Introduction: Acquired bladder diverticula (BD) are associated with bladder outlet obstruction. The aim of our study is to analyse the improvement in lower urinary tract symptoms (LUTS) in patients who underwent robot-assisted bladder diverticulectomy (RABD) combined with transurethral prostatectomy (TURP)., Material and Methods: A prospectively single-centre, single surgeon cohort of four patients with posterolateral BD due to bladder outlet obstruction (BOO) undergoing RABD combined with TURP between 2018 and 2023 was analysed., Results: Median age and maximum BD diameter were 73.5 years and 16 cm, respectively. All patients had severe LUTS and elevated postvoid residual (PVR). Preliminary uroflowmetry revealed bladder outlet obstruction with a median of maximum urine flow rate of 8.5 ml/s. The median operative time and blood loss were 212 min and 100 ml, respectively. No intraoperative complications were recorded. The median length of stay was 4 days. The International Prostate Symptom Score (IPSS) and PVR were compared between baseline, 1 month and 6 months after surgery. IPSS significantly decreased from 24 (IQR 24-25) preoperatively compared to the postoperative, at 1 month follow up 7 (IQR 6-8) ( p  < 0.0001). PVR significantly decreased too from 165 (IQR 150-187) to 35 ml (IQR 25-42) ( p  < 0.0001). In transitioning from the 1-month follow-up to the 6-month follow-up, no substantial statistical improvement was observed., Conclusion: Concomitant performance of TURP with RABD is feasible and safe. Background and Objective: Robotic surgery has contributed greatly to the shift from traditional surgery to minimally invasive surgery. Urology is the major field of application of robotic surgery. Several urological procedures, especially radical prostatectomy, benefit from the use of robotic surgery., Methods: Non-systematic research of the literature was performed using "Robot-assisted radical prostatectomy" and "Robotic platforms" as keywords to understand the actual situation and the future perspectives of this technology in prostate cancer treatment., Key Content and Findings: The robotic platform landscape is constantly evolving. DaVinci has always been the mainstay in this field, particularly after the advent of the new single port platforms. New platforms are emerging, providing an alternative option to the well-known DaVinci system. Since in literature, few studies compare the use of different robotic platforms, their application in urological procedures is not yet widely used, for both oncological and non-oncological procedures. Furthermore, artificial intelligence begins to play a role in this landscape and could be useful for future developments. So further studies are warranted to give a full comprehension of the whole scenario., Conclusions: This review aims to analyze the current state of the use of robotic platforms in urology, particularly in radical prostatectomy, and to understand the evolution.

Renal cell carcinoma (RCC) comprises various histologically distinct subtypes, each characterized by specific genetic alterations, necessitating individualized management and treatment strategies for each subtype. An exhaustive search of the PubMed database was conducted without any filters or restrictions. Inclusion criteria encompassed original English articles focusing on molecular mechanisms of kidney cancer. On the other hand, all non-original articles and articles published in any language other than English were excluded. Hereditary kidney cancer represents 5-8% of all kidney cancer cases and is associated with syndromes such as von Hippel-Lindau syndrome, Birt-Hogg-Dubè syndrome, succinate dehydrogenase-deficient renal cell cancer syndrome, tuberous sclerosis complex, hereditary papillary renal cell carcinoma, fumarate hydratase deficiency syndrome, BAP1 tumor predisposition syndrome, and other uncommon hereditary cancer syndromes. These conditions are characterized by distinct genetic mutations and related extra-renal symptoms. The majority of renal cell carcinoma predispositions stem from loss-of-function mutations in tumor suppressor genes. These mutations promote malignant advancement through the somatic inactivation of the remaining allele. This review aims to elucidate the main molecular mechanisms underlying the pathophysiology of major syndromes associated with renal cell carcinoma. By providing a comprehensive overview, it aims to facilitate early diagnosis and to highlight the principal therapeutic options available.

