Abstract Background Gastric cancer is a significant global malignancy with poor prognosis. Although the emergence of immune checkpoint inhibitors (ICIs) prolonged the duration of survival, resistance and progression are inevitable. We aim to evaluate the effectiveness of programmed death-1 (PD-1) inhibitors in immunotherapy beyond progression (IBP). Method We divided the advanced gastric cancer patients who received two lines immunotherapy into same regimen group (with same PD-1 inhibitor regime after IBP) and different regimen group (with different PD-1 inhibitor regime after IBP). Statistical analysis conducted to compare patient characteristics and evaluate survival differences between groups. Result The clinical outcome analysis showed that the same PD-1 inhibitor regime seemed to exhibit a higher disease control rate (DCR) (51.8% vs. 29.2%, P = 0.062), significantly prolonged progression-free survival 2 (PFS2) (162 vs. 75 days, P = 0.001) and overall survival (OS) (312 vs. 166 days, P = 0.022) when compared with those of cross line. In the multivariate analysis, when using different regimen group as reference, the same regimen group was found to be independently associated with improved PFS2 [hazard ratio (HR) = 0.467, 95% confidence interval (CI): 0.267–0.816, P = 0.008] and OS (HR = 0.508, 95%CI: 0.278–0.927, P = 0.027). Conclusion Continuation of the same type of PD-1 inhibitor regime in IBP shows clinical benefits and represents a promising therapeutic approach.

Abstract The sluggish kinetics of Volmer step in the alkaline hydrogen evolution results in large energy consumption. The challenge that has yet well resolved is to control the water adsorption and dissociation. Here, we develop biaxially strained MoSe2 three dimensional nanoshells that exhibit enhanced catalytic performance with a low overpotential of 58.2 mV at 10 mA cm−2 in base, and long-term stable activity in membrane-electrode-assembly based electrolyser at 1 A cm−2. Compared to the flat and uniaxial-strained MoSe2, we establish that the stably adsorbed OH engineer on biaxially strained MoSe2 changes the water adsorption configuration from O-down on Mo to O-horizontal on OH* via stronger hydrogen bonds. The favorable water dissociation on 3-coordinated Mo sites and hydrogen adsorption on 4-coordinated Mo sites constitute a tandem electrolysis, resulting in thermodynamically favorable hydrogen evolution. This work deepens our understanding to the impact of strain dimensions on water dissociation and inspires the design of nanostructured catalysts for accelerating the rate-determining step in multi-electron reactions.

Abstract Myocardial infarction (MI) is one of the leading causes of death. This is attributed to the dramatic changes in the myocardial microenvironment post-MI. Therefore, effective intervention in the early stages of MI is significant for inhibiting its progression and improving cardiac function. Herein, an injectable composite hydrogel scaffold (Gel-pBP@Mg) was developed by integrating magnesium (Mg)-modified black phosphorus nanosheets (pBP@Mg) into a reactive oxygen species-responsive hydrogel (Gel). This loose and porous Gel provides a natural platform for carrying pBP@Mg. In situ, sustained release of pBP@Mg is achieved via responsive ROS degradation in the infarct site. The high ROS reactivity of Black phosphorus nanosheets (BPNSs) can effectively inhibit the progression of oxidative stress in the infarct area and reduce inflammatory response by down-regulating the NF-κB pathway. Additionally, the sustained release of Mg loaded on the surface of BPNSs can effectively promote angiogenesis in MI, which is significant for the long-term prognosis after infarction. Our developed Gel-pBP@Mg effectively blocked infarction progression and improved myocardial function by sustainably inhibiting the “oxidative stress-inflammation” reaction chain and pro-angiogenesis. This study reveals Gel-pBP@Mg composite therapeutic potential in treating MI through In vitro and In vivo studies, providing a promising modality for MI treatment. Graphical abstract

