The effect of dietary niacinamide (NAM) on the growth performance and liver lipid metabolism of swamp eel have been poorly investigated. Therefore, a 120-day feeding trial was conducted to provide different levels of NAM in diet and an untargeted lipidome analysis was performed to address this subject. We found that dietary NAM improved the growth performance of swamp eel. It also decreased the contents of glucose and lipids, including glucose, glycogen, TG, and NEFA, as well as fat accumulation in liver, but increased HDL-C content. We speculated that NAM inhibited lipid accumulation in liver by decreasing the key enzyme activity of LPL, HL and LPS. Fatty acid profile analysis showed that the contents of MUFA and PUFA, such as palmitoleic acid, oleic acid, and eicosapentaenoic acid were significantly increased, while the contents of SFA, such as myristic acid, stearic acid, and palmitic acid were decreased in liver by dietary NAM. Lipidome analysis also indicated that the majority of differential lipid metabolites belonged to glycerophospholipids and sphingolipids, which were enriched in the pathways of insulin resistance, adipocytokine signaling pathway, cholesterol metabolism, fat digestion and absorption, lipid and atherosclerosis, and lipid cell lipolysis regulation. These findings gave scientific evidences and novel insights into the metabolic responses to dietary NAM in swamp eel, and provided basic knowledge for using NAM as a feed additive in swamp eel production.

Gonadotropin-inhibitory hormone (GnIH) is a hypothalamic neuropeptide known for inhibiting gonadotropin release. In teleosts, it is also referred to as LPXRFa. While the effects of LPXRFa on gonadotropin synthesis and release, and its impact on pituitary function in teleosts have been widely studied, its specific mechanism in fish reproduction remains relatively unexplored. In this paper, LPXRFa and LPXRFa-R genes were isolated from the swamp eel brain to assess the roles of LPXRFa in the swamp eel. Their ORFs were 612 bp and 1284 bp, encoding proteins of 203 and 427 amino acids, respectively. The LPXRFa gene encodes a polypeptide possibly cleaved into three mature peptides (LPXRFa-1, −2, and −3). The LPXRFa-R receptor protein comprises seven conserved transmembrane domains. Tissue distribution analysis revealed that LPXRFa-R expression was the highest in the hypothalamus, while LPXRFa showed the highest expression in the rest of brain, with little to no expression in other tissues. The role of LPXRFa in the reproduction of the swamp eel was investigated by studying the effect of intraperitoneal injection with each of the three peptides. The intraperitoneal injection of LPXRFa-2 down-regulated serum FSH and LH levels, while that of LPXRFa-3 down-regulated serum FSH but up-regulated LH levels. Additionally, both LPXRFa-2 and LPXRFa-3 increased the expression levels of fshr, cbx3, dmrt1, gsdf, sox9al, amh, foxl2, and rab37 in the ovaries. Moreover, LPXRFa-2 also increased the ovarian expression of lhr, while LPXRFa-3 had no effect on it. Except upregulating cbx3 and foxl2 expression, LPXRFa-1 neither impacted the serum FSH and LH levels, nor did it alter the expression of other gonadal and reproductive-related genes. In summary, our findings provide the first evidence of a LPXRFa/LPXRFa-R system in the order Synbranchiformes and suggest its involvement in sex differentiation, gonadal development, and sexual reversal in the swamp eel.

The aim of this study was to examine the effects of dietary protein and lipid levels on the growth performance and homeostasis of the intestinal flora in Paramisgurnus dabryanus. An 8-week 3 × 3 two-factorial experiment was conducted to investigate the interaction between dietary crude protein (CP: 30%, 35%, 40%) and ether extract (EE: 6%, 10%, 14%) on the growth rate and the intestinal microflora of P. dabryanus. Optimal carbohydrate level in diets could improve growth and feed efficiency，whereas HC (high carbohydrate) diets usually induce the liver injury in reared fish. While SA (sodium acetate) has demonstrated its potential to alleviate liver injury in mammals, the mechanism underlying its impact on liver health in fish remains elusive. This study aims to elucidate the effect of dietary SA on liver injury in the female Monopterus albus. In our study, groups of M. albus were fed with different diets: NC (normal, 20% carbohydrate), HC (high carbohydrate, 32% carbohydrate) and HC+A (HC supplemented with 0.2% SA) diets, in separate, for a consecutive period of 56 days. An efficient metabolism of mammals in the intestine is a crucial factor in their dietary starch utilization. Swamp eel (Monopterus albus), a carnivorous fish, has shown a remarkable ability to utilize dietary starch efficiently. However, the specific mechanisms underlying this efficient starch metabolism in the intestine remain poorly understood. Thus, we conducted a study in which juvenile swamp eels (average weight: 12.10 ± 0.03 g) were fed diets with two different starch levels (20 % -NF and 32 % -HF) for a period of 56 days. Subsequently, the intestine and digesta were extracted for analysis. This study showed that there was no significant variety on the activity of digestive enzyme and histological structure in anterior intestine of fish between HF group and NF group. However, HF diets down-regulated the expression of all differentially expressed genes of glycine--tRNA ligase concerned to damage in the most significantly enriched GO function of glycyl-tRNA aminoacylation. Besides, the HF diets significantly increased the genus of Cetobacterium with the function of producing acetic acid. Consistently, the HF diets significantly increased the concentration of short chain fatty acids (SCFA), particularly acetic acid, in the posterior intestine digesta of swamp eel. In summary, our study argued that the enhanced efficiency in utilizing dietary starch by the swamp eel might be attributed to the increased abundance of Cetobacterium and the subsequent production of acetic acid in the posterior intestine.