Bladder paraganglioma is a rare neuroendocrine neoplasm, either functional or non-functional, arising from the urinary bladder. Functional variants present with catecholamine-related symptoms, while non-functional variants pose diagnostic challenges, mimicking urothelial carcinoma. Misdiagnosis risks underscore the importance of accurate identification for appropriate patient management. In this case, a 52-year-old man, diagnosed incidentally with hypertension and reported occasional post-micturition tachycardia, underwent abdominal ultrasound for known hepatic cyst follow-up, revealing an oval hypoechoic bladder mass. Initial consideration of bladder urothelial carcinoma prompted further investigation with contrast-enhanced CT scan and cystoscopy that confirmed extrinsic mass nature, and subsequent robotic-assisted partial cystectomy was performed. Histologically, the removed mass exhibited characteristic features of bladder paraganglioma. Postoperative recovery was uneventful, with resolution of post-micturition tachycardia at 1 month. Follow-up includes endocrinological evaluation and a 6-month CT scan. In conclusion, bladder paraganglioma should be considered in para-vesical mass differentials. This case highlights the importance of meticulous history collection, even in asymptomatic patients, the need for a multidisciplinary approach for accurate diagnosis and management of this rare condition, and the robotic approach as a viable option., (© The Author(s), 2024.)

Kidney stone disease (KSD) is one of the most common urological diseases. The incidence of kidney stones has increased dramatically in the last few decades. Kidney stones are mineral deposits in the calyces or the pelvis, free or attached to the renal papillae. They contain crystals and organic components, and they are made when urine is supersaturated with minerals. Calcium-containing stones are the most common, with calcium oxalate as the main component of most stones. However, many of these form on a calcium phosphate matrix called Randall's plaque, which is found on the surface of the kidney papilla. The etiology is multifactorial, and the recurrence rate is as high as 50% within 5 years after the first stone onset. There is a great need for recurrence prevention that requires a better understanding of the mechanisms involved in stone formation to facilitate the development of more effective drugs. This review aims to understand the pathophysiology and the main molecular mechanisms known to date to prevent recurrences, which requires behavioral and nutritional interventions, as well as pharmacological treatments that are specific to the type of stone.

A frequency-modulated continuous-wave radar for short-range target imaging, assembling a transceiver, a PLL, an SP4T switch, and a serial patch antenna array, was realized. A new algorithm based on a double Fourier transform (2D-FT) was developed and compared with the delay and sum (DAS) and multiple signal classification (MUSIC) algorithms proposed in the literature for target detection. The three reconstruction algorithms were applied to simulated canonical cases evidencing radar resolutions close to the theoretical ones. The proposed 2D-FT algorithm exhibits an angle of view greater than 25° and is five times faster than DAS and 20 times faster than the MUSIC one. The realized radar shows a range resolution of 55 cm and an angular resolution of 14° and is able to correctly identify the positions of single and multiple targets in realistic scenarios, with errors lower than 20 cm.

Microwave imaging is an active area of research that has garnered interest over the past few years. The main desired improvements to microwave imaging are related to the performances of radiating systems and identification algorithms. To achieve these improvements, antennas suitable to guarantee demanding requirements are needed. In particular, they must operate in close proximity to the objects under examination, ensure an adequate bandwidth, as well as reduced dimensions and low production costs. In addition, in near-field microwave imaging systems, the antenna should provide an ultra-wideband (UWB) response. Given the relevance of the foreseen applications, many UWB antenna designs for microwave imaging applications have been proposed in the literature. In this paper, a comprehensive review of different UWB antenna designs for near-field microwave imaging is presented. The antennas are classified according to the manufacturing technology and radiative performances. Particular attention is also paid to the radiation mechanisms as well as the techniques used to reduce the size and improve the bandwidth.

We describe a nonsurgical technique for managing gastric distention in infants with type C esophageal atresia, involving intubating the trachea with an umbilical catheter and entering the stomach through the fistula as soon as a flexible bronchoscope found its wide-open orifice. Organophosphorous (OP) compounds are pesticides frequently released into the environment because of extensive use in agriculture. Among these, methyl parathion (mPT) recently received attention as a consequence of illegal use. The predominant route of human exposure to mPT is via inhalation, but inadvertent consumption of contaminated foods and water may also occur. The goal of this study was to investigate the in vitro effects of mPT on cells in the oral cavity and evaluate the potential protective role of epigallocathechin-3-gallate (EGCG) on these effects. Human gingival fibroblasts (HGF) were exposed to 10, 50, or 100 μ g/ml mPT for 24 h and assessed for oxidative stress, as evidenced by reactive generation of oxygen species (ROS), induction of apoptotic cell death, DNA damage (comet assay and cytochinesis-block micronucleus test), and nitric oxide (NO) production. The results showed that mPT produced significant oxidative stress, cytotoxicity, and genotoxicity and increased NO levels through stimulation of inducible NO synthase expression. Finally, data demonstrated that EGCG (10, 25, or 50 μ M) was able to inhibit the pesticide-induced effects on all parameters studied. Data indicate that cytotoxic and genotoxic effects may be associated with oxidative stress induced by mPT observed in HGF cultures and that EGCG plays a protective role via antioxidant activities.

