Abstract Nicotinamide Adenine Dinucleotide (NAD+) is implicated in bioenergetics, DNA repair, and senescence. Depletion of NAD+ is associated with aging and neurodegenerative disease, prompting a growing interest in NAD+ supplementation. With rising over-the-counter use of NAD, understanding their impact on anesthetic recovery becomes essential. This study investigates the effect of NADH, a common NAD+ precursor, on anesthesia in rodents. Baseline and post-anesthesia (1.5% isoflurane) open field and Y-maze activity were recorded in adult male and female C57BL/6 mice (n = 8–10/group). NADH (150 mg/kg, intraperitoneal) or vehicle (0.9% normal saline) were given at baseline or during anesthesia. The NADH-treated group exhibited a significant decrease in open-field activity relative to vehicle-treated. This diminished activity was reflected in reduced distance travelled and average velocity after emergence from anesthesia in the NADH-treated group. NADH treatment did not improve Y-maze performance after anesthesia, partly related to reduced locomotor activity in the NADH-treated group. This study demonstrates that NADH does not appear to hasten recovery from anesthesia. Instead, there was a depression in open-field activity and no change in Y-maze performance with NADH supplementation, indicators of locomotive and cognitive recovery in rodents. The broad implications of NAD+ in aging are likely to shape supplementation trends, highlighting the importance of understanding the potential influence of administering NAD+ on anesthetic sensitivity and recovery.

Aluminum nitride (AlN) is an emerging material for integrated quantum photonics with its excellent linear and nonlinear optical properties. In particular, its second-order nonlinear susceptibility χ ^(2) allows single-photon generation. We have grown AlN thin films on silicon nitride (Si _3 N _4 ) via reactive DC magnetron sputtering. The thin films have been characterized using x-ray diffraction (XRD), optical reflectometry, atomic force microscopy (AFM), and scanning electron microscopy. The crystalline properties of the thin films have been improved by optimizing the nitrogen to argon ratio and the magnetron DC power of the deposition process. XRD measurements confirm the fabrication of high-quality c -axis oriented AlN films with a full width at half maximum of the rocking curves of 3.9° for 300 nm-thick films. AFM measurements reveal a root mean square surface roughness below 1 nm. The AlN deposition on SiN allows us to fabricate hybrid photonic circuits with a new approach that avoids the challenging patterning of AlN.

Aquaculture is globally the fastest growing sector of agriculture that needs to be sustainable and must also meet bioeconomic demands. In principle, aquaponics, the combination of aquaculture and horticulture within a single recirculating aquaponic system (SRAPS), provides a sustainable approach; however, it has lower productivity of both fish and plants in comparison to separate recirculating systems. The aim of our new concept for aquaponics is to improve sustainability and productivity concomitant with lowering environmental emissions. The aquaponic system for (nearly) emission free tomato and fish production in greenhouses (ASTAF-PRO) is a new combination of systemic parts, consisting of 2 independent recirculating units: an aquaculture system (RAS) for fish and a hydroponic unit for plants. Both systems are connected by a 1-way valve to deliver fish water containing nutrients into the hydroponic reservoir, where the fish water can be optimized as fertilizer in order to meet specific demands of the plant species. Thus, our double recirculation aquaponic system (DRAPS) can provide optimum conditions for both parts to increase productivity and to prevent any adverse interactions between plant and fish units. Water use is minimized by condensing the plant evapo-transpired water via air conditioning and returning it into the RAS. Testing of the ASTAF-PRO prototype successfully demonstrated the principle using a combination of tilapia and tomato production. Tilapia productivity was similar to single RAS, while tomato production at least indicated the potential for similar efficiency to conventional hydroponics. Thus, ASTAF-PRO improves sustainability and productivity while lowering environmental emissions, and might promote future application of aquaponics for food security.

We study quantum state tomography, entanglement detection and channel noise reconstruction of propagating quantum microwaves via dual-path methods. The presented schemes make use of the following key elements: propagation channels, beam splitters, linear amplifiers and field quadrature detectors. Remarkably, our methods are tolerant to the ubiquitous noise added to the signals by phase-insensitive microwave amplifiers. Furthermore, we analyse our techniques with numerical examples and experimental data, and compare them with the scheme developed in Eichler et al (2011 Phys. Rev. Lett. 106 220503; 2011 Phys. Rev. Lett. 107 113601), based on a single path. Our methods provide key toolbox components that may pave the way towards quantum microwave teleportation and communication protocols.

Josephson parametric amplifiers (JPA) are promising devices for applications in circuit quantum electrodynamics and for studies on propagating quantum microwaves because of their good noise performance. In this work, we present a systematic characterization of a flux-driven JPA at millikelvin temperatures. In particular, we study in detail its squeezing properties by two different detection techniques. With the homodyne setup, we observe the squeezing of vacuum fluctuations by superposing signal and idler bands. For a quantitative analysis, we apply dual-path cross-correlation techniques to reconstruct the Wigner functions of various squeezed vacuum and thermal states. At 10 dB signal gain, we find 4.9 ± 0.2 dB squeezing below the vacuum. In addition, we discuss the physics behind squeezed coherent microwave fields. Finally, we analyze the JPA noise temperature in the degenerate mode and find a value smaller than the standard quantum limit for phase-insensitive amplifiers.

