture, activity, and physiological functions of mitochondria in oocytes are largely different from those in somatic cells, but the mitochondrial proteins related to oocyte quality and reproductive longevity remain largely unknown. Here, using whole-exome sequencing data from 1,024 women (characterized by oocyte maturation arrest and degenerated or morphologically abnormal oocytes) and 2,868 healthy controls, we performed a population and gene-based burden test for mitochondrial genes and identified a candidate gene, cytochrome c oxidase assembly protein 15 (COX15). We report that biallelic COX15 pathogenic variants cause human oocyte ferroptosis and female infertility in a recessive inheritance pattern. COX15 variants impaired mitochondrial respiration in Saccharomyces cerevisiae and led to reduced protein levels in HeLa cells. Oocyte-specific deletion of Cox15 led to impaired Fe2+ and reactive oxygen species homeostasis that caused mitochondrial dysfunction and ultimately sensitized oocytes to ferroptosis. In addition, ferrostatin-1 (an inhibitor of ferroptosis) could rescue the oocyte ferroptosis phenotype in vitro and ex vivo. Our findings not only provide a genetic diagnostic marker for oocyte development defects but also expand the spectrum of mitochondrial disorders to female infertility and contribute to unique insights into the role of ferroptosis in human oocyte defects.
