The ant genus Monomorium is one of the most species-rich but taxonomically problematic groups in the hyperdiverse subfamily Myrmicinae. An East Asian species, M. triviale Wheeler, produces both reproductive queens and sterile workers via obligate thelytokous parthenogenesis. Here, we describe the immature forms of M. triviale based on light and scanning electron microscopy observations, with a note on the striking caste dimorphism in the last larval instar. The last-instar queen larvae were easily recognized by their large size, “aphaenogastroid” body shape, and rows of doorknob-like tubercles on the lateral and dorsal body surface. This type of queen-specific structure has not been found in ants in general, let alone congeneric species found in Japan. In stark contrast to the queen larvae, worker larvae showed a “pheidoloid” body shape and a body surface similar to other ants. The worker larvae were estimated to have three instars, consistent with previously described congeners. The pupae of both castes had no cocoon, a characteristic commonly described in other Myrmicinae species. In total, the developmental period from egg to adult worker averaged 59 days under 25°C. We discuss possible functions of the tubercles of queen larvae based on previous studies.

Ants use chemical signals to communicate for various purposes related to colony function. Social organization in the red imported fire ant, Solenopsis invicta, is determined by the Sb supergene, with colonies of the monogyne (single-queen) form lacking the element and colonies of the polygyne (multiple-queen) form possessing it. Polygyne workers accept new reproductive queens in their nest, but only those carrying Sb; young winged queens lacking this genetic element are executed as they mature sexually in their natal nest or as they attempt to enter a foreign nest to initiate reproduction after mating and shedding their wings. It has been suggested that queen supergene genotype status is signaled to workers by unsaturated cuticular hydrocarbons, while queen reproductive status is signaled by piperidines (venom alkaloids). We used high-throughput behavioral assays to study worker acceptance of paper dummies dosed with fractions of extracts of polygyne queens, or blends of synthetic counterparts of queen cuticular compounds. We show that the queen supergene pheromone comprises a blend of monoene and diene unsaturated hydrocarbons. Our assays also reveal that unsaturated hydrocarbons elicit discrimination by polygyne workers only when associated with additional compounds that signal queen fertility. This synergistic effect was obtained with a polar fraction of queen extracts, but not by the piperidine alkaloids, suggesting that the chemical(s) indicating queen reproductive status are compounds more polar than cuticular hydrocarbons but are not the piperidine alkaloids. Our results advance understanding of the role of chemical signaling that is central to the regulation of social organization in an important invasive pest and model ant species.

The demand for stingless bee colonies in Malaysia has considerably increased due to the rapid advance of meliponiculture in using the stingless bees as agricultural pollinators, as well as the commercialization of stingless bee products (i.e. honey, bee bread and propolis). Thus, in vitro queen rearing for a large scale and rapid colony multiplication must be developed in order to fulfil the public requirements in a short period. Little is known about the in vitro rearing of native stingless bee queen, Heterotrigona itama. Therefore, in this study, we investigated the amount of larval food required by H. itama queen by comparing three different amounts of larval food, viz., 100 µL, 120 µL and 150 µL. All treatments were controlled under 100% relative humidity for the first 6 days, and 75% relative humidity for the rest of larval development until queen adult emergence, under 30 °C incubator temperature. The results showed that larvae of H. itama treated with the highest amount of larval food (150 µL) led to 78% of the queen’s emergence, whereas larvae treated with 120 µL and 100 µL of larval food resulted in 40% and 0% of queen emergence. The dynamic survival curve showed that most of the larvae died before the pupation phase and reached constant stability afterward. The queen’s body and abdominal length were significantly greater than wild workers. Microscopy analysis showed that in vitro queen had well-developed reproductive system with a huge ovary and spermatheca, whereas wild worker had much smaller ovary without spermatheca. Outcomes from this study could help increase the number of colonies on a large scale, allowing for their use both ecologically and economically, and contribute to conservation efforts in native species of stingless bees.

Bumble bees belong to the genus Bombus, order Hymenoptera and family Bombidae, which includes approximately 250 species all over the world, out of which 48 species are found in India. Bumble bees are effective pollinators with high pollination efficiency and increasing demand of pollination services has highlighted the concept of commercialization of these pollinators. They generally have an annual life cycle with three castes viz., queen, workers and drones; only the queen undergoes diapause during winters. The oviposition rate and colony initiation percentage of the bumble bees are affected by different factors viz., foundation queen, feed, hygienic conditions, nesting material and environmental conditions. Amongst these, nesting material as well as feeding methods are the most important factors that affect the success rate and queen acceptance. Hence, it is very important to know which nesting material and feed is highly accepted by queens for colony initiation and development. The present review summarizes the literature about different domiciles and feed used for invitro rearing of bumble bees and their effect on colony development.

Subterranean termite control methods using chitin synthesis inhibitors (CSIs) aim at eliminating colonies that feed upon a bait formulation. Several benzoylurea active ingredient formulations are currently commercially available as alternative termite management strategies to liquid termiticides. Individual workers need to molt on a regular basis and CSIs interfere with such molting process, allowing sufficient time for the acquisition of a colony-wide lethal dose prior to widespread mortality. As workers progressively die, the colony eventually collapses, leaving only soldiers and primary reproductives that starve to death. One common observation is that young workers often die early owing to their relatively short molting cycle. However, the absence of brood in dying colonies raises questions about the potential fate of eggs laid by the queen. This study aims to determine if CSI baits also terminate the ability of a colony to produce a new cohort of workers by disabling the ongoing brood development. Incipient termite colonies were used to test the impact of noviflumuron on the queen’s ability to lay eggs and on the eggs' ability to hatch. Our results showed that queens in colonies exposed to CSI not only initially laid less eggs than the control queens, but eggs also did not develop and were progressively cannibalized, eventually leading to colony establishment failure. This result implies that queens of mature colonies exposed to CSI would lose the ability to lay viable eggs as the colony collapses, leading to an absence of worker replacement, aiding in colony elimination.

