The development of receptive endometrium (RE) from pre-receptive endometrium (PE) for successful embryo implantation is a complex dynamic process in which the morphology and physiological states of the endometrial epithelium undergo a series of significant changes, including cell proliferation and apoptosis. However, the molecular mechanisms are not yet fully understood. In this study, a higher circRNA3669 level was observed in PE than in RE of goats. Functional assays revealed that this overexpression promoted the proliferation of goat endometrial epithelial cells (GEECs) by activating the expression of genes related to the PI3K/AKT-mTOR and MAPK pathways, thereby inhibiting apoptosis in vitro. Furthermore, circRNA3669 functioned as a competing endogenous RNA (ceRNA) to upregulate Reticulocalbin-2 (RCN2) expression at the post-transcriptional level by interacting with and downregulating miR-26a in GEECs. In addition, RCN2, which is highly expressed in the PE of goats, was found to be regulated by β-estradiol (E2) and progesterone (P4). Our results demonstrated that RCN2 also affected the key proteins PI3K, AKT, mTOR, JNK, and P38 in the PI3K/AKT-mTOR and MAPK pathways, thereby facilitating GEECs proliferation and suppressing their apoptosis in vitro. Collectively, we constructed a new circRNA3669-miR-26a-RCN2 regulatory network in GEECs, which further provides strong evidence that circRNA could potentially play a crucial regulatory role in the development of RE in goats.

Objective: Evidence suggests that calcium release from the endoplasmic reticulum (ER) can be induced to cause calcium overload, which in turn can trigger mitochondrial-dependent apoptosis. Dysregulation of systemic calcium homeostasis and changing levels of calcium-binding proteins have been shown to be associated with the malignant behavior of tumors. However, the precise molecular mechanism underlying Nasopharyngeal carcinoma (NPC) remains uncertain. Methods: Reticulocalbin (RCN2) expression in NPC was assessed using GEO database, western blot analysis and qRT-PCR. Apoptosis was assessed using flow cytometric analysis and the expression levels of apoptosis-related proteins were determined using western blot analysis. Intracellular calcium ion concentrations were measured using fluorescence imaging. The findings from these analyses were validated in vitro using nude mice models. Luciferase and ChIP assays were used to measure transcriptional regulation. Clinical significance was evaluated using tissue microarray analysis (n=150). Results: Our results showed that RCN2 promotes malignancy by causing Ca2+ flow imbalance, which leads to the initiation of the stress-mediated mitochondrial apoptosis pathway. We demonstrate that calreticulin (CALR) resides primarily in the endoplasmic reticulum and interacts with RCN2. Moreover, the transcription factors YY1 and homeobox protein goosecoid (GSC) both contribute to the initiation of RCN2 transcription by directly binding to the predicted promoter region of RCN2. Finally, high expression of RCN2 combined with high expression of GSC and YY1 may serve as an important clinical biomarker of poor prognosis in patients with NPC. Conclusion: YY1 and GSC are upstream regulators of RCN2, involved in mitochondrial calcium overload and stress-induced mitochondrial apoptosis. Moreover, miR-1297 inhibition and RCN2 overexpression could counteract the inhibitory effects of DUXAP8 knockdown on the malignant phenotypes of CC cells. Besides, enhanced RCN2 expression restored the tumor growth in vivo that was inhibited by DUXAP8 repression. Exposure of articular cartilage to excessive mechanical loading is closely related to the pathogenesis of osteoarthritis (OA). However, the exact molecular mechanism by which excessive mechanical loading drives OA remains unclear. In vitro, primary chondrocytes were exposed to cyclic tensile strain at 0.5 Hz and 10 % elongation for 30 min to simulate excessive mechanical loading in OA. In vivo experiments involved mice undergoing anterior cruciate ligament transection (ACLT) to model OA, followed by interventions on Rcn2 expression through adeno-associated virus (AAV) injection and tamoxifen-induced gene deletion. 