Conservation Agriculture (CA) is based in principles of the minimum soil disturbance, preservation of organic mulch on soil surface and crop rotation. Therefore, this study aimed to compare the CA versus conventional tillage for tomato and broccoli cropping. The experimental treatments were two tillage method (no-tillage and conventional tillage) as the main plot and species of cover crops, legumes, grasses and a mix (legume + grass) as subplot. Soil properties and vegetable agronomic performance were assessment. No-tillage provided higher soil water content and increased soil fertility and higher soil mechanical resistance to penetration than conventional tillage. Broccoli growth and yield were impaired under no-tillage, while tomato had greater root length and similar yield compared to the conventional tillage. Regarding to the cover crops there was no difference on the experimental traits. Conservation Agriculture is recommended to increase soil fertility and soil water storage as well as for ensure sustainable vegetable production.

The COVID-19 pandemic has highlighted the urgent need for the identification of new antiviral drug therapies for a variety of diseases. COVID-19 is caused by infection with the human coronavirus SARS-CoV-2, while other related human coronaviruses cause diseases ranging from severe respiratory infections to the common cold. We developed a computational approach to identify new antiviral drug targets and repurpose clinically-relevant drug compounds for the treatment of a range of human coronavirus diseases. Our approach is based on graph convolutional networks (GCN) and involves multiscale host-virus interactome analysis coupled to off-target drug predictions. Cell-based experimental assessment reveals several clinically-relevant drug repurposing candidates predicted by the in silico analyses to have antiviral activity against human coronavirus infection. In particular, we identify the MET inhibitor capmatinib as having potent and broad antiviral activity against several coronaviruses in a MET-independent manner, as well as novel roles for host cell proteins such as IRAK1/4 in supporting human coronavirus infection, which can inform further drug discovery studies.

Background While ST-Elevation Myocardial Infarction (STEMI) door-to-balloon times are often below 90 min, symptom to door times remain long at 2.5-h, due at least in part to a delay in diagnosis. Objectives To develop and validate a machine learning-guided algorithm which uses a single‑lead electrocardiogram (ECG) for STEMI detection to speed diagnosis. Methods Data was extracted from the Latin America Telemedicine Infarct Network (LATIN), a population-based Acute Myocardial Infarction (AMI) program that provides care to patients in Brazil, Colombia, Mexico, and Argentina through telemedicine. Sample: the first dataset was comprised of 8511 ECGs that were used for various machine learning experiments to test our Deep Learning approach for STEMI diagnosis. The second dataset of 2542 confirmed STEMI diagnosis EKG records, including specific ischemic heart wall information (anterior, inferior, and lateral), was derived from the previous dataset to test the STEMI localization model. Preprocessing: Detection of QRS complexes by wavelet system, segmentation of each EKG record into individual heartbeats with fixed window of 0.4 s to the left and 0.9 s to the right of main. Training & Testing: 90% and 10% of the total dataset, respectively, were used for both models. Classification: two 1-D convolutional neural networks were implemented, two classes were considered for first models (STEMI/Not-STEMI) and three classes for the second model (Anterior/Inferior/Lateral) each corresponding to the heart wall affected. These individual probabilities were aggregated to generate the final label for each model. Results The single‑lead ECG strategy was able to provide an accuracy of 90.5% for STEMI detection with Lead V2, which also yielded the best results overall among individual leads. STEMI Localization model provided promising results for anterior and inferior wall STEMIs but remained suboptimal for Lateral STEMI. Conclusions An Artificial Intelligence-enhanced single‑lead ECG is a promising screening tool. This technology provides an autonomous and accurate STEMI diagnostic alternative that can be incorporated into wearable devices, potentially providing patients reliable means to seek treatment early and offers the potential to thereby improve STEMI outcomes in the long run.