No. of bitstreams: 0 Previous issue date: 2021-12-01 Background: Much of the complex anatomy of a holometabolous insect is built from disc-shaped epithelial structures found inside the larva, i.e., the imaginal discs, which undergo a rapid differentiation during metamorphosis. Imaginal discs-derived structures, like wings, are built through the action of genes under precise regulation. Results: We analyzed 30 honeybee transcriptomes in the search for the gene expression needed for wings and thoracic dorsum construction from the larval wing discs primordia. Analyses were carried out before, during, and after the metamorphic molt and using worker and queen castes. Our RNA-seq libraries revealed 13,202 genes, representing 86.2% of the honeybee annotated genes. Gene Ontology analysis revealed functional terms that were caste-specific or shared by workers and queens. Genes expressed in wing discs and descendant structures showed differential expression profiles dynamics in premetamorphic, metamorphic and postmetamorphic developmental phases, and also between castes. At the metamorphic molt, when ecdysteroids peak, the wing buds of workers showed maximal gene upregulation comparatively to queens, thus underscoring differences in gene expression between castes at the height of the larval-pupal transition. Analysis of small RNA libraries of wing buds allowed us to build miRNA-mRNA interaction networks to predict the regulation of genes expressed during wing discs development. Conclusion: Together, these data reveal gene expression dynamics leading to wings and thoracic dorsum formation from the wing discs, besides highlighting caste-specific differences during wing discs metamorphosis. Ants are highly polyphenic Hymenoptera, with at least three distinct adult forms in the vast majority of species. Their sexual dimorphism, however, is overlooked to the point of being a nearly forgotten phenomenon. Using a multimodal approach, we interrogate the near total head microanatomy of the male of Dorylus helvolus, the "sausagefly," and compare it with the conspecific or near-conspecific female castes, the "driver ants." We found that no specific features were shared uniquely between the workers and males to the exclusion of the queens, indicating independence of male and worker development; males and queens, however, uniquely shared several features. Certain previous generalizations about ant sexual dimorphism are confirmed, while we also discover discrete muscular presences and absences, for which reason we provide a coarse characterization of functional morphology. Based on the unexpected retention of a medial carinate line on the structurally simplified mandible of the male, we postulate a series of developmental processes to explain the patterning of ant mandibles. We invoke functional and anatomical principles to classify sensilla. Critically, we observe an inversion of the expected pattern of male-queen mandible development: male Dorylus mandibles are extremely large while queen mandibles are poorly developed. To explain this, we posit that the reproductive-limited mandible phenotype is canalized in Dorylus, thus partially decoupling the queen and worker castes. We discuss alternative hypotheses and provide further comparisons to understand mandibular evolution in army ants. Furthermore, we hypothesize that the expression of the falcate phenotype in the queen is coincidental, that is, a "spandrel," and that the form of male mandibles is also generally coincidental across the ants. We conclude that the theory of ant development and evolution is incomplete without consideration of the male system, and we call for focused study of male anatomy and morphogenesis, and of trait limitation across all castes.

Research on cuticular hydrocarbons of solitary and eusocial bees has greatly contributed to our understanding of the evolution of eusociality. However, to understand the transition from simple to complex societies, understanding subsocial species is necessary. Subsociality, defined as prolonged parental care, is a pre-requisite for eusociality yet cuticular hydrocarbons of subsocial bees are not well characterized. Here we present the first characterization and analysis of cuticular hydrocarbons in a subsocial bee, Ceratina calcarata Robertson. We found that cuticular hydrocarbons signal both age and reproductive status in this species. Pre-reproductive, virgin females overexpress three compounds, heneicosane, farnesol, and nonadecane. We show that pentacosane, a known fertility signal, is likely used to signal reproduction in C. calcarata. Since we found a possible reproductive signal in a subsocial bee this suggests that queen pheromones likely evolved from pre-existing fertility signals.

Forage cactus is a perennial crop, which has been widely exploited for feeding ruminants in the semiarid region of different countries around the world. The main objective of this chapter is to describe the use and importance of spineless cactus as forage, desertification mitigation, source of water for animals and a source of income for producers in semiarid regions. The main species explored in Brazil are Opuntia spp. and Nopalea spp., due to characteristics such as resistance to pests, productivity, water-use efficiency and demand for soil fertility. The productivity of the species in a region will depend on its morphological characteristics, plant spacing, planting systems and its capacity to adapt to climatic and soil conditions. In other parts of the world, cactus species are the most cosmopolitan and destructive among invasive plants. However, the use of spineless forage cactus in areas where it can develop normally and may become the basis for ruminants’ feed would increase the support capacity production systems. Thus, specifically for Brazil’s semiarid region these species can make the difference as forage for animal feeding, cultivated as monoculture or intercropped, for soil conservation and desertification mitigation, source of water for animals, preservation of the Caatinga biome and be a potential source of income for producers if cultivated as vegetable for nutritional properties and medicinal derivative of fruits and cladodes for exports.

Two queen ant specimens, one alate and one dealate, from mid-Cretaceous (Late Albian–Early Cenomanian) Burmese amber are herein reported as belonging Haidomyrmex cerberus Dlussky, 1996. This is the first discovery and documentation of an alate queen in Haidomyrmex. Compared with workers of Haidomyrmex cerberus, alate and dealate queens are larger in body size, have smaller compound eyes, a longer antennal scape, more complex mandibles, and a relatively large-sized metasoma. It is hypothesized that these differences are due to caste differences.

The complex system of communication used by social insects is responsible for their success on Earth. Cuticular hydrocarbons (CHCs) are considered the most commonly used form of communication among social insects. The best-known function of CHCs is to distinguish nestmates, providing cues to the whole colony to identify potential threats. In some species, CHCs function as queen pheromones, which contribute to the queens’ ability to regulate the reproductive division of labor. Additionally, differences of CHCs were described in castes and they are also important in sex recognition. Here, we reinvestigated the chemical composition on the cuticle of German wasp Vespula germanica and we found that proportions of linear alkanes were more abundant in the cuticle of queens than other castes. Chemical analysis between caste and sex, including data of virgin queens and males, showed that each group possesses different compositions on their cuticle, being mostly quantitative differences. Thus, individuals of V. germanica may recognize and discriminate caste and sex through subtle chemical variations, or alternatively, based on templates used in nestmate recognition. The chemical analysis provides correlative evidence of CHCs being used as cues in the chemical communication of this species.

