ABSTRACT: As a highly infectious and contagious pathogen in chickens, infectious bronchitis virus (IBV) is currently grouped into nine genotypes (GI to GIX). However, the classification of serotypes of IBV is still not clear. In this study, 270 field strains of IBV were isolated from dead or diseased chicken flocks in eastern and southern China during January 2021 to April 2023. These isolated IBV strains could be classified into 2 genotypes, GI (including 5 lineages GI-1, GI-13, GI-19, GI-22, and GI-28) and GVI based on the complete S1 sequence. Further analysis showed that the GI-19, GI-13, GI-22, GI-28, and GVI were the dominant genotypes with the proportions of 61.48, 8.89, 8.89, 7.78, and 8.89% respectively, and the homology of S1 protein of these isolates ranged from 86.85 to 100% in GI-19, 92.22 to 100% in GI-13, 83.1 to 100% in GI-22, 94.81 to 100% in GI-28 and 90.0 to 99.8% in GVI, respectively. Moreover, cross-neutralization test with sera revealed that these isolates in GI-19 lineage could be classified into at least 3 serotypes according to the antigenic relationship. In addition, structure assay using PyMOL indicated that one mutation such as S120 in receptor binding site (RBD) of GI-19 might alter the antigenicity and conformation of S protein of IBV. Overall, our data demonstrate that not only multiple genotypes, but also multiple serotypes in a single genotype or lineage have been co-circulated in eastern and southern China, providing novel insights into the molecular evolution of the antigenicity of IBV and highlighting the significance of the selection of the dominant isolate for vaccine development in IBV endemic region.

The technique of Enhanced Gas Recovery by CO2 injection (CO2-EGR) into shale reservoirs has brought increasing attention in the recent decade. CO2-EGR is a complex geophysical process that is controlled by several parameters of shale properties and engineering design. Nevertheless, more challenges arise when simulating and predicting CO2/CH4 displacement within the complex pore systems of shales. Therefore, the petroleum industry is in need of developing a cost-effective tool/approach to evaluate the potential of applying CO2 injection to shale reservoirs. In recent years, machine learning applications have gained enormous interest due to their high-speed performance in handling complex data and efficiently solving practical problems. Thus, this work proposes a solution by developing a supervised machine learning (ML) based model to preliminary evaluate CO2-EGR efficiency. Data used for this work was drawn across a wide range of simulation sensitivity studies and experimental investigations. In this work, linear regression and artificial neural networks (ANNs) implementations were considered for predicting the incremental enhanced CH4. Based on the model performance in training and validation sets, our accuracy comparison showed that (ANNs) algorithms gave 15% higher accuracy in predicting the enhanced CH4 compared to the linear regression model. To ensure the model is more generalizable, the size of hidden layers of ANNs was adjusted to improve the generalization ability of ANNs model. Among ANNs models presented, ANNs of 100 hidden layer size gave the best predictive performance with the coefficient of determination (R2) of 0.78 compared to the linear regression model with R2 of 0.68. Our developed ML-based model presents a powerful, reliable and cost-effective tool which can accurately predict the incremental enhanced CH4 by CO2 injection in shale gas reservoirs.

Miscible natural gas injection is widely considered as a practical and efficient enhanced oil recovery technique. However, the main challenge in this process is the high minimum miscibility pressure (MMP) between natural gas and crude oil, which limits its application and recovery factor, especially in high-temperature reservoirs. Therefore, we present a novel investigation to quantify the effect of chemical-assisted MMP reduction on the oil recovery factor. Firstly, we measured the interfacial tension (IFT) of the methane-oil system in the presence of chemical or CO2 to calculate the MMP reduction at a constant temperature (373K) using the vanishing interfacial tension (VIT) method. Afterwards, we performed three coreflooding experiments to quantify the effect of MMP reduction on the oil recovery factor under different injection scenarios.The interfacial tension measurements show that adding a small fraction (1.5 wt%) of the tested surfactant (SOLOTERRA ME-6) achieved 9% of MMP reduction, while adding 20 wt% of CO2 to the methane yields 13% of MMP reduction. Then, the coreflooding results highlight the significance of achieving miscibility during gas injection, as the ultimate recovery factor increased from 65.5% under immiscible conditions to 77.2% using chemical-assisted methane, and to 79% using gas mixture after achieving near miscible condition. The results demonstrate the promising potential of the MMP reduction to significantly increase the oil recovery factor during gas injection. Furthermore, these results will likely expand the application envelop of the miscible gas injection, in addition to the environmental benefits of utilizing the produced gas by re-injection/recycling instead of flaring which contributes to reducing the greenhouse gas emissions.

The rapidly-cooling of pure molybdenum (Mo) at 1010 Ks−1 and the uniaxial tensile of the solid at a strain rate of 2 × 10 s−1 were studied by molecular dynamics simulation; then the structure evolution was investigated in terms of pair distribution function and the largest standard cluster analysis. It is found that Mo melt was cooled into a complex crystal with multiple characteristic lengths. Further analysis revealed that it is a mixture topological close packing (TCP) crystal composed of the dominated A15 phase (Mo-A15) and the less Z phase, with H phase as twin grain boundary. The Phonon spectrum, potential energy, and cohesion energy revealed that Mo-A15 is stable slightly inferior to Mo-bcc. Compared to the prevailing bcc Mo, Mo-A15 holds much higher Young’s modulus, ultimate tensile strength, and Yield strength, being a representative material for barrier layer with high hardness.

As an immunosuppressive disease virus, chicken infectious anemia virus (CIAV) mainly infects chickens, causing aplastic anemia and systemic lymphoid tissue atrophy. In recent years, the prevalence of CIAV in the poultry industry globally has caused huge economic losses. In this study, a total of 223 clinical samples, including anal swabs, tissues, blood, and vaccines, were collected from 19 broiler farms or breeding companies in Jiangsu province, with symptoms of significant anemia and immunosuppression during 2020–2022. Among them, 75 samples (75/223, 33.6%) were positive for CIAV in polymerase chain reaction (PCR) test, and 20 CIAV strains were successfully isolated. The phylogenetic trees of the 20 isolates and 42 CIAV strains deposited in GenBank formed four distinct groups (A-D). And the isolates mainly belonged to Group A but with high genetic diversity. Analysis for VP1 indicated that these isolates possess key characteristics of highly pathogenic strains. Meanwhile, VP2 and VP3 were much conserved with much fewer mutations compare to VP1. The above epidemiological study of CIAV provides novel insights into molecular characterization of CIAV and lays the foundation for developing efficient strategies for control of CIAV in China.