For gradiometric three-Josephson-junction flux qubits, we perform a systematic study on the tuning of the minimal transition frequency, the so-called qubit gap. By replacing one of the qubit's Josephson junctions by a dc superconducting quantum interference device (SQUID), the critical current of this SQUID and, in turn, the qubit gap can be tuned in situ by a control flux threading the SQUID loop. We present spectroscopic measurements demonstrating a well-defined controllability of the qubit gap between zero and more than 10 GHz. This is important for tuning the qubit into and out of resonance with other superconducting quantum circuits in scalable architectures, while still operating it at its symmetry point with optimal dephasing properties. The experimental data agree very well with model calculations based on the full qubit Hamiltonian. From a numerical fit, we determine the Josephson coupling and the charging energies of the qubit junctions. The derived values agree well with those measured for other junctions fabricated on the same chip. We also demonstrate the biasing of gradiometric flux qubits near the symmetry point by trapping an odd number of flux quanta in the gradiometer loop. In this way, we study the effect of the significant kinetic inductance, thereby obtaining valuable information for the qubit design.

Abstract We present a novel workflow for forecasting production in unconventional reservoirs using reduced-order models and machine-learning. Our physics-informed machine-learning workflow addresses the challenges to real-time reservoir management in unconventionals, namely the lack of data (i.e., the time-frame for which the wells have been producing), and the significant computational expense of high-fidelity modeling. We do this by applying the machine-learning paradigm of transfer learning, where we combine fast, but less accurate reduced-order models with slow, but accurate high-fidelity models. We use the Patzek model (Proc Natl Acad Sci 11:19731–19736, https://doi.org/10.1073/pnas.1313380110 , 2013) as the reduced-order model to generate synthetic production data and supplement this data with synthetic production data obtained from high-fidelity discrete fracture network simulations of the site of interest. Our results demonstrate that training with low-fidelity models is not sufficient for accurate forecasting, but transfer learning is able to augment the knowledge and perform well once trained with the small set of results from the high-fidelity model. Such a physics-informed machine-learning (PIML) workflow, grounded in physics, is a viable candidate for real-time history matching and production forecasting in a fractured shale gas reservoir.

Abstract Background The most severe form of human malaria is caused by the protozoan parasite Plasmodium falciparum. This unicellular organism is a member of a subgenus of Plasmodium called the Laverania that infects apes, with P. falciparum being the only member that infects humans. The exceptional virulence of this species to humans can be largely attributed to a family of variant surface antigens placed by the parasites onto the surface of infected red blood cells that mediate adherence to the vascular endothelium. These proteins are encoded by a large, multicopy gene family called var, with each var gene encoding a different form of the protein. By changing which var gene is expressed, parasites avoid immune recognition, a process called antigenic variation that underlies the chronic nature of malaria infections. Results Here we show that the common ancestor of the branch of the Laverania lineage that includes the human parasite underwent a remarkable change in the organization and structure of elements linked to the complex transcriptional regulation displayed by the var gene family. Unlike the other members of the Laverania, the clade that gave rise to P. falciparum evolved distinct subsets of var genes distinguishable by different upstream transcriptional regulatory regions that have been associated with different expression profiles and virulence properties. In addition, two uniquely conserved var genes that have been proposed to play a role in coordinating transcriptional switching similarly arose uniquely within this clade. We hypothesize that these changes originated at a time of dramatic climatic change on the African continent that is predicted to have led to significant changes in transmission dynamics, thus selecting for patterns of antigenic variation that enabled lengthier, more chronic infections. Conclusions These observations suggest that changes in transmission dynamics selected for significant alterations in the transcriptional regulatory mechanisms that mediate antigenic variation in the parasite lineage that includes P. falciparum. These changes likely underlie the chronic nature of these infections as well as their exceptional virulence.

Abstract Most of solid-state spin physics arising from spin–orbit coupling, from fundamental phenomena to industrial applications, relies on symmetry-protected degeneracies. So does the Zeeman spin–orbit coupling, expected to manifest itself in a wide range of antiferromagnetic conductors. Yet, experimental proof of this phenomenon has been lacking. Here we demonstrate that the Néel state of the layered organic superconductor κ-(BETS)2FeBr4 shows no spin modulation of the Shubnikov–de Haas oscillations, contrary to its paramagnetic state. This is unambiguous evidence for the spin degeneracy of Landau levels, a direct manifestation of the Zeeman spin–orbit coupling. Likewise, we show that spin modulation is absent in electron-doped Nd1.85Ce0.15CuO4, which evidences the presence of Néel order in this cuprate superconductor even at optimal doping. Obtained on two very different materials, our results demonstrate the generic character of the Zeeman spin–orbit coupling.

The identification of specific epigenetic regulatory proteins in infectious organisms has become a high-profile research topic and a focus for several drug development initiatives. However, studies that define specific roles for different epigenetic modifiers occasionally report differing results, and we similarly provide evidence regarding the histone methyltransferase PfSET10 that is in stark contrast with previously published results.

Though rare, fibroepithelial polyps of the ureter consistently present with symptoms such as flank discomfort and may lead to renal failure. This often affects young adults and characteristically arises in the proximal ureter. The pathogenesis is unclear. Flank pain is the most common presenting symptom, which may create diagnostic confusion. Indeed, resection provides lasting relief which otherwise may elude patients who receive other treatments. Resection modality may be selected on a patient-by-patient basis. Here we present a case in which flank pain, and historical episodes of abdominal pain, were resolved by excision of a fibroepithelial polyp by a robotic assisted laparoscopic approach.

Direct electrical brain stimulation can induce widespread changes in neural activity, offering a means to modulate network-wide activity and treat disease. Here, the authors show that the low-frequency functional connectivity profile of a stimulation target predicts where induced theta activity occurs.