The objective was to review and compare outcomes after tongue-lip adhesion (TLA) and mandibular distraction osteogenesis (MDO) in infants with severe breathing difficulties related to Pierre Robin sequence (PRS). A single-centre retrospective (2002-2012) study was carried out; 18 infants with severe breathing difficulties related to PRS resistant to conservative treatment, who underwent TLA or MDO to correct airway obstruction, were enrolled. The primary outcome measures were successful weaning from respiratory support and resumption of full oral feeding. Nine underwent TLA and nine MDO. Eight of the nine infants who underwent MDO and all those treated with TLA were successfully weaned from respiratory support. After discharge, residual respiratory distress was diagnosed more commonly after TLA than after MDO (6/9 vs 1/9, P=0.050). Infants resumed oral feeding sooner after MDO than after TLA (mean days after surgery to full oral feeds 44±24 vs 217±134, P<0.003). The length of hospital stay was longer for infants treated with MDO than for those treated with TLA. The rate of complications was similar. Infants with severe airway obstruction related to PRS can benefit safely from either TLA or MDO. Although MDO lengthens the time to discharge, this option stabilizes airway patency of infants with PRS more efficiently and achieves full oral feeding more rapidly than TLA., (Copyright © 2013 International Association of Oral and Maxillofacial Surgeons. Neonates with Pierre Robin Sequence (PRS) usually present with varying degrees of upper airway obstruction and difficulty feeding. Early treatment is important for such children in order to prevent impaired cognitive development resulting from hypoxemic episodes. Various procedures aimed at widening the pharyngeal space have been proposed, including prone position, tongue-lip adhesion, mandibular traction, non-invasive ventilation and palatal plates. Mandibular distraction osteogenesis (MDO) using external or internal devices has become increasingly popular as an alternative treatment option when other medical or surgical techniques do not prove to be satisfactory. This review summarizes current evidence on the effectiveness of MDO in infants with PRS. Because of a lack of studies comparing this treatment with other procedures, general recommendations cannot be drawn and treatment of infants with PRS still requires individualization.

Objective: To study the frequency of non-alcoholic fatty liver disease (NAFLD), its relationship to clinical and biochemical variables, and the effect 12-month's lifestyle intervention in obese children and adolescents., Methods: Thirty-six obese patients aged 7 to 18 years, 42% female and 58% male, 72.2% prepubertal and 27.8% pubertal, were selected. Anthropometric measurements and glucose, insulin (baseline and after a glucose load), lipid profile, C-reactive protein, and aminotransferase tests were performed before and 12 months after dietary and physical activity intervention. Liver ultrasound was performed to determine the presence of NAFLD., Results: NAFLD was found in 66.7% (n=24), and was mild in 30.6%, moderate in 27.8%, and severe in 8.3%. Subjects with NAFLD had higher body mass index (BMI, p=0.007), waist (p=0.005), fat area (p=0.002), basal insulin (p=0.01), and HOMA-IR (p=0.008) values and lower QUICKI (p=0.02) values than those with no NAFLD. After intervention, physical activity increased (p=0.0001) and calorie intake remained unchanged. NAFLD disappeared in 9 patients (37.5%, p=0.02) and disease severity decreased in 3 patients (12.5%). In addition, BMI Z-score (p=0.005), fat area (p=0.0001), basal insulin (p<0.05), insulin resistance (p<0.005), lipid profile (p<0.03), and transaminases decreased. Weight loss was the main variable accounting for NAFLD improvement., Conclusion: This group of obese children and adolescents showed a high frequency of NAFLD. Single-wall carbon nanotubes (SWCNTs) are one of the most extensively produced carbon materials and the environmental, public and professional exposure is therefore dramatically increasing. Consequently the studies on bio-effects and safety of SWCNTs are highly needed. The goal of this study was investigate the effects in vitro of SWCNTs in cells of the oral cavity, never employed in this research field. We exposed human gingival fibroblasts to 50, 75, 100, 125, 150 μg/ml SWCNTs for 24 h and we investigated genotoxicity (Comet assay and micronucleus test), cytotoxicity, oxidative stress, as reactive oxygen species (ROS) generation, and stress response, as Heat shock protein 70 (Hsp70) expression. SWCNTs produced genotoxic effects at all doses, even if detected with different sensitiveness by the two tests, and at the two highest doses induced a strong decrease of the cell proliferation and cell survival, causing apoptosis too. Furthermore, we proved the ability of these nanomaterials to induce oxidative stress and Hsp70 expression. Finally, by inhibition of Hsp70 expression, we demonstrated that this heat shock protein conferred protection against SWCNT geno-cytotoxicity., (Copyright © 2011 Elsevier Ltd. All rights reserved.)