The debate continues whether the expression of the same rare genetic mutation in multiple primary lung cancers suggests intrapulmonary metastasis or truly multiple primary lung cancers. We report a case of a 54-year-old female patient who presented with multiple nodules in the right lung discovered during a routine examination, persisting for six months. The patient had three central lesions in the right lung’s upper, middle, and lower lobes. She underwent thoracoscopic wedge resection, and the postoperative pathology reported two minimally invasive adenocarcinoma and one adenocarcinoma in situ. Interestingly, genetic testing for lung cancer-related driver genes revealed the presence of the rare RET mutation in all three nodules. This led us to speculate that these nodules might have the exact origin rather than being multiple primaries. To verify this hypothesis, we conducted further testing on these nodules, including whole-exome sequencing (The NGS data was generated from the Illumina sequencing platform by Novogene Co. Ltd, Beijing, China). The results indicated that although all three nodules expressed the RET mutation, there was significant heterogeneity in the gene mutations (differences in the number of cellular mutations, substitution composition levels, and clustering analysis of the three nodules). Thus, the patient was considered to have multiple primary lung cancers. In such cases, whole-exome sequencing can distinguish whether the nodules have the exact origin.

Abstract Three‐dimensional (3D) current collectors (CCs) have emerged as an effective strategy to inhibit dendrites and ensure the safety of lithium (Li) metal anodes. However, existing 3D CCs are generally too heavy (typically tens of mg cm−2) or too thick (tens to hundreds of micrometers), making large‐scale production and further application challenging. Additionally, the use of single‐component 3D CCs, whether electrically active or inert, only exhibits limited effects on stabilizing Li anodes. Here, we present a scalable screen‐printing technique for the synthesis of ultralight (~0.4 mg cm−2) and ultrathin (~0.54 μm) SiO2 grids on Cu foil to regulate both the vertical electric field and Li‐ion concentration by forming an electrically active/inert dual‐function architecture. This technology breaks the limitations of traditional 3D CCs in material/fabrication costs, weight, thickness and especially, scalability for large‐scale fabrication. By using this dual‐function architecture, our Cu@SiO2‐grid CCs (~8.31 mg cm−2), which are even lighter than the original Cu‐foil CCs (~8.85 mg cm−2), realize an ultra‐smooth anode surface without Li dendrites, and thus leads to an ultra‐long cyclic life of over 1500 h at 1 mA cm−2. The assembled Li metal batteries demonstrate excellent capacity retention of ~80% over 400 cycles at 1 C and ~ 76% over 250 cycles at 5 C, which highlight the promising 3D CCs for practical applications.

Automatic detection and recognition of safety helmet wearing based on video analysis is important to ensure production safety. It is inefficient to supervise whether workers wear safety helmets by manual means. With the advancement of deep learning, using computer vision to assist in the detection of safety helmet-wearing holds significant research and application value. However, complex environments and variable factors pose challenges in achieving accurate detection and recognition of safety helmet usage. Helmet-wearing detection methods are generally classified as traditional machine learning and deep learning methods. Traditional machine learning methods employ manually selected features or statistical features, resulting in poor model stability. Deep learning–based methods are further divided into “two-stage” and “one-stage” methods. The two-stage method has high detection accuracy but cannot achieve real-time detection, while the one-stage counterpart is the reverse. Achieving accuracy as well as real-time detection is an important challenge in the development of video-based helmet detection systems. Accurate and quick detection of helmets is essential for effective real-time monitoring of production sites. To address these challenges, this paper proposes DS-YOLOv5—a real-time helmet detection and recognition model based on the YOLOv5 models. The proposed model solves three main problems: First, insufficient global information extraction problem of convolutional neural network (CNN) models. Second, the lacking robustness of the deep SORT for multiple targets and occlusion problems in video scenes. Third, the inadequate feature extraction of multiscale targets. The DS-YOLOv5 model takes advantage of the improved Deep SORT multitarget tracking algorithm to reduce the rate of missed detections in multitarget detection and occlusion and increase the error tolerance in video detection. Further, a simplified transformer module is integrated into the backbone network to enhance the capture of global information from images and thus enhance feature learning for small targets. Finally, the bidirectional feature pyramid network is used to fuse multiscale features, which can better adapt to target scale changes caused by the photographic distance. The DS-YOLOv5 model was validated using the GDUT-HWD dataset by ablation and comparison experiments. In these experiments, the tracking capability of the improved Deep SORT is compared with the YOLOv5 model using the public pedestrian dataset MOT. The results of the comparison of the five one-stage methods and four helmet detection and recognition models revealed that the proposed model has better capability for dealing with occlusion and target scale. Further, the model achieved mean average orecision (mAP) of 95.5%, which is superior to that of the other helmet detection and recognition models.

