International audience; The existence of narrow axial volcanic zones of mid-oceanic ridges testifies of the underlying concentration of both melt distribution and tectonic strain. As a result of repeated diking and faulting, axial volcanic zones therefore represent a spectacular topographic expression of plate divergence. However, the submarine location of oceanic ridges makes it difficult to constrain the interplay between tectonic and magmatic processes in time and space. In this study, we use the Dabbahu–Manda Hararo (DMH) magmatic rift segment (Afar, Ethiopia) to provide quantitative constraints on the response of tectonic processes to variations in magma supply at divergent plate boundaries. The DMH magmatic rift segment is considered an analogue of an oceanic ridge, exhibiting a fault pattern, extension rate and topographic relief comparable to intermediate-to slow-spreading ridges. Here, we focus on the northern and central parts of DMH rift, where we present quantitative slip rates for the past 40 kyr for major and minor normal fault scarps in the vicinity of a recent (September 2005) dike intrusion. The data obtained show that the axial valley topography has been created by enhanced slip rates that occurred during periods of limited volcanism, suggestive of reduced magmatic activity, probably in association with changes in strain distribution in the crust. Our results indicate that the development of the axial valley topography has been regulated by the lifetimes of the magma reservoirs and their spatial distribution along the segment , and thus to the magmatic cycles of replenishment/differentiation (

Unravelling the volcanic history of the Dabbahu/Manda Hararo rift segment in the Afar depression (Ethiopia) using a combination of cosmogenic (36Cl and 3He) surface exposure dating of basaltic lava-flows, field observations, geological mapping and geochemistry, we show in this paper that magmatic activity in this rift segment alternates between two distinct magma chambers. Recent activity in the Dabbahu rift (notably the 2005–2010 dyking crises) has been fed by a seismically well-identified magma reservoir within the rift axis, and we show here that this magma body has been active over the last 30 kyr. However, in addition to this axial magma reservoir, we highlight in this paper the importance of a second, distinct magma reservoir, located 15 km west of the current axis, which has been the principal focus of magma accumulation from 15 ka to the subrecent. Magma supply to the axial reservoir substantially decreased between 20 ka and the present day, while the flank reservoir appears to have been regularly supplied with magma since 15 ka ago, resulting in less variably differentiated lavas. The trace element characteristics of magmas from both reservoirs were generated by variable degrees of partial melting of a single homogeneous mantle source, but their respective magmas evolved separately in distinct crustal plumbing systems.Magmatism in the Dabbahu/Manda Hararo rift segment is not focussed within the current axial depression but instead is spread out over at least 15 km on the western flank. This is consistent with magneto-telluric observations which show that two magma bodies are present below the segment, with the main accumulation of magma currently located below the western flank, precisely where the most voluminous recent (Applying these observations to slow spreading mid-ocean ridges indicates that magma bodies likely have a lifetime of a least 20 ka, and that the continuity of magmatic activity is maintained by a system of separate relaying reservoirs, which could in return control the location of spreading. This long term (>105 yr) alternation between distinct crustal reservoirs located broadly at the same location relative to the segment appears to be a key feature for organizing and maintaining active spreading centres over stable soft points in the mantle.

The composition of the mantle plumes that created large oceanic plateaus such as Ontong Java or the Caribbean is still poorly known. Geochemical and isotopic studies on accreted portions of the Caribbean plateau have shown that the plume source was heterogeneous and contained isotopically depleted and relatively enriched portions. A distinctive feature of samples from the Caribbean plateau is their unusual Sr isotopic compositions, which, at a given Nd isotopic ratio, are far higher than in samples from other oceanic plateaus. Sr, O and He isotopic compositions of whole rocks and magmatic minerals (clinopyroxene or olivine) separated from komatiites, gabbros and peridotites from Gorgona Island in Colombia were determined to investigate the origin of these anomalously radiogenic compositions. Sequentially leached clinopyroxenes have Sr isotopic compositions in the range 87Sr/86Sr=0.70271–0.70352, systematically lower than those of leached and unleached whole rocks. Oxygen isotopic ratios of clinopyroxene vary within the range ?18 O=5.18–5.35‰, similar to that recorded in oceanic island basalts. He isotopic ratios are high (R/Ra=8–19). The lower 87Sr/86Sr ratios of most of the clinopyroxenes shift the field of the Caribbean plateau in Nd–Sr isotope diagrams toward more `normal' values, i.e. a position closer to the field defined by mid-ocean ridge basalts and oceanic-island basalts. Three clinopyroxenes have slightly higher 87Sr/86Sr ratios that cannot be explained by an assimilation model. The high 87Sr/86Sr and variations of 143Nd/144Nd are interpreted as a source characteristic. Trace-element ratios, however, are controlled mainly by fractionation during partial melting. We combine these isotopic data in a heterogeneous plume source model that accounts for the diversity of isotopic signatures recorded on Gorgona Island and throughout the Caribbean plateau. The heterogeneities are related to old recycled oceanic lithosphere in the plume source; the high 3He/4He ratios may indicate that the source material once resided in the lower mantle.