Shock is a clinical disorder that challenges caregivers in the neonatal intensive care unit. The predominant cause of shock in neonates is sepsis. This article provides an overview of the current treatment of septic shock with particular emphasis on newer vasoactive drugs (milrinone, levosimendan and vasopressin) to support cardiovascular dysfunction.

Activated single wall carbon nanotubes have been used for biomedical purposes as carriers for drugs, peptides, proteins and nucleic acids. A large volume of data speaks to their suitability to act as a carrier. The ability of two differently activated SWNTs (with carboxyl groups or with carboxyl groups and polyethylenimine (PEI)) to form a complex with the hepatitis A virus was evaluated. Both types of activations permitted the formation of a virus-SWNT complex. However, their patterns were different. The carboxyl-activated nanotubes had a somewhat low adsorptive capacity that was related inversely to the concentrations of the SWNTs and viruses. Statistical analysis, using the chi(2)-test, showed no significant differences between the SWNT-PEI ratios of 1:2.5, 1:1 and 1:0.5. The addiction of PEI improved the adsorption, probably because of the electropositive charge of the molecule. Adsorption was optimal between 100 microg and 10 ng with a SWNTs-PEI weight ratio of 1:0.2 up to an inoculum of 10(5) genome equivalents of hepatitis A virus. Reducing or increasing this weight ratio reduced the adsorptive capacity of the PEI, and this adsorption activity was time and contact-dependent. Thus, SWNTs coated with PEI are able to complex with viruses, and they might be used in the future to transfect non-permissive cell lines., (Copyright 2010 Elsevier B.V. All rights reserved.)

Mechanical ventilation is considered a supportive, nontherapeutic technology used to perform the work of breathing for patients who are unable to do so on their own. In neonatology, mechanical ventilation is often used for premature neonates who are unable to sustain ventilation because of reduced functional residual capacity due to surfactant deficiency. Mechanical ventilation is thus an attempt to mimic the respiratory system's physiological function of gas exchange until the respiratory system reaches maturation. In pediatrics, mechanical ventilation is rarely used for acute respiratory distress syndrome as shown by Dahlem et al. in 2003 who found that only 9.9% of cases of respiratory failure in PICU was caused by ARDS. For this reason, ventilatory techniques in PICU are very heterogenous from the assisted to the most aggressive controlled modes associated with ventilator maneuvers. There are no specific guidelines for the use of mechanical ventilation in children and the low number of infants with ARDS in PICU makes it difficult to run randomized controlled trials in this population. Thus the algorithms are based on the results of either adult or neonatal studies. The advantage of extrapolating data from the neonatal evidence relates mainly to the prevention of ventilator induced lung injury (e.g., CPAP, HFOV, NIV, permissive hypercapnia, surfattant), of which neonatologists are particularly expert.