Abstract The indiscriminate use of nitrogen fertiliser (NF) is a obstruction to improve soil quality and crop yields. However, the effect of biochar and NF on soil microbial ecosystem (SME) and crop yields is unknown. A five-year field experiment in China aimed to evaluate the effects of biochar and nitrogen fertiliser (NF) combination on soil structure, C-to-N ratio (CNR), microbial biomass, and spring maize yield. Biochar and NF were applied at different rates, and the combined application resulted in a soil solid–liquid–gas ratio closer to the ideal value. The use of biochar alone and in combination with NF significantly increased soil's C, N, and CNR. A moderate application of biochar and NF resulted in favourable biological and chemical properties of the soil. The application of biochar and NF at moderate levels led to an increase in SME, with the B8N150 producing the highest yield. The highest yield of B8N150 represents a 24.25% increase compared to the unfertilized control and a 9.04% increase compared to B0N150. Moderate use of biochar and NF could be beneficial in areas with similar climatic conditions.

Aiming at the problem of difficult crack detection in underground urban sewage pipelines, a lightweight sewage pipeline crack detection method based on sewage pipeline robots and improved YOLOv8n is proposed. The method uses pipeline robots as the equipment carrier to move rapidly and collect high-definition data of apparent diseases in sewage pipelines with both water and sludge media. The lightweight RGCSPELAN module is introduced to reduce the number of parameters while ensuring the detection performance. First, we replaced the lightweight detection head Detect_LADH to reduce the number of parameters and improve the feature extraction of modeled cracks. Finally, we added the LSKA module to the SPPF module to improve the robustness of YOLOv8n. Compared with YOLOv5n, YOLOv6n, YOLOv8n, RT-DETRr18, YOLOv9t, and YOLOv10n, the improved YOLOv8n has a smaller number of parameters of only 1.6 M. The FPS index reaches 261, which is good for real-time detection, and at the same time, the model also has a good detection accuracy. The validation of sewage pipe crack detection through real scenarios proves the feasibility of the proposed method, which has good results in targeting both small and long cracks. It shows potential in improving the safety maintenance, detection efficiency, and cost-effectiveness of urban sewage pipes.

Objective MicroRNA-related single-nucleotide polymorphisms (miR-SNPs) can alter microRNA (miRNA) expression profiles, thereby influencing the risk of rheumatic diseases. Herrin a case control study, six miR-SNPs in miRNA processing machinery genes, namely RAN (rs14035), XPO5 (rs11077), Dicer (rs3742330), GEMIN3 (rs197412), GEMIN4 (rs2740348), and TNRC6B (rs9623117), were genotyped to assess their correlation with the risk of systemic lupus erythematosus (SLE). Methods We included 119 patients with SLE and 130 healthy controls. The genotypes of the six miR-SNPs were determined using polymerase chain reaction (PCR). Serum cytokine levels were assessed using a cytometric bead array, and fluorescent probe technology was used to determine plasma reactive oxygen species (ROS) levels. Results The AA genotype of Dicer was correlated with a 0.566-fold decreased risk of SLE compared with that of the AG + GG genotype (odds ratio, 0.566; 95% CI, 0.342–0.935; p = .026), and the rs3742330 A allele was associated with a significantly decreased risk of SLE ( p = .035) compared with that of the rs3742330G allele. Additionally, AA genotype carriers exhibited lower levels of interleukin-6 (IL-6) in the blood ( p = .013). Subsequent analysis revealed increased ROS production in patients with SLE than that in the controls (621.042 ± 425.285 vs 499.966 ± 302.273, p = .011). Conclusion Our findings suggest that ROS generation participates in SLE pathogenesis. The identification of Dicer gene SNP rs3742330 as a potential modifier of SLE risk via mediating IL-6 overproduction suggests a potential avenue for targeted interventions to manage SLE and its associated immune dysregulation.