After the earthquakes of September 26, 1997, that hit the Umbria-Marcheboundary (Apennine, Central Italy), with a maximum 6.0 Mw, aprogram of geochemical surveying together with a collection ofhydrogeological changes episodes was extended throughout theepicentre-area, taking the yearly period of the seismic sequence as a whole.After a first areal screening, the Bagni di Triponzo thermal spring wasselected for a discrete temporal monitoring (weekly and monthly basis),being the unique thermal spring throughout the epicentre area. This sitedeserves peculiar interest in deepening the knowledge about deep fluidscirculation changing during seismicity.Laboratory and on-field analyses included major, minor and trace elementsas well as dissolved gases (He, Ar, CH4, CO2, H2S,222Rn, NH4, As, Li, Fe, B, etc...) and selected isotopic ratios(C, H, O, He, Sr, Cl), meaningful from tectonic point of view.The chemistry and isotopic chemistry of the spring were fully outlined anddiscussed, pointing out the main process involving the thermal aquifer: thewater-rock interaction inside the Evaporite Triassic Basement (ETB),possibly involving also the Paleozoic Crystalline Basement. On theother hand, sudden and apparent geochemical and hydrogeologicalvariations during the seismic sequence ruled out an evolution in thewater-rock interaction processes. They occurred both at depth, i.e.,induced by fluid remobilization within the crust explained by the Coseismic Strain Model and by the Fault Valve Activity Model, and in the shallow part of the reservoir (i.e., meteoric watercontamination). A statistical multivariable analysis (Factor Analysis) wasaccomplished to better constrain the correlation between the paroxysmalphases of the seismic sequence and the observed trends and spike-likeanomalies. The groundwater variations was inferred to occur mainly insidethe ETB, from depth (1–2 km) up to surface, particularly in associationof the Sellano earthquake (14/10/1997) and of the seismic re-activationof the sequence at the end of March 1998 (Gualdo Tadino-Rigali andVerchiano areas). The lack of deeper input from below the ETB (slightsignature of PCB), as the lack of He mantle signature, during the seismicperiod as a whole, accounted for seismogenic fault segments rooted onlyin the crust. The results also provide useful information about theearthquake-related response mechanisms occurring at this site, thatrepresent the basic task for planning and managing the impendinghydro-geochemical network aimed at defining the relationships betweenseismic cycle, fluids and reliable earthquake forerunners.

International audience; Crustal extension at mature continental rifts and oceanic ridges occurs by a combination of normal faulting and magma injection, which interact to create rift morphology. Quantifying the relative roles of faulting and melt intrusion in accommodating extension at magmatic rifts remains difficult and requires studies at sufficient spatial and temporal scales to resolve the interaction between these processes. In this study we provide new chronological constraints based on cosmogenic exposure dating for the ˜100 kyr topographic evolution of a young and active magmatic rift segment in Afar, Ethiopia. We combine structural investigations, field mapping, geochemical analysis and cosmogenic 3He exposure dating of lava surfaces in order to investigate the interplay between volcanic activity and fault growth in the northern part of the axial depression, where the rift segment intersects a large stratovolcano. Our results allow us to determine the roles of the various magma reservoirs feeding this rift system and their interactions during accretion over the past 100 kyr. New age data for key lava units allow several magmatic cycles to be distinguished. Each cycle lasts 20-40 ka resulting in periods of high and low magma supply rate. The variations in magma supply rate at the segment extremity strongly affect the development of the rift depression, with the availability of melt controlling the morphological impact of faulting. Melts from different magma reservoirs feeding the segment are chemically distinct and geochemical analysis of lavas from the rift floor allows their respective contributions to maintaining magmatic accretion to be estimated. We propose that melts from the magma reservoir at the northern end of the segment contribute around one-third of the length of this portion of the segment, whereas the mid-segment reservoir is responsible for the remaining two-thirds of the segment accretion.