To understand the effects of vitamin A on lipid deposition in rice field eels, integrated liver transcriptome and metabolome were conducted and the changes in the genes and metabolites were assessed. Three groups of rice field eel were fed with 0, 200, and 16,000 IU/kg vitamin A supplementations in their diets for 70 days. The total lipid content in the whole body of the rice field eels was significantly increased with the vitamin A supplementations (p < 0.05). Comparative transcriptome analysis revealed 14 pathways and 46 differentially expressed genes involved in lipid metabolism. Sphingolipid metabolism, glycerolipid metabolism, primary bile acid biosynthesis and steroid hormone biosynthesis were significantly enriched pathways. In these pathways, three differential genes phospholipid phosphatase 1a (PLPP1a), phospholipid phosphatase 2b (PLPP2b), cytochrome P450 21a2 (CYP21a2) were consistent with the change trend of lipid content, and the other three differential genes aldo-keto reductase family 1 member D1 (AKR1D1), uridine diphosphate glucuronic acid transferase 1a1 (UGT1a1), cytochrome P450 1a (CYP1a) were opposite. Metabolomic analysis revealed that primary bile acid biosynthesis, sphingolipid metabolism, steroid hormone biosynthesis and biosynthesis of unsaturated fatty acids were all critical for rice field eel metabolic changes in response to vitamin A. Six important differential metabolites (eicosapentaenoic acid, sphinganine, 11-beta-hydroxyprogesterone, hydroxyeicosatetraenoic acid, cholic acid, and glycochenodeoxycholate) were identified and have provided new insights into how vitamin A regulates lipid deposition. Integrated transcriptome and metabolome analyses revealed that primary bile acid biosynthesis was the only remarkably enriched pathway in both the transcriptome and metabolome while that sphingosine was the main metabolite. Based on the above results, we have concluded that vitamin A promotes lipid deposition in the rice field eel through the primary bile acid synthesis pathway, and lipid deposits are widely stored in cell membranes, mainly in the form of sphingosine. These results will provide reference data to help improve our understanding of how vitamin A regulates lipid metabolism.

Lipid is an important source of energy in fish feeds, and the appropriate fat content can improve the efficiency of protein utilization. However, excessive lipid content in the feed can lead to abnormal fat deposition in fish, which has a negative effect on the growth of fish. Therefore, the effects of feed lipid levels on swamp eel were studied. Essential functional genes were screened using transcriptomics. We divided 840 fish into seven groups (four replicates). A mixture of fish and soybean oils (1:4), 0%, 2%, 4%, 6%, 8%, 10%, and 12% was added to the basic feed were named groups one to seven (L1-L7), respectively. Isonitrogenous diets were fed swamp eel for 10 weeks. Growth performance, visceral index, nutritional components, and biochemical indexes were measured and analyzed. Livers of the 0%, 6%, and 12% groups were subjected to transcriptome sequencing analysis. The results of our study showed that: the suitable lipid level for the growth of swamp eel was 7.03%; the crude fat content of whole fish, liver, intestine, muscle, and skin increased with the increase of lipid level, with some significant difference, and excess fat was deposited in skin tissue; triglyceride, total cholesterol, and free fatty acid contents increased with the increase of feed lipid level. High-density lipoprotein levels in the L3 and L4 groups were higher than in the other groups. Blood glucose concentrations in the L5, L6, and L7 groups increased; the liver tissue structure was damaged when the lipid level was too high. two-hundred-and-twenty-eight differentially expressed genes were found. Several critical pathways regulating glucose metabolism and energy balance (e.g., glycerolipid metabolism, glycolysis synthesis, degradation of ketone bodies, and Janus Kinase/Signal Transducer and Activator of Transcription signaling pathway) were enriched in swamp eel compared with the Kyoto Encyclopedia of Genes and Genomes (KEGG) database. Suitable lipid levels (7.03%) can promote the growth of swamp eel, and excessive lipid levels can cause elevated blood lipids and lead to liver cell damage. Regulatory mechanisms may involve multiple metabolic pathways for glucose and lipid metabolism in eels. This study provides new insights to explain the mechanism of fat deposition due to high levels of lipid and provides a basis for the production of efficient and environmentally friendly feed for swamp eel.