Since 2011, the Gyrovirus galga 1 (GyVg1, previously recognized as avian gyrovirus 2) strain has extensively been detected worldwide. However, because there are no up-to-date reports of examining the distribution of GyVg1 in flocks in southern China, the epidemiology of this virus is unknown. To investigate the prevalence and genetic evolution of GyVg1, a total of 2,077 field samples collected from 113 chicken farms in 6 provinces in southern China during 2020 to 2022 were tested. Among them, 315 samples (315/2,077, 15.17%) were positive for GyVg1 by PCR. The positive rate of GyVg1 detection between different regions of southern China ranged from 11.69% (Guangdong) to 22.46% (Yunnan). The correlation between GyVg1 prevalence and sample source groups was analyzed, the results showing that the highest seroprevalence of GyVg1 was observed in visceral tissues (27.34%, 187/684), significantly higher (P < 0.05) than that of feather shafts (17.22%, 31/180), serums (8.85%, 78/881), and fecal (5.72%, 19/332). Additionally, the complete genomes of 10 GyVg1 strains were sequenced and analyzed, which showed nucleotide identities of 96.2 to 99.9%, 97.0 to 100.0%, 95.2 to 100.0%, and 95.7 to 99.8% in the complete genome, ORF1, ORF2, and ORF3, respectively, and 94.4 to 100.0%, 91.3 to 100.0%, and 98.7 to 100.0% amino acid similarity in the VP2, VP3, and VP1 proteins, respectively. Phylogenetic analysis of the whole genome showed that 10 GyVg1 strains belong to genotype I, and one strain belongs to genotype III. Sequence analysis showed several amino acid substitutions in both the VP1, VP2, and VP3 proteins. Our results enhance the understanding of the molecular characterization of GyVg1 infection in southern China. In conclusion, this study reveals the high prevalence and high genetic differentiation of GyVg1 in Chinese chickens and suggests that the potential impact of GyVg1 on the chicken industry may be of concern.

ABSTRACT: In recent years, the infection rate of avian encephalomyelitis virus (AEV) infection in chickens has risen significantly, seriously endangering the development of the chicken industry. In order to study the current epidemiological status of AEV in China as well as the genetic and evolutionary patterns of the virus, we conducted a survey and genomic analysis of chicken AEV. The results showed that 46.26% (136/294) of the tissue samples tested (n = 294) were positive for AEV, with the highest positivity rate of 62.24% (61/98) among tissue samples from chickens aged 13 to 18 wk. The complete genomes of 2 representative AEV strains were determined, and the VP1 evolutionary tree results revealed that the 2 representative strains belonged to a novel AEV strain. Multiple alignment analysis showed that the ORF1 genes of the 2 representative strains differed by 82.3 to 99.9% at the amino acid level compared with the reference AEV strain, and the mutations at the key amino acid loci of VP2 and VP3 were the same as those in the chick embryo-adapted strain. The analysis makes up for the molecular epidemiological data and genetic variation of the 2 representative strains. The analysis makes up for the molecular epidemiological data and genetic variation of AEV and provides a basis for further understanding the spread of AEV in China.

BackgroundArticular cartilage and cartilage matrix degradation are key pathological changes occurring in the early stage of knee osteoarthritis (KOA). However, currently, there are limited strategies for early prevention and treatment of KOA. Duhuo Jisheng Decoction (DHJSD) is a formula quoted in Bei Ji Qian jin Yao Fang, which was compiled by Sun Simiao in the Tang Dynasty of China. As a complementary therapy, it is widely used to treat early-stage KOA in China; however, its mechanism has not been completely elucidated.ObjectiveThis study investigated the potential role of DHJSD in preventing cartilage degradation and the underlying mechanism.MethodsA rat model of KOA model was established via the Hulth method. Subsequently, 25 rats were randomized into sham (saline), model control (saline), high-DHJSD (1.9g/mL of DHJSD), medium-DHJSD (1.2g/mL of DHJSD), and low-DHJSD groups (0.6g/mL of DHJSD). After 4 weeks of treatment, all rats were sacrificed and the severity of the cartilage degeneration was evaluated by a series of histological methods. The autophagosome was observed using transmission electron microscopy, and the related functional proteins were detected by the western blotting and real-time polymerase chain reaction. Next, the mechanism by which DHJSD improves knee cartilage degeneration was further clarified the in vitro by gene silencing technology combined with a series of functional experiments. The proteins levels of PTEN, Akt, p-Akt, mTOR, and p-mTOR, as well as the marker proteins of autophagy and apoptosis were determined. Zinc levels in chondrocytes were determined using inductively coupled plasma mass spectrometry.ResultsHistopathological staining revealed that DHJSD had a protective effect on the cartilage. DHJSD increased autophagosome synthesis and the expression of autophagy proteins LC3 and Beclin-1 in chondrocytes. Moreover, it reduced the phosphorylation levels of Akt and mTOR and the levels of zinc, MMP-13, Bax, and Bcl-2. Following PTEN silencing, this DHJSD-mediated reduction in Akt and mTOR phosphorylation and Bax, Bcl-2, and zinc levels were further decreased; in addition, DHJSD-mediated increase in LC3 and Beclin-1 levels was decreased.ConclusionDHJSD inhibits the Akt/mTOR signaling pathway by targeting PTEN to promote autophagy in chondrocytes, which may help reduce MMP-13 production by regulating zinc levels in chondrocytes.

Partial discharge (PD) is a localized discharge phenomenon in the insulator of electrical equipment resulting from the electric field strength exceeding the local dielectric breakdown electric field. Partial-discharge signal identification is an important means of assessing the insulation status of electrical equipment and critical to the safe operation of electrical equipment. The identification effect of traditional methods is not ideal because the PD signal collected is subject to strong noise interference. To overcome noise interference, quickly and accurately identify PD signals, and eliminate potential safety hazards, this study proposes a PD signal identification method based on multiscale feature fusion. The method improves identification efficiency through the multiscale feature fusion and feature aggregation of phase-resolved partial-discharge (PRPD) diagrams by using PMSNet. The whole network consists of three parts: a CNN backbone composed of a multiscale feature fusion pyramid, a down-sampling feature enhancement (DSFB) module for each layer of the pyramid to acquire features from different layers, a Transformer encoder module dominated by a spatial interaction–attention mechanism to enhance subspace feature interactions, a final categorized feature recognition method for the PRPD maps and a final classification feature generation module (F-Collect). PMSNet improves recognition accuracy by 10% compared with traditional high-frequency current detection methods and current pulse detection methods. On the PRPD dataset, the validation accuracy of PMSNet is above 80%, the validation loss is about 0.3%, and the training accuracy exceeds 85%. Experimental results show that the use of PMSNet can greatly improve the recognition accuracy and robustness of PD signals and has good practicality and application prospects.

Abstract Duck adenovirus 3 (DAdV-3), a newly emerged duck adenovirus, has resulted in significant economic losses to the duck industry across China since 2014. However, little is known about the B cell epitopes in major antigen of DAdV-3 and the serological approach for detection of DAdV-3 is not available. In this study, four monoclonal antibodies (mAbs) specific to Fiber-2 protein of DAdV-3 were first generated and designated as 2G10, 3D9, 5E6, and 6B12. Indirect immunofluorescence assay (IFA) showed that all of the mAbs reacted with the Fiber-2. Moreover, mAbs 2G10, 5E6, and 6B12 demonstrated good activity with Fiber-2 in Western blot. Notably, the Fiber-2 could be immunoprecipitated efficiently by mAb 3D9. Epitope mapping revealed that mAbs 2G10, 3D9, 5E6, and 6B12 recognized 397-429aa, 463-481aa, 67-99aa, and 1-66aa of Fiber-2, respectively. Besides, a novel sandwich ELISA for efficient detection of DAdV-3 was developed based on mAb 3D9 and horseradish peroxidase (HRP) conjugated mAb 3D9. The sandwich ELISA only reacted with DAdV-3 but not with other duck-associated viruses. The limit of detection of the ELISA was 6.25 × 103 TCID50/mL. Overall, the mAbs generated laid the foundation for elucidating the critical role of Fiber-2 in mediating infection and pathogenesis, and the sandwich ELISA approach established here provided efficient and rapid serological diagnostic tool for DAdV-3.