BACKGROUND: In patients with suspected heparin-induced thrombocytopenia (HIT) who need renal replacement therapy, a nonheparin anticoagulant has to be chosen to prevent thrombosis in the extracorporeal circuit. Danaparoid, a low-molecular-weight heparinoid consisting of heparan sulphate, dermatan sulphate, and chondroitin sulphate, is recommended for systemic anticoagulation in patients with HIT. However, there are few data on the use of danaparoid in patients with acute renal failure, especially in patients dependent on renal replacement therapy such as continuous venovenous hemofiltration (CVVH). In the present study, we analyzed the pharmacokinetics and pharmacodynamics of danaparoid during CVVH in patients with suspected HIT. METHODS: Based on a mathematical model, a dosing scheme for danaparoid was designed, aiming at anti-Xa levels of 0.5 to 0.7 U/mL, with a maximum of 1.0 U/mL. This dosing scheme was prospectively tested in the first CVVH run of a cohort of five patients with suspected HIT. CVVH with a blood flow rate of 150 mL/minute and a substitution rate of 2,000 mL/hour was performed with a cellulose triacetate membrane. Danaparoid was administered as a continuous infusion of 100 anti-Xa-U/hour after a loading dose of 3,500 anti-Xa-U. Serial measurements of anti-Xa activity and prothrombin fragment F1+2 were performed at baseline, at t = 5, 15, and 30 minutes, and at t = 1, 2, 4, 8, 16, and 24 hours after the danaparoid loading dose. RESULTS: The median anti-Xa activity reached a maximum of 1.02 (0.66 to 1.31) anti-Xa-U/mL after 15 minutes and gradually declined to 0.40 (0.15 to 0.58) anti-Xa-U/mL over the span of 24 hours. Target anti-Xa levels were reached from 2 to 12 hours after the loading dose. Median prothrombin fragment F1+2 gradually decreased from 432 (200 to 768) to 262 (248 to 317) pmol/L after 24 hours. No bleeding or thromboembolic events occurred throughout the described treatment period. CONCLUSION: Danaparoid administered by a continuous infusion of 100 anti-Xa-U/hour after a loading dose of 3,500 anti-Xa-U elicited target anti-Xa levels from 2 to 12 hours after the loading dose, without bleeding or thromboembolic events during the described CVVH treatment in patients with suspected HIT

It is shown that a block lower triangular unitary operator with Kronecker index N < ∞ admits a factorization into a product of N block lower triangular unitary operators with Kronecker index equal to 1. If the block lower triangular operator is exponentially decaying with respect to the main diagonal, then the factors can be chosen exponentially decaying too. A description of the elementary factors is also given. The proofs are based on the fact that the operators involved appear as input output operators of finite dimensional time varying unitary systems, and a cascade decomposition of such systems.

Membrane vesicles derived by osmotic lysis of spheroplasts of Paracoccus denitrificans (ATCC 13543) grown aerobically in continuous culture under conditions of carbon limitation with succinate as sole carbon and energy source accumulate radioactivity in an uncoupler-sensitive respiration-dependent manner when incubated in the presence of [14C]succinate or L-[14C]malate. Membranes prepared from cells grown under conditions of sulphate limitation with succinate as the sole carbon source do not accumulate L-[14C]malate during ascorbate + N,N,N′,N′-tetramethyl-p-phenylene diamine (TMPD) oxidation. The apparent Km for succinate and L-malate uptake is 6.7 and 10 μM respectively with a Vmax for uptake of either substrate being 8.3 nmol/min per milligram of membrane protein. Intravesicular radioactivity was largely confined to C4 dicarboxylic acids, succinate, fumarate, and malate and was freely exchangeable with external unlabeiled C4 dicarboxylic acids but not aspartate. Both ubiquinol1 and ascorbate (+ TMPD) oxidation supported accumulative uptake of succinate or L-malate but NADH did not. Since energization of the dicarboxylate transport system is accomplished by the oxidation of ascorbate + TMPD in the presence of antimycin A, it is concluded that heterotrophically grown P. denitrificans containing cytochrome a3 possess a functional energy-coupling site 3 (terminal energy-transducing region of the respiratory chain) despite claims to the contrary.