Objective: Transcranial Doppler (TCD) is a sensitive technique for circulatory arrest diagnosis in brain death when patterns such as reverberant flow and short systolic spikes are observed. In infants, the nonossified fontanelles compensate for intracranial hypertension. We describe TCD patterns in infants with brain death, different from adults, with the hemodynamic modifications induced by anterior fontanelle compression., Method: TCD was performed in 2 infants with diagnosed brain death admitted to the neonatal intensive care unit., Results: TCD showed a large peak 'reverberant' flow, with a high peak systolic velocity and a consistent retrograde component, away from the brain. Compression of the anterior fontanelle induced, at first, a reduction in systolic flow with the subsequent appearance of the characteristic short systolic spikes. Upon compression removal, a brief increase in the systolic flow was observed before the prompt reappearance of the reverberant flow., Conclusion: TCD for brain death diagnosis should be done cautiously in infants. In these cases, reverberating flow may be indicative of circulatory arrest even if with a large peak and with a high peak systolic velocity. Heavy fontanelle compression may reproduce the classical adult TCD patterns of brain death, thus supporting the diagnosis of cerebral circulatory arrest., (Copyright 2010 S. Karger AG, Basel.)

Nanotechnologies hold considerable promise of advances in many sectors especially the biomedical field, since the materials used are of the appropriate dimensions to interact with important biological matter such as proteins, DNA and viruses. In this field the use of nanotechnologies will probably be second in importance only to biotechnologies. However many characteristics of nanomaterials that make them so promising from a technological point of view may also lead to negative effects on the environment and human health. It is important therefore that the environmental and work-related exposure effects to these materials be evaluated. In this article the potential uses, toxic effects and public health implications of nanobiotechnologies are discussed.

Resistance to apoptosis is essential for cancer survival and plays a critical role in carcinogenesis. Growing evidence suggests that nicotine can act as a tumor promoter, impairing apoptotic process in certain types of human cancer cell lines. Our previous study revealed in human gingival fibroblasts (HGFs) a concomitant antiapoptotic and genotoxic effect of nicotine, manifested by the attenuation of staurosporine (STP)-induced apoptosis and the increase of micronucleus frequency. The present report provides evidence that nitric oxide (NO) is critically involved in these actions. In vitro treatment with sodium nitroprusside as NO donor showed that NO produced similar effects as those observed with nicotine: it caused DNA damage and partially prevented apoptosis induced by staurosporine. Exposure of HGFs to nicotine, at concentrations similar to those found in the blood of habitual smokers, leads to the production of NO associated with the induction of inducible nitric oxide synthase (iNOS) expression. Experiments using an inhibitor of iNOS, N-monomethyl-L-arginine (NMA), together with nicotine confirmed the involvement of NO in the drug action, abrogating completely cell death and a good part of the genotoxicity. Finally, we show by different approaches that the inhibition of cell death by nicotine through NO release is related to modulation of caspase-1 activation.

The mismatch repair (MMR) system and p53 protein play a pivotal role in maintaining genomic stability and modulate cell chemosensitivity. Aim of this study was to examine the effects of either MMR-deficiency or p53 inactivation, or both, on cellular responses to bleomycin. The MMR-deficient colon carcinoma cell line HCT116 and its MMR-proficient subline HCT116/3-6, both expressing wild-type p53, were transfected with an expression vector encoding a dominant-negative p53 mutant, or with the empty vector. Four transfected clones, having the following phenotypes, MMR-proficient/p53 wild-type, MMR-proficient/p53 mutant, MMR-deficient/p53 wild-type, MMR-deficient/p53 mutant, were subjected to treatment with bleomycin. Loss of MMR function alone was associated with increased resistance to apoptosis, chromosomal damage and inhibition of colony formation caused by bleomycin. Loss of p53 alone resulted in abrogation of G1 arrest and increased sensitivity to apoptosis and chromosomal damage induced by the drug, but did not affect clonogenic survival after bleomycin treatment. Disabling both p53 and MMR function led to abrogation of G1 arrest and to a moderate impairment of drug-induced apoptosis. Chromosomal damage was reduced in the MMR-deficient/p53 mutant clone with respect to the MMR-proficient/p53 wild-type one, when evaluated 48 h after bleomycin treatment, but was comparable in both clones 96 h after drug exposure. Clonogenic survival of the MMR-deficient/p53 mutant clone was similar to that of the MMR-deficient/p53 wild-type one. The effects of MMR-deficiency on cellular responses to bleomycin were confirmed using the MMR-proficient lymphoblastoid cell line TK6 and its MMR-deficient subline MT1, both expressing wild-type p53. In conclusion, our data show that loss of MMR and p53 function exerts opposite and independent effects on apoptosis and chromosomal damage induced by bleomycin. Moreover, inactivation of MMR confers resistance to the cytotoxic activity of the anticancer agent in cells expressing either wild-type or mutant p53.