The Ti substrate, with physicochemical stability and excellent electrical conductivity, is potential to fabricate all-solid-state batteries. However, the Titanium atoms at high-temperature synthesis makes it easy to react with active materials. Modifying the titanium surface with an inert protective layer while ensuring high electrical conductivity remains a significant challenge. Herein, a pencil was used to draw on the Ti substrate and subsequently sintering at a high temperature to obtain a graphite-coated Ti substrate (Ti-Graphite). The experimental results show that Ti-Graphite has a smoother surface and fewer defects in the graphite layer. Moreover, titanium carbide is formed at the interface of graphite and Ti substrate, which will enhance the physicochemical stability of the Ti-Graphite. To gain deep understanding of the evolution mechanism in the synthesis, molecular dynamics simulations were carried out to investigate it. The results exhibit that the voids at the C/Ti interface and stress in the model were significantly decreased after sintering and cooling treatment, proving that Ti-Graphite has a smoother surface, a more stable interface, and fewer defects at the atomic scale. Therefore, we provide a simple and facile strategy to obtain a Ti-graphite substrate with superior physicochemical stability, which reaveals excellent potential to synthesize high-performance all-solid-state batteries.

ABSTRACT Recently, the infection of fowl adenoviruses (FAdVs) in chicken flocks has caused huge economic losses to the poultry industry in China. Although immunization with Fiber protein can provide autogenous protection of FAdVs, no commercial multivalent vaccines are available for the prevention and control of FAdVs. In this study, a novel recombinant virus FAdV4-F/8a-rF2 expressing the Fiber of FAdV-8a was generated through CRISPR-Cas9 and Cre-LoxP system by using the template virus FA4-EGFP. FAdV4-F/8a-rF2 not only exhibited a similar replication capacity to the wild-type FAdV-4 in Leghorn male hepatoma (LMH) cells but was also highly attenuated to specific pathogen free (SPF) chickens. Moreover, the inoculation of FAdV4-F/8a-rF2 could induce high neutralizing antibodies and provide full protection against both FAdV-4 and FAdV-8a. All these data demonstrate that the recombinant virus FAdV4-F/8a-rF2 developed here can be used as an attenuated bivalent vaccine candidate for the prevention and control of both FAdV-4 and FAdV-8a. IMPORTANCE Epidemiological data reveal that FAdV-4 and FAdV-8a are the dominant serotypes of FAdVs in the poultry industry in China. Although three commercial inactivated vaccines against FAdV-4 have been licensed in China, the bivalent vaccine against both FAdV-4 and FAdV-8a is not available. Here, we used CRISPR-Cas9 and Cre-LoxP system to generate a recombinant virus FAdV4-F/8a-rF2 expressing the Fiber of FAdV-8a. Notably, FAdV4-F/8a-rF2 was highly attenuated and could provide efficient protection against both FAdV-4 and FAdV-8a in the chicken infection model, highlighting the applaudable application of FAdV4-F/8a-rF2 as a novel live-attenuated bivalent vaccine against the diseases caused by the infection of FAdV-4 and FAdV-8a.

ABSTRACT H6 influenza A viruses are enzootic and genetically diverse in both domestic poultry and wild birds, occasionally causing spillovers to mammals, including humans, posing threat to public health. However, the antigenic sites of the hemagglutinin (HA) of H6 are poorly understood. In this study, a panel of murine monoclonal antibodies (MAbs) with different reactivity spectra against HA of H6 was generated and nine key residues of antigenic epitopes in H6 were identified through escape mutants of these MAbs. Of these, residues 139 and 140 belonged to antigenic Sa, residue 89 belonged to antigenic Cb, residue 149 belonged to Ca2, while residue 221 belonged to Ca1 regarding the H1 antigenic sites. However, the other four residues 69, 120, 124, and 246 were not located in the identified antigenic regions of H1. Although most HA variants of the escape mutants had the preference for avian-like receptor, whereas escape mutants m3B9 and m4C2 increased the binding affinity for both avian and human-like receptors, and m1A5 and m6D11 decreased the binding affinity for avian-like receptors. Moreover, a single dose of MAbs 4C2 or 6E3 could fully protect mice against lethal challenge of mouse-adapted A/Eurasian teal/Jiangxi/2018WB0417(H6N2) (MA E-Teal/417) in the prophylactic treatment and also could provide partial protection in the therapeutic treatment. Overall, the generated MAbs and the identified key residues of antigenic epitopes not only provide novel insights into the key markers for monitoring the antigenic variation of H6 but also demonstrate the potential for developing MAb-based passitive treatments for H6. IMPORTANCE Since the escape immunity of influenza A viruses (IAVs) is mainly caused by the continuous antigenic variations in HA, the identification of key antigenic epitopes is crucial for better understanding of the escape immunity and vaccine development for IAVs. The antigenic sites of several HA subtypes, including H1, H3, H5, and H9, have been well characterized, whereas those of H6 subtype are poorly understood. Here, we mapped nine key residues of antigenic epitopes in H6 through escape mutants using a panel of MAbs. Moreover, MAbs 4C2 and 6E3, targeting 140 and 89 residues, respectively, could protect mice against lethal challenge of MA E-Teal/417. These key residues of antigenic epitopes identified here provide the molecular targets for further elucidating the antigenic evolution of H6 and better preparing the vaccine against H6 IAV.

Semiconductor–metal composites are widely used in aerospace and automotive manufacturing, weapon production, and electronic packaging because of their high hardness and strength as well as low coefficient of thermal expansion. In this study, SiC–CuNi composite films produced by single-target and dual-target co-deposition were simulated using molecular dynamics. The films were subjected to high-temperature quenching to obtain an annealing model. The deformation behaviors of the deposition and annealing models during the nanoindentation process were investigated in detail. Then, a characterization method for inspecting the roundness of nanochannels was proposed. In the deposition model, the Ni–Cu model generated amorphous structures at the interface. However, the annealing treatment atomically rearranged the film, producing dense films with improved crystalline quality. Other deposition models are developed using complex twin structures, which play a role in stress distribution and strain diffusion during deformation. Comparative investigation shows that high-temperature quenching can effectively enhance the stiffness of composite films, tightly bonding the atoms at the semiconductor–metal interface. Hardness calculations were performed for all models and the result was that the Cu + Ni model had the highest hardness. Moreover, the formation of nanochannels is affected: the tighter the bond, the more uniform the nanochannels.

Functional gradient materials (FGMs) are a category of advanced materials that contain two (or more) different components with continuously varying compositional distributions. With the advancement in nanotechnology, FGM applications have evolved from traditional applications to complex micro- and nano-electronic and energy conversion devices. Therefore, studying the mechanical behavior of the nanostructures of different FGMs plays a crucial role in determining the feasibility of their various applications. Herein, the mechanical behavior of the shear deformation of Ti–Al FGM nano-bulks with a radial (Z-axis direction) gradient was investigated using molecular dynamics (MD) simulations. The distributions of Ti and Al in the Z-direction were obtained using the sigmoid function. The results demonstrate that the distribution function parameters are important in regulating the mechanical properties—elastic modulus and ultimate tensile strength—of Ti–Al FGMs. It gives us a new perspective on the analysis of the microstructure evolution and dislocation density of FGMs, providing theoretical support for the practical application of FGMs.

