Abstract Background Inflammatory bowel disease (IBD) patients experience recurrent episodes of intestinal inflammation and often follow an unpredictable disease course. Mucosal colonization with adherent-invasive Escherichia coli (AIEC) are believed to perpetuate intestinal inflammation. However, it remains unclear if the 24-year-old AIEC in vitro definition fully predicts mucosal colonization in vivo. To fill this gap, we have developed a novel molecular barcoding approach to distinguish strain variants in the gut and have integrated this approach to explore mucosal colonization of distinct patient-derived E. coli isolates in gnotobiotic mouse models of colitis. Results Germ-free inflammation-susceptible interleukin-10-deficient (Il10 −/− ) and inflammation-resistant WT mice were colonized with a consortium of AIEC and non-AIEC strains, then given a murine fecal transplant to provide niche competition. E. coli strains isolated from human intestinal tissue were each marked with a unique molecular barcode that permits identification and quantification by barcode-targeted sequencing. 16S rRNA sequencing was used to evaluate the microbiome response to E. coli colonization. Our data reveal that specific AIEC and non-AIEC strains reproducibly colonize the intestinal mucosa of WT and Il10 −/− mice. These E. coli expand in Il10 −/− mice during inflammation and induce compositional dysbiosis to the microbiome in an inflammation-dependent manner. In turn, specific microbes co-evolve in inflamed mice, potentially diversifying E. coli colonization patterns. We observed no selectivity in E. coli colonization patterns in the fecal contents, indicating minimal selective pressure in this niche from host-microbe and interbacterial interactions. Because select AIEC and non-AIEC strains colonize the mucosa, this suggests the in vitro AIEC definition may not fully predict in vivo colonization potential. Further comparison of seven E. coli genomes pinpointed unique genomic features contained only in highly colonizing strains (two AIEC and two non-AIEC). Those colonization-associated features may convey metabolic advantages (e.g., iron acquisition and carbohydrate consumption) to promote efficient mucosal colonization. Conclusions Our findings establish the in vivo mucosal colonizer, not necessarily AIEC, as a principal dysbiosis driver through crosstalk with host and associated microbes. Furthermore, we highlight the utility of high-throughput screens to decode the in vivo colonization dynamics of patient-derived bacteria in murine models. Video Abstract

Post-contact immobility (PCI) is a final attempt to avoid predation. Here, for the first time, we examine the pattern of movement and immobility when antlion larvae resume activity after PCI. To simulate contact with, and escape from, a predator we dropped the larvae onto three different substrates: Paper, Shallow sand (2.3mm-deep) and Deep sand (4.6mm-deep). The Paper lining a Petri dish represented a hard surface that antlion larvae could not penetrate to hide. The Shallow sand permitted the antlions to dig but not to submerge completely whereas the Deep sand allowed them both to dig and to submerge. We tracked their paths automatically and recorded alternating immobility and movement durations over 90min. On the impenetrable substrate, antlion larvae showed super-diffusive dispersal, their movement durations became longer, their immobility durations became shorter and their instantaneous speeds increased. This is consistent with the antlions needing to leave an area of hard substrate and quickly to find somewhere to hide. On Shallow sand, antlion larvae exhibited a modest increase in movement duration, a modest decrease in immobility duration and a concomitant diffusive dispersal. This is consistent with their use of a spiral search, presumably for a suitable depth of sand, to conceal themselves. On Deep sand, the movement and immobility durations of the antlion larvae did not change and their dispersal was sub-diffusive because they were able to bury themselves. On Paper, the distribution of immobility durations had a long tail, consistent with a log-normal distribution. On Shallow and Deep sand, most of the distribution was fitted better by a power law or a log-normal. Our results suggest that PCI in antlion larvae is a disruptive event and that post-PCI movement and immobility gradually return to the pattern typical of intermittent locomotion, depending on the scope for burying and hiding in the substrate.