Phosphoinositides (PIPs) are key regulators of membrane traffic and signaling. The interconversion of PIPs by lipid kinases and phosphatases regulates their functionality. Phosphatidylinositol (PI) and PIPs have a unique enrichment of 1-stearoyl-2-arachidonyl acyl species; however, the regulation and function of this specific acyl profile remains poorly understood. We examined the role of the PI acyltransferase LYCAT in control of PIPs and PIP-dependent membrane traffic. LYCAT silencing selectively perturbed the levels and localization of phosphatidylinositol-4,5-bisphosphate [PI(4,5)P2] and phosphatidylinositol-3-phosphate and the membrane traffic dependent on these specific PIPs but was without effect on phosphatidylinositol-4-phosphate or biosynthetic membrane traffic. The acyl profile of PI(4,5)P2 was selectively altered in LYCAT-deficient cells, whereas LYCAT localized with phosphatidylinositol synthase. We propose that LYCAT remodels the acyl chains of PI, which is then channeled into PI(4,5)P2 Our observations suggest that the PIP acyl chain profile may exert broad control of cell physiology.

Lysosomes are terminal, degradative organelles of the endosomal pathway that undergo repeated fusion-fission cycles with themselves, endosomes, phagosomes, and autophagosomes. Lysosome number and size depends on balanced fusion and fission rates. Thus, conditions that favour fusion over fission can reduce lysosome numbers while enlarging their size. Conversely, favouring fission over fusion may cause lysosome fragmentation and increase their numbers. PIKfyve is a phosphoinositide kinase that generates phosphatidylinositol-3,5-bisphosphate to modulate lysosomal functions. PIKfyve inhibition causes an increase in lysosome size and reduction in lysosome number, consistent with lysosome coalescence. This is thought to proceed through reduced lysosome reformation and/or fission after fusion with endosomes or other lysosomes. Previously, we observed that photo-damage during live-cell imaging prevented lysosome coalescence during PIKfyve inhibition. Thus, we postulated that lysosome fusion and/or fission dynamics are affected by reactive oxygen species (ROS). Here, we show that ROS generated by various independent mechanisms all impaired lysosome coalescence during PIKfyve inhibition and promoted lysosome fragmentation during PIKfyve re-activation. However, depending on the ROS species or mode of production, lysosome dynamics were affected distinctly. H2O2 impaired lysosome motility and reduced lysosome fusion with phagosomes, suggesting that H2O2 reduces lysosome fusogenecity. In comparison, inhibitors of oxidative phosphorylation, thiol groups, glutathione, or thioredoxin, did not impair lysosome motility but instead promoted clearance of actin puncta on lysosomes formed during PIKfyve inhibition. Additionally, actin depolymerizing agents prevented lysosome coalescence during PIKfyve inhibition. Thus, we discovered that ROS can generally prevent lysosome coalescence during PIKfyve inhibition using distinct mechanisms depending on the type of ROS.

Lysosomes are the terminal degradative compartment of autophagy, endocytosis and phagocytosis. What once was viewed as a simple acidic organelle in charge of macromolecular digestion has emerged as a dynamic organelle capable of integrating cellular signals and producing signal outputs. In this review, we focus on the concept that the lysosome surface serves as a platform to assemble major signaling hubs like mTORC1, AMPK, GSK3 and the inflammasome. These molecular assemblies integrate and facilitate cross-talk between signals such as amino acid and energy levels, membrane damage and infection, and ultimately enable responses such as autophagy, cell growth, membrane repair and microbe clearance. In particular, we review how molecular machinery like the vacuolar-ATPase proton pump, sestrins, the GATOR complexes, and the Ragulator, modulate mTORC1, AMPK, GSK3 and inflammation. We then elaborate how these signals control autophagy initiation and resolution, TFEB-mediated lysosome adaptation, lysosome remodeling, antigen presentation, inflammation, membrane damage repair and clearance. Overall, by being at the cross-roads for several membrane pathways, lysosomes have emerged as the ideal surveillance compartment to sense, integrate and elicit cellular behavior and adaptation in response to changing environmental and cellular conditions.

Revisar a literatura, de modo sistematizado, para esclarecimento acerca do uso de produtos naturais contra Streptococcus mutans. Material e Métodos: Foi realizada uma busca nas bases de dados PubMed, Literatura Latino-Americana e do Caribe em Ciências da Saúde (LILACS) e Bibliografia Brasileira de Odontologia (BBO) via Biblioteca Virtual em Saúde (BVS) e Biblioteca Eletrônica Científica Online (SciELO). Não foram feitas restrições quanto ao ano nem ao idioma. Empregaram-se os descritores padronizados (Medical Subject Headings – MeSH) “Streptococcus mutans”, “natural products” e “produtos naturais”, conforme a base consultada, com uso dos operadores booleanos “AND” para que se fizesse uma combinação entre o microrganismo e os produtos naturais. Resultados: De um total de 83 textos identificados, foram lidos e analisados na íntegra 38 deles. Em todos os artigos incluídos foi constatado o uso de diversos produtos naturais contra a ação de S. mutans, e a maioria teve ótima ação contra o microrganismo. Conclusão: Os produtos naturais incluídos e analisados nessa revisão podem ser empregados contra S. mutans e, assim, auxiliar na prevenção da cárie dentária.