Scientists have extensively studied metallic glasses (MGs) for their excellent properties and potential applications. However, the limited glass forming ability (GFA) of MGs poses a significant challenge in engineering fabrication, severely limiting their scope of application. Herein, we collected data from 167 peer-reviewed research articles and 2 commonly used datasets and constructed a comprehensive dataset containing 2085 data points. We employed gradient boosting algorithmic models, such as XGBoost, LGBoost, and AdaBoost, for selecting the best feature set and constructing the most effective model from among 118 feature parameters through techniques such as three-step feature selection method, ten-fold cross-validation, and grid search. As the uneven distribution of the original dataset significantly impacts the model’s performance, we implemented five nonlinear transformation techniques and three oversampling techniques to improve the model. The results indicated that the nonlinear and oversampling techniques improved the model’s performance by improving the distribution balance. Finally, we interpreted the constructed models through Shapley additive explanation, providing relevant rules concerning GFA. The techniques employed and the rules presented herein contribute to the statistical understanding of GFA and may facilitate the discovery of novel MGs.

ABSTRACT: Avian infectious bronchitis (IB) is a highly contagious disease caused by infectious bronchitis virus (IBV). Vaccination is an effective approach for controlling IBV. Therefore, reliable immune monitoring for IB is critical for poultry. In this study, a novel peptide derived from S2 protein was used to develop an enzyme-linked immunosorbent assay (ELISA) for the detection of broadly cross-reactive antibodies against IBV. The peptide-based ELISA (pELISA) showed good specificity and sensitivity in detecting IBV antibodies against different serotypes. A semilogarithmic regression method for determining IBV antibody titers was also established. Antibody titers detected by pELISA and calculated with this equation were statistically similar to those evaluated by indirect fluorescence assay (IFA). Moreover, the comparison analysis showed a 96.07% compatibility between the pELISA and IDEXX ELISA. All these data demonstrate that the pELISA generated here can be as a rapid and reliable serological surveillance tool for monitoring IBV infection or vaccination.

The glass-forming ability (GFA) of metallic glasses can be enhanced by adding trace elements, but it is difficult to explain the reasons from microcosmic viewpoint due to the limitation of experimental conditions. Therefore, in order to reveal the effects of trace elements on GFA, the rapid solidification process of Mg65Cu(35-x)Yx (x = 5,10,15,20,25) ternary alloys are simulated by molecular dynamics (MD) in this paper, then the short- and medium-range order, thermodynamics and dynamics are investigated. The average atomic potential energy shows that the GFA becomes stronger monotonically with the increase of Y element. S433 and S544 can evolve into S555 with higher fivefold symmetry and packing density during solidification, which the topologically close-packed (TCP) clusters are mainly composed of S555. And the higher TCP clusters system has stronger GFA, because TCP clusters are easy to connect with each other to form large-sized nanoclusters, and the larger size nanoclusters have a smaller average displacement, which hinders the crystallization. As TCP atoms with slower dynamic characteristics than other atoms, the dynamics with higher Y element becomes slower with the increase of TCP clusters, and thus adding Y element plays a promoting role for GFA.

Recently, the infection of serotype 4 fowl adenovirus (FAdV-4) in chicken flocks has become endemic in China, which greatly threatens the sustainable development of poultry industry. The development of recombinant FAdV-4 expressing foreign genes is an efficient strategy for controlling both FAdV-4 and other important poultry pathogens. Previous reverse genetic technique for generating the recombinant fowl adenovirus is generally inefficient. In this study, a recombinant FAdV-4 expressing enhanced green fluorescence protein (EGFP), FA4-EGFP, was used as a template virus and directly edited fiber-2 gene to develop an efficient double-fluorescence approach to generate recombinant FAdV-4 through CRISPR/Cas9 and Cre-Loxp system. Moreover, using this strategy, a recombinant virus FAdV4-HA(H9) stably expressing the HA gene of H9N2 influenza virus was generated. Chicken infection study revealed that the recombinant virus FAdV4-HA(H9) was attenuated, and could induce haemagglutination inhibition (HI) titer against H9N2 influenza virus at early time points and inhibit the viral replication in oropharynx. All these demonstrate that the novel strategy for constructing recombinant FAdV-4 expressing foreign genes developed here paves the way for rapidly developing attenuated FAdV-4-based recombinant vaccines for fighting the diseases caused by both FAdV-4 and other pathogens.

Recently, the highly pathogenic serotype 4 fowl adenovirus (FAdV-4) and duck adenovirus 3 (DAdV-3) were outbroken and widespread, causing substantial economic losses to the duck industry. Therefore, there is an urgent need to generate a recombinant genetic engineering vaccine candidate against both FAdV-4 and DAdV-3. In this study, a novel recombinant FAdV-4 expressing the Fiber-2 protein of DAdV-3, designated as rFAdV-4-Fiber-2/DAdV-3, was generated based on CRISPR/Cas9 and Cre-LoxP systems. Indirect immunofluorescence assay (IFA) and western blot (WB) showed that the Fiber-2 protein of DAdV-3 in rFAdV-4-Fiber-2/DAdV-3 was expressed successfully. Moreover, the growth curve revealed that rFAdV-4-Fiber-2/DAdV-3 replicated efficiently in LMH cells and even showed a stronger replication ability compared to the wild type FAdV-4. The generation of the recombinant rFAdV-4-Fiber-2/DAdV-3 provides a potential vaccine candidate against both FAdV-4 and DAdV-3.

ABSTRACT: Chicken infectious anemia virus (CIAV) is the pathogen of chicken infectious anemia. Currently, due to the lack of effective diagnostics technology and prevention approach, CIAV has spread globally and caused huge economic losses to poultry industry. In this study, a novel peptide-based ELISA (pELISA) for efficient detection of antibody against CIAV was developed. The peptide (25CRLRRRYKFRHRRRQRYRRRAF45) used in pELISA was highly conserved in VP1 protein of different CIAV isolates. The specificity and reproducibility showed that the pELISA only reacted with sera against CIAV, not with sera against other pathogens tested, and the CV of the intra-/inter-assay of the pELISA was 6.8 to 9.22%. Moreover, the comparison assay using 56 clinical samples showed that the positive rate of the pELISA and the commercial ELISA kit (IDEXX) was 85.7 and 80.4%, respectively. The pELISA generated here provides a rapid and efficient serological detection method for diagnosis of CIAV infection and evaluation of the efficacy of CIAV vaccination.

In this paper, we establish the structure model of multilayer Graphene (MLG)/Mg2Si/Si heterojunction photodetector (PD) based on the Technology CAD (TCAD). We further investigate the effect of Graphene layers on the photoelectric properties of MLG/Mg2Si/Si heterojunction PD. The simulation results indicate that compared with Mg2Si/Si PD, the photoelectric properties of MLG/Mg2Si/Si heterojunction PD are enhanced. The responsivity is increased from 0.51 A/W to 0.84 A/W, the external quantum efficiency (EQE) increased from 70.68% to 107.29%, and the noise equivalent power (NEP) reduced from 7.07 × 10–11 WHz–1/2 to 1.59 × 10–11 WHz–1/2. It is further shown that detectivity (D*) is increased from 1.4 × 1010 Jones to 6.3 × 1010 Jones, and the on/off ratio is increased from 141.45 to 629. After adding Graphene layers, the maximum improvement rates of the responsivity, EQE, NEP, D*, and on/off ratio of heterojunction PDs improved by 65%, 52%, 77.5%, 350%, and 345%, respectively. Moreover, it is seen that the energy band of MLG/Mg2Si/Si heterojunction PDs is shifted to the valence band, the reverse breakdown voltage and dark current are decreased. Our results indicate that the photocurrent in the spectral response is increased and the wavelength corresponding to the EQE peak is shifted towards red. A strong electric field is formed at the interface between MLG and Mg2Si, which is beneficial to the separation of photogenic carriers. The simulation results indicate that the MLG/Mg2Si/Si heterojunction PDs can detect visible light and NIR with a wavelength in the range of 400–1500 nm.