Despite the identical genomic context, trophocytes and oenocytes in worker bees exhibit aging-related phenotypes, in contrast to the longevity phenotypes in queen bees. To explore this phenomenon at the molecular level, we evaluated the age-associated transcriptomes of trophocytes and oenocytes in worker bees and queen bees using high-throughput RNA-sequencing technology (RNA-seq). The results showed that (i) while gene expression profiles were different between worker and queen bees, they remained similar between young and old counterparts; (ii) worker bees express a high proportion of low-abundance genes, whereas queen bee transcriptomes display a high proportion of moderate-expression genes; (iii) genes were upregulated to a greater extent in queen bees vs. worker bees; and (iv) distinct aging-related and longevity-related candidate genes were found in worker and queen bees. These results provide new insights into the cellular aging and longevity of trophocytes and oenocytes in honey bees. Identification of aging-associated biomarker genes also constitutes a basis for translational research of aging in higher organisms. Abstract Recent bumble bee declines have prompted the development of novel population monitoring tools, including the use of putatively non-lethal tarsal clipping to obtain genetic material. However, the potential side effects of tarsal clipping have only been tested in the worker caste of a single domesticated species, prompting the need to more broadly test whether tarsal clipping negatively affects sampled individuals. To determine if tarsal clipping reduces queen survivorship and colony establishment, we collected wild queens of Bombus vosnesenskii and clipped tarsi from a single leg of half the individuals. We reared captive queens and estimated survivorship and nest establishment success. We also clipped tarsi of workers from a subset of colonies across a range of body sizes. We found no consistent negative effect of clipping on queen survival. In the first year, clipped nest-searching queens suffered heavy mortality, but there was no effect on foraging queens. The following year, we found no effect of clipping on queen survival or establishment. Clipping did not reduce overall worker survival but reduced survivorship for those in the smallest size quartile. Implications for insect conservation Our findings suggest tarsal clipping does not have consistent negative effects on individual survival. However, our results varied with queen behavioral state, year, and worker size, suggesting differences within and among species and interactions with landscape stressors warrant further study. In the interim, we recommend researchers and conservationists minimize the use of tarsal clipping for sensitive species, populations, or small workers except in cases of exceptional scientific need.

In swarming season, honeybee queens emit two kinds of queen piping signals: tooting and quacking. These signals are considered to be important for swarming, and honeybees distinguish between the two signals through differences in their temporal structures. In this study, we revealed that the piping signals of two honeybee species, Apis cerana and Apis mellifera, had different temporal structures. The tooting of A. cerana consisted of almost one long syllable, while that of A. mellifera was constructed from several syllables. The quacking of both species comprised around 50 short syllables, but the duration and period of the syllables of A. cerana were about half those of A. mellifera. The findings provide new insights that reveal the mechanisms of signal discrimination and the functions of the signals.

Honey bee queen quality is a critical factor of colony performance. Indications of such qualities can manifest themselves through morphological traits such as wet weight and thorax width. Improving such characteristics is driven in part by nutritional provision in queen-cell-builder hives. We investigated the potential to improve queen quality by adding coenzyme Q10 (endogenous antioxidant) and caffeine (central nervous system stimulator) to feeder syrup in queen-cell-builder colonies for 15 and 20 days prior to grafting, two sets of queens were reared. We recorded subsequent wet weight, body length, head width and length, thorax and wing width and length, and spermathecae diameter. The queen-cell acceptance rate was not affected by either treatment or graft period. Coenzyme Q10 increased wet weight, body and wing length in the first graft, and thorax width, wing length and spermathecae diameter in the second graft. The caffeine treatment increased head and thorax length in first graft and thorax width in the second. A mix of the two substances (coenzyme Q10 and caffeine) increased head width in the first graft and spermathecae diameter in the second graft. This study suggests that the application of coenzyme Q10 to cell-builder colonies at least 15 days prior to grafting can increase reared wet weight (the most significant quality indicator) and thorax width of queen bees.

The endemic Tasmanian Digger Wasp Williamsita tasmanica is only known from 13 registered museum specimens, with little recorded on its life history. A nesting aggregation of females was observed over a 25-day period, enabling the first detailed observations of host log, nest design, prey and egg deposition, and behavioural patterns to be made. Cells were provisioned with large flies (order Diptera) of nine species from nine genera and four families to observe behaviour. This aggregation used a large eucalypt with an exposed core of friable brown rot for nesting. Our study showed that the day-to-day behaviour of the female is predictable, centring on nest construction, hunting large flies and egg-laying. This information has been lodged on the Atlas of Living Australia and voucher specimens collected for the Queen Victoria Museum and Art Gallery to facilitate further taxonomic study.

Queen mating frequency is an important reproductive trait of the western honeybee Apis mellifera. Yet, it demands more attention when investigated under extreme or confined ecosystems. Queen mating frequency of the Yemeni Honeybee A. m. jemenetica was estimated under Saudi Arabia desert conditions, Riyadh (24°71′36″N, 46°67′53″E). Mating of queens took place after 8–13 days from emergence. Duration of mating flight ranged between 26 and 39 min. Subsequently, six microsatellite loci were used to genotype queen's progeny (n = 30 workers/queen). The average number of drone alleles using workers genotypes ranged between 5.83 ± 0.31 and 6.33 ± 1.09. However, effective paternal allele number was extremely low and ranged between 3.35 ± 0.34 and 3.60 ± 0.40. This relatively low mating frequency of the Yemeni honeybee, A. m. jemenetica, might have striking effect on the overall colony survival. Providentially, this relatively low mating frequency does not impact colonial heterozygosity, shown in this study (0.66 ± 0.07–70 ± 0.04), adversely. These results may affect hive survivability and entails distinctive management practices under such conditions.

