Introduction: Idiopathic generalised epilepsy (IGE) refers to a group of epilepsies resulting from the activation of neurons in the whole brain. This study aimed to evaluate the metabolite changes in thalamus as diagnostic biomarkers in IGE patients compared to healthy individuals using magnetic resonance spectroscopy (MRS) technique. Methods: The MRS was performed on 35 IGE patients (26 women and 11 men) with average age of 32 (ranged from 18 to 43) and 35 healthy individuals (13 women and 22 men) with average age of 31 (ranged from 21 to 50) as the control group. The levels of N-acetylaspartate (NAA), creatine (Cr) and choline (Cho) were measured using MRS. The NAA/Cr and NAA/Cho ratios were calculated for all participants. These values were statistically compared using t-test between the groups. Results: The NAA had significant lower values in IGE patients, 9.6 (SD = 0.8) and 9.9 (SD = 0.7) for right and left thalamus, respectively, compared to 10.9 (SD = 0.9) and 10.7 (SD = 0.9) in control group. The Cr values in the left side of thalamus were significantly higher in IGE patients (6.7 [SD = 0.8] versus 5.8 [SD = 0.5]); however, there was no difference in right thalamus. Measurements showed no difference for amounts of Cho between the groups in both sides of thalamus. The NAA/Cr ratio was 1.48 (SD = 0.14) and 1.48 (SD = 0.16) for right and left thalamus, respectively, in IGE patients in comparison with 1.83 (SD = 0.2) and 1.86 (SD = 0.26) in controls. There was no meaningful variation between the NAA/Cho ratio of the right and left thalamus among the groups. Conclusion: Thalamic NAA, Cr and NAA/Cr ratio values in IGE patients showed statistical differences compared to healthy individuals. Evaluating metabolites variations in thalamus using MRS is suggested for differentiating IGE patients from healthy individuals. [ABSTRACT FROM AUTHOR]

Introduction : Apoptosis is one of the most important biological processes, which occurs through the activation of intracellular cell death pathway. Aim : The aim of this study was to determine the pattern of cell death within the early stages of spinal cord injury (SCI) and the evaluation of the changes in Bax protein expression of progressive apoptosis. Material and methods : 48 adult male Sprague Dawley rats were randomly divided into two control and experimental groups. Animals were anesthetized and then the laminectomy procedure was performed in the area of T6–8. On days 1, 7, 14 and 28 after the surgery, one-third of the middle part of their testis tissue were taken for histological and immunohistochemical analysis. Results : The immunohistochemical analysis indicated the presence of TUNEL- positive cells and cells containing the pro-apoptotic protein Bax in testicular sperms in the 1st day after SCI, as well as increased on 28 days. Discussion : 1 day after SCI, the apoptosis occurred in testicular sperm lines as well as the apoptosis can be related to Bax protein expression. Therefore inhibition of caspase activity via caspase cascades-mediated apoptosis may have a protective role in testicular injury during the acute phase of SCI. Conclusions : Our finding suggested that the damage and destruction after SCI can be controlled by therapeutic interventions at the appropriate time before the destructive changes of SCI. In order to evaluate the correlation of B7-H4 and B7-H1 with renal cell carcinoma (RCC), we analyzed B7-H1 and B7-H4 expressions and their clinical significance by immunohistochemical method. Our result indicated that B7-H4-positive staining was detected in 58.13 % of RCC tissues (25 tissues tumors), and there were 18 tissues of patients without detectable B7-H4. Furthermore, 21 cases (48.83 %) were B7-H1-positive. Positive tumor expressions of B7-H4 and B7-H1 were markedly related to advanced TNM stage ( P = 0.001; P = 0.014), high grade ( P = 0.001; P = 002), and larger tumor size ( P = 0.002; P = 024) in RCC tissues than patients with B7-H4-negative and B7-H1-negative in RCC tissues. The patients with B7-H1 and B7-H4-positive expressions were found to be markedly correlated with the overall survival of the patients ( P < 0.05) and tended to have an increased risk of death when compared with negative expression groups. Univariate analysis showed that B7-H4 and B7-H1 expressions, TNM stage, high grade, and tumor size were significantly related to the prognosis of RCC. Furthermore, multivariate analysis showed that B7-H4 and B7-H1 expressions decreased overall survival. The adjusted HR for B7-H1 was 2.83 (95 % CI 1.210-2.971; P = 0.031) and also was 2.918 (95 % CI 1.243-3.102; P = 0.006) for B7-H4 that showed these markers were independent prognostic factors in RCC patients. The expressions of B7-H1 and B7-H4 in RCC patients indicate that these markers may be as a predictor of tumor development and death risk. Further investigations can be helpful to confirm B7-H1 and B7-H4 roles as an independent predictor of clinical RCC outcome. [ABSTRACT FROM AUTHOR]