Growing evidence suggests that nicotine, the addictive component of cigarettes, can have a direct role in tumor development by enhancing cell proliferation and impairing apoptotic process in certain types of human cancer cell lines. Since the correlation between apoptosis and DNA damage is already well documented, we investigated the response of human gingival fibroblasts (HGFs) to nicotine exposure by examining its effect on DNA damage induction and apoptotic process in parallel. To assess the genotoxicity of this drug, the cytokinesis-block micronucleus (CBMN) test was performed. Treatment of HGFs with nicotine, at a concentration of 1 microM, caused a statistically significant increase of micronucleus (MN) frequency at the tested time intervals, while no change was detected in cell growth under the same conditions. Furthermore, we found that preincubation of HGFs with 1 microM nicotine strongly attenuated staurosporine (STP)-induced apoptosis. Finally, we found that cultures exposed to nicotine showed an increase of reactive oxygen species, as determined by increased levels of 2,7-dichlorofluorescein (DCF). When cells were prelabeled with N-acetyl-cysteine (NAC), a substrate for glutathione synthesis, and catalase (CAT), the oxygen free radical scavenger, a significant reduction in cytogenetic damage was observed. Thus, for the first time, we report a concomitant genotoxic and antiapoptotic effect of nicotine in HGFs.

The activation of telomerase in phytohemagglutinin-stimulated peripheral lymphocytes is thought to play a role in telomere maintenance and DNA repair. Considering the importance of this enzyme in both cancer and senescence, we studied the effects of the telomerase inhibitor 3'-azido-3'-deoxythymidine on the frequency of chromosomal aberrations and micronuclei induced in peripheral blood lymphocytes (PBL) of elderly monozygotic and dizygotic twins, evaluated with respect to the genotoxic effects induced in unrelated young subjects. Our results show that the cytogenetic damage induced by 3'-azido-3'-deoxythymidine in human PBL was mainly regulated by genetic factors and allowed the identification of hypersensitive subjects. Ageing, which did not modify the individual susceptibility to 3'-azido-3'-deoxythymidine induction of chromosome aberrations and micronuclei, nevertheless determined an overall increase in nuclear damage.

The induction of chromosome damage in cultured human lymphocytes by in vitro treatments with aphidicolin (APC) and bleomycin (BLM) has been proposed as test of sensitivity to mutagens. To assess their validity, we have investigated whether the individual expression of induced chromosome damage has a genetic rather than an environmental basis. Metaphase analysis for chromosomal aberrations (CA) and micronucleus (MN) assay in cytokinesis-blocked cells have been performed in peripheral blood lymphocytes from 19 healthy male twins (9 monozygotic and 10 dizygotic pairs), aged 70-78 years, after APC, BLM and APC+BLM treatments. Concordance between twins revealed a high genetic component in the sensitivity towards clastogenic action of APC both as percentages of CA and MN. The micronucleus assay demonstrated a genetic basis also in the expression of chromosome damage induced by BLM and APC+BLM treatments. Since twins were elderly people, to investigate the possible role of age, CA and MN frequencies were compared with those found in lymphocytes from 11 young male donors. Basal and APC-induced chromosome damage were clearly increased in the former. Following BLM and APC+BLM treatments, age significantly increased mitotic delay, as shown by the mitotic indexes (MI) and by the ratios between binucleated and mononucleated (B/M) cells.