Termites are major pests of houses and buildings, and also living plants such as agricultural crops, trees in forests, urban areas and gardens. However, in Japan, the basic ecology of termites nesting in living trees is not fully understood. In this study, we observed 255 colonies (nests) of the drywood termite Neotermes sugioi, collected in the field on Okinawa Island, and reported the frequency composition of the reproductive castes, the size of wood with termite gallery, the population size of colonies, and the relative position of the reproductive and non-reproductive castes within nests. Most colonies were headed by a primary queen and a primary king. However, colonies headed only by primary queens, primary kings, or neotenic kings, each accounted for approximately 5% of the colonies. A colony size of 1,000– 4,000 individuals (2058.2 ± 1695.0 [mean ± SD]) was common and the average length of colony branches was less than 100 cm. Queens and kings were found in the same or nearby nest areas, and more predominantly in the central or root side of nest wood areas. The termites may experience colony fragmentation and reproductive loss as a consequence of typhoons. Incipient colonies (i.e., colonies at an early stage of development) were found on 11.3% of branches of Leucaena leucocephala that did not show any obvious signs of infestation. In future research, it will be necessary to update the list of trees damaged by this termite species, compare the damage by tree species, and evaluate the economic impact.

In this study, we investigated the morphological characteristics of three glands that are associated with the labium and the maxillae in the female castes as well as males of the ant Camponotus japonicus Mayr, 1866 using serial semithin sections and scanning electron microscopy (SEM). We propose to name these glands as the postmentum base gland, the stipes base gland and the cardo base gland. The cardo base gland represents what was previously known as the maxillary gland, the two other glands are novel findings. They all belong to class-3 glands and have no reservoir for the storage of secretion. Their morphology is similar among the studied categories (major worker, minor worker, queen and male). The postmentum base gland is made up by 10–15 secretory units that open through the articulation membrane on both sides of the base of the labial postmentum with no significant differences among castes. The stipes base gland is formed by a cluster of secretory units that open at the articulation membrane between cardo and stipes. This gland is much larger in major workers (almost 140 cells) than in the other castes (approx. 50–60 cells). The cardo base gland opens through the membrane that connects the cardo and stipes to the head capsule. The number of cells of this gland is much larger in major workers and queens (approximately 190 and 150, respectively) than in minor workers and males (approximately 45 and 40, respectively). No information is yet available on the function of the glands.

Naked mole-rats are eusocial mammals that live in hierarchies consisting of one breeding female, one to three male consorts and their reproductively suppressed offspring. A ‘disperser morph’ subcaste has been suggested with a subset of nonbreeders exhibiting motivation to leave their natal colony and mate with unfamiliar conspecifics. To test the hypotheses that intrinsic colony variables (e.g. population density, sex ratio, queen temperament) influence the dispersal phenotype, and that males and females differ in responsiveness to these variables, we evaluated dispersal behaviour in 17 laboratory colonies. Queen aggression was associated with the number of female, but not male, dispersers, although dispersers were not themselves targets of queen aggression. Female dispersers were more aggressive than their nondispersing sisters, although still less aggressive than queens overall. Following outpairing with an unfamiliar opposite-sex animal, dispersers and nondispersers produced litters at similar rates, demonstrating that motivation to leave the colony, and not anticipatory reproductive maturation, is the key to successful dispersal. Collectively, these data suggest that aggressive naked mole-rat queens motivate dispersal in their daughters and that female dispersers show traits consistent with successful queens (e.g. aggression).

Milkweed butterflies in the genus Danaus are studied in a diverse range of research fields including the neurobiology of migration, biochemistry of plant detoxification, host-parasite interactions, evolution of sex chromosomes, and speciation. We have assembled a nearly chromosomal genome for Danaus chrysippus (known as the African Monarch, African Queen, and Plain Tiger) using long read sequencing data. This species is of particular interest for the study of genome structural change and its consequences for evolution. Comparison with the genome of the North American Monarch Danaus plexippus reveals generally strong synteny, but highlights three inversion differences. The three chromosomes involved were previously found to carry peaks of intra-specific differentiation in D. chrysippus in Africa, suggesting that these inversions may be polymorphic and associated with local adaptation. The D. chrysippus genome is over 40% larger than that of D. plexippus, and nearly all of the additional ∼100 Megabases of DNA comprises repeats. Future comparative genomic studies within this genus will shed light on the evolution of genome architecture.

Bumblebees are important pollinating insects, but many species have suffered declines over the last century. Long-tongued bumblebees have been identified as particularly at risk, partly due to their more selective diet. Attempts to study these species in captivity have been impeded by stress-induced behaviours which cause queens to kill or abandon their brood. Here, we attempt to further develop techniques, using queen pairing and Bombus terrestris cocoons, to successfully rear two common long-tongued bumblebee species (B. pascuorum and B. hortorum) in captivity. Approximately half of queens laid eggs and 29% produced workers. Although challenges remain, there is a great deal to be gained from optimising the captive rearing of these species.

The current study aimed to investigate the important reproductive biology and morphology of A.m. jemenitica queens and drones through measuring the weight of virgin and mated queens, size and weight of spermathecae, weight of ovaries, number of ovarioles, quantity and viability of semen in queen and drones. Accordingly, the average weights of 0.139 ± 0.01 g and 0.143 ± 0.013 g recorded for virgin and mated queens respectively. The sizes of spermathecae were 1.248 ± 0.103 mm and 1.25 ± 0.022 mm for virgin and mated queens respectively. The mean weight of ovaries was 0.013 ± 0.003 g and the numbers of ovarioles varied from 124 to 163 with the mean of 142.9 ± 9.47 and with no significant difference between virgin and mated queens. The average number of stored sperm per spermathecae of mated queen was estimated to be 4.202 ± 0.613 million with the viability of 80.39%. The average number of sperm per drone recorded was 8,763,950 ± 1,633,203.15 with viability of 79.54 ± 6.70%. In general, the current study revealed that the values recorded for reproductive biology and morphological characters of A. m. jemenitica queens and drones were relatively lower than values recorded for other Apis mellifera races. This mainly could be associated with the body size of the race which is known to be the smallest race among A. mellifera races. Moreover, the harsh environmental conditions of the regions, high temperature, low humidity and limited resources may have contributed for the smaller biological and morphological values. The information will serve as a base in future selection and breeding of program of the race. English sea wall flood defences support an important grassland habitat for bumblebees (Bombus spp.). However, annual cutting in midsummer (July–August) could negatively affect them. The mowing regime on a sea wall at Goldhanger Creek in Essex was changed to a late cut (after 15 September) and nest-searching behaviour was compared with an adjacent sea wall cut in midsummer (normal cut in August). Sward height and the number of queens nest-searching were significantly higher on the normal cut sea wall than on the late cut one. Tall grass swards resulting from a cut early in the previous summer may be important for nest-searching compared to those mown late, although there is probably an interaction with sea wall aspect and soil type. The landward slope was also more attractive for nesting than the flat folding (berm) due to the longer vegetation where nests under construction were located (of Bombus humilis and B. terrestris).