In Guyana, the coastal plains dominate agricultural production, while the hinterland is an emerging agricultural frontier. The coastal and hinterland regions have differing agro-climatic conditions, but share immediate climate change and environmental degradation pressures, including soil degradation. Even though climate change adaptation is prioritized over greenhouse gas mitigation in Guyana, soil-focused farming, otherwise known as sustainable soil management (SSM), can provide a system that creates synergies between these two facets of climate-smart agriculture and, also, promotes soil security. This article proposes a bottom-up planning process for SSM in Guyana by assessing its underlying psycho-social and physical facilitators and barriers. The main questions addressed are: what are the attitudes of Guyanese farmers to climate change? What are their capabilities for SSM, in terms of education, technology and government support? In answering these questions, inductive-derived thematic analysis of transcripts derived from in-depth telephone interviews with seventeen (17) farmers, from coastal and hinterland regions, provides an initial basis for ground truthing on the local appropriateness of SSM. Results show that hinterland farmers are more emotive and value-driven about their environment, while coastal farmers, instead, prioritize access to markets and gaining favorable prices for their commodities. Additionally, the lack of education and training are identified as severe limitations to the capabilities of farmers to practice SSM. In conclusion, a weak marketing environment is seen as a binding constraint of sustainable intensification as surplus goods attract low prices. Stronger linkages to dynamic markets, as well as increased investment opportunities are needed for sustainable farming to become economically feasible. Therefore, psychosocial capital must be strengthened before any natural capital is improved under Guyana's various agro-environmental policies.

Understanding how mobile, marine predators use three-dimensional space over time is central to inform management and conservation actions. Combining tracking technologies can yield powerful datasets over multiple spatio-temporal scales to provide critical information for these purposes. For the white shark (Carcharodon carcharias), detailed movement and migration information over ontogeny, including inter- and intra-annual variation in timing of movement phases, is largely unknown in the western North Atlantic (WNA), a relatively understudied area for this species. To address this need, we tracked 48 large juvenile to adult white sharks between 2012 and 2020, using a combination of satellite-linked and acoustic telemetry. Overall, WNA white sharks showed repeatable and predictable patterns in horizontal movements, although there was variation in these movements related to sex and size. While most sharks undertook an annual migratory cycle with the majority of time spent over the continental shelf, some individuals, particularly adult females, made extensive forays into the open ocean as far east as beyond the Mid-Atlantic Ridge. Moreover, increased off-shelf use occurred with body size even though migration and residency phases were conserved. Summer residency areas included coastal Massachusetts and portions of Atlantic Canada, with individuals showing fidelity to specific regions over multiple years. An autumn/winter migration occurred with sharks moving rapidly south to overwintering residency areas in the southeastern United States Atlantic and Gulf of Mexico, where they remained until the following spring/summer. While broad residency and migration periods were consistent, migratory timing varied among years and among individuals within years. White sharks monitored with pop-up satellite-linked archival tags made extensive use of the water column (0–872 m) and experienced a broad range of temperatures (−0.9 – 30.5°C), with evidence for differential vertical use based on migration and residency phases. Overall, results show dynamic inter- and intra-annual three-dimensional patterns of movements conserved within discrete phases. These results demonstrate the value of using multiple tag types to track long-term movements of large mobile species. Our findings expand knowledge of the movements and migration of the WNA white shark population and comprise critically important information to inform sound management strategies for the species.

Many young birds die soon after fledging, as they lack the skills to find food and avoid predation. Post-fledging parental care is assumed to assist acquisition of these vital skills. However, we still lack empirical examples examining the length of time fledglings spend with parents, how they associate during this critical time, or whether such variation in the fledgling dependency period has consequences for the survival and behaviour of young as they navigate their first year of independent life. Here, we make use of observations and radio frequency identity (RFID) logs of visits to supplementary feeding stations to investigate how condition of fledgling hihi (stitchbird, Notiomystis cincta), a New Zealand passerine, predicts dispersal behaviour and tendency to follow parents during their 2 week post-fledging dependence period. We find that thinner fledglings followed their parents more closely in time when visiting feeding stations, compared to fatter siblings (all following ranged from 3 s to 10 min). However, broods in poorer condition tended to disperse from the natal territory up to 6.5 days earlier than broods of fatter fledglings (all dispersed within 14 days). Our results did not find that sociality or survival during the first year of life differed depending on variation in fledgling behaviour; neither following parents closely nor dispersing later predicted each bird's number of associates (degree), or survival over winter. These results suggest that fledglings may be able to compensate for early differences in condition with behaviour, either during the post-fledging dependence period or when independent.