Immune Checkpoint Inhibitors (ICIs) have dramatically revolutionized the therapeutic approaches by which we treat a series of cancers accompanied by immune-related adverse events (irAEs). Herein, we reported an intrahepatic cholangiocarcinoma male patient with a history of ankylosing spondylitis developing pulmonary arterial hypertension (PAH) under ICI combined therapy with pembrolizumab and lenvatinib. The indirect measurement of cardiac ultrasound showed a pulmonary artery pressure (PAP) of 72mmHg after 21 three-week cycles of ICI combined therapy. The patient partially responded to the treatment of glucocorticoid and mycophenolate mofetil. The PAP decreased to 55mmHg 3 months after the ICI combined therapy was discontinued, but increased to 90mmHg after the ICI combined therapy was rechallenged. We treated him with adalimumab -an antitumor necrosis factor-alpha (ani-TNF-α) antibody- combined with glucocorticoid and immunosuppressants under lenvatinib monotherapy. The patient responded again with PAP decreasing to 67mmHg after 2 two-week cycles of adalimumab. Accordingly, we diagnosed him to have irAE-related PAH. Our findings supported the use of glucocorticoid disease-modifying antirheumatic drugs (DMARDs) as a treatment option in refractory PAH.

Abstract Purpose Recent studies have indicated some differences in the prognosis of patients with stage III-N2 lung adenocarcinoma, and the prognosis of patients with skip N2 lymph node metastasis (SKN2) is good. This study grouped patients with stage III-N2 lung adenocarcinoma by propensity score matching (PSM) to evaluate the impact of SKN2 on the prognosis of these patients. Methods The clinical data for patients who underwent radical lobectomy and had a postoperative pathological diagnosis of stage III-N2 lung adenocarcinoma at our centre from 2016 to 2018 were collected, and PSM was performed at a ratio of 1:1. Results A total of 456 patients were enrolled in this study. After PSM, 112 patients were included in the SKN2 group, and 112 patients were included in the non-SKN2 group. Significant progress has been achieved in the field of oriented object detection (OOD) in recent years. Compared to natural images, objects in remote sensing images exhibit characteristics of dense arrangement and arbitrary orientation while also containing a large amount of background information. Feature extraction in OOD becomes more challenging due to the diversity of object orientations. In this paper, we propose a semantic-driven rotational feature enhancement method, termed SREDet, to fully leverage the joint semantic and spatial information of oriented objects in the remote sensing images. We first construct a multi-rotation feature pyramid network (MRFPN), which leverages a fusion of multi-angle and multiscale feature maps to enhance the capability to extract features from different orientations. Then, considering feature confusion and contamination caused by the dense arrangement of objects and background interference, we present a semantic-driven feature enhancement module (SFEM), which decouples features in the spatial domain to separately enhance the features of objects and weaken those of backgrounds. Furthermore, we introduce an error source evaluation metric for rotated object detection to further analyze detection errors and indicate the effectiveness of our method. Extensive experiments demonstrate that our SREDet method achieves superior performance on two commonly used remote sensing object detection datasets (i.e., DOTA and HRSC2016).

Summary: Esophageal squamous-cell carcinoma (ESCC) is commonly treated with radiotherapy; however, radioresistance hinders its clinical effectiveness, and the underlying mechanism remains elusive. Here, we develop patient-derived xenografts (PDXs) from 19 patients with ESCC to investigate the mechanisms driving radioresistance. Using RNA sequencing, cytokine arrays, and single-cell RNA sequencing, we reveal an enrichment of cancer-associated fibroblast (CAF)-derived collagen type 1 (Col1) and tumor-cell-derived CXCL1 in non-responsive PDXs. Col1 not only promotes radioresistance by augmenting DNA repair capacity but also induces CXCL1 secretion in tumor cells. Additionally, CXCL1 further activates CAFs via the CXCR2-STAT3 pathway, establishing a positive feedback loop. Directly interfering with tumor-cell-derived CXCL1 or inhibiting the CXCL1-CXCR2 pathway effectively restores the radiosensitivity of radioresistant xenografts in vivo. Collectively, our study provides a comprehensive understanding of the molecular mechanisms underlying radioresistance and identifies potential targets to improve the efficacy of radiotherapy for ESCC.