In the fungus-growing ant genus Atta, foundress queens nourish their brood and incipient fungus gardens with nutrients derived from trophic eggs. We discovered a third kind of egg laid by Atta foundresses in addition to reproductive and trophic eggs. We use fluorescent microscopy to show that this third type of eggs represents reproductive eggs that are unviable and fail to develop. Unviable reproductive eggs are somewhat larger (≈ 490 µm × 317 µm) than regular reproductive eggs (≈ 425 µm × 240 µm), but smaller than trophic eggs (≈ 640 µm × 530 µm). Trophic eggs liquify by some endogenous process within 24 h after oviposition to release nutrient contents. Unviable reproductive eggs do not liquify, but unviable reproductive eggs can be digested by the fungus, whereas reproductive eggs are not digested by the fungus to complete their development. We also report the first observation for A. texana of nanitic males (presumably diploid males) that were killed by the foundress queen shortly after the males’ eclosion.

The study of the social organization of ants may help to understand why some species are able to persist in natural forests after fragmentation and anthropogenic disturbances. The Neotropical ant Gnamptogenys bisulca has shown to be a biological indicator of habitat quality in Andean montane forests and we proposed to explain this characteristic by investigating its social structure and the spatial distribution of colonies at fine scale. In eight 100 m2 plots located in four forests in the western Colombian Andes, the position of G. bisulca colonies was recorded for spatial distribution and their social composition described: ergatoid females were found in most of the nests, isolated or together with normal queens. As ergatoids and queens were dissected to examine their reproductive status, it appeared that apparent polygyny represented, in fact, effective monogyny, with other potential reproductive females being unmated or inhibited. In the few cases of mixed colonies, a queen was always at the head of the colony. Local spatial distribution was random and did not fit the hypothesis of nesting by fission, as generally observed in the case of the presence of ergatoids. However, the social structure was significantly different according to the site, the most preserved forest showing no or rare ergatoids (but queens) compared to the other sites. Our results indicated that the presence of ergatoids in G. bisulca may help colonies to adapt to slightly disturbed habitat contexts.

The growth of imaginal discs in holometabolous insects is coordinated with larval growth to ensure the symmetrical and proportional development of the adult appendages. In ants, the differential growth of these discs generates distinct castes-the winged male and queen castes and the wingless worker caste. In the hyperdiverse ant genus Pheidole, the worker caste is composed of two morphologically distinct subcastes: small-headed minor workers and larger, big-headed, soldiers. Although these worker subcastes are completely wingless, soldier larvae develop rudimentary forewing discs that function in generating the disproportionate head-to-body scaling and size of soldiers. It remains unclear, however, how rudimentary forewing discs in soldier larvae are coordinated with other imaginal discs. Here we show, using quantitative nano-CT imaging and three-dimensional analyses, that the increase in the volume of the soldier rudimentary forewing discs is coordinated with larval size as well as with the increase in the volume of the leg and eye-antennal (head) discs. However, relative to larval size, we found that when the rudimentary forewing discs appear during the last larval instar, they are relatively smaller but increase in volume faster than that of the head (eye-antennal) and leg discs. These findings show that the rudimentary wing disc in soldier larvae has evolved novel patterns of inter-organ coordination as compared with other insects to generate the big-headed soldier caste in Pheidole. More generally, our study raises the possibility that novel patterns of inter-organ coordination are a general feature of rudimentary organs that acquire novel regulatory functions during development and evolution.

Ophryocystis elektroscirrha McLaughlin & Myers is a specific parasite of butterflies of the genus Danaus. However, experimentally, it can complete its cycle in other butterflies. This study presents the first case of infection in the queen butterfly, Danaus gilippus (Cramer), in Central Mexico. Prevalence of the parasite in D. gilippus was 7%, in contrast in D. plexippus was almost 100%. Because of known negative effects on resident populations of monarch butterflies, it is important to monitor the prevalence of the parasite in queen butterflies and assess effects it might have on their fitness in the ecosystem.

Viruses, and in particular the deformed wing virus (DWV), are considered as one of the main antagonists of honey bee health. The ‘suppressed in ovo virus infection’ trait (SOV) described for the first time that control of a virus infection can be achieved from genetically inherited traits and that the virus state of the eggs is indicative for this. This research aims to explore the effect of the SOV trait on DWV infections in queens descending from both SOV-positive (QDS+) and SOV-negative (QDS–) queens. Twenty QDS+ and QDS– were reared from each time four queens in the same starter–finisher colony. From each queen the head, thorax, ovaries, spermatheca, guts and eviscerated abdomen were dissected and screened for the presence of the DWV-A and DWV-B genotype using qRT-PCR. Queens descending from SOV-positive queens showed significant lower infection loads for DWV-A and DWV-B as well as a lower number of infected tissues for DWV-A. Surprisingly, differences were less expressed in the reproductive tissues, the ovaries and spermatheca. These results confirm that selection on the SOV trait is associated with increased virus resistance across viral genotypes and that this selection drives DWV towards an increased tissue specificity for the reproductive tissues. Further research is needed to explore the mechanisms underlying the interaction between the antiviral response and DWV.