10 μL AAV2/5 containing AAV- Rcn2 or AAV-sh Rcn2 was administered to the mice by articular injection at 1 week post ACLT surgery, and Col2a1 -creERT: Rcn2 flox/flox mice were injected with tamoxifen intraperitoneally to obtain Rcn2 -conditional knockout mice. Finally, we explored the mechanism of Rcn2 affecting OA. Here, we identified reticulocalbin-2 (Rcn2) as a mechanosensitive factor in chondrocytes, which was significantly elevated in chondrocytes under mechanical overloading. PIEZO type mechanosensitive ion channel component 1 (Piezo1) is a critical mechanosensitive ion channel, which mediates the effect of mechanical loading on chondrocytes, and we found that increased Rcn2 could be suppressed through knocking down Piezo1 under excessive mechanical loading. Furthermore, chondrocyte-specific deletion of Rcn2 in adult mice alleviated OA progression in the mice receiving the surgery of ACLT. On the contrary, articular injection of Rcn2 -expressing adeno-associated virus (AAV) accelerated the progression of ACLT-induced OA in mice. Abstract Background Rice (Oryza sativa) panicle architecture is the major determinant of the ideal plant architecture that directly influence yield potential. Many genes influencing development of primary branches, secondary branches, spikelet and pedicel would also influence panicle architecture, which is thus a complex trait regulated by genes from various aspects. miR156, an extensively studied miRNA, has recently emerged as promising target for crop improvement because of its role in plant architecture regulation, such as the number of tillers, plant height and the panicle architecture. Increasing evidence suggests that miR156 might play an important role in panicle architecture regulation. Main body To study the detailed function of miR156 in rice panicle architecture regulation, we examined the genetic interaction or transcriptional regulation of miR156/OsSPL to other panicle regulating genes. Our results revealed that expression of many panicle related genes were influenced by miR156. Through biochemical analysis, we further proved that miR156 directly regulated the axillary meristem regulating gene, LAX1, at the transcription level. And the intimate relations between miR156 and LAX1, and miR156 and LAX2 were also uncovered by genetic analysis. On the other hand, a tight genetic linkage between miR156 and RCN2, the panicle branch promoting gene, was also detected, which suggested a buffering mechanism for the miR156 mediated panicle architecture regulation. Furthermore, genetic analysis also demonstrated that miR156 functioned in the same pathway with OsRA2 to regulate pedicel length. Short conclusion Altogether, miR156 integrates several genetic pathways mediated by genes such as LAX1, LAX2, RCN2 and OsRA2, and comprehensively regulates panicle development in rice. Endothelial cell pyroptosis is a novel cause of endothelial dysfunction in sepsis. Reticulocalbin-2 (RCN2) is involved in regulating vascular inflammation and plays an important role in the cardiovascular system. However, the role of RCN2 in inflammation-induced endothelial cell pyroptosis remains to be explored. ABSTRACT Objective:To detect the protein expressions of reticulocalbin-2 (RCN2) and pseudopodium-enriched atypical kinase 1 (PEAK1) in the colorectal cancer tissues, to evaluate their correlations with the clinicopathological characteristics and prognosis. Methods: The immunohistochemistry assay was applied to detect the protein expressions of RCN2 and PEAK1 in 90 cases of the colorectal cancer tissues and the adjacent normal colorectal tissues. The correlations of RCN2/PEAK1 expression with the clinicopathological characteristics were analyzed, and the Kaplan-Meier survival curve was utilized to analyze the influence of RCN2/PEAK1 expression on the prognosis. Moreover, the Spearman was used to examine the correlation between the RCN2 and PEAK1 expression. Result: The positive rates of RCN2 and PEAK1 protein in the colorectal cancer tissues were higher than those in the adjacent normal colorectal tissues(P<0.05). In the colorectal cancer tissues, the RCN2 expression was positively correlated with the tumor size, the depth of invasion and the TNM stages (P<0.05), besides, the PEAK1 expression was positively correlated with the depth of invasion, the lymph node metastasis and the TNM stages (P<0.05). The Log Rank results showed that the 5-year overall survival rates in the RCN2-positive group and the PEAK1-positive group were both lower than those in the RCN2-negative group and the PEAK1-negative group, respectively(P<0.05). In the colorectal cancer tissues, there was a positive correlation between the RCN2 and PEAK1 expression (r=0.586, P=0.000).Conclusion: The RCN2 and PEAK1 protein are up-regulated in the colorectal cancer tissues, which was correlated with progression