Social network research often depends on the willingness of respondents to provide personal information about themselves and alters. Survey design strategies that increase willingness to share this information are necessary for social network research to be feasible, especially when name generators are used for sampling because rosters are unavailable. We conducted an experiment in which one group of respondents (n = 94) received an online survey that included an example network map and the other group (n = 100) received one that did not. Results show the map did not increase or decrease provision of network contacts nor influence the types of ties reported. Furthermore, respondents were reluctant to provide names and contact information of alters. Our study demonstrates the difficulty of collecting network information without a previously defined roster and an attempt to improve data collection through strategic survey design.

Mikania micrantha Kunth is native to tropical America and has invaded tropical and subtropical Asia and numerous Pacific Islands. It forms dense thickets and reduces native species diversity and populations in its introduced range. This invasive vine also seriously impacts many agricultural crops and is listed as one of the world’s 100 worst invasive alien species. Its life history characteristics, such as the production of large numbers of wind-dispersed seeds, vegetative reproduction, rapid growth, and genetic diversity all contribute to its invasiveness. In this review, we focus on how mechanisms to defend against its natural enemies boost the invasiveness of M. micrantha. It possesses potent defenses against natural enemies such as pathogenic fungi, herbivorous insects, and parasitic nematodes, and exhibits allelopathic potential against plant competitors. These defensive abilities, in concert with its formidable life history characteristics, contribute to the invasiveness of M. micrantha, potentially leading to further naturalization. Several other reviews have summarized the biology and management of the species, but ours is the first review to focus on how the defensive mechanisms of M. micrantha likely enhance its invasiveness. Relatively little is known about the array of defensive capabilities of M. micrantha; therefore, there is considerable scope for further research on its chemical defenses.

ABSTRACTIntroduction Every year in the United States, emergency departments (EDs) receive approximately 2.4 million non-traumatic dental condition (NTDC) visits. The American Association of State and Territorial Dental Directors (ASTDD) developed a surveillance guideline to understand which specific conditions dental infrastructure can address before patients succumb to the ED for care.Methods Descriptive epidemiology data of ED encounters from the California Department of Health Care Access and Information were analyzed. Primary diagnostic NTDCs in California EDs between 2016 and 2019 were reviewed using ASTDD surveillance protocol. Ophthalmic condition surveillance, with guidance from California Health and Human Services data, was used as a comparison due to similar national ED utilization rates.Results In California, 82 unique codes were used for primary NTDC diagnoses for the approximate 54,000 NTDC visits annually. The most common NTDC code was “Other Specified Disorders of Teeth and Supporting Structures,” accounting for 74–80% of all NTDC diagnoses. Ophthalmic condition codes had greater specificity with 12% as their most frequent code for “laceration without a foreign body of the left eyelid.”Conclusion Relatively few dental-related ED visits are associated with highly informative NTDC codes, limiting surveillance efforts to track the prevalence of underlying conditions. There is an opportunity to guide patients to proper dental care by 1) forming dental partners with EDs and 2) establishing referral protocol through case managers. If an ED had a standardized workflow to guide patients to dental care, a significant barrier could be addressed for both patients and providers.

Abstract Quantum simulations are becoming an essential tool for studying complex phenomena, e.g. quantum topology, quantum information transfer and relativistic wave equations, beyond the limitations of analytical computations and experimental observations. To date, the primary resources used in proof-of-principle experiments are collections of qubits, coherent states or multiple single-particle Fock states. Here we show a quantum simulation performed using genuine higher-order Fock states, with two or more indistinguishable particles occupying the same bosonic mode. This was implemented by interfering pairs of Fock states with up to five photons on an interferometer, and measuring the output states with photon-number-resolving detectors. Already this resource-efficient demonstration reveals topological matter, simulates non-linear systems and elucidates a perfect quantum transfer mechanism which can be used to transport Majorana fermions.