We describe the queen caste of the Arabian endemic species Tapinoma wilsoni Sharaf & Aldawood, 2012 from the type locality (Dhi Ayn Archaeological Village) located in the southwestern mountains of the Kingdom of Saudi Arabia. We present the first illustrated key to the Arabian TapinomaFoerster, 1850 based on the queen castes. Three species are recognised from the Arabian Peninsula,T. melanocephalum (Fabricius, 1793), T. simrothi Krausse, 1911 and T. wilsoni Sharaf & Aldawood, 2012.

Simple Summary Honey bees rely on pollen and nectar to provide nutrients to support their yearly colony cycle. Specifics of the cycle differ among geographic regions as do the species of flowering plants and the nutrients they provide. We examined responses of honey bees from two different queen lines fed pollens from locations that differed in floral species composition and yearly colony cycles. We detected differences between the queen lines in the amount of pollen they consumed and the size of their hypopharyngeal glands (HPG). There were also seasonal differences between the nutrient composition of pollens. Spring pollens collected from colonies in both locations had higher amino and fatty acid concentrations than fall pollens. There also were seasonal differences in responses to the pollens consumed by bees from both queen lines. Bees consumed more spring than fall pollen, but digested less of it so that bees consumed more protein from fall pollens. Though protein consumption was higher with fall pollen, HPG were larger in spring bees. Abstract Honey bee colonies have a yearly cycle that is supported nutritionally by the seasonal progression of flowering plants. In the spring, colonies grow by rearing brood, but in the fall, brood rearing declines in preparation for overwintering. Depending on where colonies are located, the yearly cycle can differ especially in overwintering activities. In temperate climates of Europe and North America, colonies reduce or end brood rearing in the fall while in warmer climates bees can rear brood and forage throughout the year. To test the hypothesis that nutrients available in seasonal pollens and honey bee responses to them can differ we analyzed pollen in the spring and fall collected by colonies in environments where brood rearing either stops in the fall (Iowa) or continues through the winter (Arizona). We fed both types of pollen to worker offspring of queens that emerged and open mated in each type of environment. We measured physiological responses to test if they differed depending on the location and season when the pollen was collected and the queen line of the workers that consumed it. Specifically, we measured pollen and protein consumption, gene expression levels (hex 70, hex 110, and vg) and hypopharyngeal gland (HPG) development. We found differences in macronutrient content and amino and fatty acids between spring and fall pollens from the same location and differences in nutrient content between locations during the same season. We also detected queen type and seasonal effects in HPG size and differences in gene expression between bees consuming spring vs. fall pollen with larger HPG and higher gene expression levels in those consuming spring pollen. The effects might have emerged from the seasonal differences in nutritional content of the pollens and genetic factors associated with the queen lines we used.

We found that the growth rate of the rudimentary forewing discs differs from the leg discs but is similar to the growth of the head (eye-antennal) discs relative to larval size, suggesting that growth of each type of imaginal disc may be differentially regulated. In addition to their larger size, the soldier eye-antennal discs increase in width as they undergo morphogenesis to generate the characteristic shape of the large soldier head, suggesting that the rudimentary forewing discs may regulate their patterning in addition to their growth. Finally, we observe little growth of the leg and eye-antennal discs during the bipotential stage, while in minor worker development these discs grow at similar rates to one another in coordination with larval size to generate the smaller minor worker subcaste. Our results suggest that rudimentary organs with regulatory functions may participate in new patterns of inter-organ coordination and regulation to produce novel phenotypes and complex worker caste systems. We provide characterization of larval development and imaginal disc growth and morphogenesis with the aim of highlighting this as an emerging system for the study of rudimentary organs during development and evolution.

As a critical stage in the life cycle of ant colonies, nest establishment depends on external and internal factors. This study investigates the effect of the number of queens on queen and worker behavior during nest establishment in invasive Argentine ants (Linepitema humile) and native Mediterranean Tapinoma nigerrimum. We set up experimental colonies with the same number of workers but with one or six queens. At different time points, we recorded the positions of queens and workers inside and outside the nest. Our results highlight the influence of the number of queens on the position of queens and workers with between-species differences. Queens of both species entered the nests more quickly when there were six queens. During nest establishment, more workers were inside nests with six queens for both species, with this effect being greater for T. nigerrimum. Once nests were established, fewer workers of both species were engaged in nest maintenance and feeding in nests with six queens, T. nigerrimum had fewer workers engaged in patrolling. These results suggest that the number of queens is a key factor driving queen and worker behavior during and after nest establishment with different species responses.

Some species of the ant genus Forelius can be misclassified as the genus Tapinoma due to the convergent reduction of the petiolar scale. I review the taxonomic status of Tapinoma antarcticum Forel 1904 and Tapinoma heyeri Forel 1902. Morphological evidence supports the transfer of these names to the ant genus Forelius. Proposed taxonomic changes are as follows: Forelius antarcticus (Forel) comb. nov. (=Forelius fazi (Santschi 1923) comb. nov., =Forelius eidmanni Goetsch 1933 comb. rev.); Forelius heyeri (Forel) comb. nov. (=Forelius heyeri risii (Forel 1912) comb. and syn. nov., =Forelius pusillus (Santschi 1922) syn. nov., =Forelius tucumanus (Kusnezov 1953) syn. nov.). Lectotypes for Forelius antarcticus and Forelius heyeri are designated. The worker of Forelius heyeri, as well as the worker and queen of Forelius antarcticus are diagnosed and redescribed. Multifocus images for lectotype workers are provided. Morphological variation within and between species of Forelius with rounded spiracles is discussed.

In 1999, Barry Bolton postulated the presence of a basimandibular gland in the mandibles in all species of the ant genus Strumigenys, solely based on scanning microscopy observations. We now confirm the presence of this putative gland in the proximal outer part of the mandibles of 22 investigated species by histological and ultrastructural examination, including 10 short- and 12 long-mandibulate species. All species have a basimandibular gland, that is formed by 15&ndash, 25 &micro, m thick epithelial cells and belongs to class-1 following the standard classification of insect exocrine glands. We consider it a novel gland because of its peculiar bowl-shape and special arrangement of the microvilli that are confined to large vacuolar spaces instead of reaching the cuticle. The gland is most pronounced in S. mutica, particularly in the queen. In addition to this gland, we also found scattered class-3 intramandibular gland cells in the mandibles. Queens of S. mutica are peculiar in having a cluster of these cells in the distal tip of their mandibles. As this species is a social parasite, further research is required to determine whether the development of these mandibular glands is related to its parasitic lifestyle.