The involvement of the antioxidant enzymes catalase and glutathione peroxidase (both at 0.1 mg/ml) in defence against the genotoxicity of phosphamidon (80 microg/ml) and dieldrin (25 microM) was investigated in order to demonstrate that the two pesticides damage DNA through the generation of reactive oxygen species and therefore of oxidative stress. The pesticide genotoxicity was determined by the cytokinesis-block micronucleus test performed on primary mouse lung fibroblast cultures. Also, 3-aminotriazole (40 mM) and mercaptosuccinate (0.5 mM), inhibitors of catalase and glutathione peroxidase, respectively, were added to the cultures. Data indicate that catalase causes a decrease only in the damage induced by phosphamidon, while glutathione peroxidase protects against damage induced by both phosphamidon and dieldrin. Simultaneous treatment with antioxidant inhibitors and pesticides results in a decrease in micronucleus frequency and cell number, due to apoptotic death. Our results indicate that clastogenic DNA damage produced by the two pesticides is modulated by antioxidant enzymes and their inhibitors and thus could be due to oxidative stress induction.

Two pesticides, organophosphate phosphamidon (PHO) and organochlorine dieldrin (DED) were assayed by the mouse bone marrow micronucleus test, to ascertain whether they showed genotoxic activity in vivo. Two doses, sub-lethal (PHO=3 mg/kg b.wt.; DED=60 mg/kg b.wt.) and lethal (PHO=5 mg/kg b.wt.; DED=90 mg/kg b.wt.), of each substance were administered intraperitoneally to 9-10-week old CBA male mice, in acute and repeated exposure. The sub-lethal dose was also administered at two different times and twice at 24-h intervals. Both PHO and DED proved able to induce a dose-dependent increase of micronucleated polychromatic erythrocytes (PCE). The two pesticides also showed a different detoxification time. Furthermore, the CREST staining with antikinetochore antibodies allowed us to conclude that the two chemicals are clastogens., (Copyright 1999 Elsevier Science B.V.)

The equilibrium O2-binding properties of the hybrid haemoglobin (Hb) present in vivo in erythrocytes from mule and of its parent Hbs from horse and donkey were compared with special reference to the effect of heterotropic ligands such as Cl-, D-glycerate 2,3-bisphosphate (DPG) and inositol hexakisphosphate. All these Hbs display a decreased effect by polyphosphates, confirming that what has been observed for horse Hb [Giardina, Brix, Clementi, Scatena, Nicoletti, Cicchetti, Argentin & Condò (1990) Biochem. J. 266, 897-900] is common to other equine species, at least from a qualitative standpoint. However, different quantitative aspects can be detected, which can be accounted for by a different role for the two types of chain in characterizing the binding free energy for the various heterotropic effectors. In particular, it is shown that the binding mode of DPG and inositol hexakisphosphate displays different features since long-range effects can be observed clearly for inositol hexakisphosphate but not for DPG. In general terms, in spite of a different intrinsic O2 affinity, the modulation of functional properties by third ligands leads these Hbs to behave, under physiological conditions, similarly to human HbA. It might represent an interesting example of how different species with similar functional needs find different ways to produce a similar functional behaviour.

Pgm allele frequencies of 383 individuals were determined in a sample of Drosophila melanogaster from three laboratory Sardinian populations, using the techniques of standard electrophoresis, heat denaturation, and isoelectric focusing. The analysis of the progeny obtained from informative crosses showed that the isoelectric focusing patterns segregate in a Mendelian way. The Pgm1.00 and Pgm0.70 electrophoretic alleles displayed different isoelectric points, whereas the Pgm1.00,tr and Pgm1.00,ts isoelectrophoretic alleles could not be differentiated when tested by isoelectric focusing. Moreover, the Pgm0.70,ts allele was split into two classes, with isoelectric points of pH 6.4 and pH 6.6.

Despite the fact that the horse is one of the more common domesticated animals, there are few reports dealing with the properties of its blood, and no comprehensive study has been performed on the reactivity of horse haemoglobin towards organic and inorganic ions. Here we report data on the effects of the organic phosphates D-glycerate-2,3-bisphosphate (2,3-DPG) and InsP6, and of chloride on the properties of horse haemoglobin. Thus the effect of saturating concentrations of 2,3-DPG on the oxygen affinity of horse haemoglobin is about 60% lower than with human adult haemoglobin under the same experimental conditions. The same applies also to InsP6, whose effect on oxygen binding to horse haemoglobin is decreased by about 55% compared with human adult haemoglobin. On the whole, horse haemoglobin appears to be much less sensitive to organic phosphates than previously believed. These results are discussed in the light of the primary structure of the molecule.