Abstract Purpose In 2015, the World Health Organization renamed mucinous bronchioloalveolar adenocarcinoma as pulmonary invasive mucinous adenocarcinoma (IMA). Due to its low incidence and unclear prognosis with surgical treatment, previous studies have presented opposing survival outcomes. We aimed to investigate the differences in surgical prognosis and prognosis-related risk factors by comparing IMA with non-mucinous invasive adenocarcinoma (NMA). Methods A total of 20,914 patients diagnosed with IMA or NMA from 2000 to 2014 were screened from the Surveillance, Epidemiology, and End Results database. The screened patients were subjected to propensity score matching (PSM) in a 1:4 ratio to explore the survival differences between patients with IMA and NMA and the factors influencing prognosis. Phase‐change materials are of great interest in solving mismatch between energy supply and demand. However, the vulnerability of solid–liquid phase‐change materials to leakage during the phase‐change process limits their development and application in practice. Herein, the enhancement of the shape stability of phase‐change materials is achieved through an organic–inorganic composite. The mixture of phenolic resin and polyethylene glycol forms a homogeneous solution based on their excellent mutual solubility and is able to be adsorbed into the pores of the expanded graphite by means of a vacuum‐impregnation strategy. The 3D cross‐linked network structure of phenolic resin is formed within the pores of expanded graphite, enabling in situ encapsulation of polyethylene glycol. It is worth noting that the curing reaction of phenolic resin is able to be initiated by heating up without the addition of any curing agent and other auxiliary materials. A thermal conductivity enhancement of 20 times than that of polyethylene glycol is achieved along with a photothermal conversion efficiency of 63.72% and with a latent heat of 134.94 J g−1 without leakage.

Non-small-cell lung cancer (NSCLC) is the most common cancer in the world. In recent years, the incidence of synchronous multiple primary lung cancer (SMPLC) has gradually increased. Surgery is the preferred method to treat these patients. The management of SMPLC patients who cannot tolerate surgical treatment is controversial. We report a rare case in which a 70-year-old Chinese woman with no history of smoking had three primary lung adenocarcinoma lesions. Two lesions had epidermal growth factor receptor (EGFR) exon 19 deletion mutations, and one lesion had the L858R mutation. After first-generation EGFR–tyrosine kinase inhibitor (TKI) treatment, the three lesions all showed a good response until disease progression. After the corresponding drug treatments were given based on the different drug resistance mechanisms, good responsiveness was shown in each lessions. This case suggests that in the treatment of SMPLC, it is necessary to learn the molecular-biological information of each lesion due to the differences thereof, and a targeted treatment regimen should be developed on this basis.

BackgroundIt is still unclear whether stage I lung adenocarcinoma patients with tumour spread through air spaces (STAS) can benefit from postoperative adjuvant chemotherapy (ACT) after lobectomy. This study investigated the effect of ACT on the postoperative survival of patients with stage I (STAS+) lung adenocarcinoma.MethodsWe retrospectively analysed the clinical data of stage I (STAS+) invasive lung adenocarcinoma patients who underwent lobectomy in the Department of Thoracic Surgery of our hospital from January 1, 2013 to January 1, 2016. Propensity score matching (PSM) was performed to group patients to investigate whether ACT could lead to better prognosis of patients.ResultsA total of 593 patients with stage I (STAS+) lung adenocarcinoma were enrolled. The study after PSM included 406 patients. Kaplan–Meier survival analysis showed the experimental group had a better 3-year recurrence-free survival (RFS) rate (p = 0.037) and the 5-year RFS rate (p = 0.022) than the control group. It also had higher 5-year overall survival (p = 0.017). The multivariate analysis by Cox proportional hazard regression model showed that stage I STAS+ lung adenocarcinoma patients with lymphatic vessel invasion (HR: 1.711, 95% CI: 1.052-2.784; p = 0.045), vascular invasion (HR: 5.014, 95% CI: 3.154-7.969; p < 0.001), and visceral pleural invasion (HR: 2.086, 95% CI: 1.162-3.743; p = 0.014), and without ACT (HR: 1.675, 95% CI: 1.043-2.689; p = 0.033) had a significant survival disadvantage.ConclusionACT can boost the postoperative survival of patients with stage I (STAS+) lung adenocarcinoma.