A facile sol–gel spin coating method has been proposed for the synthesis of spin-coated ZnO nanofilms on ITO substrates. The as-prepared ZnO-nanofilm-based W/ZnO/ITO memory cell showed forming-free and tunable nonvolatile multilevel resistive switching behaviors with a high resistance ratio of about two orders of magnitude, which can be maintained for over 103 s and without evident deterioration. The tunable nonvolatile multilevel resistive switching phenomena were achieved by modulating the different set voltages of the W/ZnO/ITO memory cell. In addition, the tunable nonvolatile resistive switching behaviors of the ZnO-nanofilm-based W/ZnO/ITO memory cell can be interpreted by the partial formation and rupture of conductive nanofilaments modified by the oxygen vacancies. This work demonstrates that the ZnO-nanofilm-based W/ZnO/ITO memory cell may be a potential candidate for future high-density, nonvolatile, memory applications.

Avian leukosis virus subgroup J (ALV-J) generally induces hemangioma, myeloid leukosis, and immunosuppression in chickens, causing significant poultry industry economic losses worldwide. The unusual env gene of ALV-J, with low homology to other subgroups of ALVs, is associated with its unique pathogenesis. However, the exact molecular basis for the pathogenesis and oncogenesis of ALV-J is still not fully understood. In this study, ALV-J infection and the overexpression of Env could efficiently downregulate the phosphorylation of SHP-2 (pSHP-2) in vitro and in vivo. The membrane-spanning domain (MSD) in Env Gp37 was the functional domain responsible for pSHP-2 downregulation. Moreover, the overexpression of SHP-2 could effectively promote the replication of ALV-J, whereas knockout or allosteric inhibition of SHP-2 could inhibit ALV-J replication. In addition, the knockout of endogenous chicken SHP-2 could significantly increase the proliferation ability of DF-1 cells. All these data demonstrate that SHP-2 dephosphorylated by ALV-J Env could efficiently promote ALV-J replication, highlighting the important role of SHP-2 in the pathogenesis of ALV-J and providing a new target for developing antiviral drugs against ALV-J.

The outbreaks of hepatitis-hydropericardium syndrome (HPS) caused by the highly pathogenic serotype 4 fowl adenovirus (FAdV-4) have caused a huge economic loss to the poultry industry globally since 2013. Although the Fiber-2 has been identified as a key virulent related factor for FAdV-4, little is known about its molecular basis. In this study, we identified the efficient interaction of the Fiber-2 with the karyopherin alpha 3/4 (KPNA3/4) protein via its N-terminus of 1–40aa. The analysis of the overexpression and knockout of KPNA3/4 showed that KPNA3/4 could efficiently assist the replication of FAdV-4. Moreover, a fiber-2-edited virus FAV-4_Del with a deletion of 7–40aa in Fiber-2 was rescued through the CRISPR-Cas9 technique. In comparison with the wild type FAdV-4, FAV-4_Del was highly attenuated in vitro and in vivo. Notably, the inoculation of FAV-4_Del in chickens could provide full protection against the lethal challenge with the wild type FAdV-4. All these findings not only give novel insights into the molecular basis for the pathogenesis of Fiber-2 but also provide efficient targets for developing antiviral strategies and live-attenuated vaccine candidates against the highly pathogenic FAdV-4.

In recent years, hepatitis-hydropericardium syndrome (HHS) and inclusion body hepatitis (IBH) caused by serotype 4 fowl adenovirus (FAdV-4) and serotype 8b fowl adenovirus (FAdV-8b), respectively, are widely prevalent in China, causing huge economic losses to the poultry industry. Numerous studies have revealed the mechanism of the infection and pathogenesis of FAdV-4. However, little is known about the mechanism of infection with FAdV-8b. Among the major structural proteins of fowl adenoviruses, fiber is characterized by the ability to recognize and bind to cellular receptors to mediate the infection of host cells. In this study, through superinfection resistance analysis and an interfering assay, we found that Fiber-1 of FAdV-4, rather than hexon, penton, and fiber of FAdV-8b, conferred efficient superinfection resistance against the infection FAdV-8b in LMH cells. Moreover, truncation analysis depicted that the shaft and knob domains of FAdV-4 Fiber-1 were responsible for the inhibition. However, knockout of the coxsackie and adenovirus receptor (CAR) in LMH cells inhibited the replication of FAdV-8b only at early time points, indicating that CAR might not be the key cell receptor for FAdV-8b. Overall, our findings give novel insights into the infection mechanism of FAdV-8b and provide a new target for the prevention and control of both FAdV-4 and FAdV-8b.

H6 avian influenza viruses (AIVs) not only continue to circulate in both domestic poultry and wild waterfowl, but also have occasionally caused spillovers infections in pigs and humans, posing a potential threat to public health. However, the molecular mechanism of H6 AIV adaptation to mammals remains largely unknown. In this study, two mouse-adapted (MA) H6 AIV strains, named as MA E-Teal/417 and MA GWF-Goose/740, were generated through blind passages in BALB/c mice. The two MA H6 strains replicated more efficiently and showed higher virulence than the corresponding wild type (WT) H6 strains in mice. Genome sequencing revealed that MA E-Teal/417 and MA GWF-Goose/740 carried six amino acid mutations (PB2-T224A/E627K, HA-G124R, NA-F167L/Y356H and M1-M92R), and four amino acid mutations (PB1-K577E, PA-T97I/D514E and HA-T276K), respectively, when compared to the corresponding WT virus. Receptor binding assay showed MA E-Teal/417 had stronger binding activity to α-2,3 SA than WT E-Teal/417. Moreover, the polymerase activity analysis found the RNP polymerase activity of both MA H6 viruses was significantly higher than that of the corresponding WT virus in 293T cells. All these demonstrate that H6 AIV can acquire limit amino acid substitutions to adapt to mammals and increase virulence, highlighting the significance of monitoring such mutations of H6 AIV in the field for alarming the potential of its cross-transmission and pathogenesis in mammals.

Background and objectivesDiabetes mellitus is a serious metabolic disease that causes a serious economic burden worldwide. Gut flora is a major component of diabetes research, and the aim of this study was to understand the trends and major components of research related to diabetes and gut flora in the last 11 years.MethodsWe searched the Web of Science Core Collection database for articles on diabetes and gut flora related research from 2011-2021 on July 2, 2022. The literature data were analyzed for country, institution, author, steward, journal, and highly cited literature using Citespace.5.8.R3 and Vosviewer1.6.17.ResultsFinally 4834 articles that met the requirements were included. The overall trend of articles published in the last 11 years is increasing, and the trend of articles published after 2019 is increasing significantly. In total, 109 countries, 4820 institutions, and 23365 authors were involved in the field of research. The highest number of publications was 1262 articles from the United States, the institution with the most publications was the University of Copenhagen with 134 articles, and the author with the most publications was PATRICE D CANI with 52 articles.ConclusionThe number of studies related to diabetes and intestinal flora is increasing and more and more researchers are involved in this field. Intestinal flora provides a key research direction for the treatment of diabetes. In the future, gut flora will remain the focus of the diabetes field.