Individual animals can often move more safely or more efficiently as members of a group. This can be as simple as safety in numbers or as sophisticated as aerodynamic or hydrodynamic cooperation. Here, we show that individual plant–animal worms (Symsagittifera roscoffensis) can move to safety more quickly through flocculation. Flocs form in response to turbulence that might otherwise carry these beach-dwelling worms out to sea. They allow the worms to descend much more quickly to the safety of the substrate than single worms could swim. Descent speed increases with floc size such that larger flocs can catch up with smaller ones and engulf them to become even larger and faster. To our knowledge, this is the first demonstration of social flocculation in a wild, multicellular organism. It is also remarkable that such effective flocculation occurs where the components are comparatively large multicellular organisms organized as entangled ensembles.

Collective motion is a fascinating and intensely studied manifestation of collective behaviour. Circular milling is an impressive example. It occurs in fishes, processionary caterpillars and army ants, among others. Its adaptive significance, however, is not yet well understood. Recently, we demonstrated experimentally circular milling in the marine plant–animal worm Symsagittifera roscoffensis. We hypothesized that its function is to gather the worms and facilitate the dense films they form on the beach to promote the photosynthesis of their symbiotic algae. Here, we report for the first time, to our knowledge, the occurrence of S. roscoffensis circular mills in nature and show that it is by no means rare. The size and behaviour of circular mills in their natural environment is compatible with our earlier experimental results. This makes S. roscoffensis a good study system for understanding the proximate and ultimate mechanisms of circular milling.

To find useful work to do for their colony, individual eusocial animals have to move, somehow staying attentive to relevant social information. Recent research on individual Temnothorax albipennis ants moving inside their colony’s nest found a power-law relationship between a movement’s duration and its average speed; and a universal speed profile for movements showing that they mostly fluctuate around a constant average speed. From this predictability it was inferred that movement durations are somehow determined before the movement itself. Here, we find similar results in lone T. albipennis ants exploring a large arena outside the nest, both when the arena is clean and when it contains chemical information left by previous nest-mates. This implies that these movement characteristics originate from the same individual neural and/or physiological mechanism(s), operating without immediate regard to social influences. However, the presence of pheromones and/or other cues was found to affect the inter-event speed correlations. Hence we suggest that ants’ motor planning results in intermittent response to the social environment: movement duration is adjusted in response to social information only between movements, not during them. This environmentally flexible, intermittently responsive movement behaviour points towards a spatially allocated division of labour in this species. It also prompts more general questions on collective animal movement and the role of intermittent causation from higher to lower organizational levels in the stability of complex systems.

How do animals in groups organize their work? Division of labour, i.e. the process by which individuals within a group choose which tasks to perform, has been extensively studied in social insects. Variability among individuals within a colony seems to underpin both the decision over which tasks to perform and the amount of effort to invest in a task. Studies have focused mainly on discrete tasks, i.e. tasks with a recognizable end. Here, we study the distribution of effort in nest seeking, in the absence of new nest sites. Hence, this task is open-ended and individuals have to decide when to stop searching, even though the task has not been completed. We show that collective search effort declines when colonies inhabit better homes, as a consequence of a reduction in the number of bouts (exploratory events). Furthermore, we show an increase in bout exploration time and a decrease in bout instantaneous speed for colonies inhabiting better homes. The effect of treatment on bout effort is very small; however, we suggest that the organization of work performed within nest searching is achieved both by a process of self-selection of the most hard-working ants and individual effort adjustment.