Major invasions of Indo-Pacific lionfish (Pterois volitans and P. miles) are underway in the Western Atlantic Ocean and the Mediterranean Sea. While the establishment of lionfish in the Western Atlantic is perhaps the most well-studied marine fish invasion to date, the rapidly expanding invasion in the Mediterranean is more recent and has received less attention. Here we review and synthesize successes and failures from two decades of lionfish management in the Western Atlantic to give policy recommendations for their management in the Mediterranean. Two failed approaches that were attempted multiple times in the Western Atlantic and that we advise against are (1) feeding lionfish to native fish to promote predation and (2) implementing bounty programs to incentivize lionfish harvest. Broadly, the most important management lessons that we recommend include (1) conducting routine removals by spearfishing with scuba, which can effectively suppress local abundances of lionfish; (2) encouraging the development of recreational and commercial lionfish fisheries, which can promote long-term, sustainable lionfish population control; and, (3) engaging local communities and resource users (e.g., with lionfish removal tournaments), which can concurrently achieve multiple objectives of promoting lionfish removals, market-development, research, and public education. Managers in the Western Atlantic often needed to adapt current conservation policies to enable lionfish removals in areas where spearfishing with scuba was otherwise prohibited for conservation purposes. The risk of abusing these policies was mitigated through the use of gear restrictions, diver trainings, and through participatory approaches that integrated scuba divers and stakeholder organizations in lionfish research and management. Our review of policies and practices in the Mediterranean Sea found that many of our recommended lionfish management approaches are not being done and indicate potential opportunities to implement these. We expect and fully recommend that work continues towards multinational cooperation to facilitate regional coordination of research, control, and management efforts with respect to the Mediterranean lionfish invasion. As with other major biological invasions, lionfish are unconstrained by political borders and their control will require rapid and strategic management approaches with broad cooperation among and between governments and stakeholders.

Background Immunotherapy with checkpoint inhibitors has shown impressive results in patients with melanoma, but still many do not benefit from this line of treatment. A lack of tumor-infiltrating T cells is a common reason for therapy failure but also a loss of intratumoral dendritic cells (DCs) has been described.Methods We used the transgenic tg(Grm1)EPv melanoma mouse strain that develops spontaneous, slow-growing tumors to perform immunological analysis during tumor progression. With flow cytometry, the frequencies of DCs and T cells at different tumor stages and the expression of the inhibitory molecules programmed cell death protein-1 (PD-1) and T-cell immunoglobulin and mucin-domain containing-3 (TIM-3) on T cells were analyzed. This was complemented with RNA-sequencing (RNA-seq) and real-time quantitative PCR (RT-qPCR) analysis to investigate the immune status of the tumors. To boost DC numbers and function, we administered Fms-related tyrosine 3 ligand (Flt3L) plus an adjuvant mix of polyI:C and anti-CD40. To enhance T cell function, we tested several checkpoint blockade antibodies. Immunological alterations were characterized in tumor and tumor-draining lymph nodes (LNs) by flow cytometry, CyTOF, microarray and RT-qPCR to understand how immune cells can control tumor growth. The specific role of migratory skin DCs was investigated by coculture of sorted DC subsets with melanoma-specific CD8+ T cells.Results Our study revealed that tumor progression is characterized by upregulation of checkpoint molecules and a gradual loss of the dermal conventional DC (cDC) 2 subset. Monotherapy with checkpoint blockade could not restore antitumor immunity, whereas boosting DC numbers and activation increased tumor immunogenicity. This was reflected by higher numbers of activated cDC1 and cDC2 as well as CD4+ and CD8+ T cells in treated tumors. At the same time, the DC boost approach reinforced migratory dermal DC subsets to prime gp100-specific CD8+ T cells in tumor-draining LNs that expressed PD-1/TIM-3 and produced interferon γ (IFNγ)/tumor necrosis factor α (TNFα). As a consequence, the combination of the DC boost with antibodies against PD-1 and TIM-3 released the brake from T cells, leading to improved function within the tumors and delayed tumor growth.Conclusions Our results set forth the importance of skin DC in cancer immunotherapy, and demonstrates that restoring DC function is key to enhancing tumor immunogenicity and subsequently responsiveness to checkpoint blockade therapy.