Estuarine environments are suggested to be the final receivers of human pollution and are impacted by surrounding urbanization and compounds carried by the river waters that flow from the continent. Polycyclic aromatic hydrocarbons (PAHs) are among the contaminants that can reach estuaries and can directly affect marine conservation, being considered highly deleterious to organisms living in these environments. This research investigated the meiofauna of three estuaries exposed to different levels of urbanization and consequently different levels of PAH concentrations, in order to assess how these compounds and environmental factors affect the distribution, structure and diversity of these interstitial invertebrates. A total of 15 major meiofauna groups were identified, with Nematoda being the dominant taxon (74.64%), followed by Copepoda (9.55%) and Polychaeta (8.56%). It was possible to observe significant differences in all diversity indices studied in the estuaries. With the exception of average density, the diversity indices (richness, Shannon index and evenness) were higher in the reference estuary, Goiana estuarine system (GES). On the other hand, the Timbó estuarine system (TES) had the lowest Shannon index value and richness, while the Capibaribe estuarine system (CES) had the lowest evenness value. The latter two estuaries (TES and CES) presented intermediate and high levels of urbanization, respectively. The ecological quality assessment (EcoQ) in the studied estuaries was classified from Poor to Moderate and the estuary with the lowest demographic density in its surroundings, GES, showed a better ecological quality (Moderate EcoQ). A significant distance-based multivariate linear modelling regression (DistLM) was observed between the environmental variables and the density of the meiobenthic community, where PAHs and pH were the main contributors to organism variation. The sediments were characterized by predominance of very fine sand and silt-clay in the most polluted environments, while the control site environment (GES) was dominated by medium grains. The highest concentrations of PAHs were found in the most urbanized estuaries, and directly affected the structure of the interstitial benthic community. The metrics used in the present study proved to be adequate for assessing the environmental quality of the investigated estuaries.

The secondary production is the result of the functional response of populations subject to various environmental factors. Marine habitats vary in terms of quantity and quality of food supply, and the use of secondary production values, as well as renewal rates (P/B), may be used as estimates for understanding the incorporation of organic matter and energy per unit, population or community in each area. This estimative was performed for the population of Scolelepis goodbodyi in a tropical beach in the Southwestern Atlantic, located in an upwelling area. A comparison of Spionidae and non-spionid populations from different latitudes was also done. The Mass Specific Growth Rate method (MSGR) and the Production/Biomass ratio (P/B) were used to estimate the somatic annual production and average annual biomass. The mean density and biomass were 16.38 ind. m -2 and 2.78 g AFDW m -2 ,respectively. The secondary production and P/B were 8.3 g AFDW m -2 y -1 and 2.98 y 1 , respectively. The growth rate in weight was greater for the small size than the large size classes. The largest individuals (W3C = 1.0 mm) showed the lowest biomass and secondary production values. The observed high rates of secondary production and P/B suggest that this S. goodbodyi population can transfer large amounts of biomass to higher trophic levels of the local food web. Studies of the secondary production of spionidae populations in different latitudes, including the population of S . goodbodyi in the beach of Manguinhos, showed variability in their rates probably due to the differences of several factors such as life history and environmental variability