Recently, the outbreak of the infection of Duck adenovirus 3 (DAdV-3) characterized by swelling and hemorrhagic liver and kidney has caused huge economic losses to duck industry since 2014 in China. To date, the B cell epitopes in the Fiber-1 protein and the underlying infection mechanism of DAdV-3 have not been investigated. In this study, the recombinant Fiber-1 protein was first expressed in E. coli and six novel monoclonal antibodies (mAbs) against Fiber-1 were generated, designated as 1D8, 1E6, 3G6, 4G1, 4G2, and 6F10, respectively. Moreover, mAbs 3G6 and 6F10 could efficiently immunoprecipitate the Fiber-1 in LMH cells infected with DAdV-3 or transfected with pcDNA3.1-Fiber-1. Notably, mAbs 3G6 and 4G2 also showed certain neutralizing activity against DAdV-3 infection in vitro. Epitopes mapping revealed that the B cell epitope recognized by 6F10, 3G6, 4G1, 1D8, 4G2, and 1E6 was located in 34-66aa, 67-99aa, 64-296aa, 297-329aa, 330-362aa, and 363-395aa, respectively. Sequence alignments further found that the six epitopes recognized by these mAbs were highly conserved among different DAdV-3 isolates. The generated mAbs specific to Fiber-1 and their defined epitopes provide powerful tools for establishing rapid and efficient diagnostics for the detection of DAdV-3 and pave the way for further studying on the critical role of Fiber-1 in mediating the infection of DAdV-3.

Miscible CO2 injection appears to be an important enhanced oil recovery technique for improving sweep efficiency and eliminating CO2-oil interfacial tension resulting in up to 10% higher oil recovery compared to immiscible flooding, in addition to the environmental benefits of reducing greenhouse gas emissions through carbon capturing utilising and storage (CCUS). Moreover, this technique could be similarly applicable to natural gas and nitrogen projects to increase oil recovery and to reduce the associated gas flaring. However, miscible displacement may not be achievable for all reservoirs, in particular, reservoirs with high temperature where high injection pressure would be needed to reach miscibility which likely exceeds the formation fracture pressure. Therefore, to further achieve reservoirs’ potential, there is a pressing need to explore a viable means to decrease the miscibility pressure, and thus expand the application envelop of miscible gas injection in reservoirs with high temperatures.In this work, we aim to provide insights into minimum miscibility pressure (MMP) reduction by adding chemicals into CO2 phase during injection. We achieved this objective by performing a comprehensive review on chemical-assisted MMP reduction using different chemical additives (e.g., alcohols, fatty acids, surfactants) and different experimental methodologies.Previous experimental studies have shown that a fraction of chemical additives can yield up to 22% of MMP reduction in CO2-oil system. Based on results analysis, surfactant based chemicals were found to be more efficient compared to alcohol based chemicals in reducing the interfacial tension in the CO2-oil system. Based on the current experimental results, adding chemicals to improve the miscibility and reduce the MMP in the CO2-oil system appears to be a promising technique to increase oil recovery while reducing operating cost. Selection of the effective chemical additives may help to expand the application of miscible gas injection to shallow and high temperature reservoirs. Furthermore, our review provides an overall framework to screen potential chemical additives and an injection strategy to be used for miscible displacement in CO2 and/or gas systems.

Goose astrovirus (GoAstV) is a new Avastrovirus of the genus astrovirus causing gout, hemorrhage, and swellings of kidneys that have affected goslings around the major goose-producing regions in China. The GoAstV is divided into goose astrovirus type 1 (GoAstV-1) and goose astrovirus type 2 (GoAstV-2). Although GoAstV-2 is known to be the causative agent of goose gout, little published information about the relationship between GoAstV-1 and goose gout is unknown. In this study, we investigated the presence of GoAstV-1 in 293 visceral tissue/dead embryos samples with gout on different farms in Jiangxi province, China. A survey result indicated that the mono-infection of GoAstV-1 (32.08%) and co-infection of GoAstV-1 (12.28%) with GoAstV-2 in gout goslings in Jiangxi, China. JXGZ, a GoAstV-1 strain, was effectively isolated from the visceral tissue of gosling gout and serially propagated for more than 25 passages in a goose embryo. The JXGZ strain's whole genome was sequenced and investigated. Phylogenetic analysis of complete genome and capsid protein sequences of JXGZ strain show that it was more closely related to GoAstV-1 strain than GoAstV-2 strain and was grouped within the GoAstV-1 cluster. These findings will aid in the development of efficient diagnostic reagents and possible vaccinations by providing insight into the prevalence and genetic evolution of GoAstV-1 in China.

With the rapid development of society, the demand for information storage is increasing. Hence, research involving magnetic storage materials is gaining importance. In this study, Fe3Mn3Co60.66Si33.34 powders were prepared via mechanical alloying and sintering. The effect of the sintering temperature on the crystal structure was analyzed by X-ray diffraction, and the results showed that doped Co2Si powder was successfully prepared. Vibrating sample magnetometry was used to investigate the magnetic properties of Fe3Mn3Co60.66Si33.34, and showed that the coercivity of the samples decreased with increasing sintering temperature. Fe3Mn3Co60.66Si33.34 sintered at 950 °C yielded a magnetic saturation strength of 36.42 emu/g, coercivity of 135.92 Oe, and remanent magnetization of 4.97 emu/g. Electron microscopy analyses showed that the prepared particles were spherical, and the average grain size increased with increasing sintering temperature. The electromagnetic loss was analyzed by simulating electromagnetic parameters, which revealed that the electromagnetic loss tangent decreased with increasing sintering temperature. Hence, it is inferred that Fe3Mn3Co60.66Si33.34 powder materials sintered at high temperatures are expected to have suitable properties for magnetic storage applications.

A facile hydrothermal process has been developed to synthesize the α-Fe2O3 nanowire arrays with a preferential growth orientation along the [110] direction. The W/α-Fe2O3/FTO memory device with the nonvolatile resistive switching behavior has been achieved. The resistance ratio (RHRS/RLRS) of the W/α-Fe2O3/FTO memory device exceeds two orders of magnitude, which can be preserved for more than 103s without obvious decline. Furthermore, the carrier transport properties of the W/α-Fe2O3/FTO memory device are dominated by the Ohmic conduction mechanism in the low resistance state and trap-controlled space-charge-limited current conduction mechanism in the high resistance state, respectively. The partial formation and rupture of conducting nanofilaments modified by the intrinsic oxygen vacancies have been suggested to be responsible for the nonvolatile resistive switching behavior of the W/α-Fe2O3/FTO memory device. This work suggests that the as-prepared α-Fe2O3 nanowire-based W/α-Fe2O3/FTO memory device may be a potential candidate for applications in the next-generation nonvolatile memory devices.