and poor prognosis. Pituitary adenoma (PA) is a common intracranial tumor, and its incidence has been on the rise in recent years. Pituitary tumor not only causes intracranial space occupying signs, but also produces endocrine disorders, such as infertility, sexual dysfunction, facial and limb changes. Moreover, it destroys the internal environment stability and even affects the appearance of a person. However, the mechanism of PA is not fully understood. Previous research has confirmed that the expression or role of long non-coding RNA is closely connected with the occurrence of human diseases. In this study, we discovered that long intergenic non-protein coding RNA 1004 (LINC01004) was aberrantly up-regulated in PA cells. Functional assays manifested that LINC01004 promoted malignant behaviors of PA cells in vitro and growth of PA in vivo. By using bioinformatics tools and a series of mechanism assays, LINC01004 was identified to sponge miR-323a-3p/miR-136–5p to enhance the expression of RCN2 in PA. In conclusion, the results provided in this study revealed a novel regulatory mechanism of LINC01004 in PA, which might be helpful for the treatment of PA and supply a new thought for further research of PA. Abstract: Flowering time and inflorescence architecture are important and inter-relating traits in rice (Oryza sativa L.). Overexpression of each of RCN1 to RCN3 in rice could result in delayed flowering and abnormal inflorescence architecture. From a transferred DNA (T-DNA) insertion population constructed from japonica rice cv. Zhonghua 11, a mutant, A989, exhibiting dense panicle and late flowering was isolated. Genetic and molecular analysis showed that a single copy of T-DNA was inserted into the genome of the mutant, and the T-DNA was inserted into the 330 bp position after RCN2 poly A. Overexpression of RCN2 was observed in the mutant as revealed by reverse transcription-polymerase chain reaction (RT-PCR) test. When RCN2 gene was transferred into wild type driven by double 35S promoters, the transformants showed no panicle due to the suspensions of differentiation and growth of the secondary-branch primordium; however, the transition from vegetative growth to productive growth was normal in the transgenic plant. Vascular calcification (VC) is associated with the high morbidity and mortality of cardiovascular diseases in dialysis patients and is a process in which vascular smooth muscle cells (VSMCs) actively differentiate into osteoblast-like cells. Reticulocalbin-2 (RCN2) is involved in the process of osteogenic differentiation under diabetic conditions, but its regulatory role under hyperphosphatemic conditions and the related mechanisms remain unclear. In this study, the importance of the interactions among RCN2, STAT3 and miR-155-5p during the osteogenic differentiation and calcification of human aortic VSMCs (HASMCs) were investigated. RCN2 was measured in femoropopliteal artery plaque specimens from 6 peripheral arterial disease (PAD) patients with chronic kidney disease (CKD) and 6 PAD patients without CKD. RCN2 protein and mRNA expression were assessed in the high phosphate-induced aortic rings culture ex vivo model. In vitro calcification assays and molecular mechanism studies were performed in HASMCs. Immunohistochemical staining results revealed increased RCN2 expression in the calcified plaques of femoral arteries of patients with CKD and in a high phosphate-induced aortic culture ex vivo model. RCN2 promoted HASMCs osteogenic differentiation and calcification by inducing STAT3 phosphorylation. Furthermore, inhibition of STAT3 activation by cryptotanshinone (CT) promoted miR-155-5p expression in HASMCs. In turn, miR-155-5p inhibited RCN2 mRNA expression, while RCN2 overexpression partially offset the miR-155-5p-mediated inhibition of HASMC calcification, acting as a positive feedback loop. These results demonstrate that RCN2 is a crucial regulator of VC under hyperphosphatemic conditions. RCN2/STAT3/miR-155-5p feedback loop is important in VC and targeting each member of this feedback loop could potentially reverse high phosphate-induced VC. Regulation of calcineurin, a Ca2+/calmodulin-regulated phosphatase, is important for the nervous system, and its abnormal activity is associated with various pathologies, including neurodegenerative disorders. In yeast cells lacking the VPS13 gene (vps13Δ), a model of VPS13-linked neurological diseases, we recently demonstrated that calcineurin is activated, and its downregulation reduces the negative