International audience; AbstractThe queen and worker castes of the honey bee are very distinct phenotypes that result from different epigenomically regulated developmental programs. In commercial queen rearing, it is common to produce queens by transplanting worker larvae to queen cells to be raised as queens. Here, we examined the consequences of this practice for queen ovary development and genome-wide methylation. Queens reared from transplanted older worker larvae weighed less and had fewer ovarioles than queens reared from transplanted eggs. Methylome analyses revealed a large number of genomic regions in comparisons of egg reared and larvae reared queens. The methylation differences became more pronounced as the age of the transplanted larva increased. Differentially methylated genes had functions in reproduction, longevity, immunity, and metabolic functions suggesting that the methylome of larval reared queens was compromised and more worker-like than the methylome of queens reared from eggs. These findings caution that queens reared from worker larvae are likely less fecund and less healthy than queens reared from transplanted eggs.

Insects are the most numerous group of animals on earth. As such, they have developed many different ways of sexual reproduction. In most primitive species, males deposit a sac full of sperm called spermatophore lying around, waiting to be found and picked up by females. However, as this is not the most efficient way to reproduce, many insects developed copulation and internal fertilization. In many insects, males and females only mate once, and after mating, females often store the sperms for the rest of their life. Such an example is honeybees. The queen is fertilized only once and can later determine whether she will produce male or female progeny by fertilizing the eggs (thus producing females) or leaving them unfertilized (producing drones). In some insects (and also spiders), mating is a dangerous affair for the males, as females often consume males after the copulation.

Honey bee (Apis mellifera) queens have a remarkable organ, the spermatheca, which successfully stores sperm for years after a virgin queen mates. This study uniquely characterized and quantified the transcriptomes of the spermathecae from mated and virgin honey bee queens via RNA sequencing to identify differences in mRNA levels based on a queen’s mating status. The transcriptome of drone semen was analyzed for comparison. Samples from three individual bees were independently analyzed for mated queen spermathecae and virgin queen spermathecae, and three pools of semen from ten drones each were collected from three separate colonies. In total, the expression of 11,233 genes was identified in mated queen spermathecae, 10,521 in virgin queen spermathecae, and 10,407 in drone semen. Using a cutoff log2 fold-change value of 2.0, we identified 212 differentially expressed genes between mated and virgin spermathecal queen tissues: 129 (1.4% of total) were up-regulated and 83 (0.9% of total) were down-regulated in mated queen spermathecae. Three genes in mated queen spermathecae, three genes in virgin queen spermathecae and four genes in drone semen that were more highly expressed in those tissues from the RNA sequencing data were further validated by real time quantitative PCR. Among others, expression of Kielin/chordin-like and Trehalase mRNAs was highest in the spermathecae of mated queens compared to virgin queen spermathecae and drone semen. Expression of the mRNA encoding Alpha glucosidase 2 was higher in the spermathecae of virgin queens. Finally, expression of Facilitated trehalose transporter 1 mRNA was greatest in drone semen. This is the first characterization of gene expression in the spermathecae of honey bee queens revealing the alterations in mRNA levels within them after mating. Future studies will extend to other reproductive tissues with the purpose of relating levels of specific mRNAs to the functional competence of honey bee queens and the colonies they head.

Programmed cell death (PCD) in unicellular organisms is in some instances an altruistic trait. When the beneficiaries are clones or close kin, kin selection theory may be used to explain the evolution of the trait, and when the trait evolves in groups of distantly related individuals, group or multilevel selection theory is invoked. In mixed microbial communities, the benefits are also available to unrelated taxa. But the evolutionary ecology of PCD in communities is poorly understood. Few hypotheses have been offered concerning the community role of PCD despite its far-reaching effects. The hypothesis we consider here is that PCD is a black queen. The Black Queen Hypothesis (BQH) outlines how public goods arising from a leaky function are exploited by other taxa in the community. Black Queen (BQ) traits are essential for community survival, but only some members bear the cost of possessing them, while others lose the trait In addition, BQ traits have been defined in terms of adaptive gene loss, and it is unknown whether this has occurred for PCD. Our conclusion is that PCD fulfils the two most important criteria of a BQ (leakiness and costliness), but that more empirical data are needed for assessing the remaining two criteria. In addition, we hold that for viewing PCD as a BQ, the original BQH needs to include social traits. Ants exhibit a striking variety of lifestyles, including highly specialist or mutualist species. The minute blind workers of the African genus Melissotarsus chew tunnels in live trees to accommodate their obligate partner scale insects. Their modified legs are adapted for tunneling, but are unsuited for walking outside, confining these ants to their initial host tree. Here, we investigated whether this unique lifestyle results in complex patterns of genetic diversity at different scales, from the same tree to different populations. Using 19 microsatellite markers, we assessed their mating strategy and colony structure among and across populations in South Africa. We showed that only one queen reproduces within a colony, mated with up to three males. Yet, several inseminated dealate queens are present in colonies; one probably replaces the older queen as colonies age. The reproduction of a single queen per colony at a given time results in genetic differentiation between colonies, even those located on the same tree. Overall, we discussed how the slow process of colony digging under the bark and the lack of worker patrolling above the bark might result in reduced competition between colonies and allow several secluded colonies to cohabit in a cramped space on a tree.

In the United States are 37 endemic Strumigenys ant species belonging to a single temperate and recently radiated Nearctic clade formerly classified as Pyramica (non-trap jaw Strumigenys Smith1). We report the discovery of a new unrelated and morphologically distinct endemic Nearctic Strumigenys we described as Strumigenys superstes sp. nov. known only from a single queen collected in a pitfall trap at high altitude in northern Arizona. In comparing Strumigenys superstes sp. nov. morphology with queens of other closely related species, we showed Strumigenys superstes sp. nov. is the only temperate member of the otherwise tropical (Panamanian) clade of short-mandibled trap jaw species formerly belonging to the genus Glamyromyrmex (now forming the Strumigenys excisa species-group).