Currently, the outbreak of serotype 4 fowl adenovirus (FAdV-4) has spread worldwide and caused tremendous economic loss to the poultry industry. Although inactivated vaccines have been licensed against FAdV-4 in China, a rapid and efficient serological method for measuring the titer of neutralizing antibodies (NAbs) specific for FAdV-4 post-infection or vaccination is rarely reported. Classical virus neutralization test (VNT) is superior in sensitivity and specificity for detecting NAbs but is either time-consuming or laborious. In this study, a recombinant virus FA4-EGFP expressing EGFP-fiber-2 fusion protein, rather than wild type (WT) FAdV-4 was used to develop a novel VNT for detecting FAdV-4 NAbs. Specificity analysis showed that the approach only reacted with the sera against FAdV-4, not with the sera against other avian pathogens tested. The novel VNT was effective in the detection of NAbs against FAdV-4 in sera from both experimentally infected and clinically vaccinated chickens, and had good linear correlation with the classical VNT. Moreover, the novel VNT not only significantly simplifies the procedure for detection of NAbs, but also shortens the timeline to 24 h in comparison with the classical VNT with 3-4 d. All these data demonstrate that the FA4-EGFP based VNT developed here provides an efficient diagnostic method for monitoring the immunological state of the vaccination or diagnosing the clinical infection of FAdV-4 in a quick and funding-saving manner.

Abstract Recently, the outbreaks of hydropericardium-hepatitis syndrome (HHS) caused by the highly pathogenic fowl adenovirus serotype 4 (FAdV-4) have resulted in huge economic losses to the poultry industry globally. Although several inactivated or subunit vaccines have been developed against FAdV-4, live-attenuated vaccines for FAdV-4 are rarely reported. In this study, a recombinant virus FA4-EGFP expressing EGFP-Fiber-2 fusion protein was generated by the CRISPR/Cas9 technique. Although FA4-EGFP shows slightly lower replication ability than the wild type (WT) FAdV-4, FA4-EGFP was significantly attenuated in vivo compared with the WT FAdV-4. Chickens infected with FA4-EGFP did not show any clinical signs, and all survived to 14 day post-infection (dpi), whereas those infected with FAdV-4 showed severe clinical signs with HHS and all died at 4 dpi. Besides, the inoculation of FA4-EGFP in chickens provided efficient protection against lethal challenge with FAdV-4. Compared with an inactivated vaccine, FA4-EGFP induced neutralizing antibodies with higher titers earlier. All these data not only provide a live-attenuated vaccine candidate against the highly pathogenic FAdV-4 but also give a potential insertion site for developing FAdV-4-based vaccine vectors for delivering foreign antigens.

We analyze the real and imaginary parts of cBN in a broad wavelength range both 0.5-1.0 µm and 1.36-10 µm respectively, and use both Fresnel theory and Meep software package to calculate the transmittance and reflectance. The theoretical and numerical results show that the transmision and reflection on the interfaces mentioned above have an abnormal phenomenon, and the physical mechanism is analyzed. Between 0.5 μm and 1 μm, the cBN film is transparent with the transmissivity equal to the order of 1.0. When the light wavelength is in the range of 4-9 μm, the structures made of cBN and Si, such as cBN film, cBN-Si double layer film and multiple layer film, have the same optical characteristic with the highest reflectivity and lower absorptivity and zero transmissivity. These findings can be used in the structure design of the optoelectronic devices, such as the optical bandpass filter, the optical bandstop filter, the optical attenuator, the photodetector, and the solar cell.

ABSTRACT Hepatitis-hydropericardium syndrome (HHS) caused by the highly pathogenic fowl adenovirus serotype 4 (FAdV-4) has resulted in huge economic losses to the poultry industry globally. The fiber-2 gene, as a major virulence determiner, is also an important vaccine target against FAdV-4. In this study, we used a CRISPR/Cas9-based homology-dependent recombinant technique to replace the fiber-2 gene with egfp and generate a novel recombinant virus, designated FAdV4-EGFP-rF2. Although FAdV4-EGFP-rF2 showed low replication ability compared to the wild-type FAdV-4 in LMH cells, FAdV4-EGFP-rF2 could effectively replicate in LMH-F2 cells with the expression of Fiber-2. Moreover, FAdV4-EGFP-rF2 was not only highly attenuated in chickens, but also could provide efficient protection against a lethal challenge of FAdV-4. Moreover, FAdV4-EGFP-rF2 without fiber-2 could induce neutralizing antibodies at the same level as FA4-EGFP with fiber-2. These results clearly demonstrate that although fiber-2 affects the viral replication and pathogenesis of FAdV-4, it is not necessary for virus replication and induction of neutralizing antibodies; these findings provide novel insights into the roles of fiber-2 and highlight fiber-2 as an insertion site for generating live-attenuated FAdV-4 vaccines against FAdV-4 and other pathogens. IMPORTANCE Among all serotypes of fowl adenovirus, serotypes FAdV-1, FAdV-4, and FAdV-10 are unique members with two fiber genes (fiber-1 and fiber-2). Recent studies reveal that Fiber-1, not Fiber-2, directly triggers viral infection of FAdV-4, whereas Fiber-2, but not Fiber-1, has been identified as the major virulence determiner and an efficient protective immunogen for subunit vaccines. Here, we replaced fiber-2 with egfp to generate a novel recombinant virus, designated FAdV4-EGFP-rF2. In vitro and in vivo studies on FAdV4-EGFP-rF2 revealed that fiber-2 was not necessary for either virus replication or efficient protection for FAdV-4; these results not only provide a novel live-attenuated vaccine candidate against HHS, but also give new ideas for generating a FAdV-4 based vaccine vector against other pathogens.

JS001 (toripalimab) is a humanized IgG monoclonal antibody which strongly inhibits programmed cell death protein 1 (PD1). In this study, we used a different iodine isotype (nat/124/125I) to label JS001 probes to target the human PD1 (hPD1) antigen. In vitro, the half maximal effective concentration (EC50) value of natI-JS001 did not significantly differ from that of JS001. The uptake of 125I-JS001 by activated T cells was 5.63 times higher than that by nonactivated T cells after 2 h of incubation. The binding affinity of 125I-JS001 to T cells of different lineages after phytohemagglutinin (PHA) stimulation reached 4.26 nmol/L. Humanized PD1 C57BL/6 mice bearing mouse sarcoma S180 cell tumors were validated for immuno-positron emission tomography (immuno-PET) imaging. Pathological staining was used to assess the expression of PD1 in tumor tissues. The homologous 124I-human IgG (124I-hIgG) group or blocking group was used as a control group. Immuno-PET imaging showed that the uptake in the tumor area of the 124I-JS001 group at different time points was significantly higher than that of the blocking group or the 124I-hIgG group in the humanized PD1 mouse model. Taken together, these results suggest that this radiotracer has potential for noninvasive monitoring and directing tumor-specific personalized immunotherapy in PD1-positive tumors.

Abstract In recent years, hepatitis-hydropericardium syndrome (HHS) and inclusion body hepatitis (IBH) caused by fowl adenovirus (FAdV) infection have resulted in significant economic losses to the poultry industry worldwide. Epidemiological analysis revealed that serotype FAdV-8 is one of the major pathogenic FAdVs currently prevalent in domestic flocks. Although the fiber protein of FAdV plays vital roles in viral infection and pathogenesis, the B cell epitope in the fiber protein is less known. In this study, two monoclonal antibodies (mAbs) specific to fiber protein of FAdV-8, designated as 4D9 and 5F10, were prepared. Although the mAb 4D9 and 5F10 could not neutralize FAdV-8 infection, 4D9 and 5F10 showed good activities of indirect immunofluorescence, western blot and immunoprecipitation. Epitope analysis revealed that mAb 5F10 recognized 187–219aa in the fiber whereas mAb 4D9 recognized 113–149aa in the fiber. Sequence analysis showed that the epitope recognized by mAb 5F10 was conserve across serotypes FAdV-7, 8a and 8b whereas that for mAb 4D9 was only conserve in FAdV-8b. The generation of mAbs specific to fiber of FAdV-8 and the identification of the novel B cell epitopes here lay the foundation for further studying the antigenicity of the fiber and developing specific diagnosis for FAdV-8.