Background Human population growth has led to biodiversity declines in tropical cities. While habitat loss and fragmentation have been the main drivers of urban biodiversity loss, man-made interventions to reduce health risks have also emerged as an unintentional threat. For instance, insecticide fogging to control mosquito populations has become the most common method of preventing the expansion of mosquito-borne diseases such as Dengue. However, the effectiveness of fogging in killing mosquitoes has been called into question. One concern is the unintended effect of insecticide fogging on non-target invertebrates that are crucial for the maintenance of urban ecosystems. Here, we investigate the impacts of fogging on: (1) target invertebrate taxon (Diptera, including mosquitoes); (2) non-target invertebrate taxa; and (3) the foraging behavior of an invertebrate pollinator taxon (Lepidoptera) within an urban tropical forest. Methods We carried out fogging with Pyrethroid insecticide (Detral 2.5 EC) at 10 different sites in a forest situated in the state of Selangor, Peninsular Malaysia. Across the sites, we counted the numbers of knocked-down invertebrates and identified them based on morphology to different taxa. We constructed Bayesian hierarchical Poisson regression models to investigate the effects of fogging on: (1) a target invertebrate taxon (Diptera) 3-h post-fogging; (2) selected non-target invertebrate taxa 3-h post-fogging; and (3) an invertebrate pollinator taxon (Lepidoptera) 24-h post-fogging. Results A total of 1,874 invertebrates from 19 invertebrate orders were knocked down by the fogging treatment across the 10 sites. Furthermore, 72.7% of the invertebrates counted 3-h post-fogging was considered dead. Our regression models showed that given the data and prior information, the probability that fogging had a negative effect on invertebrate taxa 3-h post-fogging was 100%, with reductions to 11% of the pre-fogging count of live individuals for the target invertebrate taxon (Diptera), and between 5% and 58% of the pre-fogging count of live individuals for non-target invertebrate taxa. For the invertebrate pollinator, the probability that fogging had a negative effect 24-h post-fogging was also 100%, with reductions to 53% of the pre-fogging count of live individuals. Discussion Our Bayesian models unequivocally demonstrate that fogging has detrimental effects on one pollinator order and non-target invertebrate orders, especially taxa that have comparatively lower levels of chitinisation. While fogging is effective in killing the target order (Diptera), no mosquitos were found dead in our experiment. In order to maintain urban biodiversity, we recommend that health authorities and the private sector move away from persistent insecticide fogging and to explore alternative measures to control adult mosquito populations.

Managing invasive Indo-Pacific lionfish (Pterois volitans and P. miles) in the Western Atlantic Ocean is beyond the capacity of natural resource organizations alone. In response, organizations have mobilized members of the public and citizen scientists to help. We used a structured survey to assess the activities and perceptions of 71 organizations that engage the public and citizen scientists in lionfish research and management throughout the invaded range of the Western Atlantic. Five case studies were also conducted that exemplified varied and multi-pronged approaches to engagement of the public and citizen scientists in lionfish control, monitoring, and knowledge-sharing. The public has been engaged to some extent in every approach, but organizations most frequently indicated engaging members of the public in raising awareness, promoting consumption, organized culling/removal, tournaments, and data collection. Sixty-five percent of organizations surveyed engaged the public in data collection, and data collection was ranked as the scientific research activity in which the public is most often involved. Most organizations indicated their data has contributed to scientific publications, management, and government agency research and/or policy. Collectively these findings demonstrate the conservation value of citizen scientists to assist organizational efforts to control, manage, and study a large-scale marine invasion.

Abstract Proteoglycans are promising therapeutic targets in Multiple Sclerosis (MS), because they regulate many aspects of the immune response. This was studied using surfen, an agent that binds both heparan sulphate proteoglycans (HSPGs) and chondroitin sulphate proteoglycans (CSPGs). Initial cell culture work on bone marrow derived macrophages (BMDMs) found that surfen reduced concentrations of the chemokines CCL2, CCL4 and CCL5, with reduced messenger (m)RNA expression for Tumor Necrosis Factor, IL-6, IL-1β and inducible nitric oxide synthase. These data were further explored using Experimental Autoimmune Encephalomyelitis (EAE) in mice. Surfen reduced clinical signs during EAE when administered from disease onset, and reduced infiltration by CD4 positive T cells and macrophages into the central nervous system. These mice also showed reduced mRNA expression for the chemokines CCL3 and CCL5, with reduced concentrations of CCL2, CCL3 and CCL5. During EAE, surfen treatment induced a persistent increase in Interleukin (IL)-4 concentrations which may enhance T helper 2 responses. During EAE, surfen treatment reduced mRNA expression for HSPGs (NDST1, agrin, syndecan-4, perlecan, serglycin, syndecan-1) and the CSPG versican. By contrast, surfen increased mRNA expression for the CSPG aggrecan, with no effect on neurocan. During EAE, significant positive correlations were found between mRNA expression and clinical score for syndecan-4, serglycin and syndecan-1 and a significant negative correlation for aggrecan. These correlations were absent in surfen treated mice. Repair in the later stages of MS involves remyelination, which was modeled by injecting lysolecithin (lysophosphatidylcholine, LPC) into mouse corpus callosum to create regions of demyelination. When surfen was injected 2 days after LPC, it delayed remyelination of the lesions, but had no effect when injected 7 days after LPC. The delayed remyelination was associated with local increases in CSPG expression. Therefore surfen suppresses inflammation but inhibits remyelination in these models. A mechanism in common may be increased CSPG expression.