Ultra-dense small cell networks (UD-SCNs) will be an integral part of next generation network (NGN). How to deal with serious interference is one of the important challenges in a UD-SCN. In this paper, the cooperative interference management problem for a UD-SCN is explored by allowing small cell base stations (SBSs) to collaborate with their neighbors. The proposed coalitional structure generation among SBSs can mitigate the co-tier interference within a coalition, thus improving the network capacity. Specifically, a cooperative scheme among the neighboring SBSs is formulated as a coalitional structure generation with characteristic forms. Furthermore, the relative sub-channel resources are allocated in the process of cluster generation. Compared with the existing SBS cooperative schemes, the novelty of the proposed SBS cooperation method, which uses a hierarchical clustering algorithm (SC-HCA) to compute the pairs of members, is demonstrated. The computation can enhance the efficiency of the proposed algorithm and is especially suitable for UD-SCN scenarios with tens and even hundreds of cells. The simulation results show that the proposed SC-HCA achieves a 422.13% system data rate improvement relative to that of the non-cooperative scheme in a UD-SCN scenario.

A pulse-coupled neural network (PCNN) is a promising image segmentation approach that requires no training. However, it is challenging to successfully apply a PCNN to medical image segmentation due to common but difficult scenarios such as irregular object shapes, blurred boundaries, and intensity inhomogeneity. To improve this situation, a novel framework incorporating fuzzy connectedness (FC) is proposed. First, a comparative study of the traditional PCNN models is carried out to analyze the framework and firing mechanism. Then, the characteristic matrix of fuzzy connectedness (CMFC) is presented for the first time. The CMFC can provide more intensity information and spatial relationships at the pixel level, which is helpful for producing a more reasonable firing mechanism in the PCNN models. Third, by integrating the CMFC into the PCNN framework models, a construction scheme of FC-PCNN models is designed. To illustrate this concept, a general solution that can be applied to different PCNN models is developed. Next, the segmentation performances of the proposed FC-PCNN models are evaluated by comparison with the traditional PCNN models, the traditional segmentation methods, and deep learning methods. The test images include synthetic and real medical images from the Internet and three public medical image datasets. The quantitative and visual comparative analysis demonstrates that the proposed FC-PCNN models outperform the traditional PCNN models and the traditional segmentation methods and achieve competitive performance to the deep learning methods. In addition, the proposed FC-PCNN models have favorable capability to eliminate inference from surrounding artifacts.

BackgroundSynchronous multiple ground-glass nodules (SMGGNs) in synchronous multiple lung cancers are associated with specific imaging findings. It is difficult to distinguish whether multiple nodules are primary tumors or metastatic lesions in the lungs. The need for PET/CT and contrast-enhanced brain MRI for these patients remains unclear. This study investigated the necessity of these two imaging examinations for SMGGN patients by means of retrospective analysis.MethodsSMGGN patients who were diagnosed and treated in our hospital from October 2017 to May 2020 and underwent whole-body PET/CT(Cranial excepted) and/or contrast-enhanced brain MRI+DWI were enrolled in this study. We analyzed the imaging and clinical characteristics of these patients to evaluate SMGGN patients’ need to undergo whole-body PET/CT and brain MRI examination.ResultsA total of 87 SMGGN patients were enrolled. 51 patients underwent whole-body PET/CT examinations and did not show signs of primary tumors in other organs, metastatic foci in other organs, or metastasis to surrounding lymph nodes. 87 patients underwent whole-brain MRI, which did not reveal brain metastases but did detect an old cerebral infarction in 23 patients and a new cerebral infarction in one patient. 87 patients underwent surgical treatment in which 219 nodules were removed. All nodules were diagnosed as adenocarcinoma or atypical adenomatous hyperplasia. No lymph node metastasis was noted.ConclusionFor SMGGN patients, PET/CT and enhanced cranial MRI are unnecessary for SMGGNs patients, but from the perspective of perioperative patient safety, preoperative MRI+DWI examination is recommended for SMGGNs patients.

BackgroundLung cancer is a malignant tumor that has the highest morbidity and mortality rate among all cancers. Early diagnosis of lung cancer is a key factor in reducing mortality and improving prognosis.MethodsIn this study, we performed CTC next-generation sequencing (NGS) in early-stage lung cancer patients to identify lung cancer-related gene mutations. Meanwhile, a serum liquid chromatography-tandem mass spectrometry (LC-MS) untargeted metabolomics analysis was performed in the CTC-positive patients. To screen potential diagnostic markers for early lung cancer.Results62.5% (30/48) of lung cancer patients had ≥1 CTC. By CTC NGS, we found that > 50% of patients had 4 commonly mutated genes, namely, NOTCH1, IGF2, EGFR, and PTCH1. 47.37% (9/19) patients had ARIDH1 mutations. Additionally, 30 CTC-positive patients and 30 healthy volunteers were subjected to LC-MS untargeted metabolomics analysis. We found 100 different metabolites, and 10 different metabolites were identified through analysis, which may have potential clinical application value in the diagnosis of CTC-positive early-stage lung cancer (AUC >0.9).ConclusionsOur results indicate that NGS of CTC and metabolomics may provide new tumor markers for the early diagnosis of lung cancer.