Background In the GLOBAL LEADERS trial, ticagrelor monotherapy beyond 1 month compared with standard antiplatelet regimens after coronary stent implantation did not improve outcomes at intention‐to‐treat analysis. Considerable differences in treatment adherence between the experimental and control groups may have affected the intention‐to‐treat results. In this reanalysis of the GLOBAL LEADERS trial, we compared the experimental and control treatment strategies in a per‐protocol analysis of patients who did not deviate from the study protocol. Methods and Results Baseline and postrandomization information were used to classify whether and when patients were deviating from the study protocol. With logistic regressions, we derived time‐varying inverse probabilities of nondeviation from protocol to reconstruct the trial population without protocol deviation. The primary end point was a composite of all‐cause mortality or nonfatal Q‐wave myocardial infarction at 2 years. At 2‐year follow‐up, 1103 (13.8%) of 7980 patients in the experimental group and 785 (9.8%) of 7988 patients in the control group qualified as protocol deviators. At per‐protocol analysis, the rate ratio for the primary end point was 0.88 (95% CI, 0.75–1.03; P =0.10) on the basis of 274 versus 325 events in the experimental versus control group. The rate ratio for the key safety end point of major bleeding was 1.00 (95% CI, 0.79–1.26; P =0.99). The per‐protocol and intention‐to‐treat effect estimates were overall consistent. Conclusions Among patients who complied with the study protocol in the GLOBAL LEADERS trial, ticagrelor plus aspirin for 1 month followed by ticagrelor monotherapy was not superior to 1‐year standard dual antiplatelet therapy followed by aspirin alone at 2 years after coronary stenting. The design and synthesis of a microporous construct based on the entrapment of an emissive fluorescein derivative in a zinc 2-methylimidazolate (ZIF-8) metal-organic framework (MOF) is detailed. Synthesis of the MOF in the presence of a fluorophore enables the capture and dispersal of dye molecules within the ZIF-8 framework. Within the resulting supramolecular assemblies, the fluorophore components show excellent photophysical properties such as high emission and increased fluorescence lifetime, despite the tendency of the dye to undergo aggregation-caused quenching in the solid-state, as well as a 4-fold enhancement of the fluorophore’s photostability. The demonstration that supramolecular events can be invoked to construct solid fluorescent systems from separate components is realized. The encapsulation of the fluorescein in an enclosed subunit of the ZIF-8 framework was modelled using the density-functional tight-binding method. Furthermore, the fluorophore@MOF composite can be internalized by mammalian macrophage cells and transported to lysosomes without disrupting cell viability. In principle, this simple protocol can evolve into a general strategy for intracellular delivery of functional molecular components for targeted bioimaging or theranostic applications.

O uso de metodologias ativas no processo de ensino-aprendizagem vem sendo amplamente difundido em muitas instituições de ensino e a Odontologia não foge desse contexto. Além de trazer aos estudantes uma visão mais global, oferece mais autonomia individual e consciência coletiva, uma vez que o trabalho mais ativo, com problematizações, é benéfico para o desenvolvimento dos mesmos. Frente a isso, aqui é relatada a experiência da produção de um vídeo em 360º, de uma situação rotineira em Odontologia: o preparo do paciente e a lavagem das mãos em centro cirúrgico. O material elaborado se propõe a servir como ferramenta para que o estudante possa assisti-lo previamente às práticas subsequentes, pois assim terá a possibilidade de estar mais adaptado, o que parece favorecer a execução de algumas atividades odontológicas. A ferramenta aqui apresentada é um recurso novo que pode ser combinado com métodos tradicionais de ensino e seu impacto deve ser avaliado.

Os pacientes que necessitam de tratamento endodôntico geralmente têm uma expectativa negativa em relação a esse procedimento. Objetivo: Avaliar a expectativa e a percepção de pacientes que realizaram o tratamento endodôntico na clínica de especialização em Endodontia ou no Centro de Especialidades Odontológicas da Universidade Positivo. Material e métodos: A amostra de conveniência contou com 123 pacientes, que foram atendidos no período de setembro de 2016 a outubro de 2017. Eles responderam a um questionário com perguntas referentes a gênero, idade, expectativas quanto ao tratamento endodôntico, presença de dor e também questões relacionadas à experiência durante e após a realização do tratamento endodôntico, bem como ao atendimento prestado. Os dados foram tabulados e expressos em frequência e porcentagem. Depois utilizou-se o teste de Qui-quadrado, por meio do programa SPSS, versão 21.0. Resultados: A maioria dos pacientes eram mulheres, entre 41 e 50 anos, que já haviam realizado tratamento endodôntico anteriormente. A ausência de dor foi relatada por 76% dos pacientes, e 50% não tiveram medo durante o tratamento endodôntico. A anestesia, as agulhas e limas representaram os fatores que mais causaram medo nos pacientes. Para 28,5% a experiência foi como imaginavam; para 70,7% foi melhor do que pensavam. Dos respondentes, 93,5% consideraram o atendimento ótimo. Conclusão: O sentimento de medo antes do tratamento endodôntico foi relevante e agravado pela visualização de materiais e instrumentais. Entretanto a ausência de dor durante a intervenção favoreceu positivamente a percepção pós-operatória, com diminuição do relato de medo entre os pacientes.