Insect societies are complex systems, displaying emergent properties much greater than the sum of their individual parts. As such, the concept of these societies as single 'superorganisms' is widely applied to describe their organisation and biology. Here, we test the applicability of this concept to the response of social insect colonies to predation during a vulnerable period of their life history. We used the model system of house-hunting behaviour in the ant Temnothorax albipennis. We show that removing individuals from directly within the nest causes an evacuation response, while removing ants at the periphery of scouting activity causes the colony to withdraw back into the nest. This suggests that colonies react differentially, but in a coordinated fashion, to these differing types of predation. Our findings lend support to the superorganism concept, as the whole society reacts much like a single organism would in response to attacks on different parts of its body. The implication of this is that a collective reaction to the location of worker loss within insect colonies is key to avoiding further harm, much in the same way that the nervous systems of individuals facilitate the avoidance of localised damage.

Collective decision-making is a characteristic of societies ranging from ants to humans. The ant Temnothorax albipennis is known to use quorum sensing to collectively decide on a new home; emigration to a new nest site occurs when the number of ants favouring the new site becomes quorate. There are several possible mechanisms by which ant colonies can select the best nest site among alternatives based on a quorum mechanism. In this study, we use computational models to examine the implications of heterogeneous acceptance thresholds across individual ants in collective nest choice behaviour. We take a minimalist approach to develop a differential equation model and a corresponding non-spatial agent-based model. We show, consistent with existing empirical evidence, that heterogeneity in acceptance thresholds is a viable mechanism for efficient nest choice behaviour. In particular, we show that the proposed models show speed–accuracy trade-offs and speed–cohesion trade-offs when we vary the number of scouts or the quorum threshold.

Resolving the geographic extent and timing of coastal shark migrations, as well as their environmental cues, is essential for refining shark management strategies in anticipation of increasing anthropogenic stressors to coastal ecosystems. We employed a regional-scale passive acoustic telemetry array encompassing 300 km of the east Florida coast to assess what factors influence site fidelity of juvenile lemon sharks (Negaprion brevirostris) to an exposed coastal nursery at Cape Canaveral, and to document the timing and rate of their seasonal migrations. Movements of 54 juvenile lemon sharks were monitored for three years with individuals tracked for up to 751 days. While most sharks demonstrated site fidelity to the Cape Canaveral region December through February under typical winter water temperatures, historically extreme declines in ocean temperature were accompanied by rapid and often temporary, southward displacements of up to 190 km along the Florida east coast. From late February through April each year, most sharks initiated a northward migration at speeds of up to 64 km day(-1) with several individuals then detected in compatible estuarine telemetry arrays in Georgia and South Carolina up to 472 km from release locations. Nineteen sharks returned for a second or even third consecutive winter, thus demonstrating strong seasonal philopatry to the Cape Canaveral region. The long distance movements and habitat associations of immature lemon sharks along the US southeast coast contrast sharply with the natal site fidelity observed in this species at other sites in the western Atlantic Ocean. These findings validate the existing multi-state management strategies now in place. Results also affirm the value of collaborative passive arrays for resolving seasonal movements and habitat preferences of migratory coastal shark species not easily studied with other tagging techniques.

We present a dynamical systems analysis of a decision-making mechanism inspired by collective choice in house-hunting honeybee swarms, revealing the crucial role of cross-inhibitory 'stop-signalling' in improving the decision-making capabilities. We show that strength of cross-inhibition is a decision-parameter influencing how decisions depend both on the difference in value and on the mean value of the alternatives; this is in contrast to many previous mechanistic models of decision-making, which are typically sensitive to decision accuracy rather than the value of the option chosen. The strength of cross-inhibition determines when deadlock over similarly valued alternatives is maintained or broken, as a function of the mean value; thus, changes in cross-inhibition strength allow adaptive time-dependent decision-making strategies. Cross-inhibition also tunes the minimum difference between alternatives required for reliable discrimination, in a manner similar to Weber's law of just-noticeable difference. Finally, cross-inhibition tunes the speed-accuracy trade-off realised when differences in the values of the alternatives are sufficiently large to matter. We propose that the model, and the significant role of the values of the alternatives, may describe other decision-making systems, including intracellular regulatory circuits, and simple neural circuits, and may provide guidance in the design of decision-making algorithms for artificial systems, particularly those functioning without centralised control.