Habitat degradation and summer droughts severely restrict feeding options for the endangered southern hairy-nosed wombat (SHNW; Lasiorhinus latifrons). We reconstructed SHNW summer diets by DNA metabarcoding from feces. We initially validated rbcL and ndhJ diet reconstructions using autopsied and captive animals. Subsequent diet reconstructions of wild wombats broadly reflected vegetative ground cover, implying local rather than long-range foraging. Diets were all dominated by alien invasives. Chemical analysis of alien food revealed Carrichtera annua contains high levels of glucosinolates. Clinical examination (7 animals) and autopsy (12 animals) revealed that the most degraded site also contained most individuals showing signs of glucosinolate poisoning. We infer that dietary poisoning through the ingestion of alien invasives may have contributed to the recent population crashes in the region. In floristically diverse sites, individuals appear to be able to manage glucosinolate intake by avoidance or episodic feeding but this strategy is less tractable in the most degraded sites. We conclude that recovery of the most affected populations may require effective Carrichtera management and interim supplementary feeding. More generally, we argue that protection against population decline by poisoning in territorial herbivores requires knowledge of their diet and of those food plants containing toxic principles.

The ability of weeds to evolve is key to their success, and the relationship between weeds and humans is marked by co-evolution going back to the agricultural revolution, with weeds evolving to counter human management actions. In recent years, climate change has emerged as yet another selection pressure imposed on weeds by humans, and weeds are likewise very capable of adapting to this latest stress of human origin. This review summarizes 10 ways this adaptation occurs: (1) general-purpose genotypes, (2) life history strategies, (3) ability to evolve rapidly, (4) epigenetic capacity, (5) hybridization, (6) herbicide resistance, (7) herbicide tolerance, (8) cropping systems vulnerability, (9) co-evolution of weeds with human management, and (10) the ability of weeds to ride the climate storm humans have generated. As pioneer species ecologically, these 10 ways enable weeds to adapt to the numerous impacts of climate change, including warming temperatures, elevated CO2, frequent droughts and extreme weather events. We conclude that although these 10 ways present formidable challenges for weed management, the novelty arising from weed evolution could be used creatively to prospect for genetic material to be used in crop improvement, and to develop a more holistic means of managing agroecosystems.

Oncolytic viruses selectively target and kill cancer cells in an immunogenic fashion, thus supporting the establishment of therapeutically relevant tumor-specific immune responses. In 2015, the US Food and Drug Administration (FDA) approved the oncolytic herpes simplex virus T-VEC for use in advanced melanoma patients. Since then, a plethora of trials has been initiated to assess the safety and efficacy of multiple oncolytic viruses in patients affected with various malignancies. Here, we summarize recent preclinical and clinical progress in the field of oncolytic virotherapy.

Dendritic cells (DCs) are specialized antigen-presenting cells that have a notable role in the initiation and regulation of innate and adaptive immune responses. In the context of cancer, appropriately activated DCs can induce anti-tumor immunity by activating innate immune cells and tumor-specific lymphocytes that target cancer cells. However, the tumor microenvironment (TME) imposes different mechanisms that facilitate the impairment of DC functions, such as inefficient antigen presentation or polarization into immunosuppressive DCs. These tumor-associated DCs thus fail to initiate tumor-specific immunity, and indirectly support tumor progression. Hence, there is increasing interest in identifying interventions that can overturn DC impairment within the TME. Many reports thus far have studied oncolytic viruses (OVs), viruses that preferentially target and kill cancer cells, for their capacity to enhance DC-mediated anti-tumor effects. Herein, we describe the general characteristics of DCs, focusing on their role in innate and adaptive immunity in the context of the TME. We also examine how DC-OV interaction affects DC recruitment, OV delivery, and anti-tumor immunity activation. Understanding these roles of DCs in the TME and OV infection is critical in devising strategies to further harness the anti-tumor effects of both DCs and OVs, ultimately enhancing the efficacy of OV-based oncotherapy.

Light trapping within a light absorbing medium is a key to highly efficient thin film solar cells. We propose a large-scale procedure based on materials with low absorption for the fabrication of combined Distributed Bragg Reflector (DBR) and grating light trapping structures. Using Rigorous Coupled Wave Analysis (RCWA) numerical simulations we designed a combined DBR and 1D grating structure allowing to significantly improve the absorption in a aSi:H film as thin as 100 nm. The optimized light trapping structure was fabricated. The enhancement of light absorption in thin aSi:H film was experimentally proven and discussed quantitatively with respect to the theoretical expectations.