The Segregation Distortion (SD) phenomenon is a typical case of non-Mendelian segregation in Drosophila melanogaster, due to the dysfunction of sperm bearing a non-SD homologous chromosome. In nature, several factors involved in the expression of the SD phenomenon have been described; among these, a genetic modifier carried by chromosome 3, which enhances the distortion effect of the SD chromosomes. The analysis of natural Sardinian populations, carried out in order to evaluate the presence of chromosome 3 bearing these enhancer factors, has enabled us to ascertain that (a) also in these populations chromosomes 3 with enhancer factors are present, although with frequencies lower than those previously reported in other publications; (b) among these enhancer chromosomes 3, some increase the k of certain chromosomes 2 from values of chromosomes considered non-distorting (k less than or equal to 0.66) to value typical of SD chromosomes. The data obtained also allow us to put forward some considerations regarding the dynamics of the SD phenomenon in Sardinian populations, where the frequency of SD chromosomes is fairly elevated.

A simple procedure is described to detect genetic heterogeneity within electrophoretic classes at a locus in Drosophila, based on electrophoresis and heat denaturation studies. Temperature-resistant (tr) and temperature-sensitive (ts) isoelectrophoretic alleles at the phosphoglucomutase locus (Pgm) are present at polymorphic frequencies in natural and in laboratory populations of Drosophila melanogaster.

The discovery and the cytogenetic characterization of a new SD (Segregation Distorter) chromosome 2 from a natural population in Ranna (Sicily, Italy), SD(Ra), are reported. The main features of this chromosome are as follows: (a) it contains an Sd(Ra) gene with a moderate degree of segregation distortion (k = 0.72), (b) a recessive female sterile gene, fs(2)(TLM), responsible for modifications of the morphology and structure of the tests and ovaries is located at 89.7, (c) SD(Ra)/SD(Ra) males and females are viable but sterile, the females due to homozygosis of fs(2)(TLM) and the males because of homozygosis of a region containing the Sd locus, and (d) SDi/SDj combinations are fertile, thus suggesting that the different Sd factors found in natural populations constitute a multiple allelic series.-These data may indicate that each population containing SD chromosomes has evolved its own genetic architecture for the complex SD system, with specific modifiers and perhaps different Sd genes. The possibility of reconstructing the evolutionary pattern of the SD(Ra) chromosome in the natural Ranna population after the model of Charlesworth and Hartl (1978) and Crow (1979) is considered.

The second chromosome Co-122 (Corato-122) extracted from a natural population of Drosophila melanogaster caught in Corato (Apulia) and maintained in the laboratory over the SM5 balancer chromosome, proved to carry: (1) a Segregation distorter factor, named SdCo; (2) a recessive lethal mutation, termed mle-Co (maleless-Corato), which causes the lethality of only males; (3) another recessive lethal mutation, l(2)Co (lethal (2) Corato), probably arisen in the laboratory by mutation. This mutation accounts for the diminished recovery of homozygous females observed in the stock. The genetic features and the cytological analysis of the SD chromosome are reported, as well as the genetic localization of mle-Co and 1(2)Co and their cytogenetic mapping. An allelism test has ascertained that mle-Co is allelic to mle, a male-specific mutation described by Fukunaga et al., 1975. The tight linkage of mle-Co and l(2)Co with Sd is discussed from the standpoint of population genetics.

Homogenates of single individuals of two natural populations and five laboratory populations of Culex pipiens were examined by combining electrophoresis and heat denaturation studies on phosphoglucomutase (PGM). All populations showed a high degree of polymorphism for isoelectrophoretic temperature-resistant (tr) and temperature-sensitive (ts) alleles. Formal genetic data on the heat stability differences of the PGM are given. If both electrophoretic and isoelectrophoretic alleles are taken into account, the mean increase in the degree of heterozygosity is quite remarkable, i.e., about 65%.--The data are considered in relation to the biological significance that this new type of variability of structural genes could have in natural populations.

In 6 species of mosquitoes of the genera Aedes and Culiseta (Culicidae, Diptera) the frequency of phosphoglucomutase (PGM) heat sensitivity alleles is inversely correlated with the temperature of the environment where larvae develop. These data suggest that different selective values are associated with the PGM thermoresistant and thermosensitive genotypes in the different habitats.