Introduction Radiation protection is an important safety issue for radiographers and patients. The aim of this study was to assess the observance of radiation protection regulations in radiology departments of Kermanshah University of Medical Sciences, Kermanshah, Iran. Materials and Methods In total, 48 radiographers and 8 radiography rooms were evaluated in three hospitals of Kermanshah, Iran. Additionally, 120 patients were randomly selected in the present study. For data collection, a questionnaire on radiation protection devices, radiographers, and patients was completed. Data were analyzed, using Microsoft Excel. Results Based on the analysis, 56.8% of radiation protection devices were accessible to radiographers. Overall, 81.3% of radiographers stated that they utilized film badges for radiographic procedures, while only 71.7% had used these badges in practice. Additionally, 54.2% of radiographers claimed that they regularly performed medical check-ups; however, based on the documents available at personnel offices, only 43.8% had taken this measure into account. Also, 60.4% of radiographers claimed that they had participated in annual training courses, while based on the records, only 41.7% had participated in such courses. Objective: The purpose was to study the impact of anthropometrical parameters on portal vein diameter and liver size by ultrasound in a subset of Karachi population. Methods: Four hundred and fifty nine apparently healthy subjects were included in this cross sectional study. After recording weight and height of each subject, Portal vein diameter and both liver lobes were measured by gray scale ultrasonography. Students T test and ANOVA were applied for statistical analyses. Results: With increasing age, portal vein diameter and right lobe of liver increased significantly (p value < 0.001). Increase in portal vein diameter was also observed with rise in body mass index (0.8 cm in underweight - 1.1 cm in obese subjects). Sizes of right and left liver lobes also increased with a rise in body mass index (p value < 0.001 and 0.001). Gender, however, did not have any effect on portal vein diameter and liver size. Conclusion: Age and body mass index are reliable parameters to consider for avoiding false positive diagnosis of hepatomegaly and portal hypertension. Knowing the right and left liver size with respect to anthropometrical measurements also assist a clinician in selecting a subject for liver transplantation. [ABSTRACT FROM AUTHOR]

Introduction : Acute myeloid leukemia (AML) is a type of cancer that affects the blood and bone marrow, characterized by the rapid growth of abnormal white blood cells. Aim : The aim of the present study was to evaluate the inhibitory effect of human embryonic stem cell – conditioned medium (hESC-CM) on the proliferation of human leukemic HL-60 cells. Material and methods : We measured proliferation of myeloid leukemia cell by XTT assay. Also cell cycle and apoptosis was measured by flow cytometry after the cells stained. Results and discussion : Our experiment indicated that the human embryonic stem cells conditioned medium has anti-proliferative effects on the progression of HL60 cell. HL60 cells were treated with conditioned medium; the results of the flow cytometry demonstrate that hESC-CM was capable of increasing apoptosis of HL60 cells and inhibit the cell cycle progression. Conclusions : Our results show that Human embryonic stem cell conditioned medium contain factors that are able to inhibit the growth and inducing apoptosis of HL60 cells, which may represent a novel therapeutic approach for leukemia.