A high-carbohydrate diet lowers the rearing cost and decreases the ammonia emission into the environment, whereas it can induce liver injury, which can reduce harvest yields and generate economic losses in reared fish species. Macroalgae Saccharina japonica (SJ) has been reported to improve anti-diabetic, but the protective mechanism of dietary SJ against liver injury in fish fed a high-carbohydrate diet has not been studied. Therefore, a 56-day nutritional trial was designed for swamp eel Monopterus albus, which was fed with the normal diet [20% carbohydrate, normal carbohydrate (NC)], a high carbohydrate diet (32% carbohydrate, HC), and a HC diet supplemented with 2.5% SJ (HC-S). The HC diet promoted growth and lowered feed coefficient (FC), whereas it increased hepatosomatic index (HSI) when compared with the NC diet in this study. However, SJ supplementation increased iodine contents in muscle, reduced HSI, and improved liver injury, such as the decrease of glucose (GLU), total bile acid (TBA), and alanine aminotransferase (ALT) in serum, and glycogen and TBA in the liver. Consistently, histological analysis showed that SJ reduced the area of lipid droplet, glycogen, and collagen fiber in the liver (p < 0.05). Thoroughly, the underlying protective mechanisms of SJ supplementation against HC-induced liver injury were studied by liver transcriptome sequencing coupled with pathway analysis. The Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of the differentially expressed genes (DEGs), such as the acetyl-coenzyme A synthetase (acss1), alcohol dehydrogenase (adh), interferon-induced protein with tetratricopeptide repeats 1 (ifit1), aldo-keto reductase family 1 member D1 (akr1d1), cholesterol 7-alpha-monooxygenase (cyp7a1), and UDP-glucuronosyltransferase (ugt), indicated that the pathway of glycolysis/gluconeogenesis was the main metabolic pathway altered in the HC group compared with the NC group. Meanwhile, hepatitis C, primary BA biosynthesis, and drug metabolism-cytochrome P450 were the three main metabolic pathways altered by SJ supplementation when compared with the HC group. Moreover, the BA-targeted metabolomic analysis of the serum BA found that SJ supplementation decreased the contents of taurohyocholic acid (THCA), taurochenodeoxycholic acid (TCDCA), taurolithocholic acid (TLCA), nordeoxycholic acid (NorDCA), and increased the contents of ursocholic acid (UCA), allocholic acid (ACA), and chenodeoxycholic acid (CDCA). In particular, the higher contents of UCA, ACA, and CDCA regulated by SJ were associated with lower liver injury. Overall, these results indicate that the 2.5% supplementation of SJ can be recommended as a functional feed additive for the alleviation of liver injury in swamp eel-fed high-carbohydrate diets.

This study aimed to analyze the intestinal flora of male versus female swamp eels, which have the unique characteristic of sex reversal. Same-aged swamp eels from the same parents, kept under the same conditions in terms of environment, diet, etc., were used as the study materials. After breeding for 1 year, 18 healthy swamp eels (nine males and nine females), weighing 39.4 ± 2.3 g, were selected. The intestinal contents of three swamp eels of the same sex were combined and labeled as follows: FM1-FM3 (n = 3) for females and MM1-MM3 (n = 3) for males. High-throughput sequencing was used to research the v3-v4 region of 16S rDNA in the intestinal flora. The results suggested significant differences in the structure, composition, and diversity of the intestinal flora of male versus female swamp eels. The relative abundances of Actinobacteria and Spirochaetes in female swamp eels were significantly higher (p < 0.05) than in male swamp eels at the phylum level. The relative abundances of Mycobacterium, Bacillus, and Vagococcus in female swamp eels were considerably higher (p < 0.05) than in male swamp eels at the genus level. The Alpha diversity of females was higher than that of males, and the Shannon index of females was also significantly higher (p < 0.05, Tukey’s HSD) than that in males. Investigations of Beta diversity, including NMDS ordination, UPGMA cluster analysis, and PLS-DA analysis, showed that female and male swamp eels could be clearly distinguished based on significant differences in intestinal flora between the FM group and the MM group. LEfSe analysis showed that the dominant bacteria were Cetobacterium in male swamp eels and Clostridium_sp_ND2, Mycobacterium, Alphaproteobacteria, and Bacilli in female swamp eels. The results showed dramatic differences in the intestinal flora between the sexes, which indicates the need for a more in-depth study on sex reversal in the future.

Nannochloropsis sp. was grown to the exponential phase and transferred to the high CO2 (2,800 μl l−1) and irradiance (100 μmol photons m−2 s−1) condition with different levels of nitrate and phosphate for 72 h, then the photosynthetic activity and inorganic carbon acquisition of the alga were measured. The apparent photosynthetic efficiency ( α) of Nannochloropsis sp. decreased with increasing NO3 − concentration from 150 to 3,000 μM, and the high nitrate-grown cells showed the lowest levels of light-saturated photosynthetic rate ( Pm), while the low nitrate-grown cells showed the highest levels of dark respiration rate ( Rd). The maximal light-saturated photosynthetic rate and the minimal dark respiration rate were seen under the middle nitrate condition. When the nitrate concentration ranged from 150 to 3,000 μM, the affinity for inorganic carbons of Nannochloropsis sp. increased sharply with the increasing NO3 − concentration to 300 μM and then decreased significantly. The middle phosphate-grown cells exhibited the highest light-saturated photosynthetic rate and apparent photosynthetic efficiency, however, the affinity for inorganic carbons of Nannochloropsis sp. was the maximum under the low phosphate condition.
