Aim: To study the effect of supplementation of different levels of selenium as nanoparticles/sodium selenite on blood biochemical profile and humoral immunity in male Wistar rats. Materials and Methods: The experimental research was conducted at Division of Animal Nutrition, Indian Veterinary Research Institute, Izatnagar. 63 male Wistar rats were divided into 9 equal groups on the basis of their mean body weight (BW) (124.3±3.1 g BW) following completely randomized design. Experimental feeding was similar in all the groups except for the source and level of selenium (Se) in the diet. While Group 1 (control) was fed a basal diet with no Se supplementation, in Groups 2 and 3, 150 ppb Se was supplemented either as sodium selenite or Se nanoparticles, respectively. In Groups 4, 5, 6 and 7, Se was supplemented as its nanoparticles at 50%, 25%, 12.5% and 6.25% levels respectively i.e. at 75 ppb, 37.5 ppb, 18.75 ppb and 9.375 ppb levels respectively. In Groups 8 and 9, 300 ppb Se was supplemented either as Se nanoparticles or sodium selenite, respectively. Experimental feeding was conducted for a period of 91 days. At the end of the experimental trial, blood samples were collected to analyze the blood serum biochemical profile (serum glucose, serum total protein (TP), serum albumin, serum globulin, serum albumin: globulin ratio [A:G ratio], serum total cholesterol) and humoral immunity. Abstract Background In this study, we designed a novel genetic circuit sensitive to Cd2+, Zn2+ and Pb2+ by mimicking the CadA/CadR operon system mediated heavy metal homeostasis mechanism of Pseudomonas aeruginosa. The regular DNA motifs on natural operon were reconfigured and coupled with the enhanced Green Fluorescent Protein (eGFP) reporter to develop a novel basic NOT type logic gate CadA/CadR-eGFP to respond metal ions mentioned above. A Genetically Engineered Microbial (GEM)-based biosensor (E.coli-BL21:pJET1.2-CadA/CadR-eGFP) was developed by cloning the chemically synthesised CadA/CadR-eGFP gene circuit into pJET1.2-plasmid and transforming into Escherichia coli (E. coli)-BL21 bacterial cells. Results The GEM-based biosensor cells indicated the reporter gene expression in the presence of Cd2+, Zn2+ and Pb2+ either singly or in combination. Further, the same biosensor cells calibrated for fluorescent intensity against heavy metal concentration generated linear graphs for Cd2+, Zn2+ and Pb2+ with the R2 values of 0.9809, 0.9761 and 0.9758, respectively as compared to non-specific metals, Fe3+ (0.0373), AsO4 3− (0.3825) and Ni2+ (0.8498) making our biosensor suitable for the detection of low concentration of the former metal ions in the range of 1–6 ppb. Furthermore, the GEM based biosensor cells were growing naturally within the concentration range of heavy metals, at 37 °C and optimum pH = 7.0 in the medium, resembling the characteristics of wildtype E.coli. Conclusion Finally, the novel GEM based biosensor cells developed in this study can be applied for detection of targeted heavy metals in low concentration ranges (1–6 ppb) at normal bacterial physiological conditions.

In this study, two types of modified cassava starch samples (MCS and MWS) prepared from commercially available native cassava starch (NCS) and native cassava starch extracted using the wet method (NWS) were investigated for the removal of Pb(II) ions from aqueous solutions. MCS and MWS samples were synthesized under acidic conditions using Pluronic 123 as the structure-directing agent and tetraethylorthosilicate (TEOS) as the chemical modifying agent. Modified starch samples were characterized using Fourier transform infrared (FTIR) spectroscopy, thermogravimetric analysis (TGA), X-ray Diffraction (XRD), and a nitrogen (N2) gas adsorption–desorption analyser. MCS and MWS showed enhanced thermal stabilities upon acid hydrolysis and chemical modification. The effects of contact time and initial Pb(II) concentration were studied through batch adsorption experiments. Adsorption kinetics followed the pseudo-second-order kinetic model. The equilibrium adsorption data were analysed and compared by the Langmuir and Freundlich adsorption models. The coefficient correlation (R2) was employed as a measure of the fit. The Langmuir model fitted well with equilibrium adsorption data, giving a maximum Pb(II) adsorption capacity of 370.37 and 294.12 mg/g for MWS and MCS, respectively. Modified samples exhibited a higher desorption efficiency of over 97%. This study demonstrated that modified starch could be utilized for Pb(II) removal from industrial wastewater.

An experiment was conducted on 24 male goat kids (6.80±0.20 kg ABW) to elucidate the effect of supplemental vitamin E and/or selenium yeast on their growth, serum antioxidant enzymes, selenium and ï¡- tocopherol status. Kids were randomly divided into 4 equal groups and fed a basal diet consisted of concentrate mixture and oat straw to meet their nutrient requirement. Group 1 served as control (without any supplementation), whereas animals in groups 2, 3 and 4 were supplemented either 0.3 mg selenium (Se) as Se-yeast, 100 mg of DL-ï¡-tocopheryl acetate or both. This feeding practice lasted for 180 days, during which fortnightly body weights of kids were recorded. Blood was collected at day 0, 60, 120 and 180 of experimental feeding and analysed for ï¡-tocopherol, Se and activity of antioxidant enzymes i.e. catalase (CAT), super oxide dismutase (SOD), glutathione peroxidase (GSHPx) and malonaldialdehyde (MDA) concentration as a measure of lipid per-oxidation (LPO). Results revealed that the activity of erythrocyte GSH-Px and SOD and MDA concentration as a measure LPO and serum Se were significantly higher in all the 3 experimental groups as compared to control. Similarly, the concentration of ï¡- tocopherol in serum was significantly higher in group 2, 3 and 4 as compared to group 1. However, growth rate of the kids and concentration of CAT did not differ among the different treatments. It was concluded that vitamin E and Se-yeast supplementation had no effect on body weight gain, but improved the antioxidant status of the kids in terms of erythrocyte GSH-Px, SOD activity and LPO.