The 143 low- and middle-income countries (LMICs) of the world constitute 80% of the world’s population or roughly 5.86 billion people with much variation in geography, culture, literacy, financial resources, access to health care, insurance penetration, and healthcare regulation. Unfortunately, their burden of cardiovascular disease in general and acute ST-segment–elevation myocardial infarction (STEMI) in particular is increasing at an unprecedented rate. Compounding the problem, outcomes remain suboptimal because of a lack of awareness and a severe paucity of resources. Guideline-based treatment has dramatically improved the outcomes of STEMI in high-income countries. However, no such focused recommendations exist for LMICs, and the unique challenges in LMICs make directly implementing Western guidelines unfeasible. Thus, structured solutions tailored to their individual, local needs, and resources are a vital need. With this in mind, a multicountry collaboration of investigators interested in LMIC STEMI care have tried to create a consensus document that extracts transferable elements from Western guidelines and couples them with local realities gathered from expert experience. It outlines general operating principles for LMICs focused best practices and is intended to create the broad outlines of implementable, resource-appropriate paradigms for management of STEMI in LMICs. Although this document is focused primarily on governments and organizations involved with improvement in STEMI care in LMICs, it also provides some specific targeted information for the frontline clinicians to allow standardized care pathways and improved outcomes.

Thus, many farmers who use cover crops attempt to reduce the use of agricultural inputs, especially herbicides. Therefore, our objectives were to study the potential of different cover crop species to suppress weeds and produce an in situ organic mulch and to evaluate the effect of the organic mulch with and without spraying glyphosate on weed suppression for vegetable (tomato [Solanum lycopersicum L.] and broccoli [Brassica oleracea L. var. botrytis]) growth and yield. Five cover crop treatments (sunn hemp [Crotalaria juncea L.], jack bean [Canavalia ensiformis (L.) DC.], pearl millet [Pennisetum glaucum (L.) R. Br.], forage sorghum [Sorghum bicolor (L.) Moench ssp. bicolor], and no cover crop [control]) were used in the main plots; and glyphosate was or was not sprayed on the flattened cover crop in the subplots of this split-plot experimental design. Organic mulch from pearl millet, sorghum, and sunn hemp resulted in lower weed biomass during the early seasons of both tomato and broccoli than mulch from jack bean and no cover crop (control). Spraying glyphosate after roller-crimping reduced weed biomass by 103 g m-2 and 20 g m-2 by 45 and 60 d after transplanting (DATr) of tomato, respectively, and resulted in a better tomato yield compared with not spraying. Glyphosate reduced weed biomass by 110 g m-2 in the early season of broccoli (30 DATr) but did not affect yield. Terminating high-biomass cover crops with a roller-crimper is a promising technique for weed management in vegetable crops that has the potential to reduce or even eliminate the need for herbicide. Maldanids are tube-building polychaetes, known as bamboo-worms; inhabit diverse marine regions throughout the world. The subfamily Euclymeninae was proposed to include forms with anal and cephalic plates, a funnel-shaped pygidium, and a terminal anus. Euclymene, the type genus of Euclymeninae, has about 18 valid species. Euclymene vidali sp. nov. is defined and members of the species described from Northeastern Brazil. Members of this species have 23 chaetigers, and one pre-pygidial achaetous segment; nuchal grooves extend through three quarters of the cephalic plate, and there is one acicular spine with a denticulate tip. Euclymene africana, and E. watsoni, are here recognized, respectively, as Isocirrus africana comb. nov., and I. watsoni comb. nov. Three monotypic genera are invalid: Macroclymenella, Eupraxillella, and Pseudoclyemene; their species should be recognized as Clymenella stewartensis com. nov., Praxillella antarctica com. nov., and Praxillela quadrilobata com. nov., respectively. An identification key and a comparative table for all species of Euclymene are provided. A comparative table for all genera of Euclymeninae is also furnished. The paraphyletic status of Euclymene and Euclymeninae is discussed. The taxon Maldanoplaca is not code compliant and should only be regarded as an informal name.