Abstract The treatment regimen for the eradication of Helicobacter pylori may be best when therapy is susceptibility guided. However, it is unrealistic to use a strategy based on susceptibility testing to prioritize therapy for every patient in China. Empirical therapy of H. pylori is still widely used. The study was designed to discuss the best first‐line treatment regimen depending on empirical therapy. The focal point of the study was the optimal length of the therapy. Also, the selection of antibiotics was discussed in the article. This was a prospective, randomized, non‐inferiority trial. H. pylori‐infected patients who have no previous eradication therapy were randomly assigned to the following: 20 mg of rabeprazole, 1000 mg of amoxicillin, 500 mg of clarithromycin, and 220 mg of bismuth potassium citrate (BACPPI), administered twice a day for 10 or 14 days. The efficacy, side effects, and remission rate of clinical symptoms were determined. A total of 240 subjects were included in the study. The eradication rate with 14 and 10 days was essentially identical in both intention‐to‐treat (90.83% [95% CI, 86%–96%] vs. 87.50% [95% CI, 82%–93%]) and per‐protocol (94.78% [95% CI, 91%–99%] vs. 92.11% [95% CI, 87%–97%]) analyses. Loss of appetite and belching symptoms were significantly better in the BACPPI‐10 group than those in the control group after treatment. Side effects were generally mild and similar between groups. Our results showed that a 10‐day amoxicillin–clarithromycin‐containing bismuth quadruple therapy may be recommended for the primary empirical treatment of H. pylori infection in Beijing, China.

Chronic psychological stress is associated with an increased risk for relapse of inflammatory bowel diseases (IBD) and impedes the treatment of this condition. However, the impact of stress on the risk of IBD onset remains unclear. The goal of the present study was to examine whether chronic unpredictable mild stress (CUMS) could initiate or aggravate the onset of colon inflammation in rats which, in turn, would be capable of triggering bowel disease. We found that CUMS exposure increased infiltration of CD-45 positive cells and MPO activity, as well as augmented the expression of the inflammatory cytokines, IFN-γ and IL-6 within the colon of these rats. In addition, CUMS treatment changed the composition and diversity of gut microbiota and enhanced intestinal epithelial permeability, indicating the presence of a defect in the intestinal barrier. This CUMS-induced disruption of mucosal barrier integrity was associated with a reduction in expression of the tight junction protein, occludin 1, and an inhibition in mucosal layer functioning via reductions in goblet cells. Results from bacterial cultures revealed an increased presence of bacterial invasion after CUMS treatment as compared with that observed in controls. Thus, our data indicate that CUMS treatment induces alterations of the fecal microbiome and intestinal barrier defects, which facilitates bacterial invasion into colonic mucosa and further exacerbates inflammatory reactions within the colon. Accordingly, chronic stress may predispose patients to gastrointestinal infection and increase the risk of inflammation-related gut diseases.

The Microsoft HoloLens is a head-worn mobile augmented reality device that is capable of mapping its direct environment in real-time as triangle meshes and localize itself within these three-dimensional meshes simultaneously. The device is equipped with a variety of sensors including four tracking cameras and a time-of-flight (ToF) range camera. Sensor images and their poses estimated by the built-in tracking system can be accessed by the user. This makes the HoloLens potentially interesting as an indoor mapping device. In this paper, we introduce the different sensors of the device and evaluate the complete system in respect of the task of mapping indoor environments. The overall quality of such a system depends mainly on the quality of the depth sensor together with its associated pose derived from the tracking system. For this purpose, we first evaluate the performance of the HoloLens depth sensor and its tracking system separately. Finally, we evaluate the overall system regarding its capability for mapping multi-room environments.