In recent years, the emergence of avian orthoreovirus (ARV) has caused significant losses to the poultry industry worldwide. In this study, a novel ARV isolate, designated as AHZJ19, was isolated and identified from domestic chicken with viral arthritis syndrome in China. AHZJ19 can cause typical syncytial cytopathic effect in the chicken hepatocellular carcinoma cell line, LMH. High-throughput sequencing using Illumina technology revealed that the genome size of AHZJ19 is about 23,230 bp, which codes 12 major proteins. Phylogenetic tree analysis found that AHZJ19 was possibly originated from a recombination among Hungarian strains, North American strains, and Chinese strains based on the sequences of the 12 proteins. Notably, the σC protein of AHZJ19 shared only about 50% homology with that of the vaccine strains S1133 and 1733, which also significantly differed from other reported Chinese ARV strains. The isolation and molecular characteristics of AHZJ19 provided novel insights into the molecular epidemiology of ARV and laid the foundation for developing efficient strategies for control of ARV in China.

ABSTRACT: As important immunosuppressive viruses, chicken infectious anemia virus (CIAV) and subgroup J avian leukosis virus (ALV-J) have caused huge economic losses to the poultry industry globally. Recently, the co-infection of CIAV and ALV-J frequently occurred in the domestic chicken flocks in China. However, the synergistic pathogenesis of CIAV and ALV-J has not been fully investigated. Here, a co-infection study was performed to further understand the potential synergistic pathogenesis of CIAV and ALV-J. In vitro study showed that CIAV could promote the replication of ALV-J in HD11 cells, but ALV-J could not increase the replication of CIAV. Chicken infection study showed both CIAV and ALV-J with synergistic effects caused significant body weight loss to the infected chickens. Although ALV-J had no effect on CIAV viral shedding and tissue load, CIAV did significantly increase ALV-J viremia, viral shedding and tissue load in the co-infection group. Moreover, both CIAV and ALV-J could significantly inhibit the humoral immunity to H9N2 influenza virus and serotype 4 fowl adenovirus (FAdV-4). All these data demonstrate the synergistic pathogenesis for the co-infection of CIAV and ALV-J, and highlight the positive effect of CIAV on the pathogenesis of ALV-J.

Currently, a fatal disease of hepatitis-hydropericardium syndrome (HHS) caused by serotype 4 fowl adenovirus (FAdV-4) has spread worldwide and resulted in tremendous economic losses to the poultry industry. Various vaccines against FAdV-4 were developed to control the disease; however, few live-attenuated vaccines were available. In this study, we targeted the N-terminal of fiber-1 and rescued a recombinant virus FAdV4-RFP_F1 expressing the fusion protein of RFP and Fiber-1 based on the CRISPR/Cas9 technique. In vitro studies showed that FAdV4-RFP_F1 replicated slower than the wild type FAdV-4, but the peak viral titer of FAdV4-RFP_F1 could still reach 107.0 TCID50/ml with high stability in LMH cells. Animal studies found that FAdV4-RFP_F1 not only was highly attenuated to the 2-week-old SPF chickens, but could also provide efficient protection against lethal challenge of FAdV-4. All these demonstrate that the recombinant virus FAdV4-RFP_F1 could be as an efficient live-attenuated vaccine candidate for FAdV-4, and the N-terminal of fiber-1 could be as a potential insertion site for expressing foreign genes to develop FAdV-4-based vaccine.

Influenza A virus (IAV) is widespread in wild bird reservoirs. Sixteen hemagglutinin subtypes are associated with wild waterfowl hosts; some subtypes are isolated infrequently, one of which is H12 IAV. In this study, we detected three H12 IAVs from Anascrecca and Anas formosa in Poyang Lake, China, in 2018, one of which was isolated. Phylogenetic analysis revealed that the genome sequences of the three H12 viruses belonged to the Eurasian lineage, except for PA genes and one NP gene, which belonged to the North American lineage. The growth kinetics showed that the H12 isolate grew better in A549 than MDCK cells. Moreover, although the H12 isolate cannot efficiently replicate in BALB/c mice, it can bind to both α-2,6 sialic acid (SA) and α-2,SA-linked receptors. In addition, we examined the phylodynamics of H12 viruses by Bayesian phylogeographic analysis. The results show that two major transmission routes of H12 IAVs were from Asia to Oceania and from Europe to South America, and Anas and Arenaria genera were the major hosts of the viral transmission. Our findings help us better understand the evolution of H12 IAV and highlight the need for the continued surveillance of IAVs circulating in wild birds.

In this investigation, p–Mg2Si/n–Si heterojunction photodetector (PD) is fabricated by magnetron sputtering and low vacuum annealing in the absence of argon or nitrogen atmosphere. Multilayer Graphene (MLG)/Mg2Si/Si heterojunction PD is first fabricated by transferring MLG to Mg2Si/Si heterojunction substrate using the suspended self-help transfer MLG method. After characterizing the phase composition, morphology and detection properties of Mg2Si/Si and MLG/Mg2Si/Si heterojunction PDs, the successful fabrication of the Mg2Si/Si and MLG/Mg2Si/Si heterojunction PDs are confirmed and some detection capabilities are realized. Compared with the Mg2Si/Si heterojunction PD, the light absorption and the ability to effectively separate and transfer photogenerated carriers of MLG/Mg2Si/Si heterojunction PD are improved. The responsivity, external quantum efficiency (EQE), noise equivalent power (NEP), detectivity (D*), on/off ratio and other detection properties are enhanced. The peak responsivity and EQE of the MLG/Mg2Si/Si heterojunction PD are 23.7 mA/W and 2.75%, respectively, which are better than the previous 1–10 mA/W and 2.3%. The results illustrate that the fabrication technology of introducing MLG to regulate the detection properties of the Mg2Si/Si heterojunction PD is feasible. In addition, this study reveals the potential of MLG to enhance the detection properties of optoelectronic devices, broadens the application prospect of the Mg2Si/Si-based heterojunction PDs and provides a direction for the regulation of optoelectronic devices.

Abstract Background In China, although the ALV eradication program and the MD vaccination strategy greatly reduce the disease burdens caused by the infection of ALV and MDV, the frequent emergence of novel ALV-K or vvMDV in the vaccinated chicken flock challenges the current control strategies for both diseases. Results In Guangdong Province, an indigenous chicken flock was infected with neoplastic disease. Hematoxylin–eosin staining of the tissues showed the typical characteristics of MDV and classical ALV infection. The PCR and sequencing data demonstrated that the identified MDV was clustered into a very virulent MDV strain endemic in domestic chickens in China. Moreover, subgroups ALV-A and ALV-K were efficiently recovered from two samples. The full genome sequence revealed that the ALV-K isolate was phylogenetically close to the ALV TW3593 isolate from Taiwan Province. Conclusions A co-infection of vvMDV with multiple ALV subgroups emerged in a chicken flock with neoplastic disease in Guangdong Province. The co-infection with different subgroups of ALV with vvMDV in one chicken flock poses the risk for the emergence of novel ALVs and heavily burdens the control strategy for MDV.