effects associated with vps13Δ mutation. Here, we show that overexpression of the RCN2 gene, which encodes a negative regulator of calcineurin, is beneficial for vps13Δ cells. We studied the molecular mechanism underlying this effect through site-directed mutagenesis of RCN2. The interaction of the resulting Rcn2 variants with a MAPK kinase, Slt2, and subunits of calcineurin was tested. We show that Rcn2 binds preferentially to Cmp2, one of two alternative catalytic subunits of calcineurin, and partially inhibits calcineurin. Rcn2 ability to bind to and reduce the activity of calcineurin was important for the suppression. The binding of Rcn2 to Cmp2 requires two motifs in Rcn2: the previously characterized C-terminal motif and a new N-terminal motif that was discovered in this study. Altogether, our findings can help to better understand calcineurin regulation and to develop new therapeutic strategies against neurodegenerative diseases based on modulation of the activity of selected calcineurin isoforms. Reticulocalbin (RCN) family members could play oncogenic roles in human malignancies and facilitate tumor cell proliferation and metastasis. However, the expression pattern and potential function of Reticulocalbin 2 (RCN2) in colorectal cancer has not been addressed yet. In the present study, we investigated the protein expression of RCN2 by immunohistochemistry assay, and analyzed its association with tumor progression, recurrence and prognosis in 326 cases of patients. Results suggested that the expression of RCN2 was up-regulated in colorectal cancer compared with paired adjacent nontumor specimens. RCN2 expression was closely related to tumor size and the depth of invasion. Kaplan-Meier analysis proved that RCN2 was associated with both disease-free survival and overall survival of patients with colorectal cancer. Moreover, cox's proportional hazards analysis showed that high RCN2 expression was an independent prognostic marker of poor outcome. Consistently, overexpressing RCN2 promoted CRC cell proliferation both in vitro and in vivo and knockdown RCN2 showed the opposite results. These results provided the first evidence that RCN2 level was increased in colorectal cancer and significantly correlated with tumor growth and proliferation. It also indicated that RCN2 might serve as a potential marker of tumor recurrence and prognosis of colorectal cancer.

Candida glabrata is an emerging human fungal pathogen that is frequently drug tolerant, resulting in difficulties in treatment and a higher mortality in immunocompromised patients. The calcium-activated protein phosphatase calcineurin plays critical roles in controlling drug tolerance, hyphal growth, and virulence in diverse fungal pathogens via distinct mechanisms involving survival in serum or growth at host temperature (37° and higher). Here, we comprehensively studied the calcineurin signaling cascade in C. glabrata and found novel and uncharacterized functions of calcineurin and its downstream target Crz1 in governing thermotolerance, intracellular architecture, and pathogenesis in murine ocular, urinary tract, and systemic infections. This represents a second independent origin of a role for calcineurin in thermotolerant growth of a major human fungal pathogen, distinct from that which arose independently in Cryptococcus neoformans. Calcineurin also promotes survival of C. glabrata in serum via mechanisms distinct from C. albicans and thereby enables establishment of tissue colonization in a murine systemic infection model. To understand calcineurin signaling in detail, we performed global transcript profiling analysis and identified calcineurin- and Crz1-dependent genes in C. glabrata involved in cell wall biosynthesis, heat shock responses, and calcineurin function. Regulators of calcineurin (RCN) are a novel family of calcineurin modifiers, and two members of this family were identified in C. glabrata: Rcn1 and Rcn2. Our studies demonstrate that Rcn2 expression is controlled by calcineurin and Crz1 to function as a feedback inhibitor of calcineurin in a circuit required for calcium tolerance in C. glabrata. In contrast, the calcineurin regulator Rcn1 activates calcineurin signaling. Interestingly, neither Rcn1 nor Rcn2 is required for virulence in a murine systemic infection model. Taken together, our findings show that calcineurin signaling plays critical roles in thermotolerance and virulence, and that Rcn1 and Rcn2 have opposing functions in controlling calcineurin signaling in C. glabrata.