The steady-state levels of zinc in Candida utilis yeast grown in continuous culture under conditions of zinc limitation are 2+/mg dry weight of cells. Unlike carbon-limited cells, zinc-depleted cells from a zinc-limited chemostat possess the capacity to accumulate and store zinc at levels far in excess of the steady-state level of 4 nmol/mg dry biomass observed in carbon-limited chemostat cultures. Zinc uptake is energy-dependent and apparently unidirectional since accumulated 65Zn neither exits from preloaded cells nor exchanges with cold Zn2+. The transport system exhibits a high affinity for Zn2+ (Km = 0.36 μM) with a Vmax of 2.2 nmol per minute per milligram dry weight of cells. Growth during the period of the uptake assay is responsible for the apparent plateau level of 35 nmol Zn2+/mg dry weight of cells achieved after 20–30 min in the presence of 65Zn at pH 4.5 and 30 °C. Inhibition of growth during the uptake assay by cycloheximide results in a biphasic linear pattern of zinc accumulation where the cellular zinc is about 60 nmol/mg dry weight after 1 h. The enhanced level of accumulated zinc is not inhibitory to growth. Zinc-depleted C. utilis contains elevated amounts of polyphosphate and this anionic polymer must be considered a possible zinc-sequestering and storage agent. Present experimental evidence does not allow discrimination between possible regulation of zinc homeostasis either by inhibition of zinc efflux through control of the membrane carrier or by control of the synthesis of a cytoplasmic zinc-sequestering macromolecule.

A small-scale chemostat has been used to assess the basic nutritional requirements of curdlan producing strains of Alcaligenes faecalis. A lean, chemically-defined mineral salts medium has been formulated compatible with operation of a continuous culture at dilution rates equivalent to μxax observed in batch-culture. Molar growth yields for carbon, nitrogen, phosphorus and oxygen have been determined. The optimized minimal salts medium with 2.5% glucose and 90mM NH4Cl supported a steady-state nitrogen-limited biomass of 8.9g dry biomass/L at a max. dilution rate of 0.23hr−1. By virtue of its ability to increase μmax, the addition of yeast extract to the chemostat nutrient feed resulted in a significant increase in the max. biomass productivity of the continuous fermenter. Under these conditions curdlan is not synthesized in chemostat culture. Operation of a nitrogen-limited chemostat at Dmax resulted in the complete replacement of the original uracil-requiring mutant (ATCC 21680) by a non uracil-requiring variant strain of A. faecalis. Both the parent auxotroph and apparent revertant strain display similar physiological properties including the capacity to synthesize an extracellular, water-insoluble, neutral β-1,3-glucan polymer (curdlan) under appropriate non-growing conditions.

Lysophosphatidylinositol acyltransferase utilizing arachidonoyl CoA and lysophosphatidylcholine acyltransferase utilizing linoleoyl CoA were measured in subcellular fractions of rat brain. In general, the distribution of activities paralleled that of NADPH–cytochrome c reductase. It is concluded that the endoplasmic reticulum is the major site of these acyltransferase activities in rat brain.

The biosynthesis of a thermogelable, extracellular homo-β-(1 → 3)-glucan called "curdlan," has been studied in batch and continuous cultures of Alcaligenes faecalis var. myxogenes. Curdlan production is associated with the poststationary phase of a nitrogen-depleted, aerobic batch culture. Exopolymer is not detected in single-stage, carbon-limited continuous cultures but curdlan can be isolated from the effluent of a nitrogen-limited chemostat operating at a dilution rate (D) of −1. A spontaneous variant of strain ATCC 21680 was isolated and found to be compatible with long-term, nitrogen-limited chemostat culture. The specific rate of curdlan production is approximately four times higher in poststationary batch cultures than in single-stage continuous fermentations. The product yield (Yp/s) associated with batch processing (nongrowing cultures) is approximately 0.5 g curdlan/g glucose, with CO2 being the only detectable by-product.

Candida utilis NRRL Y-900 was grown in aerobic continuous culture in a minimal salts medium with sucrose (1% w/v) as the carbon source. Increasing the concentration of zinc in the medium from 2.3 μM to about 30 μM results in an increase in the apparent critical dilution rate from 0.3 to 0.47 h−1, and in the maximum biomass productivity from 1.5 g dry weight per litre per hour (at D = 0.33 h−1) to 2.56 g per litre per hour (at D = 0.45 h−1). The maximum steady-state level of cell-associated zinc (at D = 0.4 h−1) is 4 nmol Zn2+/mg dry weight, during carbon-limited growth, and about 9 nmol Zn2+/mg dry weight when FeCl3 is omitted from the medium. At input zinc concentrations 2+ the culture becomes zinc-limited. Zinc-limitation results in a decrease in the growth yield Ysucrose from 0.54 to 0.38 g cells/g sucrose consumed, and Y0 from 22 to 13 g cells/g-atom O2, suggesting an altered efficiency of energy metabolism. The composition of the culture biomass with respect to protein and RNA content is not affected by zinc limitation.