Bumble bees are essential, efficient pollinators of numerous crops, and scientists are increasingly concerned about their global decline. Although several potential causes have been put forward, the concurrent modification of landscape structure and pesticide use by agriculture are often pointed out as the main drivers of the decline. Yet, the mechanisms through which these factors cause species to decline remain elusive. Most field studies have been conducted at the individual level, and the few studies that addressed higher levels of organization only covered narrow ecological conditions. We undertook a field experiment in spring 2016 to overcome these limitations by placing four commercial Bombus impatiens colonies in each of 20 sites located along a gradient of agricultural intensity in southern Québec, Canada. We hypothesized that landscape habitat composition and configuration, as well as local floral resources, would affect colony development. We expected colonies surrounded by low‐intensity and/or flowering crops in heterogeneous landscapes to develop better and live longer, partly linked to more abundant and diversified food resources, than colonies embedded in landscapes dominated by high‐intensity crops. Colonies were weighed once a week, and workers were captured to assess pollen load diversity. Final colony weight, queen production, and presence of depredators (Aphomia sociella, Lepidoptera: Pyralidae) were also recorded. Landscape habitat composition and configuration were characterized within 1 km of colonies. Local availability of floral resources was assessed within 100 m of colonies every two weeks. As predicted, colony weight and longevity decreased with the proportion of intensive crops and increased with the amount of flowering crops, but queen production and the occurrence of A. sociella were not affected by landscape composition. Contrary to our prediction, local availability of floral resources also did not affect colonies. The pollen richness brought back to colonies decreased with the proportion of flowering crops and varied according to the proportion of intensive crops interacting with local floral resources. Our work contributes to generalize the more restrictive conclusion of local‐scale, crop‐specific studies that intensive crop farming undermines bumble bee colony development and that it may thus play a role in the large‐scale population decline of these insects.

Objetivo: comprobar la divergencia genética de Liometopum apiculatum con base al fragmento Citocromo oxidasa subunidad I, usando tres poblaciones diferentes.Diseño/Metodología/Enfoque: Se tomaron muestras de poblaciones de L. apliculatum (escamol) en tres sitios de recolección: Otumba, Estado de México; Pinos, Zacatecas y Matehuala, San Luis Potosí y compararon extractos de ADN amplificados con patrones taxonómicos moleculares establecidos por NCBI GenBank.Resultados: Se construyó un modelo de distancia genética de dos parámetros con las tres poblaciones de L. apiculantum que confirma la conectividad de los sitios a pesar de la distancia basada en la subunidad citocromo oxidasa I. Estas similitudes mantienen la adaptación de L. apiculatum a condiciones climáticas adversas. Limitaciones en el estudio/implicaciones: la similitud, basada en la subunidad I del citocromo oxidasa, puede permitir el uso de hormigas reinas de diferentes sitios para mejorar la reproducción de otros nidos, pero requiere pruebas de aceptabilidad de las hormigas reinas introducidas.Hallazgos/Conclusiones: el ADN de L. apiculatum mostró muy poca divergencia genética en los nidos de hormigas analizados. Es posible una estrategia para mejorar las actividades de reproducción de esta hormiga basada en la reintroducción de hormigas reina en los nidos.

The rose is the queen of flowers and is widely used as a garden plant and for the cut flower market. Roses are also used for the production of essential oil for the cosmetic and perfume industries. A lot of botanical roses are scented and use their volatiles to attract pollinators. Fragrances in garden roses are very diverse and scent has always been an important character in the selection process. Breeders have recently tried to introduce new fragrances, for instance reminiscent of fruit or spice odors. But despite their efforts, some roses on the market are not very fragrant, especially the ones selected for the cut flower market. The cause of this lack of scent is not known. In modern roses or Hybrid Tea roses, scent is mainly produced by petals, although stamens can also contribute to the emission of volatiles. Scent compounds are concentrated in rose petal epidermis, probably in small lipid droplets. In spite of numerous chemical studies, the biosynthetic pathways of many rose scent compounds are unknown. We are studying several genes involved in the biosynthesis of scent in rose. For example, we recently characterized the enzymes responsible for the so-called “tea scent” emitted by Chinese roses. Apart from “tea scent”, terpenoids, especially monoterpenes, are also major constituents of rose flowers, mostly responsible for the “typical rose scent”. Generally, terpenoid biosynthesis in plants is achieved by various terpene synthases. However, with a combination of transcriptomic and genetic approaches, our research group recently discovered a terpene synthase-independent pathway. A key enzyme of this pathway is RhNUDX1, belonging to the Nudix protein family. It has geranyl diphosphate diphosphohydrolase activity in vitro. A positive correlation was found between the expression levels of RhNUDX1 gene and the production of the monoterpene geraniol, indicating its essential role in scent production in roses.

The purpose of this study was to assess the ecological knowledge surrounding the western queen butterfly, Danaus gilippus thersippus (H. Bates). Specifically, our objectives were to synthesize existing data and knowledge on the ecology of the queen and use results of this assessment to inform the direction of future research on this understudied species. We identified six core areas for assessment: distribution, the biodiversity of plant resources, western queen and their host plant phenology, chemical ecology, and four key life history traits. We mapped the distribution of D. g. thersippus from museum specimen records, citizen science (e.g., iNaturalist) and image sharing app-based observations, along with other observational data enumerating all current known plant resources and long-range movements. We assembled 14 larval food plants, six pyrrolizidine alkaloids plants and six nectar plants distributed in the western Mojave and Sonoran Desert regions of the United States and Baja California. We report on its phenology and its long-range movement. Butterfly species have declined across the western US, and western monarch populations have declined by 97%. Danaus g. thersippus has received little research attention compared with its famous congener D. plexippus L. Danaus g. thersippus&rsquo, desert distribution may be at its temperature limits for the species distribution and for its rare host plant Asclepias nyctaginifolia.
